Objective To observe the influence of Weikang No.3 on the expression of neurotensin(NT) and calcitonin gene-related peptide(CGRP) in the gastric antrum, duodenum and brain tissues of FD model mice, and to explore the mechanism of Weikang No.3 in treatment of FD from the angle of brain gut axis. Methods Forty Kunming mice were selected and 10 were randomly selected as the control group. The remaining mice were subjected to unpredictable chronic stress to prepare the FD models. Thirty FD model were successfully established and they were randomly divided into the model group, domperidone group, and Weikang No. 3 group. The mice in the domperidone group and Weikang No. 3 group were given domperidone and Weikang No. 3 [7. 56 mL/( kg·d) ] by gavage, respectively, and the mice in the model group were given the same amount of distilled water for 7 days. Mice in the control group were fed a normal diet,no modeling. The day after the last gavage, gastric antrum, duodenum and brain tissues were taken after anesthesia, and the expression of NT and CGRP receptors were detected by immunohistochemical SP method. Results The relative expression levels of NT and CGRP receptors in the gastric antrum, duodenum and brain tissues of the model group were all higher than those of the control group( all P < 0. 01). The relative expression of NT and CGRP receptors in the gastric antrum, duodenum and brain tissues of Weikang No. 3 group and domperidone group decreased significantly as compared with that of the model group( all P < 0. 01). No significant difference was found in the expression levels of NT and CGRP receptors in different tissues between the Weikang No. 3 group and domperidone group( all P > 0. 05). Conclusion Weikang No. 3 can downregulate the expression of NT and CGRP receptors in the gastric an-trum,duodenum and brain tissues of FD model mice, which might be one of the mechanisms in treatment of FD.
