10. 多态性 BoLA Ⅰα1α2 与恒定链结合及在真核细胞共定位.

- Abstract
Objective: To demonstrate the polymorphism of α chain of bovine major histocompatibility complex (BoLA) class Ⅰmolecule and domain binding constant chain (Ii). Methods: Total 75 BoLA Iα genes were obtained from three Huaibei cattle and analyzed by molecular biology software; the genes of typical BoLA Iα domains and Ii were cloned, and then inserted into prokaryotic expression plasmid. After induced protein expression; the domains of BoLA Ⅰα chain binding to Ii were detected by pull-down method and Western blot. The recombinant eukaryotic expression plasmids were constructed and the co-localization of BoLA Iα segments with Ii was observed by laser confocal microscopy. Results: Firstly, it was found that there were at least 5 kinds of BoLA Iα in the cloned gene sequence, which were highly polymorphic and they were mainly distributed in the antigen peptide binding region (PBR) of BoLA Ⅰ (α1α2) and cytoplasmic region. Secondly, the prokaryotic recombinant plasmids containing BoLA Ⅰα1α2α3, BoLA Ⅰα1α2 or BoLA Ⅰ α3 were constructed, then they were respectively induced to express and purified, in which, the BoLA Ⅰ α1α2α3 and BoLA Ⅰα1α2 had the activity of binding to Ii. Finally, in 293T cells BoLA Ⅰα1α2α3 or BoLA Iα1α2 was found that could co-localize with Ii, while a single BoLA Iα3 could not. Conclusion: BoLA Ⅰα gene is highly polymorphic. BoLA Ⅰα1α2 is a functional fragment that binds to Ii and co-locates intracellular. [ABSTRACT FROM AUTHOR]

- Abstract
With the increasing proportion of wind power connected to the power system, accurate wind power prediction becomes more and more important. Therefore, a short-term wind power prediction model based on fuzzy entropy and complete ensemble empirical mode decomposition with adaptive noise (CEEMDAN) was proposed. The complete ensemble empirical mode decomposition was used to decompose the original wind power series to obtain a series of sub-series with different frequencies. The fuzzy entropy (FE) algorithm was then used to identify the characteristics of each frequency component, and the subsequence components were classified into high and medium frequency component classes and trend terms. The trend term was the low frequency component, which had the characteristics of smoothness and low volatility, and the sparrow search algorithm (SSA) was used to optimize the support vector regression (SVR) for prediction. The high and medium frequency components had high volatility and complex characteristics, and SSA was used to optimize long short-term memory (LSTM) and attention mechanism (AM) were introduced to better assign weights to important information. Finally, the analysis of experimental results show that the proposed method has higher accuracy in wind power prediction. [ABSTRACT FROM AUTHOR]

An accurate extraction of the crop spatial distribution is of great significance for the decision-making on management measures in modern agriculture. Fortunately, the remote sensing images can be widely used as the important data sources for the spatial distribution of crops at present. It is a high demand to extract the high-quality features from the spatial distribution of crops using the remote sensing images. In this study, the Sentinel-2A images were selected to extract the high-precision spatial distribution of winter wheat, in order to avoid the data scale reduction and feature fusion. Firstly, the red edge resource was utilized to classify the important features of winter wheat. The visible light and red edge bands were also combined to effectively reduce the misclassification of pixels for the high accuracy. A downscale model Red Edge Down Scale (REDS) was then established to balance the spatial scale of the data in the Sentinel-2A images, due to the different band resolution between the red edge (20m) and the visible light (10m). The generative countermeasure network was constructed using the three red edge bands of B5, B6 and B7. More importantly, the spatial resolution of B11 shortwave infrared band was reduced from 20 to 10 m, in order to obtain the better consistence in the spatial resolution of visible light and red edge band. The edge blur of image was also prevented from the interpolation (nearest neighbor interpolation). Secondly, the inputs of REDS consisted of the low- and high-resolution channel, correspondingly to the spectral and texture information, respectively. The spatial structure information was then input from the high- into the low-resolution channel. As such, the improved model was achieved in the image data from the high-resolution red edge and short-wave infrared (SWIR) channel. Secondly, the original data was extracted, and then combined into the basic input data, including the three red edge bands after scaling down, the visible light band with a resolution of 10m, and three remote-sensing index products, namely Enhanced Vegetation Index (EVI), Normalized Difference Vegetation Index (NDVI), and Normalized Difference Red-Edge1 (NDRE1). Thirdly, the semantic feature extraction model was constructed as the Red Edge and Vegetation Index Feature Network (REVINet) using convolutional neural network. The coding and decoding units were constructed in the REVINet model using residual network. The linear model was used to fuse the multi-scale features for the output by the decoding units. SoftMax function was used as a classifier for the pixel-by-pixel classification. Finally, the segmentation, and the spatial distribution of winter wheat were generated to verify the REVINet model, compared with the ERFNet, U-Net, and RefineNet models. The experimental results show that the smoother contour edge was extracted from the planting area of winter wheat, particularly with the less misclassification. Meanwhile, the recall (92.15%), precision (93.74%), accuracy (93.09%), and F1 score (92.94%) were better than the rest models, indicating the ideal performance. The spatial distribution of the whole research area demonstrated that the winter wheat in China was mainly distributed in the south of the Great Wall in 2022. The relatively high accuracy of extracted areas was achieved with the better coincidence degree, compared with the standard released by the National Statistical Department in 2021. Therefore, the data organization and feature extraction can be expected to serve as the spatial distribution of winter wheat using the Sentinel-2A. Hemicellulose is a rich renewable resource, which is one of the most promising raw materials for the production of biofuels. α-L-arabinofuranosidase(EC 3.2.1.55) is an auxiliary enzyme in hemicellulose hydrolase system which catalyzes the α-L-1,2, α-L-1,3 and α-L-1.5-arabinofuranoside residues in non-reducing terminal of various oligosaccharides and polysaccharides. Bacillus pumilus is a widely used, bio-friendly feed microbial strain that can degrades mannan, xylan, cellulose and so on. Therefore, cloning the α-L-arabinofuranosidase gene xyn43 from B. pumilus and recombinant expression is beneficial to the separation and purification of the enzyme and it can be applied to the hydrolysis of other hemicellulose polysaccharide. In this study, we used the E. coli BL21 expression system to express the α-L-arabinofuranosidase gene xyn43 isolated from the B. pumilus and then analyzed the enzymatic properties of recombinant enzyme Xyn43. And we used Xyn43 and commercial Xylanase derived from the mutant strain of Aspergillve clavatus to degrade the oat spelt xylan. The results showed that the optimal temperature of Xyn43 was 50 °C and the optimum pH was 6.0. It was stable over a pH range of 5.0 -10.0 and temperatures of 45-55 °C. Compared with Xylanase alone, the reducing sugar content in the hydrolyzate of Xyn43 and Xylanase added simultaneously and Xyn43 added after Xylanase increased 16% and 20% respectively, and the xylose content increased of 35% and 48%. The studies indicate that Xyn43 is able to synergistically with commercial Xylanase for oat spelt xylan degradation and can improve the hydrolysis efficiency, produced more xylosaccharides, arabinose and xylose.
