This paper summarized the treatment and nursing experience of a patient with late-stage acute lactational mastitis. Nursing measures included Traditional Chinese Medicine external application, nursing evaluation, massage for milk lactation, dressing change, health education. After intervention, patient had resumed breastfeeding and wound was well-healed, representing the characteristic of disease prevention in Traditional Chinese Medicine. Objective: To analyze the pregnancy weight gain and glycolipid levels during pregnancy in normal pregnant women who delivered macrosomia without pregnancy complications, obesity, and advanced age, and explore their influence on the risk of macrosomia. Methods: Normal pregnant women who underwent regular prenatal examination were selected as the research object in the International Peace Maternal and Child Health Hospital from September 2020 to December 2020, and divided into macrosomia group (104 cases) and normal control group (258 cases) according to neonatal weight. The difference of basic situation and the levels of glucose and lipid metabolism were compared, and Logistic regression models were adopted to explore the risk factors of macrosomia in normal pregnant women. Results: Compared with the normal control group, the macrosomia group had higher levels of fasting blood glucose, triglyceride (TG) and lower levels of high density lipoprotein (HDL) at all stages of pregnancy (P＜0.05), there are lager difference of TG level and smaller difference of HDL level in macrosomia group (both P＜0.05). Compared with those who gained proper weight during pregnancy, those who gained too much weight had 142% increased risk of delivery of macrosomia (OR=2.42, 95%CI: 1.34-4.39) and those who had adequate weight gain had 73% decreased risk of delivery of macrosomia (OR=0.27, 95%CI: 0.10-0.77). For every 1 mmol/L increase in HDL in late pregnancy, the risk of delivery of macrosomia decreased by 79% (OR=0.21, 95%CI: 0.08-0.57). Conclusions: Normal pregnant women should control their weight gain strictly during pregnancy to reduce the risk of macrosomia, and lipid metabolism needs to be paid attention to, in which the level and change of HDL in late pregnancy may assist in the screening of cryptogenic macrosomia. 【Objective】 To identify the pathogen species causing gray mold on facility tomato. 【Methods】 Suspected gray mold disease samples were isolated from tomato cultivation plots in three facilities in Hohhot City and Ulanqab City, Inner Mongolia. Morphological and molecular biology methods were used to identify the isolated strains. The pathogenicity of the pathogens was determined and re-isolated on detached stems and fruits according to Koch′s rule. 【Results】 Seven strains of pathogen were isolated from diseased stems and fruits. The ITS rDNA sequences were 99.60%-100% homologous with Botrytis cinerea (GenBank registration number KP151610, KX426964, and ON566787), indicating that all the seven strains were Botrytis cinerea. Two strains, G4 and JW1, were selected and inoculated on the stems and fruits of healthy facility tomato. Polycystic ovary syndrome (PCOS) is a common reproductive endocrine disorder in women, and serum anti-Müllerian hormone (AMH), as a new indicator for evaluating ovarian reserve, is widely used in the evaluation, diagnosis and treatment of PCOS. With the development of the research on AMH, the research of AMH of PCOS patients during pregnancy has made some progress. It is found that AMH decreases during pregnancy and fluctuates as pregnancy progresses. There is a certain correlation between AMH and androgens during pregnancy of PCOS patients. AMH can be used as a candidate factor for predicting preterm birth in PCOS patients, and provides new ideas and methods for the management of PCOS patients during pregnancy. In addition, it should be noted that the intrauterine environment with high AMH may be an adverse interfering factor in the early life of the offspring of pregnant women with PCOS. PCOS patients belong to a high-risk pregnancy group after pregnancy and face the challenges of various complications during pregnancy. Furthermore, AMH can better guard the maternal and fetal health of PCOS pregnant women, and has broad application prospects in the field of obstetrics. A method for quantitative determination of triethylene diamine in activated carbon by ultrasonic extraction and gas chromatography with internal standard was established.The method used methanol as solvent and tetramethylenediamine as internal standard for quantitative detection by gas chromatography.The extraction method was optimized, through experiments, the optimal conditions of ultrasonic extraction were determined as extraction temperature of 50 ℃,extraction solvent volume of 20 mL and extraction time of 10 min, and the methodology was verified.The results show that the linearity of the method is good under this condition, R²＞0.995 0.The limit of detection was 0.10 g/L and the limit of quantitation was 0.33 g/L.The recoveries were 84.22%～93.08%,the intra-day relative standard deviations were 4.60%～6.29%,and the intra-day relative standard deviations were 5.07%～7.03%.The method is simple and fast to operate, and the quantitative results are accurate and reliable, which can meet the needs of actual sample detection and provide an effective reference for guaranteeing the monitoring efficiency of triethylene diamine in activated carbon. Objective To observe the cognitive dysfunction of elderly rats intraperitoneally injected with oxycodone by increasing-dose method versus stable-dose method. Methods Thirty clean-grade male Sprague-Dawley rats aged 20 months were selected and randomly divided into 3 groups with 10 rats in each. Rats in the group A were injected intraperitoneally with oxycodone at a dose of 0. 2 mg/ (kg·d) once per day for 14 consecutive days. Rats in the group B were injected intraperitoneally for 7 consecutive days, once a day, the dose increased from 0. 1 mg/ (kg·d) to 0. 7 mg/ (kg·d), and the daily increased dose was 0. 1 mg/Kg. Rats in the group C were intraperitoneally injected with normal saline 2 mL/ d, once a day for 7 consecutive days. After the administration or normal saline, the training and testing of water maze swimming started, and the training lasted for 3 days. After training, we carried out the place navigation test (PNT) for 3 days (PNT1-3) . After PNT, the underwater platform was taken out for space probe test (SPT) . At 1 week after SPT, we put the rats back on the platform and perform PNT again (PNT4) . Results Compared with group C, rats in the group B had longer platform escape latency and fewer platform crossing times on PNT2 and PNT4, and rats in the group A had longer platform escape latency and fewer platform crossing times on PNT4 (all P<0. 05) . Compared with group B, rats in the group A had a short platform escape latency and more platform crossing times on PNT4 (both P<0. 05) . Conclusions Long-term use of oxycodone may cause cognitive dysfunction in rats. The stable dose method has less damage to learning and memory function than the incremental dose method, and the cognitive dysfunction can be recovered more quickly after the drug is stopped. Objective To analyze the gene expression of hamster kidney normal BHK21 cells after inhibiting serine threonine tyrosine kinase 1 (STYK1/NOK) by transcriptome analysis, and analyze its possible signaling pathways. Methods BHK21 cells were divided into empty vector group (transfected with 6 μg empty vector pBs/U6), low concentration transfection group (2 μg si-STYK1/NOK plasmid + 4 μg empty vector pBs/U6) and high concentration transfection group ( 6 μg si-STYK1/NOK plasmid). Western blot assay was used to detect STYK1 / NOK protein expression in 48 hours after transfection. Based on the detection results, the empty RNA group and high transfection concentration group were selected to extract total RNA for the transcriptome sequencing. The sequences obtained by sequencing were filtered and compared to obtain differentially expressed genes. After obtaining the differentially expressed genes, R software was used to analyze the biological functions of Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG). The interactions in the STRING protein interaction database were used to analyze the differential gene-protein interaction networks. QRT-PCR was used to verify the key differentially expressed genes methylenetetrahydrofolate dehydrogenase 2 (Mthfd2), activating transcription factor 5 (Atf5), phospholipase A2 group Ⅱ A (Pla2g2a) and 6-phosphofructo-2-kinase/fructose-2, 6- biphosphatase 3 (Pfkfb3). CCK-8 method was used to detect cell proliferation in empty vector group and high transfection concentration group. Results Western blot results showed that the expression of STYK1/NOK protein was significantly inhibited in the high-concentration transfection group. Analysis of the transcriptome sequencing results revealed a total of 44 differentially expressed genes, including 19 up-regulated genes and 25 down-regulated genes in the empty vector group and the high-concentration transfection group. GO enrichment analysis found that the differentially expressed genes mainly affected biological regulation, cellular processes, metabolic regulation, cellular biological processes, cells, cellular parts, extracellular regions, ligands and catalytic activity. KEGG pathway analysis found that the differentially expressed genes were mainly concentrated in the immune system, cancer, cell cycle, signal transduction and amino acid metabolism. The qRT-PCR results showed that the relative expression levels of Mthfd2 and Atf5 were significantly increased in the high concentration transfection group (P＜0.05), and the relative expression levels of Pfkfb3 and Pla2g2a were significantly decreased (P＜0.05), which was consistent with the sequencing results. Cell proliferation experiments showed that the cell proliferation was significantly higher in the high-concentration transfection group than that in the empty vector group (P＜ 0.01). Conclusion The inhibition of STYK1 / NOK expression can result in the enhanced expression of proto-oncogenes, the promoted expression of tumor suppressor genes and the decreased cell proliferation in BHK21 cells. Antibiotic resistance genes (AR Gs) are closely related to anti biotic resistance in bacteria, and their existence in food has attracted more and more attention. Cheese is a kind of instant fermented food, and its consumptions g row rapidly in China. In this study, quantitative PCR was used to detect 8 kinds of antibiotic resistance genes in domestic and foreign cheese and processed cheese samples, and to characterize their existence and distribution in cheese. The results indicated that, the re we re abundant ARGs detected in the two types of cheese, except for vancomycin resistant genes. In processed cheese, the abundances of resistance genes were significantly lower than regular cheese samples. In addition, resistance genes of aminoglycoside and tetracycline class and the big ring lipid-forests amides-chain positive fungu selement class B (MLSB) we redetected at higher rates in processed cheese samples. In regular cheese, resistance genes against (fluoroqu inolone, quinolone, florfenico l, chloramp henicol and amphen icol) and aminoglycoside were detected at higher rates. In our study, lactic acid bacteria were also isolated from the cheese with their antibiotic susceptibility tested. With in the 18 strains of lactic acid bacteria tested, it indicated that the resistant rate against sulfonoxazole were higher ( 90. 9 % ), and t hat against benzoxicillin was 86. 36 % . In addition, within the strains of lactic acid bact eria tested, the susceptibilities against tetracycline as well as ampicillin were 100%, and chloramphenicol (81.8%). The highest intermediate rates were ciprofloxacin (72.7%) and against norfloxacin? was 67.9% . Objective: To investigate the effect and potential mechanism of silencing TCAB1 on the proliferation of human aortic smooth muscle cells(HASMC). Methods: Three si RNA sequences(siTCAB1-331, si TCAB1-619, si TCAB-749) targetingTCAB1 and one negative sequence(NC) were designed, synthesized and then transfected into HASMC with Lipofectamine-TM2000. HASMC were divided three groups: interference group, blank control group, negative control group. Cell transfection was observed under fluorescence microscope. The m RNA and protein expression levels of TCAB1 were detected by RT-q PCR and Western blotting, the best target was selected from the three interfering targets. The effective si RNA(si TCAB1-749) was further transfected into HASMC with LipofectamineTM2000. The changes of cell cycle and CyclinD1 expression of HASMC cells were assessed using flow cytometry,RT-qPCR and Western blotting, respectively. The proliferation of HASMC was assessed using MTS assay after transfecting si TCAB1-749 at 24 h, 48 h,72 h. Results: RT-qPCR and Western blot indicated that the si TCAB1-749 was the best target in the three interfering targets. MTS revealed that the number of cell growth in si TCAB1-749 group was significantly lower than BC group and NCgroup at 24 h, 48 h, 72 h(P<0.05); the percentages of S phases in the cell cycle of si TCAB1-749 group was decreased and G1 phases was increased at 48 h(P < 0.05). In addition, the expression of cyclinD1 was decreased in si TCAB1-749 group(P<0.05). 目的 观察不同年龄良性胆囊疾病患者腹腔镜胆囊切除前后肠道菌群变化。方法 126 例良性胆囊疾病患者按年龄分为青年组(<45岁) 37例、中年组(45～60岁) 70例、老年组(> 60岁) 19例,3组均行腹腔镜胆囊切除术(LC),比较各组手术前后粪便优势菌双歧杆菌(Lgg)、乳酸菌(Lac)、大肠杆菌(E. coli)和肠球菌(Ecc)计数及差值。结果 与同组术前比较,青年组术后 Lgg 计数减少,中年组和老年组 Lgg、Lac 计数减少和E. coli、Ecc计数增加(P均<0. 05)。与青年组比较,中年组E. coli、Ecc 及老年组 Lgg、Lac、E. coli、Ecc 计数差值增加 (P均<0. 05); 与中年组比较,老年组Lgg、Lac、E. coli、Ecc 计数差值增加(P均<0.05)。结论 LC可以引起良性胆囊疾病患者肠道菌群变化,这种变化随着年龄的增长更加明显,老年组 LC 术后肠道菌群失调较中、青年组严重,且表现为Lgg和Lac等益生菌减少,因此肠道微生态制剂应注重制剂种类和剂量对不同人群的针对性,以获得更好疗效。 [ABSTRACT FROM AUTHOR]
