Genetic diversity influences the evolutionary potential of forest trees under changing environmental conditions, thus indirectly the ecosystem services that forests provide. European beech (Fagus sylvatica L.) is a dominant European forest tree species that increasingly suffers from climate change-related die-back. Here, we conducted a systematic literature review of neutral genetic diversity in European beech and created a meta-data set of expected heterozygosity (He) from all past studies providing nuclear microsatellite data. We propose a novel approach, based on population genetic theory and a min–max scaling to make past studies comparable. Using a new microsatellite data set with unprecedented geographic coverage and various re-sampling schemes to mimic common sampling biases, we show the potential and limitations of the scaling approach. The scaled meta-dataset reveals the expected trend of decreasing genetic diversity from glacial refugia across the species range and also supports the hypothesis that different lineages met and admixed north of the European mountain ranges. As a result, we present a map of genetic diversity across the range of European beech which could help to identify seed source populations harboring greater diversity and guide sampling strategies for future genome-wide and functional investigations of genetic variation. Our approach illustrates how to combine information from several nuclear microsatellite data sets to describe patterns of genetic diversity extending beyond the geographic scale or mean number of loci used in each individual study, and thus is a proof-of-concept for synthesizing knowledge from existing studies also in other species.

Societal Impact Statement Biodiversity is essential to food security and nutrition locally and globally. By reviewing the global state of edible plants and highlighting key neglected and underutilized species (NUS), we attempt to unlock plant food resources and explore the role of fungi, which along with the wealth of traditional knowledge about their uses and practices, could help support sustainable agriculture while ensuring better protection of the environment and the continued delivery of its ecosystem services. This work will inform a wide range of user communities, including scientists, conservation and development organizations, policymakers, and the public of the importance of biodiversity beyond mainstream crops. Summary As the world's population is increasing, humanity is facing both shortages (hunger) and excesses (obesity) of calorie and nutrient intakes. Biodiversity is fundamental to addressing this double challenge, which involves a far better understanding of the global state of food resources. Current estimates suggest that there are at least 7,039 edible plant species, in a broad taxonomic sense, which includes 7,014 vascular plants. This is in striking contrast to the small handful of food crops that provide the majority of humanity's calorie and nutrient intake. Most of these 7,039 edible species have additional uses, the most common being medicines (70%), materials (59%), and environmental uses (40%). Species of major food crops display centers of diversity, as previously proposed, while the rest of edible plants follow latitudinal distribution patterns similarly to the total plant diversity, with higher species richness at lower latitudes. The International Union for Conservation of Nature Red List includes global conservation assessments for at least 30% of edible plants, with ca. 86% of them conserved ex situ. However, at least 11% of those species recorded are threatened. We highlight multipurpose NUS of plants from different regions of the world, which could be key for a more resilient, sustainable, biodiverse, and community participation‐driven new “green revolution.” Furthermore, we explore how fungi could diversify and increase the nutritional value of our diets. NUS, along with the wealth of traditional knowledge about their uses and practices, offer a largely untapped resource to support food security and sustainable agriculture. However, for these natural resources to be unlocked, enhanced collaboration among stakeholders is vital.

Remote detection of aerial parasitic plants in forests is imperative in precision forestry, as it can help to manage tree stands and to monitor forest ecosystem health. The plain forests located in Noor and Hyrcanian forests (Iran), characterized by mixed broadleaved forests, host large populations of mistletoe (Viscum album). In this study, aiming to delineate trees infected by mistletoe, an unmanned aerial vehicle (UAV) equipped with an RGB camera was used to acquire series of images in winter (February of 2020) and summer (September of 2020). The canopy height model (CHM) was generated from UAV images then Gaussian filter was applied to the CHM. Two sites were selected and the individual tree crowns in these sites were delineated manually and automatically (using a region growing algorithm based on the filtered canopy height model). A range of UAV-based RGB vegetation indices (VIs) was generated. Individual trees were classified into two classes (i.e., infected and non-infected) using a random forest classification algorithm, and based on six image processing scenarios (i.e., three scenarios where tree crowns were delineated manually, followed by identification of infected trees in leaf-off, leaf-on, and combined leaf-off and leaf-on seasons, and three scenarios with the same identification procedure applied to automatically delineated tree crowns using a regional growing algorithm). Optimal classification results using manual and automatic crown delineation were obtained by leaf-off and combined leaf-off + leaf-on season data with the overall accuracy of 87% and 76% for site 1, respectively. Also, the overall accuracy of 80% and 69% was obtained from combined leaf-off + leaf-on season data for site 2. The study demonstrates the potential of using UAV-based RGB data for studying mistletoe infection and distribution in a complex forest ecosystem.

Fire blight disease, caused by the bacterium Erwinia amylovora (E. amylovora), is responsible for substantial losses in cultivated apples worldwide. An important mechanism of plant immunity is based on the recognition of conserved microbial molecules, named pathogen-associated or microbe-associated molecular patterns (PAMPs or MAMPs), through pattern recognition receptors (PRRs), leading to pattern-triggered immunity (PTI). The interspecies transfer of PRRs represents a promising strategy to engineer broad-spectrum and durable disease resistance in crops. EFR, the Arabidopsis thaliana PRR for the PAMP elf18 derived from the elongation factor thermal unstable (EF-Tu) proved to be effective in improving bacterial resistance when expressed into Solanaceae and other plant species. In this study, we tested whether EFR can affect the interaction of apple with E. amylovora by its ectopic expression in the susceptible apple rootstock M.26. Stable EFR expression led to the activation of PAMP-triggered immune response in apple leaves upon treatment with supernatant of E. amylovora, as measured by the production of reactive oxygen species and the induction of known defense genes. The amount of tissue necrosis associated with E. amylovora infection was significantly reduced in the EFR transgenic rootstock compared to the wild-type. Our results show that the expression of EFR in apple rootstock may be a valuable biotechnology strategy to improve the resistance of apple to fire blight.

Societal Impact Statement Bioenergy is a major component of the global transition to renewable energy technologies. The plant and fungal kingdoms offer great potential but remain mostly untapped. Their increased use could contribute to the renewable energy transition and addressing the United Nations Sustainable Development Goal 7 “Ensure access to affordable, reliable, sustainable and modern energy for all.” Current research focuses on species cultivated at scale in temperate regions, overlooking the wealth of potential new sources of small‐scale energy where they are most urgently needed. A shift towards diversified, accessible bioenergy technologies will help to mitigate and adapt to the threats of climate change, decrease energy poverty, improve human health by reducing indoor pollution, increase energy resilience of communities, and decrease greenhouse gas emissions from fossil fuels. Summary Bioenergy derived from plants and fungi is a major component of the global transition to renewable energy technologies. There is rich untapped diversity in the plant and fungal kingdoms that offers potential to contribute to the shift away from fossil fuels and to address the United Nations Sustainable Development Goal 7 (SDG7) “Ensure access to affordable, reliable, sustainable and modern energy for all.” Energy poverty—the lack of access to modern energy services—is most acute in the Global South where biodiversity is greatest and least investigated. Our systematic review of the literature over the last 5 years (2015–2020) indicates that research efforts have targeted a very small number of plant species cultivated at scale, mostly in temperate regions. The wealth of potential new sources of bioenergy in biodiverse regions, where the implementation of SDG7 is most urgently needed, has been largely overlooked. We recommend next steps for bioenergy stakeholders—research, industry, and government—to seize opportunities for innovation to alleviate energy poverty while protecting biodiversity. Small‐scale energy production using native plant species in bioenergy landscapes overcomes many pitfalls associated with bioenergy crop monocultures, such as biodiversity loss and conflict with food production. Targeted trait‐based screening of plant species and biological screening of fungi are required to characterize the potential of this resource. The benefits of diversified, accessible bioenergy go beyond the immediate urgency of energy poverty as more diverse agricultural landscapes are more resilient, store more carbon, and could also reduce the drivers of the climate and environmental emergencies.

In addition to two known ecdysteroids, 20-hydroxyecdysone and turkesterone, three previously undescribed stigmastane-type ecdysteroids were isolated from the underground parts of Rhaponticum acaule (L.) DC. by chromatographic techniques (CC, VLC, MPLC). The structures of the compounds were established by chemical (acetylation) and spectroscopic methods including UV, IR, HRMS, 1D-NMR: 1H-NMR, 13C-NMR, DEPT-135. and 2D-NMR: COSY, NOESY, HSQC, HMBC. Two compounds were isolated as an isomeric mixture and each of them was purified and converted to the corresponding acetylated derivative. Based on all of the evidence, the structures of three undescribed stigmastane-type ecdysteroids were established as 2β,3β,11α,20β,22α,24,28-heptahydroxy-6-oxo-stigmast-7-en-25,29-lactone and the cyclic 22,29-hemiacetals 22R and 22S stigmast-7-en-29-al,2β,3β,11α,20α,22,28-hexahydroxy-6-oxo, and the trivial names acaulesterone and rhapocasterones A and B are suggested, respectively. The structures and absolute configurations of 20-hydroxyecdysone and cyclic-22,29-hemiacetal-22R-stigmast-7-en-29-al,2β,3β,11α,20α,22,28-hexahydroxy-6-oxo were confirmed by X-ray crystal-structure analyses of their acetyl derivatives.

High occurrence of Fusarium poae (FP) and Fusarium langsethiae (FL) and their mycotoxins nivalenol (NIV) and T‐2/HT‐2 have been observed in Swiss oats. Early prediction of mycotoxin levels is important for farmers and the cereal industry to minimize the risk of contaminated food and feed. Therefore, climate chamber experiments were conducted to investigate the influence of different temperatures (10, 15, 20 °C) and durations (4, 8, 12 h) at 99% relative humidity (RH) on the infection of oats with FP and FL. In addition, to discover the most susceptible period of oats, artificial FL inoculations were conducted at different growth stages. Field experiments were performed to observe the dispersal of these fungal species within the field and to investigate the weather conditions that influence the dispersal. The climate chamber experiments revealed higher contamination with NIV and T‐2/HT‐2 in the 10 °C treatments and with a prolonged humidity duration of 12 h 99% RH. Inoculations of oat plants at early (DC 61) and mid (DC 65) anthesis, led to higher FL infection and T‐2/HT‐2 accumulation in the grains compared with treatments at earlier growth stages, which might be due to an increased susceptibility during anthesis. No indication for spore dispersal was observed in the field experiments. The results obtained, together with the cropping factors that influence infection and mycotoxin production, could be used as a first step in developing forecasting models to predict the contamination of oats with the mycotoxins NIV and T‐2/HT‐2.

Fusarium head blight is one of the most noxious cereal diseases. Worldwide, F. graminearum (FG) and the mycotoxin deoxynivalenol (DON) is the most dominant species/mycotoxin in barley and wheat. Barley is often produced as on farm feed and thus routine mycotoxin analyses similar to those of cereals for human consumption are not performed. Hence, an early prediction of mycotoxin levels is important for farmers to minimise the risk of contaminated feed but also of contaminated cereals entering the cereal supply chain. Therefore, climate chamber experiments with artificial FG infection of barley investigating the influence of different temperatures (10 °C, 15 °C, 20 °C) and durations (4 h, 8 h, 12 h) at 99% relative humidity were conducted to accumulate data to develop a forecasting system. An up to three times higher DON contamination in the 15 °C treatments for the feed barley variety Ascona was detected compared with the 10 °C and 20 °C treatments. For the malting barley variety Concerto, the prolonged humidity durations had a stronger effect under all tested temperatures and resulted in up to two times higher DON contaminations. In addition, field experiments where spore deposition during anthesis as well as disease incidence, fungal amount and mycotoxins were observed, showed that the overwintered straw treatment resulted, depending on the year, in a three times higher FG incidence and DON content compared with the control and freshly inoculated straw treatment.

This study reappraised traditional knowledge (TK) about palms (Arecaceae) by the Chachi indigenous group in northwestern Ecuador, 30 years after the first study in 1985 on Chachi palm ethnobotany (Barfod and Balslev 1988). We wished to gain insight about which palm species the Chachi people use today, and how palm TK has changed among the Chachi since 1985. In 2015, using semi-structured interviews and participant observation, we documented nine useful palm species and 457 use reports. The 1985 methods were less formalized, based on open-ended interviews and recorded 14 palm species with 38 use descriptions. Most uses fell into the categories Food (13 use descriptions), Utensils/Tools (10), and Construction (7). In 2015, most of the use descriptions similarly fall into the categories Food (38), Construction (20), and Utensils and tools (19). As in 1985, the most important species harvested today are Iriartea deltoidea and Wettinia quinaria. Four understory palm species reported as useful in 1985 were not recorded in 2015. Still, most of the uses documented among the Chachi in 1985 were also registered in 2015. Knowledge about blowguns, blowgun darts, and marimba keys, however, seems to have vanished. Although palms still provide important ecosystem services for the Chachi, (e.g., food and construction), better management of natural resources and land-use is pivotal to meet the Sustainable Development Goals that Ecuador is committed to through their participation in the United Nation's Sustainable Development Knowledge Platform. This is particularly complicated because of the rapid human population growth in the coastal lowland of Ecuador and the impending threats from climate change.

Occurrence and genomic organization of dispersed elements containing ZpS1 satellite repeats have been investigated in a wide representation of species of the old plant genus Zamia (Zamiaceae, Cycadales). In Z. paucijuga, the ZpS1 repeat is organized as long satellite DNA arrays and as short arrays inserted into AT-rich dispersed elements. A comparative study by Southern analysis shows that these unusual dispersed elements containing the ZpS1 repeat are present with different organizations in all investigated Zamia species. In some species these elements are present with a low copy number, while in other species secondary amplification events, involving specific sequence clusters, appear to have generated characteristic dispersed elements in a high copy number. Among Zamia species, several groups share similar restriction patterns, as the Zamia loddigesii complex and the Caribbean species suggesting a general correlation between organization and genomic representation of the dispersed repeated sequence and the pattern of phyletic relationships in the genus. However, the finding of different patterns also among closely related species suggests a complex history of amplifications and losses of these dispersed repetitive elements that cannot be always easily traced through the phylogenetic reconstruction of this ancient plant group.

Given the rapid ability of many pathogens to overcome genetic resistance, the identification and practical implementation of new sources of resistance is essential. Landraces and wild relatives of wheat have played an important role as genetic resources for the improvement of disease resistance. The use of molecular approaches, particularly molecular markers, has allowed better characterization of the genetic diversity in wheat germplasm. In addition, the molecular cloning of major resistance (R) genes has recently been achieved in the large, polyploid wheat genome. For the first time this allows the study and analysis of the genetic variability of wheat R loci at the molecular level and therefore, to screen for allelic variation at such loci in the gene pool. Thus, strategies such as allele mining and ecotilling are now possible for characterization of wheat disease resistance. Here, we discuss the approaches, resources and potential tools to characterize and utilize the naturally occurring resistance diversity in wheat. We also report a first step in allele mining, where we characterize the occurrence of known resistance alleles at the wheat Pm3 powdery mildew resistance locus in a set of 1,320 landraces assembled on the basis of eco-geographical criteria. From known Pm3 R alleles, only Pm3b was frequently identified (3% of the tested accessions). In the same set of landraces, we found a high frequency of a Pm3 haplotype carrying a susceptible allele of Pm3. This analysis allowed the identification of a set of resistant lines where new potentially functional alleles would be present. Newly identified resistance alleles will enrich the genetic basis of resistance in breeding programmes and contribute to wheat improvement

To investigate bioactive compounds potentially involved in the biotic interactions exhibited by Phlomis purpurea (Lamiaceae) in rhizospheres infested with Phytophthora cinnamomi, the plant rhizome was chemically analysed. The nortriterpenoid (175)-2 alpha,3 alpha,11 alpha,23,24-pentahydroxy-19(18 -> 17)-abeo-28-norolean-12-en-18-one, was isolated and its structure was elucidated by comprehensive spectroscopic analysis, chiefly using 2D NMR experiments, and X-ray analysis. It was shown to be exuded by roots and to exhibit anti-Phytophthora and antitumor activities. (C) 2016 Published by Elsevier Ltd.

Healing Animals, Feeding Souls: Ethnobotanical Values at Sacred Sites in Central Italy. Ethnobotanical knowledge is a fundamental repository of the values and applications of different plants. This knowledge is often related to spiritual beliefs and religious sites, where plants have been nurtured and conserved for their use in rituals and traditional practices. While this link is well known for different areas of the global south, it has hardly been investigated in relatively more secular and modernized Western contexts. Here, we use first-hand vegetation surveys and published records to examine the occurrence of ethnobotanical values at 30 Catholic shrines in Central Italy, and compare them with an equal number of non-sacred control sites. We ask this: to what extent is there an association of useful plants with sacred places in Italy, as found in other cultural contexts? We show that a greater number of useful plants are found at sacred sites. While this is mainly a consequence of the higher species richness of sacred sites, an association with plants used in animal husbandry is particularly evident, and likely related to the deep historical connection between sacred places and pastoralist traditions in Central Italy. Also, we show that there are significant variations in the distribution of old trees; the largest specimens are found at the center of sacred sites, while tree size visibly decreases away from the shrines. This indicates also that individual trees have been actively managed and conserved at sacred sites, probably driven by the symbolic values that old trees frequently embody.

Flower emasculation is commonly used to make flowers unattractive to pollinating insects and to carry out controlled pollinations. In sweet cherry, we have observed recurrent low fruit set after flower emasculation and compatible pollination without apparent causes. The definitive version is available at: http://www.sciencedirect.com/science/journal/03044238, Despite the extensive research work characterizing pollen performance in several plant species, less effort has been made to characterize it in some economically important species as Citrus, in which the failure of the sexual reproductive process, and subsequent parthenocarpic fruit development and seedlessness are prized characters. In this work we characterized pollen–pistil interaction in the three ancestral true-type Citrus species in order to determine the influence of the male and female genotypes, as well as of the temperature regime on pollen performance. Specifically, we characterized the effect of temperature on pollen grain germination in vitro, and on pollen tube growth in vivo in cut flowers and in planta under field temperature conditions. Results obtained showed that temperature variation has a strong effect on pollen germination, and on pollen tube kinetics and on their population size depending on the specific male–female combination. The optimum temperature for pollen germination in vitro was of 25 °C, while the most favorable temperature to accelerate in vivo pollen tube growth depended on the particular male–female interaction and ranged between 15 and 25 °C. Furthermore, temperature appears to have an effect on self-incompatibility reaction by affecting the place where pollen tubes are arrested. Over all, our results show that pollen performance is not only an inherent characteristic of the pollen genotype, but is largely dependent on the particular male–female combination and on genotype–temperature interactions.

Microarrays offer virtually unlimited diagnostics capability, and have already been developed into diagnostic chips for many different plant pests. The full capacity of such chips, however, has lagged far behind their full potential. The main reason for this is that current chip design relies on a priori genetic information for target organisms and on a consensus on the genetic sequences to be used in particular organism groups. Such information is often unavailable and laborious to obtain. Thus, broad-application diagnostic microarrays have been limited to narrow organism groups focused on Genera of pests/pathogens or those affecting individual host crops, without applicability for simultaneous detection of diverse pests affecting many crops. This paper describes the development of a diagnostic microarray platform that has universal application based on genomic fingerprinting of any organism without a need for a priori sequence information. Taxon-specific hybridization patterns are obtained by unique hybridisation of genomic DNA to 100s–1000s of short random oligonucleotide probes. Taxon identification is then achieved by comparison of hybridisation patterns from an unknown sample against a reference-pattern database. Using bacteria as a model pathogen group, these methods deliver highly reproducible hybridisation patterns with high resolution power and enable discrimination at the species and subspecies level.

We provide a short discussion of how the use of molecular data and sophisticated analytical methods has expanded our knowledge about the phylogenetic relationships among flowering plants and how this affects the familial and suprafamilial classification of succulents. A tree diagram illustrates the current hypothesis on their interrelationships and a table lists all 83 families that include succulent representatives (c.12,500 species from c.690 genera), together with information on taxonomic diversity (i.e. number of estimated species and genera) and architectural types of succulence. Furthermore, we briefly discuss some important recent modifications to the family classification of flowering plants and provide arguments for the proposed changes as far as succulents are concerned. In particular, we focus on the controversially discussed family classification of the monocotyle-donous order Asparagales and provide arguments for a revised classification that considers the distinct variation patterns in this clade.

The effect of soil collected from long-term (1007 years of duration of monoculture) and short-term (55 years) grapevine monoculture on production of volatile organic compounds (VOC’s) and on functional parameters of photosystem II in grapevine leaves was analyzed. Grapevine plantlets grown in tested soils showed differences in VOC’s production after five months cultivation. Chlorophyll a fluorescence measurements by JIP-test revealed that the photosystem II was less efficient but the fluorescence intensity increased in plant growing in soil from the long-term monoculture compared to plants growing in the short term monoculture soil. Pseudomonas spp. carrying the biocontrol genes phlD and hcnAB were isolated from long-term monoculture soil. A consortium of ten of these isolates was added to the short term monoculture soil. The plants grown in this inoculated soil showed similar changes in fluorescence intensity and photosystem efficacy as the plants growing in long term monoculture. In this study, simple tools for measurement of a “soil effect” by measuring only a leaf have been tested successfully. To investigate the dynamics of precursor compounds of green leaf volatiles (GLV)s and other biogenic compounds released by mechanically damaged Brassica oleracea leaves, plants were exposed for two consecutive 16h light phases to highly enriched (13)CO(2). Analysis by GC-MS indicated (1) biogenic compounds released upon wounding, (2) a different labelling pattern between and (3) within compounds, and (4) evidence for spatial heterogeneity of the precursor pool extrapolated from points (1)-(3). First, GLVs comprised C(5) and C(6) molecules, with the GLV pentenyl acetate being reported here for the first time from higher plants. Second, the labelling pattern found in most GLVs indicates a low turnover of the precursor alpha-linolenic acid. Moderate labelling of dimethyldisulphide indicates a connection to an active plastidic methyl pool closely connected to CO(2) fixation, and very weak labelling of terpenes indicates a constitutive monoterpene pool. Third, not all GLVs exhibit similarly strong labelling patterns (hexenyl acetate vs. hexyl acetate), indicating different precursors. As the labelling patterns of alcohol and acetate moieties in the esters differ, with only the former being strongly labelled, the precursor of the acetate moiety, acetyl-CoA, is likely to derive from a different cellular pool to that used in chloroplastic fatty acid synthesis, or was rapidly synthesised after the end of labelling. Fourth, the exceptionally high relative abundance of labelled GLV and the low concentration of unlabelled molecules are likely to occur because recently synthesized alpha-linolenic acid is bound in lipids that are organised in distinct areas, or are chemically different from the older lipids. They must be preferentially used as precursors.

Four γ-glutamyl peptides were isolated from a hydrophilic extract of garlic bulbs. γ- l -Glutamyl- S -( trans -1-propenyl)- l -cysteine has been isolated as a new genuine constituent of Allium sativum . For the first time detailed NMR data for γ- l -glutamyl- S -allyl- l -cysteine, γ- l -glutamyl- S -( trans -1-propenyl)- l -cysteine and γ- l -glutamyl- S -allylthio- l -cysteine are reported. The structures of the four compounds were established on the basis of their spectral data (IR, FABMS, 1 H and 13 C NMR). Extensive 2D heteronuclear correlated NMR experiments allowed the assignment of all proton and carbon resonances for all reported compounds.
