The application of seaweed extract-based biostimulants is a promising approach for achieving sustainable agriculture, with an enormous potential of improving crop yield and mitigating climate change effects. Abiotic stressors, such as drought, are major factors resulting in tomato (Solanum lycopersicum L.) yield losses and seaweed-based biostimulants have been proposed as an eco-friendly strategy to counteract this negative impact. Chondrus crispus is a common red seaweed widely used as source of carrageenans, not yet explored as a plant biostimulant. In this study, a protein hydrolysate-rich C. crispus extract, by-products of the carrageenan extraction, was tested on tomato plants under well-watered condition and water shortage. The foliar application of the protein-rich C. crispus extract conferred drought tolerance to tomato plants resulting in less noticeable visual stress symptoms. Treated plants showed higher shoot height and biomass under both well-watered and water deficit conditions, evidencing the double effect exerted by this new biostimulant, as plant growth promoter and drought stress protector. The treatment with the biostimulant had an effect on levels of abscisic acid and proline, and triggered the expression of Solyc02g084840, a drought marker gene. Finally, a label-free mass spectrometric approach allowed us to identify phycoerythrins and phycocyanins as major bioactive proteins contained in the extract. Altogether, these results indicate that the foliar application of protein hydrolysate-rich extracts from C. crispus improved tomato plant growth and tolerance to drought stress, suggesting a new opportunity for further applications in the agriculture and horticultural sectors. The composition of seaweeds is complex, with vitamins, phenolic compounds, minerals, and polysaccharides being some of the factions comprising their structure. The main polysaccharide in brown seaweeds is fucoidan, and several biological activities have been associated with its structure. Chitosan is another marine biopolymer that is very popular in the biomedical field, owing to its suitable features for formulating drug delivery systems and, particularly, particulate systems. In this work, the ability of fucoidan to produce nanoparticles was evaluated, testing different amounts of a polymer and using chitosan as a counterion. Nanoparticles of 200–300 nm were obtained when fucoidan prevailed in the formulation, which also resulted in negatively charged nanoparticles. Adjusting the pH of the reaction media to 4 did not affect the physicochemical characteristics of the nanoparticles. The IC50 of fucoidan was determined, in both HCT−116 and A549 cells, to be around 160 µg/mL, whereas it raised to 675–100 µg/mL when nanoparticles (fucoidan/chitosan = 2/1, w/w) were tested. The development of greener processes for the sustainable utilization of raw materials is increasingly demanded for environmental and economic reasons. A rapid and chemical-free technique was proposed for the extraction of hybrid kappa/iota (6/4) carrageenan from Sarcopeltis (ex Gigartina) skottsbergii. After separation, carrageenans were analyzed by Fourier transform infrared attenuated total reflectance, high-performance size-exclusion chromatography, and rheology. Maximum carrageenan extraction yields up to 63–64% were obtained operating at 110 or at 160 °C, for 5–7 min considering the sum of the heating and cooling periods, but the extraction of the phenolic fraction was favored at 220 °C. The recovered carrageenan showed apparent viscous values around 103 mPa at the lowest tested shear rates (0.1 1/s) and could be suitable to formulate films. Furthermore, those carrageenans obtained under 140 °C showed gel characteristics without previous separation from the liquid extract, avoiding ethanolic precipitation and energy consumption. The antiradical properties correlated with the phenolic content in the liquid phase, but no influence of temperature on the reducing properties was observed. The microwave-assisted hydrothermal treatment could be an efficient tool without needing chemicals for the extraction of carrageenans, which showed adequate rheological properties for commercial uses. This study focusses on the valorization of the red seaweed Mastocarpus stellatus, collected in the Northern coast of Portugal, as a natural source of high value compounds due to its beneficial properties. An annual monitoring of the seaweed was performed by determining its lipids, ash, carbohydrates, phycobiliproteins, total phenolic compounds, antioxidant capacity and carrageenan from three different rocky shores located in the north of Portugal. The results showed a seasonal and spatial variability of the studied compounds between October 2018 and September 2019 depending on the climatic variables of temperature, precipitation, and solar radiation. The most productive season coincided with the warmest months, except for carbohydrates and phycobiliproteins, which were promoted in the colder season. The spatial variation also could be explained by the proximity to water channel discharges at the sampling sites. Complementary studies on the carrageenan fraction were conducted in one of the shores due to the high biopolymer content, to determine their carrageenan proportion between the summer and winter period and establish their rheological capabilities for the formulation of gelling matrices. The extracted biopolymers exhibited typical structural and viscoelastic characteristics of kappa/iota-hybrid carrageenans, suggesting notably differences depending on the harvest season, which is critically relevant to define future applications. Novel personal care products are necessary to cope with the growing market demand for sustainable green products. In this context, this work deals with the formulation and fundamental physicochemical and rheological characterization of different natural personal care products using bioactive fractions from Chondrus crispus red macroalgae extracted under optimized green conditions. Body milks, body oils and shampoos were supplemented with soluble extracts with antioxidant features recovered after hydrothermal (200 °C) and microwave (170 °C)- and ultrasound (80 °C)-assisted extraction of the red macroalgae used as raw material. Volatile organic compounds (VOCs) include a broad range of compounds. Their production influences a large number of processes, having direct and secondary effects on different fields, such as climate change, economy and ecology. Although our planet is primarily covered with water (~70% of the globe surface), the information on aquatic VOCs, compared to the data available for the terrestrial environments, is still limited. Regardless of the difficulty in collecting and analysing data, because of their extreme complexity, diversification and important spatial-temporal emission variation, it was demonstrated that aquatic organisms are able to produce a variety of bioactive compounds. This production happens in response to abiotic and biotic stresses, evidencing the fundamental role of these metabolites, both in terms of composition and amount, in providing important ecological information and possible non-invasive tools to monitor different biological systems. The study of these compounds is an important and productive task with possible and interesting impacts in future practical applications in different fields. This review aims to summarize the knowledge on the aquatic VOCs, the recent advances in understanding their diverse roles and ecological impacts, the generally used methodology for their sampling and analysis, and their enormous potential as non-invasive, non-destructive and financeable affordable real-time biomonitoring tool, both in natural habitats and in controlled industrial situations. Finally, the possible future technical applications, highlighting their economic and social potential, such as the possibility to use VOCs as valuable alternative source of chemicals and as biocontrol and bioregulation agents, are emphasized. This research did not receive any specific grant from funding agencies in the public, commercial, or not-for-profit sectors.

Several bioactive compounds that hold a potential interest in the food industry as phenolic compounds, polysaccharides, proteins and vitamins, among others, are present in seaweeds. Green extraction technologies are the preferred way to obtain these compounds. Pressurized hot water extraction, from 160 to 220 °C, was tested to achieve high yields of these components from the edible brown seaweed, Undaria pinnatifida. The maximum fucoidan content was recovered at 160 °C, while the phloroglucinol content and antioxidant activity were maximum at 220 °C. The possibility of encapsulating these bioactive fractions using mannitol was assessed. The highest production yield of the polymeric particles was found using the 220 °C fraction (close to 75%). In order to formulate microparticles with bioactive potential, several ratios of liquid phases were assessed, 3:1, 1:1 and 1:3 (w:w), using the liquid fractions obtained at 160 °C and 220 °C. The yield production was always above 67%, being in the 1:3 ratio (160 °C:220 °C) and close to 75%. The rheological results indicated that the presence of microparticles enhanced the apparent viscosity of the aqueous dispersions with non-Newtonian profiles, achieving the highest viscosity for those formulated with microparticles from 160 °C:200 °C (3:1). El creciente interés en la obtención de antioxidantes de origen natural ha posicionado a las microalgas, y entre ellas a la cianobacteria Arthrospira platensis, como una potencial fuente de compuestos antioxidantes. Los procesos de extracción utilizados tradicionalmente son poco eficientes en términos de tiempo y requerimiento energético, ya que a menudo se necesita la aplicación de elevadas temperaturas durante varias horas. Por ello, se requiere del desarrollo y optimización de metodologías de extracción más respetuosas con el medio ambiente, que permitan a su vez reducir costes y el tiempo requerido en el proceso analítico. Siguiendo con este principio, en el estudio se han optimizado los parámetros del proceso de extracción asistida por ultrasonidos de diferentes compuestos antioxidantes presentes en A. platensis. (polifenoles, pigmentos y polisacáridos). Los parámetros estudiados fueron: Cantidad de alga, Volumen de solvente, Tipo de solvente, Porcentaje de solvente, Tiempo, Potencia y Temperatura de extracción. Para cada condición estudiada, se analizó el contenido de fenoles totales (mediante el método de Folin-Ciocalteu) y la actividad antioxidante (mediante el método DPPH). Posteriormente, se utilizó la técnica de cromatografía líquida de alta resolución (HPLC-DAD) para determinar el perfil individual de polifenoles, y se caracterizaron los pigmentos y polisacáridos presentes en los extractos. El método optimizado proporcionó un extracto hidroalcohólico con alta actividad antioxidante (>85%) en condiciones experimentales muy convenientes. The growing interest in obtaining antioxidants from natural sources has positioned microalgae, including the cyanobacteria Spirulina (Arthrospira platensis), as a potential source of antioxidant compounds. Traditional methods used for the extraction of these compounds are inefficient in terms of time and energy requirements, since the application of high temperatures for several hours is often necessary. For this reason, the development and optimization of extraction methodologies that are more respectful for the environment are required, which also reduce the costs and the time required in the analytical processes. Therefore, the optimisation of an ultrasound-assisted extraction method is proposed to obtain a crude extract of Arthrospira platensis with the highest possible antioxidant activity. For this purpose, the following parameters were optimised: amount of algae, solvent volume, solvent type and concentration, ultrasound extraction time and power, as well as temperature. For each condition studied, the total phenolic content (using the Folin-Ciocalteu method) and the antioxidant activity (using the DPPH method) were analysed. Subsequently, the high-performance liquid chromatography technique (HPLC-DAD) was used to determine the individual profile of polyphenols. The pigments and polysaccharides present in the extracts were also characterized. The optimized method provided a hydroalcoholic extract with high antioxidant activity (>85%) under very convenient experimental conditions. Biotecnología

The recovery of biopolymers from natural resources using eco-friendly extraction technologies that enhance their mechanical properties has gained attention in recent years. In this context, this work deals with the isolation of hybrid carrageenans from Mastocarpus stellatus red seaweed using subcritical water extraction operating in a wide range of thermal conditions (70–190 °C). The extracted biopolymers were analyzed by means of either Fourier-Transform infrared, nuclear magnetic resonance, rheological or cell viability assays. In parallel, the fundamental chemical composition of the seaweed used as raw material, as well as the main phytochemical properties of the soluble liquid extracts, were also studied. Results indicated that thermal extraction conditions significantly affected the rheological behavior of the recovered hybrid carrageenans. The hybrid carrageenan extraction yields varied, with results between 10.2 and 30.2% being the highest values obtained at hydrothermal treatment of 130 °C. A wide palette of viscous features was identified for recovered hybrid carrageenans, with the strongest rheology properties observed at the same temperature. It should be remarked that the maximum inhibitory effect was also obtained at 130 °C for both the ovarian carcinoma cell line (A2780) (65%, IC50: 0.31 mg/mL) and lung carcinoma cell line (A549) (59%, IC50: 0.41 mg/mL). En la presente tesis se investigaron dos nuevas estrategias de operación para minimizar el contenido de N2 y O2 en el biometano. La primera estrategia operacional consistió en la desgasificación del caldo de cultivo antes de alimentarlo a la columna de lavado de biogás interconectada al HRAP. Para ello se instaló una membrana de contacto líquido-gas de fibra hueca de polidimetilsiloxano (PDMS), la cual se operó a diferentes presiones de vacío. Se obtuvieron eficiencias de eliminación de oxígeno disuelto del caldo de cultivo ≥60 % operando a una presión de vacío entre 100 – 300 mbar, confirmándose la capacidad de la tecnología de membranas acoplada a la limpieza fotosintética del biogás para reducir el contenido de O2 del biometano. La segunda estrategia operacional reveló una mejora en la eliminación de CO2 y H2S al aumentar la presión de operación en la columna de lavado de biogás. Sin embargo, fue necesaria una sobrepresión mínima de 2 atm para limitar la desorción de N2 y O2. Los resultados de ambas estrategias operativas alcanzaron un biometano de alta calidad (CH4 > 87 %, CO2 0.9 – 9 %, N2 0.7 – 9 %, y O2 0 – 2 %) .La segunda parte de la investigación de esta tesis se centró en evaluar el rendimiento de la purificación fotosintética de biogás junto con la producción de biomasa algal en un FBR tubular interconectado a una columna externa de lavado de biogás. Durante 395 días se evaluó y optimizó la influencia de la alcalinidad y de la limitación de nitrógeno (N) en el caldo de cultivo en la calidad del biometano generado y en la composición de la biomasa obtenida. Una alta alcalinidad (~1700 mg L– 1) en el caldo de cultivo aseguró una composición de biometano que cumplía con las regulaciones internacionales para su inyección en redes de gas natural o su uso como combustible vehicular. Por otra parte, no se observó un efecto perjudicial sobre la calidad del biometano cuando se implementaron los ciclos de limitación de N. Además, la limitación de N promovió un aumento en la cantidad de carbohidratos almacenada en la biomasa algal, alcanzando valores hasta 2 veces superiores a los obtenidos en condiciones nutricionales normales. The interest in seaweeds for cosmetic, cosmeceutics, and nutricosmetics is increasing based on the demand for natural ingredients. Seaweeds offer advantages in relation to their renewable character, wide distribution, and the richness and versatility of their valuable bioactive compounds, which can be used as ingredients, as additives, and as active agents in the formulation of skin care products. Bioactive compounds, such as polyphenols, polysaccharides, proteins, peptides, amino acids, lipids, vitamins, and minerals, are responsible for the biological properties associated with seaweeds. Seaweed fractions can also offer technical features, such as thickening, gelling, emulsifying, texturizing, or moistening to develop cohesive matrices. Furthermore, the possibility of valorizing industrial waste streams and algal blooms makes them an attractive, low cost, raw and renewable material. El biogás procedente de la digestión anaerobia de aguas residuales o residuos sólidos orgánicos representa una fuente de energía renovable importante para mitigar el uso de los combustibles fósiles. La purificación de biogás es necesaria antes de su utilización como combustible para vehículos o su inyección en las redes de gas natural. En este contexto, existen diversas normativas internacionales que establecen las concentraciones máximas y mínimas para cada componente del biometano permitidas en función de su aplicación final. Actualmente se dispone de una gran variedad de tecnologías físicoquímicas y biológicas comerciales para eliminar el CO2 y el H2S del biogás. Sin embargo, la mayoría de estas tecnologías deben ser implementadas secuencialmente para eliminar el H2S, el CO2 y contaminantes traza como los siloxanos o los contaminantes orgánicos volátiles. Carrageenan and carrageenan oligosaccharides are red seaweed sulfated carbohydrates with well-known antiviral properties, mainly through the blocking of the viral attachment stage. They also exhibit other interesting biological properties and can be used to prepare different drug delivery systems for controlled administration. The most active forms are λ-, ι-, and κ-carrageenans, the degree and sulfation position being determined in their properties. They can be obtained from sustainable worldwide available resources and the influence of manufacturing on composition, structure, and antiviral properties should be considered. This review presents a survey of the antiviral properties of carrageenan in relation to the processing conditions, particularly those assisted by intensification technologies during the extraction stage, and discusses the possibility of further chemical modifications. El objetivo principal de este proyecto consiste en analizar y cuantificar el contenido en ácidos grasos poliinsaturados (PUFA´s) de una muestra de microalgas liofilizadas del género Nannochloropsis gaditana, así como la realización de ensayos previos de extracción sólido-líquido utilizando disolventes eutécticos (DES´s) con el objeto de evaluar la capacidad extractora y selectiva de los PUFA´s presentes en la muestra de microalgas. Para alcanzar este objetivo global se han planteado los siguientes objetivos específicos: - Caracterización de la masa microalgal. - Desarrollo de un método cromatográfico para el análisis de lípidos (PUFA´s). - Síntesis y ensayos de extracción empleando DES´s. - Análisis de los resultados obtenidos. De acuerdo con estos objetivos específicos, el trabajo se va a articular en los siguientes capítulos: En el capítulo 3 se plantea una descripción de algas y microalgas incluyéndose los métodos de extracción de compuestos bioactivos, haciendo hincapié en las técnicas de extracción actuales y convencionales de lípidos y llevando a cabo una revisión exhaustiva de los ácidos grasos, así como de los disolventes eutécticos profundos. En el capítulo 4 se recogen los materiales y métodos utilizados. En el capítulo 5 se muestran los resultados del estudio. En el capítulo 6 se presentan las principales conclusiones extraídas de los capítulos anteriores. Durante las últimas décadas se ha producido un incremento exponencial en la generación de aguas residuales agroalimentarias como consecuencia del incremento de la producción industrial. Por ello, en la actualidad se pueden encontrar aguas residuales de elevada carga orgánica y una mayor concentración de nutrientes y productos tóxicos, por lo que deben ser tratadas previamente a su vertido para no incurrir en daños medioambientales graves. En este contexto, a día de hoy existen diversas tecnologías que se emplean para tratar las aguas residuales agroalimentarias, como son los tratamientos físico-químicos y los procesos biológicos (aerobios, anóxicos, anaerobios y sus combinaciones). Estas tecnologías conllevan, en muchos casos, costes energéticos y de operación elevados y no permiten valorizar los nutrientes presentes en el agua residual. Una alternativa a los tratamientos biológicos más convencionales es el uso de consorcios de microalgas y bacterias en simbiosis, donde las bacterias oxidan la materia orgánica, liberando CO2, amonio, nitratos, nitritos y fósforo soluble y mediante la fotosíntesis oxigénica las microalgas asimilan estos compuestos en forma de biomasa, liberando O2. Una vez llevado a cabo el tratamiento del agua, las microalgas producidas pueden ser valorizadas en forma de biocombustibles (biodiesel, bioetanol, biogás, etc.) y otros compuestos de alto valor añadido, mejorando la viabilidad económica del proceso global. En los últimos años, la búsqueda de nuevas materias primas renovables ha aumentado exponencialmente debido al gran consumo de materiales y los limitados recursos fósiles. Al mismo tiempo, un importante esfuerzo de investigación se centra en la gestión de las grandes cantidades de desechos generadas por las actividades humanas. Combinando ambos objetivos de investigación, la recuperación y la valorización de los componentes procedentes de desechos parecen ser la única solución sostenible. Las aguas residuales, que contienen gran cantidad de materia orgánica y nutrientes, son uno de los mayores residuos generados en nuestra sociedad. Se están estudiando procesos biotecnológicos con consorcios de microalgas y bacterias para el tratamiento de aguas residuales con el objetivo de mejorar los rendimientos de recuperación de los nutrientes (C, N, P, S ...), así como con el fin de obtener agua limpia para otras aplicaciones. Por lo tanto, esta tesis tiene como objetivo abordar la valorización de la biomasa de microalgas-bacterias cultivadas en fotobiorreactores de tratamiento de aguas residuales. Este trabajo se centra en la valorización de la fracción de carbohidratos de esta biomasa para producir biogás y monosacáridos fermentables, pero considerando el efecto de este proceso en otras fracciones valiosas de la biomasa, como proteínas y lípidos, y la generación de otros subproductos. Due to the possible depletion of fossil fuels in the near future and the necessity of finding new food sources for a growing world population, marine microalgae constitutes a very promising alternative resource, which can also contribute to carbon dioxide fixation. Thus, seven species (Chaetoceros calcitrans, Chaetoceros gracilis, Isochrysis galbana, Nannochloropsis gaditana, Dunaliella salina, Tetraselmis suecica, and Tetraselmis chuii) were grown in five serial batch cultures at a bench scale under continuous illumination. The batch cultures were inoculated with an aliquot that was extracted from a larger-scale culture in order to obtain growth data valid for the entire growth cycle with guaranteed reproducibility. Thus, measurements of optical density at several wavelengths and cell counting with a haemocytometer (Neubauer chamber) were performed every one or two days for 22 days in the five batch cultures of each specie. Modeling of cell growth, the relationship between optical density (OD) and cell concentration and the effect of wavelength on OD was performed. The results of this study showed the highest and lowest growth rate for N. gaditana and T. suecica, respectively. "Sargassum muticum" is a native macroalgae from the Northeastern Pacific coasts, but in the Atlantic coasts is an invasive. No commercial utilization strategies are established. The development of valorization processes could be a good alternative to compensate the collection costs. Marine macroalgae are generating interest, they contain a variety of bioactives with chemical composition and structure not found in any other terrestrial source. The fractions with more potential as a raw material for the production of biologically active compounds are polysaccharides, phlorotannins and terpenoids. The production of three components: fucoxanthin, fucoidans and phlorotannins was proposed. Fucoxanthin. This carotenoid present antioxidant properties and the activities against cancer, obesity, diabetes and malaria were confirmed. In addition to the pharmacological applications, fucoxanthin is highly demanded in weight loss products. Polysaccharides. The polysaccharides have a reserve function and they can provide resistence and flexibility to withstand waves and avoid dessication. These compounds present antioxidant, antiinflamatory, anticancer, inmunomodulatory, neuroprotective, hepatoprotective, healing and antiviral activities. Phlorotannins. They have structural, signalling and defense functions, particularly against the environmental stress, radiation and microbial. They present antioxidant, anticancer, antidiabetes, antialergenic, antihypertensive, anti-infammatory, antimicrobial and antiviral action. The fucoidan and phlorotannin solubilization stages and the evaluation of chemical composition, structure and functions will be addressed, with the aim of developing processing technologies suited for the extraction and concentration of the active components. The recovery of fucoxanthin will also be assessed in this thesis. The concentrated products and fractions will be evaluated for their possible use in food, cosmetic and pharmacological applications. No se han establecido vías de aprovechamiento comercial por lo que conviene desarrollar procesos de valorización. Las macroalgas marinas poseen una variedad de compuestos bioactivos con estructuras que no se encuentran en fuentes terrestres. Las fracciones con más potencial para obtener compuestos biológicamente activos son los polisacáridos, florotaninos y terpenoides. Se ha propuesto la obtención de tres componentes de interés: fucoxantina, fucoidanos y florotaninos. Fucoxantina. Este carotenoide presenta acción antioxidante y se ha confirmado su actividad frente al cáncer, obesidad, diabetes, angiogénesis y malaria. Además de los usos farmacológicos, destacan los productos adelgazantes con fucoxantina. Polisacáridos. Tienen función de reserva y proporcionan resistencia y flexibilidad para soportar el oleaje y evitar la desecación. Los más importantes son laminaranos, alginatos y fucoidanos. Se han confirmado las actividades antioxidante, antiinflamatoria, anticáncer, inmunomodulatoria, neuroprotectora, hepatoprotectora, cicatrizante, antiviral. Florotaninos. Desempeñan funciones estructurales, de señalización y de defensa frente a estrés ambiental, radiación y ataque microbiano. Presentan actividad antioxidante, anticáncer, antidiabetes, antialérgica, antihipertensiva, antin?amatoria, antimicrobiana y antiviral. Se estudiarán los procesos de solubilización de fucoidanos y florotaninos y se abordará el estudio de la composición, elucidación estructural y función de los mismos, para posteriormente desarrollar las tecnologías adecuadas para la concentración de los componentes más activos dependiendo de la actividad biológica deseada. Se evaluará también el grado de recuperación de fucoxantina. Se obtendrán productos concentrados y fracciones enriquecidas en fucoidanos y florotaninos para su posible uso alimentario, cosmético o farmacológico. Téñense confirmado as actividades antioxidante, antiinflamatoria, anticancro, inmunomodulatoria, neuroprotectora, hepatoprotectora, cicatrizante e antiviral. Florotaninos. Desempeñan funciones estructurais, de sinalización e de defensa fronte ao estrés ambiental, radiación e ataque microbiano ou herbívoro. Presentan actividade antioxidante, anticancro, antidiabetes, antialérxica, antihipertensiva, antiinflamatoria, antimicrobiana e antiviral. Estudaranse os procesos de solubilización de fucoidanos e florotaninos e abordaráse o estudo da composición, elucidación estructural e función de los mismos, para posteriormente desenvolver as tecnoloxías máis axeitadas para a concentración dos compoñentes máis activos dependendo da actividad biolóxica desexada. Avaliarase tamén o grao de recuperación de fucoxantina. Se obterán produtos concentrados e fraccións enriquecidas en fucoidanos e florotaninos de algas marróns para o seu posible uso alimentario, cosmético ou farmacolóxico.

El cultivo masivo de microalgas para la producción de biocombustible alternativo al petróleo, compuestos de valor añadido (CAVA) con aplicaciones químicas o farmacéuticas y como fuente de alimento para una población mundial creciente, sigue siendo uno de los campos más prometedores de la bioingeniería y biotecnología. Sin embargo, todavía queda mucho trabajo por realizar en investigación y desarrollo para optar a todas las magníficas posibilidades que ofrece esta fuente renovable. De este modo, en esta revisión se pretende mostrar los principios referentes al cultivo de microalgas: su importancia, los problemas tecnológicos que todavía quedan por solucionar, todos los aspectos relacionados con sus tipos y sistemas de cultivo, las fases del crecimiento, las ecuaciones de la cinética de duplicación celular, características importantes referentes a la selección y aislamiento de cepas, escalado, métodos de caracterización bioquímica (determinación de clorofilas, proteínas, lípidos, carbohidratos, sílice, etc.) y, finalmente, las especies de microalgas más relevantes. However, much work must be done still in research and development to achieve all the possibilities offered by this renewable source. Thus, this review intends to show the principles concerning the cultivation of microalgae: its importance, technological problems still to be solved, all aspects related to their types and systems of cultivation, growth phases, kinetics equations for cell duplication, important aspects related to the selection and isolation of strains, scaling, biochemical characterization methods (determination of chlorophylls, proteins, lipids, carbohydrates, silica, etc.) and finally the most relevant microalgae species. In this work, a very useful and accurate procedure, based on the spectrophotometric method published by the American Public Health Association in the Standards Methods for the Examination of Water and Wastewater, was developed to determine cell density of Isochrysis galbana performing a single direct absorbance measurement in exponential phase of growth, which is the desirable operating mode for any microalgae production plant. Thus, Isochrysis galbana was cultured in serial batch cultures from a larger scale fed-batch culture. The growth performance of this species of microalgae under laboratory conditions was analysed by spectrophotometry at different wavelengths and cell counting in a haemocytometer (Neubauer chamber) showing that doubling times and cell death increased with increasing initial cell density. Besides, it was demonstrated that the absorbance of these cultures followed a linear trend as a function of time and cell density during the exponential phase of growth, results in which the developed direct method is based on. En este trabajo, se ha determinado un procedimiento muy útil y preciso basado en el método espectrofotométrico descrito por la Asociación Americana Pública de Salud en los Métodos Estándar para el Análisis de Aguas y Aguas Residuales, para calcular la densidad celular de cultivos de Isochrysis galbana realizando una única medida de absorbancia en fase exponencial de crecimiento que es el modo de trabajo deseable de cualquier planta de producción de microalgas. De este modo, se realizó una serie de cultivos de Isochrysis galbana en discontinuo a partir de otro cultivo a mayor escala con alimentación intermitente. Se ha analizado el nivel de crecimiento de esta especie de microalga por espectrofotometría a diferentes longitudes de onda y recuento de células en un hemocitómetro (cámara de Neubauer) mostrando que el tiemplo de duplicación y la muerte celular aumentó con el aumento de la densidad celular inicial. Además, se ha demostrado que la absorbancia de estos cultivos sigue una tendencia lineal en función del tiempo y densidad celular durante la fase de crecimiento exponencial, resultados en los cuales se basa el método directo desarrollado. Actualmente, en varios procesos industriales de extracción las nuevas tecnologías para reducir tiempo, espacio y energía, así como la cantidad de los disolventes utilizados, son bienvenidas como candidatas a la intensificación de la producción. Esto es especialmente importante en los casos complejos de materiales naturales donde la ruptura de las pareced celulares es la etapa limitante que conduce a cinéticas lentas y rendimientos bajos. Entre varios tratamientos mecánicos, químicos, físicos o fisicoquímicos, está el uso de microondas en materias naturales. Debido a la irradiación electromagnética de la materia, el agua intracelular se calienta y se evapora rápidamente, y el gradiente de presión generado causa probablemente el daño de la pared celular. En consecuencia, los compuestos intracelulares de interés llegan a ser más asequibles con un pretratamiento eficaz, y se logra mejorar la cinética del posterior proceso de extracción convencional.
