However, there are still concerns about the potential adversities that these nanoparticles may present in an organism. In particular, whether they could cause an exacerbated cytotoxic response in susceptible tissues due to damage or disease, such as the heart, liver, spleen, or kidneys. In this regard, this study aims to evaluate the cytotoxicity of mono- and bimetallic nanoparticles of gold and silver (Au:Ag NPs) on healthy and hypertrophic cardiac H9c2 cells, and on healthy and metabolically activated macrophages derived from U937 cells. The main objective of this work is to explore the susceptibility of cells due to exposure to Au:Ag NPs in conditions representing cardiometabolic diseases.Methods: Au:Ag NPs were synthesized in different molar ratios (Au:Ag, 100:0, 75:25, 50:50, 25:75, 0:100) using starch as a capping and reducing agent. Their physicochemical properties were characterized through UV-vis spectroscopy, X-ray diffraction (XRD), Fourier transform infrared (FTIR) spectroscopy, dynamic light scattering (DLS), ζ-potential measurements, and transmission electron microscopy (TEM). Moreover, the effect of the metal-based nanoparticle exposure on healthy and hypertrophic H9c2 cells was measured by analyzing the cellular vitality, the loss of mitochondrial membrane potential (∆Ψm), and the production of mitochondrial reactive oxygen species (mROS).Results: The Au:Ag NPs did not affect the cell vitality of healthy or metabolically activated macrophages. On the contrary, healthy H9c2 cells showed decreased mitochondrial metabolism when exposed to NPs with higher Ag concentrations. Furthermore, hypertrophic H9c2 cells were more susceptible to the same NPs compared to their non-hypertrophic counterparts, and presented a pronounced loss of ∆Ψm. In addition, these NPs increased the production of mROS and regulated cell death in both cardiac cells.Conclusion: In conclusion, low doses of high-Ag load in Au:Ag NPs produced cytotoxicity on H9c2 cardiac cells, with hypertrophic cells being more susceptible. These results suggest that cardiac hypertrophic conditions are more prone to a cytotoxic response in the presence of bimetallic Au:Ag NPs compared to healthy cells. In addition, this work opens the door to explore the nanotoxicity of noble metal-based NPs in biological disease conditions.Keywords: cardiovascular disease, hypertrophic cardiomyoblasts, bimetallic nanoparticles, metabolically activated macrophages

Abstract Diabetic nephropathy is one of the most frequent complications of diabetic patients and is the leading cause of end-stage renal disease worldwide. The complex physiopathology of this complication raises a challenge in the development of effective medical treatments. Therefore, a better understanding of this disease is necessary for producing more targeted therapies. In this work we propose human amylin as a possible mediator in the development of diabetic nephropathy. Islet amyloid polypeptide or amylin is a hormone co-secreted with insulin. The human isoform has the ability to fold and form amyloid aggregates in the pancreas of patients with type 2 diabetes mellitus, disrupting cellular homeostasis due to its ability to form pores in lipid bilayers. It has been described that hIAPP can be secreted and exported in extracellular vesicles outside the pancreas, being a plausible connecting mechanism between the β-cell and other peripheral tissues such as the kidney. Here, we demonstrate that tubular, podocytes and mesangial cells can incorporate hIAPP coming from β-cells. Then, this hIAPP can form aggregates inside these kidney cells, contributing to its failure. In order to study the consequences in vivo, we found amylin aggregates in the kidney of mice overexpressing hIAPP after feeding a high fat diet. In addition, we observed an increase in glomerulosclerosis index and inflammation. Specifically, there were significant changes in signalling pathways directly involved in the diabetic nephropathy such as an increased in mTORC1 signaling pathway, an alteration in mitochondrial dynamics and an increased in endoplasmic reticulum stress. All these results demonstrate the importance of hIAPP in the kidney and its possible contribution in the development of diabetic nephropathy.

Eggplant is a vegetable grown worldwide, and due to quality standards, large amounts of biomass are generated after harvest. Biomass is considered a source of bioactive compounds with antioxidant properties. Therefore, this research aimed to evaluate the bioaccessibility (BA) and antioxidant capacity of microencapsulated alkaloids from eggplant fruit biomass. Eggplant biomass was collected, and the total alkaloid content, antioxidant capacity (TEAC, FRAP, and ORAC), and alkaloid profile (UPLC/MS) were determined before and after the in vitro digestion of encapsulated and non-encapsulated alkaloids. In vitro digestion significantly reduced the total alkaloid content and antioxidant capacity of alkaloid-rich extracts. Microencapsulation increased the bioaccessibility of alkaloid-rich extracts threefold, and the antioxidant capacity increased by up to 50%. The antioxidant capacity of digested microcapsules increased, and their bioaccessibility was higher than that of non-encapsulated alkaloids. Solamargine and solasonine decreased by 17 and 15% BA, respectively, during in vitro digestion; however, microencapsulation protected these alkaloids during in vitro digestion and enhanced their content. This study demonstrates that microencapsulation is a feasible option to protect alkaloids and preserve their antioxidant capacity during gastrointestinal digestion, as well as to give added value to eggplant plant biomass.

Polyphenols have attracted attention for their anti-inflammatory, antidiabetic, and anticancer properties. Due to the antioxidant and anti-inflammatory potential of these molecules, they are also proposed as a potential therapeutic tool to prevent complications of cancer and decrease the secondary effects of conventional chemotherapeutic drugs. Nonetheless, polyphenols such as flavonoids and phenolic acids have low bioavailability, as they are highly metabolized. Thus, administration strategies have been developed to enhance the anticancer properties of polyphenols. Most of these strategies involve different encapsulation techniques, such as nanoencapsulation, nanoemulsion, and the use of other polymeric matrixes. These techniques can increase the activity of these compounds after going through the gastrointestinal process and improve their solubility in an aqueous medium. This review comprises recent studies regarding encapsulation techniques to enhance the bioactivity of polyphenols against cancer and their current state in clinical studies. Overall, micro- and nanoencapsulation techniques with different polymers enhanced the anticancer properties of polyphenols by inhibiting tumor growth, modulating the expression of genes related to metastasis and angiogenesis, decreasing the expression of pro-inflammatory biomarkers.

Chitosan is a non-toxic, biodegradable, and biocompatible natural biopolymer widely used as a nanocarrier, emulsifier, flocculant, and antimicrobial agent with potential applications in industry. Recently, chitosan has been used as an encapsulating agent for bioactive plant compounds and agrochemicals by different technologies, such as spray-drying and nanoemulsions, to enhance antimicrobial activity. Chitosan nanocomposites have been shown to increase potential biocidal, antibacterial, and antifungal activity against pathogens, presenting higher stability, decreasing degradation, and prolonging the effective concentration of these bioactive compounds. Therefore, the objective of this work is to review the most outstanding aspects of the most recent developments in the different methods of encapsulation of bioactive compounds (phenolic compounds, essential oils, among others) from plants, as well as the applications on phytopathogenic diseases (fungi and bacteria) in vitro and in vivo in cereal, fruit and vegetable crops. These perspectives could provide information for the future formulation of products with high efficacy against phytopathogenic diseases as an alternative to chemical products for sustainable agriculture.

Epizootic hemorrhagic disease (EHD) is a non-contagious arthropod-transmitted viral disease and a World Organization for Animal Health (WOAH)-listed disease of domestic and wild ruminants since 2008. EHDV is transmitted among susceptible animals by a few species of midges of genus Culicoides. During the fall of 2021, a large outbreak caused by the epizootic hemorrhagic disease virus (EHDV), identified as serotype 8, was reported in Tunisian dairy and beef farms with Bluetongue virus (BTV)-like clinical signs. The disease was detected later in the south of Italy, in Spain, in Portugal and, more recently, in France, where it caused severe infections in cattle. This was the first evidence of EHDV-8 circulation outside Australia since 1982. In this study, we analyzed the epidemiological situation of the 2021–2022 EHDV outbreaks reported in Tunisia, providing a detailed description of the spatiotemporal evolution of the disease. We attempted to identify the eco-climatic factors associated with infected areas using generalized linear models (GLMs). Our results demonstrated that environmental factors mostly associated with the presence of C. imicola, such as digital elevation model (DEM), slope, normalized difference vegetation index (NDVI), and night-time land surface temperature (NLST)) were by far the most explanatory variables for EHD repartition cases in Tunisia that may have consequences in neighboring countries, both in Africa and Europe through the spread of infected vectors. The risk maps elaborated could be useful for disease control and prevention strategies.
