Science - Abstract
Abstract Due to their exceptional solubility and stability, nanobodies have emerged as powerful building blocks for research tools and therapeutics. However, their generation in llamas is cumbersome and costly. Here, by inserting an engineered llama immunoglobulin heavy chain (IgH) locus into IgH-deficient mice, we generate a transgenic mouse line, which we refer to as ‘LamaMouse’. We demonstrate that LamaMice solely express llama IgH molecules without association to Igκ or λ light chains. Immunization of LamaMice with AAV8, the receptor-binding domain of the SARS-CoV-2 spike protein, IgE, IgG2c, and CLEC9A enabled us to readily select respective target-specific nanobodies using classical hybridoma and phage display technologies, single B cell screening, and direct cloning of the nanobody-repertoire into a mammalian expression vector. Our work shows that the LamaMouse represents a flexible and broadly applicable platform for a facilitated selection of target-specific nanobodies.

The reaction $\gamma p \rightarrow p \pi^0 \eta$ has been studied with the CBELSA/TAPS detector at the electron stretcher accelerator ELSA in Bonn for incident photon energies from threshold up to 3.1 GeV. This paper has been motivated by the recently claimed observation of a narrow structure in the M$_{N\eta}$ invariant mass distribution at a mass of 1678 MeV/$c^2$. The existence of this structure cannot be confirmed in the present work. Instead, for E$_{\gamma}$ = 1400 - 1500 MeV and the cut M$_{p\pi^0} \le 1190 $ MeV/$c^2$ a statistically significant structure in the M$_{p\eta}$ invariant mass distribution near 1700 MeV/$c^2$ is observed with a width of $\Gamma\approx 35$ MeV/$c^2$ while the mass resolution is $\sigma_{res}$ = 5 MeV/$c^2$. Increasing the incident photon energy from 1420 to 1540 MeV this structure shifts in mass from $\approx$ 1700MeV/c$^2$ to $\approx$ 1725 MeV/$c^2$; the width increases to about 50 MeV/$c^2$ and decreases thereafter. The cross section associated with this structure reaches a maximum of $\approx$ 100 nb around E$_{\gamma} \approx$ 1490 MeV (W $\approx $ 1920 MeV), which coincides with the $p a_0$ threshold. Three scenarios are discussed which might be the origin of this structure in the M$_{p\eta}$ invariant mass distribution. The most likely interpretation is that it is due to a triangular singularity in the $\gamma p \rightarrow p a_0 \rightarrow p \pi^0 \eta$ reaction, Comment: 16 pages, 18 figure

We report the first measurement of the average of the electron-proton and positron-proton elastic scattering cross sections. This lepton charge-averaged cross section is insensitive to the leading effects of hard two-photon exchange, giving more robust access to the proton's electromagnetic form factors. The cross section was extracted from data taken by the OLYMPUS experiment at DESY, in which alternating stored electron and positron beams were scattered from a windowless gaseous hydrogen target. Elastic scattering events were identified from the coincident detection of the scattered lepton and recoil proton in a large-acceptance toroidal spectrometer. The luminosity was determined from the rates of M{\o}ller, Bhabha and elastic scattering in forward electromagnetic calorimeters. The data provide some selectivity between existing form factor global fits and will provide valuable constraints to future fits., Comment: 6 pages, 3 figures, submitted to PRL

The polarization observables $T, E, P, H$, and $G$ in photoproduction of $\eta$ mesons off protons are measured for photon energies from threshold to $W=2400\,$MeV ($T$), 2280 MeV ($E$), 1620 MeV ($P, H$), or 1820 MeV ($G$), covering nearly the full solid angle. The data are compared to predictions from the SAID, MAID, J\"uBo, and BnGa partial-wave analyses. A refit within the BnGa approach including further data yields precise branching ratios for the $N\eta$ decay of nucleon resonances. A $N\eta$-branching ratio of $0.33\pm 0.04$ for $N(1650)1/2^-$ is found, which reduces the large and controversially discussed $N\eta$-branching ratio difference of the two lowest mass $J^P=1/2^-$-resonances significantly., Comment: 10 pages, 11 figures

A measurement of the double-polarization observable $E$ for the reaction $\gamma p\to \pi^0 p$ is reported. The data were taken with the CBELSA/TAPS experiment at the ELSA facility in Bonn using the Bonn frozen-spin butanol (C$_4$H$_9$OH) target, which provided longitudinally-polarized protons. Circularly-polarized photons were produced via bremsstrahlung of longitudinally-polarized electrons. The data cover the photon energy range from $E_\gamma =600$~MeV to $E_\gamma =2310$~MeV and nearly the complete angular range. The results are compared to and have been included in recent partial wave analyses., Comment: 21 pages

Local and national governments around the world are currently considering the elimination of the annual switch to and from Daylight Saving Time (DST). As an international organization of scientists dedicated to studying circadian and other biological rhythms, the Society for Research on Biological Rhythms (SRBR) engaged experts in the field to write a Position Paper on the consequences of choosing to live on DST or Standard Time (ST). The authors take the position that, based on comparisons of large populations living in DST or ST or on western versus eastern edges of time zones, the advantages of permanent ST outweigh switching to DST annually or permanently. Four peer reviewers provided expert critiques of the initial submission, and the SRBR Executive Board approved the revised manuscript as a Position Paper to help educate the public in their evaluation of current legislative actions to end DST.

Photoproduction of $\eta$ mesons from neutrons} \abstract{Results from measurements of the photoproduction of $\eta$ mesons from quasifree protons and neutrons are summarized. The experiments were performed with the CBELSA/TAPS detector at the electron accelerator ELSA in Bonn using the $\eta\to3\pi^{0}\to6\gamma$ decay. A liquid deuterium target was used for the measurement of total cross sections and angular distributions. The results confirm earlier measurements from Bonn and the MAMI facility in Mainz about the existence of a narrow structure in the excitation function of $\gamma n\rightarrow n\eta$. The current angular distributions show a forward-backward asymmetry, which was previously not seen, but was predicted by model calculations including an additional narrow $P_{11}$ state. Furthermore, data obtained with a longitudinally polarized, deuterated butanol target and a circularly polarized photon beam were analyzed to determine the double polarization observable $E$. Both data sets together were also used to extract the helicity dependent cross sections $\sigma_{1/2}$ and $\sigma_{3/2}$. The narrow structure in the excitation function of $\gamma n\rightarrow n\eta$ appears associated with the helicity-1/2 component of the reaction.

Summary There is an ongoing debate in the United Kingdom and in other countries about whether twice‐yearly changes into and out of Daylight Saving Time should be abolished. Opinions are divided about whether any abolition of Daylight Saving Time should result in permanent Standard Time, or year‐long Daylight Saving Time. The British Sleep Society concludes from the available scientific evidence that circadian and sleep health are affected negatively by enforced changes of clock time (especially in a forward direction) and positively by the availability of natural daylight during the morning. Thus, our recommendation is that the United Kingdom should abolish the twice‐yearly clock change and reinstate Standard Time throughout the year. [ABSTRACT FROM AUTHOR]

Abstract Early school times fundamentally clash with the late sleep of teenagers. This mismatch results in chronic sleep deprivation posing acute and long-term health risks and impairing students' learning. Despite immediate short-term benefits for sleep, the long-term effects of later starts remain unresolved. In a pre-post design over 1 year, we studied a unique flexible school start system, in which 10–12th grade students chose daily between an 8:00 or 8:50AM-start. Missed study time (8:00–8:50) was compensated for during gap periods or after classes. Based on 2 waves (6–9 weeks of sleep diary each), we found that students maintained their ~ 1-h-sleep gain on later days, longitudinally (n = 28) and cross-sectionally (n = 79). This gain was independent of chronotype and frequency of later starts but attenuated for boys after 1 year. Students showed persistently better sleep quality and reduced alarm-driven waking and reported psychological benefits (n = 93) like improved motivation, concentration, and study quality on later days. Nonetheless, students chose later starts only infrequently (median 2 days/week), precluding detectable sleep extensions in the flexible system overall. Reasons for not choosing late starts were the need to make up lost study time, preference for extra study time and transport issues. Whether flexible systems constitute an appealing alternative to fixed delays given possible circadian and psychological advantages warrants further investigation.

Abstract The mismatch between teenagers’ late sleep phase and early school start times results in acute and chronic sleep reductions. This is not only harmful for learning but may reduce career prospects and widen social inequalities. Delaying school start times has been shown to improve sleep at least short-term but whether this translates to better achievement is unresolved. Here, we studied whether 0.5–1.5 years of exposure to a flexible school start system, with the daily choice of an 8 AM or 8:50 AM-start, allowed secondary school students (n = 63–157, 14–21 years) to improve their quarterly school grades in a 4-year longitudinal pre-post design. We investigated whether sleep, changes in sleep or frequency of later starts predicted grade improvements. Mixed model regressions with 5111–16,724 official grades as outcomes did not indicate grade improvements in the flexible system per se or with observed sleep variables nor their changes—the covariates academic quarter, discipline and grade level had a greater effect in our sample. Importantly, our finding that intermittent sleep benefits did not translate into detectable grade changes does not preclude improvements in learning and cognition in our sample. However, it highlights that grades are likely suboptimal to evaluate timetabling interventions despite their importance for future success.

The OLYMPUS collaboration reports on a precision measurement of the positron-proton to electron-proton elastic cross section ratio, $R_{2\gamma}$, a direct measure of the contribution of hard two-photon exchange to the elastic cross section. In the OLYMPUS measurement, 2.01~GeV electron and positron beams were directed through a hydrogen gas target internal to the DORIS storage ring at DESY. A toroidal magnetic spectrometer instrumented with drift chambers and time-of-flight scintillators detected elastically scattered leptons in coincidence with recoiling protons over a scattering angle range of $\approx 20\degree$ to $80\degree$. The relative luminosity between the two beam species was monitored using tracking telescopes of interleaved GEM and MWPC detectors at $12\degree$, as well as symmetric M{\o}ller/Bhabha calorimeters at $1.29\degree$. A total integrated luminosity of 4.5~fb$^{-1}$ was collected. In the extraction of $R_{2\gamma}$, radiative effects were taken into account using a Monte Carlo generator to simulate the convolutions of internal bremsstrahlung with experiment-specific conditions such as detector acceptance and reconstruction efficiency. This paper reports on a measurement of the double-polarization observable G in $\pi^0$ photoproduction off the proton using the CBELSA/TAPS experiment at the ELSA accelerator in Bonn. The observable G is determined from reactions of linearly-polarized photons with longitudinally-polarized protons. The polarized photons are produced by bremsstrahlung off a properly oriented diamond radiator. A frozen spin butanol target provides the polarized protons. The data cover the photon energy range from 617 to 1325 MeV and a wide angular range. The experimental results for G are compared to predictions by the Bonn-Gatchina (BnGa), J\"ulich-Bonn (J\"uBo), MAID and SAID partial wave analyses. Implications of the new data for the pion photoproduction multipoles are discussed.

Data on the polarization observables T, P, and H for the reaction $\gamma p\to p\pi^0$ are reported. Compared to earlier data from other experiments, our data are more precise and extend the covered range in energy and angle substantially. The results were extracted from azimuthal asymmetries measured using a transversely polarized target and linearly polarized photons. The data were taken at the Bonn electron stretcher accelerator ELSA with the CBELSA/TAPS detector. Within the Bonn-Gatchina partial wave analysis, the new polarization data lead to a significant narrowing of the error band for the multipoles for neutral-pion photoproduction.

The first measurements of the beam-target-helicity-asymmetries $E$ and $G$ in the photoproduction of $\omega$-mesons off protons at the CBELSA/TAPS experiment are reported. $E$ ($G$) was measured using circularly (linearly) polarised photons and a longitudinally polarised target. $E$ was measured over the photon energy range from close to threshold ($E_\gamma = 1108$~MeV) to $E_\gamma = 2300$~MeV and $G$ at a single energy interval of $1108 < E_\gamma <1300$~MeV. Both measurements cover the full solid angle. The observables $E$ and $G$ are highly sensitive to the contribution of baryon resonances, with $E$ acting as a helicity filter in the $s$-channel. The new results indicate significant $s$-channel resonance contributions together with contributions from $t$-channel exchange processes. A partial wave analysis reveals strong contributions from the partial waves with spin-parity $J^P=3/2^+, 5/2^+$, and $3/2^-$., Comment: 6 pages, 6 figures

Nuclear Experiment - Abstract
In a search for $\omega$ mesic states, the production of $\omega$-mesons in coincidence with forward going protons has been studied in photon induced reactions on $^{12}$C for incident photon energies of 1250 - 3100 MeV. The $\pi^0 \gamma$ pairs from decays of bound or quasi-free $\omega$-mesons have been measured with the CBELSA/TAPS detector system in coincidence with protons registered in the MiniTAPS forward array. Structures in the total energy distribution of the $\pi^0 \gamma$ pairs, which would indicate the population and decay of bound $\omega~^{11}$B states, are not observed. The $\pi^0 \gamma$ cross section of 0.3 nb/MeV/sr observed in the bound state energy regime between -100 and 0 MeV may be accounted for by yield leaking into the bound state regime because of the large in-medium width of the $\omega$-meson. A comparison of the measured total energy distribution with calculations suggests the real part $V_0$ of the $\omega~^{11}$B potential to be small and only weakly attractive with $V_0(\rho=\rho_0) = -15\pm$ 35(stat) $\pm$20(syst) MeV in contrast to some theoretical predictions of attractive potentials with a depth of 100 - 150 MeV., Comment: 13 pages, 8 figures

The first measurement of the helicity dependence of the photoproduction cross section of single neutral pions off protons is reported for photon energies from 600 to 2300\,MeV, covering nearly the full solid angle. The data are compared to predictions from the SAID, MAID, and BnGa partial wave analyses. Strikingly large differences between data and predictions are observed which are traced to differences in the helicity amplitudes of well known and established resonances. Precise values for the helicity amplitudes of several resonances are reported.

The OLYMPUS experiment was designed to measure the ratio between the positron-proton and electron-proton elastic scattering cross sections, with the goal of determining the contribution of two-photon exchange to the elastic cross section. Two-photon exchange might resolve the discrepancy between measurements of the proton form factor ratio, $\mu_p G^p_E/G^p_M$, made using polarization techniques and those made in unpolarized experiments. OLYMPUS operated on the DORIS storage ring at DESY, alternating between 2.01~GeV electron and positron beams incident on an internal hydrogen gas target. The experiment used a toroidal magnetic spectrometer instrumented with drift chambers and time-of-flight detectors to measure rates for elastic scattering over the polar angular range of approximately $25^\circ$--$75^\circ$. Symmetric M{\o}ller/Bhabha calorimeters at $1.29^\circ$ and telescopes of GEM and MWPC detectors at $12^\circ$ served as luminosity monitors. A total luminosity of approximately 4.5~fb$^{-1}$ was collected over two running periods in 2012. The excitation function and momentum distribution of $\eta^\prime$ mesons have been measured in photon induced reactions on $^{12}{}$C in the energy range of 1250-2600 MeV. The experiment was performed with tagged photon beams from the ELSA electron accelerator using the Crystal Barrel and TAPS detectors. The data are compared to model calculations to extract information on the sign and magnitude of the real part of the $\eta^\prime$-nucleus potential. Within the model, the comparison indicates an attractive potential of -($37 \pm 10(stat)\pm10(syst)$) MeV depth at normal nuclear matter density. Since the modulus of this depth is larger than the modulus of the imaginary part of the $\eta^\prime$-nucleus potential of -($10\pm2.5$) MeV, determined by transparency ratio measurements, a search for resolved $\eta^\prime$-bound states appears promising., Comment: 13 pages, 6 figures

Nuclear Experiment - Abstract
The first measurement is reported of the double-polarization observable G in photoproduction of neutral pions off protons, covering the photon energy range from 620 to 1120 MeV and the full solid angle. G describes the correlation between the photon polarization plane and the scattering plane for protons polarized along the direction of the incoming photon. The observable is highly sensitive to contributions from baryon resonances. The new results are compared to the predictions from SAID, MAID, and BnGa partial wave analyses. In spite of the long-lasting efforts to understand {\gamma}p -> p{\pi} 0 as the simplest photoproduction reaction, surprisingly large differences between the new data and the latest predictions are observed which are traced to different contributions of the N (1535) with spin-parity J^P = 1/2^- and N (1520) with J^P = 3/2^- . In the third resonance region, where N (1680) with J^P = 5/2^+ production dominates, the new data are reasonably close to the predictions., Comment: Submitted for publication in PRL

The use of GEM foils for the amplification stage of a TPC instead of a con- ventional MWPC allows one to bypass the necessity of gating, as the backdrift is suppressed thanks to the asymmetric field configuration. This way, a novel continuously running TPC, which represents one option for the PANDA central tracker, can be realized. A medium sized prototype with a diameter of 300 mm and a length of 600 mm will be tested inside the FOPI spectrometer at GSI using a carbon or lithium beam at intermediate energies (E = 1-3AGeV). This detector test under realistic experimental conditions should allow us to verify the spatial resolution for single tracks and the reconstruction capability for displaced vertexes. In response to critiques of the ‘slavery versus freedom’ binary and its limitations, researchers at the international Bonn Center for Dependency and Slavery Studies (BCDSS—www.dependency.uni-bonn.de) at the University of Bonn tentatively employ the analytical concept of ‘asymmetrical dependency’ in their investigations of coercive social relations, such as slavery, debt bondage, and servitude. In this paper, we discuss some basic theoretical assumptions that undergird this analytical concept. In outlining an approach to asymmetrical dependency that is grounded in social and cultural theory, our goal is to provide a framework within which individual researchers can situate their projects and further develop their theoretical understanding of this phenomenon. To this end, we first introduce the analytical concept of asymmetrical dependency and explore its potential in light of the current state of research of slavery studies and related fields. We then conceptualize asymmetrical dependency as a dynamic relational process and employ a chiefly praxeological methodology to identify and describe some fundamental dynamics of these relations. Finally, we argue that the interdisciplinary study of asymmetrical dependency requires a broad practice of comparative analyses. We, therefore, consider several recent critiques of and models for comparison while relating them to the analytical concept of asymmetrical dependency we propose.

BackgroundMelatonin modulates circadian rhythms in physiology and sleep initiation. Genetic variants of the MTNR1B locus, encoding the melatonin MT2 receptor, have been associated with increased type 2 diabetes (T2D) risk. Carriers of the common intronic MTNR1B rs10830963 T2D risk variant have modified sleep and circadian traits such as changes of the melatonin profile. However, it is currently unknown whether rare variants in the MT2 coding region are also associated with altered sleep and circadian phenotypes, including meal timing.Materials and MethodsIn this pilot study, 28 individuals [50% male; 46–82 years old; 50% with rare MT2 mutations (T2D MT2)] wore actigraphy devices and filled out daily food logs for 4 weeks. We computed circadian, sleep, and caloric intake phenotypes, including sleep duration, timing, and regularity [assessed by the Sleep Regularity Index (SRI)]; composite phase deviations (CPD) as well a sleep timing-based proxy for circadian misalignment; and caloric intake patterns throughout the day. Using regression analyses, we estimated age- and sex-adjusted mean differences (MD) and 95% confidence intervals (95%CI) between the two patient groups. Secondary analyses also compare T2D MT2 to 15 healthy controls.ResultsPatients with rare MT2 mutations had a later sleep onset (MD = 1.23, 95%CI = 0.42;2.04), and midsleep time (MD = 0.91, 95%CI = 0.12;1.70), slept more irregularly (MD in SRI = −8.98, 95%CI = −16.36;−1.60), had higher levels of behavioral circadian misalignment (MD in CPD = 1.21, 95%CI = 0.51;1.92), were more variable in regard to duration between first caloric intake and average sleep offset (MD = 1.08, 95%CI = 0.07;2.08), and had more caloric episodes in a 24 h day (MD = 1.08, 95%CI = 0.26;1.90), in comparison to T2D controls. Secondary analyses showed similar patterns between T2D MT2 and non-diabetic controls.ConclusionThis pilot study suggests that compared to diabetic controls, T2D MT2 patients display a number of adverse sleep, circadian, and caloric intake phenotypes, including more irregular behavioral timing. A prospective study is needed to determine the role of these behavioral phenotypes in T2D onset and severity, especially in view of rare MT2 mutations.

The polarization observables T,E,P,H, and G in photoproduction of η mesons off protons are measured for photon energies from threshold to W=2400 MeV (T), 2280 MeV (E), 1620 MeV (P,H), or 1820 MeV (G), covering nearly the full solid angle. The data are compared to predictions from the SAID, MAID, JüBo, and BnGa partial-wave analyses. A refit within the BnGa approach including further data yields precise branching ratios for the Nη decay of nucleon resonances. A Nη-branching ratio of 0.33±0.04 for N(1650)1/2− is found, which reduces the large and controversially discussed Nη-branching ratio difference of the two lowest mass JP=1/2−-resonances significantly. Many regions and countries are reconsidering their use of Daylight Saving Time (DST) but their approaches differ. Some, like Japan, that have not used DST over the past decades are thinking about introducing this twice-a-year change in clock time, while others want to abolish the switch between DST and Standard Time, but don’t agree which to use: California has proposed keeping perennial DST (i.e., all year round), and the EU debates between perennial Standard Time and perennial DST. Related to the discussion about DST is the discussion to which time zone a country, state or region should belong: the state of Massachusetts in the United States is considering switching to Atlantic Standard Time, i.e., moving the timing of its social clock (local time) 1 h further east (which is equivalent to perennial DST), and Spain is considering leaving the Central European Time to join Greenwich Mean Time (GMT), i.e., moving its social timing 1 h further west. A wave of DST discussions seems to periodically sweep across the world. Although DST has always been a political issue, we need to discuss the biology associated with these decisions because the circadian clock plays a crucial role in how the outcome of these discussions potentially impacts our health and performance. Here, we give the necessary background to understand how the circadian clock, the social clock, the sun clock, time zones, and DST interact. We address numerous fallacies that are propagated by lay people, politicians, and scientists, and we make suggestions of how problems associated with DST and time-zones can be solved based on circadian biology.

Internal clocks driving rhythms of about a day (circadian) are ubiquitous in animals, allowing them to anticipate environmental changes. Genetic or environmental disturbances to circadian clocks or the rhythms they produce are commonly associated with illness, compromised performance or reduced survival. Nevertheless, some animals including Arctic mammals, open sea fish and social insects such as honeybees are active around-the-clock with no apparent ill effects. The mechanisms allowing this remarkable natural plasticity are unknown. We generated and validated a new and specific antibody against the clock protein PERIOD of the honeybee Apis mellifera (amPER) and used it to characterize the circadian network in the honeybee brain. We found many similarities to Drosophila melanogaster and other insects, suggesting common anatomical organization principles in the insect clock that have not been appreciated before. Time course analyses revealed strong daily oscillations in amPER levels in foragers, which show circadian rhythms, and also in nurses that do not, although the latter have attenuated oscillations in brain mRNA clock gene levels. The oscillations in nurses show that activity can be uncoupled from the circadian network and support the hypothesis that a ticking circadian clock is essential even in around-the-clock active animals in a constant physical environment.

The first measurements of the beam-target-helicity-asymmetries E and G in the photoproduction of ω-mesons off protons at the CBELSA/TAPS experiment are reported. E (G) was measured using circularly (linearly) polarised photons and a longitudinally polarised target. E was measured over the photon energy range from close to threshold (Eγ=1108 MeV) to Eγ=2300 MeV and G at a single energy interval of 1108
