Background: Dental caries is one of the most common chronic diseases, and it is caused by the acid fermentation of bacteria that have become attached to the teeth. Streptococcus mutans (S. mutans) and Lactobacillus acidophilus (L. acidophilus) anchor surface proteins to the cell wall and form a biofilm to aid adhesion to the tooth surface. Some natural plant products, particularly several flavonoids, are effective inhibitors. However, given the scarcity of inhibitors and the emergence of drug resistance, the development of new inhibitors is critical. The high-throughput virtual screening approach was used in this study to identify new potential inhibitor of against S. mutans and L. acidophilus by using ligand (Ellagic acid). Aim: To evaluate the drug interaction ligand (Ellagic acid) and protein [A3VP1 of AgI/II] of Streptococcus mutans (PDB ID: 3IPK), glucan-1,6 - alpha-glucosidase from Lactobacillus acidophilus NCFM (PDB ID: 4AIE). Materials and Methodology: The pdb format of two selected proteins was retrieved from the RCSB protein database. Then inhibitors were docked with protein (A3VP1 of AgI/II) and glucan-1,6-alpha-glucosidase to identify the potent inhibitor. An evaluation criterion was based on the binding affinities by using AutoDock. Results: The binding energy of Ellagic acid - Streptococcus mutans docked complex-10.63 kcal/mol and with Ellagic acid – Lactobacillus acidophilus docked complex was -7.30 kcal/mol. Conclusion: In this study, Showed that lesser binding energy better is the binding of the ligand and protein. These findings can provide a new strategy for dental caries disease therapy by using Ellagic acid as a inhibitor against Streptococcus mutans and Lactobacillus acidophilus

Aim: To evaluate the clinical success of Calcium Hydroxide (CH), Mineral trioxide aggregate (MTA) and Biodentine as pulp capping materials for Direct pulp capping in carious molars. Materials and Methods: Thirty-six molars of thirty-six patients with deep caries lesions, diagnosed with reversible pulpitis were subjected to direct pulp capping treatment. They were randomly divided into three groups, Biodentine (12 teeth) or MTA group (12 teeth) or CH group (12teeth). Simple randomization of three was employed to allocate the treatment materials. Patients were recalled at one, three and six months to evaluate the clinical success of the treatment outcome. Results: In clinical trial/study, the pulp capping materials gave different success rate, 91.67% success in the Biodentine group, 75% success in the MTA group and 41.67% success in Calcium Hydroxide group. Conclusion: In our study the materials tested at 1 month, 3 month and 6-month follow-up, Calcium Silicate materials are better than calcium hydroxide and comparably Biodentine is better than MTA for Direct pulp capping. Clinical Significance: The findings of this clinical trial could promote the reliability of pulp capping materials for treatment of deep carious lesions by conservative approach rather than opting endodontic management.

Nigella sativa (kalonji) has been used since ancient times as a nutritional supplement and for treating various infections and chronic ailments. As pathogens become resistant to most drugs, kalonji can be used as an alternative compound in modern medicines. The use of herbal extracts as endodontic irrigants might be beneficial as a part of a growing trend to seek natural remedies for dental treatment. Aim: To compare the antibacterial potency of Aqueous Nigella sativa extract, Aqueous Neem leaf extract, and 3% Sodium Hypochlorite. Materials and Methods: Test solutions were tested against E. faecalis (ATCC 29212) to check for their Minimum Inhibitory Concentration (MIC) by double dilution method and Kill time to measure their antimicrobial potency to be used as an intracanal irrigant. Results: The study's limitations show that Aqueous Nigella sativa extract has a better antimicrobial effect than Aqueous Neem leaf extract and 3% Sodium Hypochlorite solution against E. faecalis.

The present study evaluated the antimicrobial property of green synthesized nanosilver particles against dental cariogenic microorganisms such as Streptococcus mutans and Lactobacillus acidophilus. Materials and Methods: An in vitro study had been designed to meet the objectives. Galla Chinensis ellagic acid powder synthesized nanosilver particles (GCAgNPs) synthesized nanosilver particles were used in this study. The cariogenic bacteria S. mutans (ATCC 25175) and L. acidophilus (ATCC 4356) were used in this study. The antimicrobial activity was detected at different concentrations (1000 μg/ml, 500 μg/ml, 250 μg/ml, 125 μg/ml, and 62.5 μg/ml) by means of qualitative and quantitative methods. Results: The results show a statistically significant difference between all the concentration (1000 μg/ml, 500 μg/ml, 250 μg/ml, 125 μg/ml, 62.5 μg/ml) in Galla Chinensis synthesized silver nanoparticles (GCAgNPs) in S. mutans and L. acdiophilus. Intergroup comparison of GCAgNPs shows a statistically significant difference among all the concentrations against S. mutans and L. acidophilus. Conclusion: GCAgNPs show antimicrobial and antibiofilm activity against S. mutans and L. acidophilus microorganisms.

Pulp capping should always be considered as the primary treatment of choice for teeth without irreversible pulpitis in lesions approaching dental pulp. The predictability of vital pulp therapy has improved with the introduction of newer bioceramic materials.The aim of this study was to evaluate the clinical success of Biodentine, calcium hydroxide (CH), and mineral trioxide aggregate (MTA) as pulp capping materials for indirect pulp capping in carious permanent teeth.Indirect pulp capping was done for 36 molars of 36 patients with deep caries lesions. They were randomly divided into three groups: Biodentine group (12 teeth), MTA group (12 teeth), and CH group (12 teeth). Patients were recalled at 1, 3, and 6 months to evaluate the clinical success of the treatment outcome.All statistical analysis was performed using SPSS software version 21.0. Pearson's Chi-square test was used to compare the success and failure rates between Biodentine, MTA, and Ca(OH)In a statistical trial/study, the pulp capping materials gave different success rates, 91.67% success in the Biodentine group, 83.33% success in the MTA group, and 58.33% success in the CH group. The results were not statistically significant.Indirect pulp capping with calcium silicate materials provided better results compared to that of calcium hydroxide.

AIM AND OBJECTIVE To chemically characterize restorative composite resin polymerized with 20 wt.% and 40 wt.% dipentaerythritol penta-/hexaacrylate (DPEPHA) comonomer. Furthermore, this study aimed to evaluate the conversion degree (DC) and glass transition temperature (Tg) of the newly formed copolymer. MATERIALS AND METHODS The trial groups were photo-polymerized with DPEPHA comonomer, whereas the control group was photo-polymerized only with the propriety resin monomers. Infrared (FTIR) and nuclear magnetic resonance (NMR) spectroscopies were used for establishing copolymerization. The characteristics and composition (mass %) of the surface were explained by field-emission scanning electron microscopy (FESEM) and energy-dispersive X-ray (EDX) spectroscopy, respectively. The DC and Tg of the resultant copolymers were evaluated through FTIR and differential scanning calorimetry, respectively. Appropriate statistical tests with corresponding post hoc tests were employed to compare the medians and means of DC and Tg, respectively. RESULTS The formation of a new copolymer P(GEU-Co-DPEPHA) was evident. The DC and Tg of the P(GEU-Co-DPEPHA) copolymer were greater than the control. DPEPHA in the copolymer at 40 wt.% concentration showed the highest DC and Tg. CONCLUSION DPEPHA comonomer addition leads to the formation of a new P(GEU-Co-DPEPHA) copolymer with improved DC and Tg. CLINICAL SIGNIFICANCE The novel P(GEU-Co-DPEPHA) copolymer may improve the physico-mechanical and biological properties of the restorative composite resin. This would improve the quality of restoration and its in vivo serviceability, thereby imparting a good living quality to the entailed population.

It is crucial for discovering new treatments and ensuring that existing treatments are used to the best of their ability. Research can provide answers to questions that are unknown, filling knowledge gaps and changing the way health care professionals practice. Research studies are often conducted to diagnose diseases and health problems efficiently, to reduce the number of people who suffer from illness, and to prevent further development or recurrence of the illness.

Introduction: The purpose of the present study was to evaluate the short-term clinical outcomes of pulp capping using mineral trioxide aggregate (MTA) Angelus (Angelus Londrina, PR, Brazil) or Biodentine (Septodont, Saint Maur des Fosses, France) as capping materials in a prospective randomized controlled study. Subjects and Methods: A total of 50 teeth were confirmed to eligible for pulp capping randomly assigned into either the MTA Angelus or the Biodentine Group (25 teeth per group). Direct\indirect pulp capping was performed using these two materials, and clinical and radiographical evaluations were performed at 0, 3, and 6 months after the treatments. Teeth with no response to pulp vitality test and those exhibiting clinical or radiographic signs and/or symptoms of irreversible pulpitis or pulp necrosis were considered to be failures. Results: Forty-five patients (47 teeth) were examined at the 6-month follow-up (patient recall rate = 92%); 23 of these teeth were in the MTA Angelus group, and 24 were in the Biodentine group. The overall success rate was 93%, and the success rates in the MTA Angelus and Biodentine groups were 95.5% (21/23 teeth) and 96% (23/24 teeth), respectively. Statistical analyses of these success rates did not reveal any significant difference between the groups. Conclusions: In this randomized controlled study, no significant difference in the short-term clinical outcomes of direct\indirect pulp capping using MTA Angelus or Biodentine as the capping material was found.

Aims: This study aims to demonstrate the bone regeneration in the periapical region using platelet-rich fibrin (PRF) and nanocrystalline hydroxyapatite (NcHA) with collagen in combination with PRF and their effects on healing in periapical tissues. Subjects and Methods: Fourteen patients (15 surgical sites) were selected and distributed into three groups: Group I – Replacement with PRF, Group II – Replacement with PRF and NcHA with collagen bone graft (SYBO GRAF – C, Eucare pharmaceuticals ltd., Chennai, Tamil Nadu, India), and Group III – Control group. In all the three groups, patient recall visits were scheduled after 1-, 3-, and 6-month time interval for clinical and radiological examination. Radiovisiography image was analyzed using RADIANT Digital Imaging and Communications in Medicine VIEWER. Results: A significantly higher rate of healing was observed after 6 months in Group II (100%) followed by Group I (92%) and Group III (86%). Conclusion: It was concluded that combination of PRF and nanocrystalline hydroxyapatite with collagen produced a significantly faster bone regeneration. The conventional technique and PRF were less predictable with its healing response.

Background: Restorative dentistry is impossible without composite resins. Toxicity of these resins is common though possessing favorable properties. Modifications of the resin matrix are being executed to improve the properties of the material. Dipentaerythritol penta-/hexa-acrylate (DPEPHA) monomer has been recently identified as a cross-linking comonomer with the conventional resin matrix. Purpose: This study aimed to investigate the histocompatibility of DPEPHA comonomer in photopolymerized resin at 20 and 40 wt.% concentrations in rats. Materials and Methods: Eighteen male Wister rats were subjected to subcutaneous implantation of conventional resin specimens without DPEPHA (G0) and with DPEPHA at 20 wt.% (G20) and 40 wt.% (G40) concentrations. Then, the rats were subdivided according to the time of resection of the implantation sites at 1, 2, and 4 weeks (n = 6 rats). Tissue specimens were submitted to histopathological analysis. Results: Except for 4 weeks' time interval, there were significant differences (P < 0.05) in the inflammation among the groups at each time interval. The order of inflammation was NC

Objectives: The objective of this study is to investigate the efficacy of bioactive glass containing product on remineralization of artificial induced carious enamel lesion and to compare its efficiency with other remineralization products using an in-vitro pH cycling method. The null hypothesis tested was bioactive glass has no effect on enamel remineralization. Materials and Methods: A total of 20 enamel samples of human molar teeth were subjected to artificial caries lesion formation using pH cycling method and was verified using high resolution scanning electron microscope (HRSEM). Each demineralized sample was then divided into five test groups each containing twenty. Group A - Bioactive glass (SHY-NM), Group B - Fluoride tooth paste (Amflor), Group C - CPP-ACP (Tooth mousse), Group D - CPP-ACPF (Tooth mousse plus), Group E - control. All the test groups were exposed to the pH cycling regime, the remineralizing agents were applied for 10 min except control. After 10 days period, the entire test groups were evaluated with HRSEM and quantitative assessment by energy dispersive X-ray spectroscopy. The obtained data was analyzed statistically using one-way ANOVA, Student's t-test and Tukey's multiple comparison tests. P ≤ 0.05 was considered to be significant. Results: Rejection of the null hypothesis and highlights the concept of biomimetic bioactive glass as an effective remineralizing agent. Clinical Relevance: To focus on the importance of minimal invasive treatment on incipient carious lesion by remineralization.

Introduction The aim of this study was to evaluate and compare the antimicrobial activity of calcium hydroxide, triantibiotic mixture (TAM), and an ethanol extract of propolis as intracanal medicaments on Enterococcus faecalis–infected root canals. Methods One hundred twenty extracted intact human permanent incisors were decoronated, and chemomechanical preparation of the root canal was performed. After sterilization of the samples, they were inoculated with pure culture of E. faecalis and incubated. After incubation, colony-forming units were recorded before medication. Then, samples were divided randomly into five groups (n = 24). Each group was then exposed to various intracanal medicaments, namely calcium hydroxide (group 1), TAM (group 2), propolis (group 3), ethanol (group 4), and saline as the control group (group V). The antibacterial effectiveness of the different intracanal medicament was recorded by determining the percentage reduction in colony counts (%RCC) at the end of days 1, 2, and 7. The data were statistically analyzed using one-way analysis of variance and Tukey Honestly Significant Difference (HSD) post hoc test. Results The %RCC was highest for propolis showing 100% reduction on day 2 followed by TAM showing 82.5%, 92.2%, and 98.4% of reduction on days 1, 2, and 3, respectively. Calcium hydroxide showed a gradual increase in antibacterial activity with a maximum of 59.4% on day 7. Conclusions Propolis was more effective than TAM against E. faecalis at a 2-day time period, and both were equally effective at 7 days.

Aims and Objectives: The aim of this study was to evaluate and compare the effect of power bleaching on human enamel microhardness activated by three different light sources, namely the light emitting diode (LED), diode laser, and neodymium: Yttrium-aluminium-garnet (Nd: YAG) laser. Materials and Methods: Fifty enamel samples were randomly divided into five test groups each containing 10 samples (n = 10). The enamel surfaces of samples were polished and baseline Vickers microhardness values were evaluated. Control group samples were untreated and stored in artificial saliva. Experimental groups were subdivided into four groups based on bleaching treatment carried out. Enamel samples were treated with 40% hydrogen peroxide gel for 2 cycles of 20 min per bleach cycle (with and without light activation). Microhardness was reassessed on 1 st , 7 th , and 21 st day after storage in artificial saliva. Results were subjected to statistical analysis using one-way analysis of variance (ANOVA) and t-test. Results: There was a statistically significant difference present in enamel microhardness values compared to the unbleached and bleached group. On comparing 1 st and 7 th day to the baseline values there was a statistically significant difference in all the experimental groups. No statistically significant difference existed between the baseline values and 21 st day. Conclusion: Activation with various light sources (LED/diode laser/Nd: YAG laser) do not have additional deleterious effects on enamel microhardness and thus can be used safely to hasten the bleaching procedure.

Background: Application of laser energy onto exposed dentinal tubules provides instantaneous relief from dentin hypersensitivity. The aim of the present study was to compare between CO 2 and Nd:YAG laser irradiation for their sealing ability on human dentinal tubules when treated with and without fluoride varnish, using Scanning Electron Microscopy. Materials and Methods: Twenty extracted human intact third molar teeth were selected. A total of six samples were obtained from each tooth. One hundred and twenty dentin samples from 20 teeth were then divided into six groups with 20 samples in each group. These slabs were treated with 17% ethylenediaminetetraacetic acid (EDTA) for 5 min and with 5% NaOCl for the next 2 min for removal of the smear layer. Each of the samples from a single tooth was then subjected to six different treatment protocols according to the groups given below. Group 1 (n = 20) : CO 2 laser Group 2 (n = 20) : Nd:YAG laser Group 3 (n = 20) : Varnished with fluoride and lased with CO 2 laser Group 4 (n = 20) : Varnished with fluoride and lased with Nd:YAG laser Group 5 (n = 20) : Varnished with fluoride Group 6 (n = 20) (control) : Not subjected to lasing and fluoride application All samples were examined under scanning electron microscope. Results were subjected to statistical analysis using one-way analysis of variance (ANOVA) and the multiple comparisons among groups were carried out by Student's t-test. Results: Compared to conventional fluoridation or laser irradiation, in the treatment of dentin hypersensitivity, CO 2 lasers in combination with fluoride varnish appear to show better efficacy than either treatment modality alone.
