ABSTRACT: Cow milk, although rich in essential nutrients, is a well-known food allergen that can cause allergic reactions in infants and young children. β-Lactoglobulin accounts for 10% of the total protein in milk and 50% of the whey protein, which has high nutritional value and excellent functional properties but is also the main allergen leading to milk protein allergy. Exploring the mechanism of milk allergy and selecting suitable separation and purification methods to obtain high-purity β-LG is the premise of research on reducing allergenicity. In this review, the research progress in membrane technology, gel filtration chromatography, ion exchange chromatography, affinity chromatography, precipitation, and aqueous 2-phase system separation for the separation and purification of milk β-LG is reviewed in detail to promote the further development of milk β-LG separation and purification methods and provide a new method for the development of hypoallergenic dairy products in the future. Among these methods, ion exchange chromatography and gel chromatography are widely used, precipitation is generally used as a crude purification step, and HPLC and membrane technology are used for further purification to improve the purity of allergens.

Abstract Background Heat stress (HS) can affect the physiology and metabolism of animals. HS-induced intestinal inflammation in pigs is a common disease, causing severe diarrhea, that can result in substantial economic losses to the pig industry, but the molecular mechanisms and pathogenicity of this disease are not fully understood. The objective of this study was to identify the differentially expressed genes (DEGs) and long noncoding RNAs (DELs) related to inflammation in the colon tissues of pigs under constant (1, 7, and 14 days) HS. Results LncRNA and targeted gene interaction networks were constructed. GO annotation and KEGG pathway analyses were subsequently performed to determine the functions of the DEGs and DELs. The results revealed 57, 212, and 54 DEGs and 87, 79, and 55 DELs in the CON/H01, CON/H07, and CON/H14 groups, respectively. KRT85, CLDN1, S100A12, TM7SF2, CCN1, NR4A1, and several lncRNAs may be involved in regulating the development of intestinal inflammation. GO analysis indicated that the DEGs and DELs were enriched in a series of biological processes involved in the innate immune response, RAGE receptor binding, and positive regulation of the ERK1 and ERK2 cascades. KEGG pathways related to inflammation, such as the tight junction (TJ) and MAPK signaling pathways, were enriched in DEGs and DELs. Conclusions This study have expanded the knowledge about colon inflammation-related genes and lncRNA biology in pigs under HS; analyzed the the lncRNA‒mRNA interaction for HS-induced intestinal inflammation. These results may provide some references for our understanding of the molecular mechanism of the intestinal response to HS in pig.

Abstract In this work, the rapid and efficient preparation of isolated galloyl glucoside tautomer free radical inhibitors was investigated using Saxifraga tangutica as a raw material. Four highly polar galloyl glucoside tautomers, 3-O-galloyl-α-d-glucose ⇌ 3-O-galloyl-β-d-glucose (Fr2-1-1), 2-O-galloyl-α-d-glucose ⇌ 2-O-galloyl-β-d-glucose (Fr2-1-2/2-1-3), 1-O-galloyl-β-d-glucose (Fr2-2-1), and 6-O-galloyl-α-d-glucose ⇌ 6-O-galloyl-β-d-glucose (Fr2-3-1/Fr2-3-2), were obtained via two-step medium-pressure liquid chromatography (with solid loading instead of conventional liquid injection) and one-step high-performance chromatography coupled with on-line RPLC-DPPH techniques for targeted isolation. This separation integration technique not only increases sample intake and reduces time cost but also visualizes each step of targeted separation. All four compounds were isolated from the plant for the first time. In vitro antioxidant activity assays by DPPH (1,1‑diphenyl-2-picrylhydrazyl) revealed that Fr2-1-2/Fr2-1-3 (IC50: 5.52 ± 0.32 μM), Fr2-2-1 (IC50: 7.22 ± 0.57 μM), and Fr2-3–1/Fr2-3-2 (IC50: 7.36 ± 0.25 μM) had superior free radical scavenging abilities and that both were superior to that of quercetin (IC50: 18.61 ± 3.55 μM). Abstract The process of removing sulfur compounds and aromatic compounds to produce clean fuel is an important and effective contribution to the processes of mitigating and adapting to climate change. In contrast, it is necessary to find an innovative way to remove sulfur and carcinogenic aromatic compounds because clean, low-sulfur diesel is commonly used in all countries of the world at the present time. Therefore, in this work, we have studied the effect of the microwave radiation power and the irradiation time with the use of more than one type of organic solvent; methanol, acetonitrile and ethyl acetoacetate; as an extractant and solvent to feed ratio impact on the removal of sulfur and aromatic compounds of a real diesel fuel feed which has 450 ppm sulfur content and 16 wt% aromatic Content. The results showed that the best solvent used during this work was ethyl acetoacetate. According to the results, high sulfur removal (≈ 92%) was accomplished with microwave-assisted extractive desulfurization technique under the following ideal conditions: the irradiation time is 7 min, the solvent feed ratio is 3:1 and the microwave intensity is 180 W. To reveal the mechanism of microwave-assisted extractive desulfurization via different organic solvents, a theoretical study including structural examination and interaction energy analysis on the interaction between dibenzothiophene (DBT) or dimethyl dibenzothiophene (DMDBT) and the different organic solvents was also conducted.

Abstract In three-dimensional free-bending forming (3D-FBF), the tube is not overly constrained, and the plastic deformation behavior and forming quality of the bent tube are significantly affected by the critical structure of the forming die lining. However, the effects of die-lining structural parameters on the tube quality, and a method to determine the combination of die-lining structural parameters is yet to be devised. This study aims to propose a new framework that allows one to understand the effects of various die-lining structural parameters on tube quality and to propose the best combination of die-lining structural parameters. First, finite-element modeling is performed to simulate 3D-FBF and examine the effects of individual die-lining structures on the quality of tube formation. The simulation results show that the deformation-zone length and die gap are positively correlated with the tube-section distortion and wall-thickness variation, whereas it shows an opposite trend with respect to the bending radius. Additionally, the lining chamfer radius of the bending die and the guide lining chamfer radius minimally affect the tube forming quality. Subsequently, the optimal die-lining structure is obtained using the response-surface method. The tube cross-sectional distortion rate reduced from 2.73 to 2.53% after the die lining is optimized. Additionally, the average inner-wall thickness reduced to 1.06 mm, whereas the average outer-wall thickness increased to 0.97 mm. This paper proposes a method for optimizing the forming-die-lining mechanism and for improving the tube forming quality in 3D-FBF.

Introduction: Previous research has shown that an aqueous extract of Humulus japonicus (EH) can ameliorate hypertension, nonalcoholic fatty liver disease, and oxidative stress in adipocytes by activating the thermogenic pathway. However, the effects of an ethanol (30%) extract of EH on obesity are unknown. Methods: Various protein expression levels in fully differentiated 3T3-L1 adipocytes were assessed by Western blotting. Lipid deposition in 3T3-L1 adipocytes was examined by oil red O staining. The MTT assay was used to evaluate adipocyte viability. Caspase 3 activity and glycerol release were determined using commercial assay kits. Results: In this study, we discovered that EH treatment inhibited lipogenesis and promoted lipolysis in both differentiated 3T3-L1 adipocytes and adipose tissue of mice fed a high-fat diet. EH treatment also increased phosphorylated protein kinase A (PKA) levels while reducing p38 phosphorylation. When H89, a PKA inhibitor, was used, the effects of EH on lipogenic lipid accumulation and lipolysis in 3T3-L1 adipocytes were eliminated. Treatment with luteolin 7-O-β-d-glucoside (LU), the major active compound in EH, also suppressed lipid deposition and p38 phosphorylation but enhanced lipolysis in 3T3-L1 adipocytes. These changes were abrogated by H89. Conclusion: These findings indicate that EH containing LU reduces lipogenesis and stimulates lipolysis via the PKA/p38 signaling pathway, leading to an improvement in obesity in mice. Therefore, our study suggested that EH could be a promising therapeutic agent for treating obesity.

Cancer is a major global health challenge, and current treatment modalities, such as surgery, chemotherapy, radiation therapy, immunotherapy, and targeted therapies, are often associated with various adverse effects. Dietary strategies have emerged as potential avenues for attenuating cancer growth, progression, and metastasis in numerous solid tumor models. Early clinical studies suggest that dietary interventions hold promise for improving cancer outcomes. This review examines potential dietary interventions that target specific aspects of tumor metabolism. It first examines research on carbohydrates, proteins, lipids, and other interventions, such as vitamins and nutritional supplements, in the context of cancer treatment or management. In addition, it outlines recent developments in cancer interventions involving different dietary patterns and specific dietary restrictions on the basis of preclinical investigations and clinical trials. Finally, this review provides insights into the future prospects of dietary interventions in cancer management.

Abstract TC21 alloy is a high-strength titanium alloy that has been gaining attention in various industries for its excellent combination of strength, toughness, and corrosion resistance. Given that this alloy is hard to cut material, therefore this study aims to optimize the process parameters of Turing this alloy under different conditions (i.e. as-received alloy, and heat-treated alloy). The L9 Taguchi approach-base orthogonal array is used to determine the optimum cutting parameters and the least number of experimental trials required. The achievement of this target, three different cutting parameters are used in the experimental work; each cutting parameter has three levels. The cutting speeds are chosen as 120, 100, and 80 m/min. The feed rates’ values are 0.15, 0.1, and 0.05, mm/rev, and the depth of cut values are 0.6, 0.4, and 0.2 mm. After applying three steps of heat treatment (First step: is heating the sample to 920 °C for 1 h then decreasing to 820 °C also for 1 h, second step: cooling the sample to room temperature by water quenching (WQ), the third step: holding the sample at 600 °C for 4 h (Aging process)). The results revealed that the triple heat treatment led to the change in the microstructure from (α + β) to (α + β) with secondary α platelets (αs) formed in residual β matrix leading to a decreased surface roughness by 56.25% and tool wear by 24.18%. The two most critical factors that affect the tool insert wear and surface roughness are the death of cut and cutting speed, which contribute 46.6% and 46.7% of the total, respectively. Feed rate, on the other hand, has the least importance, contributing 20.2% and 31.9% respectively.

Abstract Neuroendocrine neoplasms (NENs) are highly heterogeneous and potentially malignant tumors arising from secretory cells of the neuroendocrine system. Gastroenteropancreatic neuroendocrine neoplasms (GEP-NENs) are the most common subtype of NENs. Historically, GEP-NENs have been regarded as infrequent and slow-growing malignancies; however, recent data have demonstrated that the worldwide prevalence and incidence of GEP-NENs have increased exponentially over the last three decades. In addition, an increasing number of studies have proven that GEP-NENs result in a limited life expectancy. These findings suggested that the natural biology of GEP-NENs is more aggressive than commonly assumed. Therefore, there is an urgent need for advanced researches focusing on the diagnosis and management of patients with GEP-NENs. In this review, we have summarized the limitations and recent advancements in our comprehension of the epidemiology, clinical presentations, pathology, molecular biology, diagnosis, and treatment of GEP-NETs to identify factors contributing to delays in diagnosis and timely treatment of these patients.

Free bending forming (FBF) technology is widely used for manufacturing complex-shaped Ti pipes without changing the forming die. Nevertheless, the characteristics of Ti alloys, such as low ductility and high yield stress, pose challenges in achieving high-quality components at room temperature. Therefore, the hot FBF technology is used to manufacture and optimize the process parameters to produce complex-shaped TA18 pipes. Combining theoretical analysis, FE modeling, and experimentation, the forming characteristics of TA18 alloy pipes at elevated temperatures were revealed, and the process parameters were optimized. The Johnson-Cook constitutive model was established in this study and coupled with FE modeling providing accurate simulations of the hot FBF process under various conditions. The FE model of the TA18 alloy pipe was verified by comparing its results with the actual TA18 pipe trials. The hot FBF process can improve the wall thickening and cross-sectional distortion of TA18 bending components and reduce the bending radius of the pipe by weakening the dynamic rebound at the end of the bending and forming of titanium pipes. The increase in heating length is also generally conducive for forming TA18 pipe, and the component's external wall thickness reduction and cross-sectional distortion can be inhibited when the heating length is 250 mm. Moreover, the increase in axial propulsion speed improves the thickening of the inner wall thickness of the TA18 bending components but also increases the thinning of the wall thickness and the distortion of the cross-section. Finally, the process parameters were optimized, and the forming quality of the complex-shape TA18 pipe can be improved. The microstructure of the TA18 pipe hardly changes during the hot FBF process.

Developing high strain rate (HSR) forming technologies is significant for efficient and high-quality aerospace sheet metal parts manufacturing. Research on the deformation behaviors and micromechanism of materials under HSRs will contribute to the further application of HSR forming technologies. In this study, the mechanical behavior of AA1060 pure aluminum at low strain rates (LSRs, 0.001 s−1 and 0.1 s−1) and high strain rates (HSRs, 2400 s−1 and 3300 s−1) was tested utilizing quasistatic tensile tests and Hopkinson bar tests at room temperature. The results of the mechanical properties showed that the elongations were 30.01 %, 37.91 %, and 46.61 %, corresponding to strain rates of 0.1 s−1, 2400 s−1, and 3300 s−1, respectively, which were 29.86 % at a strain rate of 0.001 s−1. The maximum amplification of AA1060 elongation reached 56.10 % at the HSR. Microstructure characteristics, including the Schmid factor distribution, geometrically necessary dislocation (GND) distribution, slip band, and dislocation cell, were observed by electron backscattering diffraction (EBSD) and transmission electron microscopy (TEM) under the condition of the same elongation with strain rates of 0.001 s−1 and 3300 s−1. In addition, the reasons for the plastic improvements during HSR loading were also investigated. The hard-oriented grains participate in plastic deformation at HSRs, providing additional space for dislocation multiplication and movement, increased dislocation density, and interaction between dislocations. In addition, the deformed hard-oriented grains also coordinated the strain gradient with soft-oriented grains. This improved the overall deformation uniformity, thus avoiding the premature occurrence of strain localization, namely, the elongation increase.

Abstract Background Age-related macular degeneration (AMD) is an irreversible eye disease that can cause blurred vision. Regular exercise has been suggested as a therapeutic strategy for treating AMD, but how exercise improves AMD is not yet understood. This study investigated the protective effects of developmental endothelial locus-1 (DEL-1), a myokine upregulated during exercise, on endoplasmic reticulum (ER) stress-induced injury in retinal pigment epithelial cells. Methods We evaluated the levels of AMPK phosphorylation, autophagy markers, and ER stress markers in DEL-1-treated human retinal pigment epithelial cells (hRPE) using Western blotting. We also performed cell viability, caspase 3 activity assays, and autophagosome staining. Results Our findings showed that treatment with recombinant DEL-1 dose-dependently reduced the impairment of cell viability and caspase 3 activity in tunicamycin-treated hRPE cells. DEL-1 treatment also alleviated tunicamycin-induced ER stress markers and VEGF expression. Moreover, AMPK phosphorylation and autophagy markers were increased in hRPE cells in the presence of DEL-1. However, the effects of DEL-1 on ER stress, VEGF expression, and apoptosis in tunicamycin-treated hRPE cells were reduced by AMPK siRNA or 3-methyladenine (3-MA), an autophagy inhibitor. Conclusions Our study suggests that DEL-1, a myokine, may have potential as a treatment strategy for AMD by attenuating ER stress-induced injury in retinal pigment epithelial cells.
