Reproductive phasiRNAs (phased, small interfering RNAs) are broadly present in angiosperms and play crucial roles in sustaining male fertility. While the premeiotic 21-nt (nucleotides) phasiRNAs and meiotic 24-nt phasiRNA pathways have been extensively studied in maize (Zea mays) and rice (Oryza sativa), a third putative category of reproductive phasiRNAs-named premeiotic 24-nt phasiRNAs-have recently been reported in barley (Hordeum vulgare) and wheat (Triticum aestivum). To determine whether premeiotic 24-nt phasiRNAs are also present in maize and related species and begin to characterize their biogenesis and function, we performed a comparative transcriptome and degradome analysis of premeiotic and meiotic anthers from five maize inbred lines and three teosinte species/subspecies. Our data indicate that a substantial subset of the 24-nt phasiRNA loci in maize and teosinte are already highly expressed at the premeiotic phase. The premeiotic 24-nt phasiRNAs are similar to meiotic 24-nt phasiRNAs in genomic origin and dependence on DCL5 (Dicer-like 5) for biogenesis, however, premeiotic 24-nt phasiRNAs are unique in that they are likely i) not triggered by microRNAs, ii) not loaded by AGO18 proteins, and iii) not capable of mediating PHAS precursor cleavage. In addition, we also observed a group of premeiotic 24-nt phasiRNAs in rice using previously published data. Together, our results indicate that the premeiotic 24-nt phasiRNAs constitute a unique class of reproductive phasiRNAs and are present more broadly in the grass family (Poaceae) than previously known.

Model species continue to underpin groundbreaking plant science research. At the same time, the phylogenetic resolution of the land plant Tree of Life continues to improve. The intersection of these two research paths creates a unique opportunity to further extend the usefulness of model species across larger taxonomic groups. Here we promote the utility of the Arabidopsis thaliana model species, especially the ability to connect its genetic and functional resources, to species across the entire Brassicales order. We focus on the utility of using genomics and phylogenomics to bridge the evolution and diversification of several traits across the Brassicales to the resources in Arabidopsis, thereby extending scope from a model species by establishing a model clade. These Brassicales-wide traits are discussed in the context of both the model species Arabidopsis thaliana and the family Brassicaceae. We promote the utility of such a model clade and make suggestions for building global networks to support future studies in the model order Brassicales.

Abstract The Knight-Alzheimer Disease Research Center (Knight-ADRC) at Washington University in St. Louis has pioneered and led worldwide seminal studies that have expanded our clinical, social, pathological, and molecular understanding of Alzheimer Disease. Over more than 40 years, research volunteers have been recruited to participate in cognitive, neuropsychologic, imaging, fluid biomarkers, genomic and multi-omic studies. Tissue and longitudinal data collected to foster, facilitate, and support research on dementia and aging. The Genetics and high throughput -omics core (GHTO) have collected of more than 26,000 biological samples from 6,625 Knight-ADRC participants. Samples available include longitudinal DNA, RNA, non-fasted plasma, cerebrospinal fluid pellets, and peripheral blood mononuclear cells. The GHTO has performed deep molecular profiling (genomic, transcriptomic, epigenomic, proteomic, and metabolomic) from large number of brain (n = 2,117), CSF (n = 2,012) and blood/plasma (n = 8,265) samples with the goal of identifying novel risk and protective variants, identify novel molecular biomarkers and causal and druggable targets. Overall, the resources available at GHTO support the increase of our understanding of Alzheimer Disease.

Genome sizes of plants have long piqued the interest of researchers due to the vast differences among organisms. However, the mechanisms that drive size differences have yet to be fully understood. Two important contributing factors to genome size are expansions of repetitive elements, such as transposable elements (TEs), and whole-genome duplications (WGD). Although studies have found correlations between genome size and both TE abundance and polyploidy, these studies typically test for these patterns within a genus or species. The plant order Brassicales provides an excellent system to further test if genome size evolution patterns are consistent across larger time scales, as there are numerous WGDs. This order is also home to one of the smallest plant genomes, Arabidopsis thaliana-chosen as the model plant system for this reason-as well as to species with very large genomes. With new methods that allow for TE characterization from low-coverage genome shotgun data and 71 taxa across the Brassicales, we confirm the correlation between genome size and TE content, however, we are unable to reconstruct phylogenetic relationships and do not detect any shift in TE abundance associated with WGD.

Summary: There is a need for affordable, scalable, and specific blood-based biomarkers for Alzheimer’s disease that can be applied to a population level. We have developed and validated disease-specific cell-free transcriptomic blood-based biomarkers composed by a scalable number of transcripts that capture AD pathobiology even in the presymptomatic stages of the disease. Accuracies are in the range of the current CSF and plasma biomarkers, and specificities are high against other neurodegenerative diseases.

Scorpions are predatory arachnids whose venomous sting primarily affects people in tropical and subtropical regions. Most scorpion stings can only cause localized pain without severe envenomation. Less than one-third of the stings cause systemic envenoming and possibly lead to death. About 350,000 scorpion stings in Northern Africa are recorded yearly, resulting in about 810 deaths. In Eastern/Southern Africa, there are about 79,000 stings recorded yearly, resulting in 245 deaths. Farmers and those living in poverty-stricken areas are among the most vulnerable to getting stung by scorpions. However, compared to adults, children are at greater risk of severe envenomation. Scorpion venom is made up of complex mixtures dominated by peptides and proteins that confer its potency and toxicity. These venom toxins have intra- and interspecies variations associated with the scorpion's habitat, sex, diet, and age. These variations alter the activity of antivenoms used to treat scorpion sting envenomation. Thus, the study of the proteome composition of medically important scorpion venoms needs to be scaled up along their geographical distribution and contributions to envenomation in Southern and Northern Africa. This will help the production of safer, more effective, and broad-spectrum antivenoms within these regions. Here, we review the clinical implications of scorpion sting envenomation in Southern and Northern Africa. We further highlight the compositions of scorpion venoms and tools used in scorpion venomics. We discuss current antivenoms used against scorpion sting envenomation and suggestions for future production of better antivenoms or alternatives. The main aim is to compare oncological outcomes and patterns of recurrence of patients with early-stage endometrioid endometrial cancer according to lymphovascular space invasion (LVSI) status. The secondary objective is to determine preoperative predictors of LVSI. We performed a multicenter retrospective cohort study. A total of 3546 women diagnosed with postoperative early-stage (FIGO I-II, 2009) endometrioid endometrial cancer were included. Co-primary endpoints were disease-free survival (DFS), overall survival (OS), and pattern of recurrence. Cox proportional hazard models were used for time-to-event analysis. Univariate and multivariate logistical regression models were employed. Positive LVSI was identified in 528 patients (14.6%) and was an independent prognostic factor for DFS (HR 1.8), OS (HR 2.1) and distant recurrences (HR 2.37). Distant recurrences were more frequent in patients with positive LVSI (78.2% vs. 61.3%, p < 0.01). Deep myometrial invasion (OR 3.04), high-grade tumors (OR 2.54), cervical stroma invasion (OR 2.01), and tumor diameter ≥ 2 cm (OR 2.03) were independent predictors of LVSI. In conclusion, in these patients, LVSI is an independent risk factor for shorter DFS and OS, and distant recurrence, but not for local recurrence. Deep myometrial invasion, cervical stroma invasion, high-grade tumors, and a tumor diameter ≥ 2 cm are independent predictors of LVSI.

Objective: Intrauterine device fracture, as we know it today, is an infrequent event, usually described as isolated cases. The purpose of this study was to look for factors influencing intrauterine fragment retention after device rupture. Study design: Retrospective cohort study. A total of 135 patients were recruited, and the cohort follow-up ran for three full years from 2018 to 2020. Results: Thirty-three percent of patients had a retained intrauterine fragment compared to 82 of 123 (66.7%) who had expelled it spontaneously. In the group of patients who had at least one intercurrent period between device fracture and confirmatory fragment persistent test, we found persistence of intrauterine fragment in 18 of 71 (25.4%) patients compared to 53 of 71 (74.6%) who did not (p = 0.047). A total of 6 of 39 (15.4%) of the patients with spontaneous rupture of the device presented with persistence of the intrauterine fragment compared to 32 of 81 (39.2%) of the group with fracture after manipulation (p = 0.006). The mean time elapsed from the fracture to the confirmatory test in the patients who had persistence of the fragment was 26.97 days (range from 0 to 116), while in those who expelled it spontaneously, a mean of 45.59 days (range 7–267) had elapsed (p = 0.003). Conclusions: The main factors positively influencing complete expulsion of the fragmented IUD were elapsed time of 45 days or more, intercurrent menstruation or spontaneous fracture. Therefore, the proposed protocol calls for expectant management for at least 1.5 months after fracture, allowing at least one intercurrent period to elapse prior to any therapeutic manoeuvre.

Parasites and environmental features could synergistically act as stressors to the health of their hosts. The objectives of this study were to evaluate the effect of: (i) water quality, host sex, size and body condition on adult digenean parasite infections; (ii) digenean infections and host sex and size on the oxidative stress biomarkers and body condition of hosts; and (iii) water quality on the oxidative stress biomarkers and body condition in Clarias gariepinus. Water quality variables were measured and C. gariepinus were collected each month for a year for examination of two intestinal digeneans, Masenia nkomatiensis and Glossidium pedatum, and determination of body condition and measurement of biomarkers in the host. The results indicated that the intensity of M. nkomatiensis was positively correlated with electrical conductivity and total dissolved solids. Prevalence of G. pedatum was negatively correlated with electrical conductivity, salinity and total dissolved solids. High summer water temperature was strongly associated with high digenean infections. There was no host body condition, sex or size bias for any of the parasite infection variables. Differences in the biomarker levels and body condition between uninfected fish and those infected with M. nkomatiensis or G. pedatum were insignificant indicating a low effect of the digenean parasites on oxidative stress biomarkers and body condition in the fish. However, total protein levels were positively associated with host size, and lipid peroxidation was negatively related to host body condition; total protein levels were also positively correlated with temperature and negatively correlated with dissolved oxygen. Host body condition was only negatively correlated with dissolved oxygen. Overall the trends observed in the data showed that the parasites have a negligible effect on oxidative stress in host fish and the trends observed for all variables (water quality, stress biomarkers, body condition and parasite infections) showed a strong seasonal pattern.

Stable isotope analysis offers a unique tool for comparing trophic interactions and food web architecture in ecosystems based on analysis of stable isotope ratios of carbon (13C/12C) and nitrogen (15N/14N) in organisms. Clarias gariepinus were collected from six sites along the Vaal River, South Africa and were assessed for ectoparasites and endoparasites. Lamproglena clariae (Copepoda), Tetracampos ciliotheca and Proteocephalus glanduligerus (Cestoda), and larval Contracaecum sp. (Nematoda) were collected from the gills, intestine and mesenteries, respectively. Signatures of δ13C and δ15N were analysed in host muscle tissue and parasites using bulk stable isotope analysis. Variable stable isotope enrichment between parasites and host were observed; L. clariae and the host shared similar δ15N signatures and endoparasites being depleted in δ13C and δ15N relative to the host. Differences in stable isotope enrichment between parasites could be related to the feeding strategy of each parasite species collected. Geographic and spatial differences in enrichment of stable isotopes observed in hosts were mirrored by parasites. As parasites rely on a single host for meeting their nutritional demands, stable isotope variability in parasites relates to the dietary differences of host organisms and therefore variations in baseline stable isotope signatures of food items consumed by hosts.

Monogenea is a Class of ecologically and economically significant platyhelminth parasites of freshwater and marine hosts. Contamination of the environment has been shown to variably affect parasites resulting in alterations at the population and organismal levels. The aim of this study was to perform a meta-analysis of published data to assess similarities among responses of monogeneans to pollution and estimate effect sizes of contaminant exposure on these parasites. Effect size was calculated according to Cohen’s d based on reported mean abundance for monogeneans from individual studies. A total of 57 studies were used in the present study. Each Monogenea–contaminant record represented a single data point in the meta-analysis, and this accounted for a total of 217 useable records. Most studies were concerned with species belong to Gyrodactylidae, Diplozoidae, Ancyrocephalidae and Dactylogyridae. Assessment of the effects of specific pollutants on monogeneans have mostly been restricted to exposure to eutrophication, pulp-mill effluent and metals. Comparison of effect sizes showed strong negative effects for metals and strong positive effects for eutrophication on monogeneans. Other contaminant types had weak effect sizes on monogeneans. Comparison of effect sizes among monogenean families showed that none of the effect sizes calculated were significant but metals and disturbance contaminants showed strong negative and positive effects respectively for Gyrodactylidae. The current comparisons were based on changes in the population structure and infections of monogeneans, however, organ level effects associated with exposure to poor water quality have also been noted. In this instance, exposure has been suggested to result in asymmetry of the attachment organs of some monogeneans. Despite numerous records of impacts of pollution exposure at the population level, understanding of the physiological effects and mechanisms employed by Monogenea to survive and adapt to changes in the environment are yet to be explored. Study of these aspects would enable better understanding of the variation in the trends observed at a population level for monogeneans in relation to the quality of the macroenvironment.

Snakebite envenoming is a neglected tropical disease (NTD) that results from the injection of snake venom of a venomous snake into animals and humans. In Africa (mainly in sub-Saharan Africa), over 100,000 envenomings and over 10,000 deaths per annum from snakebite have been reported. Difficulties in snakebite prevention and antivenom treatment are believed to result from a lack of epidemiological data and underestimated figures on snakebite envenoming-related morbidity and mortality. There are species- and genus-specific variations associated with snake venoms in Africa and across the globe. These variations contribute massively to diverse differences in venom toxicity and pathogenicity that can undermine the efficacy of adopted antivenom therapies used in the treatment of snakebite envenoming. There is a need to profile all snake venom proteins of medically important venomous snakes endemic to Africa. This is anticipated to help in the development of safer and more effective antivenoms for the treatment of snakebite envenoming within the continent. In this review, the proteomes of 34 snake venoms from the most medically important snakes in Africa, namely the Viperidae and Elipdae, were extracted from the literature. The toxin families were grouped into dominant, secondary, minor, and others based on the abundance of the protein families in the venom proteomes. The Viperidae venom proteome was dominated by snake venom metalloproteinases (SVMPs–41%), snake venom serine proteases (SVSPs–16%), and phospholipase A2 (PLA2–17%) protein families, while three-finger toxins (3FTxs–66%) and PLA2s (16%) dominated those of the Elapidae. We further review the neutralisation of these snake venoms by selected antivenoms widely used within the African continent. The profiling of African snake venom proteomes will aid in the development of effective antivenom against snakebite envenoming and, additionally, could possibly reveal therapeutic applications of snake venom proteins.

The upper reaches of the Crocodile River (West) system are located in the western basin of the Witwatersrand mountain chain, an area heavily influenced by gold mining. After the spill of highly acidic and contaminated mining-influenced water into the river system in 2002, chemical water treatment was implemented to compensate for the consequences of acid mine drainage. Some studies thereafter have shown metal concentrations of concern in the system, but never considered longer periods of time. For this study, we investigated water and sediments over a period of 13 months from 7 sampling sites in the system. In addition to physico-chemical parameters such as temperature, pH, and electrical conductivity, a multi-element analysis was conducted. We used various versions of atomic-absorption-spectroscopy, total reflection x-ray fluorescence spectroscopy, and inductively coupled plasma mass spectrometry in filtered and acidified water samples as well as sediment leachates. Concentrations of Ni, Zn, As, Pb, and U in the sediment were clearly elevated at the site closest to the mine as well as further downstream, some of them far exceeding quality guidelines. Moreover, dissolved Mn, Fe, Ni, Zn and U occurred irregularly in concentrations of concern at the site of mining-influenced water inflow. Our findings clearly indicate a risk of further and chronic mobilization of toxic elements from this site and a possible threat to the connected river system.

Knowledge and skills for global health program design, implementation and monitoring is an expectation for practicing public health professionals. Major health education professional organizations including American Association for Health Education (AAHE), Society of Public Health Education (SOPHE) and International Union for Health Promotion and Education (IUHPE) participated in the 2008 Galway Conference to develop standards and quality assurance systems for enhancing workforce capacity in global health education and promotion. To help promote the health of all people, eight Millennium Development Goals (MDGs) were ratified by United Nations (UN) Member States to be achieved by 2015. Heads of state met at the General Assembly and the Secretary General reported worldwide progress concerning the achievement of MDGs during the summer of 2010. This article presents implications of MDGs for public health professionals including the topics of child and maternal mortality and women's reproductive health, HIV/AIDS, malaria and other major diseases, and environmental sustainability. Specifically, this article also discusses how health education specialists can contribute to the achievement of MDGs 3, 4, 5, 6, 7 and 8, due to their unique professional training and demonstrated competencies.
