We present an analysis of two seasons of archival, multi-frequency VLA monitoring of the quad lens system JVAS B1422+231, the 15-GHz data of which have previously been published. The 8.4- and 15-GHz variability curves show significant variability, especially in polarization, but lack features on short time-scales that would be necessary for an accurate measurement of the very short predicted time delays ($\le$1 d) between the three bright images. Time delays can only realistically be measured to the very faint image D and for the first time we detect its long-term variability and determine its polarization properties. However, image-dependent (extrinsic) variability (including variations on time-scales of hours) is present in multiple images and the magnitude of this is largest in image D at 15 GHz ($\pm$10 per cent). As the variations appear to increase in amplitude with frequency, we suggest that the most likely cause is microlensing by compact objects in the lensing galaxy. Combining the monitoring data allows us to detect a faint arc of emission lying between images B and C and the jets responsible for this are imaged using archival VLBA data. Finally, we have also measured the rotation measure of the three bright images and detected the polarization properties of image D., 12 pages, 9 figures, accepted for publication in MNRAS

Wide, deep, blind continuum surveys at submillimetre/millimetre (submm/mm) wavelengths are required to provide a full inventory of the dusty, distant Universe. However, conducting such surveys to the necessary depth, with sub-arcsec angular resolution, is prohibitively time-consuming, even for the most advanced submm/mm telescopes. Here, we report the most recent results from the ALMACAL project, which exploits the 'free' calibration data from the Atacama Large Millimetre/submillimetre Array (ALMA) to map the lines of sight towards and beyond the ALMA calibrators. ALMACAL has now covered 1,001 calibrators, with a total sky coverage around 0.3 deg2, distributed across the sky accessible from the Atacama desert, and has accumulated more than 1,000h of integration. The depth reached by combining multiple visits to each field makes ALMACAL capable of searching for faint, dusty, star-forming galaxies (DSFGs), with detections at multiple frequencies to constrain the emission mechanism. Based on the most up-to-date ALMACAL database, we report the detection of 186 DSFGs with flux densities down to S870um ~ 0.2mJy, comparable with existing ALMA large surveys but less susceptible to cosmic variance. We report the number counts at five wavelengths between 870um and 3mm, in ALMA bands 3, 4, 5, 6 and 7, providing a benchmark for models of galaxy formation and evolution. By integrating the observed number counts and the best-fitting functions, we also present the resolved fraction of the cosmic infrared background (CIB) and the CIB spectral shape. For high-resolution imaging implementations, the spacecraft attitude tracking control accuracy is crucial to determining the imaging quality. This investigation addresses the attitude tracking issue of imaging spacecraft subject to system uncertainties (unavailable inertia tensor, unexpected disturbances, and actuator faults). An adaptive sliding mode control (SMC) strategy is proposed to guarantee practical fixed-time closed-loop stability even in the presence of system uncertainties. Unlike existing methodologies, the sliding mode surface is developed to satisfy a novel sufficient condition of fixed-time stability. The sliding manifold design also circumvents the unwinding phenomenon arising in quaternion representations. Particularly, this controller is developed to generate a smooth control profile by using a new parameter update law. Rigorous Lyapunov analyses are further employed to ensure the fixed-time closed-loop stability irrespective of the system initial states. Finally, numerical examples are performed to demonstrate the feasibility and highlight the inherent features of the derived control law.

In this paper, we bound the number of solutions to a general Vinogradov system of equations $x_1^j+\dots+x_s^j=y_1^j+\dots+y_s^j$, $(1\leq j\leq k)$, as well as other related systems, in which the variables are required to satisfy digital restrictions in a given base. Specifically, our sets of permitted digits have the property that there are few representations of a natural number as sums of elements of the digit set---the set of squares serving as a key example. We obtain better bounds using this additive structure than could be deduced purely from the size of the set of variables. In particular, when the digits are required to be squares, we obtain diagonal behaviour with $2k(k+1)$ variables.

Purpose:Metaplastic breast cancer (MpBC) is a rare aggressive subtype that responds poorly to cytotoxics. Median survival is approximately 8 months for metastatic disease. We report results for advanced MpBC treated with ipilimumab + nivolumab, a cohort of S1609 for rare cancers (DART: NCT02834013).Patients and Methods:Prospective, open-label, multicenter phase II (two-stage) trial of ipilimumab (1 mg/kg i.v. every 6 weeks) plus nivolumab (240 mg i.v. every 2 weeks) for advanced MpBC. Primary endpoint was objective response rate (ORR). Secondary endpoints included progression-free survival (PFS), overall survival (OS), and toxicity.Results:Overall, 17 evaluable patients enrolled. Median age was 60 years (26–85); median number of prior therapy lines was 2 (0–5). ORR was 18%; 3 of 17 patients achieved objective responses (1 complete, 2 partial responses; 2 spindle cell, 1 chondromyxoid histology), which are ongoing at 28+, 33+, and 34+ months, respectively. Median PFS and OS were 2 and 12 months, respectively. Altogether, 11 patients (65%) experienced adverse events (AE), including one grade 5 AE. Eight patients (47%) developed an immune-related AE (irAE), with adrenal insufficiency observed in all 3 responders. Responses occurred in tumors with low tumor mutational burden, low PD-L1, and absent tumor-infiltrating lymphocytes.Conclusions:The ipilimumab and nivolumab combination showed no new safety signals and met its primary endpoint with 18% ORR in advanced, chemotherapy-refractory MpBC. All responses are ongoing at >2 to almost 3 years later. The effect of ipilimumab and nivolumab was associated with exceptional responses in a subset of patients versus no activity. This combination warrants further investigation in MpBC, with special attention to understanding mechanism of action, and carefully designed to weigh against the significant risks of irAEs.

We present a pilot, untargeted extragalactic carbon monoxide (CO) emission-line survey using ALMACAL, a project utilizing ALMA calibration data for scientific purposes. In 33 deep (Texp > 40 min) ALMACAL fields, we report six CO emission-line detections above S/N > 4, one-third confirmed by MUSE observations. With this pilot survey, we probe a cosmologically significant volume of ∼105 cMpc3, widely distributed over many pointings in the southern sky, making the survey largely insusceptible to the effects of cosmic variance. We derive the redshift probability of the CO detections using probability functions from the Shark semi-analytical model of galaxy formation. By assuming typical CO excitations for the detections, we put constraints on the cosmic molecular gas mass density evolution over the redshift range 0 < z < 1.5. The results of our pilot survey are consistent with the findings of other untargeted emission-line surveys and the theoretical model predictions and currently cannot rule out a non-evolving molecular gas mass density. Our study demonstrates the potential of using ALMA calibrator fields as a multi-sightline untargeted CO emission-line survey. Applying this approach to the full ALMACAL database will provide an accurate, free of cosmic variance, measurement of the molecular luminosity function as a function of redshift.

This paper presents an indirect estimation based active disturbance rejection control (ADRC) for solar sail station-keeping. In contrast to previous applications of ADRC to spacecraft station-keeping the proposed approach estimates the deviation of the orbit state from the reference orbit, instead of estimating the orbit state directly. Since the deviation of the orbit state varies more slowly than the state itself, the indirect estimation significantly improves the estimation accuracy and hence the station-keeping performance. Moreover, this approach enables a large control time step which is two orders of magnitude larger than those in previous applications. This is beneficial to the implementation of attitude control and power consumption in practice. In addition, errors in the optical properties of the solar sail are taken into account which is a major challenge for solar sail station-keeping that has not been considered in previous station-keeping control design. The simulation results show that the presented ADRC station-keeping is robust to errors in these optical properties.

Current control approaches for solar sail station-keeping on libration point orbits have not considered the degradation of the sail’s optical properties. However, significant optical degradation could lead to poor station-keeping performance or even complete failure. This paper presents an integrated guidance and control strategy to address this problem by updating the reference orbit based on in situ estimation. An exponential optical degradation model is incorporated into the solar radiation acceleration model, and an on-line reference orbit update approach is incorporated into the station-keeping, coupled with an active disturbance rejection controller. The reflection coefficient is estimated on-line and the reference orbit is updated discretely when the optical properties have degraded by a prescribed amount. This strategy provides discrete updates to the reference orbits such that the perturbation due to the optical degradation is maintained within a small range. These smaller perturbations can be dealt with by the controller’s robustness and station-keeping can be sustained for long durations even in the presence of large optical degradation.

This paper addresses the attitude control of nano-spacecraft with three reaction wheels and three magnetorquers in low Earth orbits, and develops algorithms to maintain control when any one of the reaction wheels completely fails. In the event of a reaction wheel failure, it is shown that the key challenge of maintaining accurate autonomous three-axis tracking is a problem of singularity avoidance in control allocation. In particular, control law singularities occur depending on the type of orbit, the environment, and the attitude, where the system is no longer controllable. Two algorithmic singularity avoidance techniques based on singularity prediction and artificial potential functions are proposed. In addition, an optimized reaction wheel configuration is proposed to enhance singularity avoidance capability. To illustrate the effectiveness of the singularity avoidance control laws, they are tested in simulations that include the effects of perturbing environmental torques, reaction wheel jitter, and actuator misalignment.

On-orbit assembly strategies are proposed for an orb-shaped solar array which forms the main structure of a space solar power station. A dynamic model for the structural vibration under the influence of the gravity gradient is proposed by combining the Tschauner-Hempel equation and a finite element model of an elastic beam. The motion of the robot, used for on-orbit assembly, is described analogously to a classical vehicle-bridge coupled dynamic model. Assembly strategies are presented and analyzed, consisting of a single-robot assembly strategy, an assembly strategy using two robots and structural reinforcements. The proposed methods are verified in simulation and tuned to minimize the structural vibration during assembly.

A supercavitating underwater vehicle uses gas or vapor to reduce its skin friction enabling it to travel up to speeds of 200 m/s. However, traveling at extreme speeds can be challenging for control design since sudden disturbances or faults, even for a short time, can lead to large deviations from the reference trajectory. This paper addresses the problem of controlling a supercavitating vehicle which incorporates the actuator constraints and loss of effectiveness of the cavitator. An adaptive control law is designed based on the backstepping control method which includes an adaptive fault-tolerant component. The bounded adaptive update law can estimate the upper bound of the unknown total uncertainties including those induced by actuator faults. The proposed control is shown to guarantee stability in the presence of faults by using Lyapunov theory.

The opioid crisis has continued to progress in the United States and the rest of the world. As this crisis continues, there is a pressing need for a rapid and cost-effective method for detecting fentanyl. Recent studies have suggested that lateral flow immunoassays (LFIs) could fill this technology gap. These qualitative paper-based assays contain antibodies designed to react with fentanyl and provide positive or negative results within a matter of minutes. In this study, two different LFI configurations for the detection of fentanyl were examined (dipsticks and cassettes) for effectiveness of detection using seized drug samples and postmortem urine samples. In the current study, 44 seized drug samples (32 fentanyl-positive, 12 fentanyl-negative) and 14 postmortem urine samples (10 fentanyl-positive, 4 fentanyl-negative) were analyzed. All 32 fentanyl-containing seized drug samples and 10 postmortem fentanyl-positive urine samples displayed positive LFI results with both LFI configurations. The fentanyl dipsticks displayed a sensitivity of 100%, a specificity of 75%, and an efficiency of 93.2% for seized drug samples and a sensitivity, specificity, and efficiency of 100% for postmortem urine. Analysis of the fentanyl cassettes displayed a sensitivity, specificity, and efficiency of 100% for seized drug samples and a sensitivity of 100%, a specificity of 75%, and an efficiency of 92.9% for postmortem urine samples. These data point to the utility of LFIs as a quick and low resource-dependent option for presumptive detection of fentanyl in real-world situations.

Objective The aim of this study was to describe associations between peripheral leukocyte gene expression and early postpartum disease in dairy cows with clinical metritis. Materials and Methods The study was conducted in a conventional dairy in Washington state. Diseased cows ≤14 DIM were enrolled (n = 11) based on evidence of clinical metritis with or without comorbidity or prior treatments. Healthy cows (n = 9) were selected based on comparative DIM and lactation numbers to diseased cows. Blood was collected for a complete blood count, serum biochemistry, leukocyte isolation, and RNA extraction at enrollment and twice more at 6- to 8-d intervals. Results and Discussion A total of 34,625 genes were considered and 55 were associated with health status by the Boruta all-relevant feature machine learning selection algorithm. These were filtered to the 15 most representative genes within the diseased cohort as compared with the healthy cohort. The top ranked gene during every rank or error test was PGLYRP1. Its associated protein, peptidoglycan recognition protein 1, and other immunomodulatory molecules associated with this study selectively enhance or alter host innate immune defense mechanisms and modulate pathogen-induced inflammatory responses. Implications and Applications Host-defense peptides such as those associated with this study provide insight into individual and population-level markers of disease resilience that may help improve therapeutics, guide genetic selection, and clarify the burden of disease on animal well-being. This is particularly relevant to the dairy cow transition period with its metabolic and physiologic changes and associated increase in disease susceptibility.

With the spatial resolution of the Atacama Large Millimetre Array (ALMA), dusty galaxies in the distant Universe typically appear as single, compact blobs of dust emission, with a median half-light radius, $\approx$ 1 kpc. Occasionally, strong gravitational lensing by foreground galaxies or galaxy clusters has probed spatial scales 1-2 orders of magnitude smaller, often revealing late-stage mergers, sometimes with tantalising hints of sub-structure. One lensed galaxy in particular, the Cosmic Eyelash at $z=$ 2.3, has been cited extensively as an example of where the interstellar medium exhibits obvious, pronounced clumps, on a spatial scale of $\approx$ 100 pc. Seven orders of magnitude more luminous than giant molecular clouds in the local Universe, these features are presented as circumstantial evidence that the blue clumps observed in many $z\sim$ 2-3 galaxies are important sites of ongoing star formation, with significant masses of gas and stars. Here, we present data from ALMA which reveal that the dust continuum of the Cosmic Eyelash is in fact smooth and can be reproduced using two S\'ersic profiles with effective radii, 1.2 and 4.4 kpc, with no evidence of significant star-forming clumps down to a spatial scale of $\approx$ 80 pc and a star-formation rate of $, Comment: 5 pages; 3 figures; in press as a Letter to MNRAS

This paper addresses the problem of optimally repointing the optical axis of a spacecraft to align with the target direction. A new metric defining the repointing error is proposed where the corresponding kinematic equations provide a simple and convenient form for control design. The proposed control integrates a Control Lyapunov Function (CLF) approach with a sliding mode controller which simultaneously guarantees the optimality and robustness of the closed-loop system. Firstly, a CLF based control scheme is used to ensure that the state optimally converges to the sliding surface. Then a fixed-time non-singular terminal sliding mode controller is employed to provide robust convergence to the origin along the sliding surface. The convergence time is finite for any initial states and is thus useful for applications with critical time constraints. The region of attraction and convergence time is analyzed. Finally, numerical investigations are conducted to verify the effectiveness and superiority of the proposed algorithm with respect to the classical CLF method.

We present results of the MUSE-ALMA Halos, an ongoing study of the Circum-Galactic Medium (CGM) of low redshift galaxies (z < 1.4), currently comprising 14 strong HI absorbers in five quasar fields. We detect 43 galaxies associated with absorbers down to star formation rate (SFR) limits of 0.01-0.1 solar masses/yr, found within impact parameters (b) of 250 kpc from the quasar sightline. Excluding the targeted absorbers, we report a high detection rate of 89 per cent and find that most absorption systems are associated with pairs or groups of galaxies (three to eleven members). We note that galaxies with the smallest impact parameters are not necessarily the closest to the absorbing gas in velocity space. Using a multi-wavelength dataset (UVES/HIRES, HST, MUSE), we combine metal and HI column densities, allowing for derivation of the lower limits of neutral gas metallicity as well as emission line diagnostics (SFR, metallicities) of the ionised gas in the galaxies. We find that groups of associated galaxies follow the canonical relations of N(HI) -- b and W_r(2796) -- b, defining a region in parameter space below which no absorbers are detected. The metallicity of the ISM of associated galaxies, when measured, is higher than the metallicity limits of the absorber. In summary, our findings suggest that the physical properties of the CGM of complex group environments would benefit from associating the kinematics of individual absorbing components with each galaxy member., Comment: Accepted for publication in MNRAS; 23 pages, 25 figures

Gene co-expression networks (GCNs) provide multiple benefits to molecular research including hypothesis generation and biomarker discovery. Transcriptome profiles serve as input for GCN construction and are derived from increasingly larger studies with samples across multiple experimental conditions, treatments, time points, genotypes, etc. Such experiments with larger numbers of variables confound discovery of true network edges, exclude edges and inhibit discovery of context (or condition) specific network edges. To demonstrate this problem, a 475-sample dataset is used to show that up to 97% of GCN edges can be misleading because correlations are false or incorrect. False and incorrect correlations can occur when tests are applied without ensuring assumptions are met, and pairwise gene expression may not meet test assumptions if the expression of at least one gene in the pairwise comparison is a function of multiple confounding variables. The ‘one-size-fits-all’ approach to GCN construction is therefore problematic for large, multivariable datasets. Recently, the Knowledge Independent Network Construction toolkit has been used in multiple studies to provide a dynamic approach to GCN construction that ensures statistical tests meet assumptions and confounding variables are addressed. Additionally, it can associate experimental context for each edge of the network resulting in context-specific GCNs (csGCNs). To help researchers recognize such challenges in GCN construction, and the creation of csGCNs, we provide a review of the workflow.

In recent decades, the Venice Charter of 1964 [1] has provided the guiding principles for the conservation and restoration of ancient monuments. However, many interpret these principles as applying to historic structures in general, and not just monuments. The articles in the Restoration section of the Charter have several interesting statements (underlines are for emphasis) that are open to interpretation. In many cases, these statements cause a conflict of priorities, especially with funding being the overriding issue. In addition, local and national heritage agencies sometimes take a more liberal approach to restoration, particularly regarding authenticity. The statements under discussion are: “ARTICLE 9. The process of restoration is a highly specialized operation. Its aim is to preserve and reveal the aesthetic and historic value of the monument and is based on respect for original material and authentic documents. It must stop at the point where conjecture begins, and in this case moreover any extra work which is indispensable must be distinct from the architectural composition and must bear a contemporary stamp.” “ARTICLE 10. Where traditional techniques prove inadequate, the consolidation of a monument can be achieved by the use of any modem technique for conservation and construction, the efficacy of which has been shown by scientific data and proved by experience.” “ARTICLE 12. Replacements of missing parts must integrate harmoniously with the whole, but at the same time must be distinguishable from the original so that restoration does not falsify the artistic or historic evidence.” Each of these statements affects the authenticity of the restoration. But, maintaining authenticity of the restoration has to be balanced with the reality of maintaining our heritage buildings on limited resources. Can it be done? This paper discusses these challenges in the context of the 1996 restoration of a threestory, 19th century brownstone. The paper will include the conflicts with recommendations for an authentic restoration in accordance with the Charter principles. The work was performed on a limited budget and attempted to address the Owner’s desire for an aesthetic solution. Finally, an assessment of the restoration after 23 years will be included

Background Quantification of gene expression from RNA-seq data is a prerequisite for transcriptome analysis such as differential gene expression analysis and gene co-expression network construction. Individual RNA-seq experiments are larger and combining multiple experiments from sequence repositories can result in datasets with thousands of samples. Processing hundreds to thousands of RNA-seq data can result in challenges related to data management, access to sufficient computational resources, navigation of high-performance computing (HPC) systems, installation of required software dependencies, and reproducibility. Processing of larger and deeper RNA-seq experiments will become more common as sequencing technology matures. Results GEMmaker, is a nf-core compliant, Nextflow workflow, that quantifies gene expression from small to massive RNA-seq datasets. GEMmaker ensures results are highly reproducible through the use of versioned containerized software that can be executed on a single workstation, institutional compute cluster, Kubernetes platform or the cloud. GEMmaker supports popular alignment and quantification tools providing results in raw and normalized formats. GEMmaker is unique in that it can scale to process thousands of local or remote stored samples without exceeding available data storage. Conclusions Workflows that quantify gene expression are not new, and many already address issues of portability, reusability, and scale in terms of access to CPUs. GEMmaker provides these benefits and adds the ability to scale despite low data storage infrastructure. This allows users to process hundreds to thousands of RNA-seq samples even when data storage resources are limited. GEMmaker is freely available and fully documented with step-by-step setup and execution instructions.

This paper describes the trajectory tracking of an underactuated autonomous underwater vehicle (AUV) with three control inputs (surge, yaw and pitch moment) that operates in the presence of time-varying disturbances. The AUV kinematics are described in global coordinates as a Hamiltonian system on the Lie group SE(3) and the boundary-value problem arising from the geometric framing of Pontryagin’s Maximum Principle is applied to the vehicle kinematics. This 6-dimensional boundary value problem is solved using a numerical shooting method and a novel semi-analytical Lie group integrator. The integrator uses Rodrigue’s formula to express the exact solution of the solution curves, is symplectic and preserves energy and momentum. Inverse dynamics is applied to construct an inner-loop controller, which accounts for constraints on maximum torque and force via time reparametrization. This inner-loop control, which would drive the AUV along the reference trajectory in perfect conditions, is combined with a disturbance observer to construct an outer-loop controller, which ensures stability in the presence of bounded disturbances. Simulation results complete the work.

We present results of MUSE-ALMA Halos, an ongoing study of the Circumgalactic Medium (CGM) of galaxies ($z \leq$ 1.4). Using multi-phase observations we probe the neutral, ionised and molecular gas in a sub-sample containing six absorbers and nine associated galaxies in the redshift range $z \sim 0.3-0.75$. Here, we give an in-depth analysis of the newly CO-detected galaxy Q2131-G1 ($z=0.42974$), while providing stringent mass and depletion time limits for the non-detected galaxies. Q2131-G1 is associated with an absorber with column densities of $\textrm{log}(N_\textrm{HI}/\textrm{cm}^{-2}) \sim 19.5$ and $\textrm{log}(N_{\textrm{H}_2}/\textrm{cm}^{-2}) \sim 16.5$, has a star formation rate of $\textrm{SFR} = 2.00 \pm 0.20 \; \textrm{M}_{\odot} \textrm{yr}^{-1}$, a dark matter fraction of $f_\textrm{DM}(r_{1/2}) = 0.24 - 0.54$ and a molecular gas mass of $M_\textrm{mol} = 3.52 ^{+3.95}_{-0.31} \times 10^9 \; \textrm{M}_{\odot}$ resulting in a depletion time of $\tau_\textrm{dep} < 4.15 \; \textrm{Gyr}$. Kinematic modelling of both the CO (3--2) and [OIII] $\lambda 5008$ emission lines of Q2131-G1 shows that the molecular and ionised gas phases are well aligned directionally and that the maximum rotation velocities closely match. These two gas phases within the disk are strongly coupled. The metallicity, kinematics and orientation of the atomic and molecular gas traced by a two-component absorption feature is consistent with being part of the extended rotating disk with a well-separated additional component associated with infalling gas. Compared to emission-selected samples, we find that HI-selected galaxies have high molecular gas masses given their low star formation rate. In geostationary orbit, long eclipses and the seasonal variations in the direction and intensity of the solar input can cause damage to sensitive equipment during attitude maneuvers, which may inadvertently point the equipment towards the Sun. The requirement that transmitting and receiving antennae remain pointed towards the Earth creates further restrictions to pointing directions. The aim of the study is to construct a novel geometric and reinforcement-learning-based method to determine attitude guidance maneuvers that maintain the equipment in safe and operational orientations throughout an attitude maneuver. The attitude trajectory is computed numerically using the geometric framing of Pontryagin’s maximum principle applied to the vehicle kinematics using the global matrix Lie group representation on SO(3), and the angular velocities are shaped using free parameters. The values of these free parameters are determined by a reinforcement learning algorithm to avoid the forbidden areas while maintaining the pointing in operational areas (modeled as subsets of the two-sphere of all possible pointing directions of a particular axis). The method is applied to a model geosynchronous satellite and demonstrated in a simulation.
