In the present work, Chlorella vulgaris, a well-known microalga was used as a carbonic precursor, to synthesis the water-soluble carbon dots (CDs) via hydrothermal method. To improve the properties, acidic carbon dots (ACDs) were also synthesized via ultrasoncation of microalgal biomass after acidic hydrolysis (using diluted acidic solution). The aqueous solution of ACDs was used as a sensor for glucose determination based on glucose oxidase enzyme (GOx) reaction and fluorescence quenching of ACDs in the presence of Fe3+ ions under the optimized conditions. The linear range of glucose in the blood serum was measured from a range of 5 µM to 500 µM with a limit of detection (LOD) 2.84 µM. Additionally, the hydrogen peroxide, as the conventional reactive oxygen species (ROS) was determined based on the Fenton reaction. The linear range of H2O2 was obtained from 10 µM to 200 µM with LOD of 975 nM. In all of these applications, fluorescence quenching of ACDs was followed according to photo-induced electron transfer (PET) mechanism in the presence of generated Fe3+ ions by Fenton reaction.

The increasing use of nanoparticles and their many applications increases the likelihood of their presence in the environment. This possibility of presence necessitates the study of the effect of these substances on aquatic species. In this research, CoWO4 nanoparticles were synthesized by the ultrasonic method. Various conditions in the synthesis process were investigated to obtain the appropriate size of the nanoparticles. After selecting the optimum particles, these nanoparticles were used to investigate their effect on the growth of Dunaliella salina. For this purpose, the algal cells were subjected to three different concentrations of nanoparticles (15, 30, and 60 mg/L). The study results on algae growth parameters showed that these parameters depend on the value of nanoparticles. At 15 and 30 mg/L concentrations of the nanoparticles, numbers of cells, specific growth, biomass, and pigments showed a significant boost compared to the mentioned parameters of the control treatment. Measurement of malondialdehyde (MDA) showed that this parameter was directly related to the increase in the concentration of nanoparticles. At 60 mg/L of the nanoparticles, the MDA level was higher than the control and other treatments. This increase reflects the destructive effect of the nanoparticles on algal cells. Finally, the results showed that algae could be useful for studying the environmental effects of nanoparticles and their safety.

The presence of nanoparticles in the environment and their impact on existing organisms is one of the main concerns of researchers working in this field. In this research, Nd2WO6 nanoparticles were prepared by an ultrasonic procedure for the first time. X-ray powder diffraction (XRD), Fourier-transform infrared spectroscopy (FT-IR), and energy-dispersive X-ray spectroscopy (EDS) analyses were applied to identify and prove the purity of these particles. In addition to increasing the reaction rate and efficiency with the help of a radical generation mechanism, ultrasound was able to aid the synthesis of these particles. After confirming nanoparticle formation, the optimal nanoparticles in view of scale and morphology were selected by scanning electron microscopy (SEM) and transmission electron microscopy (TEM). Optimal particles at three concentrations (25, 50, and 100 ppm) were mixed into the algae growth medium to investigate the effects of the nanoparticles on Dunaliella salina growth. Biological parameters, including the number of cells, biomass, specific growth rate, pigments, and malondialdehyde (MDA), were measured after ten days. Growth parameters showed an increasing trend in concentrations up to 50 ppm; however, at a concentration of 100 ppm, a significant decrease was observed in contrast to the nanoparticles-free treatment. The MDA content showed a linear relationship with enhanced concentration of the nanoparticles. The examination of biological parameters showed that the algae response to stress was dependent on the concentration of nanoparticles. The results showed that 50 ppm of nanoparticles are suitable for increasing algae and achieving a suitable growth rate for commercial purposes. However, in higher concentrations, algal growth inhibition occurs, which is of great importance from a biotechnological point of view.

Drought stress was evaluated with polyethylene glycol 6000 (PEG 6000) treatment in Dunaliella salina, a microalga known for its great ability to withstand salinities of more than 30%. The aim was to explore the acclimation mechanisms used by the microalga to regulate its growth and physiology during coping with drought stress. The microalga was subjected to culture mediums containing 2 and 5% PEG for 25 days and was compared with a control culture medium. Significant decrease in growth parameters such as specific growth rate, biomass and number of divisions per day was demonstrated in PEG-treated algae. During PEG treatment, chlorophylls slightly increased, while β-carotene and total protein were not affected. Osmolytes, as well as carbohydrates, were found to be significantly higher in PEG-treated algae than in control. Increased catalase and ascorbate peroxidase activities were proportionally related to PEG concentrations in the cultures. The PEG-treated cells accumulated a considerable amount of hydrogen peroxide and malondialdehyde, especially at higher PEG concentrations. Electrolyte leakage increased, regardless of the PEG concentrations applied, while DNA fragmentation was not observed after 25 days of treating with PEG. It was concluded that Dunaliella cells could respond to the drought stress, probably by using a higher accumulation of a range of osmolytes and also more stimulation of the antioxidant enzymatic system.

The 70-kDa heat shock proteins (HSP70s) are a conserved class of chaperones that play critical roles during the normal life cycle of plants. HSP70s are particularly involved in the regulation of biotic and abiotic stress responses. In this paper, the potential roles of this protein were investigated. A reverse genetic approach was employed for transient silencing of hsp70 gene in tomato (Solanum lycopersicum L.) to evaluate different growth and physiological parameters under normal conditions and during the response to drought stress. A combined ANOVA (analysis of variance) and HCA (hierarchical clustering analysis) showed that hsp70 silencing led to severe growth retardation and mortality, significant membrane damage and leakage, decline in relative water content, low rate of pigment accumulation, and reduced antioxidant enzyme activity under normal and drought stress conditions. Among the different parameters, proline was the only trait that was unaffected by gene silencing and accumulated by similar amounts to that of nonsilent plants. In conclusion, HSP70 played critical roles in maintaining the cellular homeostasis of plants during adaptation to drought and under normal plant life conditions. It was speculated that proline was, to some extent, involved in improving the loss of protein folding or function resulting from HSP70 deficiency, and played a crucial role in the adaptation of plants on exposure to stress.

Microalgae as one of the key components of food chains in aquatic environments are promising biological models for investigating gamma irradiation effects on eukaryotic organisms. Understanding resistance mechanisms in these organisms as simple models might illuminate how gamma irradiation resistance improves in these algae and even more complex organisms. The present study aimed to investigate the effects of ionizing irradiation on Chlorella vulgaris as a stress-tolerant microalga and to find how the eukaryotic cells would tolerate such stressful conditions. The physiological responses and biochemical alterations of C. vulgaris were analyzed at three different time points (0, 16, and 48 h) after 600 Gy gamma irradiation. Compared to the control, gamma-irradiated algae had slower growth rate with significantly longer lag phase, less chlorophyll and protein contents at time 0, which were compensated and recovered during the next 48 h. The results also showed spontaneous H2O2 burst accompanied by a higher rate of lipid peroxidation and electrolyte leakage, a rapid increase of catalase activity and more ferric reducing antioxidant power immediately after irradiation. During a 48-h period, most alterations stabilized. Raising trends were observed in the carotenoid contents, the ratios of carbohydrates, amide I and amide II to fatty acids. Principal component analysis and hierarchical clustering suggest two possible distinct mechanisms: a “quick” one including spontaneous responses by boosting H2O2, amplifying enzymatic antioxidant systems, and increasing compatible solutes like proline and a “delayed” responsive strategy including the increase of soluble carbohydrates, carotenoids, and stress-related proteins triggered several hours after irradiation.

To characterize the resistance patterns of uropathogenic Escherichia coli (UPEC) in a Tertiary Teaching Hospital in Iran, we conducted a descriptive epidemiology study using molecular techniques. The subjects consisted of patients having acute urinary tract infection, who were enrolled in the study from 2014 to 2017. The antimicrobial susceptibility profile of 101 UPEC isolates was determined by Kirby-Bauer disc diffusion method. Extended spectrum β-lactamase (ESBL) was detected by the double-disk synergy test. Biofilm formation was done using microtiter plates. The presence of virulence genes (pai, pap, hly, traT, pai, cnf-1, sfa, and afa) was evaluated by a PCR. Molecular typing of UPEC E. coli isolates was performed with fimH and multilocus sequence typing (MLST). 70.3% of isolates were multidrug-resistant. 37.6% of isolates were Extended spectrum β-lactamases (ESBLs) producer. Strong biofilm formation was seen in 27.7%. Forty-seven different fimH allelic variants were identified. Among identified fimH allelic variants, the most common types were f1 (18.8%) and f14 (18.8%). ST131 (54.5%) was the most prevalent clonal group significantly correlated with the pai gene. Seven sequence types (STs) were detected only once (ST405, ST410, ST450, ST636, ST648, ST1193, and ST6451). Clonal groups showed no significant differences in terms of antibiotic resistance patterns. There was no significant difference between virulence genes and antibiotic resistance patterns in the studied clonal groups. To our knowledge, the present study is the first study in Iran that investigated the genotypic diversity of UPEC isolates by MLST and fimH typing methods. The two methods might serve as a useful molecular test for surveillance and epidemiological studies of isolates.

The natural variation of drought tolerance among eight Iranian bermudagrass (Cynodon dactylon L.) accessions was evaluated and compared with a commonly used variety by measuring various morphological, growth, physiological and molecular responses during mild and severe drought stress conditions. A combined multivariable analysis using principal component analysis, hierarchical clustering analysis, as well as drought-tolerance ranking measurement showed that the accessions were clustered into three groups in terms of drought tolerance. This method evaluated three accessions (Maragh, Abyaneh, and Badrood) as superior drought-tolerant, three (Karshahi, Josheghan, and Kashan) as relatively drought-tolerant, and three (Ardeha, SWI-7, and Khonb) as drought-sanative genotypes. Drought-tolerant accessions generally showed improved growth and relative water content (RWC), minor electrolyte leakage (EL) and lipid peroxidation (MDA), higher content of pigments (Chlorophyll and carotenoid) and dark green color index (DGCI), more significant up-regulation of two stress-related genes (ECERIFERUM1 and Delta 1-pyrroline-5-carboxylate synthetase), as well as higher accumulation of proline content compared to sensitive accessions under drought stress. On the other hand, drought-sensitive accessions typically had more activity of antioxidant enzymes (catalase and ascorbate peroxidase), and higher amounts of soluble sugars (glucose, mannose, and rhamnose), compared to tolerant accessions under drought stress. In conclusion, different defensive strategies may be recruited by tolerant or sensitive bermudagrass genotypes during encountering to drought stress.

A simple and one-pot glycothermal method was developed for the synthesis of Ni/ZnO nanocomposites. Ethylene glycol, zinc and nickel complexes were utilized as a solvent and new precursors to prepare Ni/ZnO nanocomposites, respectively. The effect of different reaction time and temperature were examined in order to approach to the optimal sample. The Ni/ZnO nanocomposites were characterized by X-ray diffraction (XRD) analysis, Transmission Electron Microscopy (TEM), Fourier transform infrared (FT-IR) spectroscopy, Scanning Electron Microscopy (SEM), and ultraviolet–visible (UV–Vis) spectroscopy. Vibrating sample magnetometer analysis was exhibited the ferromagnetic behavior of the nanocomposites, which is appropriate for nanoparticle recovery. The antibacterial effect of the selected sample was investigated by monitoring the growth of Pseudomonas aeruginosa, Bacillus subtilis, and Candida albicans, where, after 24 h, the diameter of the zone of inhibition was obtained to be around 15, 14 and 9 mm at 37 °C, respectively. Interestingly, the diameter of the inhibition zone for the Pseudomonas aeruginosa was remarkably comparable to conventional antibiotics.

Heat shock proteins (HSPs) have vital roles during plant adaptation to biotic and abiotic stresses, as well as stress-free conditions. In the present study, we used a heterologous strategy of virus induced gene silencing to investigate the role of different classes of HSPs in Nicotiana benthamiana. Different growth and physiological parameters in silenced plants were evaluated under both control and salt stress conditions. Among the treatments, silencing of HSP70, especially under salinity regime, was found to have stronger impacts on growth, protein concentration, the accumulation of photosynthetic pigments, proline and total soluble carbohydrates content, malondialdehyde, the activity of antioxidant enzymes, performance index, relative water content and the ratio of K+/Na+, suggesting a more prominent role for HSP70 in both stressful and stress-free life cycle of the plants. Principal component analysis and hierarchical clustering indicated that HSP70 gene was silenced, the plants might effectively respond to stress by a higher accumulation of compatible solutes, like proline to protect the cell membranes and proteins from damage.

Calcineurin-B-like proteins (CBLs) are essential components of the calcium signaling network and act during plant's response to stress and normal conditions. A combined research strategy of in-silico analysis and gene silencing experiment was employed to investigate the role of different classes of CBLs in tomato (Solanum lycopersicum L.) during the response to drought stress. Two different classes of CBL genes, including SlCBL3-1, and SlCBL3-2, with the minimum and a maximum number of drought-responsive cis-elements, were selected and were targeted for transient gene silencing in tomato followed by the drought treatment. The effect of silencing events was evaluated by determining of further growth and physiological traits in plants under both control and drought stress conditions. The results showed that silencing of SlCBL3-1 significantly reduced shoot and root growth, relative water content (RWC), and the concentration of pigments while increased free radical accumulation, lipid peroxidation, and leakage from the cells. On the other hand, no antioxidant enzyme activity or proline induction was triggered in plants after SlCBL3-1 silencing. Some of these adverse events were more significantly enhanced when the silenced plants were exposed to drought stress. Overall, a significant role for SlCBL3-1 in the life cycle of plant suggested under both normal and stress conditions. The SlCBL3-2 silencing showed more efficient plants recovery from silencing or drought stress conditions. Therefore, SlCBL3-2 gene may act as a negative regulator under stress conditions. The results might provide new theoretical insight and genetic resources for developing resistant crops against environmental stresses.

The growth and physiological parameters of fourteen tomato cultivars were measured during both mild and severe drought. The tomato cultivars showed decrease in growth traits, relative water content and pigments content under both drought treatments compared with the control. By contrast, proline accumulation, malondialdehyde and electrolyte leakage increased in the cultivars. The activity of superoxide dismutase was more correlated to drought stress than those of ascorbate peroxidase or catalase. The combined approach of principal component analysis, hierarchical clustering analysis and calculating mean tolerance value showed relative differences in drought tolerance among the cultivars. The studied cultivars were clustered into four groups. Four cultivars were placed in cluster I as the drought-tolerant group. Moderately tolerant cultivars were grouped into Cluster II. The clusters III and IV represented the sensitive and highly sensitive cultivars, respectively. Early orbana, Roma and Cal-j cultivars were indicated as the highly sensitive tomato cultivars. It was concluded that such an approach could be used as a useful tool to screen the other abiotic or even biotic stress tolerance in tomato.

Myrtle (Myrtus communis L.) is an evergreen and aromatic plant species with therapeutic and ornamental values. The effect of water stress induced by three concentrations of Polyethylene glycol (PEG), 0% (control), 3% (3%PEG) and 6% (6% PEG) was evaluated on in vitro-regenerated myrtle shoots. After 30 days of culture, all regeneration, growth and physiological parameters were measured at the end of the experiment. Murashige and Skoog (MS) medium contained 1.5 mgl-1 6-benzyladenine (BAP) and 0.2 mgl-1 Naphthaleneacetic acid (NAA) was optimized for the in vitro experiment. Increasing PEG concentration in the culture media resulted in fewer regenerated shoots, and higher rate of browning in the apical tissues. Chlorophylls and total carotenoids content significantly decreased in the shoots treated with PEG, however there were no significant differences in the pigment content under moderate or severe water stress conditions. PEG-induced water stress resulted in reduced plant growth and relative water content of newly-formed shoots. Lipid peroxidation and electrolyte leakage were lower in myrtle shoots treated with 6%PEG than 3%PEG, but they were still higher than that of the control. Proline accumulation significantly increased under 3%PEG treatment, while a little difference were observed between proline content under 6%PEG and the control. Apoptosis did not occur in the myrtle shoots grown under moderate or severe stress treatments which correlated with more increased Catalase and Superoxide dismutase activities in response to higher stress levels. The results showed a possible contribution of enzymatic antioxidant system in myrtle when counteracting severe water stress.

Type 2 diabetes is one of the most prevalent diseases, which increases resistance to insulin in target tissues. The measurement of miRNAs quantity is a molecular approach for diagnosis of diabetes. miRNAs are small non-coding RNA strings of 21-23 long nucleotides that act as inhibitors in proteins translation. Several methods including Northern blot, qRT-PCR and Microarray have been used for diagnosis of miRNA molecules. Real time PCR is an expensive and accurate quantitative method that is widely used in miRNA studies. The miR-21 is an important miRNA in diabetes. In this study, for the first time, a semi-quantitative protocol was developed to quantify different amounts of a synthetic miR-21. In addition to semi-quantitative method, the miR-21 quantity was determined by quantitative method in several patients with type 2 diabetes and healthy people. The results indicated that there was a direct relationship between the amount of synthetic miR-21 and the intensity of the PCR bands. We also showed that the expression of miR-21 in people with type 2 diabetes increased compared to healthy people. The results were observed by both quantitative and semi-quantitative methods. The real-time RT-PCR was more sensitive than semi-quantitative PCR in identification of miRNAs. However, semi-quantitative PCR method benefited from higher simplicity and lower costs for defining general patterns of miRNA expression.

In this work, Fe2WO6 nanoparticles were synthesized by the ultrasound-assisted precipitation method. Various conditions were applied, including the change of the pH factor and reaction time for the synthesis of nanoparticles. After confirming the synthesis of the nanoparticles by various analyzes and evaluating their size and morphology, one of the conditions for the synthesis of the nanoparticles were selected as the optimum condition. The samples were added to the growth medium of a well-known microalga, Dunaliella salina at three concentrations of 20, 40 and 80 ppm to evaluate the effect of nanoparticles on biological systems. After 10 days different biological parameters were measured and compared with those of the control sample. According to the results, at concentration of 20 ppm the number of cells, the amount of chlorophyll a, and b, and biomass increased compared to the control samples. The Carotenoid level was higher in the treatment with 40 ppm of nanoparticles than that in the control samples. Compared to the control sample, the level of lipid peroxidation and the ratio of carbohydrate to amide II showed to be higher under 80 ppm treatment of particles. According to HCA analysis, both the evaluated parameters and concentrations of nanoparticles were divided into two general categories. Overall results showed that the effect of Fe2WO6 nanoparticles on microalgae could be a dose-dependent phenomenon, so that the addition of 20 ppm nanoparticles in the culture media helped the growth and the physiological status of algae. On the other hand, the application of a higher concentration of nanoparticles negatively affects algal biology. The results showed that the algae could be successfully used to precise screen of various nanoparticles in terms of safety especially in aquatic environments and also biotechnological applications.

The objective of the present study was to investigate the effect of CaCl2 pre-treatment at different concentrations on the responses of microalga C. vulgaris under gamma radiation toxicity. Changes in growth, physiological parameters and biochemical compositions of the algae pretreated with 0.17 (normal), 5, and 10 mM CaCl2 were analyzed under 300 Gy gamma irradiation and compared to those of gamma-free control. The results showed that parameters including specific growth rate, cell size, chlorophyll and protein contents, ascorbate peroxidase (APX), and superoxide dismutase (SOD) activity, Ferric Reducing Antioxidant Power (FRAP), and the ratios of nucleic acid to protein negatively affected by gamma irradiation. All these parameters, except for the ratios of nucleic acid to protein significantly increased in the algae when pretreated with a CaCl2 content higher than normal concentration. The analysis also showed that parameters including catalase activity, proline, and carotenoid content, the level of lipid peroxidation, and electrolyte leakage (EL) significantly increased under gamma irradiation but not affected significantly under different CaCl2 pre-treatments. Additionally, specific growth rate, chlorophyll a and protein content, APX and SOD activity, FRAP, lipid peroxidation, electrolyte leakage, and the ratios of nucleic acid to protein were the only parameters that significantly affected by the interaction of gamma toxicity and CaCl2 pretreatment. Overall, the results suggested that regardless of the CaCl2 effect, the algal cells responded to gamma radiation more efficiently by increasing proline, carotenoids content, and CAT activity. More important, it was concluded that calcium had an essential role in modifying the detrimental effect of gamma toxicity on the algae mainly by increasing the activity of ascorbate peroxidase and superoxide dismutase and maintaining the reducing antioxidant power (FRAP) of the cells at a high level.

In this work, we investigated the importance of phytoene desaturase (PDS) during salt stress in plants. For this aim, we transiently suppressed the corresponding gene of Nicotiana benthamiana using a Tobacco Rattle Virus silencing system. After salinity stress, the silenced plants were assayed for different growth and physiological parameters. The silenced plants were found to be induced more stunted and feeble shoots and roots than the negative controls. The results indicated that the concurrent incidence of salinity stress and viral infection did not have an additive influences on the plant growth and physiological parameters. Importantly, our results showed a more detrimental influence of salinity on plant growth and physiology after PDS silencing. Compared to the control, salt-stressed plants contained more proline, and malondialdehyde, less amount of photosynthetic pigments, higher Na+/K+ ratios in shoots and roots and lower relative water content after PDS silencing. The activity of antioxidant enzymes were also decreased in the plants coped with salinity or Tobacco Rattle Virus. It was also represented that simultaneous silencing of the PDS gene and salinity treatment resulted in a significant decrease of the performance index (PIABS) and an increase of dissipation per active reaction center (DIo/RC) reflecting severe injuries in photosynthetic machinery under such a condition. Overall results suggested that PDS silencing could increase the sensitivity of plants to salinity.
