We study the dynamics of a seeding experiment where a fibrous scaffold material is colonized by two types of cell populations. The specific application that we have in mind is related to the idea of meniscus tissue regeneration. In order to support the development of a promising replacement material, we discuss certain rate equations for the densities of human mesenchymal stem cells and chondrocytes and for the production of collagen-containing extracellular matrix. For qualitative studies, we start with a system of ordinary differential equations and refine then the model to include spatial effects of the underlying nonwoven scaffold structure. This study is aimed to contribute to a more comprehensive understanding of the molecular hydrogen distribution in the galaxy M33 by introducing novel methods for generating high angular resolution (18.2$''$, equivalent to 75 pc) column density maps of molecular hydrogen ($N_{\rm H_2}$). M33 is a local group galaxy that has been observed with Herschel in the far-infrared wavelength range from 70 to 500 $\mu$m. Previous studies have presented total hydrogen column density maps ($N_{\rm H}$), using these FIR data (partly combined with mid-IR maps), employing various methods. We first performed a spectral energy distribution fit to the 160, 250, 350, and 500 $\mu$m continuum data obtain $N_{\rm H}$, using a technique similar to one previously reported in the literature. We also use a second method which involves translating only the 250 $\mu$m map into a $N_{\rm H}$ map at the same angular resolution. An $N_{\rm H_2}$ map via each method is then obtained by subtracting the HI component. Distinguishing our study from previous ones, we adopt a more versatile approach by considering a variable emissivity index, $\beta$ and dust absorption coefficient, $\kappa_0$. This choice enables us to construct a $\kappa_0$ map, thereby enhancing the depth and accuracy of our investigation of the hydrogen column density. We address the inherent biases and challenges within both methods (which give similar results) and compare them with existing maps available in the literature. Moreover, we calculate a map of the carbon monoxide CO-to-H$_2$ conversion factor ($X_\mathrm{CO}$ factor), which shows a strong dispersion around an average value of $1.8\times10^{20}\,\mathrm{cm^{-2}/(K\,km\,s^{-1})}$ throughout the disk. We obtain column density probability distribution functions (N-PDFs) from the $N_{\rm H}$, $N_{\rm H_2}$, and $N_{HI}$ maps and discuss their shape.

Understanding the biophysical and biochemical properties of molecular nanocarriers under physiological conditions and with minimal interference is crucial for advancing nanomedicine, photopharmacology, drug delivery, nanotheranostics and synthetic biology. Yet, analytical methods struggle to combine precise chemical imaging and measurements without perturbative labeling. This challenge is exemplified for azobenzene-based photoswitchable lipids, which are intriguing reagents for controlling nanocarrier properties on fast timescales, enabling, e.g., precise light-induced drug release processes. Here, we leverage the chemical recognition and high spatio-temporal resolution of scattering-type scanning near-field optical microscopy (s-SNOM) to demonstrate non-destructive, label-free mid-infrared (MIR) imaging and spectroscopy of photoswitchable liposomes below the diffraction limit and the tracking of their dynamics down to 50 ms resolution. The vesicles are adsorbed on an ultrathin 10-nm SiN membrane, which separates the sample space from the tip space for stable and hour-long observations. By implementing a transient nanoscopy approach, we accurately resolve, for the first time, photoinduced changes in both the shape and the MIR spectral signature of individual vesicles and reveal abrupt change dynamics of the underlying photoisomerization process. Photonic metasurfaces offer exceptional control over light at the nanoscale, facilitating applications spanning from biosensing, and nonlinear optics to photocatalysis. Many metasurfaces, especially resonant ones, rely on periodicity for the collective mode to form, which makes them subject to the influences of finite size effects, defects, and edge effects, all of which have considerable negative impact at the application level. These aspects are especially important for quasi-bound state in the continuum (BIC) metasurfaces, for which the collective mode is highly sensitive to perturbations due to high quality factors and strong near-field enhancement. Here, we quantitatively investigate the mode formation in quasi-BIC metasurfaces on the individual resonator level using scattering scanning near-field optical microscopy (s-SNOM) in combination with a new image processing technique. We find that the quasi-BIC mode is formed at a minimum size of 10 x 10-unit cells much smaller than expected from far-field measurements. Furthermore, we show that the coupling direction of the resonators, defects and edge states have pronounced influence on the quasi-BIC mode. This study serves as a link between the far-field and near-field responses of metasurfaces, offering crucial insights for optimizing spatial footprint and active area, holding promise for augmenting applications such as catalysis and biospectroscopy., Comment: 30 pages, 6 figures, 8 supplementary figures

High-latitude intermediate-velocity clouds (IVCs) are part of the Milky Way's HI halo and originate from either a galactic fountain process or extragalactic gas infall. They are partly molecular and can most of the time be identified in CO. Some of these regions also exhibit high-velocity cloud (HVC) gas, which is mostly atomic, and gas at local velocities (LVCs), which is partly atomic and partly molecular. We conducted a study on the IVCs Draco and Spider, both were exposed to a very weak UV field, using the receiver upGREAT on SOFIA. The 158 micron line of ionized carbon (CII) was observed, and the results are as follows: In Draco, the CII line was detected at intermediate velocities (but not at local or high velocities) in four out of five positions. No CII emission was found at any velocity in the two observed positions in Spider. To understand the excitation conditions of the gas in Draco, we analyzed complementary CO and HI data as well as dust column density and temperature maps from Herschel. The observed CII intensities suggest the presence of shocks in Draco that heat the gas and subsequently emit in the CII cooling line. These shocks are likely caused by the fast cloud's motion toward the Galactic plane that is accompanied by collisions between HI clouds. The nondetection of CII in the Spider IVC and LVC as well as in other low-density clouds at local velocities that we present in this paper (Polaris and Musca) supports the idea that highly dynamic processes are necessary for CII excitation in UV-faint low-density regions., Comment: in press Astronomy and Astrophysics

Scattering scanning near-field optical microscopy (s-SNOM) is a powerful technique for mid-infrared spectroscopy at nanometer length scales. By investigating objects in aqueous environments through ultrathin membranes, s-SNOM has recently been extended towards label-free nanoscopy of the dynamics of living cells and nanoparticles, assessing both the optical and the mechanical interactions between the tip, the membrane and the liquid suspension underneath. Here, we report that the tapping AFM tip induces a reversible nanometric deformation of the membrane manifested as either an indentation or protrusion. This mechanism depends on the driving force of the tapping cantilever, which we exploit to minimize topographical deformations of the membrane to improve optical measurements. Furthermore, we show that the tapping phase, or phase delay between driving signal and tip oscillation, is a highly sensitive observable for quantifying the mechanics of adhering objects, exhibiting highest contrast for low tapping amplitudes where the membrane remains nearly flat. We correlate mechanical responses with simultaneously recorded spectroscopy data to reveal the thickness of nanometric water pockets between membrane and adhering objects. Besides a general applicability of depth profiling, our technique holds great promise for studying mechano-active biopolymers and living cells, biomaterials that exhibit complex behaviors when under a mechanical load., Comment: 31 pages, 7 figures, 7 supplementary figures

We develop a model the dynamics of human mesenchymal stem cells (hMSCs) and chondrocytes evolving in a nonwoven polyethylene terephtalate (PET) scaffold impregnated with hyaluron and supplied with a differentiation medium. The scaffold and the cells are assumed to be contained in a bioreactor with fluid perfusion. The differentiation of hMSCs into chondrocytes favors the production of extracellular matrix (ECM) and is influenced by fluid stress. The model takes deformations of ECM and PET scaffold into account. The scaffold structure is explicitly included by statistical assessment of the fibre distribution from CT images. The effective macroscopic equations are obtained by appropriate upscaling from dynamics on lower (microscopic and mesoscopic) scales and feature in the motility terms an explicit cell diffusion tensor encoding the assessed anisotropic scaffold structure. Numerical simulations show its influence on the overall cell and tissue dynamics.

It has long been discussed whether stellar feedback in the form of winds and/or radiation can shred the nascent molecular cloud, thereby controlling the star formation rate. However, directly probing and quantifying the impact of stellar feedback on the neutral gas of the nascent clouds is challenging. We present an investigation doing exactly that toward the RCW 79 HII region using the ionized carbon line at 158 $\mu$m ([CII]) from the FEEDBACK Legacy Survey. We combine this data with information on the dozen ionizing O stars responsible for the evolution of the region, and observe in [CII] for the first time both blue- and red-shifted mostly neutral high-velocity gas which reaches velocities up to 25 km s$^{-1}$ relative to the bulk emission of the molecular cloud. This high-velocity gas mostly contains neutral gas and partly forms a fragmented shell, similar to recently found shells in a few Galactic HII regions. However, this shell does not account for all of the observed neutral high-velocity gas. We also find high-velocity gas streaming out of the nascent cloud through holes and obtain a range of dynamical timescales below 1.0 Myr for the high-velocity gas which is well below the 2.3$\pm$0.5 Myr age of the OB cluster. This suggests a different scenario for the evolution of RCW 79, where the high-velocity gas is not solely stemming from a spherical expanding bubble, but also from gas recently ablated at the edge of the turbulent molecular cloud into the surrounding interstellar medium through low-pressure holes or chimneys. The resulting mass ejection rate estimate for the cloud is 0.9-3.5$\times$10$^{-2}$ M$_{\odot}$~yr$^{-1}$, which leads to short erosion timescales, i.e. $<$5 Myr, for the nascent molecular cloud. Elongated anisotropic Gaussian filters are used for the orientation estimation of fibers. In cases where computed tomography images are noisy, roughly resolved, and of low contrast, they are the method of choice even if being efficient only in virtual 2D slices. However, minor inaccuracies in the anisotropic Gaussian filters can carry over to the orientation estimation. Therefore, this paper proposes a modified algorithm for 2D anisotropic Gaussian filters and shows that this improves their precision. Applied to synthetic images of fiber bundles, it is more accurate and robust to noise. Finally, the effectiveness of the approach is shown by applying it to real-world images of sheet molding compounds.

We present combined experimental and numerical work on light-matter interactions at nanometer length scales. We report novel numerical simulations of near-field infrared nanospectroscopy that consider, for the first time, detailed tip geometry and have no free parameters. Our results match published spectral shapes of amplitude and phase measurements even for strongly resonant surface phonon-polariton (SPhP) modes. They also verify published absolute scattering amplitudes for the first time. A novel, ultrabroadband light source enables near-field amplitude and phase acquisition into the far-infrared spectral range. This allowed us to discover a strong SPhP resonance in the polar dielectric SrTiO3 (STO) at approximately 24 micrometer wavelength of incident light.

We explore the sensitivity of Tevatron data to heavy new physics effects in differential dijet production rates using the SMEFT in light of the fact that consistent and conservative constraints from the LHC cannot cover relatively low cutoff scales in the EFT. In contrast to the results quoted by the experimental collaborations and other groups, we find that, once consistency of the perturbation expansion is enforced and reasonable estimates of theoretical errors induced by the SMEFT series in $\frac{E}{\Lambda}$ are included, there is no potential to constrain four-quark contact interactions using Tevatron data. This shows the general difficulty of constraining physics model-independently using fairly imprecise measurements, limited by low luminosity and/or systematic errors inherent to the precision of the detectors., Comment: 13 pages, 3 figures

The crystal structure of solid-state matter greatly affects its electronic properties. For example in multilayer graphene, precise knowledge of the lateral layer arrangement is crucial, since the most stable configurations, Bernal and rhombohedral stacking, exhibit very different electronic properties. Nevertheless, both stacking orders can coexist within one flake, separated by a strain soliton that can host topologically protected states. Clearly, accessing the transport properties of the two stackings and the soliton is of high interest. However, the stacking orders can transform into one another and therefore, the seemingly trivial question how reliable electrical contact can be made to either stacking order can a priori not be answered easily. Here, we show that manufacturing metal contacts to multilayer graphene can move solitons by several $\mu$m, unidirectionally enlarging Bernal domains due to arising mechanical strain. Furthermore, we also find that during dry transfer of multilayer graphene onto hexagonal Boron Nitride, such a transformation can happen. Using density functional theory modeling, we corroborate that anisotropic deformations of the multilayer graphene lattice decrease the rhombohedral stacking stability. Finally, we have devised systematics to avoid soliton movement, and how to reliably realize contacts to both stacking configurations.

We report on the first implementation of ultrafast near field nanoscopy carried out with the transient pseudoheterodyne detection method (Tr-pHD). This method is well suited for efficient and artifact free pump-probe scattering-type near-field optical microscopy with nanometer scale resolution. The Tr-pHD technique is critically compared to other data acquisition methods and found to offer significant advantages. Experimental evidence for the advantages of Tr-pHD is provided in the Near-IR frequency range. Crucial factors involved in achieving proper performance of the Tr-pHD method with pulsed laser sources are analyzed and detailed in this work. We applied this novel method to time-resolved and spatially resolved studies of the photo-induced effects in the insulator-to-metal transition system vanadium dioxide with nanometer scale resolution., Comment: 37 pages, 6 figures

We investigated emission and propagation of polaritons in a two dimensional van der Waals material hexagonal boron nitride (hBN). Our specific emphasis in this work is on hyperbolic phonon polariton emission that we investigated by means of scattering-type scanning near-field optical microscopy. Real-space nano-images detail how the polaritons are launched in several common arrangements including: light scattering by the edges of the crystal, metallic nanostructures deposited on the surface of hBN crystals, as well as random defects and impurities. Notably, the scanned tip of the near-field microscope is itself an efficient polariton launcher. Our analysis reveals that the scanning tips are superior to other types of emitters we have investigated. Furthermore, the study of polariton emission and emission efficiency may provide insights for development of polaritonic devices and for fundamental studies of collective modes in other van der Waals materials., Comment: 19 pages, 3 figures
