Over the last decade, supercell simulations and observations with ever-increasing resolution have provided new insights into the vortex-scale processes of tornado formation. This article incorporates these and other recent findings into the existing three-step model by adding an additional fourth stage. The goal is to provide an updated and clear picture of the physical processes occurring during tornadogenesis. Specifically, we emphasize the importance of the low-level wind shear and mesocyclone for tornado potential, the organization and interaction of relatively small-scale pretornadic vertical vorticity maxima, and the transition to a tornado-characteristic flow. Based on these insights, guiding research questions are formulated for the decade ahead. SIGNIFICANCE STATEMENT: This article provides a nontechnical overview of how tornadoes form. Sequentially, the most important processes include the initial creation of rotating updrafts, the development of disorganized patches of rotation at the surface, the organization of these patches into a more defined, symmetric vortex, and the final transition into a fully developed tornado in which air turns abruptly upward very near the surface. The COVID-19 pandemic has prompted a renewed interest in intergenerational letter and email writing. Evidence shows that expressive writing, including letter writing, has a number of benefits including improved literacy and perceived well-being, and it can also facilitate a deep connection with another person. This scoping review provides an overview of the existing research on letter and email writing between different age cohorts. Of the 471 articles retrieved from Scopus, CINAHL, PsycINFO, MEDLINE, Academic Search Premier, and Web of Science, 17 studies met the inclusion criteria and were critically appraised and synthesized in this review. The studies were grouped into two themes according to their stated aims and outcomes: (a) studies exploring changes in perceptions, and (b) studies relating to skills development and bonds. The results showed a range of benefits for intergenerational letter writers, from more positive perceptions of the other age group, through improved writing skills and subject knowledge, to forming intergenerational memories and bonds. The review also highlights some of the limitations of the current research and formulates recommendations for future studies in the fields of writing studies, intergenerational research, and educational gerontology.

The aim of the research is to assess the synchronization of business and financial cycles in EU countries, taking into account the division into “old” and “new” members of this group and the impact of the crisis phenomena of 2020–2022. The study used quarterly time series representing the dynamics of real GDP, credit to the private non-financial sector and main stock indices, which were subjected to a decomposition procedure. Cycle convergence was examined using spectral analysis and recursive correlation. As a result of the research, it was observed that EU countries differ in terms of the degree of impact of financial variables on the economic cycle. The group of “new” and “old” EU countries differs statistically significantly in terms of the average level of synchronization of the economic and stock market cycles. The crisis phenomena of 2020–2022 had a different impact on the degree of cycle synchronization. No clearly different reaction of the level of convergence was observed in countries geographically close to the armed conflict in Ukraine, with the exception of Poland.

Abstract Designing machines and equipment for post-harvest operations of agricultural products requires information about their physical properties. The aim of the work was to evaluate the possibility of introducing a new approach to predict the moisture content in bean and corn seeds based on measuring their dimensions using image analysis using artificial neural networks (ANN). Experimental tests were carried out at three levels of wet basis moisture content of seeds: 9, 13 and 17%. The analysis of the results showed a direct relationship between the wet basis moisture content and the main dimensions of the seeds. Based on the statistical analysis of the seed material, it was shown that the characteristics examined have a normal or close to normal distribution, and the seed material used in the investigation is representative. Furthermore, the use of artificial neural networks to predict the wet basis moisture content of seeds based on changes in their dimensions has an efficiency of 82%. The results obtained from the method used in this work are very promising for predicting the moisture content.

IntroductionRenal cell carcinoma (RCC) represents cancer originating from the renal epithelium and accounts for > 90% of cancers in the kidney. Prostate-specific membrane antigen (PSMA) is overexpressed in tumor-associated neovascular endothelial cells of many solid tumors, including metastatic RCC. Although studied in several small clinical studies, PSMA-based imaging and therapy have not been pursued rigorously in preclinical RCC. This study aimed to evaluate the preclinical performance of PSMA-based radiotheranostic agents in a relevant murine model.MethodsA PSMA-overexpressing murine cell line, PSMA+ RENCA, was developed by lentiviral transduction. PSMA-based theranostic agents, 68Ga-L1/177Lu-L1/225Ac-L1, were synthesized in high radiochemical yield and purity following our reported methods. Immunocompetent BALB/c mice were used for flank and orthotopic tumor inoculation. 68Ga-L1 was evaluated in small animal PET/CT imaging in flank and PET/MR imaging in orthotopic models. Cell viability studies were conducted for 177Lu-L1 and 225Ac-L1. Proof-of-concept treatment studies were performed using 225Ac-L1 (0, 37 kBq, 2 kBq × 37 kBq, 1 week apart) using PSMA+ RENCA in the flank model.ResultsCellular uptake of 68Ga-L1, 177Lu-L1, and 225Ac-L1 confirmed the specificity of the agents to PSMA+ RENCA cells rather than to RENCA (wt) cells, which are low in PSMA expression. The uptake in PSMA+ RENCA cells at 1 h for 68Ga-L1 (49.0% incubated dose [ID] ± 3.6%ID/million cells), 177Lu-L1 (22.1%ID ± 0.5%ID)/million cells), and 225Ac-L1 (4.1% ± 0.2% ID)/million cells), respectively, were higher than the RENCA (wt) cells (~ 1%ID–2%ID/million cells). PET/CT images displayed > 7-fold higher accumulation of 68Ga-L1 in PSMA+ RENCA compared to RENCA (wt) in flank implantation at 1 h. A twofold higher accumulation of 68Ga-L1 was observed in orthotopic tumors than in normal kidneys during 1–3 h postinjection. High lung uptake was observed with 68Ga-L1 PET/MR imaging 3 weeks after orthotopic implantation of PSMA+ RENCA due to spontaneous lung metastases. The imaging data were further confirmed by immunohistochemical characterization. 225Ac-L1 (0-37 kBq) displayed a dose-dependent reduction of cell proliferation in the PSMA+ RENCA cells after 48 h incubation; ~ 40% reduction in the cells with treated 37 kBq compared to vehicle (p < 0.001); however, no effect was observed with 177Lu-L1 (0–3700 kBq) up to 144 h postinoculation, suggesting lower efficacy of β-particle-emitting radiations in cellular studies compared to α-particle-emitting 225Ac-L1. Animals treated with 225Ac-L1 at 1 week posttumor inoculation in flank models displayed significant tumor growth delay (p < 0.03) and longer median survival of 21 days and 24 days for the treatment groups 37 kBq and 2 kBq × 37 kBq, respectively, compared to the vehicle group (12 days).ConclusionThe results suggest that a theranostic strategy targeting PSMA, employing PET and α-emitting radiopharmaceuticals, enabled tumor growth control and enhanced survival in a relevant immunocompetent murine model of RCC. These studies provide the rationale for clinical studies of PSMA-targeted theranostic agents in patients with RCC.

Imagery rescripting (IR) is a cognitive-experiential technique used for the treatment of various mental conditions through the re-working of aversive memories. It has been widely applied in psychotherapy to address Post-Traumatic Stress Disorder (PTSD) and personality, anxiety, and eating disorders. Despite usually being effective, IR's underlying mechanism has not been clarified yet. Consistently, different assumptions may correspond to different IR versions and outcomes. Among these versions, active IR (AIR) – meaning a rescripting where the patient is the active ‘rescriptor’ – seems to have particularly positive effects. So far, IR use has been almost exclusively limited to in vivo settings. But the increasing availability of Virtual Reality (VR) in the last few years has favored the in virtuo implementation of safe and effective psychological treatments. On these grounds – and given that virtual scenarios can offer the possibility to perform any necessary actions and even more than real ones – we hypothesize that implementing AIR in VR can be effective in treating psychological conditions. As a preliminary evaluation of this assumption, we systematically searched the literature and reviewed the studies concerning VR realizations of AIR addressing mental issues. Our literature analysis provides the first evidence supporting this hypothesis, yet urging further research and testing.

The current state of aquatic ecosystems was assessed for lakes in two river–lake systems: Święta Struga (Święte and Ocypelek) and Kałębnica (Czarne Południowe and Kałębie). Trophic states were determined using the Vollenweider, Nürnberg, and Carlson methods. Additionally, shoreline changes were analyzed for all lakes concerning their natural character, and physicochemical parameters were utilized as auxiliary elements in the assessment of the ecological state following the guidelines of Directive 2000/60/EC. Supplementary to these methods, the ecological evaluation of the littoral zone utilized meiobenthic organism grouping as an indicator. The results indicate significant differences among the studied reservoirs. Lake Święte is a reservoir bordering mesotrophy and eutrophy, with the ecological state of the littoral zone showing ongoing trophic development. Lake Ocypelek exemplifies a eutrophic reservoir undergoing progressive degradation. The lakes in the Kałębnica system also exhibit advanced trophic conditions, with Lake Kałębie being a eutrophic reservoir. Its shoreline has been significantly altered due to the development of the direct catchment area. In contrast, Lake Czarne Południowe serves as an example of a mesotrophic reservoir, with a substantially changed shoreline. Considering physicochemical elements and chlorophyll a concentration, the water quality for all examined lakes was found to be below the good category. The diagnosis of the condition of lake aquatic ecosystems requires an individualized approach to each of them. Therefore, employing various methods for their assessment, encompassing physico-chemical, biological, and morphological parameters, provides the opportunity to initiate potential remedial actions that will effectively contribute to their improvement. Conducting low-cost research involving a multidisciplinary approach to diagnose the state of aquatic ecosystems is becoming a crucial element in lake monitoring.

The objective of this paper was to determine the impact of EEG-biofeedback training on the motivation and efficiency of powerlifters during the bench press exercise in relation to the external load and the level of training. The study included 18 trained powerlifters who were divided into the intermediate (IG) and the advanced (AG) groups. EEG-biofeedback training was conducted every three days, lasting 27 minutes each time (5×3-minute intervals with recovery periods – lying on a bench – between them 4×3 minutes), and ended with a final EEG measurement in the second cycle of research. The repeated measures ANOVA showed intra group differences due to external loading for the FAI (Frontal Alpha Asymmetry) obtained in the EEG both before and after biofeedback training. In AG group analysis revealed significant differences between 65%1RM and 35%1RM. In the IG group between 35%1RM and 50, 65 and 80%1RM. One of the major variables influencing the efficiency of strength training, including bench press workouts, is the level of training. The more successfully an athlete uses motivation when exercising, the better their training, which translates into greater results and a lower chance of injury.

Abstract Cyclic high-dose testosterone administration, known as bipolar androgen therapy (BAT), is a treatment strategy for patients with metastatic castration-resistant prostate cancer (mCRPC). Here, we report the results of a multicenter, single arm Phase 2 study (NCT03554317) enrolling 45 patients with heavily pretreated mCRPC who received BAT (testosterone cypionate, 400 mg intramuscularly every 28 days) with the addition of nivolumab (480 mg intravenously every 28 days) following three cycles of BAT monotherapy. The primary endpoint of a confirmed PSA50 response rate was met and estimated at 40% (N = 18/45, 95% CI: 25.7–55.7%, P = 0.02 one-sided against the 25% null hypothesis). Sixteen of the PSA50 responses were achieved before the addition of nivolumab. Secondary endpoints included objective response rate (ORR), median PSA progression-free survival, radiographic progression-free survival (rPFS), overall survival (OS), and safety/tolerability. The ORR was 24% (N = 10/42). Three of the objective responses occurred following the addition of nivolumab. After a median follow-up of 17.9 months, the median rPFS was 5.6 (95% CI: 5.4–6.8) months, and median OS was 24.4 (95% CI: 17.6–31.1) months. BAT/nivolumab was well tolerated, resulting in only five (11%) drug related, grade-3 adverse events. In a predefined exploratory analysis, clinical response rates correlated with increased baseline levels of intratumoral PD-1 + T cells. In paired metastatic tumor biopsies, BAT induced pro-inflammatory gene expression changes that were restricted to patients achieving a clinical response. These data suggest that BAT may augment antitumor immune responses that are further potentiated by immune checkpoint blockade.

Relationships and Sex Education (RSE) in schools are predominantly heterocentric. Consequently, lesbian, gay, bisexual, transgender, and questioning young people have reported feeling excluded. This exclusion results in feelings of being "different" and "other," which in turn leads to further disengagement in the sex education classroom, contributing to poor sexual health literacy, greater risk of abusive relationships, and higher rates of sexually transmitted infections. A rapid review was undertaken to identify the impact of non-inclusive sex education. The review makes recommendations for policy and practice, which includes the provision of training courses to school teaching staff with an emphasis on inclusive RSE, appropriate online resources for lesbian, gay, bisexual transgender, and questioning (LGBTQ) young people, as well as offering 1:1 emotional health support for LGBTQ young people as they begin to question their sexual orientation.

The dynamic development of modern technologies allows the construction of devices that change the current approach to the design and inventory of buildings. The Terrestrial Laser Scanning (TLS) of large-scale objects improves the existing state’s documentation and the building reconstruction design phase, especially in the interdisciplinary Architecture, Engineering, and Construction sector(AEC) approach. Understanding these applications’ wide range of technical requirements and considerations is crucial in projects. This research aims to investigate the validity and quality of using TLS for the structural expertise of a metro tunnel. The analysis exploits and focuses on laser scanning technology applications for testing the analysis accuracy of monolithic concrete walls in a building of extended length. The study results indicate that the technological advancement in Building Information Modelling (BIM) presents extraordinary opportunities for AEC professionals to use laser scanning in a holistic, collaborative workflow based on 3D model-based design and data quality.

In this paper, a general framework for deal with uncertainties in each stage of consequence modeling is presented. In the first part of the framework, the sources of uncertainty are identified and confirmed by sensitivity analysis for the source term, dispersion, physical effects and consequence analysis. While the second part comprises an application of the fuzzy logic system to each step of the consequence modeling. The proposed procedure is verified by the case study for a liquefied natural gas (LNG) release on water. The results in terms of hazard zones are compared with calculations obtained using the Monte Carlo method and with experimental data. The fuzzy logic approach provides less uncertain and more precise results in comparison to the deterministic approach.

Abstract Fuzzy logic models are increasingly used to control simple and complex devices, as well as entire operating systems. In this study, a fuzzy logic model was applied to assess the performance a boom stabilization system in a field sprayer. The model was tested on a field sprayer with a trapezoid system for stabilizing the sprayer boom with a length of 21 m. Measuring cables for registering the displacement of the boom's terminal segments (right and left) in the vertical and horizontal plane were installed on the sprayer. The field sprayer was connected to a tractor. The model was based on two linguistic variables: "absolute displacement of the boom's terminal segments" and "boom stability index". It was assumed that the sprayer boom was stable when the displacement of the boom's terminal segments did not exceed 0.25% of boom length. The study demonstrated that the proposed model can be reliably used to assess boom stability in real time (during field operations). The time required to achieve boom stability was more than 2.5 times shorter in the vertical than in the horizontal plane, which can be attributed mainly to the structure of the stabilization system. The proposed model is universal, and it can be applied to evaluate other boom stabilization systems in field sprayers.

Food neophobia, defined as fear or aversion to eating new or unfamiliar foods, is a significant challenge, especially in the context of preschool children. In the scientific literature, this phenomenon is often described as a natural developmental stage, but its severity and impact on preferences and eating patterns still raise many questions. The purpose of the present study was to assess the prevalence of food neophobia in children aged 1 to 6 years and to analyze its relationship with eating habits, preferences, and eating patterns. The study was conducted using a proprietary questionnaire and validated research tools such as the Child Feeding Scale (MCH-FS) and Food Neophobia Scale (FNS). The study included 345 children, of whom 59.1% were observed to be at significant risk for food neophobia. The results of the study suggest that food neophobia is not a common phenomenon in children aged 1–2 years but becomes more pronounced later in childhood. Another important finding was that food neophobia shows a stronger association with established eating patterns than with individual taste preferences. Considering these results, this phenomenon should be considered not only as a natural part of child development, but also as a potential indicator of eating disorders that may require intervention. These findings underscore the need for further research that could deepen the understanding of the mechanisms governing food neophobia and its long-term consequences for child health.

Some species of the Gentianaceae family are a valuable source of secondary metabolites. However, the phytochemical knowledge of some of these species remains insufficient. Therefore, this work focused on the isolation of the two main secondary metabolites in the methanolic extract from a Gentiana capitata cell suspension using preparative HPLC and the determination of their structure using UHPLC–DAD–IT–MS/MS and NMR methods. Their content in the methanolic extract was quantified using a previously validated HPLC method. The toxicity of the extract and two isolated compounds was also tested on the PC-12 cell line. The structures of the main secondary metabolites were identified as isosaponarin and 3,7,8-Trimethoxy-9-oxo-9H-xanthen-1-yl 6-O-β-D-ribopyranosyl-β-D-allopyranoside by comparing the UHPLC–DAD–IT–MS/MS and NMR results with the literature data. The content of isosaponarin was determined to be 0.76 ± 0.04%, and the content of 3,7,8-trimethoxy-9-oxo-9H-xanthen-1-yl 6-O-β-D-ribopyranosyl-β-D-allopyranoside was found to be 0.31 ± 0.02% in the dry extract. Additionally, a two-fold increase in the viability of the PC-12 cell line was observed compared to the control after treatment with the methanolic extract at a concentration of 500 µg/mL. These results suggest the potential use of G. capitata cell suspension methanolic extract as a new source of isosaponarin and 3,7,8-trimethoxy-9-oxo-9H-xanthen-1-yl 6-O-β-D-ribopyranosyl-β-D-allopyranoside, highlighting their lack of toxicity to the PC-12 (rat pheochromocytoma) cell line.

Backgrounds/Objectives: Parotid gland tumors (PGTs) with parapharyngeal space (PPS) involvement have a specific clinical course and they can be a great challenge for surgeons, especially due to more difficult approaches and the risk of serious complications. The aim of this study is to present the characteristics of PGTs with PPS involvement. Methods: Retrospective, multicenter analysis of 1954 primary PGTs from 5 years (2017–2021) was performed. Comparative analysis was performed between groups with and without PPS involvement and included the following clinical and histopathological data: age, sex, place of residence, tumor size, FNAC result, percentage of malignant tumors, histological diagnosis, radicality of resection, and postoperative facial nerve (FN) dysfunction. Results: PPS involvement was found in 114 patients (5.83%). Secondary tumors affecting the deep lobe or the entire gland were predominant (46 and 60 cases, respectively). In a univariate analysis of tumors with and without PPS involvement, statistically significant differences were found in their size > 4 cm (12.97% vs. 37.72%), percentage of malignant tumors (7.12% vs. 17.55%), incidence of Warthin Tumors (WTs) (43.58% vs. 24.56%), percentage of R1 resection (5.53% vs. 12.50%), and rate of FN paresis (17.15% vs. 53.34%). Multivariate analysis showed that tumors with PPS involvement were statistically significantly characterized by larger size (tumors > 4 cm were 2.9 times more frequent), 2 times less frequent occurrence of WTs, and 1.6 times higher risk of FN paresis. Conclusion: PGTs with PPS involvement show certain clinical and histological differences and require more complex surgical accesses. Therefore, they cannot be treated as "ordinary" tumors occupying the deep lobe.

Traumatic brain injury (TBI), Alzheimer’s disease (AD), and epilepsy share proposed mechanisms of injury, including neuronal excitotoxicity, cascade signaling, and activation of protein biomarkers such as tau. Although tau is typically present intracellularly, in tauopathies, phosphorylated (p-) and hyper-phosphorylated (hp-) tau are released extracellularly, the latter leading to decreased neuronal stability and neurofibrillary tangles (NFTs). Tau cleavage at particular sites increases susceptibility to hyper-phosphorylation, NFT formation, and eventual cell death. The relationship between tau and inflammation, however, is unknown. In this review, we present evidence for an imbalanced endoplasmic reticulum (ER) stress response and inflammatory signaling pathways resulting in atypical p-tau, hp-tau and NFT formation. Further, we propose tau as a biomarker for neuronal injury severity in TBI, AD, and epilepsy. We present a hypothesis of tau phosphorylation as an initial acute neuroprotective response to seizures/TBI. However, if the underlying seizure pathology or TBI recurrence is not effectively treated, and the pathway becomes chronically activated, we propose a “tipping point” hypothesis that identifies a transition of tau phosphorylation from neuroprotective to injurious. We outline the role of amyloid beta (Aβ) as a “last ditch effort” to revert the cell to programmed death signaling, that, when fails, transitions the mechanism from injurious to neurodegenerative. Lastly, we discuss targets along these pathways for therapeutic intervention in AD, TBI, and epilepsy.

The main goal of this research is to create nanocomposites based on unmodified and iron-modified (FeNP) montmorillonite (Mt) and carbon nanotubes (CNT) synthesized using the chemical vapor deposition method. The target area for the application of these materials is the creation of water treatment systems. This paper compares the efficiency of the CNT synthesis process on Mt before and after modification with FeNP of different concentrations and provides the characterization of the CNT microstructure and structure using different methods, such as scanning electron microscopy, high-resolution transmission electron imaging, and Raman spectroscopy. For initial verification of properties important for water purification, Mt+CNT and Mt+FeNP+CNT nanocomposites on a carbon nonwoven fabric (CF) are tested in this work. Incubation of the above-mentioned samples in a water–oil mixture reveals complex adsorption dynamics. The CF+Mt+FeNP+CNT sample shows a very good oil adsorption capacity due to its superhydrophobic and oleophilic properties.
