The aim of the study was to investigate local biocompatibility and systemic effects of nonwoven polylactide (PLA) matrices on blood and liver parameters after their subcutaneous implantation in Wistar rats.Materials and methods. Bioabsorbable fibrous PLA matrices were produced by electrospinning and had dimensions (10 × 10 mm², thickness of no more than 0.5 mm; fiber diameter in the matrix ~1 μm) appropriate for subcutaneous implantation in white laboratory rats. Polymer implants were sterilized in ethylene oxide vapor. Thirty days after the implantation of PLA matrices, local biocompatibility according to GOST ISO 10993-6-2011, cellular parameters (total leukocyte count, hemogram, erythrocyte count, hemoglobin concentration), and biochemical blood parameters (lactate concentration, alanine aminotransferase (ALT) and aspartate aminotransferase (AST) levels) were studied, and a standard histologic evaluation of the liver was performed.Results. PLA matrix samples were mild local irritants on a scale of 1–1.9 points according to GOST ISO 10993-6-2011 criteria 30 days after the subcutaneous implantation. The median density of distribution of multinucleated giant cells (MNGCs) in the connective tissue around and in PLA matrices was 1,500 (1,350; 1,550) per 1 mm² of a slice. Pronounced leukocytic reaction due to lymphocytosis was noted (an increase by 1.7 times compared with a sham-operated (SO) control group, р < 0.02). The absence of a significant neutrophil count in the blood revealed sterile inflammation proceeding in the subcutaneous tissue around the PLA materials. Normalization of hepatic cytolysis markers (ALT and AST activity) in the blood without pronounced changes in the structure of the liver and the number of binuclear hepatocytes was noted. These markers were increased in SO controls (ALT up to 123% and AST up to 142%, p < 0.001 compared with values in the intact group).Conclusion. Nonwoven PLA matrices are biocompatible with subcutaneous tissue, undergo bioresorption by MNGCs, and have a distant protective effect on the functional state of the liver in laboratory animals. Hypotheses on the detected systemic effect during subcutaneous implantation of PLA matrices were discussed; however, specific mechanisms require further study.

Aim. To identify early morphological changes in the heart and aorta of rats with experimental metabolic syndrome induced by a high-fat and high-carbohydrate diet (HFHCD).Materials and methods. The study was carried out on male Wistar rats. The animals were divided into two groups: a control group (n = 10) and an experimental group (n = 10). The rats from the control group were fed with a standard laboratory diet. The rats from the experimental group received HFHCD for 12 weeks. Body weight, blood pressure (BP), and individual parameters of carbohydrate and lipid metabolism were assessed in the rats. A histologic examination of the heart and aorta in the animals was performed.Results. Feeding rats with HFHCD led to an increase in body weight, elevation of BP, obesity, hyperglycemia, and triglyceridemia. The histologic examination of the heart in the rats of the experimental group showed signs of vascular disease, lipomatosis, and focal myocardial degeneration. Lipid accumulation in the cells of the media, hyperplasia of adipocytes in the adventitia, and depletion and fragmentation of the elastic lamina were revealed in the aortic wall of the rats receiving HFHCD.Conclusion. The study indicated that HFHCD is an effective way to model metabolic syndrome. Structural disorders in the heart and aorta may be the mainstay for the development of cardiomyopathy and arterial hypertension in diet-induced metabolic syndrome.

The aim of the research is to study the possibility of using track-etched membrane, including track-etched membranes modified with cold plasma, followed by layering prenatal stem cells (PSC) on the material surface in surgical treatment of bullous keratopathy (BK). Materials and methods. The track membranes made of polyethylene terephthalate were obtained by irradiating the polymeric film with the 40Ar+8ion beams and by chemical etching. The study was conducted on 16 rabbits (Sylvilagus bachmani), which after BK modelling were divided into 4 groups: the 1st group was a control group of 4 animals (4 eyes); the 2nd group was a group of 4 animals (4 eyes) into which were implanted TM; the 3rd group was a group of 4 animals (4 eyes) into which were implanted TM with PSC; the 4th group was a group of 4 animals (4 eyes) into which were implanted plasma modified TM with PSC. TM was obtained by irradiating the PET film with 40Ar + 8 ions and subsequent chemical etching. The eyes were enucleated for histological examination after 8 weeks from the start of the experiment. Results. As a result of the research, it was found that the implantation of TM with a preliminary layeringof human PSC promotes the growth of the fibroblast population in the cornea stroma and intensifies leukocyte (lymphocytes and eosinophilic granulocytes) infiltration as opposed to the implantation of PET TM without a cellular component. In addition, the implantation of TM contributes to a twofold decrease in the cornea edema induced by BK. Modification of TM with cold plasma did not affect the studied histomorphometric parameters. Conclusion. The implantation of TM based on PET during bullous keratopathy contributed to the development of the productive phase of infiltrative inflammation in the cornea of the eye. Pre-layering of human PSC reduced the severity of destructive changes in the rabbit cornea after BK modeling. The modification of TM by cold plasma did not affect the studied histomorphometric parameters.

Aim. This research studies the effect of a low-temperature atmospheric plasma and the subsequent γ-ray sterilization on topography and properties of track membranes (TM) based on polyethylene terephthalate (PET).Materials and methods. TM were obtained by irradiating a PET film with a 40Ar+8 ion beam and then by chemical etching in an aqueous solution of 1.5N NaOH. Modification of the membrane surface was carried out by exposure to an atmospheric low-temperature plasma. The gamma radiation of the radionuclide 60Со with the dosages of 1kGy (SI) and 10 kGy (SI) was used to sterilize the membranes. In vitro studies of the TM biocompatibility were performed by using a culture of prenatal stromal cells isolated from a lung of an 11-week human embryo and maintained ex vivo.Results. It has been established that the treatment of the membranes with the low-temperature atmospheric plasma leads to an increase in the roughness and hydrophilization of the TM surface. The change in the physical-chemical state of the TM surface as a result of the exposure of cold plasma and subsequent sterilization had practically no effect on the morphofunctional state of the culture of human prenatal stromal cells. In vitro tests on the TM cellular-molecular biocompability with a short-term culture of in vitro fibroblast-like cells have made it possible to indicate their relative bioinerticity with respect to human stromal cells. The conclusion is made about the relative bioinerticity of TM and the proposed regimes for their sterilization with respect to the culture of human stromal cells, the prospects for further research in applying the material to the areas of surgical practice (cardiology, ophthalmology).

A state of titanium oxide and oxynitride coatings on L316 steel has been studied before and after their contact with model biological fluids. Electrokinetic investigation in 1 mmol potassium chloride showed significant (more than 10 times) fall of magnitude of electrostatic potential of thin (200–300 nm) titanium films at pH changing in the range of 5–9 units during 2 h. Nevertheless, zeta-potential of all samples had negative charge under pH > 6.5. Long-term (5 weeks) contact of samples with simulated body fluid (SBF) promoted steel corrosion and titanium oxide and oxynitride films dissolution. On the other hand, sodium and chloride ions precipitation and sodium chloride crystals formation occurred on the samples. Of positive fact is an absence of calcification of tested artificial surfaces in conditions of long-term being in SBF solution. It is supposed decreasing hazard of fast thrombosis and loss of materials functional properties. According to in vitro experiment conducted, prospective biocompatibility of materials tested before and after their contact with SBF lines up following manner: Ti–O–N (1/3) > Ti–O–N (1/1), TiO2 > Steel. It may be explained by: 1) the corrosion-preventive properties of thin titanium oxide and oxynitride films;2) a store of surface negative charge for Ti–O–N (1/3) film; 3) minor augmentation of mass and thickness of titanium films connected with speed of mineralization processes on the interface of solution/solid body. At the same time, initial (before SBF contact) differences of samples wettability became equal. Modifying effect of model biological fluids on physicochemical characteristics of materials tested (roughness enhancement, a reduction or reversion of surface negative potential, sharp augmentation of surface hydrofilicity) should took into account under titanium oxide and oxynitride films formation and a forecast of their optimal biological properties as the materials for cardiovascular stents.

Very little research is devoted to the study of communication systemic immunological changes and local immunological changes in the tissue of the thyroid in autoimmune thyrophaties. The goal of the research was to establish immunologic and morphologic predictors of clinical course and functional outcomes of Graves’s disease (GD).Material and methods. We examined 45 patients with GD (14 men and 31 women) aged 18–55 years (47.0 (35.0–53.0)) years, matching the following criteria: a verified diagnosis GD, accommodation in Tomsk oblast and the Tomsk not less than 10 years, the patient's consent for the study. The control group included 30 people, with an average age of (45.3 ± 5.6) years and was comparable by age and sex with GD patients. The study included: definition of the standard for GD hormonal and serological profile (free T4, free T3, and TSH, antibodies to TPO, antibodies to the TSH receptor), the cultivation of mononuclear leukocytes in complete culture medium within 24 hours with subsequent determination of interleukins concentrations (IL-2, IL-4, TNF-alpha) in culture medium using enzyme-linked immunosorbent assay, determination of the number of blood cells carrying membrane-bound receptors to IL-2, IL-4, TNF-R1 by flow laser cytometry on cytometer BD FACS Canto tmΙΙ (USA) using labeled monoclonal antibody, the standard postoperative histological examination of thyroid gland tissue samples and immunohistochemical detection in samples of thyroid tissue receptors to IL-2, IL-4, TNF-alpha.Results. On the basis of the received results it is possible to allocate 2 clinical-immunologic and morphologic cluster of autoimmune hyperthyroidism syndrome in patients with clinical diagnosis of GD. The first includes the formation of 1 and 2 histological options GD (minimally expressed monocytes infiltration, lack of oncocytic transformation of thyroid epithelium), with some clinical characteristics (persistent and pronounced hyperthyroidism, the large size of goiter, higher titer of receptor TSH-antibody and smaller TPO-antibody, diffuse nature of the lesion), and the second – is represented by 3d histological option with oncocytic restructuring of follicular epithelium, expressed the monocyte/macrophage infiltration with the TNF-RI expression and clinical patterns, including: the older age group of patients, the smaller size of goiter, the emergence of “pseudo nodes” ultrasound, reflecting the presence of lymphoid follicles-infiltrates, more lenient for hyperthyroidism. Probably, that the 2nd cluster is a combination of “classical GD” with autoimmune thyroiditis, however, a set of clinical and laboratory-instrumental signs led to the fact that these patients were in the group of GD patients.Conclusion. This approach (combining study of indicators of systemic and local tissue-specific autoimmune inflammation) is a promising from the point of view of separate parts coverage in the autoimmune thyroids diseases pathogenesis, approximating researchers to develop immune based therapeutic technologies.

The complex immunologic estimation of inflammatory reactions in bronchial contents at 30 patients with various phenotypes of a serious bronchial asthma is spent. It is established that the population structure of lymphocytes and cytokines a profile of the induced phlegm reflects expression and character of inflammatory disturbances in respiratory tracts that will allow to use the given non-invasive method for revealing of molecular markers of serious forms of a bronchial asthma. At the serious terapevticheskirefractory form of a bronchial asthma «brittle» a phenotype in respiratory tracts total СD4+ -, СD19+ -cell and concentration interleukina-4 against depression interleukina-10 is enlarged. At the serious form of a bronchial asthma of a phenotype «the chronic asthma with the fixed bronchial obstruction» in a bronchial secret prevails CD8+ -lymphocytes, concentration interleukina-8, the transforming growth factor-β increases.

In the present research the complex cytologic and biochemical estimation of a condition induced sputum at 19 patients with the heavy therapeutic-sensitive form of a bronchial asthma and 18 patients with chronic obstructive disease of lungs is lead. Dominating morphological and biochemical markers of an asthma became the raised number of macrophages, lymphocytes, and eosinophils at decrease in functional activity neutrophil elastase and increases activation properties. Progressing of destructive and fibrous processes at patients with heavy form chronic obstructive pulmonary disease occurs on a background active neutrophil and lymphocytes reactions in a mucous membrane of bronchial tubes that proves to be true high activity elastase and falling of functioning proteinase inhibitor.

The inflammatory-reparative reactions of sclera after different injury were studied. In animals of the 1st group carried out conjunctivotomy and surgical diathermy. In animals of the 2nd group carried out conjunctivotomy. Biomicroscopy, histologic and morphometrical studies were carried out. The results demonstrate surgical diathermy of sclera leads to intensification of local inflammatory reaction and is a risk factor of excessive regeneration in glaucoma filtration surgery.

This study aims to determine the effectiveness of administering 80 ppm nitric oxide in reducing kidney injury, mitochondrial dysfunction and regulated cell death in kidneys during experimental perfusion. Twenty-four sheep were randomized into four groups: two groups received 80 ppm NO conditioning with 90 min of cardiopulmonary bypass (CPB + NO) or 90 min of CPB and hypothermic circulatory arrest (CPB + CA + NO), while two groups received sham protocols (CPB and CPB + CA). Kidney injury was assessed using laboratory (neutrophil gelatinase-associated lipocalin, an acute kidney injury biomarker) and morphological methods (morphometric histological changes in kidney biopsy specimens). A kidney biopsy was performed 60 min after weaning from mechanical perfusion. NO did not increase the concentrations of inhaled NO2 and methemoglobin significantly. The NO-conditioning groups showed less severe kidney injury and mitochondrial dysfunction, with statistical significance in the CPB + NO group and reduced tumor necrosis factor-α expression as a trigger of apoptosis and necroptosis in renal tissue in the CPB + CA + NO group compared to the CPB + CA group. The severity of mitochondrial dysfunction in renal tissue was insignificantly lower in the NO-conditioning groups. We conclude that NO administration is safe and effective at reducing kidney injury, mitochondrial dysfunction and regulated cell death in kidneys during experimental CPB.
