Pump–probe experiments with subfemtosecond resolution are the key to understanding electronic dynamics in quantum systems. Here we demonstrate the generation and control of subfemtosecond pulse pairs from a two-colour X-ray free-electron laser. By measuring the delay between the two pulses with an angular streaking diagnostic, we characterize the group velocity of the X-ray free-electron laser and show control of the pulse delay down to 270 as. We confirm the application of this technique to a pump–probe measurement in core-ionized para-aminophenol. These results reveal the ability to perform pump–probe experiments with subfemtosecond resolution and atomic site specificity.

The case study of the monumental complex of San Francesco le Moniche, built in the ancient Norman county Aversa, in northern Campania (Italy), is analyzed here. The cultural heritage of the complex and the additional value associated with geoheritage (building stones and underground extraction cavities) have been highlighted. The building stratification of the complex was reconstructed based on documentary sources, including historical cartography and photographic documentation of postcards from the early 1900s. It began around 1200 and ended in the 20th century; part of the original citadel was dismantled and incorporated into the urban fabric in the Fascist era. The building stratification of the complex was also read from the distribution and architecture of the existing underground cavities from which the building stones were extracted. The subsoil, reconstructed on the basis of geological and geophysical data, is made up of a tuff substrate, starting from approximately 5–6 m from the ground level, which represents the main building stone as well as being an important testimony to the long volcanological history of northern Campania. Laser scanner surveys of the known and accessible cavities were carried out to obtain a three-dimensional view of the entire monumental complex and its underground spaces. The results provide a clear example of a geoheritage–cultural heritage interface which reminds us of the importance of an integrated approach in their valorization, specifically in urban areas. Additionally, the results of the study allowed us to improve the knowledge of the complex and the site, and provide useful tools for the planning of future targeted investigations.

Abstract Background Glioblastoma (GBM) stands as the most prevalent and aggressive form of adult gliomas. Despite the implementation of intensive therapeutic approaches involving surgery, radiation, and chemotherapy, Glioblastoma Stem Cells contribute to tumor recurrence and poor prognosis. The induction of Glioblastoma Stem Cells differentiation by manipulating the transcriptional machinery has emerged as a promising strategy for GBM treatment. Here, we explored an innovative approach by investigating the role of the depolarized resting membrane potential (RMP) observed in patient-derived GBM sphereforming cell (GSCs), which allows them to maintain a stemness profile when they reside in the G0 phase of the cell cycle. Methods We conducted molecular biology and electrophysiological experiments, both in vitro and in vivo, to examine the functional expression of the voltage-gated sodium channel (Nav) in GSCs, particularly focusing on its cell cycle-dependent functional expression. Nav activity was pharmacologically manipulated, and its effects on GSCs behavior were assessed by live imaging cell cycle analysis, self-renewal assays, and chemosensitivity assays. Mechanistic insights into the role of Nav in regulating GBM stemness were investigated through pathway analysis in vitro and through tumor proliferation assay in vivo. Results We demonstrated that Nav is functionally expressed by GSCs mainly during the G0 phase of the cell cycle, suggesting its pivotal role in modulating the RMP. The pharmacological blockade of Nav made GBM cells more susceptible to temozolomide (TMZ), a standard drug for this type of tumor, by inducing cell cycle re-entry from G0 phase to G1/S transition. Additionally, inhibition of Nav substantially influenced the self-renewal and multipotency features of GSCs, concomitantly enhancing their degree of differentiation. Finally, our data suggested that Nav positively regulates GBM stemness by depolarizing the RMP and suppressing the ERK signaling pathway. Of note, in vivo proliferation assessment confirmed the increased susceptibility to TMZ following pharmacological blockade of Nav. Conclusions This insight positions Nav as a promising prognostic biomarker and therapeutic target for GBM patients, particularly in conjunction with temozolomide treatment.

Abstract Environmental and physiological situations can challenge the balance between protein synthesis and folding capacity of the endoplasmic reticulum (ER) and cause ER stress, a potentially lethal condition. The unfolded protein response (UPR) restores ER homeostasis or actuates programmed cell death (PCD) when ER stress is unresolved. The cell fate determination mechanisms of the UPR are not well understood, especially in plants. Here, we integrate genetics and ER stress profiling with natural variation and quantitative trait locus analysis of 350 natural accessions of the model species Arabidopsis thaliana. Our analyses implicate a single nucleotide polymorphism to the loss of function of the general PCD regulator BON-ASSOCIATED PROTEIN2 (BAP2) in UPR outcomes. We establish that ER stress-induced BAP2 expression is antagonistically regulated by the UPR master regulator, inositol-requiring enzyme 1 (IRE1), and that BAP2 controls adaptive UPR amplitude in ER stress and ignites pro-death mechanisms in conditions of UPR insufficiency.

Attosecond physics enables the study of ultrafast coherent electron dynamics in matter upon photoexcitation and photoionization, revealing spectacular effects such as hole migration and coherent Auger dynamics in molecules. In the photoionization scenario, there has been a strong focus on probing the physical manifestations of internal quantum coherence within the individual parent ion and photoelectron systems. However, quantum correlations between these two subsystems emerging from attosecond photoionization events have thus far remained much more elusive. In this work, we design theoretically and model numerically a direct probe of quantum entanglement in attosecond photoionization in the form of a Bell test. We simulate from first principles a Bell test protocol for the case of noble gas atoms photoionized by ultrashort, circularly polarized infrared laser pulses in the strong-field regime predicting robust violation of the Bell inequality. This theoretical result paves the way for the direct observation of entanglement in the context of ultrafast photoionization of many-electron systems. Our work provides a novel perspective on attosecond physics directed toward the detection of quantum correlations between systems born during attosecond photoionization and unraveling the signatures of entanglement in ultrafast coherent molecular dynamics, including in the chemical decomposition pathways of molecular ions.

The contribution of sediment transport and accumulation to soil formation was investigated in an area characterized by continental sedimentary activity since the Late Pleistocene. The area was the north-eastern portion of the large Quaternary graben represented by the Campania Plain, which is rimmed to the north–east–south by the Mesozoic carbonate Apennine nappes. The plain was filled mainly by products generated by eruptions from the Phlegrean Fields, which were also distributed on the slopes bordering the plain and remobilized toward the adjacent surfaces. Five sites were selected in the area in question. They were studied using morphological features and pertinent characteristics of the mineral soil fraction >2.0 mm, such as their volume and lithological description. Soils were compared to selected lithostratigraphic sequences characterizing the studied area, which were collected from literature and reinterpreted in pedological keys. The results showed that soils derived from the emplacement of Phlegrean primary volcanic materials, such as Campania Ignimbrite (~39–40 ky B.P.) and Neapolitan Yellow Tuff (~15 ky B.P.), with the related weathering products, and from volcanic materials reworked and transported by alluvial/colluvial episodes. The latter formed contrasting soil horizons which, differing in both rock fragment content and lithological composition, testified to the presence of lithological discontinuities. The formation of the horizons in question interrupted the genetic sequence derived from the in situ alteration of the volcanic substrata, suggesting that processes of transport and redistribution of sediments from the adjacent mountain slopes contributed to soil formation. The comparison of the pedostratigraphies with the lithostratigraphic sequences indicated a strong relation between geomorphic and pedogenetic events.

Schistosomiasis is a major neglected parasitic disease that affects more than 240 million people worldwide caused by Platyhelminthes of the genus Schistosoma. The treatment of schistosomiasis relies on the long-term application of a single safe drug, praziquantel (PZQ). Unfortunately, PZQ is very effective on adult parasites and poorly on larval stage and immature juvenile worms; this can partially explain the re-infection in endemic areas where patients are likely to host parasites at different developmental stages concurrently. Moreover, the risk of development of drug resistance because of the widespread use of a single drug in a large population is nowadays a serious threat. Hence, research aimed at identifying novel drugs to be used alone or in combination with PZQ is needed. Schistosomes display morphologically distinct stages during their life cycle and epigenetic mechanisms are known to play important roles in parasite growth, survival, and development. Histone deacetylase (HDAC) enzymes, particularly HDAC8, are considered valuable for therapeutic intervention for the treatment of schistosomiasis. Herein, we report the phenotypic screening on both larvae and adult Schistosoma mansoni stages of structurally different HDAC inhibitors selected from the in-house Siena library. All molecules have previously shown inhibition profiles on human HDAC6 and/or HDAC8 enzymes. Among them we identified a quinolone-based HDAC inhibitor, NF2839, that impacts larval and adult parasites as well as egg viability and maturation in vitro. Importantly, this quinolone-based compound also increases histone and tubulin acetylation in S. mansoni parasites, thus representing a leading candidate for the development of new generation anti-Schistosoma chemotherapeutics.

In Campania (southern Italy), the widespread presence of anthropogenic cavities in the subsoil of the Neapolitan and Caserta provinces is well known. In these towns, the underground quarrying activities were performed for centuries to extract volcanic tuffs for buildings. The urban developments have sealed many signals of the presence of cavities and their real extent is almost unknown, thus representing a geological hazard and contributing to the subsoil instability of many places. This contribution will show the main cavity typologies recognized across an area north of Naples according to the geological characteristics of the subsoil. The main aim of the study is the cataloging of the cavities and the analysis of the city subsoil as their presence may easily trigger the collapse of the shallow or deeper soils. Moreover, the recognition and sustainable reuse of cavities contributes to enhancing the cultural and touristic promotion of a territory. In this study, a database framework was elaborated that includes all the possible architectural, geological and geotechnical elements of the cavities. Data were managed in a GIS environment in order to provide a useful tool for monitoring and managing the cavities for risk mitigation and tourism enhancement.

The harvest of the edible sea urchin Paracentrotus lividus is intensively practiced in some regions of the Western Mediterranean Sea. The removal of the largest individuals can determine an overall reduction in population size and a size class truncation that can lead to a drastic drop the self-sustenance. The aim of this study is to evaluate the variability of the population reproductive potential across 5 years in one of the main harvest hotspots of Sardinia (Western Mediterranean Sea). The breeding stock consists of commercial and under-commercial size individuals which were sampled on a monthly basis to estimate their GonadoSomatic Index (GSI) and the Individual Gamete Output (IGO). In addition, the reproductive potential of the population—Total Gamete Output (TGO)—was calculated across the 5-year period in relation with the variation of the density of the breeding stock. During the last year, the reproductive potential was also estimated in a well-conserved population of a nearby Marine Protected Area. No significant variability in GSI and IGO was found over the 5 years nor when compared with the ones of protected population in the last year. However, the intensive harvest drastically rescaled the population body-size: although density of the commercial size class remained low, density of the under-commercial size-class halved from the beginning to the end of the study. Accordingly, the proportional decrease of their gamete output contribution led to a 40% loss of the reproductive potential of the whole population in the 5-year period. Interestingly, despite the loss of reproductive potential due to the decrease of the breeding stock density, the average values of IGO slightly increased across the years leading to the highest Annual Gamete Output (AGO) during the fourth year of sampling. This positive pattern could suggest a mechanism of reproductive investments of the survivors in terms of gonad production rate or increase in spawning intensity. This work provides evidence of the direct effect of size-selective harvesting on the rapid loss of population self-sustenance. Furthermore, it lays new prospective for future research of the indirect effects of the rescaling population body-size in functional traits of the sea urchin P. lividus and that could become important for both, sustainable exploitation and ecosystem conservation management.

BackgroundLung cancer remains the leading cause of cancer-related death worldwide. Targeted therapies with tyrosine kinase inhibitors (TKIs) result in improvement in survival for non-small cell lung cancer (NSCLC) with activating mutations of the epidermal growth factor receptor (EGFR). Unfortunately, most patients who initially respond to EGFR-TKI ultimately develop resistance to therapy, resulting in cancer progression and relapse. Combination therapy is today a common strategy for the treatment of tumors to increase the success rate, improve the outcome and survival of patients, and avoid the selection of resistant cancer cells through the activation of compensatory pathways. In NSCLC, the phosphoinositide-3-kinase/protein kinase B (AKT)/mammalian target of rapamycin (mTOR) pathway has been heavily implicated in both tumorigenesis and the progression of disease.ObjectivesIn this study, we investigated the efficacy of a PI3K δ-sparing inhibitor, MEN1611, in models of NSCLC sensitive and resistant to EGFR inhibitors (erlotinib and gefitinib) with a wild-type PIK3CA gene.MethodsWe performed functional, biochemical, and immunohistochemistry studies.ResultsWe demonstrated good efficacy of MEN1611 in NSCLC devoid of PIK3CA gene mutations but with constitutive activation of the PI3K/AKT pathway and its synergistic effect with gefitinib both in vitro and in vivo.ConclusionsOverall, this preclinical study indicates that the inhibitor could be a candidate for the treatment of NSCLC with an erlotinib/gefitinib-resistant phenotype and constitutive activation of the PI3K/AKT pathway, a phenotype mimicked by our model system.
