High capacity immunoadsorbents (400–1000 mg of antibodies/g) were derived from oxidized cellulose preparations in the form of suspensions and porous spherical beads. For coupling proteins, the matrices were activated with NaIO 4 . The numbers of aldehyde groups did not diminish when the activated matrices were stored in water during one year. Porous spherical beads had good flow characteristics and were useful for column methods.

A method is described for immunizing mice with a protein-cellulose complex obtained by covalent coupling of antigenic protein molecules to suspended cellulose particles. Primary immunization of the animals with the complex led to a pronounced immune response persisting for 20–30 days. Subsequent administration of the same antigen in a soluble form resulted in extremely active antibody formation equal to or better than that induced with Freund's completed adjuvant. In BALB/c mice the antibody response was about ten times greater than in the C57BL/6 strain.

1. 1. A method for the investigation of histone-nucleic acid and histone-protein interactions in agarose is proposed. 2. 2. The formation of precipitin lines take place in those zones of the agarose where the reacting substances meet in suitable concentrations. Histones form 4 to 5 precipitin lines with nucleic acids and 6 lines with rabbit serum. 3. 3. The number, character and position of the precipitin lines depend on the heterogeneity and the nature of the reacting components. 4. 4. In conjunction with a reference system the proposed method can be used for the identification of nucleic acids, histones and other proteins.

A nature of non-specific immunoglobulins (Ig) forming in response to immunization was studied. These were suggested to be antibodies of very low affinity, reversibly reacting with the respective antigen. To detect such antibodies immunoexchange chromatography was employed. Non-specific Ig synthetized in vitro by spleen cells of normal rabbits and rabbits immunized with horse gamma-globulin (HGG) or egg albumin (EA) were applied on the column with specific immunoadsorbents (Sehadex G-200 or Sepharose 4B coupled with HGG). If antibodies to HGG reversibly reacting with the antigen were present in the sample applied, they should be eluted from the column later than normal Ig, or Ig obtained from EA-immunized rabbits. No differences were observed, however, when comparing the distribution of various non-specific Ig on the columns. It was concluded that the main bulk of antigen dependent non-specific Ig do not consist of antibodies reversibly reacting with the antigen.
