Plant growth-promoting rhizobacteria (PGPRs) represent a promising strategy for enhancing plant resilience and yields under drought-stress conditions. This study isolated and characterized PGPR from wheat rhizosphere soil in Egypt. Four PGPR strains were evaluated for an array of plant growth-promoting traits, including IAA production, biofilm formation, siderophore production, nitrogen fixation, ACC deaminase activity, phosphate solubilization, and antagonistic potential. Molecular identification via 16S rRNA sequencing classified three isolates (MMH101, MMH102, and MMH103) within the Bacillus genus and one isolate (MMH104) as Myroides sp. Greenhouse experiments examined the effects of PGPR inoculation on the drought-stressed Egyptian wheat cultivar, Gimmeza-9. Wheat plants inoculated with PGPR isolates showed dramatic improvements in growth parameters and stress tolerance indicators compared to non-inoculated controls when subjected to a 10-day drought period, with Bacillus rugosus (MMH101) inoculation resulting in increases of 61.8% in fresh biomass, 77.2% in dry biomass, 108.5% shoot length, and 134.9% root length. PGPR treatments also elevated the chlorophyll and proline content while reducing malondialdehyde levels. The findings demonstrate the effectiveness of PGPR inoculation in enhancing the morphology, physiology, and drought stress resilience of wheat. Potato tuber starch characteristic is influenced by the ratio of amylose to amylopectin, which is affected by the granule-bound starch synthase (GBSS) gene. GBSS gene expression was decreased in the leaves of the Desiree potato variety utilizing genome editing using CRISPR-Cas9. Constructs encoding Cas9 gene and sgRNAs targeting GBSS gene were inserted into plant leaves using Agrobacterium-mediated transformation delivery. The results obtained lines with mutations in GBSS genes accounting for 21% of regenerated shoots. The identification of mutations within one base pair of the used guide sequence provided further evidence of the considerable similarity between the target region around the protospacer adjacent motif (PAM) position and the used guide sequence. Transforming DNA into potato leaves produced mutants that lacked the Cas9 gene. Worldwide, potato (Solanum tuberosum L.) is one of the most essential agro-economically food crops. To ensure its presence throughout the year for food processors and to extend its shelf life, it should be stored in cold temperatures. Despite the benefits of storage at low temperatures, it causes undesirable phenomena; one of them is cold-induced sweetening, which reduces the quality and the commercial value of the potato tubers. In this study, the CRISPR technology ″Clustered Regularly Interspaced Short Palindromic Repeats″ has been used in potato cultivar Desiree via Agrobacterium-mediatied transformation to edit starch phosphorylase gene that is responsible for starch degradation in cold temperature storage, after regeneration steps. The regenerated plants were used in the screening of genome editing by Sanger sequencing and the ICE program. Screening results showed that Desiree 2 (DE2), Desiree 7 (DE7) and Desiree 27 (DE27) lines have mutations in sgRNA sequence that indicated a significant reduction in reduced sugar contents of tubers after cold storage for 90 and 120 days. The reduction mean values were 29.224, 35.078 and 35.862 in DE2, DE7 and DE27 respectively after 90 days and 83.212, 56.674 and 34.109 after 120 days without visible differences in plant morphology and tubers. Recent advances in genome sequencing have led to an increase in the number of sequenced genomes. However, the presence of repetitive sequences complicates the assembly of plant genomes. The LTR assembly index (LAI) has recently been widely used to assess the quality of genome assembly, as a higher LAI is associated with a higher quality of assembly. Here, we assessed the quality of assembled genomes of 1664 plant and algal genomes using LAI and reported the results as data repository called PlantLAI (https://bioinformatics.um6p.ma/PlantLAI). A number of 55 117 586 pseudomolecules/scaffolds with a total length of 988.11 gigabase-pairs were examined using the LAI workflow. A total of 46 583 551 accurate LTR-RTs were discovered, including 2 263 188 Copia , 2 933 052 Gypsy , and 1 387 311 unknown superfamilies. Consequently, only 1136 plant genomes are suitable for LAI calculation, with values ranging from 0 to 31.59. Based on the quality classification system, 476 diploid genomes were classified as draft, 472 as reference, and 135 as gold genomes. We also provide a free webtool to calculate the LAI of newly assembled genomes and the ability to save the result in the repository. The data repository is designed to fill in the gaps in the reported LAI of existing genomes, while the webtool is designed to help researchers calculate the LAI of their newly sequenced genomes. Mycobacterium tuberculosis (Mtb) complex, responsible for tuberculosis (TB) infection, continues to be a predominant global cause of mortality due to intricate host-pathogen interactions that affect disease progression. MicroRNAs (miRNAs), essential posttranscriptional regulators, have become pivotal modulators of these relationships. Recent findings indicate that miRNAs actively regulate immunological responses to Mtb complex by modulating autophagy, apoptosis, and immune cell activities. This has resulted in increased interest in miRNAs as prospective diagnostic indicators for TB, especially in differentiating active infection from latent or inactive stages. Variations in miRNA expression during Mtb infection indicate disease progression and offer insights into the immune response. Furthermore, miRNAs present potential as therapeutic targets in host-directed therapy (HDT) techniques for TB infection. This work examines the function of miRNAs in TB pathogenesis, with the objective of identifying particular miRNAs that regulate the immune response to the Mtb complex, evaluating their diagnostic value and exploring their therapeutic implications in host-directed therapy for TB infection. To meet the critical demand of LTR-RTs data-driven research, we updated the CicerSpTEdb database to version 2.0, which includes more accurate intact LTR-RT elements with annotation of internal domains. We also added the ability to BLAST against TEs of Cicer species. As a result, 3701 intact LTR-RTs were detected in the studied genomes, including 2840 Copia and 861 Gypsy elements. Of the 3701 intact LTR-RTs, 588 were in C. arietinum, including 475 Copia and 113 Gypsy. While 1373 were detected in C. reticulatum, including 1041 Copia and 332 Gypsy. Furthermore, 1740 were found in C. echinospermum, including 1324 Copia and 416 Gypsy. Based on LTR-RT clades, the analysis classified the 3701 identified intact LTR-RTs in the studied genomes as Ale (850), SIRE (740), unknown (455), Ikeros (323), Reina (290), Tork (290), Ivana (282), Tekay (197), Athila (128), TAR (99), CRM (31), and Ogre (16) elements.
