Abstract

The study investigated theefficacy of five entomopathogenic fungi [Aspergillussp., Beauveria sp., Fusarium sp., Trichoderma sp. (1) and Trichodermasp. (2)] isolated from soil samples which were taken in the wheat farming of INPV (National Institute ofPlant Protection of Constantine, Constantine-Algeria) on the second-stagelarvae of Tuta absoluta (Lepidoptera: Gelechiidae). Spore suspensions of fungi were applied by spraying method afterdilution with sterile distilled water containing 0.02% Tween 80, so that 107spores/ml concentration. Individuals exposed to distilled water containingTween 80 were used as the control group in order to compare the effect ofentomopathogenic fungi. Evaluations of infection development were made at the 1st,3rd, 5th and 7th days following applications.According to the results obtained, the highest mortality at the 7th DAA (DayAfter Application) was found as 75.00% and 73.00% from the individuals exposedto of with Trichoderma sp. (1) andwith Trichoderma sp. (2)entomopathogenic fungi, respectively. These entomopathogenic fungi followed%62.00, %45.00 and %41.00 mortality of Aspergillussp., Fusarium sp. and Beauveria sp., respectively., Bu çalışmada, Ulusal BitkiKoruma Enstitüsü (INPV-National Institute of Plant Protection of Constantine,Constantine-Algeria) çevresinden alınan toprak örneklerinden izole edilen 5farklı entomopatojen fungus [Aspergillus sp.,Beauveria sp., Fusarium sp., Trichodermasp. (1) ve Trichoderma sp. (2)]’un Tuta absoluta Meyrick (Lepidoptera:Gelechiidae) 2. dönem larvaları üzerine etkinliği araştırılmıştır.Fungusların spor süspansiyonları 107 spor/ml olacak şekilde %0,02oranında Tween 80 içeren steril saf su ile seyreltildikten sonra püskürtmeyöntemiyle uygulanırken, Tween 80 içeren steril saf su uygulanmış bireyler ise kontrolgrubu olarak kabul edilmiştir. Enfeksiyon gelişimi değerlendirmeleri,uygulamalar yapıldıktan 1, 3, 5 ve 7 gün sonra canlı birey sayıları üzerindenyapılmıştır. Elde edilen sonuçlara göre, uygulamadan sonraki 7. günde, kontrolgrubuna göre en yüksek yüzde ölüm oranı %75,00 ile Trichoderma sp. (1) ve %73,00 ölüm oranı ile Trichoderma sp. (2) entomopatojen funguslarında elde edilmiştir. Bufungusları sırasıyla %62,00, %45,00 ve %41,00 ölüm oranı ile - Aspergillus sp., Fusarium sp. ve Beauveria sp.fungusları takip etmiştir.

4. Pathogenicity of three entomopathogenic fungi, to the aphid species, Metopolophium dirhodum(Walker) (Hemiptera: Aphididae), and their Alkaline protease activities

Abstract

The aim of the present study is to investigate the effect of three entomopathogenic fungi (EPF) (Beauveria bassiana, Cladosporium cladosporioides, and Verticillium alfalfae) on the aphid species, Metopolophium dirhodum(Walker) (Hemiptera: Aphididae).The selected EPF were isolated from the agricultural soil of the National Institute of Plant Protection (INPV) in Constantine, Algeria, and were tested against the aphid insects that were collected from the same area. The aphid species M. dirhodumwere exposed to each fungal spore suspensions 107conidia/ml for 10 s. Percent mortality was recorded at 1, 3, 5, and 7 days post treatment. Percentage mortalities, 7 days post treatment, were 95.83, 63.98, and 51.83% by B. bassiana, C. cladosporioides, and V. alfalfae, respectively. The higher protease activities were observed for isolate V. alfalfaewith 95 U/ml, followed by B. bassianawith 38.26 U/ml, and finally C. cladosporioideswith 35, 65 U/ml. The results presented in this study revealed that there was no relation between high alkaline protease activities and high virulence isolates.

5. Effects of some entomopathogenic fungi on the aphid species, Aphis gossypiiGlover (Hemiptera: Aphididae)

Abstract

Background: Effects of the entomopathogenic fungi Beauveria bassiana, Verticillium alfalfae, and Trichoderma viride; secondary metabolites of MS1 (B. bassiana) and MS2 (V. alfalfae); and Dimethoate active substances on the aphid species, Aphis gossypiiGlover (Hemiptera: Aphididae), were tested. Main body of the abstract: Fungus isolates were prepared as 107conidia ml−1of spore suspensions and applied on the 2nd instar nymphs of A. gossypii. After the applications, evaluations were made on the 1st, 3rd, 5th, and 7th days by counting the live individuals. Obtained data were 100 and 93% mortality rate at MS1 (B. bassiana) and MS2 (V. alfalfae), and secondary metabolites were recorded in the 3rd day count results. On the 5th day counts, the highest mortality rates after secondary metabolites were statistically at the same group with B. bassiana, T. viride, and dimethoate. On the 7th day, counting results of all experiment groups were analyzed statistically and were found effective. Short conclusion: Obtained results showed that the fungal secondary metabolites might be useful when utilized as a biocontrol agent against the aphids.
