KTRs’ compromised immune response to COVID-19 vaccines indicates urgent revision of immunisation policies.Methods: A cross-sectional study was conducted in Madinah, Saudi Arabia of 84 KTRs who had received at least one dose of a COVID-19 vaccine. ELISA was used to evaluate anti-spike SARS-CoV-2 IgG and IgM antibody levels in blood samples obtained one month and seven months after vaccination. Univariate and multivariate analyses were performed to identify associations between seropositive status and factors such as the number of vaccine doses, transplant age, and immunosuppressive therapies.Results: The mean age of KTRs was 44.3 ± 14.7 years. The IgG antibody seropositivity rate (n=66, 78.5%) was significantly higher than the seronegativity rate (n=18, 21.4%) in the whole cohort (p< 0.001). In KTRs seroconverting after one month (n=66), anti-SARS-CoV-2 IgG levels declined significantly between one month (median [IQR]:3 [3– 3]) and seven months (2.4 [1.7– 2.6]) after vaccination (p< 0.01). In KTRs with hypertension, IgG levels significantly decreased between one and seven months after vaccination (p< 0.01). IgG levels also decreased significantly in KTRs with a transplant of > 10 years (p=0.02). Maintenance immunosuppressive regimens (triple immunosuppressive therapy and steroid-based and antimetabolite-based regimens) led to a significant decrease in IgG levels between the first and second sample (p< 0.01). KTRs receiving three vaccine doses showed higher antibody levels than those receiving a single dose or two doses, but the levels decreased significantly between one (median [IQR]: 3 [3– 3]) and seven months (2.4 [1.9– 2.6]) after vaccination (p< 0.01).Conclusion: KTRs’ humoral response after SARS-CoV-2 vaccination is dramatically inhibited and wanes. Antibody levels show a significant decline over time in KTRs with hypertension; receiving triple immunosuppressive therapy or steroid-based or antimetabolite-based regimens; receiving mixed mRNA and viral vector vaccines; and with a transplant of > 10 years.Keywords: SARS-CoV-2, kidney transplant recipients, booster, immunosuppression, COVID-19 vaccines, humoral immune response

A cost-effective, selective, sensitive, and operational TLC-densitometric approach has been adapted for the concurrent assay of Hydroxyzine Hydrochloride (HYX), Ephedrine Hydrochloride (EPH), and Theophylline (THP) in their pure powder and pharmaceutical forms. In the innovative TLC-densitometric approach, HYX, EPH, and THP were efficaciously separated and quantified on a 60F 254 silica gel stationary phase with chloroform-ammonium acetate buffer (9.5:0.5, v / v ) adjusted to pH 6.5 using ammonia solution as a mobile liquid system and UV detection at 220 nm. The novel TLC method validation has been performed in line with the international conference for harmonization (ICH) standards and has been effectively used for the estimation of the researched medicines in their pharmaceutical formulations without intervention from excipients. Additionally, parameters affecting the chromatographic analysis have been investigated. The new TLC approach's functionality and greenness were appraised using three modern and automated tools, namely the Blue Applicability Grade Index (BAGI), the Analytical Greenness metric (AGREE), and the Green Analytical Procedure Index (GAPI) tools. In short, the greenness characteristics were not achieved as a result of using mandatory, non-ecofriendly solvents such as ammonia and chloroform. On the contrary, the applicability and usefulness of the novel TLC approach were attained via concurrent estimation for the three drugs using simple and straightforward procedures. Moreover, the novel TLC method outperforms previously published HPLC ones in terms of the short run time per sample and moderate pH value for the liquid system. According to the conclusions of comparisons with previously recorded TLC methods, our novel HPTLC method has the highest AGREE score, so it is the greenest HPTLC strategy. Moreover, its functionality and applicability are very appropriate because of the simultaneous assessment of three drugs in one TLC run. Furthermore, no tedious and complicated extraction and evaporation processes are prerequisites.

Purpose: To evaluate the effect of a Simulation-based training (SBT) program on neonatal and paediatric nurses' knowledge regarding infant safe sleep practices., Background: Sudden infant death syndrome (SIDS) presents a major public health concern, preventable through the promotion of optimal safe sleep practices, particularly among neonatal and paediatric nurses. Despite its effectiveness in enhancing nurses' knowledge and clinical skills, SBT is not an adopted training method for nurses in Egypt., Design and Methods: A single-group pre- and post-test design involved 57 nurses from Neonatal Intensive Care Unit, Paediatric Intensive Care Unit, and Paediatric In-patient Unit. The study consisted of two stages. In the first stage, knowledge assessment to identify deficiencies. The second stage, researchers developed four SBT scenarios. Two of these scenarios were recorded for training purposes, while the other two were intended for nurses to actively participate in. Data were collected from May 2022 to January 2023., Results: A significant improvement in nurses' knowledge of infant safe sleep practices and SIDS prevention was observed (p = 0.000). This study investigated and explored the availability of micro-flora and micro-fauna in the ruminal contents of Arabian camel ( Camelus dromedarius ) from three different regions in Saudi Arabia along with two seasons. Samples were prepared and tested by conventional polymerase chain reaction (PCR). This study confirmed that the bacterial flora were dominating over other microbes. Different results of the availability of each microbe in each region and season were statistically analyzed and discussed. There was no significant effect of season on the micro-flora or micro-fauna however, the location revealed a positive effect with Ruminococcus flavefaciens (p  < 0 0.03) in the eastern region. This study was the first to investigate the abundance of micro-flora and micro-fauna in the ruminal contents of camels of Saudi Arabia. This study underscores the significance of camel ruminal micro-flora and micro-fauna abundance, highlighting their correlation with both seasonality and geographic location. This exploration enhances our comprehension of camel rumination and digestion processes. Preserving the environment, reducing the amount of waste resulting from chemical trials, and reducing the amount of energy consumed have currently become a pivotal global trend. An analytical quality by design (AQbD) based eco-friendly TLC-densitometric method was implemented for quantifying two antihypertensive agents, captopril (CPL) and hydrochlorothiazide (HCZ), along with their impurities; captopril disulphide (CDS), chlorothiazide (CTZ) and salamide (SMD). The analytical target profile (ATP) was first identified, followed by selecting the critical analytical attributes (CAAs), such as retardation factors and resolution between the separated peaks. Critical method parameters (CMPs) that may have a crucial influence on CAAs were identified and emanated through the quality risk assessment phase. A literature survey-based preliminary studies were performed, followed by optimization of the selected CMPs through a custom experimental design to attain the highest resolution with optimum retardation factors. Moreover, method robustness was also tested by testing the design space. Complete separation of the drugs and their impurities was achieved using ethyl acetate: glacial acetic acid (6: 0.6, v/v) as a developing system applied to a 12 cm length TLC plate at room temperature with UV scanning at 215 nm. Calibration graphs were found to be linear in the ranges of (0.70-6.00), (0.10-2.00), (0.20-1.00), (0.07-1.50) and (0.05-1.00) µg/band corresponding to CPL, HCZ, CDS, CTZ, and SMD, respectively. Four different green metric tools were used to evaluate the greenness profile of the proposed method, and results showed that it is greener than the reported HPLC method. Method whiteness assessment was also conducted. Moreover, the method performance was evaluated following the ICH guidelines, and the outcomes fell within the acceptable limits. The developed method could be approved for routine assay of the cited components in their pharmaceutical formulations and bulk powder without interference from the reported impurities. Two accurate, sensitive, and selective methods for simultaneous determination of miconazole nitrate (MIC), nystatin (NYS), and metronidazole (MET) in pure state or drug product were established and verified. First, RP-HPLC-DAD was designed. Separation was accomplished using a ZOBRAX Eclipse Plus RP-C 8 column that was running under an isocratic elution of methanol: 0.05% aqueous solution of sodium dodecyl sulphate (40: 60 v/v), with a flow rate that was regulated at 0.8 mL/min. The column temperature was adjusted at 25 °C and diode array detector was monitored at 220 nm. The linearity range of the proposed method was achieved at the concentration of 5-50, 4-50, and 4-40 µg/mL and the attained retention time for the studied drugs was 2.52, 3.52 and 4.99 min for MIC, NYS, and MET, correspondingly. Second, a TLC-densitometric approach was used to resolve the three compounds. Resolution of the three cited drugs was carried out using TLC aluminum plates pre-coated with 0.25 mm silica gel 60 F 254 . A developing solvent comprised ethyl acetate: toluene: methanol: triethyl amine: formic acid (3: 1: 7: 0.3: 0.1 by volume) (pH = 5.5) was utilized and scanning of the resolved bands at 215 nm. Linearity of the developed TLC method was evaluated and evident to be 0.4-2, 0.4-2.2, and 0.4-2 μg/band for MIC, NYS, and MET, in that order. The suggested chromatographic methods were verified according to ICH directives. The findings of the developed chromatographic procedures were statistically compared with the results of the reported ones using student's t-test and F-test. Hypertension is described by the world health organization (WHO) as a serious medical problem that significantly affects the heart, brain and kidneys. It is a major cause of premature death worldwide. The present study aims to quantify the combination of captopril (CPL), hydrochlorothiazide (HCZ) and their harmful impurities; captopril disulphide (CDS), chlorothiaizde (CTZ) and salamide (SMD). In-silico study was conducted for estimation of pharmacokinetic parameters (ADMET) as well as toxicity profile of the proposed impurities. The results showed that the three impurities under investigation had poor permeability to CNS and cannot pass the blood-brain barrier (BBB), reducing the likelihood of causing side effects in the brain. On the other hand, all studied impurities were found to be hepatotoxic. In consequence, a highly sensitive and green ultra-performance liquid chromatography- tandem mass spectrometric (UPLC/MS/MS) method was developed and validated for separation of the cited drugs in the presence of their harmful impurities; methanol and 0.1% formic acid (90:10, v/v) mixture was used as a mobile phase, eluted at a constant flow rate of 0.7 mL/min at room temperature. Detection was adopted using a tandem mass spectrometer in a positive mode only for CPL and negative mode for HCZ, CDS, CTZ and SMD. Separation was performed within 1 min. Calibration graphs were found to be linear in the ranges of (50.0-500.0 ng mL -1 ), (20.0-500.0 ng mL -1 ), (10.0-250.0 ng mL -1 ), (5.0-250.0 ng mL -1 ) and (20.0-400.0 ng mL -1 ) corresponding to CPL, HCZ, CDS, CTZ and SMD, respectively. Additionally, comparative study of greenness profile was established for the proposed and reported methods using five green metric tools. The proposed method was found to be greener than the reported HPLC method. The developed (UPLC/MS/MS) method was validated according to (ICH) guidelines and it was found to has greater sensitivity, shorter analysis time and lower environmental impact compared to the reported methods., (© 2023. Background: Chronic gastritis (CG) is a very common disease. More than half of the worldwide population suffers from symptoms of CG. This disease has received great attention since the discovery of H. pylori as the most important cause of CG. Symptoms experienced by patients with CG are attributed to H. pylori -induced inflammatory reactions. Heparanase (HPSE) is a mammalian β-endoglucoronidase. In inflammation; HPSE degrades and remodels the extracellular matrix's heparan sulfate polysaccharide chains liberating heparan sulfate-bound cytokines and chemokines, HPSE also facilitates movement of inflammatory cells., Aims: This study aimed to detect the function of HPSE in CG by correlating levels of HPSE expression with histopathological features of CG, including H. pylori infection, acute and chronic inflammatory cells, mucosal atrophic and/or metaplastic features., Methods: Ninety-five upper endoscopic-guided gastric punch biopsies were enrolled in this study. From each specimen, formalin-fixed and paraffin-embedded tissue blocks were prepared. Tissue sections were stained by Hematoxylin and eosin, Giemsa, and anti-heparanase antibody., Results: HPSE expression was statistically associated with H. pylori infection ( P -value < .000), and intensity of chronic lymphocytic inflammatory infiltrate in the gastric mucosal tissues ( P  = .004). High levels of HPSE expression were also related to the presence of neutrophils in the gastric surface epithelium and lamina propria ( P -value < .009)., Conclusions: HPSE expression was upregulated in H. pylori -associated chronic gastritis. Background: Using natural zeolites as a food additive in poultry diets offers an intriguing perspective. The objective of this study was to investigate the effects of zeolite addition and particle size on broiler performance, carcass characteristics, meat quality, moisture of excreta and litter, and intestinal measurements during 35 days., Methods: A total of 560 1-day-old female Ross-308 broilers were divided into five treatment levels (0, 5, 10, 15, and 20 g zeolite/kg diet) ( n = 16 replicates/treatment, n = 8 replicates /particle size of each treatment). Performance was calculated weekly. Carcass characteristics, meat quality, small intestine (SI) measurements, litter pH, and moisture content were determined on day 35., Results: Litter pH, breast redness, cooking loss, chewiness, total weight, and SI length were all affected by zeolite treatments ( p < 0.05). Particle size had an impact on the gastric pH and texture analysis. Their interaction had an effect on color redness, litter pH, and cooking loss. Performance was unaffected by either the main or interaction effects., Conclusion: Zeolite as a feed additive may be useful in broiler diets, particularly large particles. The performance and production efficiency factor improved numerically ( p > 0.05) with increasing zeolite doses up to 10 g zeolite/kg diet.

Introduction: Lasalocid is a feed additive widely used in ruminant nutrition and plays a crucial role in improving livestock productivity, digestibility, immunity, and overall wellbeing. The current study was conducted to investigate the effect of different levels of lasalocid (LAS) supplementation on growth performance, serum biochemistry, ruminal fermentation profile, in vitro nutrient digestibility, and gas production of growing goats., Methods: A total of 60 growing Aardi male goats with an average body weight of ~17.12 kg (3-month-old) were used for an 84-day trial. Animals were randomly divided into four treatment groups with 5 replicates of 3 goats each. All four groups were provided with a basal diet supplemented with lasalocid (LAS) at 0 (without supplementation; LAS0), 10 (LAS10), 20 (LAS20), or 30 (LAS30) ppm LAS/kg dry matter (DM). Feed intake was measured weekly, and goats were weighed every 2 weeks for an evaluation of the performance parameters. Blood samples were collected for the measurement of biochemical variables. In vitro nutrient digestibility and gas production were evaluated., Results and Discussion: The supplementation of LAS at level 30 ppm/kg DM increased ( P < 0.05) the body weight gain and average daily gain without linear or quadratic effect. The serum concentrations of high-density lipoprotein were significantly ( P < 0.05) higher in the LAS20 group than in other groups with linear and quadratic effects, while low-density lipoprotein concentration was significantly lower in the LAS20 group than in LAS0 and LAS30 with a linear effect. Different levels of lasalocid supplementation had no effect on the ruminal fermentation profile, in vitro gas production, and nutrient digestibility. The aim of this study was to investigate the serum level of fat-soluble vitamins A, D and E in clinically healthy lactating female camel ( Camelus dromedarius ) and suckling calf > one-year-old during winter and summer seasons in five main regions of Saudi Arabia. 60 sera samples were collected and tested for vitamins A, D and E levels and the results were statistically analyzed. The statistical mean value of vitamin A was within the reported range but for D and E, there were minor variations. The effect of season was insignificant ( p  > 0.05) for vitamins A and E in the combined results of the dam and newborn together. This seasonal effect was highly significant in dam serum ( p  < 0.05). Region effect was significant for vitamin A in the northern area ( p  < 0.05) and for vitamin E in the southern region ( p  < 0.05). Correlations analysis revealed significant results in the season vs vitamin A and E p  < 0.05. Mean values of vitamins A, D and E in dam and newborn did not observe significant variations however, in the season and regions there were significant variations which can be attributed to the climate difference, availability of balanced rations and camel management in each location of the five main regions of Saudi Arabia. Severe COVID-19 disease was linked to a severe proinflammatory response and cytokine storm interleukin 17 (IL-17) is one of these cytokines, was associated with severe acute lung injury and multiorgan dysfunction. Single nucleotide polymorphisms (SNPs) in genes coding IL-17 can affect level of IL-17 hence its role in diseases. Also, SNPs in IL-23 R which control IL-23 is the main activator of IL-17 production. This study aimed to determine whether the IL-17A (G/A-rs2275913), IL-23R (A/G rs11209026) SNPs and serum levels of IL-17 were related to the risk of severe COVID-19. This case-control study included 120 confirmed COVID-19 patients, divided into two categories according to the severity of the disease and 74 normal subjects as controls. COVID-19 patients were SARS-CoV-2 positive by a reverse transcription-polymerase chain reaction and subjected to full clinical examinations, routine laboratory tests, and radiographic evaluations. The IL-17 levels were assessed using ELISA method, and genotyping of IL-17A (197 A/G; rs2275913) and IL-23R rs11209026 (A/G) was performed by the TaqMan Genotyping Assay. There were no differences in the distribution of IL-17A or IL-23R genotypes between COVID-19 groups and the control group (p=0.93 and p=0.84, respectively). Severe COVID-19 patients had significantly higher IL-17 serum levels than non-severe COVID-19 (p=0.0001). The GG genotypes of IL-17A were significantly higher in severe COVID-19 patients (p=0.004). Multivariate logistic regression analysis revealed that AG, GG genotypes of IL-17 and IL-17A were independent predictors of COVID-19 disease severity (p < 0.0001, p=0.06 and p=0.04, respectively). ROC curve analysis for IL-17, as predictor of severe COVID-19 disease revealed a sensitivity of 87.9% and specificity of 66.1% at a cutoff point of 114 pg/ml with AUC = 0.799. In conclusion, these findings indicated that IL-17 may be considered a marker of severe COVID-19. IL-17A SNPs may have a role in COVID-19 severity. Soil, water, and forage contaminated with toxic heavy metals such as Pb, Cd, and Co can affect the meat and liver quality of camels raised in this area which affect human health. This study aimed to determine the concentrations of Pb, Cd and Co in soil, water, feed and young camels' carcass tissues (meat, liver, whole blood, rumen fluid and rumen tissue) from five districts in Saudi Arabia during the spring. All samples were wet-digested and analyzed by ICP-OES for heavy metals. In the liver, a significantly higher ( p < 0.05) concentration of Pb and Co was observed in camels reared in the central and southern districts, while Cd was found significantly higher in the western and northern districts. The concentration of Pb, Cd and Co in meat of camels in the northern district was significantly higher ( p < 0.05), and the meat of camels in the west had a higher ( p < 0.05) concentration of Co. In addition, the Cd and Co concentrations in rumen fluid samples from camels in the eastern district were significantly higher than in the central district. A negative correlation between the concentration of Pb and Cd in rumen fluid and rumen tissue was reported. The accumulation of Cd, Pb, and Co in meat and liver was below the recommended maximum limit. Therefore, the harmful risk of human consumption of camel meat and liver is not possible.

The objective of the present study was to investigate the effects of sprouted barley inclusion level on the growth performance, digestibility, volatile fatty acids, bacterial quantification, and rumen morphology of growing lambs. Five dietary treatments with sprouted barley (0, 25, 50, 75 and 100%) and nine replicates per dietary treatment were performed on forty-five Awassi lambs (90 days old). The average weight gain, intake, and digestibility of dry and organic matter were recorded. The pH, color, volatile fatty acids, bacterial quantification, and rumen histomorphometry were also determined. The results showed that the average dry and organic matter intake in T2 to T4 and the average weight gain in T4 decreased linearly. In contrast, the digestibility of dry and organic matter by sprouted barley (T2 to T4) was higher. The pH values and rumen color were not affected. Concentrations of formic acid, acetic acid, butyric acid, and the ratio of acetic acid to propionic acid were increased, while lactic acid and total volatile fatty acids were lower in all levels of sprouted barley. In addition, lambs fed T4 had a higher quantification of Anaerovibrio Lipolytica , Butyrivibrio Fibrisolvens , and Streptococcus Bovis quantification. Selenomonas Ruminantium was higher in T1, T2, and T4, whereas Megashpaera Elsdenii was lower in T1 to T3. The rumen histomorphometric was improved by sprouted barley (T2 and T3). Sprouted barley improved digestibility and rumen histomorphometry and increased the concentration of some volatile fatty acids and rumen bacteria but resulted in a decrease in average dry and organic matter intake, which negatively affected weight gain in lambs fed 100 % sprouted barley. Further studies are required to determine the potential effects on growing lambs fed sprouted barley.

Background: Cyclizine (CYZ), a commonly used antiemetic drug, has two pharmacopeial toxic impurities, 1-methylpiperazine (MPZ) and diphenylmethanol (DPM). When CYZ parenteral formulations are administered intravenously, both impurities are poisonous, toxic, and harmful to the human body., Objective: Cyclizine was determined along with its hazardous impurities MPZ and DPM by green multivariate calibration using UV-spectroscopic data., Methods: Three multivariate algorithms were used to resolve and quantify overlapped spectral signals: principal component regression (PCR), partial least squares (PLS), and synergistic intervals partial least squares (siPLS). A concentration set containing 16 distinct combinations of CYZ, MPZ, and DPM was randomly prepared, and the absorbance values of the concentration set were determined using the 376 point-wavelength set with an interval of 0.2 nm between 200 and 275 nm., Results: Good linear correlations were established for CYZ, MPZ, and DPM in the concentration ranges of 5.00-25.0, 0.50-2.50, and 0.50-2.50 µg/mL, respectively. The ideal spectral range and associated combinations were chosen based on the lowest root mean error of prediction (RMSEP) and correlation coefficient values (r). The siPLS approach performed better than the PCR and PLS models. The combination of four subintervals, 1, 3, 4, and 7, demonstrated the greatest effect, with RMSEP values of 0.0272, 0.0053, and 0.0315 for CYZ, MPZ, and DPM, respectively, and correlation coefficients of 0.9991, 0.9999, and 0.9997, in order. Various assessment tools were used to evaluate and measure the greenness profile of the established methods. The proposed methods were validated using internal and external validation sets., Conclusions: The three methods were effectively used to determine CYZ in its pure form and parenteral formulations, as well as its toxic impurities. Background: Pathogenesis and endothelial function in subclinical hypogonadism (SCH) remain unclear. Undercarboxylated osteocalcin (ucOC) participates in atherosclerosis and reproduction. We explored the underlying mechanisms and interplay of endothelial dysfunction, unOC and reproductive hormones in SCH and primary late-onset hypogonadism (LOH)., Methods: In the SCH, LOH, and healthy eugonadal male groups, we measured serum unOC, calculated luteinizing hormone/testosterone (LH/T), LH.T product, and estradiol/T (E/T) as indicators of impaired Leydig cells, androgen sensitivity index (ASI), and aromatase activity, respectively (LH set-point regulators), and assessed flow-mediated dilation of the brachial artery (FMD%), carotid-intima media thickness (CIMT), and aortic stiffness (AS)., Results: ↑LH/T, ↑ASI, ↓aromatase activity, normal T, follicle-stimulating hormone (FSH) and sex hormone-binding globulin (SHBG) levels, ↑unOC, and enhanced atherosclerotic markers (↓FMD%, ↑CIMT, ↑AS) are characteristics of SCH. Testosterone was positively correlated with FMD% in SCH. The independent predictors were: SHBG and LH for FMD% and CIMT, respectively, and LH/T, ucOC, FSH, estradiol, and E/T ratio for AS in the LOH group; and LH for FMD% & AS and LH and LH/T for CIMT in all study subjects., Conclusions: SCH is a distinct clinical entity characterized by impaired androgen sensitivity and aromatase activity, compensatory elevated unOC, endothelial dysfunction, and anti-atherogenic role of testosterone.

Camel products are receiving great interest worldwide because of their high functional properties and nutritive values. Therefore, this study was focused on the variation of copper [Cu], zinc [Zn], manganese [Mn], selenium [Se], iron [Fe], iodine [I], and some heavy metals, cobalt [Co], lead [Pb], and cadmium [Cd], in the blood and tissues of slaughtered camels from five regions in Saudi Arabia [SA] during the summer and winter seasons, because environmental factors vary from region to region. Whole blood, meat, liver, rumen fluid, and rumen tissues were collected from the slaughterhouse in each region during the two seasons. Moreover, samples were prepared and analyzed for trace mineral and heavy metal concentrations using ICP-MS. The data were statistically analyzed as part of a complete randomized design and correlation analysis for season and location using SAS. The findings revealed a pattern in the minerals, with Ca being the only mineral that was unrelated to other minerals in the liver. For lead and cadmium, our mean value in liver [0.40 µg/g] was below the limit of the EU standard for cadmium [0.50 µg/g], while in meat and liver, lead contents [1.62 µg/g and 2.57 µg/g, respectively] were above the limit of the EU standard [0.10 and 0.20 µg/g, respectively]. For meat, the significantly highest positive correlations were observed between P and Mg [ R 2 = 0.928], Fe and Mn [ R 2 = 0.860], and Co and Mn [ R 2 = 0.821]. For rumen tissues, P and Mg were highly correlated [ R 2 = 0.958] as well as Zn and Mg [ R 2 = 0.857], Zn and P [ R 2 = 0.836], and Fe and Ca [ R 2 = 0.802]. As a result, a region and season reflect variations in mineral concentrations in SA during the summer and winter seasons. Further intensive research is needed to investigate the minerals' biological mechanisms in camels under different environmental conditions.

The objective of the present study was to evaluate the impact of inclusion of wild cumin seeds (WCS) on performance, nutrient digestibility and blood profile in broilers during the finisher phase. For this purpose, 360, 14 days old chicks were randomly divided into four groups designated as CONT (control), 0.5WCS, 1.0WCS and 1.5WCS with 5 replicates, which were supplemented with WCS at the rate of 0, 0.5, 1, and 1.5% respectively. On the overall, feed intake was significantly ( p  < 0.01) higher in the CONT compared to the 1.0WCS. At the end of the finisher phase, and overall basis, body weight and feed conversion ratio were significantly ( p  < 0.05) higher in 0.5WCS. Crude protein apparent digestibility in the ileum was significantly ( p  < 0.05) higher in 0.5WCS compared to the CONT, while crude fat digestibility was significantly ( p  < 0.01) higher in 0.5WCS and 1.5WCS compared to the control. Similarly, serum triglyceride was significantly ( p  < 0.05) lower in 0.5WC but high density lipoprotein was significantly ( p  < 0.05) higher in the same group. It was concluded that wild Cumin at the rate of 0.5% was superior compared to the other treatments in the diet to improve the performance, nutrient digestibility and blood metabolites in broiler during the finisher phase.

Trace minerals play an important role in animal health and productivity. They are involved also in many physiological activities, and their deficiency causes a variety of pathological problems and metabolic defects, reducing consequently the animal productivity. The demand for animal products in semi-arid areas is rapidly increasing, and the supply is still below the required level, partially due to low animal productivity. Camels ( Camelus dromedarius and Camelus bactrianus ) are considered one of the main sources of healthy, high-quality meat and milk for human consumption within most of the countries in the semi-arid regions. Despite their efficient adaptation to their environment, camels can suffer from the growth retardation of newborns, low feed efficiency, anemia, poor fertility, poor reproduction and many other metabolic disorders. It is well known that trace mineral deficiencies and trace mineral toxicities can influence camels' production and reproductive efficiency, as well as many aspects of their growth and metabolism. Evaluating the trace minerals status of camels and their variability is an obvious step toward improving camels' productivity and health. Thus, the present article reviews the data regarding the status of trace minerals (copper, zinc, iron, selenium, manganese, cobalt, iodine, fluorine, molybdenum, sulfur, bromide and nickel) in camel blood and their physiological variability, with a focus on their deficiency and toxicity effects.

Backgrounds: The incidence of non-alcoholic fatty liver disease (NAFLD) has been significantly growing in recent years. Although the pathophysiology of fibrosis progression in NAFLD is not yet known, oxidative stress and inflammation have been known to have a major role in the development of NASH. Understanding the impact of micronutrients in NAFLD could potentially help us better understand NAFLD pathogenesis., Aims: Assessing the serum levels of Zn, Se, and Vitamin E and their relation to the development of hepatic fibrosis in NAFLD patients., Methods: This study included 80 NAFLD patients and 40 healthy controls. All of the patients were subjected to abdominal ultrasound and FibroScan examination (to estimate hepatic fibrosis and steatosis degree), and the serum levels of Zn, Se, and vitamin E were evaluated., Results: A statistically significant difference in the serum levels of Zn and Se was observed between the NAFLD group and the control group (P-value = 0.04 and 0.05, respectively). The serum levels of Zn and Se were independently related to the presence of hepatic fibrosis in NAFLD. However, serum vitamin E was not related to the severity of NAFLD. Furthermore, no significant difference in the levels of Zn, Se, and vitamin E was observed between the different groups of NAFLD patients categorized according to the degree of steatosis and the control group., Conclusions: Reduced serum levels of Zn and Se can be considered a possible risk factor for hepatic fibrosis in NAFLD. Deficiency in these micronutrients could play a role in the pathogenesis of NAFLD.

This work implements a combined experimental approach of analytical quality-by-design (AQbD) and green analytical chemistry (GAC) to develop an HPLC method for simultaneous determination of the two thalassemia drugs, deferasirox (DFX) and deferiprone (DFP), in biological fluid for the first time. This integration was designed to maximize efficiency and minimize environmental impacts, as well as energy and solvent consumption. To accomplish this goal, an analytical quality-by-design approach was performed, beginning with quality risk assessment and scouting analysis, followed by Placket-Burman design screening for five chromatographic parameters. Critical method parameters were thoroughly recognized and then optimized by using a two levels-three factors custom experimental design to evaluate the optimum conditions that achieved the highest resolution with acceptable peak symmetry within the shortest run time. The desirability function was used to define the optimal chromatographic conditions, and the optimal separation was achieved using an XBridge® HPLC RP-C18 (4.6 × 250 mm, 5 μm) column with ethanol : acidic water at pH 3.0 adjusted by phosphoric acid in the ratio of (70 : 30, v/v) as the mobile phase at a flow rate of 1 mL min -1 with UV detection at 225 nm at a temperature of 25 °C. Linearity was obtained over the concentration range of 0.30-20.00 μg mL -1 and 0.20-20.00 μg mL -1 for DFX and DFP, respectively, using 20.00 μg mL -1 ibuprofen (IBF) as an internal standard. The established method's greenness profile was evaluated and measured using various assessment tools, and the developed method was green. For the validation of the developed method, FDA recommendations were followed, and all the results obtained met the acceptance criteria. The suggested method was successfully used to study the pharmacokinetic parameters of DFX and DFP in rat plasma. The main objective of this study was to investigate the effects of freshly sprouted barley on the growth of lambs, in addition to its nutritional value and digestibility. In addition, sprouted barley digestibility and rumen fermentation were studied in vitro on a dry matter (DM) basis. A total of 45 three-month-old Awassi lambs were randomly assigned to five treatments of sprouted barley (0, 25, 50, 75, 100%) diets. Bodyweight, weight gain, feed intake and feed efficiency were recorded every two weeks. Nutrient analyses were performed on feed, faecal, and urine samples. DM and non-fibrous carbohydrates were measured. Digestibility of DM, organic matter (OM), and neutral detergent fiber (NDF), as well as gas production, pH value, ammonia-N, and volatile fatty acids (VFAs), were determined in vitro using continuous culture. The results showed that final bodyweight was lower (p < 0.05), while feed intake and the feed-to-gain ratio were increased (p < 0.05) in sprouted barley treatments. Nutrient analysis indicators of sprouted barley treatments (25 to100%) were lower (p < 0.05) for DM, crude protein, acid detergent fiber, lignin and ash, and higher for total digestible nutrients, NDF, fat, phosphorus, zinc, copper, and net energy than the traditional diet. In the in vivo study, the digestibility of nutrients in sprouted barley treatments was improved (p < 0.05), while the diet (sprouted barley 100%) had the lowest digestibility of DM, OM, and NDF compared with the other treatments in the in vitro study. In conclusion, the addition of sprouted barley improved digestibility, and fermentation characteristics, while having a negative effect on growth. Further studies are recommended for optimal growth performance.

Background: Pain due to inoperable upper abdominal malignancies is a challenging condition that needs a multimodal analgesic regimen to be managed properly. Celiac plexus alcohol neurolysis was proved to be effective in relieving such type of pain; however, there is no consistent data about the optimal volume to be used to maintain the balance between the neurolytic effect and the destructive effect of alcohol., Objectives: We aim to compare the analgesic effect of 2 different volumes of alcohol to improve the outcome of interventional management., Study Design: This was a randomized controlled double-blinded interventional clinical trial., Setting: Single university hospital., Methods: Thirty-two patients who suffered from abdominal pain due to unresectable abdominal malignancies were randomly allocated to receive in a single injection ultrasound-guided celiac plexus neurolysis (CPN) with injection of either 20 mL 70% alcohol (CPN20 group) or 40 mL 70% alcohol (CPN40 group). The primary outcome was the post-procedure pain score, while the secondary outcomes included the post-procedure total daily opioid consumption and quality of life (QOL)., Results: There was no statistically significant difference between both groups regarding visual analog scale (VAS) scores at all time points (P-value > 0.05); however, comparisons in each group revealed significantly reduced VAS scores at all time points following the intervention when compared to the baseline. Daily morphine equivalent consumption doses showed statistically significant differences between the baseline and each time point in both groups (P value < 0.05), with no significant difference between both groups at each time point (P value > 0.05). There was no statistically significant difference between the study groups regarding all domains in quality of life assessment at all time points (P value > 0.05). The scores of most time points in all domains were different significantly when compared to the baseline readings in both groups, with a tendency to decline over time in both groups approaching the baseline values., Limitations: This was a single-center study with a relatively small sample size. Further prospective, multicenter, randomized, and controlled studies with a larger sample size are required to confirm the effects in this study., Conclusions: During ultrasound-guided CPN for patients with inoperable upper abdominal cancers who failed medical management, a volume of 20 mL is as effective as 40 mL of 70% alcohol regarding pain control, opioid consumption, quality of life, and procedure-related complications.

Cyclizine (CYZ); an antiemetic compound; is widely misused for its euphoric or hallucinatory effects, either by oral or intravenous routes. The concomitant abuse of CYZ among addicted adolescents contributes to neuromuscular disorders that are life-threatening. Consequently, with the company of 1-Methylpiperazine (MPZ) and diphenylmethanol (DPM, Benzhydrol) as pharmacopoeia-reported CYZ impurities, a novel spectrofluorimetric assay for the detection of CYZ, has been established either in human plasma samples or in its parenteral formulation. The native fluorescence of CYZ has been investigated under various conditions. Different parameters affecting relative fluorescence intensity of CYZ including diluting solvent, surfactant, plasma protein solvent, and pH were studied and optimized. The linearity obtained between the fluorescence intensity at emission wavelength 350 nm after excitation at 244 nm and the corresponding CYZ concentrations was in the range 10-1000 ng/mL for measurement of CYZ either in pure form or in human plasma samples, with a appropriate correlation coefficient (r = 0.9999) and 3.10 ng/mL as the limit of detection and 9.41 ng/mL as the limit of quantitation. The suggested procedure was created and validated in accordance with ICH guidelines for quantification of CYZ either in its pure form or its dosage form, and FDA guidelines for the assay of CYZ in human plasma. Finally, in silico study and ADMET predictions were conducted for the studied drug impurities to estimate their pharmacokinetic behaviors. Background: Sulphasalazine (SZ) is a prodrug. Its active metabolite (mesalazine, MZ), which is also available in pharmaceutical formulations, and the major active metabolite of MZ (N-acetyl-5-aminosalicylic acid, AS) are commonly used for the treatment of inflammatory bowel diseases., Objective: Two accurate, precise, sensitive, and specific spectrophotometric methods were developed and validated for determination of the studied components., Methods: The first method is a modified ratio difference spectrophotometric method. In this method, SZ was determined by measuring the peak area from 410-500 nm, while MZ and AS were determined by measuring the difference of the selected amplitude values. The second method is a mean centering of ratio spectra spectrophotometric method., Results: The developed methods were linear over the concentration ranges of 2-35, 2-30 and 1-25 µg/mL for SZ, MZ and AS, respectively., Conclusion: The developed methods were validated according to International Conference on Harmonization guidelines. Soft conductive elastomers with low hysteresis over a wide range of stretchability are desirable in various applications. Such applications include soft sensors with a long measurement range, motion recognition, and electronic skin, just to name a few. Even though the measurement capability of the sensors based on soft materials has been greatly improved compared to the traditional ones in recent years, hysteresis in the loading and unloading states has limited the applications of these sensors, thereby negatively affecting their accuracy and reliability. In this work, conductive elastomers with near-zero hysteresis have been formulated and fabricated using 3D printing. These elastomers are made by combining highly stretchable dielectric elastomer formulations with a polar hydrophobic ionic liquid and polymerizing under ultraviolet light. High-performance piezoresistive sensors have been fabricated and characterized, with a 10-fold stretchability and low hysteresis (1.2%) over long-term stability (more than 10 000 cycles under cyclic stress) with a 20 ms response time. Additionally, the current elastomers displayed fast mechanical and electrical self-healing properties. Using 3D printing in conjunction with some of our structural innovations, we have fabricated smart gloves to show this material's wide range of applications in soft robots, motion detection, wearable devices, and medical care.

In a 90-day study, 32 growing lambs aged 3 months were utilized to evaluate the effects of various levels of spirulina dietary supplement on productive performance, nutritional digestibility, and meat quality in growing Najdi lambs. The lambs were put into 4 groups of 8 lambs each at random. The diet consisted of a total mixed ration (TMR) without spirulina (CONT), and the TMR diet supplemented with spirulina at the levels of 2 ppm (SPP2), 4 ppm (SPP4), and 8 ppm (SPP8). The treatment groups, especially SPP8, showed a significant (p < 0.05) increase in body weight and average daily gain (p < 0.05) compared to the CONT group. Dry matter intake and acid detergent fiber were also significantly (p ≤ 0.05) higher in SPP8 compared to other treated groups and CONT. The N absorption, N retention, and percentage of N digestibility coefficient were greater (P < 0.05) in lambs in the treatment groups than in the CONT. Blood biochemistry variables were not significantly (p ≤ 0.05) affected by the treatments, with the exception of the serum concentration of triglyceride and bilirubin. Carcass profile and meat quality, including back fat, body wall fat, and cooking loss, were increased significantly (p ≤ 0.05) with dietary spirulina. From the results of the present study, it was concluded that spirulina dietary supplementation at the level of 8 ppm increased weight gain, nutritional digestibility, nitrogen utilization, and meat composition in growing Najdi lambs., (© 2022. Point of care (POC), also identified as on-site testing, has evolved as a rapid and accurate technique for drug of abuse screening and analysis. The aim of this work is to detect tropicamide (TPC) abuse in biological fluids; we selected rat plasma as example. We developed a disposal miniaturized, portable, green, and budget-friendly POC solid-state electrochemical sensor based on potentiometric transduction. To attain that, an innovative microfabricated electrode modified with conducting polymer poly(3-octylthiophene) (POT) has been placed on sensitized printed circuit board (PCB). A two-stage optimization process was implemented to develop the fabricated electrode. The first stage of the optimization process depends on screening various ionophores in order to enhance the sensor selectivity towards tropicamide. Copper nanoparticles exhibited the highest selectivity towards TPC. The second stage was utilizing a polymer as an ion-to-electron transducer layer between the copper nanoparticles impregnated ion sensing membrane and the microfabricated solid-contact ion-selective electrode. This polymer was added to boost the stability of the potential drift (1.2 mV/h) due to the hydrophobic behavior of the POT, which precludes the formation of an aqueous layer at the Cu electrode/polymeric membrane interface and improve the limit of detection (1.1 × 10 -8  M). Nernstian potentiometric response was accomplished for TPC with a slope of 58.46 ± 0.43 mV/decade and E 0 ∼ 189.39 ± 2.12 over the concentration range 1.0 × 10 -7 to 1.0 × 10 -2  M. The suggested sensor's intrinsic figure of merits include a quick response time (13 ± 2 s) and long life time (45 days). The proposed sensor has been successfully employed in the selective determination of TPC in pharmaceutical formulations, and biological fluids. The chromatographic and lipophilicity characters of seven cephalosporins of different four classes (cephradine, cefaclor, cefprozil, cefixime, cefotaxime, ceftazidime and cefepime) were examined by salting out thin-layer chromatography (SOTLC). SOTLC using ammonium sulfate salt was employed to predict the lipophilicity of the proposed drugs via their retention behavior. The calculated R M0 values showed liner relationship with the molar concentration of ammonium sulfate in mobile phase in the range of 0.5-2.5 mol/L. Additionally, quantitative structure retention relationship (QSRR) was generated to figure out the relationship between the calculated chromatographic parameters (R M0 and C 0 ) and log P of the studied cephalosporins. Good correlations were found between the chromatographically obtained retention parameters (R M0 and C 0 ) and some molecular descriptors of the examined drugs. Furthermore, an efficient QSAR model was carried out using the calculated chromatographic parameters (R M0 and C 0 ) and log P of the studied cephalosporins to predict minimum inhibitory concentration (MIC) and blood brain barrier (BBB) penetration of the examined drugs. The study was extended to separate and quantify the selected antibiotics in their pure forms and pharmaceutical formulations. Normal phase thin layer chromatographic (NP-TLC) method using a usable developing system of acetone: methanol: water: ammonium hydroxide: glacial acetic acid (90: 10: 18: 3: 2, by volume) was successfully applied to resolve the studied cephalosporins. Linearity was achieved in the range of 0.2-3 µg/mL for most of the studied antibiotics. The developed SOTLC method can be considered as a good start alternative to reversed phase thin layer chromatography (RP-TLC) for prediction of the lipophilic properties of examined cephalosporins. Background: Predictive markers represent a solution for the proactive management of severe dengue. Despite the low mortality rate resulting from severe cases, dengue requires constant examination and round-the-clock nursing care due to the unpredictable progression of complications, posing a burden on clinical triage and material resources. Accordingly, identifying markers that allow for predicting disease prognosis from the initial diagnosis is needed. Given the improved pathogenesis understanding, myriad candidates have been proposed to be associated with severe dengue progression. Thus, we aim to review the relationship between the available biomarkers and severe dengue., Methodology: We performed a systematic review and meta-analysis to compare the differences in host data collected within 72 hours of fever onset amongst the different disease severity levels. We searched nine bibliographic databases without restrictive criteria of language and publication date. We assessed risk of bias and graded robustness of evidence using NHLBI quality assessments and GRADE, respectively. This study protocol is registered in PROSPERO (CRD42018104495)., Principal Findings: Of 4000 records found, 40 studies for qualitative synthesis, 19 for meta-analysis. We identified 108 host and viral markers collected within 72 hours of fever onset from 6160 laboratory-confirmed dengue cases, including hematopoietic parameters, biochemical substances, clinical symptoms, immune mediators, viral particles, and host genes. Overall, inconsistent case classifications explained substantial heterogeneity, and meta-analyses lacked statistical power. Still, moderate-certainty evidence indicated significantly lower platelet counts (SMD -0.65, 95% CI -0.97 to -0.32) and higher AST levels (SMD 0.87, 95% CI 0.36 to 1.38) in severe cases when compared to non-severe dengue during this time window., Conclusion: The findings suggest that alterations of platelet count and AST level-in the first 72 hours of fever onset-are independent markers predicting the development of severe dengue., Competing Interests: The authors have declared that no competing interests exist.

With the number of coronavirus disease 2019 (COVID-19) infected patients increasing all over the world, a large number of survivors have reported changes in their quality of life or experienced re-infection. So, we aimed to detect the percentage, type, and risk factors of persistent symptoms after improvement from acute COVID-19 infection and to detect the percentage of COVID-19 re-infection and degree of severity of the second infection. One hundred seventy-two (59 male, 113 female) patients who tested positive for severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) were followed up via mobile phone every 2 months for 8 to 10 months. After recovery, 105 patients (61%) (30 male, 75 female) reported one or more COVID-19 persistent symptoms. Fatigue, dyspnea, and depression were the most common persistent symptoms representing 37.3%, 22%, 22%, respectively. We found that age was independently related to the persistence of symptoms. During the follow-up, six females (3.5%) had laboratory-confirmed COVID-19 re-infection. Their mean age was 35.7 ± 11 years. The mean interval from the complete recovery of the first infection to the onset of the second one was 53 ± 22.2 days and ranged from 30 to 90 days. The second infection was milder in severity than the first infection in 83.33% of cases. There was a high percentage of patients who complained of persistent symptoms after recovery from COVID-19. Fatigue and headache were the most common persistent symptoms. Age was considered a risk factor for persistent symptoms. The present study was designed to study the effects of conventional and free range systems on egg quality attributes, fatty acid profile, and cholesterol contents of laying hen. Two hundred and forty R1 cross of Rhode Island Red (RIR) × Fyoumi (F) layers during the peak production were divided into two groups and reared in two different housing systems (free range vs. conventional systems). The statistical analysis of the results revealed that the housing system (modification of diet) significantly (p < 0.05) affected egg quality, fatty acid profile, and cholesterol contents. The highest (p < 0.05) whole egg and albumen weights were recorded for eggs produced in a conventional system and higher yolk weight and shell thickness were recorded in free range hens. Significantly (p < 0.05) lower concentration of saturated and higher concentration of mono- and polyunsaturated fatty acids were observed in eggs produced under a free range system. In addition, higher level of total omega-3 fatty acids and higher ratio of omega-6 to omega-3 fatty acids were observed in free range eggs as compared to those in the conventional system. From the results of the present study, we concluded that eggs produced from Rhode Island Red × Fyoumi laying hens in a free range system are qualitatively superior compared to those in the conventional system., (© 2021. Aim of the Study: We aimed to study liver function test abnormalities in our COVID-19 patients and factors affecting them and to evaluate whether liver function test abnormalities are related to the severity of COVID-19., Material and Methods: Our retrospective study included 118 patients who were SARS-CoV-2 positive. Their median age was 40 years. Fifty percent were male. Clinical and biochemical data were collected from patient records during the period from the start of June 2020 to the end of July 2020. Liver function test abnormalities included: alanine aminotransferase (ALT) > 40 U/l, aspartate aminotransferase (AST) > 40 U/l, serum albumin < 3.5 mg/dl, total bilirubin > 1.2 mg/dl, and international normalized ratio (INR) > 1.2., Results: Forty-four percent of COVID-19 patients had liver function test (LFT) abnormalities. In patients with severe SARS-CoV-2, AST, total bilirubin and INR levels were significantly higher than in patients with the non-severe disease. Levels of hemoglobin, C-reactive protein (CRP), erythrocyte sedimentation rate (ESR), serum ferritin, D-dimer, and serum glucose were significantly higher in SARS-CoV-2 patients with LFT abnormalities than those with normal liver function., Conclusions: LFT abnormalities are very common in SARS-CoV2 positive patients, especially those with the severe form. Levels of ESR, CRP, serum ferritin, and D-dimer were higher in COVID-19 patients with LFT abnormalities than those with normal LFT. Forty-eight growing Awassi lambs were used in a 70-day trial to investigate the effects of different levels of dietary sunflower hulls (SFH) on growth, rumen morphology, fiber digestibility and meat characteristics of lambs. Animals were randomly allocated to 4 groups with 3 replicates of 4 lambs each. The diet was composed of total mixed ration (TMR) without SFH (control group), and the TMR diet supplemented with SFH at a level of 5% (SFH5), 10% (SFH10) and 15% (SFH15). Lambs in the treatment groups had greater BW changes ( p = 0.04) and ADG ( p = 0.04) than the lambs in the control group. Intake of dry matter, acid detergent fiber (ADF) and neutral detergent fiber (NDF) were also significantly ( p < 0.05) higher in SFH15 compared to SFH10. Digestibility of ADL and empty stomach weight were also significantly ( p < 0.05) higher in SFH10 and SFH15, respectively. Cooking loss, blood total cholesterol and total protein decreased significantly ( p < 0.05) in SFH15. Ruminal lightness (L) and yellowness (b) also increased significantly ( p < 0.05) in SFH15. We concluded that the TMR diet supplemented with up to 15% SFH improved weight gain, digestibility, meat cooking loss and rumen color in Awassi lambs.

Two simple, sensitive and validated chromatographic methods were developed for quantitative determination of bromhexine hydrochloride (BHX) in presence of its major impurities, impurity B (IMB) and impurity C (IMC), as specified by British Pharmacopoeia. First method (I) was high-performance thin layer chromatography-densitometry at which the chromatographic separation was performed using silica gel plates and developing system consisted of hexane:acetone:ammonia solution (9:0.5:0.08, by volume) with ultraviolet scanning at 240 nm and linearity was achieved in the ranges of 0.40-10.00, 0.20-2.00 and 0.20-2.00 μg/band of BHX, IMB and IMC, respectively. Also, second chromatographic method (II) was high-performance liquid chromatography (HPLC) where the separation was carried out on C18 column at isocratic mode at flow rate 1.5 mL/min. The mobile phase consisted of methanol:water (90:10, v/v) adjusted to pH 2.5 with O-phosphoric acid and temperature was adjusted to 40°C. The scanning wavelength was 240 nm. The chromatographic run time was 6 min. Linearity of this method was achieved in the ranges of 4.00-40.00, 0.20-10.00 and 0.50-10.00 μg/mL for BHX, IMB and IMC, respectively. The validation of these chromatographic methods was made according to International Conference on Harmonization guidelines. These methods were successfully applied for determination of BHX in its pharmaceutical formulation. Background: Nutritional management in the dry period can alter body condition score (BCS) in dairy cows, a subjective measure of body fat. As such, differences in BCS during late-pregnancy not only mirror nutrient utilization by fat depots, but also can play important roles on the metabolic and hormonal environment. We investigated the association between cow BCS during late-pregnancy on developmental parameters and blood variables of neonatal calves. Forty-nine multiparous Holstein cows were retrospectively divided by prepartal BCS into normal BCS ≤3.25 (NormBCS; 3.02 ± 0.17, n = 30) or high BCS ≥3.75 (HighBCS; 3.83 ± 0.15, n = 19) groups. Plasma samples were collected from cows at - 10 d relative to parturition. Body weight, hip and wither height, hip width and body length were measured at birth and weekly through weaning (42 d of age) and until 9 weeks of age. Calf blood samples were collected from the jugular vein at birth (before receiving colostrum, 0 d), 24 h after first colostrum and at 7, 21, 42 and 50 d of age. The data were subjected to ANOVA using the mixed procedure of SAS. The statistical model included day, BCS, and their interactions., Results: Dry matter intake (kg/d or % of body weight) during the last 4 weeks of pregnancy was lower (P ≤ 0.06) in HighBCS cows. Plasma concentrations of fatty acids, ceruloplasmin, and nitric oxide were greater overall (P < 0.05) at d - 10 prior to calving in HighBCS cows, and they tended (P = 0.08) to have greater concentrations of reactive oxygen metabolites. Birth body weight was lower (P = 0.03) in calves born to dams with HighBCS. In addition, plasma concentrations of fatty acids, albumin and urea (P < 0.05) were greater in those calves. Although calves born to cows with HighBCS maintained a lower postnatal body weight (P = 0.04), hip and wither height, hip width, and body length, there was no difference (P > 0.05) in daily starter intake and average daily gain due to maternal BCS., Conclusions: Overall, results highlight an association between BCS during late-gestation on in utero calf development and postnatal growth. A high maternal BCS during late-gestation was associated with lower calf body weights, which could be due to lower maternal intakes and a state of inflammation and metabolic stress.

Vinburnine (VNB) is a vinca alkaloid used as a vasodilator to enhance cerebral circulatory insufficiency. It is a cyclic amide containing drug which is expected to be sensitive to hydrolytic degradation. The degradation profile of VNB was studied in this work following ICH recommendations for stability study. The drug was sensitive only to degradation with NaOH with the formation of the carboxylic acid derivative, identified by IR and 1 H NMR analyses as 2-((1S,12bS)-1-ethyl-1,2,3,4,6,7,12,12b-octahydroindolo[2,3-a] quinolizin-1-yl) acetic acid, (DEG). In this study five simple, smart and univariate stability indicating spectrophotometric methods were developed and validated for simultaneous determination of VNB and DEG for the first time. The developed methods include; Dual Wavelength Method (DWM), Dual Wavelength Resolution Method (DWRM), Factorized Absorbance Difference Method (FADM), Advanced Absorbance Subtraction Method (AASM), and Derivative Amplitude Factor Method (DAFM). These methods were capable of determination of VNB and DEG over the ranges of 1-30 and 3-50 µg/mL, respectively. The proposed methods were simple, smart, specific, and could be applied for analyzing synthetic mixtures of VNB and DEG and were successfully applied for determination of the drug in commercially available capsules. The obtained results of these methods were statistically compared with the reported HPLC one using student's-t and F- tests, where no significant difference was observed. This study investigated the effects of supplementing different levels of sunflower hulls (SFH) to the complete feed of late-gestation pregnant ewes on the nutritive and fatty acids profile of colostrum at three and 48 h postpartum. In this study, 24 primiparous lactating Naemi ewes were randomly selected at parturition from four dietary groups as follows: (1) C (control), total mixed ration (TMR; 0% SFH), (2) S12, TMR1 with 12% SFH (level 1), (3) S20, TMR2 with 20% SFH (level 2), and (4) S28, TMR3 with 28% SFH (level 3). The body condition scores were estimated 30 days prepartum. Colostrum samples were collected at three and 48 h postpartum to measure the nutritive values and fatty acids profiles. Data were analyzed as a complete randomized design with repeated measures and via Pearson's correlation and regression analyses. The results indicated a numerical correlation (R 2 = 0.42; p < 0.09) between the body condition score and both colostrum fat and total solids. SFH increased the colostrum fat % ( p < 0.05), especially for S12 and S20, following a cubic trend. Additionally, the colostrum from S12 and S20 ewes had a higher proportion of unsaturated fatty acids (USFAs), omega3 and lower levels of saturated fatty acids (SFAs), other than margaric acid (C17) and stearic acid (C18), SFA/USFA, and omega 6/omega 3. Furthermore, the regression analysis of the fatty acid classes and indices indicated a quadratic relationship between the parameters and SFH levels. The results confirm that the inclusion of SFH at levels greater than 20% may have a negative effect on some fatty acid parameters as a result of the high lignin intake. In conclusion, up to 20% SFH can be used in a complete feed for pregnant ewes without negative effects on the colostrum fat content and fatty acids profiles.

A reliable, selective and sensitive stability-indicating RP-HPLC assay was established for the quantitation of bromazepam (BMZ) and one of the degradant and stated potential impurities; 2-(2-amino-5-bromobenzoyl) pyridine (ABP). The assay was accomplished on a C18 column (250 mm × 4.6 mm i.d., 5 μm particle size), and utilizing methanol-water (70: 30, v/v) as the mobile phase, at a flow rate of 1.0 ml min-1. HPLC detection of elute was obtained by a photodiode array detector (DAD) which was set at 230 nm. ICH guidelines were adhered for validation of proposed method regarding specificity, sensitivity, precision, linearity, accuracy, system suitability and robustness. Calibration curves of BMZ and ABP were created in the range of 1-16 μg mL-1 with mean recovery percentage of 100.02 ± 1.245 and 99.74 ± 1.124, and detection limit of 0.20 μg mL-1 and 0.24 μg mL-1 respectively. BMZ stability was inspected under various ICH forced degradation conditions and it was found to be easily degraded in acidic and alkaline conditions. The results revealed the suitability of the described methodology for the quantitation of the impurity (ABP) in a BMZ pure sample. The determination of BMZ in pharmaceutical dosage forms was conducted with the described method and showed mean percentage recovery of 99.39 ± 1.401 and 98.72 ± 1.795 (n = 6), respectively. Structurally related carbamazepine (CBZ) and oxcarbazepine (OX) are two of the most commonly used antipsychotic drugs. The main impurities of CBZ, as described in both the USP and the BP, are iminodibenzyl (IMD) and iminostilbene (IST). Meanwhile, for non-pharmacopeial OX, the declared impurities include CBZ and IST. Prescribed oral suspensions of CBZ and OX contain additives including methyl paraben (MP), propyl paraben (PP) and sorbic acid (SA) as preservatives. An HPTLC method was introduced and developed for resolving the interference between CBZ, OX, their impurities, and the suspension additives in a single run, in addition to their quantitation with a high sensitivity that satisfies the USP requirements for the detection and quantitation of drug impurities. In the developed HPTLC method, CBZ and OX were measured in the range of 40-4000 ng per band, while IMD, IST, MP, PP and SA were in the range of 20-2000 ng per band, using a mixture of hexane : ethylacetate : formic acid : acetic acid (8 : 2 : 0.5 : 0.3, by volume) and UV scanning at 254 nm. The greenness profile of the method was evaluated by two different tools, the analytical Eco-Scale and the Green Analytical Procedure Index (GAPI), then a comparison between their results was conducted. The aim of this work is to develop and validate a simple, rapid, highly sensitive and selective spectrofluorometric method for determination of Metformin HCl (MFH) and Glibenclamide (GLB) in their binary mixture without prior separation. The proposed method based on measuring the native fluorescence intensity of GLB at λ emission  = 348 nm, after excitation at λ excitation  = 226 nm and measuring the fluorescence intensity of the fluorescent product produced from the derivatization of MFH using 9,10-phenanthraquinone in alkaline media at λ emission  = 416 nm after excitation at λ excitation  = 240 nm. The proposed spectrofluorometric method allowed sensitive determination of the studied drugs with a limit of quantitation of 0.04 and 0.01 μg mL -1 for MFH and GLB, respectively, providing greater sensitivity than the reported one. Validation of the proposed method was carried out according to ICH guidelines with respect to linearity, accuracy, precision, and selectivity. An experiment was conducted in a completely randomized design to evaluate the effect of including graded levels (0, 20, 30, 40 and 50% of diet) of dried distiller's grains with solubles (DDGS) on growth performance, slaughter parameters, blood serum metabolites and rumen microbiota in weaned Najdi male lambs. Thirty-five lambs, initial body weight of 33.45 ± 0.75 kg, and approximately three-month old were used in a 94-day feeding experiment. Performance measurements were conducted biweekly and blood samples were collected monthly. Inclusion of DDGS in the diets of growing Najdi lambs at levels up to 50% did not affect body weight gain (BWG) compared with the lambs fed the control diet (CON, 0% DDGS). Lambs fed the 50% DDGS diet consumed less feed compared with lambs in other groups (98 vs 112.5 kg DM) but had no adverse effect on BWG. Rumen pH values at 0, 6, 12 and 18 h post feeding and concentrations of blood serum total proteins, glucose, triglycerides, urea-N or albumin were similar across treatments. Slaughter parameters including slaughter weight, hot and cold carcass weights and dressing % (hot and cold carcass) were not affected by the treatments. There was no difference in the weights of full compartmental stomach and intestines, liver, omental fat, Kidney Knob and Channel Fat (KKCF) and tail fat between DDGS treatments and CON. The study concluded that the inclusion of DDGS in the diets of growing Najdi lambs had no adverse effects on growth performance and slaughter parameters. This work presents a sensitive, accurate and selective RP-HPLC method for simultaneous determination of cyproheptadine HCl (CPH), its impurity B (dibenzosuberone) and CPH oxidative degradation product (10,11-dihydroxy-dibenzosuberone) in bulk powder and in pharmaceutical formulation. The RP-HPLC method depends on isocratic elution using C8 column and mobile phase consisting of 0.05 M KH2PO4 buffer:methanol (35:65, v/v, pH = 4.5) at a flow rate of 2 mL/min, and the eluant was monitored at 245 nm. Good resolution was obtained with tR values of 3.05, 7.54 and 6.17 min for CPH, impurity and oxidative degradate, respectively. The proposed method has been validated as per ICH guidelines using pure forms of CPH, its impurity and degradation product in pharmaceutical formulation with an accuracy of 100.48, 100.16 and 100.11, respectively. Additional spiking experiments yielded an accuracy of 100 ± 1.6%. Repeatability and intermediate precision results indicated acceptable low <2% RSD values. Moreover, the developed method's statistical results were favorably compared to the previously reported method results regarding both accuracy and precision. Background Self-care and peritoneal dialysis (PD) benefits have been underutilized in patients with end-stage renal disease (ESRD). The pre-dialysis education program (PDEP) has been generally introduced as an acceptable tool in increasing the rates of PD and has been reportedly recommended for ESRD patients as part of the introduced care. We aim to study the effect of PDEP on ESRD and whether they would prefer PD of center-based hemodialysis (HD). Methods This is a retrospective cohort study that was done at King Fahad Armed Forces Hospital in Jeddah, Saudi Arabia, in the dialysis center. Data were collected on patients and included demographics, preference of renal replacement therapy modality, and other possible factors that may affect patient choices such as educational level, economic status, and age. Results A total of 213 ESRD patients that met our criteria were included, with a total of 75 patients receiving PDEP. Out of those who received the PDEP, 57.3% and 42.7% of patients decided to perform HD and PD, respectively. There was a significant impact of PDEP on reducing HD choice [OR (95% CI) = 0.11 (0.05-0.24); P-value < 0.001]. Infections did not occur in 50.5% of the included patients while 45.8%, 3.3%, and 0.5% had central line-associated bloodstream infection (CLABSI), other infections, and peritonitis, respectively. Most of the PD patients (81.8%) did not have an infection as compared to 42.3% of the HD patients. HD was also associated with increased admission days [OR (95% CI) = 1.27 (1.07-1.51); P-value = 0.007].
