This retrospective case-control study to assess the impact of Cyclin D1 and CDK4 expression along with EBV infection with Hodgkin’s lymphoma (HL).80 lymph node included in this study; (40) formalin-fixed, paraffin- embedded biopsies from Hodgkin’s lymphoma (HL), and (40) apparently healthy lymph nodes as control group. EBV detection was done by CISH whereas IHC detection system was used to evaluate both Cyclin D1 & CDK4 gene expression. The positive –EBERs CISH reactions detected in 16 / 40 (40 %) of Hodgkin lymphoma tissues while positive - EBERs reactions detected in 3 out of 40 tissues (7.5%) in control lymph nodes tissues. Statistically was found highly significant between study groups (P value = < 0.05). Cyclin D1 & CDK4 -IHC were detected in 40% and 52.5% of HL, respectively. Statistically, significant differences noticed among negative, low and moderate scoring of tissues (P<0.05). Significant EBV gene expressions along with higher percentage of tissues in which Cyclin D1 & CDK4 genes being expressed may be indicator their possible role in the pathogenesis and / or the carcinogenesis of a subset of our Hodgkin’s lymphoma cases. Background: Viral, dietary and genetic factors are implicated in the etiology ofnasopharyngeal cancer, a rare type of head and neck cancers. Unlike other viruses, HHV-8 encodes several human cytokines homologues and regulatory genes that play important roles in the viral pathogenesis. Objective: To analyze the rates of HHV-8 infection in tissues obtained from a group of patients with nasopharyngeal carcinoma and inflammatory nasal polyps (INP) . Patients and Method: One hundred- thirty formalin-fixed, paraffin- embedded nasopharyngeal carcinoma and nasal inflammatory polyps tissues enrolled in this study; 65 nasopharyngeal tissue biopsies from nasopharyngeal carcinoma; 35 tissue biopsies from nasal inflammatory polyps and 30 nasopharyngeal tissues with unremarkable pathological changes, as apparently healthy tissuecontrol. Detection of HHV-8 was done by chromogenic in situ hybridization (CISH) technique detection system. Results: In nasopharyngeal carcinoma tissues, the HHV-8- DNA positive CISH reactions were detected in 23.1% while in nasal inflammatory polyps tissues HHV-8-positive CISH reactions were found in 8.6% of the examined tissues. The correlation between HHV-8 and NPC & INP was highly significant (P= 0.001). Conclusion: Significant HHV-8 detection in nasopharyngeal carcinoma & inflammatory nasal polypstissues could point for their possible role in either pathogenesis or carcinogenesis of both these lesions. Systemic lupus erythematosus is a chronic, systemic, idiopathic autoimmune disease. One of suggested environmental mechanisms leads to development Systemic lupus erythematosus is infection with Epstein barr virus. The objectives of the study are First, detection, quantification of Epstein–barr virus in peripheral blood of Systemic lupus erythematosus patients compared to control. Second, estimation mRNA level of latent, lytic genes and compare them with control groups. Third, the comparison is between the rate of EBV detected and level of gene expression of latent and lytic phase with severity of the disease. The current study investigated on Lupus patients during period from December 2018 to March 2019. Total of 40 patients involved in this study.Blood samples are taken from Baghdad Teaching Hospital of Medical City.On the other hand, 40 blood samples were collected from apparently healthy subjects, as control from Alimamein Alkadhimein Medical city. 11/40 (27.5%) of Lupus patients were with Epstein Barr virus at mean viral load 815.72 copy/ml. And rate of viral detection in blood of patients group was highest in severe cases rather than in less severe ones. Only 2/40 (5%) of control subjects were with Epstein Barr virus at much lower mean viral load, 64.75 copy/ml. Expression of the latent genes in patients versus control groups, was 100% versus 77.5% for EBNA2, 50% versus 15% for EBNA3C, 82.5% versus 97.5% for LMP-1, and 85% versus 75% for EA/D, respectively. EBNA2 expression showed significant direct proportional correlation with viral load.And the higher expression of these latent genes of EBV was associated with severity of the disease as well. Increased rate of Epstein Barr virus DNA detection in systemic lupus erythematosus group compared to control group and higher rate of viral DNA detection within severe cases might indicate possible defect in controlling viral infection. And also increased positive EBNA-2 and EBNA-3C expression in Lupus group rather than control group indicates that these proteins of the Epstein Barr might have ability to disrupt the normal immune system and might trigger and/or promote the autoimmune status.
