Recently time series forecasting has become one of the prime application areas of climatology, economics and industries. Many research works are conducted to forecast the time series more accurately. But few of them are concentrated on predicting the time series over an extended future horizon, and there is also a scope to improve their forecasting accuracy. This work proposes a multi-step-ahead foresting method to produce a stable and accurate forecasting result for the extended future horizon. Firstly, a deep learning-based forecasting model is proposed to predict the time series. Secondly, a fuzzy time series-based error correction model is implemented to enhance the prediction performance of the deep learning model. Here to optimize all the fuzzy time series (FTS) parameters in an integrated way, an integrated butterfly optimization (FTS-IBO) algorithm is proposed. In this study, two different types of real-world multivariate time series datasets are used to analyze the forecasting performance of the proposed model. The performance of the proposed FTS-IBO algorithm is compared with the traditional butterfly optimization (FTS-BO) algorithm. The experimental results show that the FTS-IBO technique is superior to the FTS-BO technique. The forecasting performance of the proposed model has also compared the other state-of-the-art models, and the simulation results exhibit that the proposed model produces a more accurate prediction performance for multi-step-ahead time series forecasting problems compared to other models.

Knowledge, Attitudes, and Practices (KAP) studies offer valuable insights into the socio-cultural, psychological, and behavioral determinants of nutrition. This study aims to assess nutrition KAP during pregnancy and lactation in selected areas of Adilabad and Kumaram Bheem Asifabad districts, Telangana, India. A cross-sectional KAP survey was conducted on 536 individuals from February to March 2020, employing tablet-based data collection for pregnant women, lactating mothers, young mothers, and frontline staff. Departing from the standard equal-weight approach, we assigned different weights to responses based on existing literature and stakeholder consultation, acknowledging the varying significance of each response. This novel approach aims to mitigate the risk of overestimating or underestimating factors, thereby designing more effective policy interventions tailored to societal complexities. The results reveal that regarding the three micronutrients (Iron, vitamin A, and Iodine), the knowledge levels of pregnant women, lactating mothers, and young mothers were low, that is, less than 50% when compared with the functionaries/frontline staff. Pregnant women scored low on attitudes regarding micronutrients compared to lactating mothers, young mothers, and functionaries/frontline staff. Attitudes translate into practices; however, the data revealed that pregnant women, lactating mothers, and young mothers were not adopting appropriate dietary and nutrition practices except for the micronutrient iodine. The inadequate knowledge of all categories of respondents indicates a gap in nutrition literacy and education. The results point to the urgent need for policy action for nutrition education interventions among the vulnerable population.

Amid rapid urbanization in Kolkata, a city in India, forecasting air pollution, particularly PM2.5 concentrations, stands as a critical challenge. In response, this study introduces a novel approach harnessing a hybrid data processing component that enables lightweight spatiotemporal models, including ConvLSTM and BiConvLSTM, to demonstrate noteworthy competitiveness. This component addresses challenges related to data availability, resource-intensive training, and the integration of meteorological factors. Leveraging a relatively small yet comprehensive dataset focusing on the Kolkata region (from February 2, 2020, to April 11, 2023), this method effectively captures nuanced pollution dynamics with spatial and temporal considerations. The results of this approach show the BiConvLSTM model consistently outperformed several models, demonstrating its superiority at stations like Bidhannagar and Fort William, with R2 values 0.901 and 0.899 respectively. The ConvLSTM model, while not as dominant overall, exhibited significant advantages in specific scenarios, particularly evident in R2 values reaching up to 0.890 at Jadavpur and 0.887 at Victoria. These findings highlight the efficacy of the proposed hybrid data processing component in enabling lightweight spatiotemporal models to achieve competitive performance in forecasting PM2.5 concentrations across urbanized landscape of Kolkata.

BackgroundPost-operative infections are a common cause of morbidity following major surgery. Little is understood about how major surgery perturbs immune function leading to heightened risk of subsequent infection. Through analysis of paired blood samples obtained immediately before and 24 h following surgery, we evaluated changes in circulating immune cell phenotype and function across the first 24 h, to identify early immune changes associated with subsequent infection.MethodsWe conducted a prospective observational study of adult patients undergoing major elective gastrointestinal, gynecological, or maxillofacial surgery requiring planned admission to the post-anesthetic care unit. Patients were followed up to hospital discharge or death. Outcome data collected included mortality, length of stay, unplanned intensive care unit admission, and post-operative infections (using the standardized endpoints in perioperative medicine–core outcome measures for perioperative and anesthetic care criteria). Peripheral blood mononuclear cells were isolated prior to and 24 h following surgery from which cellular immune traits including activation and functional status were assessed by multi-parameter flow cytometry and serum immune analytes compared by enzyme-linked immunosorbent assay (ELISA).ResultsForty-eight patients were recruited, 26 (54%) of whom developed a post-operative infection. We observed reduced baseline pre- and post-operative monocyte CXCR4 and CD80 expression (chemokine receptors and co-stimulation markers, respectively) in patients who subsequently developed an infection as well as a profound and selective post-operative increase in CD4+ lymphocyte IL-7 receptor expression in the infection group only. Higher post-operative monocyte count was significantly associated with the development of post-operative infection (false discovery rate < 1%; adjusted p-value = 0.001) with an area under the receiver operating characteristic curve of 0.84 (p < 0.0001).ConclusionLower monocyte chemotaxis markers, higher post-operative circulating monocyte counts, and reduced co-stimulatory signals are associated with subsequent post-operative infections. Identifying the underlying mechanisms and therapeutics to reverse defects in immune cell function requires further exploration.

Abstract Computational catalysis is playing an increasingly significant role in the design of catalysts across a wide range of applications. A common task for many computational methods is the need to accurately compute the adsorption energy for an adsorbate and a catalyst surface of interest. Traditionally, the identification of low-energy adsorbate-surface configurations relies on heuristic methods and researcher intuition. As the desire to perform high-throughput screening increases, it becomes challenging to use heuristics and intuition alone. In this paper, we demonstrate machine learning potentials can be leveraged to identify low-energy adsorbate-surface configurations more accurately and efficiently. Our algorithm provides a spectrum of trade-offs between accuracy and efficiency, with one balanced option finding the lowest energy configuration 87.36% of the time, while achieving a ~2000× speedup in computation. To standardize benchmarking, we introduce the Open Catalyst Dense dataset containing nearly 1000 diverse surfaces and ~100,000 unique configurations.

Solidification cracking is a prevalent defect in welding aluminium alloys which can be effectively mitigated by employing grain refinement strategies. This paper investigates the feasibility of tailoring the weld zone grain structure of 6xxx series aluminium alloys using an adjustable ring mode (ARM) laser beam to reduce the centreline cracking susceptibility. An in-process monitoring approach, comprising a high-speed camera and an ad-hoc Digital Image Correlation, was adopted to examine the strain development during laser welding on a self-restraint test rig. Results revealed that transverse strain in the weld centre exhibits a parabola-like relationship with the core/ring power ratio, with the minimum strain determined at the power ratio of ∼1.5, regardless of the total power employed. The microstructural analysis demonstrated that increasing the core/ring power ratio can lead to the refinement of columnar and equiaxed grains. Furthermore, the formation of secondary equiaxed grains is promoted when the power ratio increases, and this is proved to be related to the characteristic of the ARM laser by the like-for-like comparison. Overall, a fine-tuned power ratio of 1.5 is determined in the studied material and welding configuration for achieving a centreline crack free weld, benefiting from the improved microstructure and reduced thermal strain.

Abstract Distraction disrupts Working Memory (WM) performance, but how the brain filters distraction is not known. One possibility is that neural activity associated with distractions is suppressed relative to a baseline/passive task (biased competition). Alternatively, distraction may be denied access to WM, with no suppression. Furthermore, behavioural work indicates separate mechanisms for ignoring distractions which occur (1) while we put information into WM (Encoding Distraction, ED) and (2) while we maintain already encoded information during the WM delay period (Delay Distraction, DD). Here we used fMRI in humans to measure category-sensitive cortical activity and probe the extent to which ED/DD mechanisms involve enhancement/suppression during a WM task. We observed significant enhancement of task-relevant activity, relative to a passive view task, which did not differ according to whether or when distractors appeared. For both ED and DD we found no evidence of suppression, but instead a robust increase in stimulus specific activity in response to additional stimuli presented during the passive view task, which was not seen for the WM task, when those additional stimuli were to be ignored. The results indicate that ED/DD resistance does not necessarily involve suppression of distractor-related activity. Rather, a rise in distractor-associated activity is prevented when distractors are presented, supporting models of input gating, and providing a potential mechanism by which input-gating might be achieved.

Introduction: Biologic aggressiveness of OSCC (Oral Cavity Squamous Cell Carcinoma), has intrigued research in various prognosticating histopathological markers over past few decades. DOI (Depth of Invasion) is one such histopathological factor which affects outcomes and was included in the AJCC 8th edition TNM staging. Pattern of Invasion (POI) has been widely reported as an adverse prognostic factor associated with higher locoregional failure and poor prognosis. However, these factors are not utilized for treatment decision making and for outcome assessment. Materials and methods: This is a retrospective analysis of 320 patients with OSCC who underwent treatment, from October 2018–February 2020. Clinic demographic details were extracted from electronic medical records. Univariate and multivariate analysis was done for the parameters. WPOI (Worst Pattern of Invasion) was correlated with all histopathological prognostic factors. Survival analysis was done using Kaplan Meier for WPOI type's I–V. DFS (Disease free Survival) was evaluated for different grades of WPOI. Results: We analyzed the results comparing, early and advanced T (Tumor) stages, cohesive WPOI I-III, non-cohesive WPOI IV-V. Univariate analysis showed a significant association of T-stage (p = 0.001), N (Nodal) -stage (p = 0.002), DOI (p = 0.008), PNI (Peri-neural invasion) (0.001) and Tumor differentiation Grade (p = 0.001). On multivariate analysis, non-cohesive WPOI (IV & V) showed significant association with grade, PNI, DOI (0.002, 0.033 & 0.033 respectively). Non-cohesive WPOI had significantly higher locoregional failures and short DFS. Conclusion: Presence of invasive WPOI is associated with advanced T stage, poor differentiation, PNI, greater depth of invasion, and higher chances of nodal metastasis. WPOI is associated with poor DFS, treatment intensification in early stage disease with WPOI type IV & V may improve survival.

An attempt was made to study the humic acid (HA) quality and clay humus complex in order to generate valuable information regarding soil carbon (C) and recalcitrant carbon variations under conservation agriculture (CA) practices. It is worthwhile to mention that CA has got wider acceptance among researchers and farmers nowadays. A field experiment was conducted in an Inceptisol with three treatments, namely conventional tillage (CT), zero tillage (ZT) without residue and zero tillage with residue (ZT+R) in a maize (Zea mays L.)-wheat (Triticum aestivum L.) (M-W) and pigeon pea (Cajanus cajan L.)-wheat (P-W) cropping system at ICAR-Indian Agricultural Research Institute, New Delhi, with a view to characterize the HA by E4/E6 ratio and total acidity, and to specify the functional groups of clay humus complex. In ZT+R based treatments, lower E4/E6 ratio and total acidity of extracted HA showed higher degree of humification and stability of humic acid carbon (HA-C). The FTIR spectroscopy of the clay-humus complex (as extracted from soil) displayed the presence of a large number of functional groups in ZT+R treatment followed by ZT and CT. It was also observed that the yield of crops was also significantly higher in ZT+R than CT in both the cropping systems except in wheat crops in the M-W system. Therefore, it can be concluded that ZT+R has the potential to enrich the organic carbon (C) quality in soil and increase the aromaticity of HA, leading to carbon stabilization in soils.

The detection of high-energy neutrino signals from the nearby Seyfert galaxy NGC 1068 provides us with an opportunity to study nonthermal processes near the center of supermassive black holes. Using the IceCube and latest Fermi-LAT data, we present general multimessenger constraints on the energetics of cosmic rays and the size of neutrino emission regions. In the photohadronic scenario, the required cosmic-ray luminosity should be larger than ∼1%−10% of the Eddington luminosity and the emission radius should be ≲15 R _S in low- β plasma and ≲3 R _S in high- β plasma. The leptonic scenario overshoots the NuSTAR or Fermi-LAT data for any emission radii we consider, and the required gamma-ray luminosity is much larger than the Eddington luminosity. The beta-decay scenario also violates not only the energetics requirement but also gamma-ray constraints, especially when the Bethe–Heitler and photomeson production processes are consistently considered. Our results rule out the leptonic and beta-decay scenarios in a nearly model-independent manner and support hadronic mechanisms in magnetically powered coronae if NGC 1068 is a source of high-energy neutrinos.

The global ambitions to end hunger, achieve food security and improved nutrition, and promote sustainable agriculture demand a complex transition of the current food environments for enabling sustainable healthy diets. The food environments in Low and Middle-Income Countries (LMICs) have been experiencing rapid and dynamic transitions across the globe, necessitating a system-level thinking and systemic approach to understand opportunities for improvement. There is a need for valid, reliable measures of food and nutrition environments for reorienting thinking and data collection toward determinants of food demand, especially the food environment components, which are critical to understand the transforming food systems. Food environment transformations are urgently required to provide consumers with more affordable and nutritious diets capable of meeting social and environmental challenges. In the present perspective, we aim to provide insights on prioritizing research on understanding and designing evidence based inclusive food environments which is crucial for promoting long-term food system innovations that are economically, socially, and environmentally sustainable and, above all, contribute to sustainable healthy diets.

World agricultural production has seen significant growth in the past four decades, yet malnutrition remains a persistent problem, particularly in the global south and more so in the rural areas. Need for a holistic approach to food systems is becoming crucial in designing policies that support the transition to sustainable and healthy diets. The present study is aimed to understand the rural food environment in the Telangana state in southern India by analyzing the combination of external and personal factors affecting food choices, attitudes, and consumption behavior. We developed a scoring-based methodology to assess the external and personal domains and dimensions to understand the food environment. The results showed that rural households favored carbohydrate-rich food groups obtained mostly from their own production or subsidized sources. On the other hand, protein and micronutrient-rich food groups were neglected due to affordability and preference for taste, cultural factors, and the limitations of external food environment. The findings of this study provide a deeper understanding of the food environment in low and middle-income countries (LMICs) conext. By highlighting the interplay between agriculture, food environments, and nutrition outcomes, this study contributes to the ongoing effort to address the global malnutrition crisis and support the development of healthier and more sustainable food systems. These findings can be useful to guide policy actions towards achieving food security and nutrition in the rural regions where food environments are under rapid transitions in the LMICs.

Groundnut crop is one of the major sources of financial and food security for a large number of Tanzanian smallholder farmers. However, the production of groundnuts in Tanzania is underdeveloped, and yields are reportedly 2.5 to 3 times lower than in other African nations such as Nigeria. There are a number of factors that contribute to lower yields including the cultivation of outdated plant varieties, increased climate variability, the infestation of pests and diseases, and the use of outdated farming techniques. To analyze the scope for increasing groundnut production, this study investigates and compares the technical efficiencies (TEs) and technological gap ratios (TGRs) in Tanzania's four main groundnut-producing regions, namely the Central zone, Lake Zone, Southern zone, Southern highland zone, by using a two-step meta frontier model. We used ICRISAT data collected under the Tropical Legume-III project during 2017–18. Our results show a very low level of technical efficiency of groundnut production in the regions and significant regional differences in TEs, TGRs, and Meta Technical Efficiencies (MTEs). The study identifies a tremendous scope to increase groundnut productivity and production in Tanzania by enhancing its production efficiency and the key drivers that may help harness this potential.

Insect artifacts produced by necrophagous adult flies over a variety of depositional surfaces are problematic in crime scene investigations. Assessment of bloodstains through morphological features combined with contextual and presumptive chemical analysis, as traditional practices, are often inconclusive. Absorbance of bloodstains and substantial wicking on fabrics due to underlying texture and repeated stitch loops in the fabric makes the diagnosis even more difficult than stain morphology. Literature related to the application of modern techniques in the diagnosis of fly artifacts was critically reviewed and presented along with working efficacies and challenges. Apart from the traditional morphological comparison of bloodstains, findings on immunoassay, scanning electron microscopy and DNA-based molecular discrimination have been considered more confirmatory differential diagnosis in crime scenes. Scanning electron microscopic methods reveal distinctive features of small crystal-like deposits with fly artifacts absent in bloodstains. The immunodetection method tested positive with antiserum (anti-md3) against defecatory and regurgitate stains. Moreover, mitochondrial DNA cytochrome c oxidase I (CO1) gene-based molecular diagnosis hold promises in discrimination. However, these modern techniques may be applied along with traditional methods to overcome confusion as per suitability of sampling, following conservative and non-conservative approaches that may offer a real help to crime scene investigators.

Congenital diaphragmatic hernia is a life-threatening anomaly with the high mortality rate in infancy. Very rarely they remain silent and manifest in the adult life. Once they manifest, the symptoms vary and are not easy to diagnose. The entity also has associated problems in the form of hypoplastic lungs and loss of domain of the abdomen. In such cases, the diagnosis is most often based on clinical suspicion and radiological confirmation. The treatment options are open abdominothoracic procedures or laparoscopic procedures. Once hernia is reduced, the defect is closed and should be reinforced with prosthetic mesh. Here we present a case of congenital diaphragmatic hernia in a teenager presenting with cardiac symptoms. The challenges faced in managing the case are discussed.

The POTE family comprises 14 paralogues and is primarily expressed in Prostate, Placenta, Ovary, Testis, Embryo (POTE), and cancerous cells. The prospective function of the POTE protein family under physiological conditions is less understood. We systematically analyzed their cellular localization and molecular docking analysis to elucidate POTE proteins' structure, function, and Adaptive Divergence. Our results suggest that group three POTE paralogs (POTEE, POTEF, POTEI, POTEJ, and POTEKP (a pseudogene)) exhibits significant variation among other members could be because of their Adaptive Divergence. Furthermore, our molecular docking studies on POTE protein revealed the highest binding affinity with NCI-approved anticancer compounds. Additionally, POTEE, POTEF, POTEI, and POTEJ were subject to an explicit molecular dynamic simulation for 50ns. MM-GBSA and other essential electrostatics were calculated that showcased that only POTEE and POTEF have absolute binding affinities with minimum energy exploitation. Thus, this study’s outcomes are expected to drive cancer research to successful utilization of POTE genes family as a new biomarker, which could pave the way for the discovery of new therapies.

Ethical practice is always considered to be a safe practice. Pandemic is one situation where the execution of ethical principles becomes a bit difficult as differences of opinion are to be taken into consideration. We have faced dilemmas previously during the AIDS outbreak or Avium Influenza pandemic. Similar ethical dilemmas if not more we are facing during the COVID-19 pandemic. Ethical concerns are mainly related to duty of care towards COVID patients, scarce resource allocation, telemedicine service, and COVID death cases. In this article, we will discuss ethical issues in relation to the duty of care and scarce resource allocation. We will also highlight guidelines to minimize ethical dilemmas and the role of the administration to make ethically correct policies as much as possible. This will not only help us to face the current pandemic but also will help to create a root map for making policies if any such pandemic hits the community in future.

In this study, detailed structural and vibrational analysis of merocyanine has been investigated using Raman, surface enhanced Raman scattering (SERS) and surface-enhanced resonance Raman scattering (SERRS). The Raman, SERS and SERRS studies aided by density functional theoretical (DFT) calculations clearly established the prevalence of the trans- and cis-conformers of the protonated form of merocyanine (MCH+) in solid and acetonitrile solution. The binding characteristics of merocyanine adsorbed on nanostructured silver-coated films (SCFs) were investigated using excitation-dependent SERS, concentration-dependent SERRS and DFT studies. The conformers of merocyanine involved in the surface adsorption processes were recognized. The prominent marker bands observed at 1538 (ethylenic C=C stretch) and 1133 cm−1 (pyridinium C-N stretch) in the Raman spectrum of merocyanine in acetonitrile shifted to 1540 and 1126 cm−1, respectively on the nanostructured SCFs. The shift in the marker bands is associated with either the preferential binding of selective conformer or change in resonance equilibrium between the benzenoid and quinoid forms. The excitation wavelength dependent SERS spectrum infers that in addition to the major contribution from the electromagnetic enhancement, chemical (resonance) effect leads to the amplification of the 1540 cm−1 band. The concentration-dependent SERRS study showed maximum enhancement for the nanostructured SCFs functionalized with 1 μM concentration of merocyanine, indicative of monolayer coverage. For lower concentrations of merocyanine, the SERRS signal intensity reduced without any alteration in the peak positions. The SERRS study thus, revealed sub-nanomolar (0.1 nM) sensing of merocyanine using nanostructured SCFs with the analytical enhancement factor (AEF) of ∼ 1010 for the 1126 cm−1 and 1540 cm−1 Raman bands for MC concentration of 0.1 nM. In this study, combination of SERRS and DFT have clearly established the predominance of trans-MCH+ on the nanostructured silver surface with minor contribution from cis-MCH+, which remain exclusively bound to the surface via the phenoxyl ring O atom. This conformational surface selectivity of geometrical isomers of merocyanine using nanostructured surfaces can be further explored for energy efficient and economical separation of geometrical isomers.

In order to migrate over large distances, cells within tissues and organisms rely on sensing local gradient cues which are irregular, conflicting, and changing over time and space. The mechanism how they generate persistent directional migration when signals are disrupted, while still remaining adaptive to signal’s localization changes remain unknown. Here, we find that single cells utilize a molecular mechanism akin to a working memory to satisfy these two opposing demands. We derive theoretically that this is characteristic for receptor networks maintained away from steady states. Time-resolved live-cell imaging of Epidermal growth factor receptor (EGFR) phosphorylation dynamics shows that cells transiently memorize position of encountered signals via slow-escaping remnant of the polarized signaling state, a dynamical ‘ghost’, driving memory-guided persistent directional migration. The metastability of this state further enables migrational adaptation when encountering new signals. We thus identify basic mechanism of real-time computations underlying cellular navigation in changing chemoattractant fields.

It is now established that immune maturation occurs along a defined trajectory in the weeks and months after birth, but the immediate changes that occur within immune cells following birth is less clear. In this study, we monitored the immune profile of neonates via analysis of paired samples (n= 28) of cord blood and heel prick blood taken at varying times post term delivery by planned elective caesarean section. This paired approach accounted for the between-subject variability often observed over the first week of life. We identified rapid changes in immune cell populations within hours of birth. Specifically, we observed increased proliferation in effector T cells (but not regulatory T cells) that exhibited an increase in cytokine producing ability and also an increase in the percentage of CD3 T cells over this short time frame. This indicates that the mobilisation of the immune system is immediate post birth, presumably as a response to sudden exposure to the external environment, antigen or stress. Hence, immune development may start to occur more rapidly than previously proposed and as such, to study this trajectory, blood sampling should begin as soon after birth as possible.

Background: Cleft lip/palate (CLP) is a congenital orofacial anomaly appearing in approximately one in 700 births worldwide. While in high-income countries CLP is normally addressed surgically during infancy, in developing countries CLP is often left unoperated, potentially impacting multiple dimensions of life quality. Previous research has frequently compared CLP outcomes to those of the general population. But because local environmental and genetic factors contribute to the risk of CLP and also may influence life outcomes, such studies may downwardly bias estimates of both CLP status and correction. Objectives: This research represents the first study to use causal econometric methods to estimate the effects of both CLP status and CLP correction on the physical, social, and mental well-being of children. Methods: Data were collected first-hand from 1,118 Indian children, where we obtained first-hand data on height, weight, grip strength, cognitive ability, reading, and math ability. A professional speech therapist reviewed digital recordings of speech taken at the interview to obtain four measures of speech quality. Using this data, the household fixed-effects model we employ jointly estimates effects of CLP status and CLP surgical intervention. Findings: Our results indicate that adolescents with median-level CLP severity show statistically significant losses in indices of speech quality (-1.59σ), physical well-being (0.32σ), academic and cognitive ability (-0.37σ), and social integration (-0.32σ). We find strong evidence that CLP surgery significantly restores speech if performed before five years of age. The first surgeries performed on less-severe CLP cases significantly restore social integration, psychological well-being, academic/cognitive ability, and a general index of human flourishing. Conclusions: Children born with CLP in India face statistically significant losses in speech, physical health, mental health, and social inclusion. CLP surgical intervention significantly restores speech quality if carried out at an early age. Surgeries with the most significant impact on life outcomes are the first surgeries performed on less-severe CLP cases.

Introduction: Undergraduate medical students are adult learners facing various assessments through examination during academic course, but the result is unsatisfactory. This study aimed at identifyingthe gaps in academics and finding out areas for improvement among the identified factors affecting the students’ performance.Methods: A questionnaire survey prospective, cross-sectional, observational, questionnaire- based study was conducted among the 3rd semester undergraduate students (N=215) in a Government Medical College of Eastern India by census method using predesigned, pretested, validated tool designed by Ron Fry. A total of 200 students participated in this study. A closed questionnaire containing 28 questions with dichotomous options was distributed. Data collected were tabulated in MS Excel spreadsheet and evaluated according to the proposed guidelines on seven factors: concentration, comprehension, test anxiety, organization, research aptitude, computer skill, and taking notes. Statistical significance of the data distribution among different groups was estimated using Chi-square tests in MS Excel 2010 software, and p Results: Among 200 participants (response rate=93%), 196 were (Male=142, Female=54) accepted; the respondats’ age range was 18-22 years. Of them, 48 students obtained honours marks (≥75% inany subject). 137 (69.9%) students had lacunae in any of the abovementioned domains, comprehension (97.9%) being the highest. The major determining factors were test anxiety along with note taking and concentration. The differences between the males and females regarding concentration (P=0.008) and note taking (P=0.009) were statistically significant. Test anxiety was the differentiating factor (P=0.013) between honours and non-honours candidates.Conclusion: Researches worldwide have identified extrinsic, intrinsic, personal, and miscellaneous factors affecting the students’ performances. This is a multifaceted issue which can be managed individually. Few of the most important determinants were dealt with in this study. To perform well, every student should understand what to learn, what to remember, and how to represent in examinations. This study will help the education authorities to guide the students and implement Competency Based Medical Education (CBME) based on Attitude, Ethics, and Communication (AETCOM) module in India.

Abstract Background Cellular reprogramming in response to environmental stress involves alteration of gene expression, changes in the protein and metabolite profile for ensuring better stress management in plants. Similar to other plant species originating in tropical and sub-tropical areas, indica rice is highly sensitive to low temperature that adversely affects its growth and grain productivity. Substantial work has been done to understand cold induced changes in gene expression in rice plants. However, adequate information is not available for early gene expression, especially in indica variety. Therefore, a transcriptome profile was generated for cold shock treated seedlings of IR64 variety to identify early responsive genes. Results The functional annotation of early DEGs shows enrichment of genes involved in altered membrane rigidity and electrolytic leakage, the onset of calcium signaling, ROS generation and activation of stress responsive transcription factors in IR64. Gene regulatory network suggests that cold shock induced Ca2+ signaling activates DREB/CBF pathway and other groups of transcription factors such as MYB, NAC and ZFP; for activating various cold-responsive genes. The analysis also indicates that cold induced signaling proteins like RLKs, RLCKs, CDPKs and MAPKK and ROS signaling proteins. Further, several late-embryogenesis-abundant (LEA), dehydrins and low temperature-induced-genes were upregulated under early cold shock condition, indicating the onset of water-deficit conditions. Expression profiling in different high yielding cultivars shows high expression of cold-responsive genes in Heera and CB1 indica varieties. These varieties show low levels of cold induced ROS production, electrolytic leakage and high germination rate post-cold stress, compared to IR36 and IR64. Collectively, these results suggest that these varieties may have improved adaptability to cold stress. Conclusions The results of this study provide insights about early responsive events in Oryza sativa l.ssp. indica cv IR64 in response to cold stress. Our data shows the onset of cold response is associated with upregulation of stress responsive TFs, hydrophilic proteins and signaling molecules, whereas, the genes coding for cellular biosynthetic enzymes, cell cycle control and growth-related TFs are downregulated. This study reports that the generation of ROS is integral to the early response to trigger the ROS mediated signaling events during later stages.

Enteric fever is a severe systemic infection caused by Salmonella enterica serovar Typhi (ST) and Salmonella enterica serovar Paratyphi A (SPA). Detection of ST and SPA in wastewater can be used as a surveillance strategy to determine burden of infection and identify priority areas for water, sanitation, and hygiene interventions and vaccination campaigns. However, sensitive and specific detection of ST and SPA in environmental samples has been challenging. In this study, we developed and validated two methods for concentrating and detecting ST/SPA from wastewater: the Moore swab trap method for qualitative results, and ultrafiltration (UF) for sensitive quantitative detection, coupled with qPCR. We then applied these methods for ST and SPA wastewater surveillance in Kolkata, India and Dhaka, Bangladesh, two enteric fever endemic areas. The qPCR assays had a limit of detection of 17 equivalent genome copies (EGC) for ST and 25 EGC for SPA with good reproducibility. In seeded trials, the Moore swab method had a limit of detection of approximately 0.05–0.005 cfu/mL for both ST and SPA. In 53 Moore swab samples collected from three Kolkata pumping stations between September 2019 and March 2020, ST was detected in 69.8% and SPA was detected in 20.8%. Analysis of sewage samples seeded with known amount of ST and SPA and concentrated via the UF method, followed by polyethylene glycol precipitation and qPCR detection demonstrated that UF can effectively recover approximately 8, 5, and 3 log10 cfu of seeded ST and SPA in 5, 10, and 20 L of wastewater. Using the UF method in Dhaka, ST was detected in 26.7% (8/30) of 20 L drain samples with a range of 0.11–2.10 log10 EGC per 100 mL and 100% (4/4) of 20 L canal samples with a range of 1.02–2.02 log10 EGC per 100 mL. These results indicate that the Moore swab and UF methods provide sensitive presence/absence and quantitative detection of ST/SPA in wastewater samples.

The inspection of the quality of welds in battery packs plays an important role in ensuring safety during the manufacturing and operation of energy-storage devices in automotive vehicles during service. This research investigated the novel application of laser Doppler vibrometry, a widely used non-destructive optical technique for modal analysis, to the post-weld evaluation of micro-TIG-welded interconnections in lithium-ion supercells. The experimental modal analysis showed features in the modal models of the supercells that were unique to their welding conditions. The comparisons between the supercells showed an absence of linear correlations between the modal parameters and the welding current, as well as differences in the welding parameters obtained from the negative and positive terminals of the cylindrical cells. These findings suggested that the modal parameters of the supercells were more strongly influenced by the rigidity of the structural materials than by the localized compliance of the welded interconnections. While this investigation demonstrated a method for using laser Doppler vibrometry to distinguish between different welding conditions in lithium-ion supercells at a structural level, further development is needed to identify the weld quality of individual interconnections.

This study evaluated a potential transition of India’s power sector to 100% wind and solar energy sources. Applying a macro-energy IDEEA (Indian Zero Carbon Energy Pathways) model to 32 regions and 114 locations of potential installation of wind energy and 60 locations of solar energy, we evaluated a 100% renewable power system in India as a concept. We considered 153 scenarios with varying sets of generating and balancing technologies to evaluate each intermittent energy source separately and their complementarity. Our analysis confirms the potential technical feasibility and long-term reliability of a 100% renewable system for India, even with solar and wind energy only. Such a dual energy source system can potentially deliver fivefold the annual demand of 2019. The robust, reliable supply can be achieved in the long term, as verified by 41 years of weather data. The required expansion of energy storage and the grid will depend on the wind and solar energy structure and the types of generating technologies. Solar energy mostly requires intraday balancing that can be achieved through storage or demand-side flexibility. Wind energy is more seasonal and spatially scattered, and benefits from the long-distance grid expansion for balancing. The complementarity of the two resources on a spatial scale reduces requirements for energy storage. The demand-side flexibility is the key in developing low-cost supply with minimum curtailments. This can be potentially achieved with the proposed two-level electricity market where electricity prices reflect variability of the supply. A modelled experiment with price signals demonstrates how balancing capacity depends on the price levels of guaranteed and flexible types of loads, and therefore, can be defined by the market.

A lawful disposal of human dead body is only possible after establishment of proper and valid cause of death. If the cause is obscure, autopsy is the only mean of search. Inadequacy and unavailability of health care facility often makes this situation more complicated in developing countries where many deaths remain unexplained and proper mortality statistics is missing, especially for infant and children. Tissue sampling by needle autopsy or use of various imaging technique in virtopsy have been tried globally to find out an easier alternative. An exclusive and unique initiative, by limited autopsy through tissue biopsy and body fluid analysis, has been taken to meet this dire need in African and some of Asian developing countries, as worldwide accepted institutional data are even missing or conflicting at times. Traditional autopsy has changed little in last century, consisting of external examination and evisceration, dissection of organs with identification of macroscopic pathologies and injuries, followed by histopathology. As some population groups have religious objections to autopsy, demand for minimally invasive alternative has increased of late. But assessment of cause of death is most important for medico-legal, epidemiological and research purposes. Thus minimally invasive technique is of high importance in primary care settings too. In this article, we have made a journey through different autopsy methods, their relevance and applicability in modern day perspective considering scientific research articles, textbooks and interviews.

Genome and transcriptome sequencing data are extremely useful resources for researchers in carrying out biological experiments that involves cloning and characterizing genes. We are presenting here genome sequence data from different clades of life including photosynthetic prokaryotes; oomycetes pathogens; probiotic bacteria; endophytic yeasts and filamentous fungus and pathogenic protozoa Leishmania donovani. In addition, we are also presenting paired control and treated stress response transcriptomes of Cyanobacteria growing in extreme conditions. The Cyanobacterial species that are included in this dataset were isolated from extreme conditions including desiccated monuments, hot springs and saline archipelagos. The probiotic Lactobacillus paracasei was isolated from Indian sub-continent. The Kala azar causing protozoan Leishmania donovani, whose early infectious stage is also included in this dataset. The endophyte Arthrinium malaysianum was isolated as a contaminant has significant bio-remediation property. Our collaborators have isolated endophyte Rhodotorula mucilaginosa JGTA1 from Jaduguda mines, West Bengal, India infested with Uranium. Our collaborators have isolated a heterozygous diploid oomycetes pathogen, Phytophthora ramorum causing sudden oak death in CA, USA coast is also part of the data. These dataset presents a unique heterogeneous collection from various sources that are analyzed using “Genome Annotator Light (GAL): A Docker-based package for genome analysis and visualization” (Panda et al., 2019) and are presented in a web site automatically created by GAL at http://www.eumicrobedb.org/cglab. To assist in light weighting of electric vehicles by improving the volumetric and gravimetric energy density and the structural performance of the battery pack, a modelling methodology based on a macro finite element model of a pouch cell has been developed. This model treats the core cell structure as a homogeneous orthotropic honeycomb block with the pouch material being defined as an orthotropic fabric with compressive stress elimination. The model considers five compression and bending load cases simultaneously and allows a level of element discretisation that is computationally efficient and appropriate for inclusion in full vehicle and sub-system simulations. The methodology is scalable in that it can be applied to a range of chemistries, external geometries and internal cell constructions. When considering stacks of cells, the model is predictive for both lateral compression and three-point bend, but further work is required to improve the confined compression response.

Recent uptake in the use of lithium-ion battery packs within electric vehicles has drawn significant attention to the selection of busbar material and corresponding thickness, which are usually based on mechanical, electrical and thermal characteristics of the welded joints, material availability and cost. To determine joint behaviour corresponding to critical-to-quality criteria, this study uses one of the widely used joining technologies, ultrasonic metal welding (UMW), to produce tab-to-busbar joints using copper and aluminium busbars of varying thicknesses. Joints for electrical and thermal characterisation were selected based on the satisfactory mechanical strength determined from the T-peel tests. Electrical contact resistance and corresponding temperature rise at the joints were compared for different tab-to-busbar joints by passing current through the joints. The average resistance or temperature increase from the 0.3 mm Al tab was 0.6 times higher than the 0.3 mm Cu[Ni] tab, irrespective of busbar selection.

This study examines the performance of several active extension strategies, commonly known as 130/30, in the Australian equities market. A detailed analysis of the factors affecting performance is explored using Monte Carlo simulations based on eight years of historical returns for the constituents of the S&P/ASX 200 index under a variety of realistic cost assumptions. We find evidence of a statistically significant increase in performance of active extension strategies over equivalent long-only portfolios, holding all other factors constant. The observed increase is highest for managers with greater levels of skill, where any tracking error limit is high and total costs are low. This is one of the first studies in the Australian market and is expected to have a high degree of relevance to institutional investors considering active extension strategies.

Free iron typically leads to the formation of excess free radicals, and additional iron deposition in the liver contributes to the oxidative pathologic processes of liver disease. Many pharmacological properties of the insectivorous plant Drosera burmannii Vahl. have been reported in previous studies; however, there is no evidence of its antioxidant or hepatoprotective potential against iron overload. The antioxidant activity of 70% methanolic extract of D. burmannii (DBME) was evaluated. DBME showed excellent DPPH, hydroxyl, hypochlorous, superoxide, singlet oxygen, nitric oxide, peroxynitrite radical and hydrogen peroxide scavenging activity. A substantial iron chelation (IC50 = 40.90 ± 0.31 μg/ml) and supercoiled DNA protection ([P]50 = 50.41 ± 0.55 μg) were observed. DBME also displayed excellent in vivo hepatoprotective activity in iron-overloaded Swiss albino mice compared to the standard desirox treatment. Administration of DBME significantly normalized serum enzyme levels and restored liver antioxidant enzymes levels. DBME lowered the raised levels of liver damage parameters, also reflected from the morphological analysis of the liver sections. DBME also reduced liver iron content by 115.90% which is also seen by Perls' staining. A phytochemical analysis of DBME confirms the presence of various phytoconstituents, including phenols, flavonoids, carbohydrates, tannins, alkaloids and ascorbic acid. Alkaloids, phenols and flavonoids were abundantly found in DBME. An HPLC analysis of DBME revealed the presence of purpurin, catechin, tannic acid, reserpine, methyl gallate and rutin. Purpurin, tannic acid, methyl gallate and rutin displayed excellent iron chelation but exhibited cytotoxicity toward normal (WI-38) cells; while DBME found to be non-toxic to the normal cells. These findings suggest that the constituents present in DBME contributed to its iron chelation activity. Additional studies are needed to determine if DBME can be used as a treatment for iron overload diseases.

The antioxidant and restoration potentials of hepatic injury by Prunus nepalensis Ser. (Steud), a wild fruit plant from the Northeastern region of India, were investigated. The fruit extract (PNME) exhibited excellent antioxidant and reducing properties and also scavenged the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical (IC50 = 30.92 ± 0.40 μg/ml). PNME demonstrated promising scavenging potency, as assessed by the scavenging of different reactive oxygen and nitrogen species. Moreover, the extract revealed an exceptional iron chelation capacity with an IC50 of 25.64 ± 0.60 μg/ml. The extract induced significant improvement of hepatic injury and liver fibrosis against iron overload induced hepatotoxicity in mice in a dose-dependent manner, and this effect was supported by different histopathological studies. The phytochemical constitutions and their identification by HPLC confirmed the presence of purpurin, tannic acid, methyl gallate, reserpine, gallic acid, ascorbic acid, catechin and rutin. The identified compounds were investigated for their individual radical scavenging and iron chelation activity; some compounds exhibited excellent radical scavenging and iron chelation properties, but most were toxic towards normal cells (WI-38). On the other hand, crude PNME was found to be completely nontoxic to normal cells, suggesting its feasibility as a safe oral drug. The above study suggests that different phytochemicals in PNME contributed to its free radical scavenging and iron chelation activity; however, further studies are required to determine the pathway in which PNME acts to treat iron-overload diseases.

Enhancers-the non-coding regions of genomes’ are responsible for regulation of transcription of interacting genes. In different cell lines different regions act as enhancers. Enhancer-promoter interaction (EPI) models suggest that enhancer helps in the assembly of transcription factors along with RNA polymerase II and interacts with promoters to increase the expression of corresponding genes. During transcription, enhancer itself undergoes transcription giving rise to small RNAs, known as enhancer RNA. Presence of Transcription Start Sites (TSS) in annotated enhancer regions is also defined as active enhancers. Three different human cell-lines namely, Gm12878, K562 and H1-hesc which are normal, cancerous and stem cell-lines, respectively were studied. K-medoids algorithm was used to segregate EPI in all the cell-lines. Three clusters were derived on the basis of expression of enhancer, expression of their interacting promoters and distance between the two. Statistical t-test analysis showed that all clusters were different from each other. Cluster-1 (expression of enhancer Mean (eeMean) =59.12, Median (eeMedian) =12.09) and cluster-2 (eeMean=1799.9, eeMedian=1468) differ from each other on the basis of enhancer’s expression. Cluster-2 (distance mean=20521, eeMean=1799.9; distance median=7984, eeMedian=1468.5) was different from cluster-3 (distance mean=180798, distance median=162626) on the basis of distance and the expression of TSS at enhancer. Finally cluster-1 (distance mean=18030, distance median=6966) and cluster-3 (distance mean=180798, distance median=162626) differ from each other on the basis of distance. RNAseq analysis showed 7 upregulated genes in K562 compared to Gm12878. Further, EPI distributions of MYC, RAD23B and Insulin like growth factors showed similar pattern in K562 and H1hesc, and they were present in cluster-1. Whereas EPI of MDN1, CDKN1C, and eukaryotic translation elongation factor2 in K562 were present in cluster-1 and EPI of H1hesc were present in cluster-1 and cluster-3. EPI of Erythrocyte membrane protein were segregated into cluster-1 and cluster-3 for both K562 and H1hesc, whereas all of these interactions were absent in Gm12878. Overall these results suggests that enhancer activities are mainly responsible for carcinogenesis in K562 cell-lines otherwise absent in normal cell-lines.
