Formation of gas hydrates are a serious concern in oil and gas production systems. The formation and possible deposition of hydrates usually occur in suitable conditions, specifically as the gas-phases of reservoir fluid interact with water molecules. Hydrate formation represents a significant risk to process safety as it can result in the plugging of both pipes and instruments. Hydrates typically form in process where light hydrocarbons, water vapor and low temperatures or high pressures are present. We have studied the conditions under which hydrates form, how formation may be prevented and what can be done once hydrates have formed.

Introduction Patients with SLE (systemic lupus erythematosus) have a higher risk of infection due to dysregulated immune system as well as long-term use of immunosuppressants (IS). This could influence the risk of COVID-19 and its outcome. Methods We conducted a longitudinal prospective study across 15 rheumatology centres during the first wave of the pandemic to understand the risk factors contributing to COVID-19 in SLE patients. During the 6 months follow-up, those who tested positive for COVID-19, their clinical course and outcome information were recorded. Results Through the study period (April–December 2020), 36/1379 lupus patients (2.9%) developed COVID-19. On analysing the COVID-19 positive versus negative cohort during the study period, male gender (adjusted RR 3.72, 95% C.I. 1.85,7.51) and diabetes (adjusted RR 2.94, 95% C.I. 1.28, 6.79) emerged as the strongest risk factors for COVID-19, in the adjusted analysis. There was no significant influence of organ involvement, hydroxychloroquine, glucocorticoid dosage (prednisolone< 7.5 mg or ≥ 7.5 mg/day) or IS on the risk of COVID-19. There was only one death (1/36) among the lupus patients due to COVID-19. Conclusion Traditional risk factors rather than lupus disease process or IS influenced the risk of COVID-19 in our cohort.

The technical and fundamental or the time series analysis is used by the most of the stockbrokers while making the stock predictions. The programming language is used to predict the stock market using machine learning is Python. In this paper we propose a Machine Learning (ML) approach that will be trained from the available stocks data and gain intelligence and then uses the acquired knowledge for an accurate prediction. In this context this study uses a machine learning technique called Support Vector Machine (SVM) to predict stock prices for the large and small capitalizations and in the three different markets, employing prices with both daily and up-to-the-minute frequencies.

This paper proposes an algorithm that provides operational strategies for multiple tethered autonomous underwater vehicle (T-AUV) systems for entanglement-free navigation. T-AUVs can perform underwater tasks under reliable communication and power supply, which is the most substantial benefit of their operation. Thus, if one can overcome the entanglement issues while utilizing multiple tethered vehicles, the potential applications of the system increase including ecosystem exploration, infrastructure inspection, maintenance, search and rescue, underwater construction, and surveillance. In this study, we focus on developing strategies for task allocation, path planning, and scheduling that ensure entanglement-free operations while considering workload balancing among the vehicles. We do not impose restrictions on the size or shape of the vehicles at this stage; our primary focus is on efficient tether management as an initial work on the topic. To achieve entanglement-free navigation, we propose a heuristic based on the primal-dual technique, which enables initial task allocation and path planning while minimizing the maximum travel cost of the vehicles. Although this heuristic often generates sectioned paths due to its workload-balancing nature, we also propose a mixed approach to provide feasible solutions for non-sectioned initial paths. This approach combines entanglement avoidance techniques with time scheduling and sectionalization methods. To evaluate the effectiveness of our algorithm, extensive simulations were conducted with varying problem sizes. The computational results demonstrate the potential of our algorithm to be applied in real-time operations, as it consistently generates reliable solutions within a reasonable time frame.

Due to technological developments in last decade, new methods of seismic evaluation are in use like simulation based, algorithm based, probabilistic, software based etc. These developments have enabled researchers to move from linear to non-linear methods of analysis. Incremental dynamic analysis (IDA) is performance evaluation method where a suite of ground motions applied to structure are further scaled to particular levels of seismic intensity. Seismic fragility curves become significant in estimation of structures risk possibility from the point of view of potential earthquakes and helps in predicting the economic consequences for forthcoming earthquakes. The paper reflects IDA and seismic fragility analysis of ground storey + 7 floor (G + 7) reinforced concrete frame subjected to suite of eleven ground motions. Primary objective was to perform equivalent static and linear-dynamic analysis to meet the National and International codal requirements. Then, pushover analysis is carried out by introducing parametric auto hinges as per ASCE 41-13 tables. To carry out pushover analysis, both geometric and material non-linearity was introduced. Strong ground motions were selected as per suitable criteria of seismic intensity. IDA is then carried out as per SeismoStruct 2022 software and using IDA curves, the fragility analysis was carried out. The results of study found useful for researchers to predict the probability of damage of the structure under earthquakes.

Internal Combustion engines have the capacity to exploit its increase in performance. Part ofwhich can be boosted by the technical solution that is turbocharging. To optimize the turbocharging to react as fast as possible and to adjust the volumetric flow of exhaust gases in the turbine, different alterations to turbochargers are done. To optimally support downsizing of Internal Combustion engines, these alterations are necessary. In a VGT there are vanes on the compressor that change the A/R of the turbo as needed. VGTs show a great improvement over conventional single turbocharger. Sequential turbocharger has an edge over VGT, but they are much more complicated. A 2.2L sequential turbocharged engine is same in performance as a 2.7L VGT engine. Exhaust gas turbocharging is a major technology for reducing fuel consumption and emissions in internal combustion engines, improving engine performance while cutting CO2 emissions. There are also electric turbochargers that almost totally eliminate turbo lag by using a motor to spin the compressor and use a generator on the turbine to recover exhaust gas energy. This application reduces fuel usage by up to 5% for driving cycles. EATC reduced the time it took to reach the optimum boost level during a load increase by up to 30%. Turbocharging is a major technology for reducing fuel consumption and emissions in internal combustion engines, improving engine performance while cutting CO2 emissions. Hybrid Turbocharging application leads to remarkable improvement at lowend torques, better engine performance and increased overall engine efficiency which paves way for downsizing of Internal Combustion engines. Proximal humerus fractures account for about 4% to 5% of all fractures. Following distal forearm fracture, proximal humeral fracture is the second most common upper extremity fracture. Kirschner (K)-Wires, external fixation, tension band wiring, and rush pins, intramedullary nails, ORIF with plates, and shoulder hemiarthroplasty have all been offered as therapeutic options. The purpose of this study was to assess the functional outcome and complication rates following internal fixation of proximal humerus fractures using a locking compression plate. Patients over the age of 18 with closed proximal humerus fractures or open proximal humerus fractures (Gustilo and Anderson type I, Type II) received open reduction and internal fixation with locking compression plate in the current study. The participants in this study were 60 people who had a proximal humerus fracture and were treated with open reduction and a proximal humerus locking compression plate. Patients ranged in age from 20 to 85 years old, with a mean age of 51.14 +/- 17.30 years. The age group 36–55 years (37 percent) was the most prevalent, followed by 56–75 years (30 %). The majority of patients (76.7 %) were men who had been in automobile accidents on the right side (83 %) (63 %). According to the Neer classification, the most prevalent type of fracture was a three-part fracture (53 %). The maximum union time observed in this study was 16 weeks, with a minimum of 10 weeks. The average time to union is 12.52 +/- 1.14 weeks. The mean flexion at the end of six months was 121.660 +/- 19.84. The mean internal rotation was 57.330 +/- 8.48, the mean external rotation was 530 +/- 11.98, and the mean abduction was 1180 +/- 19.36. 75.28 +/- 9.66 [Mean +/- SD] is the Mean Constant score. Pain 14.10, ADL 18.26, range of motion 22.46, and power 20 were the mean scores observed on Constant Score for its various aspects. According to the Constant score, the functional outcome of the 60 patients was as follows: 8 patients had excellent outcomes (13%), 34 patients had good outcomes (56.7%), 16 patients had moderate outcomes (26.7%), and two patients had poor outcomes (26.7%) and (3 %). Internal fixation of proximal humerus fractures with a locking compression plate results in anatomical reduction and secure fixation, resulting in a favorable functional outcome.

Endosomal maturation is a critical and fundamental process for robust transport of cargo (such as activated receptors) to specific cellular compartments in a timely manner. The most prominent model of endosomal maturation involves a phosphoinositide-driven gain or loss of specific proteins at the level of individual endosomes, emphasising an autonomous and stochastic model of maturation. However, to date, direct whole cell-level measurements of absolute number of maturation events have not been performed, owing to limitations in fast, volumetric imaging. Here, we use lattice light-sheet imaging to track individual very early and early endosomes over the entire population. We demonstrate that direct inter-endosomal contact drives the maturation from very early (APPL1-positive) to early (EEA1-positive) endosomes. Using fluorescence lifetime, we show that this endosomal interaction is underpinned by the asymmetric binding of EEA1 to very early and early endosomes through the N- and C-termini, respectively. Thus, stochastic microtubule-mediated inter-endosomal interactions through EEA1 provide a mechanism to bring temporal and population-level control to the process of endosome maturation. Our findings indicate that APPL1- to EEA1-positive endosomal maturation is not a result of autonomous endosomal events but is driven by heterotypic EEA1-mediated endosomal interactions.

The nanosponge is the novel and emerging technology which plays a vital role in targeting drug delivery in the controlled manner. The nanosponges are tiny sponges having size of about a virus and it can easily penetrate through the skin. The tiny sponges circulate around the body until they reach to the specific target site and stick on the surface and start to release the drug in controlled manner. It releases the drug at specific site instead of circulating overall body it is more effective for targeted drug delivery system as they enhance solubility, bioavailability and reduces side effects. Nanosponges prevent drug and protein degradation. Both hydrophilic and lipophilic drugs are incorporated in Nanosponge. In this review mechanism, method of preparation, evaluation, stabilization and applications of nanosponges are discussed.

This study aimed to assess the distance and angular location of the common peroneal nerve (CPN) on axial magnetic resonance imaging (MRI) in the valgus knees and compare the measurements with those obtained from the control group. We compared the location of the CPN according to the type of alignment by performing a subgroup analysis. From January 2009 to December 2019, we identified 41 knees with preoperative MRI in patients who underwent total knee arthroplasty (TKA) for valgus deformity (valgus group). We performed one-to-two matched-pair analysis to a cohort of patients who underwent MRI but were not candidates for TKA (control group), according to sex and age. The valgus group was classified according to the grading system reported by Ranawat et al, and the control group was also subdivided according to the hip-knee-ankle (HKA) angle obtained from lower extremity scanography: neutral (–3 to +3 degrees from the neutral mechanical axis), valgus (> +3 degrees), and varus alignment (< –3 degrees). Distance between the CPN and posterolateral cortex of the tibia at the knee joint (distance J) and tibial cut level (distance C) were measured. Background We conducted this study to identify the influence of prolonged use of hydroxychloroquine (HCQ), glucocorticoids and other immunosuppressants (IS) on occurrence and outcome of COVID-19 in patients with autoimmune rheumatic diseases (AIRDs). Methods This was a prospective, multicenter, non-interventional longitudinal study across 15 specialist rheumatology centers. Consecutive AIRD patients on treatment with immunosuppressants were recruited and followed up longitudinally to assess parameters contributing to development of COVID-19 and its outcome. Results COVID-19 occurred in 314 (3.45%) of 9212 AIRD patients during a median follow up of 177 (IQR 129, 219) days. Long term HCQ use had no major impact on the occurrence or the outcome of COVID-19. Glucocorticoids in moderate dose (7.5–20 mg/day) conferred higher risk (RR = 1.72) of infection. Among the IS, Mycophenolate mofetil (MMF), Cyclophosphamide (CYC) and Rituximab (RTX) use was higher in patients with COVID 19. However, the conventional risk factors such as male sex (RR = 1.51), coexistent diabetes mellitus (RR = 1.64), pre-existing lung disease (RR = 2.01) and smoking (RR = 3.32) were the major contributing risk factors for COVID-19. Thirteen patients (4.14%) died, the strongest risk factor being pre-existing lung disease (RR = 6.36, p = 0.01). The endosomal system provides rich signal processing capabilities for responses elicited by growth factor receptors and their ligands. At the single cell level, endosomal trafficking becomes a critical component of signal processing, as exemplified by the epidermal growth factor (EGF) receptors. Activated EGFRs are trafficked to the phosphatase-enriched peri-nuclear region (PNR), where they are dephosphorylated and degraded. The details of the mechanisms that govern the movements of stimulated EGFRs towards the PNR, are not completely known. Here, exploiting the advantages of lattice light-sheet microscopy, we show that EGFR activation by EGF triggers a transient calcium increase causing a whole-cell level redistribution of Adaptor Protein, Phosphotyrosine Interacting with PH Domain And Leucine Zipper 1 (APPL1) from pre-existing endosomes within one minute, the rebinding of liberated APPL1 directly to EGFR, and the dynein-dependent translocation of APPL1-EGF-bearing endosomes to the PNR within ten minutes. The cell spanning, fast acting network that we reveal integrates a cascade of events dedicated to the cohort movement of activated EGF receptors. Our findings support the intriguing proposal that certain endosomal pathways have shed some of the stochastic strategies of traditional trafficking and have evolved processes that provide the temporal predictability that typify canonical signaling., York et al employ lattice light-sheet microscopy to study the initial events following activation of the epidermal growth factor receptor (EGFR) in whole live cells. They find that APPL1 relocates from pre-existing endosomes, triggered by calcium, to EGFR puncta which results in a cohort, dynein mediated movement of EGFR towards the peri-nuclear region of the cell, expanding our understanding of EGFR trafficking dynamics.

BackgroundMembrane protrusions that occur on the dorsal surface of a cell are an excellent experimental system to study actin machinery at work in a living cell. Small GTPase Rac1 controls the membrane protrusions that form and encapsulate extracellular volumes to perform pinocytic or phagocytic functions.ResultsHere, capitalizing on rapid volumetric imaging capabilities of lattice light-sheet microscopy (LLSM), we describe optogenetic approaches using photoactivable Rac1 (PA-Rac1) for controlled ruffle generation. We demonstrate that PA-Rac1 activation needs to be continuous, suggesting a threshold local concentration for sustained actin polymerization leading to ruffling. We show that Rac1 activation leads to actin assembly at the dorsal surface of the cell membrane that result in sheet-like protrusion formation without any requirement of a template. Further, this approach can be used to study the complex morpho-dynamics of the protrusions or to investigate specific proteins that may be enriched in the ruffles. Deactivating PA-Rac1 leads to complex contractile processes resulting in formation of macropinosomes. Using multicolour imaging in combination with these approaches, we find that Myo1e specifically is enriched in the ruffles.ConclusionsCombining LLSM and optogenetics enables superior spatial and temporal control for studying such dynamic mechanisms. Demonstrated here, the techniques implemented provide insight into the complex nature of the molecular interplay involved in dynamic actin machinery, revealing that Rac1 activation can generate untemplated, lamellar protrusions.

Nailfold capillaroscopy (NFC) has developed into an indispensable tool for rheumatologists in the evaluation of rheumatic diseases. It offers various advantages in being rapid, noninvasive, and inexpensive. With NFC we are able to visualize the microcirculatory changes in the nail beds. These changes are key to the pathogenesis of connective tissue diseases such as systemic sclerosis. Hence NFC helps in early diagnosis of various connective tissue diseases. There is a lack of standardization in the techniques used and various capillary parameters studied, which could lead to variation in the reporting of the parameters studied. In this chapter we shall try to highlight the most common parameters studied in capillaroscopy and its utility in various connective tissue diseases.

Objective Several studies have reported hydroxychloroquine (HCQ) retinal toxicity in East Asian patients. However, at present, there are limited data from Indian patients. The aim of this study was to investigate the prevalence and pattern of HCQ retinal toxicity in Indian population and to see if screening protocol for retinal toxicity in Indian patients should be any different to others. Methods This was a cross-sectional study involving adult patients with autoimmune rheumatic diseases who had received HCQ for at least 5 years or a cumulative dose of at least 500 g. Retinal toxicity was evaluated using fundus examination, visual fields 10-2 and 30-2 protocol, and spectral domain optical coherence tomography. Results Of 110 patients screened, retinal toxicity was found in 7 patients (6.36%). A parafoveal pattern was found in 4 patients, whereas a mixed parafoveal and perifoveal pattern was found in 3 patients. None of the patients had isolated perifoveal pattern. Except for the one patient, all the patients with retinal toxicity had more than 10 years (mean, 13 ± 4.89 years; range, 5-20 years) of HCQ usage with a mean cumulative dose of 1573.7 ± 771.5 g. The mean daily dose was 5 ± 1.6 mg/kg per day. Conclusions Hydroxychloroquine retinal toxicity is more common than previously recognized in patients who have used the drug for more than 5 years. The toxicity manifests as a parafoveal or a mixed parafoveal and perifoveal pattern in Indian patients.

Renewable energy conversion is already gained momentum and the installations have been increasing for the past few decades. The continuing trend in research and developments of renewable energy technologies, cost effectiveness and market dynamics reveals the awareness of the climate change and associated impacts on the society. Purpose Polymer-particle composites, which have demonstrated wide applications ranging from energy harvesting and storage, biomedical applications, electronics and environmental sensing to aerospace applications, have been investigated for decades. However, fabricating polymer-particle composites with controlled distribution of particles in polymer continues to be a fundamental challenge. As to date, a few additive manufacturing (AM) technologies can fabricate composites, however, with a limited choice of materials or limited dispersion control. Against this background, this research investigated a hybrid polymer-particle composite manufacturing process, projection electro-stereolithography (PES) process, which integrates electrostatic deposition and projection based stereolithography (SL) technologies. Design/methodology/approach In PES process, a photoconductive film collects charged particles in the regions illuminated by light. Then, collected particles are transferred from the film to a polymer layer with defined patterns. Lastly, a digital mask is used to pattern the light irradiation of the digital micromirror device chip, selectively curing the photopolymer liquid resin and particles of that layer. By transferring particles from the photoconductive film to the photopolymer in a projection-based SL system, multi-material composites with locally controlled dispersions could be produced. A proof-of-concept PES testbed was developed. Various test cases have been performed to verify the feasibility and effectiveness of the developed approach. Findings Challenges in this novel AM process, including process design, particle patterning and transferring, are addressed in this paper. It is found that particles can be transferred to a layer of partially cured resin completely and accurately, by using the stamping approach. The transferring rate is related to stamping force and degree of conversion of the recipient layer. The developed hybrid process can fabricate polymer-particle composites with arbitrary dispersion pattern, unlimited printable height and complicated geometries. Originality/value Although an electrostatic deposition process has been investigated as a 3D printing technology for many years, it is the first attempt to integrate it with projection SL for fabricating multi-material polymer composite components. The novel hybrid process offers unique benefits including local dispersion control, arbitrary filling patterns, wide range of materials, unlimited printable height and arbitrary complicated geometries.
