Antibiotics are widely used to treat infectious diseases. This leads to the presence of antibiotics and their metabolic products in the ecosystem, especially in aquatic environments. In many countries, the growth of pathogen resistance to antibiotics is considered a threat to national security. Therefore, methods for determining the sensitivity/resistance of bacteria to antimicrobial drugs are important. This review discusses the mechanisms of the formation of antibacterial resistance and the various methods and sensor systems available for analyzing antibiotic effects on bacteria. Particular attention is paid to acoustic biosensors with active immobilized layers and to sensors that analyze antibiotics directly in liquids. It is shown that sensors of the second type allow analysis to be done within a short period, which is important for timely treatment. [ABSTRACT FROM AUTHOR]

The formation of gas vesicles has been investigated in bacteria and haloarchaea for more than 50 years. These air-filled nanostructures allow cells to stay at a certain height optimal for growth in their watery environment. Several gvp genes are involved and have been studied in Halobacterium salinarum, cyanobacteria, Bacillus megaterium, and Serratia sp. ATCC39006 in more detail. GvpA and GvpC form the gas vesicle shell, and additional Gvp are required as minor structural proteins, chaperones, an ATP-hydrolyzing enzyme, or as gene regulators. We analyzed the Gvp proteins of Hbt. salinarum with respect to their protein–protein interactions, and developed a model for the formation of these nanostructures. Gas vesicles are also used in biomedical research. Since they scatter waves and produce ultrasound contrast, they could serve as novel contrast agent for ultrasound or magnetic resonance imaging. Additionally, gas vesicles were engineered as acoustic biosensors to determine enzyme activities in cells. These applications are based on modifications of the surface protein GvpC that alter the mechanical properties of the gas vesicles. In addition, gas vesicles have been decorated with GvpC proteins fused to peptides of bacterial or viral pathogens and are used as tools for vaccine development. [ABSTRACT FROM AUTHOR]

The main idea of the book is that, depending on the addressed problem, different approaches are to be used; macro constructs are to be worked with in some cases, micro and nano in others. Biosensors considered are electrochemical, optical, atomic force microscopy-based; biofuel cells that develop the idea of electrochemical biosensors are intended for a double purpose of cleaning up the environment and working out electrical energy.

Ultrasound imaging is one of the most widely used modalities in clinical practice, revealing human prenatal development but also arterial function in the adult brain. Ultrasound waves travel deep within soft biological tissues and provide information about the motion and mechanical properties of internal organs. A drawback of ultrasound imaging is its limited ability to detect molecular targets due to a lack of cell-type specific acoustic contrast. To date, this limitation has been addressed by targeting synthetic ultrasound contrast agents to molecular targets. This molecular ultrasound imaging approach has proved to be successful but is restricted to the vascular space. Here, we introduce the nascent field of biomolecular ultrasound imaging, a molecular imaging approach that relies on genetically encoded acoustic biomolecules to interface ultrasound waves with cellular processes. Abstract The present study showed that Deep Oscillation Magnetron Sputtering (DOMS) has an excellent potential to produce AlN films for biosensing applications. However, careful control of the process parameters is required to attain the desired features. Al/AlN/Al films deposited via DOMS on Si wafers were extensively characterized to assess this potential. The resulting microstructures were examined by glancing angle x-ray diffraction, scanning electron microscopy coupled with energy dispersive microanalysis, atomic force microscopy, and x-ray photoelectron spectroscopy. The results indicated that there is a strong effect of the bias voltage (V b) applied to the substrate on the structure of the Al/AlN/Al films and that it can be divided in two regimes. The low V b regime produced the best features of the AlN films for the present application. These included a strong (002) orientation, low residual stress, and uniform surface roughness. In the high V b regime, these features degraded. The effects were associated to the nature and energy of the impinging species as a function of V b. Highlights • h-AlN (002) films can be readily obtained by deep oscillation magnetron sputtering. • The bias voltage applied to the substrate has a strong effect on film deposition. • There is a small window of bias voltage to attain the best AlN film features. Development of biosensors requires a well-organized template to enhance the sensing specificity. Biological molecules that have the ability to self-assemble into ordered structures can be exploited to construct nanostructures and nanodevices. In this regard, bacterial surface layer proteins (SbpA) have been used as an underlying matrix to bind molecules on the nanometer scale. The present study provides evidence that a gold substrate functionalized with folate-modified SbpA can be used as a sensor for breast cancer cells with superior efficacy over traditional sensors. The developed sensor uses folate to recognize folate receptors, which are highly expressed on the cell membranes of some cancer cells, such as breast cancer cells. To examine the specificity of the developed sensor, folate was used as a binding partner for folate receptors on breast cancer cells (MCF-7), and liver cancer cells (HepG2) were used as the control because they lack folate receptors. The fabricated biosensor comprised a gold surface and a recrystallized S-layer protein lattice functionalized with folate and addressed acoustically and electrochemically. Acoustic studies performed using quartz crystal microbalance with dissipation monitoring (QCM-D) showed the efficiency of the developed biosensor in distinguishing between MCF-7 and HepG2 cells. Despite the ability of QCM-D to recognize cells in situ and in real-time, this biosensor had a limited detection range (1 × 10 5  cells/mL). To confirm the functionality of the developed platform, electrochemical measurements were performed and the obtained results were in agreement with the acoustic sensor and proving the efficiency of the designed sensing layer. The S-layer lattice-folate modified biosensor provides a promising strategy for designing efficient sensing platforms that can be used to diagnose early stage cancers with highly expressed folate receptors. Acoustic wave resonator techniques are widely used in in-liquid biochemical applications. The main challenges remaining are the improvement of sensitivity and limit of detection, as well as multianalysis capabilities and reliability. The sensitivity improvement issue has been addressed by increasing the sensor frequency, using different techniques such as high fundamental frequency quartz crystal microbalances (QCMs), surface generated acoustic waves (SGAWs) and film bulk acoustic resonators (FBARs). However, this sensitivity improvement has not been completely matched in terms of limit of detection. The decrease on frequency stability due to the increase of the phase noise, particularly in oscillators, has made it impossible to increase the resolution. A new concept of sensor characterization at constant frequency has been recently proposed based on the phase/mass sensitivity equation: ∆φ/∆m ≈ −1/mL, where mL is the liquid mass perturbed by the resonator. The validation of the new concept is presented in this article. An immunosensor application for the detection of a low molecular weight pollutant, the insecticide carbaryl, has been chosen as a validation model.

Abstract: This work describes the application of a newly emerged biosensing configuration incorporating a Surface Acoustic Wave device integrated with a multi-channel microfluidic module for the rapid and efficient analysis of cardiac markers. The examined cardiac markers of creatine kinase MB (CK-MB), cardiac reactive protein (CRP), D-dimer and pregnancy-associated plasma protein A (PAPP-A), comprise a group of both established and emerging heart disease proteins, that has never been probed before with any kind of biochip-related platform. The four markers were successfully detected; kinetics and affinity studies on their interactions with the surface immobilized antibodies are also presented. A concentration detection limit of less than 1nM was achieved, with a dynamic range of more than two orders of magnitude, covering some of the pathological and healthy areas of interest. Mixtures of biomarkers applied to the device surface were used to prove the specificity of each binding event and investigate the microsystem''s performance in the presence of complex fluids, towards future utilization with real samples. The simplicity and multiplexing ability of the integrated platform render the system ideal as a potential diagnostic tool for cardiovascular risk assessment, where simultaneous analysis of various protein markers is required. Abstract: The current work describes a novel setup for multi-sample biomolecular analysis. It is based on the assembly of a dual acoustic device chip with a four-channel microfluidic module, forming an array of eight available domains for experiments. Initially, multiple detection was demonstrated via the specific interaction of neutravidin with four different biotinylated proteins, namely protein G, protein A, bovine serum albumin, and immunoglobulin G; results revealed a reproducibility between the microchannel domains better than 90%. Real-time analysis of the binding interactions was used to calculate the affinity and kinetic constants of the four biotinylated molecules binding to surface-immobilized neutravidin; this was the first time that this information was derived using a biosensing device and four biotinylated molecules. Interestingly, all calculated kinetic and affinity constants resemble those typical of antibody-antigen interactions, although the investigated specific binding was of avidin–biotin nature. Finally, under device pre-functionalization conditions, it was possible to probe eight interactions all together, exploiting the full capacity of the microsystem and reducing significantly the analysis time, contrary to the use of the standard acoustic device configuration. The outcome of this full-scale validation opens the way for the integrated acoustic platform to be implemented in even higher throughput detection for future diagnostic/biomedical applications, as well as in fundamental research studies regarding biomolecular interaction investigation and characterization. Resonant and acoustic wave devices have been researched for several decades for application in the gravimetric sensing of a variety of biological and chemical analytes. These devices operate by coupling the measurand (e.g. analyte adsorption) as a modulation in the physical properties of the acoustic wave (e.g. resonant frequency, acoustic velocity, dissipation) that can then be correlated with the amount of adsorbed analyte. These devices can also be miniaturized with advantages in terms of cost, size and scalability, as well as potential additional features including integration with microfluidics and electronics, scaled sensitivities associated with smaller dimensions and higher operational frequencies, the ability to multiplex detection across arrays of hundreds of devices embedded in a single chip, increased throughput and the ability to interrogate a wider range of modes including within the same device. Additionally, device fabrication is often compatible with semiconductor volume batch manufacturing techniques enabling cost scalability and a high degree of precision and reproducibility in the manufacturing process. Integration with microfluidics handling also enables suitable sample pre-processing/separation/purification/amplification steps that could improve selectivity and the overall signal-to-noise ratio.
