Abstract Background Autism spectrum disorder (ASD), a neurodevelopmental disorder defined by social communication deficits plus repetitive behaviors and restricted interests, currently affects 1/36 children in the general population. Recent advances in functional brain imaging show promise to provide useful biomarkers of ASD diagnostic likelihood, behavioral trait severity, and even response to therapeutic intervention. However, current gold-standard neuroimaging methods (e.g., functional magnetic resonance imaging, fMRI) are limited in naturalistic studies of brain function underlying ASD-associated behaviors due to the constrained imaging environment. Compared to fMRI, high-density diffuse optical tomography (HD-DOT), a non-invasive and minimally constraining optical neuroimaging modality, can overcome these limitations. Herein, we aimed to establish HD-DOT to evaluate brain function in autistic and non-autistic school-age children as they performed a biological motion perception task previously shown to yield results related to both ASD diagnosis and behavioral traits. Methods We used HD-DOT to image brain function in 46 ASD school-age participants and 49 non-autistic individuals (NAI) as they viewed dynamic point-light displays of coherent biological and scrambled motion. We assessed group-level cortical brain function with statistical parametric mapping. Additionally, we tested for brain-behavior associations with dimensional metrics of autism traits, as measured with the Social Responsiveness Scale-2, with hierarchical regression models. Results We found that NAI participants presented stronger brain activity contrast (coherent > scrambled) than ASD children in cortical regions related to visual, motor, and social processing. Additionally, regression models revealed multiple cortical regions in autistic participants where brain function is significantly associated with dimensional measures of ASD traits. Limitations Optical imaging methods are limited in depth sensitivity and so cannot measure brain activity within deep subcortical regions. However, the field of view of this HD-DOT system includes multiple brain regions previously implicated in both task-based and task-free studies on autism. Conclusions This study demonstrates that HD-DOT is sensitive to brain function that both differentiates between NAI and ASD groups and correlates with dimensional measures of ASD traits. These findings establish HD-DOT as an effective tool for investigating brain function in autistic and non-autistic children. Moreover, this study established neural correlates related to biological motion perception and its association with dimensional measures of ASD traits.

Abstract In our changing world, understanding plant community responses to global change drivers is critical for predicting future ecosystem composition and function. Plant functional traits promise to be a key predictive tool for many ecosystems, including grasslands; however, their use requires both complete plant community and functional trait data. Yet, representation of these data in global databases is sparse, particularly beyond a handful of most used traits and common species. Here we present the CoRRE Trait Data, spanning 17 traits (9 categorical, 8 continuous) anticipated to predict species’ responses to global change for 4,079 vascular plant species across 173 plant families present in 390 grassland experiments from around the world. The dataset contains complete categorical trait records for all 4,079 plant species obtained from a comprehensive literature search, as well as nearly complete coverage (99.97%) of imputed continuous trait values for a subset of 2,927 plant species. These data will shed light on mechanisms underlying population, community, and ecosystem responses to global change in grasslands worldwide.

Abstract Men tend to eat more meat than women, but it is not clear why. We tested three hypotheses in a cross-cultural design (20,802 individuals in 23 countries across four continents): that gender differences are (a) universal, (b) related to gender roles and thus weaker in countries with higher gender equality and human development, or (c) related to opportunities to express gender roles and thus stronger in countries with higher gender equality and human development. Across all countries, men tended to consume more meat than women. However, this difference increased significantly in countries with greater human development and gender equality. The paradoxical gender gap in meat consumption aligns with previous research that suggests greater differences in behavior across genders in contexts that are more developed and gender equal. We discuss implications for theories of culture and gender as well as practical implications for global meat reduction.

Introduction: Healthcare organizations are under increasing pressure from policymakers, payers, and advocates to screen for and address patients’ health-related social needs (HRSN). The emergency department (ED) presents several challenges to HRSN screening, and patients are frequently not screened for HRSNs. Predictive modeling using machine learning and artificial intelligence, approaches may address some pragmatic HRSN screening challenges in the ED. Because predictive modeling represents a substantial change from current approaches, in this study we explored the acceptability of HRSN predictive modeling in the ED. Methods: Emergency clinicians, ED staff, and patient perspectives on the acceptability and usage of predictive modeling for HRSNs in the ED were obtained through in-depth semi-structured interviews (eight per group, total 24). All participants practiced at or had received care from an urban, Midwest, safety-net hospital system. We analyzed interview transcripts using a modified thematic analysis approach with consensus coding. Results: Emergency clinicians, ED staff, and patients agreed that HRSN predictive modeling must lead to actionable responses and positive patient outcomes. Opinions about using predictive modeling results to initiate automatic referrals to HRSN services were mixed. Emergency clinicians and staff wanted transparency on data inputs and usage, demanded high performance, and expressed concern for unforeseen consequences. While accepting, patients were concerned that prediction models can miss individuals who required services and might perpetuate biases. Conclusion: Emergency clinicians, ED staff, and patients expressed mostly positive views about using predictive modeling for HRSNs. Yet, clinicians, staff, and patients listed several contingent factors impacting the acceptance and implementation of HRSN prediction models in the ED.

Abstract Background Hypothermia is a cause of neonatal calf death in cold climates. Practical and effective rewarming methods are important for bovine health within affected regions. Hypothesis/Objectives To compare the rewarming rate and blood analytes (glucose, lactate, and cortisol) of calves resuscitated with forced air with warm water bath, with or without oral administration of caffeine. Animals Twenty healthy neonatal Holstein bull calves. Methods In this randomized, prospective study, calves born healthy and without history of dystocia were cooled to 32°C rectal temperature then thermally resuscitated using either forced air rewarming or warm water bath (40°C) with or without oral administration of caffeine. Rectal temperatures were used to quantify recovery rate. Measurements of glucose, lactate, and cortisol were recorded for every 2°C change in rectal temperature. Results Rectal temperature decline (0.03°C per minute) and total cooling time (191.0 ± 33.3 minutes) did not significantly differ among treatment groups. Remote sensing (RS) technologies are extensively exploited by scientists and a vast audience of local authorities, urban managers, and city planners. Coastal regions, geohazard-prone areas, and highly populated cities represent natural laboratories to apply RS technologies and test new methods. Over the last decades, many efforts have been spent on improving Earth’s surface monitoring, including intensifying Earth Observation (EO) operations by the major national space agencies. They oversee to plan and make operational constellations of satellite sensors providing the scientific community with extensive research and development opportunities in the geoscience field. For instance, within this framework, the European Space Agency (ESA) and the Ministry of Science and Technology of China (MOST) have sponsored, since the early 2000s, the DRAGON initiative jointly carried out by the European and Chinese RS scientific communities. This manuscript aims to provide a synthetic overview of some research activities and new methods recently designed and applied and trace the route for further developments. The main findings are related to i) the analysis of flood risk in China, ii) the potential of new methods for the estimation and removal of ground displacement biases in small-baseline oriented interferometric Synthetic Aperture Radar (SAR) methods, iii) the analysis of the inundation risk in low-lying regions using coherent and incoherent SAR methods; and iv) the use of SAR-based technologies for marine applications.

IntroductionShooting simulations provide an excellent opportunity to train use-of-force decisions in controlled environments. Recently, military and law enforcement organizations have expressed a growing desire to integrate physiological measurement into simulations for training and feedback purposes. Although participants can easily wear physiological monitors in these scenarios, direct implementation into training may not be simple. Theoretical problems exist in the ultra-short heart rate variability windows associated with use-of-force training, and practical problems emerge as existing scenario libraries at training organizations were not designed for physiological monitoring.MethodsThe current study explored the challenges and possibilities associated with direct implementation of physiological monitoring into an existing library of firearms training scenarios. Participants completed scenarios in a shooting simulator using existing military training scenarios while wearing a device to monitor their heart rate.ResultsThe results revealed lower heart rate variability (approximately 6%) occurred in scenarios where participants did not have to fire weapons, indicating that don't-shoot scenarios may actually impose more cognitive stress on shooters. Additional evidence further demonstrated how both behavioral and physiological factors could be used concomitantly to predict unintentionally firing on non-hostile actors. However, behavioral measures were more predictive (e.g., β = .221) than physiological measures (e.g., β = −.132) when the latter metrics were limited to specific scenarios. Qualitative results suggest that simply applying physiological monitoring to existing shooting simulations may not yield optimal results because it would be difficult to directly integrate physiological measurement in a meaningful way without re-designing some elements of the simulations, the training procedure, or both.DiscussionFuture use-of-force shooting simulations should consider designing novel scenarios around the physiological measurement rather than directly implementing physiological assessments into existing libraries of scenarios.

Abstract Background Bacterial cancer therapy was first trialled in patients at the end of the nineteenth century. More recently, tumour-targeting bacteria have been harnessed to deliver plasmid-expressed therapeutic interfering RNA to a range of solid tumours. A major limitation to clinical translation of this is the short-term nature of RNA interference in vivo due to plasmid instability. To overcome this, we sought to develop tumour-targeting attenuated bacteria that stably express shRNA by virtue of integration of an expression cassette within the bacterial chromosome and demonstrate therapeutic efficacy in vitro and in vivo. Results The attenuated tumour targeting Salmonella typhimurium SL7207 strain was modified to carry chromosomally integrated shRNA expression cassettes at the xylA locus. The colorectal cancer cell lines SW480, HCT116 and breast cancer cell line MCF7 were used to demonstrate the ability of these modified strains to perform intracellular infection and deliver effective RNA and protein knockdown of the target gene c-Myc. In vivo therapeutic efficacy was demonstrated using the Lgr5creER T2 Apc flx/flx and BlgCreBrca2 flx/fl p53 flx/flx orthotopic immunocompetent mouse models of colorectal and breast cancer, respectively. Abstract Background The purpose of this meta-analysis was to provide a pooled prevalence estimate of self-reported disordered eating (SRDE) in athletes based on the available literature, and to identify risk factors for their occurrence. Methods Across ten academic databases, an electronic search was conducted from inception to 7th January 2024. The proportion of athletes scoring at or above predetermined cutoffs on validated self-reporting screening measures was used to identify disordered eating (DE). Subgroup analysis per country, per culture, and per research measure were also conducted. Age, body mass index (BMI), and sex were considered as associated/correlated factors. Results The mean prevalence of SRDE among 70,957 athletes in 177 studies (132 publications) was 19.23% (17.04%; 21.62%), I 2 = 97.4%, τ2 = 0.8990, Cochran's Q p value = 0. Australia had the highest percentage of SRDE athletes with a mean of 57.1% (36.0%-75.8%), while Iceland had the lowest, with a mean of 4.9% (1.2%-17.7%). The SRDE prevalence in Eastern countries was higher than in Western countries with 29.1% versus 18.5%. Anaerobic sports had almost double the prevalence of SRDE 37.9% (27.0%-50.2%) compared to aerobic sports 19.6% (15.2%-25%). Gymnastics sports had the highest SRDE prevalence rate, with 41.5% (30.4%-53.6%) while outdoor sports showed the lowest at 15.4% (11.6%-20.2%). Abstract Metastatic breast cancer leads to poor prognoses and worse outcomes in patients due to its invasive behavior and poor response to therapy. It is still unclear what biophysical and biochemical factors drive this more aggressive phenotype in metastatic cancer; however recent studies have suggested that exposure to fluid shear stress in the vasculature could cause this. In this study a modular microfluidic platform capable of mimicking the magnitude of fluid shear stress (FSS) found in human vasculature was designed and fabricated. This device provides a platform to evaluate the effects of FSS on MCF-7 cell line, an estrogen receptor positive (ER+) breast cancer cell line, during circulation in the vessels. Elucidation of the effects of FSS on MCF-7 cells was carried out utilizing two approaches: single cell analysis and bulk analysis. For single cell analysis, cells were trapped in a microarray after exiting the serpentine channel and followed by immunostaining on the device (on-chip). Bulk analysis was performed after cells were collected in a microtube at the outlet of the microfluidic serpentine channel for western blotting (off-chip). It was found that cells exposed to an FSS magnitude of 10 dyn/cm2 with a residence time of 60 s enhanced expression of the proliferation marker Ki67 in the MCF-7 cell line at a single cell level. To understand possible mechanisms for enhanced Ki67 expression, on-chip and off-chip analyses were performed for pro-growth and survival pathways ERK, AKT, and JAK/STAT. Results demonstrated that after shearing the cells phosphorylation of p-AKT, p-mTOR, and p-STAT3 were observed. However, there was no change in p-ERK1/2. AKT is a mediator of ER rapid signaling, analysis of phosphorylated ERα was carried out and no significant differences between sheared and non-sheared populations were observed. Taken together these results demonstrate that FSS can increase phosphorylation of proteins associated with a more aggressive phenotype in circulating cancer cells. These findings provide additional information that may help inform why cancer cells located at metastatic sites are usually more aggressive than primary breast cancer cells.

Transcranial electrical stimulation has demonstrated the potential to enhance cognitive functions such as working memory, learning capacity, and attentional allocation. Recently, it was shown that periodic stimulation within a specific duration could augment the human brain’s neuroplasticity. This study investigates the effects of repetitive transcranial alternating current stimulation (tACS; 1 mA, 5 Hz, 2 min duration) on cognitive function, functional connectivity, and topographic changes using both electroencephalography (EEG) and functional near-infrared spectroscopy (fNIRS). Fifteen healthy subjects were recruited to measure brain activity in the pre-, during-, and post-stimulation sessions under tACS and sham stimulation conditions. Fourteen trials of working memory tasks and eight repetitions of tACS/sham stimulation with a 1-minute intersession interval were applied to the frontal cortex of the participants. The working memory score, EEG band-wise powers, EEG topography, concentration changes of oxygenated hemoglobin, and functional connectivity (FC) were individually analyzed to quantify the behavioral and neurophysiological effects of tACS. Our results indicate that tACS increases: i) behavioral scores (i.e., 15.08, ${p} < 0.001$ ) and EEG band-wise powers (i.e., theta and beta bands) compared to the sham stimulation condition, ii) FC of both EEG-fNIRS signals, especially in the large-scale brain network communication and interhemispheric connections, and iii) the hemodynamic response in comparison to the pre-stimulation session and the sham condition. Conclusively, the repetitive theta-band tACS stimulation improves the working memory capacity regarding behavioral and neuroplasticity perspectives. Additionally, the proposed fNIRS biomarkers (mean, slope), EEG band-wise powers, and FC can be used as neuro-feedback indices for closed-loop brain stimulation.

ABSTRACTBackground Tuberculosis (TB) has a significant treatment burden for patients, requiring at least six months of anti-TB treatment (ATT) with multiple medicines. Ensuring good adherence to ATT is central to global TB strategies, including those in high-income, low-TB incidence (HILI) settings. For adherence interventions to be successful and deliverable, they need to address the personal and environmental factors influencing patient and provider behaviour.Purpose This paper describes the application of theory and research evidence to inform the design process of the IMPACT manualised intervention to support ATT adherence for adults with TB disease in the United Kingdom (UK). It also provides a full description of the resulting intervention.Methods We synthesised findings from our formative research (qualitative and quantitative scoping reviews and patient and carer interviews) and supplemented these with clinic observations, a literature review, and healthcare provider interviews. Findings were mapped to the guiding theoretical framework (Perceptions and Practicalities Approach) which was operationalised to design the intervention components and content. An Intervention Development Group (IDG) of relevant stakeholders were consulted to adapt the intervention to local clinical settings.Results The pragmatic, deliverable components and content for the IMPACT intervention included: (1) an enhanced, structured, risk assessment to systematically identify risk factors for non-adherence plus locally-adapted guidance to mitigate these; and (2) patient educational materials (an animated video and interactive patient booklet) about TB and its treatment, to communicate the need for treatment and address common concerns.Conclusions Using a theory– and evidence– based approach incorporating stakeholder input, we have developed a multi-component, pragmatic, manualised intervention, which addresses patients’ personal barriers to adherence within local service resources to improve adherence to ATT within UK TB services.

Deafness-causing deficiencies in otoferlin (OTOF) have been addressed preclinically using dual adeno-associated virus (AAV)-based approaches. However, timing of transduction, recombination of mRNA, and protein expression with dual hybrid AAV methods methods have not previously been characterized. Here, we have established an ex vivo assay to determine the kinetics of dual-AAV mediated expression of OTOF in hair cells of the mouse utricle. We utilized two different recombinant vectors that comprise DB-OTO, one containing the 5′ portion of OTOF under the control of the hair cell-specific Myo15 promoter, and the other the 3′ portion of OTOF. We explored specificity of the Myo15 promoter in hair cells of the mouse utricle, established dose response characteristics of DB-OTO ex vivo in an OTOF-deficient mouse model, and demonstrated tolerability of AAV1 in utricular hair cells. Furthermore, we established deviations from a one-to-one ratio of 5′ to 3′ vectors with little impact on recombined OTOF. Finally, we established a plateau in quantity of recombined OTOF mRNA and protein expression by 14 to 21 days ex vivo with comparable recovery timing to that in vivo model. These findings demonstrate the utility of an ex vivo model system for exploring expression kinetics and establish in vivo and ex vivo recovery timing of dual AAV-mediated OTOF expression.

Anaphylaxis is an acute, potentially fatal, systemic hypersensitivity reaction that warrants prompt diagnosis and management. It continues to be challenging to anticipate who may be at risk of a severe, life-threatening allergic reaction. Anaphylaxis can be caused by a range of allergens, such as certain foods, medications, latex, insect stings, etc. Cofactors that augment the severity of clinical symptoms and increase the risk of poor outcomes include exercise, stress, infectious diseases, underlying mast cell disease, active allergic disease such as asthma, advanced age, intake of certain medications, history of previous anaphylaxis, and delayed or missed administration of adrenaline. According to the European Anaphylaxis Registry, food is the major elicitor of anaphylaxis, especially eggs, cow milk, and nuts, in children and adolescents. Reaction to insect venom has also been noted in young adulthood. Early recognition of signs and symptoms and prompt treatment are crucial in anaphylaxis management to avoid serious and even fatal outcomes. It is crucial for both individuals and clinicians to identify the cause of anaphylaxis. Biomarkers of anaphylaxis, such as histamine, tryptase, platelet activation factor (PAF), chymase, carboxypeptidase A3, dipeptidyl peptidase I (DPPI), basogranulin, CCL-2, hsa-miR-451a, may be useful in diagnosis and management. The purpose of this review article is to present a comprehensive overview of current evidence and expert opinions regarding the risk factors that predispose individuals to anaphylaxis. Additionally, it provides insights into potential biomarkers and genetic markers for accurate diagnosis and management. This review underscores the significance of expert guidance in enhancing patient outcomes and enabling self-management of anaphylactic episodes.

We report the use of suboxide molecular-beam epitaxy (S-MBE) to grow α-(AlxGa1−x)2O3 films on (110) sapphire substrates over the 0 < x < 0.95 range of aluminum content. In S-MBE, 99.98% of the gallium-containing molecular beam arrives at the substrate in a preoxidized form as gallium suboxide (Ga2O). This bypasses the rate-limiting step of conventional MBE for the growth of gallium oxide (Ga2O3) from a gallium molecular beam and allows us to grow fully epitaxial α-(AlxGa1−x)2O3 films at growth rates exceeding 1 µm/h and relatively low substrate temperature (Tsub = 605 ± 15 °C). The ability to grow α-(AlxGa1−x)2O3 over the nominally full composition range is confirmed by Vegard’s law applied to the x-ray diffraction data and by optical bandgap measurements with ultraviolet–visible spectroscopy. We show that S-MBE allows straightforward composition control and bandgap selection for α-(AlxGa1−x)2O3 films as the aluminum incorporation x in the film is linear with the relative flux ratio of aluminum to Ga2O. The films are characterized by atomic-force microscopy, x-ray diffraction, and scanning transmission electron microscopy (STEM). Abstract 1. Conserving species' ability to traverse the landscape is vital for maintaining biodiversity in the face of global change. Connectivity conservation requires identifying important pathways for species' movements and aligning societal support for conservation of those pathways. Contemporary connectivity analyses emphasize the impacts of topography, vegetation and human footprint on species' movements; but largely ignore the role that attitudes, economics and institutions play in practitioners' ability to conserve those movements. 2. We introduce implementation resistance as an analogue of biophysical resistance that combines social, economic and institutional factors that promote or impede connectivity conservation. We demonstrate the utility of integrating implementation resistance as a means of choosing between competing connectivity conservation strategies using wolves in Colorado (USA) as a case study. 3. Our analysis of five potential corridor locations based on biophysical costs revealed substantial differences in the social costs associated with implementing each corridor despite relatively minimal differences in the biophysical costs. 4. Our comparison of hypothetical interventions to reduce implementation resistance illustrates that interventions that reduce conflicts between land use and wolves may substantially reduce overall resistance, those reductions are not as well aligned with connectivity priorities as those resulting from changes in land management agency policy. 5. Our results highlight the need to design conservation interventions that fit both the social and ecological landscape and provide a framework for developing robust, interdisciplinary methods that facilitate implementable connectivity conservation. Abstract Ocean waves excite continuous globally observable seismic signals. We use data from 52 globally distributed seismographs to analyze the vertical component primary microseism wavefield at 14–20 s period between the late 1980s and August 2022. This signal is principally composed of Rayleigh waves generated by ocean wave seafloor tractions at less than several hundred meters depth, and is thus a proxy for near-coastal swell activity. Here we show that increasing seismic amplitudes at 3σ significance occur at 41 (79%) and negative trends occur at 3σ significance at eight (15%) sites. The greatest absolute increase occurs for the Antarctic Peninsula with respective acceleration amplitude and energy trends ( ± 3σ) of 0.037 ± 0.008 nm s−2y−1 (0.36 ± 0.08% y−1) and 4.16 ± 1.07 nm2 s−2y−1 (0.58 ± 0.15% y−1), where percentage trends are relative to historical medians. The inferred global mean near-coastal ocean wave energy increase rate is 0.27 ± 0.03% y−1 for all data and is 0.35 ± 0.04% y−1 since 1 January 2000. Strongly correlated seismic amplitude station histories occur to beyond 50∘ of separation and show regional-to-global associations with El Niño and La Niña events.

Self-lubricating aluminium matrix composites with enhanced tribological properties are sought for weight critical applications. In previous studies, the Al composites incorporating the solid lubricant WS2 have been shown to reduce both the coefficient of friction and wear rate, positioning them as promising candidates in various tribological applications (e.g. automotive industry). However, the impact of interfacial reactions between Al and WS2 during composite production on tribological performance has still not yet been explored. This study highlights the hardening effect of the reaction products. Despite some literature assuming a negative impact of these reactions as they consume WS2 in the composites, this study presents evidence that this cannot be generalised for the overall outcome. Interestingly, a controlled amount is shown to be beneficial for tribological properties. In this work, the tribological influence of the Al–W intermetallic structure forming during spark plasma sintering of the Al–WS2 composites was investigated. The microstructure was tailored by adjusting the manufacturing temperature between 500 and 600 °C. The Al–WS2 fabricated at 580 °C exhibited the lowest coefficient of friction and specific wear rate (reduced by 20 % and 97 %, respectively, compared to the one fabricated at 500 °C. Furthermore, the worn surface morphology in different stages during friction was evaluated by a novel wear track evolution analysis. This study confirmed that offering a balance between the fraction of solid lubricants and in-situ formed hard intermetallic structure is crucial to the effective formation a protective layer on the worn surface.

Farmed animal policy and advocacy efforts both attempt to generate and depend upon public support. However, relatively little is known about the factors that predict support for animal protection legislation and advocacy across the globe. We analyse data from a large international survey (23 countries, n = 20,966) alongside other data sources on animal advocacy to investigate knowledge of factory farming, the connection between attitudes towards animals and the strength of animal protection legislation, attitudes towards animals based on their food status in different countries, and the connection between personal support for policy, animal advocacy, civic activism, and animal advocacy organisations. We found that higher support for animal welfare is associated with stronger farm animal protection legislation across countries and that concerns about specific animals can vary depending on cultural and religious factors. Contrary to study hypotheses, we did not find greater support for advocacy in countries with more advocacy organisations, suggesting important opportunities to pursue advocacy in relatively neglected regions. Results are interpreted in terms of how farmed animal advocates can take advantage of and potentially generate support for animal welfare throughout the world.

IntroductionMost children cease napping between 2 and 5 years old. Little is known about the predictors of this cessation. Parents' sleep-related beliefs aid in understanding children's nighttime sleep behaviors, but few index daytime sleep beliefs.MethodsTwo measures of parents' napping beliefs were developed and evaluated-the Parents' Nap Beliefs Scale (14 items) and the Reasons Children Nap Scale (19 items). Canadian parents of 1–5-year-old children completed these questionnaires and other sleep-related measures in independent pilot (n = 201) and replication (n = 702) samples. In the replication sample, a subsample of parents also completed 1–3 weeks of daily sleep diaries. The samples were representative of the Canadian population by ethnicity and region.ResultsIn both samples, both measures demonstrated strong construct validity, convergent and divergent validity, and internal consistency. The Parents' Nap Beliefs Scale was composed of two factors: (a) Positive Beliefs and (b) Negative Beliefs about napping. The Reasons Children Nap Scale was composed of two higher order factors and five lower order factors: (a) Encouragement Reasons (Child related; Parent related) and (b) Discouragement Reasons (Child prefers not to nap; Child functions well without a nap; Scheduling).DiscussionFuture research should (a) test these scales as longitudinal determinants of children's nap behavior and cessation, (b) evaluate parental Nap Beliefs in non-Western cultures, and (c) adapt these scales for use with childcare providers.

Abstract The reintroduction of wildlife can have significant ecological impacts by altering the flow of energy in food webs. Recently, plains bison were reintroduced to part of Banff National Park after a 150‐year absence. The large herbivore's reintroduction was expected to have far‐reaching effects on the ecosystem due to its significant energy requirements and interactions with habitat and other sympatric species. This study explores the impacts of bison reintroduction on the movement and resource use of another large‐bodied grazer, the Rocky Mountain bighorn sheep. Between 2018 and 2021, we collected data from GPS collars fit on 39 bighorn sheep and 11 bison. We analyzed home range patterns, resource selection, and interactions to investigate the potential for interspecific competition, facilitation, and resource complementarity. At the population level, bison and bighorn sheep exhibited low levels of spatial overlap and there was strong evidence of resource separation in all seasons. Interactions between species did not appear to affect sheep movement rates; however, we did see differences in forage selection patterns for sheep with overlapping home ranges with bison. Collectively, results did not support the potential for competition or facilitation between bison and bighorn sheep and instead provided the strongest evidence of complementarity.

In addition to being the most magnetic objects in the known Universe, magnetars are the only objects observed to generate fast-radio-burst-like emissions. The formation mechanism of magnetars is still highly debated and may potentially be probed with the magnetar velocity distribution. We carried out a 3 yr long astrometric campaign on Swift J1818.0−1607, the fastest-spinning magnetar, using the Very Long Baseline Array. After applying the phase-calibrating 1D interpolation strategy, we obtained a small proper motion of 8.5 mas yr ^−1 mag and a parallax of 0.12 ± 0.02 mas (uncertainties at 1 σ confidence throughout the Letter) for Swift J1818.0−1607. The latter is the second magnetar parallax and is among the smallest neutron star parallaxes ever determined. From the parallax, we derived the distance ${9.4}_{-1.6}^{+2.0}$ kpc, which locates Swift J1818.0−1607 at the far side of the Galactic central region. Combined with the distance, the small proper motion leads to a transverse peculiar velocity ${v}_{\perp }={48}_{-16}^{+50}$ km s ^−1 —a new lower limit to magnetar v _⊥ . Incorporating previous v _⊥ estimates of seven other magnetars, we acquired ${v}_{\perp }={149}_{-68}^{+132}$ km s ^−1 for the sample of astrometrically studied magnetars, corresponding to the three-dimensional space velocity $\sim {190}_{-87}^{+168}$ km s ^−1 , smaller than the average level of young pulsars. Additionally, we found that the magnetar velocity sample does not follow the unimodal young pulsar velocity distribution reported by Hobbs et al. at >2 σ confidence, while loosely agreeing with more recent bimodal young pulsar velocity distributions derived from relatively small samples of quality astrometric determinations.

Fast radio bursts (FRBs) are brilliant short-duration flashes of radio emission originating at cosmological distances. The vast diversity in the properties of currently known FRBs and the fleeting nature of these events make it difficult to understand their progenitors and emission mechanism(s). Here we report high time resolution polarization properties of FRB 20210912A, a highly energetic event detected by the Australian Square Kilometre Array Pathfinder (ASKAP) in the Commensal Real-time ASKAP Fast Transients survey, which show intraburst position angle (PA) variation similar to Galactic pulsars and unusual variation of Faraday rotation measure (RM) across its two sub-bursts. The observed intraburst PA variation and apparent RM variation pattern in FRB 20210912A may be explained by a rapidly spinning neutron star origin, with rest-frame spin periods of ∼1.1 ms. This rotation timescale is comparable to the shortest known rotation period of a pulsar and close to the shortest possible rotation period of a neutron star. Curiously, FRB 20210912A exhibits a remarkable resemblance to the previously reported FRB 20181112A, including similar rest-frame emission timescales and polarization profiles. These observations suggest that these two FRBs may have similar origins.

FRB 20220610A is a high-redshift fast radio burst (FRB) that has not been observed to repeat. Here, we present rest-frame UV and optical Hubble Space Telescope observations of the field of FRB 20220610A. The imaging reveals seven extended sources, one of which we identify as the most likely host galaxy with a spectroscopic redshift of z = 1.017. We spectroscopically confirm three additional sources to be at the same redshift and identify the system as a compact galaxy group with possible signs of interaction among group members. We determine the host of FRB 20220610A to be a star-forming galaxy with a stellar mass of ≈10 ^9.7 M _⊙ , mass-weighted age of ≈2.6 Gyr, and star formation rate (integrated over the last 100 Myr) of ≈1.7 M _⊙ yr ^−1 . These host properties are commensurate with the star-forming field galaxy population at z ∼ 1 and trace their properties analogously to the population of low- z FRB hosts. Based on estimates of the total stellar mass of the galaxy group, we calculate a fiducial contribution to the observed dispersion measure from the intragroup medium of ≈90–182 pc cm ^−3 (rest frame). This leaves a significant excess of ${515}_{-272}^{+122}$ pc cm ^−3 (in the observer frame); further observation will be required to determine the origin of this excess. Given the low occurrence rates of galaxies in compact groups, the discovery of an FRB in one demonstrates a rare, novel environment in which FRBs can occur. As such groups may represent ongoing or future mergers that can trigger star formation, this supports a young stellar progenitor relative to star formation.

Information and communication technologies (ICTs) play a significant role in transportation systems as they provide the connection between transportation system users, system planners, operators, and managers. Over the last few years, with advancements in ICTs, there has been a rapid change in travel behaviour. However, little is known about how ICTs and other emerging digital technologies are being utilised in transportation-related communication for public, private, and active transportation. To bridge this gap, this study aims to explore academic and grey literature focused on the use of digital technologies in transportation-related communication. The methodological approach employs a systematic literature review, with the PRISMA (Preferred Reporting Items for Systematic Reviews and Meta-Analysis) protocol, to identify digital technologies that are found promising among scholars and practitioners for research, testing and deployment. The study analysed 158 publications and identified some common themes of how these digital technologies can be implemented to improve access to transportation communication. The study finds that: (a) The main literature focus is how information is gathered and shared with transportation users; (b) Advances in mobile applications afford real-time route updates, easy navigation of complex routes, and personalised journey planning; (c) Some notable knowledge gaps exist in how these technologies are implemented—including, willingness to use and digital literacy to understand them; (d) Many digital technologies are at the trial or development phases, which have potential to improve how transportation information can be made readily accessible to all users.

In this study we present an inducible biosensor model for the Estrogen Receptor Beta (ERβ), GFP-ERβ:PRL-HeLa, a single-cell-based high throughput (HT) in vitro assay that allows direct visualization and measurement of GFP-tagged ERβ binding to ER-specific DNA response elements (EREs), ERβ-induced chromatin remodeling, and monitor transcriptional alterations via mRNA fluorescence in situ hybridization for a prolactin (PRL)-dsRED2 reporter gene. The model was used to accurately (Z’ = 0.58–0.8) differentiate ERβ-selective ligands from ERα ligands when treated with a panel of selective agonists and antagonists. Next, we tested an Environmental Protection Agency (EPA)-provided set of 45 estrogenic reference chemicals with known ERα in vivo activity and identified several that activated ERβ as well, with varying sensitivity, including a subset that is completely novel. We then used an orthogonal ERE-containing transgenic zebrafish (ZF) model to cross validate ERβ and ERα selective activities at the organism level. Using this environmentally relevant ZF assay, some compounds were confirmed to have ERβ activity, validating the GFP-ERβ:PRL-HeLa assay as a screening tool for potential ERβ active endocrine disruptors (EDCs). These data demonstrate the value of sensitive multiplex mechanistic data gathered by the GFP-ERβ:PRL-HeLa assay coupled with an orthogonal zebrafish model to rapidly identify environmentally relevant ERβ EDCs and improve upon currently available screening tools for this understudied nuclear receptor.

Abstract Identifying spatially variable genes (SVGs) is a key step in the analysis of spatially resolved transcriptomics data. SVGs provide biological insights by defining transcriptomic differences within tissues, which was previously unachievable using RNA-sequencing technologies. However, the increasing number of published tools designed to define SVG sets currently lack benchmarking methods to accurately assess performance. This study compares results of 6 purpose-built packages for SVG identification across 9 public and 5 simulated datasets and highlights discrepancies between results. Additional tools for generation of simulated data and development of benchmarking methods are required to improve methods for identifying SVGs.

Abstract Platelet activation may contribute to age‐related cerebrovascular dysfunction by interacting with the endothelial cells that regulate the response to vasodilatory stimuli. This study evaluated the relationship between a platelet inhibitor, prostacyclin, and cerebrovascular reactivity (CVR) in healthy young (n = 35; 25 ± 4 years; 17 women, 18 men) and older (n = 12; 62 ± 2 years; 8 women, 4 men) adults, who were not daily aspirin users, before and after cyclooxygenase inhibition. Prostacyclin was determined by levels of 6‐keto‐prostaglandin F1α (6‐keto PGF1α) in the blood. CVR was assessed by measuring the middle cerebral artery blood velocity response to hypercapnia using transcranial Doppler ultrasound before (CON) and 90 min after cyclooxygenase inhibition with indomethacin (INDO). In young adults, there were no associations between prostacyclin and middle cerebral artery CVR during CON (r = −0.14, P = 0.415) or INDO (r = 0.27, P = 0.118). In older adults, associations between prostacyclin and middle cerebral artery CVR during CON (r = 0.53, P = 0.075) or INDO (r = –0.45, P = 0.136) did not reach the threshold for significance. We also evaluated the relationship between prostacyclin and the change in CVR between conditions (ΔCVR). We found no association between ΔCVR and prostacyclin in young adults (r = 0.27, P = 0.110); however, in older adults, those with higher baseline prostacyclin levels demonstrated significantly greater ΔCVR (r = –0.74, P = 0.005). In conclusion, older adults with higher serum prostacyclin, a platelet inhibitor, may rely more on cyclooxygenase products for cerebrovascular reactivity to hypercapnia.

Abstract Objective Estimate the prevalence, and associated risk factors, of high school students who are considered at risk for an eating disorder based on screening measures. Methods An electronic search of nine databases was completed from their inception until 1st September 2022. A random-effects meta-analysis was conducted, and confounder (moderator) analyses and meta-regressions examined whether the overall prevalence estimate for of screen-based disordered eating (SBDE) was moderated by student age, BMI, or gender, as well as culture and type of SBDE assessment. Results The mean estimate of the prevalence of SBDE among high school students (K = 42 (66 datapoints), N = 56282] in the sample of 25 countries was 13% ([95% CI] = 10.0–16.8%, I 2 = 99.0%, Cochran's Q p = 0.001). This effect was not moderated by features of the samples such as gender, BMI, or age. Among cultures, non-Western countries had a higher prevalence of SBDE prevalence than Western countries, but the difference was not significant. There was considerable variability in the prevalence estimates as a function of the assessment measure, but no meaningful pattern emerged. Conclusion The estimated figure of 1 in 8 high school students with SBDE—unmoderated by gender and BMI—stands out as a problem in need of attention from public health officials, psychologists, psychiatrists, pediatricians, parents, and educators. There is a great need for innovative, integrated policy and program development all along the spectrum of health promotion and universal, selective, and indicated prevention. Further research is also needed to validate and refine this estimate by (a) conducting basic research on the accuracy of eating disorder screening measurements in samples ages 14 through 17; (b) examining representative samples in more countries in general and Latin American countries in particular; (c) clarifying the relationships between SBDE and age throughout the different phases of late childhood, adolescence, and emerging adulthood; and (d) investigating whether there are meaningful forms of disordered eating and whether these are associated with variables such as gender, ethnicity, and BMI.

Abstract The mechanisms governing chemotaxis in Chlamydomonas reinhardtii are largely unknown compared to those regulating phototaxis despite equal importance on the migratory response in the ciliated microalga. To study chemotaxis, we made a simple modification to a conventional Petri dish assay. Using the assay, a novel mechanism governing Chlamydomonas ammonium chemotaxis was revealed. First, we found that light exposure enhances the chemotactic response of wild-type Chlamydomonas strains, yet phototaxis-incompetent mutant strains, eye3-2 and ptx1, exhibit normal chemotaxis. This suggests that Chlamydomonas transduces the light signal pathway in chemotaxis differently from that in phototaxis. Second, we found that Chlamydomonas collectively migrate during chemotaxis but not phototaxis. Collective migration during chemotaxis is not clearly observed when the assay is conducted in the dark. Third, the Chlamydomonas strain CC-124 carrying agg1 −, the AGGREGATE1 gene (AGG1) null mutation, exhibited a more robust collective migratory response than strains carrying the wild-type AGG1 gene. The expression of a recombinant AGG1 protein in the CC-124 strain suppressed this collective migration during chemotaxis. Altogether, these findings suggest a unique mechanism; ammonium chemotaxis in Chlamydomonas is mainly driven by collective cell migration. Furthermore, it is proposed that collective migration is enhanced by light and suppressed by the AGG1 protein.

Following metastatic spread, many hormone receptor positive (HR+) patients develop a more aggressive phenotype with an observed loss of the HRs estrogen receptor (ER) and progesterone receptor (PR). During metastasis, breast cancer cells are exposed to high magnitudes of fluid shear stress (FSS). Unfortunately, the role for FSS on the regulation of HR expression and function during metastasis is not fully understood. This study was designed to elucidate the impact of FSS on HR+ breast cancer. Utilizing a microfluidic platform capable of exposing breast cancer cells to FSS that mimics in situ conditions, we demonstrate the impact of FSS exposure on representative HR+ breast cancer cell lines through protein and gene expression analysis. Proteomics results demonstrated that 540 total proteins and 1473 phospho-proteins significantly changed due to FSS exposure and pathways of interest included early and late estrogen response. The impact of FSS on response to 17β-estradiol (E2) was next evaluated and gene expression analysis revealed repression of ER and E2-mediated genes (PR and SDF1) following exposure to FSS. Western blot demonstrated enhanced phosphorylation of mTOR following exposure to FSS. Taken together, these studies provide initial insight into the effects of FSS on HR signaling in metastatic breast cancer.

Almond trees are the most cultivated nut tree in the world. The production of almonds generates large amounts of by-products, much of which goes unused. Herein, this study aimed to develop a green chemistry approach to identify and extract potentially valuable compounds from almond by-products. Initially, a screening was performed with 10 different Natural Deep Eutectic Solvents (NADESs). The mixture lactic acid/glycerol, with a molar ratio 1:1 (1:50 plant material to NADES (w/v) with 20% v/v of water) was identified as the best extraction solvent for catechin, caffeoylquinic acid, and condensed tannins in almond hulls. Subsequently, a method was optimized by a Design of Experiment (DoE) protocol using a miniaturized extraction technique, Microwave-Assisted Extraction (MAE), in conjunction with the chosen NADESs. The optimal conditions were found to be 70 °C with 15 min irradiation time. The optimal extraction conditions determined by the DoE were confirmed experimentally and compared to methods already established in the literature. With these conditions, the extraction of metabolites was 2.4 times higher, according to the increase in total peak area, than the established literature methods used. Additionally, by applying the multiparameter Analytical Greenness Metric (AGREE) and Green Analytical Process Index (GAPI) metrics, it was possible to conclude that the developed method was greener than the established literature methods as it includes various principles of green analytical chemistry.

Background: Patients with supratentorial cavernous malformations (SCMs) commonly present with seizures. First-line treatments for cavernoma-related epilepsy (CRE) include conservative management (antiepileptic drugs (AEDs)) and surgery. We compared seizure outcomes of CRE patients after early (≤6 months) vs. delayed (>6 months) surgery. Methods: We compared outcomes of CRE patients with SCMs surgically treated at our large-volume cerebrovascular center (1 January 2010–31 July 2020). Patients with 1 sporadic SCM and ≥1-year follow-up were included. Primary outcomes were International League Against Epilepsy (ILAE) class 1 seizure freedom and AED independence. Results: Of 63 CRE patients (26 women, 37 men; mean ± SD age, 36.1 ± 14.6 years), 48 (76%) vs. 15 (24%) underwent early (mean ± SD, 2.1 ± 1.7 months) vs. delayed (mean ± SD, 6.2 ± 7.1 years) surgery. Most (32 (67%)) with early surgery presented after 1 seizure; all with delayed surgery had ≥2 seizures. Seven (47%) with delayed surgery had drug-resistant epilepsy. At follow-up (mean ± SD, 5.4 ± 3.3 years), CRE patients with early surgery were more likely to have ILAE class 1 seizure freedom and AED independence than those with delayed surgery (92% (44/48) vs. 53% (8/15), p = 0.002; and 65% (31/48) vs. 33% (5/15), p = 0.03, respectively). Conclusions: Early CRE surgery demonstrated better seizure outcomes than delayed surgery. Multicenter prospective studies are needed to validate these findings.

Racial and socioeconomic health disparities are well documented in the literature. This study examined patient demographics, including socioeconomic status (SES), among individuals presenting with aneurysmal subarachnoid hemorrhage (aSAH) and unruptured intracranial aneurysm (UIA) to identify factors associated with aSAH presentation. A retrospective assessment was conducted of all patients with aSAH and UIA who presented to a large-volume cerebrovascular center and underwent microsurgical treatment from January 2014 through July 2019. Race and ethnicity, insurance type, and SES data were collected for each patient. Comparative analysis of the aSAH and UIA groups was conducted. Logistic regression models were also employed to predict the likelihood of aSAH presentation based on demographic and socioeconomic factors. A total of 640 patients were included (aSAH group, 251; UIA group, 389). Significant associations were observed between race and ethnicity, SES, insurance type, and aneurysm rupture. Non-White race or ethnicity, lower SES, and having public or no insurance were associated with increased odds of aSAH presentation. The aSAH group had poorer functional outcomes and higher mortality rates than the UIA group. Patients who are non-White, have low SES, and have public or no insurance were disproportionately affected by aSAH, which is historically associated with poorer functional outcomes.

Abstract Temperature and biodiversity changes occur in concert, but their joint effects on ecological stability of natural food webs are unknown. Here, we assess these relationships in 19 planktonic food webs. We estimate stability as structural stability (using the volume contraction rate) and temporal stability (using the temporal variation of species abundances). Warmer temperatures were associated with lower structural and temporal stability, while biodiversity had no consistent effects on either stability property. While species richness was associated with lower structural stability and higher temporal stability, Simpson diversity was associated with higher temporal stability. The responses of structural stability were linked to disproportionate contributions from two trophic groups (predators and consumers), while the responses of temporal stability were linked both to synchrony of all species within the food web and distinctive contributions from three trophic groups (predators, consumers, and producers). Our results suggest that, in natural ecosystems, warmer temperatures can erode ecosystem stability, while biodiversity changes may not have consistent effects.

Abstract Mechanical forces drive critical cellular processes that are reflected in mechanical phenotypes, or mechanotypes, of cells and their microenvironment. We present here “Rupture And Deliver” Tension Gauge Tethers (RAD-TGTs) in which flow cytometry is used to record the mechanical history of thousands of cells exerting forces on their surroundings via their propensity to rupture immobilized DNA duplex tension probes. We demonstrate that RAD-TGTs recapitulate prior DNA tension probe studies while also yielding a gain of fluorescence in the force-generating cell that is detectable by flow cytometry. Furthermore, the rupture propensity is altered following disruption of the cytoskeleton using drugs or CRISPR-knockout of mechanosensing proteins. Importantly, RAD-TGTs can differentiate distinct mechanotypes among mixed populations of cells. We also establish oligo rupture and delivery can be measured via DNA sequencing. RAD-TGTs provide a facile and powerful assay to enable high-throughput mechanotype profiling, which could find various applications, for example, in combination with CRISPR screens and -omics analysis.

Abstract Background Anaphylaxis is a sudden multisystem allergic reaction which may result in a fatal outcome if not treated promptly. Guidelines worldwide suggest intramuscular adrenaline as the first‐line treatment for anaphylaxis outside a perioperative reaction. Adrenaline autoinjectors (AAIs) are widely used self‐administrable devices, especially in community settings. Different commercial AAIs have been authorized to be marketed in Europe. For an AAI to be efficacious, a rapid adrenaline delivery in patients, including those who are overweight or obese, resulting in an optimal cardiovascular (CV) response, is a key feature. AAIs are designed to achieve this requirement, which is reflected in their differing functional properties such as primary container selection, drug delivery mechanism (cartridge‐or syringe‐based), needle length, needle gauge, and adrenaline dose (150 μg, 300 μg, or 500 μg). However, the differences in functional properties across these devices may play a critical role in achieving these requirements as well as the differences in ergonomics in the handling of these devices. The purpose of this review Considering the dynamic pharmacokinetic/pharmacodynamic (PK/PD) profiles of different AAIs marketed in Europe and their effect on adrenaline delivery, the expert panel, also serving as author for this paper have carried out a detailed analysis of the PK/PD profiles of four AAIs, namely, Anapen, Emerade, EpiPen, and Jext, to delineate the adrenaline delivery and their subsequent physiological effects on the backdrop of device characteristics, dose strength, and the skin‐to‐muscle distances of the participants.

Most children cease napping between 2 and 5-years-old, with considerable inter-child variability. We tested the predictors of early nap cessation (i.e., children who cease napping before three years old) using longitudinal data from 5504 Canadian children (51.1% male; 89.8% White) in three cohorts with two timepoints each. Children were 0–1-years-old at baseline (M = 10.19 months SD = 3.95 months) and 2–3-years-old at follow-up (M = 30.83 months, SD = 4.60 months). Parents reported on demographic, perinatal, growth, developmental, child and parent functioning, and child sleep variables. At follow-up, 10.9% ± 0.8% had ceased napping. Multigroup multivariate logistic regression was conducted using a model building approach to identify predictors of early nap cessation. Early nap cessation was predicted by older child age (ORs range from 1.15 to 1.24, moderated by cohort), female sex (OR = 1.29; 95% CI: 1.07–1.55), having an older sibling (OR = 1.33; 95% CI: 1.10–1.62), achieving more developmental milestones (OR = 1.08; 95% CI: 1.03–1.13), and longer nighttime sleep duration (OR = 1.06; 95% CI: 1.01–1.11). Non-White ethnicity (OR = 0.41; 95% CI: 0.28–0.60), birthweight < 2500 gs (OR = 0.60; 95% CI: 0.37–0.96), parent working/in school (ORs range from 0.50 to 0.58, moderated by cohort), and the birth mother consuming alcohol during pregnancy (OR = 0.56; 95% CI: 0.40–0.79) were related to a lower likelihood of nap cessation. Findings suggest nap cessation is influenced by developmental and socio-environmental factors.

Abstract The management objectives of many protected areas must meet the dual mandates of protecting biodiversity while providing recreational opportunities. It is difficult to balance these mandates because it takes considerable effort to monitor both the status of biodiversity and impacts of recreation. Using detections from 45 camera traps deployed between July 2019 and September 2021, we assessed the potential impacts of recreation on spatial and temporal activity for 8 medium‐ and large‐bodied terrestrial mammals in an isolated alpine protected area: Cathedral Provincial Park, British Columbia, Canada. We hypothesised that some wildlife perceive a level of threat from people, such that they avoid ‘risky times’ or ‘risky places’ associated with human activity. Other species may benefit from associating with people, be it through access to anthropogenic resource subsidies or filtering of competitors/predators that are more human‐averse (i.e., human shield hypothesis). Specifically, we predicted that large carnivores would show the greatest segregation from people while mesocarnivores and ungulates would associate spatially with people. We found spatial co‐occurrence between ungulates and recreation, consistent with the human shield hypothesis, but did not see the predicted negative relationship between larger carnivores and humans, except for coyotes (Canis latrans). Temporally, all species other than cougars (Puma concolor) had diel activity patterns significantly different from that of recreationists, suggesting potential displacement in the temporal niche. Wolves (Canis lupus) and mountain goats (Oreamnos americanus) showed shifts in temporal activity away from people on recreation trails relative to off‐trail areas, providing further evidence of potential displacement. Our results highlight the importance of monitoring spatial and temporal interactions between recreation activities and wildlife communities, in order to ensure the effectiveness of protected areas in an era of increasing human impacts.

Cutaneous T-cell lymphoma (CTCL) is a rare type of non-Hodgkin lymphoma of the skin, where at later stages skin-homing malignant T-cells affect lymph nodes, blood, and visceral organs. Even though early CTCL does not affect survival, it can progress to more advanced stages of disease and have a significant effect on the quality of life of patients. Although expectant management is a treatment consideration in early disease stages, most patients cycle through different skin-directed therapies throughout their lifetime. It can become a challenge to manage the serious and accumulating risk of side effects of these therapies, including various skin cancers and skin damage. Adverse effects from topical therapies limit their long-term utility. Thus, there is an unmet need for well-characterized therapies that have a rapid onset of action and minimal long-term/cumulative side effect profile. Most recently, the results of a Phase 3 study of topical HyBryte™ as a potential treatment for CTCL demonstrated its efficacy and safety profile. This article summarizes what is known about HyBryte™, focuses on its mechanism of action, and highlights its effectiveness, safety, and tolerability in the context of other current FDA-approved topical therapies for CTCL.

Effective species management relies on evidence-based goals that address the processes influencing population demography. These goals therefore require understanding how stressors affect the species of interest, and the food web within which the focal species is embedded. For woodland caribou, a conservation priority across much of its range, habitat loss and alteration is a primary cause of decline. Governments have identified maximum disturbance targets whereby a maximum of 35% total disturbance provides a 60% likelihood of self-sustaining caribou populations, but targets specific for linear features such as roads and seismic lines — which play a key role in altering ecological processes and are a primary focus for habitat restoration — have not been established. We created a framework to conceptually link stressors (linear features) to caribou declines via ecological mechanisms, and used this framework to guide a literature review to support the development of a linear feature-based management target for Southern Mountain Caribou. Despite the vast amount of research investigating the mechanisms leading to the negative relationship between caribou demography and linear features, our review of 54 peer-reviewed manuscripts and 99 grey-literature reports found little to no evidence of a threshold in these relationships in which to inform a linear feature-based management target for caribou population growth. Most studies evaluated how linear features modify space use of caribou, their predators, and the primary prey of those predators, whereas few mechanistically linked linear features to caribou demography. Our work provides a foundation in which other systems can conceptualize how stressors are linked to species declines, and to support the development of evidence-based management goals.

Abstract Estimating the strength of interactions among species in natural communities has always been a challenge for empirical ecologists. Sessile organisms, like plants or corals, often occur in metacommunities where they compete only with their immediate neighbours but disperse propagules over a wider area. To estimate the strength of competitive interactions, ecologists often count abundances in cells on a spatial grid for at least two time‐points. This data is then analysed using regression, by modelling the change in population size as a function of local densities, using cells as independent data‐points: a technique known as space‐for‐time substitution. These analyses generate estimates of competition coefficients; however, the method ignores dispersal among cells. To determine the impact of ignoring dispersal, we derived the bias that would arise when we apply regression methods to a metacommunity in which a fraction of seeds disperse beyond their natal cells but this dispersal is ignored in the model fitting process. We present results from a range of population models that make different assumptions about the nature of competition and assess the performance of our bias formulae by analysing data from simulated metacommunities. We reveal that: estimates of competition coefficients are biased when dispersal is not properly accounted for; and the resulting bias is often correlated with abundance, with rare species suffering the greatest overestimation. We also provide a standardized metric of competition that allows the bias to be calculated for a broad range of other population models. Our study suggests that regression methods that ignore dispersal produce biased estimates of competition coefficients when using space‐for‐time substitution. Our analytical bias formula allows empirical ecologists to potentially correct for biases, but it requires either tailored experiments in controlled conditions or an estimate of the average dispersal rate in a natural community, so may be challenging to apply to real datasets.

Background: Autism spectrum disorder (ASD) is a neurodevelopmental disorder diagnosed based on social impairment, restricted interests, and repetitive behaviors. Contemporary theories posit that cerebellar pathology contributes causally to ASD by disrupting error-based learning (EBL) during infancy. The present study represents the first test of this theory in a prospective infant sample, with potential implications for ASD detection. Methods: Data from the Infant Brain Imaging Study (n = 94, 68 male) were used to examine 6-month cerebellar functional connectivity magnetic resonance imaging in relation to later (12/24-month) ASD-associated behaviors and outcomes. Hypothesis-driven univariate analyses and machine learning–based predictive tests examined cerebellar–frontoparietal network (FPN; subserves error signaling in support of EBL) and cerebellar–default mode network (DMN; broadly implicated in ASD) connections. Cerebellar-FPN functional connectivity was used as a proxy for EBL, and cerebellar-DMN functional connectivity provided a comparative foil. Data-driven functional connectivity magnetic resonance imaging enrichment examined brain-wide behavioral associations, with post hoc tests of cerebellar connections. Results: Cerebellar-FPN and cerebellar-DMN connections did not demonstrate associations with ASD. Functional connectivity magnetic resonance imaging enrichment identified 6-month correlates of later ASD-associated behaviors in networks of a priori interest (FPN, DMN), as well as in cingulo-opercular (also implicated in error signaling) and medial visual networks. Post hoc tests did not suggest a role for cerebellar connections. Conclusions: We failed to identify cerebellar functional connectivity–based contributions to ASD. However, we observed prospective correlates of ASD-associated behaviors in networks that support EBL. Future studies may replicate and extend network-level positive results, and tests of the cerebellum may investigate brain-behavior associations at different developmental stages and/or using different neuroimaging modalities.

Forest roads provide access to remote wooded areas, serving as key transportation routes and contributing to human impact on the local environment. However, large animals, such as bears (Ursus sp.), moose (Alces alces), and caribou (Rangifer tarandus caribou), are affected by their presence. Many publicly available road layers are outdated or inaccurate, making the assessment of landscape objectives difficult. To address these gaps in road location data, we employ CubeSat Imagery from the Planet constellation to predict the occurrence of road probabilities using a SegNet Convolutional Neural Network. Our research examines the potential of a pre-trained neural network (VGG-16 trained on ImageNet) transferred to the remote sensing domain. The classification is refined through post-processing, which considers spatial misalignment and road width variability. On a withheld test subset, we achieve an overall accuracy of 99.1%, a precision of 76.1%, and a recall of 91.2% (F1-Score: 83.0%) after considering these effects. We investigate the performance with respect to canopy coverage using a spectral greenness index, topography (slope and aspect), and land cover metrics. Results found that predictions are best in flat areas, with low to medium canopy coverage, and in the forest (coniferous and deciduous) land cover classes. The results are vectorized into a drivable road network, allowing for vector-based routing and coverage analyses. Our approach digitized 14,359 km of roads in a 23,500 km2 area in British Columbia, Canada. Compared to a governmental dataset, our method missed 10,869 km but detected an additional 5774 km of roads connected to the network. Finally, we use the detected road locations to investigate road age by accessing an archive of Landsat data, allowing spatiotemporal modelling of road access to remote areas. This provides important information on the development of the road network over time and the calculation of impacts, such as cumulative effects on wildlife.
