- Abstract
The current in vitro study aimed to investigate the effects of a processed sugarcane extract on the viability of avian Eimeria sporozoites. Treatments were applied to hatched sporozoites: 1) without additives (no-treatment control); 2) with ethanol; 3) with salinomycin; 4) with Polygain™. All treatments were incubated in RPMI media containing live sporozoites at 37 °C for 14 h and then the number of viable sporozoites were counted. Compared to the no-treatment control, Polygain™ decreased (P < 0.001) the counts of E. maxima, E. acervulina, E. bruneti, and E. mitis sporozoites to a level similar to salinomycin (P > 0.05). In conclusion, Polygain™ could be a potential candidate as an anticoccidial agent.

The measurement of anti-streptokinase antibodies can distinguish the patients who may benefit from streptokinase and those who should be treated with some other thrombolytic regimen. Neutralising titration test is a commonly used classical assay for measuring anti-streptokinase antibodies and in this assay the ability of anti-streptokinase antibodies in patients' sera in preventing the lytic effect of streptokinase is assessd. As we showed previously the presence of r-tPA in the serum may interefere with the neutralising titration test, we investigated this interference in vitro. The level of neutralising anti-streptokinase antibodies in a serum sample with known levels of the antibodies were measured in absence and presence of increasing amount of r-tPA including therapeutic values. Increasing amount of r-tPA in vitro induced a sudden reduction in the measurable titre of anti-streptokinase antibody levels in a serum with elevated levels of anti-streptokinase antibodies. This effect of r-tPA, which is through activation of plasminogen has not been reported previously. We suggest this assay is unsuitable for clinical diagnosis of anti-streptokinase antibodies.

- Abstract
Cheese microbiota and metabolites and their inter-relationships that underpin specific cheese quality attributes remain poorly understood. Here we report that multi-omics and integrative data analysis (multiple co-inertia analysis, MCIA) can be used to gain deeper insights into these relationships and identify microbiota and metabolite fingerprints that could be used to monitor product quality and authenticity. Our study into different brands of artisanal and industrial cheddar cheeses showed that Streptococcus, Lactococcus and Lactobacillus were the dominant taxa with overall microbial community structures differing not only between industrial and artisanal cheeses but also among different cheese brands. Metabolome analysis also revealed qualitative and semi-quantitative differences in metabolites between different cheeses. This also included the presence of two compounds (3-hydroxy propanoic acid and O-methoxycatechol-O-sulphate) in artisanal cheese that have not been previously reported in any type of cheese. Integrative analysis of multi-omics datasets revealed that highly similar cheeses, identical in age and appearance, could be distinctively clustered according to cheese type and brand. Furthermore, the analysis detected strong relationships, some previously unknown, which existed between the cheese microbiota and metabolome, and uncovered specific taxa and metabolites that contributed to these relationships. These results highlight the potential of this approach for identifying product specific microbe/metabolite signatures that could be used to monitor and control cheese quality and product authenticity.

20. Microbiota and Metabolite Profiling Combined With Integrative Analysis for Differentiating Cheeses of Varying Ripening Ages

Cheese maturation and flavor development results from complex interactions between milk substrates, cheese microbiota and their metabolites. In this study, bacterial 16S rRNA-gene sequencing, untargeted metabolomics (gas chromatography-mass spectrometry) and data integration analyses were used to characterize and differentiate commercial Cheddar cheeses of varying maturity made by the same and different manufacturers. Microbiota and metabolite compositions varied between cheeses of different ages and brands, and could be used to distinguish the cheeses. Individual amino acids and carboxylic acids were positively correlated with the ripening age for some brands. Integration and Random Forest analyses revealed numerous associations between specific bacteria and metabolites including a previously undescribed positive correlation between Thermus and phenylalanine and a negative correlation between Streptococcus and cholesterol. Together these results suggest that multi-omics analyses has the potential to be used for better understanding the relationships between cheese microbiota and metabolites during ripening and for discovering biomarkers for validating cheese age and brand authenticity.

The expression of genes that control germination-related processes in corn (Zea mays L.) is influenced by environmental factors. Germination of seeds may be facilitated by hormonal priming. The purpose of this investigation was to quantify the effects of different germination temperatures [(5, 10, 15, 20, 25, 30, 35, and 40°C), NaCl-induced stress (0, −0.4, −0.8, and −1.2 MPa), and priming solutions (control, hydropriming, abscisic acid (ABA), gibberellic acid (GA), and salicylic acid (SA)] (Experiment 1). Effects of germination temperatures, PEG 6000-induced stress (0, −0.4, −0.8, and −1.2 MPa), and priming solutions were also evaluated separately (Experiment 2). In both cases, a completely randomized design with four replications was used. Increasing temperatures from 5 to 25°C gradually improved germination percentage and rate, whereas temperatures > 25°C decreased these indices. After imposing drought (PEG 6000-induced stress) or salinity (NaCl-induced stress) treatments, hormonal priming caused germination to occur at a lower base temperature, compared with the non-priming treatment. However, the effect of hormonal priming was dependent on temperature. At sub-optimal temperatures (< 25°C), the highest germination percentage and rate were recorded after GA priming. At above-optimal temperatures (> 25°C), ABA priming resulted in the highest germination percentage and rate. Moreover, hydrothermal time constant decreased in hormone-treated seeds. Based on coefficient of determination (R2) and root mean square error (RMSE), a dent-like model predicted cardinal temperatures more accurately than a beta model did. Generally, GA-, SA-, and ABA-priming were recommended under sub-optimal, optimal, and above-optimal temperatures, respectively. [ABSTRACT FROM AUTHOR]

Objectives. 1) To investigate ECG changes following co-proxamol (paracetamol 325 mg and dextropropoxyphene 32.5 mg) overdose in comparison to co-codamol (paracetamol and codeine) or co-dydramol (paracetamol and dihydrocodeine) in a prospective study. 2) To examine the relationship between estimated dextropropoxyphene dose and ECG changes in a larger patient population. Background. Co-proxamol is a common cause of drug-induced death and hospital admission in the United Kingdom. ECG changes following dextropropoxyphene have been reported in animals and man, including QRS prolongation. Methods. The prospective study was conducted on 15 patients and controls with overdose. A retrospective study of a cohort of 159 co-proxamol overdoses from a combined data set from Edinburgh and Newcastle, Australia was also conducted. The measured or estimated 'four hour' plasma paracetamol level was used as a surrogate of the amount of dextropropoxyphene ingested. Results. In the prospective study co-proxamol overdose caused statistically significant QRS prolongation (mean [95% CI] 99.36 [96.19, 102.53] msec), compared to the other combination opioid-paracetamol products (82.84 [80.81, 84.88] msec) but no effect on PR or QTc. QRS duration increase was evident soon after exposure and remained prolonged and stable over the following 24 h. In the retrospective cohort study a dose dependency of effect on QRS was documented, although the correlation coefficient relating paracetamol level to effect was relatively weak (r=0.338, Sig. [2-tailed] 0.003, n=74). Conclusions. QRS is significantly prolonged in co-proxamol overdose, and this prolongation is dose dependent. These findings have clinical relevance to the management of patients with coproxamol poisoning. Drugs and chemicals are almost easily available in Iran. Natural toxins as poisonous plants and animals also exist in most parts of the country. Therefore, acute poisonings, either intentional or accidental and also drug abuse/addiction are common in Iran. In spite of these difficulties there is no center for poison control and surveillance in this country to gather information and analyse data. The files of a systematic randomised ten percent of all hospital-referred poisoned patients from 21 March 1993 to 20 March 2000 in Imam Reza (p) University Hospital of Mashhad (71589 cases) were screened retrospectively. Young adults (40.3%) and school children (22.9%) were the most vulnerable group. Mean age was 22.3 (S.D. 14.38) years with a minimum of less than one and a maximum of 98 years old. A female predominance was found (53.4%). Intentional poisoning was more common (54.4%) than accidental exposures (45.2%). Fourteen cases were classified as criminal poisoning. 79.7% of exposures were via ingestion, followed by dermal exposures (14.1%), and inhalation (6.2%). The majority (83.7% of patients were from urban areas. Most patients (68.6%) were treated in the Emergency Toxicology Clinic and discharged, 19.2% were temporarily hospitalized and 11.3% were hospitalized for 24 hr. Main groups of poisons were pharmaceuticals (61.4%), chemicals (22.8%), and natural toxins (16.6%). The overall number of poisoned patients was higher in spring and summer (62.8%). In conclusion, acute poisonings, particularly self-poisonings, are common in Iran. Since medical documentation is not routinely provided in this country the results of this retrospective study can be used for surveillance. Establishment of fluent data gathering and analysis within the local health system are challenges for the future. The conventional advanced process yield index ST pk is widely applied in industry to provide an exact measure of the overall production yield whose quality characteristics are mutually independent and multivariate normally distributed. While one can find numerous studies that consider a crisp estimation of STpk to evaluate and test the overall process yield, the recorded measurements of product quality characteristics are not always reported precisely. This paper presents a new fuzzy-based method to assess the overall process yield in the presence of a specified degree of ambiguity for the sample data. After finding a fuzzy estimator of STpk based on Buckley's approach, a new fuzzy three-decision testing rule is proposed to evaluate process performance based on critical values and fuzzy p-values. Subsequently, this work extends the application of the proposed method to the class of correlated characteristics by adopting the principal component analysis technique. The introduced fuzzy testing procedure includes the existing customary binary-decision testing rule as a spacial case. In addition, comparative studies are conducted to display the benefits of the proposed rule. Seed aging or deterioration is considered as one of the seed capacity reducing factors and limiting seed germination, hence, realizing the effective physiological factors on seed deterioration is very important. In order to study the germination indices and enzymatic and non-enzymatic antioxidants in wheat (Triticumaestivum L. cv. Pishtaz) seed as affected by accelerated seed aging and gibberellic acid (GA
3 ) priming, a laboratory experiment was conducted in Ferdowsi University of Mashhad, Mashhad, Iran, during 2017–2018. For this purpose, different levels of seed aging (100% relative humidity at 40°C for 4, 6 and 7 days, respectively) and GA3 -primed seeds (0, 25, 50 and 100 mg/L) were the first and second experimental factors, respectively. The results indicated that seed germination percentage and indices, superoxide dismutase and ascorbate peroxidase activities, vitamin E, glutathione and ascorbic acid contents were reduced with severe deterioration, compared with low deterioration, whereas, the mentioned traits were improved by increasing the levels of GA3 seed priming. Overall, the results of this experiment suggest that the GA3 -primed seed can be an effective approach to improve the seed germination and seedling growth in wheat under aging conditions. Although there have been a substantial number of reviews published on the health effects of lead, these reviews have predominantly focused on recent publications and rarely look at older, more historical articles. Old documents on lead can provide useful insight in establishing the historical context of lead usage and its modes of toxicity. The objective of this review is to explore historical understandings and uses of lead prior to the 20th century. One hundred eighty-eight English language articles that were published before the year 1900 were included in this review. Major themes in historical documentation of lead toxicology include lead's use in medical treatments, symptoms of lead poisoning, treatments for lead poisoning, occupational lead poisonings, and lead contamination in food and drinking water. The results of this review indicate that lead's usage was widespread throughout the 19th century, and its toxic properties were well-known. Common symptoms of lead poisoning and suggested treatments were identified during this time period. This review provides important insight into the knowledge and uses of lead before the 20th century and can serve as a resource for researchers looking at the history of lead. [ABSTRACT FROM AUTHOR]

Carbon nanotube-natural biopolymer nanovectors have important potential applications in delivery system for drugs and biomolecules. In this work, the use of multi-wall CNTs as nanoreserviors for drug loading and controlled release is demonstrated .We synthesized CNT-based Drug delivery systems; MWCNT-CS nanoparticles based on an ionotropic gelation method as a sustained-release systems for the delivery of Tenofovir (hydrophilic anti-retroviral drug). Molecularly imprinted polymer used as shell for encapsulating the synthesized polymer to reduce the toxicity of CNT and improved theit application in Drug Delivery System. The prepared nanoparticles were characterized by FTIR spectroscopy. TGA was applied to study the thermal stabilities, and SEM to investi-gate the morphology. - Abstract
We investigated the effects of postharvest application of carbon disulphide (CS2) in various concentrations (0, 15, 25, 35, 45 and 55 ml m-3) and with different exposure duration (24, 48, 72 and 96 h) on breaking of dormancy and sprouting of potato (Solanum tuberosum L., cv. Marfona) minitubers of two ages (freshly harvested and one week after harvest) and two weight classes (1.5 and 12 g). In comparison with the control minitubers, CS2 treated minitubers showed significantly shorter dormancy and better sprouting. More rotting and weaker responses were observed in freshly harvested treated minitubers compared with minitubers treated one week after harvest. The number of sprouts per minituber and their length were significantly enhanced by treating minitubers with CS2 compared with the untreated control minitubers, but there were strong interactions with minituber weight. Results showed that duration of CS2 treatment was more important than concentration and longer duration of CS2 treatment exhibited a stronger action on breaking dormancy and sprouting of potato minitubers than shorter treatments. But, when longer duration was accompanied with higher concentration, treatment with CS2 led to formation of needle sprouts, which are undesirable as they do not produce vigorous stems

Responses to prolonged low-temperature treatment of imbibed seeds (vernalization) were examined in barley (Hordeum vulgare). These occurred in two phases: the perception of prolonged cold, which occurred gradually at low temperatures, and the acceleration of reproductive development, which occurred after vernalization. Expression of the VERNALIZATION1 gene (HvVRN1) increased gradually in germinating seedlings during vernalization, both at the shoot apex and in the developing leaves. This occurred in darkness, independently of VERNALIZATION2 (HvVRN2), consistent with the hypothesis that expression of HvVRN1 is induced by prolonged cold independently of daylength flowering-response pathways. After vernalization, expression of HvVRN1 was maintained in the shoot apex and leaves. This was associated with accelerated inflorescence initiation and with down-regulation of HvVRN2 in the leaves. The largest determinant of HvVRN1 expression levels in vernalized plants was the length of seed vernalization treatment. Daylength did not influence HvVRN1 expression levels in shoot apices and typically did not affect expression in leaves. In the leaves of plants that had experienced a saturating seed vernalization treatment, expression of HvVRN1 was higher in long days, however. HvFT1 was expressed in the leaves of these plants in long days, which might account for the elevated HvVRN1 expression. Long-day up-regulation of HvVRN1 was not required for inflorescence initiation, but might accelerate subsequent stages of inflorescence development. Similar responses to seed vernalization were also observed in wheat (Triticum aestivum). These data support the hypothesis that VRN1 is induced by cold during winter to promote spring flowering in vernalization-responsive cereals.

Background: Acute poisoning is a common health problem worldwide. Traditionally, the frequency of admitted cases, number of deaths or online resources were used to measure the volume of this problem. In this study, the burden of acute poisoning is addressed for the first time. Methods: Data related to 44,340 poisoned patients from 2004 to 2013 were analysed. Two sources including hospital information and forensic medicine data of the Khorasan Razavi catchment area. The disability-adjusted life years (DALY) method – including years of life lost (YLL) and years lived with disability (YLD) – was used to estimate the burden. Results: In this period and for all acute poisonings, the mean (SD, minimum–maximum) incidence of acute poisoning was 30,485 (3049 per year). In total, 29,208 DALY were lost, including 8499 YLD and 20,709 YLL. DALY have increased from 1808 in 2004 to 4283 in 2013. Among the total DALY over this 10-year period, 17,592 (60%) were related to males. Narcotic use accounted for the largest percentage of DALY (19.6%), followed by analgesics–sedative use (16.3%) and illegal drug use (11.41%). The health loss due to these causes was large (YLD = 8499 and YLL = 20,709) and showed a rapidly increasing trend. Conclusions: Estimating DALY is more informative than using the frequency of death. If we expect the same ratio in the country, we would expect a DALY rate of 729,750 years for the 75,000,000 inhabitants of this country in this time period or 72,975 DALY lost per year for poisonings.
