Abstract

Fungi belonging to the genus Tuber produce edible ascocarps known as truffles. Tuber magnatum Picco may be the most appreciated truffle species given its peculiar aroma. While its life cycle is not yet fully elucidated, some studies demonstrated an active role of microorganisms. The main goal of this study was to determine how the T. magnatum microbiome varies across space and time. To address this, we characterized microbial communities associated with T. magnatum through high-throughput amplicon sequencing of internal transcribed spacer (ITS) and 16S rDNAs in three productive natural sites in Italy across 2 years. At each site, four truffles were sampled as well as the soil underneath and at 40, 100, and 200 cm from the harvesting points, to assess for microbial variation between substrates, years, and sites. A statistically significant site-related effect on microbial communities was identified, whereas only the prokaryotic community was significantly affected by the distance of soil from the truffle. Significant differences between sampling years were also found, demonstrating a possible relation among rainfall precipitation and Firmicutes and Actinobacteria. Thirty-six bacterial OTUs in truffles and 11 bacterial OTUs in soils beneath truffles were identified as indicator taxa. As shown for other truffle species, the dominance of Bradyrhizobium, Rhizobium, and Ensifer spp. within the truffle fruiting body suggests an evolutionary adaptation of this microorganism to the genus Tuber. The present work offers novel and relevant insights into the microbial ecology of T. magnatum ecosystems and fruiting bodies. The function and role of these bacteria in the truffle microbiome and life cycle are in need of further investigation.

Abstract

The organic fraction of soils is critically important to soil health and optimal ecosystem functioning. Traditional analysis of soil organic horizons (O horizons) has been dependent upon laboratory-based instrumentation. Simultaneously, the use of proximal sensors such as portable X-ray fluorescence (PXRF) spectrometry along with visible near infrared diffuse reflectance spectroscopy (VisNIR DRS) has gained popularity for providing rapidly acquired spectral and elemental data useful for soil physicochemical property quantification. However, PXRF and VisNIR DRS have mostly been applied to the assessment of mineral soils. This preliminary study evaluated 136 organic laden soil samples (most aptly described as upland, non-saturated O horizons) using both laboratory based instrumentation (CN analyzer) and proximal sensors to evaluate total carbon (TC) and total nitrogen (TN). Results revealed that combining model outcomes using model fusion improved TC and TN prediction accuracies relative to using an individual instrument (PXRF or VisNIR DRS) or model averaging with improvements in root mean square error (RMSE) on the order of 10–47% and 10–67% for TC and TN, respectively. Partial least squares+random forest (PLS+RF) approaches emerged as the best model for predicting both TC and TN in organic laden soil samples. These results suggest that the strong predictive applications of proximal sensors extensively documented on mineral soils, may show similar promise for determination of a wide number of physicochemical properties on organic soil matrices, yet further exploration with a larger and more diverse dataset is recommended.

Abstract

Subaqueous soils have to be studied with proper methodologies, according to a pedological approach and considering the key role of animal bioturbation. Morphological and chemical characteristics of submerged soils found in the MPA of "Torre del Cerrano", Adriatic sea (Italy), have been studied in this way, and we conclude that submarine soils of the MPA are important examples of pedogenesis promoted by animals. Soils from the highly protected marine area hosted a major biodiversity than those from the sub- and non-protected areas, while no striking differences in terms of soil physical and chemical characteristics among protected, sub-protected and non-protected areas occurred., EQA - International Journal of Environmental Quality, Vol 19 (2015)

Abstract

Physical and chemical soil properties are generally correlated with the parent material, as its composition may influence the pedogenetic processes, the content of nutrients, and the element biocycling. This research studied the chemical and biochemical properties of the A horizon from soils developed on different rocks like amphibolite, serpentinite, phyllite, and granite under a relatively similar climatic regime from Galicia (northwest Spain). In particular, the effect of the parent material on soil evolution, organic carbon sequestration, and the hormone-like activity of humic and fulvic acids were tested. Results indicated that all the soils were scarcely fertile because of low concentrations of available P, exchangeable Ca (except for the soils on serpentinite and phyllite), and exchangeable K, but sequestered relevant quantities of organic carbon. The scarce soil fertility was common to all soils independently of the parent material, and we attributed this similarity to the pedogenetic pressure induced by the climatic conditions. Also, the hormone-like activity of humic and fulvic acids, similar for all the soils, was probably due to pedogenesis. We hypothesized that the hormone-like activity of the humic substances helps growth and diffusion of vegetation in low fertile soils and, consequently, soil organic carbon sequestration too.
