The aim was to investigate the specific sensitivity of endothelial cells to diamond nanoparticles dependent on exposure to nanoparticles.Methods: Diamond nanoparticles were characterized with Raman spectroscopy, Fourier-transform infrared spectroscopy (FTIR) and dynamic light scattering (DLS). Toxicity of diamond nanoparticles was assessed for endothelial cells (HUVEC), human mammary epithelial cells (HMEC) and HS-5 cell line. The effect of diamond nanoparticles on the level of ROS, NO, NADPH and protein synthesis of angiogenesis-related proteins of endothelial cells was evaluated.Results and Discussion: Our studies demonstrated severe cell type-specific toxicity of diamond nanoparticles to endothelial cells (HUVEC) depending on nanoparticle surface interaction with cells. Furthermore, we have assessed the effect on cytotoxicity of the bioconjugation of nanoparticles with a peptide containing the RGD motive and a serum protein corona. Our study suggests that the mechanical interaction of diamond nanoparticles with the endothelial cell membranes and the endocytosis of nanoparticles lead to the depletion of NADPH, resulting in an intensive synthesis of ROS and a decrease in the availability of NO. This leads to severe endothelial toxicity and a change in the protein profile, with changes in major angiogenesis-related proteins, including VEGF, bFGF, ANPT2/TIE-2, and MMP, and the production of stress-related proteins, such as IL-6 and IL-8.Conclusion: We confirmed the presence of a relationship between the toxicity of diamond nanoparticles and the level of cell exposure to nanoparticles and the nanoparticle surface. The results of the study give new insights into the conditioned toxicity of nanomaterials and their use in biomedical applications.Keywords: diamond nanoparticles, endothelial cells, nanotoxicity, oxidative toxicity

This article presents the microstructure, hardness, fracture toughness coefficient KIC and phase composition of submicron WC-4Co carbides. The carbides were sintered using the innovative U-FAST (Upgraded Field Assisted Sintering Technology) method, from mixtures of WC-Co powders with an average WC grain size of 0.4 µm and 0.8 µm. The obtained sinters were characterized by a relative density above 99% of the theoretical density. The hardness of the obtained composites was above 2000 HV30, while the KIC coefficient was about 8 MPa m1/2.

Zymoseptoria tritici is a fungal pathogen causing losses in wheat yields. Here, we present new primer sets for species-specific identification of this microorganism in wheat leaf samples using conventional PCR. Primer sets were validated in silico using tools available in genetic databases. Furthermore, in vitro tests were also carried out on 190 common wheat samples with visual symptoms of Septoria tritici blotch (STB) collected in Poland in three growing seasons (2015, 2016, 2017). The designed primer sets showed full hybridization to the available genetic resources deposited in the NCBI GenBank database, and their high specificity and sensitivity were demonstrated on wheat leaf samples and selected fungal strains.

The aim of the study was to investigate specific responses of spring wheat (C3 photosynthesis) and maize (C4 photosynthesis) to drought and flooding stress. Analyses of water content, gas exchange intensity, photosynthetic apparatus activity, chlorophyll content, plant height and biological membrane integrity were performed on the 10th day of drought and flooding in both species at the third leaf stage. A specific response of wheat under both drought and flooding conditions involved an increase in ETo/RC ratio, describing electron transport flux converted into a single reaction center in PSII. Correlations between electrolyte leakage and the probability of electron transport beyond the plastoquinone QA, and the amount of energy used for the electron transport were also found. A specific response of maize during flooding was the increase of stomatal conductance. Additionally, a significant correlation between PN/Ci and relative water content was exhibited. Furthermore, the parameters differentiating the studied species only under stressful conditions were rendered. The application of such parameters can be widely used, e.g., for studying the reaction of a potential cultivars to drought and flooding. Providing such information to potential farmers can help better select cultivars for their environmental conditions.

The effectiveness of exogenously applied zearalenone (ZEN) in alleviating water deficiency stress of pea (Pisum sativum L.) and yellow lupine (Lupinus luteus L.) was analyzed in the pot experiment. ZEN was applied in the form of spraying in the flowering phase on the first day of induced drought. The effectiveness of ZEN was evaluated based on physiological (electrolyte leakage, greenness, and photosystem II activity) and biochemical (protein, proline, ascorbic acid contents, and antioxidant enzyme activity) parameters after 14 days of drought. The yield and yield quality defined as yield components, total protein, fats, sugars, and antioxidants (tocopherols and β-carotene) were measured in newly formed seeds. ZEN residue in the seeds was analyzed employing UHPLC-MS/MS to exclude its accumulation. The results showed the possibility of reducing the effects of drought stress through the use of ZEN. It was manifested by increased cell membranes stability and antioxidant enzyme activity and above all ZEN increased crop yield, compared to untreated plants. ZEN modified seed composition by inducing the accumulation of fats and antioxidants. There was no accumulation of exogenous ZEN in seeds.

The study aimed to determine the phytohormone profile of sweet briar rose (Rosa rubiginosa L.) seedlings and privileged synthesis pathways of individual hormones including gibberellins, cytokinins and auxins in response to long-term soil drought. We detected eight gibberellins, nine auxins and fifteen cytokinins. Abscisic acid (ABA) was also detected as a sensitive indicator of water stress. Thirty days of soil drought induced significant increase of ABA content and species-specific quantitative changes of other phytohormones. We established preferred synthesis pathways for three gibberellins, six auxins and eight cytokinins. Both an increase and decrease in gibberellin and cytokinin levels may modulate sweet briar's response to soil water shortage. In the case of auxins, induction of effective adaptation mechanisms to extremely dry environments is mostly triggered by their rising levels. Under drought stress, sweet briar seedlings increased their gibberellin pool at the expense of reducing the pool of cytokinins and auxins. This may indicate a specific role of gibberellins in adaptation mechanisms to long-term soil water deficit developed by sweet briar.

The resistance of microorganisms to antibiotics is a crucial problem for which the application of nanomaterials is among a growing number of solutions. The aim of the study was to create a nanocomposite (composed of graphene oxide and silver nanoparticles) with a precise mode of antibacterial action: what enables textiles to be coated in order to exhibit antibacterial properties. A characterization of nanomaterials (silver nanoparticles and graphene oxide) by size distribution, zeta potential measurements, TEM visualization and FT-IR was performed. The biological studies of the nanocomposite and its components included the toxicity effect toward two pathogenic bacteria species, namely Pseudomonas aeruginosa and Staphylococcus aureus, interaction of nanomaterials with the outer layer of microorganisms, and the generation of reactive oxygen species and lipid peroxidation. Afterwards, antibacterial studies of the nanocomposite’s coated textiles (cotton, interlining fabric, polypropylene and silk) as well as studies of the general toxicity towards a chicken embryo chorioallantoic membrane model were conducted. The toxicity of the nanocomposite used was higher than its components applied separately (zones of growth inhibition for P. aeruginosa for the final selected concentrations were as follows: silver nanoparticles 21 ± 0.7 mm, graphene oxide 14 ± 1.9 mm and nanocomposite 23 ± 1.6 mm; and for S. aureus were: silver nanoparticles 27 ± 3.8 mm, graphene oxide 14 ± 2.1 mm, and nanocomposite 28 ± 0.4 mm. The viability of P. aeruginosa and S. aureus after treatment with selected GO-Ag decreased to 27% and 31%, respectively, compared to AgNPs, when the viability of both species was 31% and 34%, accordingly). The coated textiles showed encouraging antibacterial features without general toxicity towards the chicken embryo chorioallantoic membrane model. We demonstrated that graphene oxide might constitute a functional platform for silver nanoparticles, improving the antibacterial properties of bare silver. Due to the application of the nanocomposite, the textiles showed promising antibacterial features with a low general toxicity, thereby creating a wide possibility for them to be used in practice.

Little is known about mechanisms of sweet briar adaptation to dry habitats. The species is highly invasive and displaces native plants from dry lands of the southern hemisphere. This study evaluates physiological basis of Rosa rubiginosa L. adaptation to soil drought. We performed a pot soil drought experiment and assessed water relations, water use efficiency, gas exchange and photosynthetic apparatus activity. The study also measured the content of chlorophyll, soluble carbohydrates and proline and analyzed plant biomass growth. We hypothesized that the drought stress induced an effective mechanism enabling adaptation of young sweet briar roses to soil water deficit. The study identified several adaptation mechanisms of R. rubiginosa allowing the plant to survive soil drought. These included limiting transpiration and stomatal conductance, increasing the level of soluble sugars, reducing chlorophyll content, accumulating CO2 in intercellular spaces, and increasing the quantum yield of electron transport from QA− to the PSI end electron acceptors. As a result, young sweet briar roses limited water loss and photoinhibition damage to the photosynthetic apparatus, which translated into consumption of soluble sugars for growth purposes. This study showed that photosynthesis optimization and increased activity of the photosynthetic apparatus made it possible to avoid photoinhibition and to effectively use water and sugars to maintain growth during water stress. This mechanism is probably responsible for the invasive nature of R. rubiginosa and its huge potential to displace native plant species from dry habitats of the southern hemisphere.

Drought has become an essential environmental factor limiting plant productivity. In order to detect differences in chlorophyll (Chl) a fluorescence, Chl concentration (in SPAD units), and yield parameters of yellow lupin ('Morocco 4' and 'Taper') and pea ('Wenus' and 'SZD165'), drought stress was initiated by withholding water for 14 d (25% of field water capacity). A significant decrease of leaf relative water content was found in tested cultivars. SPAD values did not show significant changes in all cultivars. The maximum photochemical efficiency and area over the Chl a fluorescence induction curve decreased in both species under drought. Performance index in lupin did not change significantly under drought while it declined in pea. Among the tested cultivars, lupin cv. 'Morocco 4' maintained the highest efficiency of the photosynthetic apparatus as well as the highest number of pods, seeds per plant, height, and dry mass of shoots under drought conditions.

Ten doubled haploid (DH) lines of winter barley with an increased range of freezing/drought tolerance were used to identify phytohormones involved in plant stress acclimation. Cold hardening and drought stress were applied at the most critical stages of plant development on young seedlings and heading plants, respectively. The level of the phytohormones was significantly higher at heading, more than 5-fold in respect of salicylic acid (SA) and total brassinosteroids (BRs) and 1.7-fold in the case of abscisic acid (ABA). Moreover, the spectrum of detectable BRs increased from one—homocastasterone (HCS)—found in seedlings to four BRs identified in heading plants [HCS, castasterone (CS), teasterone and dolicholide], with the last one detected for the first time in cereal species. To some extent freezing tolerance seems to be determined by native hormonal status as control seedlings of tolerant DH lines contained 1.4- and 2.3-fold lower amount of ABA and HCS and 2.3-fold higher amount of SA in comparison to freezing-sensitive ones. Such dependency was not observed in heading plants as significant variation in CS content was the only detected difference. Under stress treatments, tolerant DH lines accumulated significantly lower (75–81%) amount of ABA, which probably reflected lower stress intensity resulting from another defence strategy. In contrast, stress-induced significant almost 2-fold increase in HCS/CS and 2–3-fold decrease in SA content specific for tolerant DH lines of barley suggest the involvement of these molecules in freezing/drought defence. Detected correlations suggest their interaction with nonspecific peroxidase and low molecular weight antioxidants.

5-Aminolevulinic acid relieves the effects of environmental stresses in plants. Therefore, the aim of our study was to evaluate the effects of 5-aminolevulinic acid (5-ALA) on the activity of the photosynthetic apparatus in spring wheat. Other analyzed parameters involved plant height, relative turgidity, membrane status, and chlorophyll level. The plant material consisted of three genotypes of spring wheat (J × Z, R × K, K × M), subjected to mild and severe drought in the early phase of vegetative development. 5-ALA showed a positive effect on the activity of the photosynthetic apparatus under water stress. The relieving action of 5-ALA on PSII was the most evident in J × Z genotype during severe soil drought. 5-ALA positively influenced the maximum photochemical efficiency of PSII (Fv/Fm), the overall performance index of PSII photochemistry (PI) and the effective quantum field of PSII (φEo). In the same genotype, the investigated acid stimulated light energy absorption (ABS/CSm), and enhanced the amount of excitation energy trapped in PSII reaction centers (TRo/CSm) and the amount of energy used for electron transport (ETo/CSm). Moreover, 5-aminolevulinic acid showed its potential to overcome the adverse effects of water deficit on Triticum aestivum L. by increasing plant growth, relative turgidity, and chlorophyll content and reducing the degree of damage to cell membranes at the early phase of vegetative development.

Soil drought is a major problem in plant cultivation. This is particularly true for thermophilic plants, such as maize, which grow in areas often affected by precipitation shortage. The problem may be alleviated using plant growth and development stimulators. Therefore, the aim of the study was to analyze the effects of 5-aminolevulinic acid (5-ALA), zearalenone (ZEN), triacontanol (TRIA) and silicon (Si) on water management and photosynthetic activity of maize under soil drought. The experiments covered three developmental stages: three leaves, stem elongation and heading. The impact of these substances applied during drought stress depended on the plant development stage. 5-ALA affected chlorophyll levels, gas exchange and photochemical activity of PSII. Similar effects were observed for ZEN, which additionally induced stem elongation and limited dehydration. Beneficial effects of TRIA were visible at the stage of three leaves and involved leaf hydration and plant growth. A silicon preparation applied at the same developmental stage triggered similar effects and additionally induced changes in chlorophyll levels. All the stimulators significantly affected transpiration intensity at the heading stage.

The aim of this study was to investigate the accumulation of silicon in oilseed rape and to characterize the changes in chosen water balance parameters in response to drought. The following parameters were estimated: water content, osmotic and water potential, evapotranspiration, stomatal conductance and abscisic acid level under optimal and drought conditions. It was shown that oilseed rape plants accumulate silicon after its supplementation to the soil, both in the case of silicon alone and silicon together with iron. It was revealed that silicon (without iron) helps maintain constant water content under optimal conditions. While no silicon influence on osmotic regulation was observed, a transpiration decrease was detected under optimal conditions after silicon application. Under drought, a reduction in stomatal conductance was observed, but it was similar for all plants. The decrease in leaf water content under drought was accompanied by a significant increase in abscisic acid content in leaves of control plants and those treated with silicon together with iron. To sum up, under certain conditions, silicon is accumulated even in non-accumulator species, such as oilseed rape, and presumably improves water uptake under drought stress.

Climate changes, which result in the occurrence of periods with relatively high temperatures during the winter, can lead to the deacclimation of cold-hardened plants and cause problems with their winter survival. The aim of these studies was to investigate the physiological changes (photosynthesis and water relations including aquaporin expression) that accompany deacclimation process in the economically important winter oilseed rape plants. The effect of deacclimation on frost tolerance was also estimated for two tested cultivars: semi-dwarf and plants of a normal height. After cold acclimation at 4 &deg, C (compared to the unacclimated control) the typical changes that accompany cold acclimation such as an increase in the content of water-soluble sugars or a lower water content in the leaves, which lead to an increased frost tolerance, were observed. Deacclimation partially or completely reversed these changes, which resulted in a decreased frost tolerance that was accompanied by a decrease in the content of sugars and an increase of the osmotic potential. The chemical composition of the leaves, which was measured using FT-Raman spectroscopy also clearly confirmed the metabolic differences between the cold-acclimated and deacclimated plants. The plants were significantly different in regard to the content of the various pigments as well as fatty acids and polysaccharides. The phenomenon of a deacclimation-induced decrease in aquaporin PIP1 accumulation, which was accompanied by unchanged PIP1 transcript accumulation, will be discussed in the aspects of the water relations and decreased frost tolerance in deacclimated plants.

This review synthesizes knowledge on epigenetic regulation of leaf senescence and discusses the possibility of using this knowledge to improve crop quality. This control level is implemented by different but interacting epigenetic mechanisms, including DNA methylation, covalent histone modifications, and non-covalent chromatin remodeling. The genetic and epigenetic changes may act alone or together and regulate the gene expression, which may result in heritable (stress memory) changes and may lead to crop survival. In the review, the question also arises whether the mitotically stable epigenetic information can be used for crop improvement. The barley crop model for early and late events of dark-induced leaf senescence (DILS), where the point of no return was defined, revealed differences in DNA and RNA modifications active in DILS compared to developmental leaf senescence. This suggests the possibility of a yet-to-be-discovered epigenetic-based switch between cell survival and cell death. Conclusions from the analyzed research contributed to the hypothesis that chromatin-remodeling mechanisms play a role in the control of induced leaf senescence. Understanding this mechanism in crops might provide a tool for further exploitation toward sustainable agriculture: so-called epibreeding.

Plant tolerance to environmental stress is determined by a very complicated network composed of many intra- and extracellular factors. The aim of this study was to select candidate genes involved in responses to freezing and drought in barley on the basis of previous proteomic studies and to analyze changes in their expression caused by application of both stress factors. Six candidate genes for freezing tolerance (namely the genes encoding elongation factor 1 alpha (EF1A), ferredoxin-NADP reductase, a 14-3-3a protein, &beta, fructofuranosidase, CBF2A and CBF4B) and six for drought tolerance (encoding transketolase, periplasmic serine protease, triosephosphate isomerase, a protein with a co-chaperon region (GroEs), pfam14200 and actin) were chosen arbitrarily on the basis of in silico bioinformatic analyses. The expression levels of these genes were measured under control and stress conditions in six DH (doubled haploid) lines with differing freezing and drought tolerance. The results of gene expression analysis confirmed the roles of the candidate genes preselected in this study on the basis of previous proteome analysis in contributing to the differences in freezing and drought tolerance observed in the studied population of DH lines of winter barley.

The aim of the study was to determine molecular basis of plant photosynthetic activity during soil drought and under rapid (unfavorable) and gradual (favorable) rehydration. We analyzed the content of proteins associated with the photosynthetic apparatus and photosynthetic fixation of CO2. The experiment involved two genotypes of triticale showing complete and incomplete recovery from drought stress. The reason for irreversible changes in the activity of the photosynthetic apparatus was stress-induced decrease in the content of Rieske protein (PetC) of cytochrome complex b6f. Intensification of metabolic processes during rehydration and following overload the electron transport chain led to electron transfer from primary acceptors QA/QB in PSII and ferredoxin in PSI to oxygen. The resulting reactive oxygen species oxidized proteins and inhibited synthesis of those controlling the photosynthetic apparatus and carbon fixation. The consequence was a reduction of quantum yield of electron transport to their final acceptors in PSI and lowering of plant photosynthetic activity and biomass. Simultaneous utilization of H2O2 during saturation of cell wall with phenolic compounds ensured restoration of desired level of proteins controlling the photosynthetic apparatus and photosynthetic carbon fixation, as well as high photosynthetic activity.

Modern anthropological research includes very sophisticated diagnostic methods. They allow us to obtain information that has not been available so far. The aim of this paper is to analyze, using current microscopic technologies, the Mesolithic dental material of one adult individual from Woźna Wieś (Poland). The present case study will focus on the analysis of enamel hypoplasia. A scanning electron microscope (SEM) was used to count the number of perikymata building on the hypoplastic line. Linear enamel hypoplasia (LEH) was diagnosed only on the right mandibular canine. The time of occurrence of environmental disturbance was estimated between about 4.2 and 4.9 years of age. The occlusal wall built the enamel hypoplasia with no more than three to four perikymata, meaning that the physiological stress had to have occurred over a fairly short period of time (about 30–40 days).

The alimentary tract of oxudercine gobies is characterized by a lack of an anatomically distinct stomach, owing to which they are classified as stomachless. Since the environment, food requirements, and feeding habits have a significant impact on the anatomy of the alimentary tract of fish, it was assumed that predominantly carnivorous, semi-terrestrial mudskippers would have a stomach. In order to verify this hypothesis, anatomical, histological, histochemical and ultrastructural analysis of the alimentary tract of the Atlantic mudskipper Periophthalmus barbarus was performed. The results revealed that despite a lack of clear anatomical distinction within the alimentary tract, there were four well-distinguished sections visible at the histological level: oesophagus, stomach, intestine, and rectum. The division was enhanced by the presence of a pyloric sphincter and an ileorectal valve. The stomach contained tubular glands composed of oxynticopeptic cells. Gland cells had pepsinogen granules and a well-developed tubulovesicular network of smooth membranes, which indicates the secretion of gastric juice. The presence of neutral mucus in the apical region of surface epithelial cells as protection against hydrochloric acid as well as the presence of active pepsin also confirm gastric function. However, low pepsin activity seems to implies low protein digestion. The results of this study indicate that the Atlantic mudskipper P. barbarus has a functional stomach.

We developed new PCR assays that target beta-tubulin (TUB2) and 14 alpha-demethylase (CYP51) genes and used them for the species-specific detection of Blumeria graminis f. sp. tritici (Bgt). Based on fungi DNA sequences available in the NCBI (National Center for Biotechnology Information) GenBank database we developed simplex and duplex PCR assays. The specificities of the primer sets were evaluated using environmental samples of wheat leaves collected during the 2015/2016 growing season across Poland. Primer sets LidBg17/18 and LidBg21/22 strongly amplified fragments of the expected length for all 67 tested samples. Primer specificity was confirmed using field samples of Zymoseptoria tri­tici, Puccinia triticina (syn. P. recondita f. sp. tritici), P. striiformis f. sp. tritici, and Pyrenophora tritici-repentis.

Cell wall-bound phenolics (CWP) play an important role in the mechanisms of plant acclimation to soil drought. The study involved CWP analyses in 50 strains and 50 doubled haploid (DH) lines of winter triticale exposed to drought at their vegetative and generative stages. CWP in the plants experiencing drought at the generative stage positively correlated with their leaf water contents. The strains and DH lines characterized by high content of CWP showed higher leaf water content and higher activity of photosynthetic apparatus when exposed to drought at the generative stage compared to the strains and DH lines with the low CWP content. Furthermore, when drought subsided at the generative stage, the strains and DH lines richer in CWP demonstrated higher regeneration potential and their grain yield loss was smaller.

The aim of the study was to analyze the activity of the photosynthetic apparatus during induced dehydration of leaves harvested from perennial shrubs of Rosa rubiginosa growing at wet and dry sites and differing in the level of drought memory. The markers of drought memory included accumulation of D1 (PsbA) protein, chaperone Cpn60, Rubisco activase (RA), and Rubisco larger subunit (RbcL). The study investigated the levels of soluble carbohydrates and phenolic compounds, of cell wall-bound phenolics, non-enzymatic antioxidants, and assimilation pigments. During induced dehydration, leaves from the dry site exhibited slower water loss and greater activity of the photosynthetic apparatus, particularly in terms of effectiveness of electron transport between PSII and PSI. They also effectively dissipated excessive excitation energy in the form of heat and far-red fluorescence. Higher level of D1 protein in the leaves collected from the dry site was accompanied by lower accumulation of Cpn60, RA and RbcL proteins and considerably higher content of soluble carbohydrates and carotenoids. Drought memory in R. rubiginosa may be associated with a low level of proteins responsible for photosynthetic fixation of CO2 and reduced content of chlorophyll a and b, and also with enhanced accumulation of D1 protein and soluble carbohydrates and carotenoids.

The species Puccinia triticina (Pt) and Puccinia striiformis f. sp. tritici (Pst) are devastating cereal pathogens that cause leaf and stripe rust diseases. We developed PCR assays for the species-specific detection of Pt and Pst, 2 biological agents that cause wheat rust disease. For each pathogen, we validated 3 primer sets that target the second largest subunits of the RNA polymerase II (rpb2) and β-tubulin 1 (tub1) genes. The specificities of the primers were verified using naturally infected plant materials with visual symptoms of disease. All primer sets amplified a single DNA fragment of the expected length. The primer sets LidPr15/16, LidPr1/2, and LidPs13/14 were able to detect small amounts of pure fungal DNA with sensitivities of 0.1, 1, and 10 pg/μL, respectively. A sufficient detection limit (1 pg/μL to 5 ng/μL) was observed for all assays when the sensitivity test was performed with host plant DNA. The study also evaluated the simultaneous detection of both rust pathogens, and the multiplex PCR assay generated amplicons of 240 and 144 bp in length for Pts (LidPs9/10) and Pt (LidPr1/2), respectively.

The present study aimed at identifying the regions of triticale genome responsible for cell wall saturation with phenolic compounds under drought stress during vegetative and generative growth. Moreover, the loci determining the activity of the photosynthetic apparatus, leaf water content (LWC) and osmotic potential (Ψ o) were identified, as leaf hydration and functioning of the photosynthetic apparatus under drought are associated with the content of cell wall-bound phenolics (CWPh). Compared with LWC and Ψ o, CWPh fluctuations were more strongly associated with changes in chlorophyll fluorescence. At the vegetative stage, CWPh fluctuations were due to the activity of three loci, of which only QCWPh.4B was also related to changes in F v/F m and ABS/CSm. In the other QTLs (QCWPh.6R.2 and QCWPh.6R.3), the genes of these loci determined also the changes in majority of chlorophyll fluorescence parameters. At the generative stage, the changes in CWPh in loci QCWPh.4B, QCWPh.3R and QCWPh.6R.1 corresponded to those in DIo/CSm. The locus QCWPh.6R.3, active at V stage, controlled majority of chlorophyll fluorescence parameters. This is the first study on mapping quantitative traits in triticale plants exposed to drought at different stages of development, and the first to present the loci for cell wall-bound phenolics.

Carbohydrates are used in plant growth processes, osmotic regulation and secondary metabolism. A study of the allocation of carbohydrates to a target set of metabolites during triticale acclimation to soil drought was performed. The study included a semi-dwarf cultivar 'Woltario' and a long-stemmed cultivar 'Moderato', differing in the activity of the photosynthetic apparatus under optimum growth conditions. Differences were found in the quantitative and qualitative composition of individual carbohydrates and phenolic compounds, depending on the developmental stage and water availability. Soluble carbohydrates in the semi-dwarf 'Woltario' cv. under soil drought were utilized for synthesis of starch, soluble phenolic compounds and an accumulation of cell wall carbohydrates. In the typical 'Moderato' cv., soluble carbohydrates were primarily used for the synthesis of phenolic compounds that were then incorporated into cell wall structures. Increased content of cell wall-bound phenolics in 'Moderato' cv. improved the cell wall tightness and reduced the rate of leaf water loss. In 'Woltario' cv., the increase in cell osmotic potential due to an enhanced concentration of carbohydrates and proline was insufficient to slow down the rate of leaf water loss. The mechanism of cell wall tightening in response to leaf desiccation may be the main key in the process of triticale acclimation to soil drought.

Ectromelia virus (ECTV, the causative agent of mousepox), which represents the same genus as variola virus (VARV, the agent responsible for smallpox in humans), has served for years as a model virus for studying mechanisms of poxvirus-induced disease. Despite increasing knowledge on the interaction between ECTV and its natural host-the mouse-surprisingly, still little is known about the cell biology of ECTV infection. Because pathogen interaction with the cytoskeleton is still a growing area of research in the virus-host cell interplay, the aim of the present study was to evaluate the consequences of ECTV infection on the cytoskeleton in a murine fibroblast cell line. The viral effect on the cytoskeleton was reflected by changes in migration of the cells and rearrangement of the architecture of tubulin, vimentin, and actin filaments. The virus-induced cytoskeletal rearrangements observed in these studies contributed to the efficient cell-to-cell spread of infection, which is an important feature of ECTV virulence. Additionally, during later stages of infection L929 cells produced two main types of actin-based cellular protrusions: short (actin tails and "dendrites") and long (cytoplasmic corridors). Due to diversity of filopodial extensions induced by the virus, we suggest that ECTV represents a valuable new model for studying processes and pathways that regulate the formation of cytoskeleton-based cellular structures. © 2016 Wiley Periodicals, Inc.

The reasons for partial recovery after a soil drought are not fully understood and have not been studied so far. This study investigated the physiological and biochemical responses of triticale cultivars with differential recovery ability after soil water deficit. Activity of the photosynthetic apparatus under soil drought followed by rehydration was estimated. Plant antioxidant potential was determined based on the measurement of catalase and peroxidase activity. The levels of hydrogen peroxide and superoxide radical were assessed. Under rehydration, the not fully-recovered cultivar experienced further significant increase in the content of H2O2 and inhibited activity of the photosynthetic apparatus, as compared to the drought period. On 42nd day of the rehydration, the not fully-recovered cultivar showed also a reduced photosynthetic activity in the flag leaves, which resulted in a significant decrease in its grain yield. The first week of a rapid rehydration involved a decrease in total peroxidase and catalase activities. The increased content of H2O2 was compensated only when leaf water content was gradually restored in the first week of the rehydration and no further decrease in the activity of the photosynthetic apparatus was noticed. A destructive effect of the rapid rehydration was manifested in an intensification of the physiological processes associated with reactive oxygen species (ROS) overproduction. An important cause of hydrogen peroxide overproduction seems to be the electron leakage due to overloading of the electron transport chain (ETC) in the PSI and PSII.

Rdza koronowa, powodowana przez grzyb Puccinia coronata, jest jedną z najpowszechniej występujących chorób grzybowych owsa w Polsce. Do tej pory u owsa zidentyfikowano ponad 100 genów odporności na ten patogen, jednak są one szybko przełamywane przez nowe patotypy Puccinia coronata. Celem pracy była ocena efektywności genów warunkujących odporność na izolaty Puccinia coronata skolekcjonowane w centralnej i południowo-wschodniej Polsce w latach 2010-2011. W badaniach wykorzystano 31 linii referencyjnych owsa z pojedynczymi genami odporności na rdzę koronową. Analizy przeprowadzono z wykorzystaniem testów żywiciel-patogen w oparciu o 6 izolatów Puccinia coronata. Izolaty pochodzące z różnych rejonów uprawy owsa różniły się między sobą wirulencją w stosunku do 21 z 31 linii referencyjnych. Przeprowadzone badania wykazały, że najbardziej efektywne w stadium siewki są geny Pc14, Pc39, Pc51, Pc52, Pc53, Pc57, Pc60, Pc68, Pc71 i Pc91.

We developed new PCR assays that target beta-tubulin (TUB2) and 14 alpha-demethylase (CYP51) genes and used them for the species-specific detection of Blumeria graminis f. sp. tritici (Bgt). Based on fungi DNA sequences available in the NCBI (National Center for Biotechnology Information) GenBank database we developed simplex and duplex PCR assays. The specificities of the primer sets were evaluated using environmental samples of wheat leaves collected during the 2015/2016 growing season across Poland. Primer sets LidBg17/18 and LidBg21/22 strongly amplified fragments of the expected length for all 67 tested samples. Primer specificity was confirmed using field samples of Zymoseptoria tri-tici, Puccinia triticina (syn. P. recondita f. sp. tritici), P. striiformis f. sp. tritici, and Pyrenophora tritici-repentis.

Acquiring competences in the area of physical culture depends on the intention of physical activity, as well as on the level of physical fitness. It is important to learn the factors determining physical fitness; therefore the purpose of this research was to determine the relationship between the stages of readiness for regular physical activity and the level of physical fitness of children from Poland and Turkey. The research included pupils aged 14 living in the Raciborz area in Poland and in the area of the city of Balikesir in Turkey. Readiness for regular physical activity was assessed using a special algorithm in accordance with the concept of the transtheoretical model (TM). In this research the interpretation of TM was accepted for the assessment of intentional behavior. To determine the level of physical fitness, the European Fitness Test - Eurofit was used. Statistical analyses showed statistically significant differences. Of the 8 tests carried out, in six - a larger level was obtained by respondents from Poland and in five of them these were statistically significant differences. In 2 of the tests carried out, the young people from Turkey reached an average higher level. The correlation coefficient between the stages of readiness to undertake physical activity and physical fitness tests was calculated to achieve the main objective. The vast majority were low dependencies. On the basis of the conducted research, it was found that the studied group of Polish youth was characterized by a higher level of physical fitness than their peers in Turkey. There was no significant relationship between the stages of readiness to undertake physical activity and the results of fitness tests. The need for further research was concluded.
