Abstract

The number of patients affected by neurodegenerative diseases is increasing worldwide, and no effective treatments have been developed yet. Although precision medicine could represent a powerful tool, it remains a challenge due to the high variability among patients. To identify molecules acting with innovative mechanisms of action, we performed a computational investigation using SAFAN technology, focusing specifically on HuD. This target belongs to the human embryonic lethal abnormal visual-like (ELAV) proteins and plays a key role in neuronal plasticity and differentiation. The results highlighted that the molecule able to bind the selected target was (R)-aloesaponol-III-8-methyl ether [(R)-ASME], a metabolite extracted from Eremurus persicus. Notably, this molecule is a TNF-alpha inhibitor, a cytokine involved in neuroinflammation. To obtain a suitable amount of (R)-ASME to confirm its activity on HuD, we optimized the extraction procedure. Together with ASME, another related metabolite, germichrysone, was isolated. Both ASME and germichrysone underwent biological investigation, but only ASME confirmed its ability to bind HuD. Given the multifactorial nature of neurodegenerative diseases, we decided to investigate ASME as a proteasome activator, being molecules endowed with this kind of activity potentially able to counteract aggregations of dysregulated proteins. ASME was able to activate the considered target both in enzymatic and cellular assays. Therefore, ASME may be considered a promising hit in the fight against neurodegenerative diseases.

Abstract

Introduction: Hereditary spastic paraplegia is a clinically and genetically heterogeneous neurological entity that includes more than 80 disorders which share lower limb spasticity as a common feature. Abnormalities in multiple cellular processes are implicated in their pathogenesis, including lipid metabolism; but still 40% of the patients are undiagnosed. Our goal was to identify the disease-causing variants in Sudanese families excluded for known genetic causes and describe a novel clinico-genetic entity.Methods: We studied four patients from two unrelated consanguineous Sudanese families who manifested a neurological phenotype characterized by spasticity, psychomotor developmental delay and/or regression, and intellectual impairment. We applied next-generation sequencing, bioinformatics analysis, and Sanger sequencing to identify the genetic culprit. We then explored the consequences of the identified variants in patients-derived fibroblasts using targeted-lipidomics strategies.Results and Discussion: Two homozygous variants in ABHD16A segregated with the disease in the two studied families. ABHD16A encodes the main brain phosphatidylserine hydrolase. In vitro, we confirmed that ABHD16A loss of function reduces the levels of certain long-chain lysophosphatidylserine species while increases the levels of multiple phosphatidylserine species in patient's fibroblasts.Conclusion:ABHD16A loss of function is implicated in the pathogenesis of a novel form of complex hereditary spastic paraplegia.

Abstract

To overcome the increasing price and various operational issues, the 'Benefit-Sharing Model' can be an ideal solution as it improves resource optimization and maximizes cost efficiency. In particular, a Japan-Bangladesh joint venture medical device manufacturing company had been making loss since its inception. It reported an annual net profit for the first time after the introduction of the 'Benefit-Sharing Model' in the supply chain operations of the organization. Price fluctuations, limited supplier sources, below-par planning process and less supplier collaboration were resulting in continuous business losses which the company needed to solve to ensure profitability. Here we discuss the utilization of a strategic model to improve process parameters and resource optimization with ideal EOQ which was adjusted with supplier MOQ ensuring perpetual shipments. Furthermore, shipment optimization to achieve cost effectiveness, research opportunities for slow moving material and Covid impact minimization are reported in this paper. Utilizing this overall approach, raw-material price reduced 6% and a smooth order trend was achieved which reduced 41% inventory holding cost, 24% lead time, 67% port demurrage cost and increased capacity utilization by 25%. In addition, R&D development to utilize slow-moving quantity reduced purchase by 7% and new source development reduced cost 45%. Moreover, during Covid period, future risk assessment and strategic business negotiation helped to secure business from high freight and price hike up to 9 months and increased order by 20%. 10. The effects of Bone Morphogenetic Protein 4 on adult neural stem cell proliferation, differentiation and survival in an in vitro model of ischemic stroke

Abstract

The subventricular zone (SVZ) of the lateral ventricles represents a main region where neural stem cells (NSCs) of the mature central nervous system (CNS) reside. Bone Morphogenetic Proteins (BMPs) are the largest subclass of the transforming growth factor-β (TGF-β) superfamily of ligands. BMP4 is one such member and plays important roles in adult NSC differentiation. However, the exact effects of BMP4 on SVZ adult NSCs in CNS ischemia are still unknown. Using oxygen and glucose deprivation (OGD) as an in vitro model of ischemia, we examined the behavior of adult NSCs. We observed that anoxia resulted in reduced viability of adult NSCs, and that BMP4 treatment clearly rescued apoptotic cell death following anoxia. Furthermore, BMP4 treatment exhibited a strong inhibitory effect on cellular proliferation of the adult NSCs in normoxic conditions. Moreover, such inhibitory effects of BMP4 treatment were also found in OGD conditions, despite the enhanced cellular proliferation of the adult NSCs that was observed under such ischemic conditions. Increased neuronal and astroglial commitment of adult NSCs were found in the OGD conditions, whereas a reduction in differentiated neurons and an increase in differentiated astrocytes were observed following BMP4 treatment. The present data indicate that BMP4 modulates proliferation and differentiation of SVZ-derived adult NSCs and promotes cell survival in the in vitro model of ischemic stroke.

Abstract

Hereditary spinocerebellar degenerations (SCDs) is an umbrella term that covers a group of monogenic conditions that share common pathogenic mechanisms and include hereditary spastic paraplegia (HSP), cerebellar ataxia, and spinocerebellar ataxia. They are often complicated with axonal neuropathy and/or intellectual impairment and overlap with many neurological conditions, including neurodevelopmental disorders. More than 200 genes and loci inherited through all modes of Mendelian inheritance are known. Autosomal recessive inheritance predominates in consanguineous communities; however, autosomal dominant and X-linked inheritance can also occur. Sudan is inhabited by genetically diverse populations, yet it has high consanguinity rates. We used next-generation sequencing, genotyping, bioinformatics analysis, and candidate gene approaches to study 90 affected patients from 38 unrelated Sudanese families segregating multiple forms of SCDs. The age-at-onset in our cohort ranged from birth to 35 years; however, most patients manifested childhood-onset diseases (the mean and median ages at onset were 7.5 and 3 years, respectively). We reached the genetic diagnosis in 63% and possibly up to 73% of the studied families when considering variants of unknown significance. Combining the present data with our previous analysis of 25 Sudanese HSP families, the success rate reached 52–59% (31–35/59 families). In this article we report candidate variants in genes previously known to be associated with SCDs or other phenotypically related monogenic disorders. We also highlight the genetic and clinical heterogeneity of SCDs in Sudan, as we did not identify a major causative gene in our cohort, and the potential for discovering novel SCD genes in this population.

Abstract

Introduction Increased mortality has been demonstrated in older adults with coronavirus disease 2019 (COVID-19), but the effect of frailty has been unclear. Methods This multi-centre cohort study involved patients aged 18 years and older hospitalised with COVID-19, using routinely collected data. We used Cox regression analysis to assess the impact of age, frailty and delirium on the risk of inpatient mortality, adjusting for sex, illness severity, inflammation and co-morbidities. We used ordinal logistic regression analysis to assess the impact of age, Clinical Frailty Scale (CFS) and delirium on risk of increased care requirements on discharge, adjusting for the same variables. Results Data from 5,711 patients from 55 hospitals in 12 countries were included (median age 74, interquartile range [IQR] 54–83; 55.2% male). The risk of death increased independently with increasing age (>80 versus 18–49: hazard ratio [HR] 3.57, confidence interval [CI] 2.54–5.02), frailty (CFS 8 versus 1–3: HR 3.03, CI 2.29–4.00) inflammation, renal disease, cardiovascular disease and cancer, but not delirium. Age, frailty (CFS 7 versus 1–3: odds ratio 7.00, CI 5.27–9.32), delirium, dementia and mental health diagnoses were all associated with increased risk of higher care needs on discharge. The likelihood of adverse outcomes increased across all grades of CFS from 4 to 9. Conclusion Age and frailty are independently associated with adverse outcomes in COVID-19. Risk of increased care needs was also increased in survivors of COVID-19 with frailty or older age.

The widespread health impacts on millions of people from consumption of arsenic (As) contaminated groundwater of the Ganges delta necessitate an effort to locate As-safe aquifer zones and to develop a sustainable aquifer management policy. We look into the aquifer geometries, sediment characteristics and groundwater As concentrations to develop an understanding of the factors that inhibit As invasion/mobilization within the As-safe aquifers. However, the sustainability of the present-day safe aquifer is at risk due to the heavy groundwater pumping for irrigation and thus, promoting water conserving agricultural practices is extremely vital. We suggest that, formulating a sustainable As mitigation plan should also involve the non-scientific communities such as the local tubewell drillers and farmers for widespread implementation of the plan., QC 20211020

Despite many efforts over the last 25 years, millions of people are still drinking groundwater having arsenic above Bangladesh and World Health Organization limits. This can be linked, to some extent, to a lack of pragmatic policies and implementation strategies. Important lessons have been learned from extensive studies in Araihazar and Matlab in Bangladesh. Also better mitigation strategies for reducing arsenic exposure have been demonstrated at both locations. New policy and mitigation strategies should focus on mapping using mobile technology for village scale mapping. Dissemination of test result is extremely important in increasing awareness of the users and a new three-color scheme (Blue, Green, Red corresponding to <10, 10–50, and >50 µg L−1) instead of existing Red-Green color scheme should be adopted to lower arsenic intake. Abstract

The WASH Sector Development Plan (2011–2025) recognizes the absence of harmonized approaches as the root challenge to scaling up drinking water safety in Bangladesh. UNICEF is supporting the Government of Bangladesh (GoB) through the Department of Public Health Engineering (DPHE) and Policy Support Branch to re-engineer its approach to drinking water safety by integrating policy, systems strengthening & sustainable services delivery at national and sub-national levels. Some of the key contributions to the sector include the implementation of the UNICEF-DPHE Arsenic Safe Union model with the declaration of 106 Arsenic Safe Villages, reduction in arsenic contamination rates of new tubewells, a system for preventing elite capture of water points, and the adoption of the ASU model in a $240 million arsenic mitigation drinking water project using domestic resources. 40. Arsenic and manganese in shallow tubewells : validation of platform color as a screening tool in Bangladesh

Abstract

This study aimed to evaluate the potential of handpump tubewell platform color as a low-cost, quick and convenient screening tool for As and Mn in drinking water tubewells. For this study, groundwater samples and corresponding tubewell platform pictures were collected from 272 shallow tubewells in Matlab Upazila of South-Eastern Bangladesh. The result shows that arsenic concentration within the surveyed (n = 272) tubewells, 99% (n = 269) exceeded the World Health Organization (WHO) guideline value of 10 µg/L, and 98% (n = 267) exceeded the Bangladesh drinking water standards (BDWS) of 50 µg/L. In relation to the platform color concept, within 233 (total 272) red colored platform tubewells, 230 (99%) exceeded the WHO guideline value of 10 µg/L, and 229 (98%) tubewells exceeded BDWS of 50 µg/L. This result shows a strong correlation between the development of red color stain on tubewell platform and As concentrations in the corresponding tubewell water. This study suggests that red-colored platform can be used for primary identification of tubewells with an elevated level of As and thus could prioritize sustainable As mitigation management in developing countries where water comes from reductive shallow aquifers. This study did not confirm the potential for Mn screening, as red discoloration by Fe oxides was found to mask the black discoloration of Mn oxides. Deep Convolutional Neural Networks (CNNs) have recently demonstrated the state-of-the-art classification performance on ImageNet Large Scale Visual Recognition Challenge (ILSVRC) since 2012, yet there is relatively no clear understanding of the reasons behind their outstanding performance, or how they might be improved, In this paper we present a novel benchmarking of multiple state-of-the-art deep CNN architectures by providing an analysis of important performance metrics: speed, memory consumption, and network parameters utilization. Key findings are: (1) fully connected layers are of a high cost on speed and memory consumption compared to the sparsely connected layers; (2) the depth and the performance of an architecture are in a nonlinear relationship and constrained by layers transformation types; (3) 1×1 convolutions are an efficient way to reduce dimensionality and pooling features; (4) addition units as in residual and densely connected networks accelerate the backpropagation time by distributing the gradients through the graph, we believe our set of benchmarks are a step towards better realization of the best architectural design choices of Deep CNNs.

Abstract

Groundwater-level fluctuations represent hydraulic responses to changes in groundwater storage due to aquifer recharge and drainage as well as to changes in stress that include water mass loading and unloading above the aquifer surface. The latter ‘poroelastic’ response of confined aquifers is a well-established phenomenon which has been demonstrated in diverse hydrogeological environments but is frequently ignored in assessments of groundwater resources. Here we present high-frequency groundwater measurements over a twelve-month period from the tropical, fluvio-deltaic Bengal Aquifer System (BAS), the largest aquifer in south Asia. The groundwater level fluctuations are dominated by the aquifer poroelastic response to changes in terrestrial water loading by processes acting over periods ranging from hours to months; the effects of groundwater flow are subordinate. Our measurements represent the first direct, quantitative identification of loading effects on groundwater levels in the BAS. Our analysis highlights the potential limitations of hydrogeological analyses which ignore loading effects in this environment. We also demonstrate the potential for employing poroelastic responses in the BAS and across other tropical fluvio-deltaic regions as a direct, in-situ measure of changes in terrestrial water storage to complement analyses from the Gravity and Climate Experiment (GRACE) mission but at much higher resolution.

Abstract

Pollen morphology of ten Enkianthus species was examined using light (LM) and scanning electron microscopy (SEM) and previous infrageneric classification of Enkianthus was discussed in the light of the palynological characters obtained in this study. The pollen grains are monads, oblate spheroidal to prolate in shape and 3‐ to 5‐colpor(oid)ate. Exine sculpture varies from granulate to coarsely rugulate‐psilate with faint to distinct minute granules in SEM. Infraspecific geographical differences in palynological characters were found in E. deflexus. The species of Enkianthus are clearly differentiated into two pollen morphological groups that correspond to the sections Enkiantella + Meisteria and Andromedina + Enkianthus, respectively. The members of the sections Enkiantella and Meisteria have pollen grains typically with three apertures, prolate spheroidal to prolate, ratio of colpus length to polar axis (L/P) 0.69–0.84, relatively thinner exine and exine sculpturing tendency to granulate, character state...

Abstract

The aim of the present study was to investigate the effects of xenoestrogens, like nonylphenol (NP) on growth performance and gonads development of male Nile tilapia. Fish were randomly distributed into four groups in 16 fiberglass tanks with carrying capacity of 15 fish/tank and four replicates. Fish were exposed to different concentrations of NP. The contaminations of NP were 0, 25, 50 and 100 μgL-1 administrated for 126 days. After exposure period, fish weight and survival were measured. Gonads and liver were dissected for calculating both Gonado-Somatic Index (GSI) and Hepato- Somatic Index (HSI) and processed for histological examination. At the end of the exposure period in all treated groups, the average survival rate was significantly lower than that of the control. The average body weights were ranged between 73.41 ± 1.37g - 78.14 ± 6.86g for the control and 50 μg NP L-1, respectively with no statistical significant differences. Results showed slight changes in liver weight among treated group compared to the control. A significant (P<0.05) reduction in gonads weight and GSI occurred in exposed fish compared to the control. Testicular sections from the control group appeared fully mature where, lumens and ducts were loaded with spermatozoa. However, testicular sections from NP exposed fish showed germinal epithelium degeneration, which correlated with exposure level of NP and reflected on low weight of the gonads. The presence of oocytes within the testicular tissue was pronounced, especially in high dose exposed fish. Therefore, severity of testis-ova was mild in 25 μg NP L-1, moderate to pronounced in 50 and 100 μg NP L-1 exposed groups, respectively. From the obtained results, it could be concluded that the environmental pollutants with estrogenic activity such as nonylphenol can alter the development of gonads and disrupt reproduction of wild and farmed fish. Nearly 90% of the estimated 10 million tubewells in Bangladesh are installed privately by local drillers for rural drinking water supplies in Bangladesh. The awareness of local drillers on elevated Arsenic (As) concentrations in tubewell water at shallow depths have made them change their practice of installation of tubewells. Using the visual color attributes of the shallow sediments (<100 m) and content of dissolved iron, generally associated with high As concentrations, the local drillers presently install community tubewells at depths targeting red/brownish or off-white sediments. This study recognizes the local tubewell drillers as important stakeholder in the business of tubewell installation. Abstract

Background Epidemiological data about congenital rubella syndrome (CRS) are scarce and rubella vaccine is not yet included in the childhood immunization schedule in Sudan. This study aimed to identify and describe CRS cases among Sudanese infants with congenital eye or heart defects. Methods Between February and September 2010, paired oral fluid and dried blood spot samples were collected from 98 infants aged up to 12 months. These infants were enrolled during their visits to five hospitals in Khartoum, Sudan. Clinical samples were screened for rubella IgM and for ≥ 6 months old infants also for IgG antibodies by ELISA. The oral fluid of IgM and/or IgG positive patients was tested for rubella RNA by reverse transcriptase PCR. Results Our findings revealed that two children (2.0%) were IgM positive and another five children (5.1%) were positive for IgG antibodies. None of the five infants of which enough oral fluid was available for RNA investigation was PCR positive. Conclusions This study documented the presence of CRS in Sudan and highlighted the importance of rubella vaccine introduction for preventing future CRS cases in the country.
