Fish feed quality is the main determinant of fish flesh quality, so it is important for successful aquaculture. The current study determines the concentration of heavy metals in fish feeds (A and B), water, and their bioaccumulation in gills, liver, and muscle of C. carpio cultured in different environments (biofloc technology and earthen pond systems). In addition, the correlation between heavy metals in fish feeds with bioaccumulated metals in fish tissues was also determined. Results revealed that most heavy metal concentration was significantly greater (P 0.05) in feed B than in feed A but in permissible range, while all the heavy metal concentration was notably higher in earthen ponds than in biofloc technology. Result from the bioaccumulation factor and concentration of the metals showed that heavy metals were highly accumulated in the fish liver followed by gills. The metal concentration in fish feeds and fish edible parts (muscle) was lower than the WHO standard level; however, the amount of Pb was higher in the fish muscle, liver, and gills, which is harmful for human consumption and also for fish health. Though the correlation test revealed that all of the metals from the feeds were positively correlated to the metals in fish tissues, but most of the estimated correlation was significant and linearly correlated. It can be concluded that producers must measure feed quality correctly to avoid heavy metal contamination because it may assimilate and accumulate in the food chain.

For successful aquaculture, the primary need is the quality of fish feed, which determines fish flesh quality. The current study was conducted to evaluate the number and concentration of heavy metals in commonly used fish feeds and fish gills, liver, and muscle of biofloc technology and earthen pond systems. Besides this, the correlation between heavy metals in fish feeds with detected metals in the gills, liver, and muscle of fish was also determined. Results revealed that heavy metals concentration, including Cu and Cd, in feed B was significantly greater than in feed A, but the Zn level in feed A was significantly higher than in feed B. Furthermore, the concentration of heavy metals in fish of both aquaculture systems was significantly higher in the liver than in the gills and muscle. From the current study, it can be concluded that the fish feed producers need to measure feed quality adequately to avoid hazardous contamination by heavy metals in the feed. The ultimate consumer, fish and humans, may, otherwise, be predisposed to assimilate and accumulate these heavy metals.

The current study aims to determine the effect of four different anesthetics such as MS-222 (100 mg/L), tobacco extract (50 mg/L), propiscin (1.0 ml/L) and clove oil (40 mg/L) on the hematology and blood biochemical parameters of Labeo rohita. Blood was sampled 10 minutes after the anesthesia was administered and again after 24 hours. Except for the clove oil 24h measurements, there was a significant difference in the number of blood cells in all anesthetic applications (P0.05) with tobacco, Change has been detected in ALP with MS-222 and tobacco calcium and magnesium showed non-significant differences with anesthetics. Overall all the parameters were affected by anesthetics used in the current research, however, it was established that 24 hours following the administration of clove oil, the discrepancies in blood values had vanished to a great extent.

Coal miners are continuously exposed to coal mine dust and airborne particulate that act as a potential risk to their health. The present study evaluates the DNA damage in coal miners using the Buccal Micronucleus Cytome (BMCyt) assay. The samples of the blood and buccal epithelial cells of 40 coal miners and 20 control subjects were taken from coal mines of Pail and Padhrar, Pakistan, to establish buccal anomaly frequencies of metal levels in the blood. Besides this, work history and duration hours were also analyzed. Results revealed that micronucleus frequencies positively correlated with the metal concentrations in the miner's blood. The change in the extent of nuclear damage per unit change in the year was 0.170 for micronuclei; however, with addition in each year of working experience, nuclear buds and broken egged nuclei increased by 0.316 and 0.194 units, where each year increases karyolysis by 0.349 units and karyorrhexis by 0.308 units, respectively. An increase in work hours and working years was positively correlated with cytogenetic damage. Nuclear damage in coal miners due to occupational exposure is obvious and increases with increasing work experience. Hence, the Buccal Micronucleus Cytome assay has proved to be an effective cytogenetic biomonitoring tool for assessing genetic and nuclear damage in coal miners.

Colorectal cancer (CRC) is one of the most common malignancies worldwide. As current therapies toward CRC, including chemotherapy and radiotherapy, pose limitations, such as multidrug resistance (MDR) as well as the intrinsic and potential cytotoxic effects, necessitating to find more effective treatment options with fewer side effects, traditional Chinese medicine (TCM) has an advantage in complementary therapies. In the present study, 3-(4,5-dimethylthiozol-2-yl)-2,5-diphenyltetrazolium bromide (MTT assays), trypan blue staining, colony formation, 4,6-diamidino-2-phenylindole dihydrochloride (DAPI) staining, cell cycle determination, and Annexin V-FITC/PI staining were used to examine the efficacy of Sanjie Yiliu Formula (SJYLF) against CRC proliferation and to investigate its underlying molecular mechanisms through protein expression of various proapoptotic factors by quantitative polymerase chain reaction (q-PCR) and Western blotting. This four-herb-TCM SJYLF can be suggested as one of the decoctions clinically effective in late-stage cancer treatment. Our results suggest that SJYLF robustly decreased the viability of only CRC cell lines (HCT-8, SW-480, HT-29, and DLD-1) and not the normal human kidney cells (HK-2). Moreover, SJYLF significantly suppressed proliferation and induced apoptosis in HCT-8 and downregulated cyclin D1, CDK4, and BCL-2, while Bax expression was upregulated at both mRNA and protein expression levels.

Continuous inhalation of coal dust among coal workers leads to a variety of disorders. The present study aims to evaluate the potential oxidative stress associated with coal dust generated from coal mining activities among exposed workers through the antioxidant enzyme system, including superoxide dismutase (SOD), catalase (CAT), and glutathione (GSH). In this study cohort, intensive coal mine workers were assessed for antioxidant variations. Blood samples were collected from dust-exposed workers (engaged in different activities at coal mines; n = 311) and residents of the same city (nonexposed, control group; n = 50). The workers' exposure to coal dust was categorized based on working area (administrative group, surface workers, underground workers), working hours (up to 8 h and more than 8 h), and time of service. The results showed significantly altered activities of SOD, CAT, and GSH among the whole exposed group and its categories compared to the control group. A significant difference was also observed between high- and low-exposure groups. Statistical analysis revealed a negative correlation between antioxidant activity (catalase and SOD) and coal dust levels. Besides, coal exposure was associated with the time of service, smoking status, and dietary habits. The findings of this study reveal higher oxidative stress among highly exposed coal mine workers (underground workers > surface workers > administrative group > nonexposed group), and longer working hours have more pronounced adverse effects on workers' health.

Irrigation using sewage water can be beneficial, as it can increase the productivity of crops but has negative consequences on crops, soil contamination, and human health. It contains a variety of toxins, such as chemicals and heavy metals, which damage the soil and crops. In this regard, the aim of the research was to assess the potential health hazards of iron (Fe) metal in food crops (leafy and root crops) treated with wastewater (T_1), canal water (T_2), and tube well water (T_3). Water, soil, and edible components of food crops were collected at random from three distinct locations. Fe concentration in samples was estimated using atomic absorption spectrophotometer, following wet digestion method. The Fe concentrations, ranged from 0.408 to 1.03 mg/l in water, 31.55 to 187.47 mgkg-1 in soil and 4.09 to 32.583 mgkg-1 in crop samples; which were within permissible limits of the World Health Organization (WHO). There was a positive correlation between soils and crops. The bioconcentration factor, enrichment factor (EF), daily intake of metals (DIM), health risk index (HRI), and target hazard quotient (THQ) all values were 1, which indicated soil contamination, but there was no Fe toxicity in crops, no health risk, and no-carcinogenic risk for these food crops in humans. To prevent the excessive accumulation of Fe metal in the food chain, regular monitoring is needed.

Extremozymes are thermophilic enzymes active in harsh environments. These enzymes have a more compact structure with an increased number of hydrophobic interactions, hydrogen bond networks, ion pairs, and disulfide bonds than their counterparts functional in mesophilic organisms. Extremophilic organisms surviving at extreme temperatures, pH, and pressure, express various stress-induced proteins that interact with extremozymes to keep them functional. Here, we summarize the structural features of extremozymes that can work well in the presence of nonaqueous organic solvents, in high salinity, in extreme temperatures, pHs, and cold temperatures. We also discuss how structures of these enzymes differ from mesophilic enzymes and how the understanding of structural and functional properties of such enzymes help to design new generation of genetically engineered enzymes with novel biotechnological applications.

The SARS CoV-2 pandemic has affected millions of people around the globe. Despite many efforts to find some effective medicines against SARS CoV-2, no established therapeutics are available yet. The use of phytochemicals as antiviral agents provides hope against the proliferation of SARS-CoV-2. Several natural compounds were analyzed by virtual screening against six SARS CoV-2 protein targets using molecular docking simulations in the present study. More than a hundred plant-derived secondary metabolites have been docked, including alkaloids, flavonoids, coumarins, and steroids. SARS CoV-2 protein targets include Main protease (MPro), Papain-like protease (PLpro), RNA-dependent RNA polymerase (RdRp), Spike glycoprotein (S), Helicase (Nsp13), and E-Channel protein. Phytochemicals were evaluated by molecular docking, and MD simulations were performed using the YASARA structure using a modified genetic algorithm and AMBER03 force field. Binding energies and dissociation constants allowed the identification of potentially active compounds. Ligand-protein interactions provide an insight into the mechanism and potential of identified compounds. Glycyrrhizin and its metabolite 18-β-glycyrrhetinic acid have shown a strong binding affinity for MPro, helicase, RdRp, spike, and E-channel proteins, while a flavonoid Baicalin also strongly binds against PLpro and RdRp. The use of identified phytochemicals may help to speed up the drug development and provide natural protection against SARS-CoV-2.

Hesperidin and naringenin are heteropolycyclic, aromatic bioflavonoids with a wide range of therapeutic and physiological effects. Hesperidin is a flavanone glycoside, while naringenin exists as an aglycon. Both compounds are widely present in citrus fruits, grapefruits, tomatoes, and some other plants. The antioxidant and free radical scavenging activities are found to be the most important properties of these compounds and complement their protective effects against a plethora of diseases including cancer, cardiovascular disease, hyperlipidemia, hypertension, neurodegeneration, diabetes, renal diseases, and skin damage. This chapter discusses the properties and therapeutic mechanisms/targets of hesperidin and naringenin in the treatment of various diseases. We have also discussed the metabolism and bioavailability of these flavanones in human and animal models. Besides, recent developments regarding the modulation and regulation of disease markers, clinical studies, and trial data assembled encourage future studies to develop improved clinical formulations for hesperidin and naringenin with better efficacy, bioavailability, delivery methods, and therapeutic potentials.

Cadmium (Cd) is one of the major hazardous elements that is very toxic to the health of both human and plants. The toxicity of Cd causes plants to suffer by disabling their overall physiological mechanisms. Therefore, present study was intended to investigate the synergistic role of AgNPs and IAA in improving the resilience against Cd toxicity and underlaying physiological and biochemical mechanisms in carrot (Daucus carota L.) plants. Also, the existence of genotypic variation for Cd tolerance in D. carota was also studied. The results revealed that Cd stress decreased plant growth attributes like root diameter, root length, root weight, shoot weight, shoot length, leaves fresh weight and leaves dry weight. Nonetheless, AgNPs and IAA mitigated Cd stress by detoxifying reactive oxygen species (ROS). Additionally, the application of AgNPs and IAA boosted plant growth through reducing the level of malondialdehyde (MDA). Enhancement in the activity of phenol synthesizing and oxidizing enzymes including peroxidase, polyphenol oxidase and phenylalanine ammonia-lyase was also observed by application of AgNPs and IAA. The increased activities of antioxidant enzymes including POX, PPO and PAL by the combined application of AgNPs and IAA advocate stress ameliorative role against Cd stress in plants. The enhanced Cd content was detected in the roots as compared to shoots of treated plants. Pre breed 22 was found as a Cd tolerant genotype.

Arsenic (As) polluted food chain has become a serious issue for the growth and development of humans, animals and plants. Nitric oxide (NO) or silicon (Si) may mitigate As toxicity. However, the combined application of NO and Si in mitigating As uptake and phytotoxicity in Brassica juncea is unknown. Hence, the collegial effect of sodium nitroprusside (SNP), a NO donor and Si application on B. juncea growth, gas exchange parameters, antioxidant system and As uptake was examined in a greenhouse experiment. Arsenic toxicity injured cell membrane as signposted by the elevated level of malondialdehyde (MDA) and hydrogen peroxide (H2O2), thus decreasing the growth of stressed plants. Moreover, As stress negatively affected gas exchange parameters and antioxidative system of plants. However, NO or/and Si alleviated As induced oxidative stress through increasing the activity of superoxide dismutase (SOD), ascorbate peroxidase (APX), glutathione reductase (GR), glutathione S-transferase (GST), glutathione (GSH), along with thiol and proline synthesis. Furthermore, plants treated with co-application of NO and Si showed improved growth, gas attributes and decreased As uptake under As regimes. The current study highlights that NO and Si synergistically interact to mitigate detrimental effects of As stress through reducing As uptake. Our findings recommend combined NO and Si application in As spiked soils for improvement of plant growth and stress alleviation.

Levels of Mo, As, Se, Fe, Cu, Zn, Ni, and Pb were determined in a vegetable commonly consumed in Pakistan. Samples were collected from three different sites (Ratokala, Phularwan, and Mailowal) supplied with three different water sources. Mo and Pb in water and Mo and As in C. sativum were higher than the suggested standards at the international level. Lower bioconcentration factor and pollution load index were seen at Site-I than at other sites. A positive association was found between the rhizosphere metals and those of the vegetable at all three sites. Enrichment factor at three sites ranged from 0.28 to 10.39. The highest value of daily intake of metals was found for Fe (0.245 mg/kg/day) and of the health risk index for As (70.41) at the wastewater inundated site. It is inferred that uptake of Mo, As, Cu, Ni, and Pb through C. sativum represents a high health danger to the individuals using this vegetable.

Environmental changes as well as a worsening environmental quality, strongly affect women lives. A woman goes through a wide range of changes throughout her lifetime, from fetal development through her post-menopause years, which involves a direct relationship between body metallic ion concentration and development and function. When this relationship is balanced, it helps to create conditions for good health. However, when this relationship is out of balance, it can lead to a range of health problems that can be painful and devastating. With exposure, women and girls are at greater risk for developing reproductive health problems including early puberty, uterine fibroids, endometriosis, polycystic ovarian syndrome (PCOS) and breast cancer. These health problems can be devastating to woman’s fertility, overall health and quality of life. In this scenario, the present investigation study was carried out to analyze the concentration of different metallic elements in female body by using scalp hairs. Hairs being noninvasive material are good indicator of metallic element in human body. Three categories of females viz. unmarried, pregnant and menopause were selected. Concentration of iron, copper and zinc were found lower among pregnant ones as compared to unmarried and menopausal females. Moreover, concentration of iron, zinc and copper were significantly higher among unmarried females. Key words: Pregnant, unmarried, menopause, hairs.
