This study investigated the effects of pre-heating fertile Japanese quail eggs during storage on embryonic development, incubation performance, hatched chick quality, and chick performance up to 35 days of age. The experiment was laid out in a completely randomized design in a 3 × 2 + 1 factorial arrangement, totaling seven treatments. These treatments included three storage periods (3.5, 6.5, and 9.5 days) and two pre-heating periods (0 and 4 h at 37.5°C and 60% RH). Pre-heating was conducted on the third day of egg storage inside the incubator machine, with the control treatment involving the storage of fertile eggs for 12 h without pre-heating. Extended egg storage for more than six days led to an increase in the percentages of yolk and shell, elevated albumen pH, reduced albumen percentage, diminished hatchability rate, and an increased embryonic mortality rate. Pre-heating the eggs resulted in a reduction in the weight, length, and amount of residual yolk sac of the newly hatched chick. No significant effects of pre-heating fertile Japanese quail eggs during storage were observed on the studied variables. Therefore, the protocol involving a 4-h pre-heating at 37.5 ºC and 60% relative humidity during the storage period does not yield improvements in incubation rates or chick quality in Japanese quail. Further studies are warranted to determine the optimal protocol for pre-heating Japanese quail eggs.

Um animal é considerado saudável quando se encontra em um estado de bem-estar biopsicossocial. A domesticação e a forma de criação influenciam de maneira significativa na vida dos animais, determinando aspectos comportamentais e fisiológicos. Hamsters costumam ser criados em gaiolas, que são ambientes muito reduzidos em relação ao que esses roedores costumam viver quando livres na natureza. O objetivo do presente trabalho foi avaliar as mudanças que ocorreram em hamsters devido a domesticação e, conforme isso, determinar qual é a melhor condição de criação para que esses animais vivam em bem-estar.

Physical activity has been widely investigated in horses to elucidate locomotion characteristics and behavior. However, research in real environment of training stables is limited. The objective of this study was to evaluate the effect of training regimen on night time physical activity of racehorses. Physical activities of twenty animals were monitored during the night time using accelerometers. The animals were compared in terms of training regimen: horses subjected to training on continual days, and horses subjected to training on intermittent days; age and sex were also considered. The variables analyzed were: counts per minute, percentage of time in (sedentary, light, moderate and vigorous) physical activity. Statistical analysis was performed by the PROCGLM procedure (ANOVA) and the means were calculated by Tukey's test. The training regimen impacts the physical activity of thoroughbreds. The mean counts per minute showed a higher physical activity in the intermittent trained animals in relation with continuously trained animals. The continuously trained animals presented a greater percentage of time in sedentary physical activity than those intermittent trained. However, intermittently trained horses spent a larger percentage of time in moderate and vigorous physical activity, in comparison with continuously trained animals. In conclusion, racehorses subject to training on continual days have lower physical activity in the night time than those that train on intermittent days.

There is insufficient information about germination, seed biology and seedling establishment of native grass species of the cerrado ( Brazilian savannah) to recommend their use in restoration of disturbed areas. We evaluated the production, percentage of fertile seeds, germination, seedling growth and establishment in the field of eighteen native grass species and one exotic invasive species ( Melinis minutiflora) and also the effects of different storage periods on germination. The percentage of fertile seeds varied from 6·0 to 94·2% among native species. Ten species produced a minimum of 800 fertile seeds per gram. Germination rate was in the range of 11·0 to 98·8%. Storage for 1 year reduced the germination rate in one species, did not affect germination of three species and increased germination (by 2-10 times) in 14 species. After 2 years of storage, six species showed reductions in germination. Three different patterns of resource allocation were observed and might provide mechanisms for increasing seedling establishment under different environmental conditions. In the field, only two species flowered in the first year after sowing and an additional eight in the second year of observation. Melinis minutiflora is an aggressive species with more than three times the fertile seed production of native species (2820 seeds per gram), high germination rate (55%) and a high number of germinable seeds (1551 per gram). Despite the wide variation in seed characteristics and the delay in flowering, it is concluded that most native grass species can be used in projects aiming at the restoration and management of degraded areas. Cancer genetics has uncovered many tumor-suppressor and oncogenic pathways, but few alterations have revealed mechanisms involved in tumor spreading. Here, we examined the role of the third most significant chromosomal deletion in human melanoma that inactivates the adherens junction gene NECTIN1 in 55% of cases. We found that NECTIN1 loss stimulates melanoma cell migration in vitro and spreading in vivo in both zebrafish and human tumors specifically in response to decreased IGF1 signaling. In human melanoma biopsy specimens, adherens junctions were seen exclusively in areas with low IGF1 levels, but not in NECTIN1-deficient tumors. TGF-β signaling is involved in pancreatic ductal adenocarcinoma (PDAC) tumorigenesis, representing one of the four major pathways genetically altered in 100% of PDAC cases. TGF-β exerts complex and pleiotropic effects in cancers, notably via the activation of SMAD pathways, predominantly SMAD2/3/4. Though SMAD2 and 3 are rarely mutated in cancers, SMAD4 is lost in about 50% of PDAC, and the role of SMAD2/3 in a SMAD4-null context remains understudied. We herein provide evidence of a SMAD2/3 oncogenic effect in response to TGF-β1 in SMAD4-null human PDAC cancer cells. We report that inactivation of SMAD2/3 in SMAD4-negative PDAC cells compromises TGF-β-driven collective migration mediated by FAK and Rho/Rac signaling. Moreover, RNA-sequencing analyses highlight a TGF-β gene signature related to aggressiveness mediated by SMAD2/3 in the absence of SMAD4. Using a PDAC patient cohort, we reveal that SMAD4-negative tumors with high levels of phospho-SMAD2 are more aggressive and have a poorer prognosis. Thus, loss of SMAD4 tumor suppressive activity in PDAC leads to an oncogenic gain-of-function of SMAD2/3, and to the onset of associated deleterious effects., (© 2022. The Author(s).)

It is of utmost importance to decipher the role of chronic exposure to low doses of environmental carcinogens on breast cancer progression. The early-transformed triple-negative human mammary MCF10AT1 cells were chronically (60 days) exposed to low doses (10 -10 M) of Benzo[a]pyrene (B[a]P), a genotoxic agent, and/or Bisphenol A (BPA), an endocrine disruptor. Our study revealed that exposed MCF10AT1 cells developed, in a time-dependent manner, an acquired phenotype characterized by an increase in cancerous properties (anchorage independent growth and stem-like phenotype). Co-exposure of MCF10AT1 cells to B[a]P and BPA led to a significantly greater aggressive phenotype compared to B[a]P or BPA alone. This study provided new insights into the existence of a functional interplay between the aryl hydrocarbon receptor (AhR) and the G protein-coupled receptor 30 (GPR30) by which chronic and low-dose exposure of B[a]P and/or BPA fosters the progression of MCF10AT1 cells into a more aggressive substage. Experiments using AhR or GPR30 antagonists, siRNA strategies, and RNAseq analysis led us to propose a model in which AhR signaling plays a "driver role" in the AhR/GPR30 cross-talk in mediating long-term and low-dose exposure of B[a]P and/or BPA. Retrospective analysis of two independent breast cancer cohorts revealed that the AhR/GPR30 mRNA expression signature resulted in poor breast cancer prognosis, in particular in the ER-negative and the triple-negative subtypes. Finally, the study identified targeting AhR and/or GPR30 with specific antagonists as a strategy capable of inhibiting carcinogenesis associated with chronic exposure to low doses of B[a]P and BPA in MCF10AT1 cells. Recombination systems represent a major breakthrough in the field of genetic model engineering. The Flp recombinases (Flp, Flpe, and Flpo) bind and cleave DNA Frt sites. We created a transgenic mouse strain ([Fsp1-Flpo]) expressing the Flpo recombinase in fibroblasts. This strain was obtained by random insertion inside mouse zygotes after pronuclear injection. Flpo expression was placed under the control of the promoter of Fsp1 (fibroblast-specific protein 1) gene, whose expression starts after gastrulation at Day 8.5 in cells of mesenchymal origin. We verified the correct expression and function of the Flpo enzyme by several ex vivo and in vivo approaches. The [Fsp1-Flpo] strain represents a genuine tool to further target the recombination of transgenes with Frt sites specifically in cells of mesenchymal origin or with a fibroblastic phenotype., (© 2020 The Authors. Objective: To evaluate longterm drug survival (proportion of patients still receiving treatment) and discontinuation of etanercept (ETN), infliximab (IFX), adalimumab (ADA), certolizumab pegol (CZP), and golimumab (GOL) using observational data from patients with rheumatoid arthritis (RA)., Methods: Following a systematic literature review, drug survival at 12 and 12-24 months of followup was estimated by summing proportions of patients continuing treatment and dividing by number of studies. Drug survival at ≥ 36 months of followup was estimated through Metaprop., Results: There were 170 publications included. In the first-line setting, drug survival at 12 months with ETN, IFX, or ADA was 71%, 69%, and 70%, respectively, while at 12-24 months the corresponding rates were 63%, 57%, and 59%. In the second-line setting, drug survival at 12 months with ETN, IFX, or ADA was 61%, 69%, and 55%, respectively, while at 12-24 months the corresponding rates were 53%, 39%, and 43%. Drug survival at ≥ 36 months with ETN, IFX, or ADA in the first-line setting was 59% (95% CI 46-72%), 49% (95% CI 43-54%), and 51% (95% CI 41-60%), respectively, while in the second-line setting the corresponding rates were 56% (95% CI 52-61%), 48% (95% CI 40-55%), and 41% (95% CI 36-47%). Discontinuation of ETN, IFX, and ADA at 36 months of followup was 38-48%, 42-62%, and 38-59%, respectively. Data on CZP and GOL were scarce., Conclusion: After > 12 months of followup, more patients with RA receiving ETN remain on treatment compared with other tumor necrosis factor inhibitors.

Transforming growth factor (TGFβ) is a secreted factor, which accumulates in tissues during many physio- and pathological processes such as embryonic development, wound healing, fibrosis and cancer. In order to analyze the effects of increased microenvironmental TGFβ concentration in vivo, we developed a conditional transgenic mouse model (Flpo/Frt system) expressing bioactive TGFβ in fibroblasts, a cell population present in the microenvironment of almost all tissues. To achieve this, we created the genetically-engineered [Fsp1-Flpo; FSF TGFβ CA ] mouse model. The Fsp1-Flpo allele consists in the Flpo recombinase under the control of the Fsp1 (fibroblast-specific promoter 1) promoter. The FSF TGFβ CA allele consists in a transgene encoding a constitutively active mutant form of TGFβ (TGFβ CA ) under the control of a Frt-STOP-Frt (FSF) cassette. The FSF TGFβ CA allele was created to generate this model, and functionally validated by in vitro, ex vivo and in vivo techniques. [Fsp1-Flpo; FSF TGFβ CA ] animals do not present any obvious phenotype despite the correct expression of TGFβ CA transgene in fibroblasts. This [Fsp1-Flpo; FSF TGFβ CA ] model is highly pertinent for future studies on the effect of increased microenvironmental bioactive TGFβ concentrations in mice bearing Cre-dependent genetic alterations in other compartments (epithelial or immune compartments for instance). These dual recombinase system (DRS) approaches will enable scientists to study uncoupled spatiotemporal regulation of different genetic alterations within the same mouse, thus better replicating the complexity of human diseases.

Pancreatic ductal adenocarcinoma (PDAC) is one of the solid tumors with the poorest prognosis. The stroma of this tumor is abundant and composed of extracellular matrix and stromal cells (including cancer-associated fibroblasts and immune cells). Nerve fibers invading this stroma represent a hallmark of PDAC, involved in neural remodeling, which participates in neuropathic pain, cancer cell dissemination and tumor relapse after surgery. Pancreatic cancer-associated neural remodeling is regulated through functional interplays mediated by physical and molecular interactions between cancer cells, nerve cells and surrounding Schwann cells, and other stromal cells. In the present study, we show that Schwann cells (glial cells supporting peripheral neurons) can enhance aggressiveness (migration, invasion, tumorigenicity) of pancreatic cancer cells in a transforming growth factor beta (TGFβ)-dependent manner. Indeed, we reveal that conditioned medium from Schwann cells contains high amounts of TGFβ able to activate the TGFβ-SMAD signaling pathway in cancer cells. We also observed in human PDAC samples that high levels of TGFβ signaling activation were positively correlated with perineural invasion. Secretome analyses by mass spectrometry of Schwann cells and pancreatic cancer cells cultured alone or in combination highlighted the central role of TGFβ in neuro-epithelial interactions, as illustrated by proteomic signatures related to cell adhesion and motility. Altogether, these results demonstrate that Schwann cells are a meaningful source of TGFβ in PDAC, which plays a crucial role in the acquisition of aggressive properties by pancreatic cancer cells.

Objective: To evaluate the diagnostic value of color Doppler ultrasound (CDUS) for the detection of sacroiliitis, in patients with inflammatory back pain (IBP)., Methods: Consecutive patients with IBP and suspected axial spondyloarthritis (SpA), but without a definitive diagnosis, were included. Consecutive patients with defined SpA and axial involvement were included as a control group. All patients underwent clinical evaluation, magnetic resonance imaging (MRI), and CDUS of sacroiliac joints (SIJ) within the same week. Sensitivity, specificity, positive predictive value (PPV) and negative predictive value (NPV) for the diagnosis of sacroiliitis by CDUS were calculated, using MRI as the gold standard., Results: There were 198 SIJ evaluated in 99 patients (36 with previous SpA). There were 61 men (61.6%), with a mean age of 39.8 years (SD 11.3) and median disease duration of 24 months (IQR 12-84). At the patient level, CDUS had a sensitivity of 63% (95% CI 48.7-75.7%) and a specificity of 89% (95% CI 76-96%). The PPV was 87.2% (95% CI 72.6-95.7%) and the NPV was 66.7% (95% CI 53.3-78.3%). At joint level, CDUS had a sensitivity of 60% (95% CI 49-70%) and a specificity of 93% (95% CI 88-98%). The PPV was 83% (95% CI 78-95%) and the NPV was 43% (95% CI 33-56%). The sensitivity of CDUS for the diagnosis of axial SpA was 54% (95% CI 36.6-71.2%), specificity was 82% (95% CI 63.1-93.9%), PPV was 79% (95% CI 57.8-92.9%), and NPV was 59% (95% CI 42.1-74.4%)., Conclusion: CDUS showed adequate diagnostic properties for detection of sacroiliitis and is a useful tool in patients with IBP.

Objective: To evaluate components of the minimal disease activity (MDA) criteria in psoriatic arthritis (PsA)., Methods: In patients achieving and not achieving MDA, fulfillment of each of the 7 criteria was evaluated., Results: Among 41 patients with MDA, 7.4% did not fulfill the tender/swollen joint count whereas 49% did not fulfill the skin criteria. Of the 42 patients not fulfilling MDA, 100%, 76.5%, and 65% did not fulfill the patient pain score, the patient's global assessment, and the Psoriasis Area and Severity Index (PASI), respectively., Conclusion: A minority of patients with PsA fulfilling the MDA criteria presented active joints, but half had active skin. Visual analog scale scores and the PASI prevented patients from achieving MDA.

Turner syndrome (TS) is an important cause of short stature in girls. Patients with TS most often do not have growth hormone deficiency (GHD). Testing GH secretion is not indicated despite the presence of short stature. In the last 20 years only three cases were reported with this association in Pubmed. We describe a case of an 11 year old girl with short stature and karyotype confirmed TS: 45,X(16)46,X,i(X)(ql0)(13). Because her growth velocity was low (-3 SD), we evaluated the GH response with stimulating tests and the results were under the normal range. These findings were compatible with GHD. It is important to check for GHD in patients with TS whenever the growth velocity is low for age and sex.

Introduction: The Standard Precautions are measures to prevent healthcare workers' exposure to infectious agents, like HIV, HBV and HCV, through the routine use of techniques and appropriate barriers that reduce the exposure probability. These measures intend to prevent contact with blood and other fluids, potentially infected, contributing to the reduction of nosocomial infection., Objectives: To evaluate the knowledge, attitudes and adherence to the standard precautions for infection control among the healthcare workers of the Hospital Geral de Santo António, through the application of a questionnaire. To identify the gaps of each professional group and clinical departments, in order to better define the needs to planning future education., Results: Of the total of 172 questionnaires, 7% did not know about the standard precautions, the majority from medical staff. Importantly 21% affirm recapping needles. Globally, a low level knowledge about the ways HIV, HBV and HCV is transmitted was verified, mostly regarding the possibility of transmission from dishes and other material necessary to give food to the patients. About 95% considered it important to wash the hands in several different situations but 21% are unaware of alternative alcoholic solutions., Conclusions: There is a precarious knowledge of these measures, enhancing the risk of nosocomial infection and pointing to the need of specific education for all different health professionals.

It has been demonstrated that respiratory resistance and elastance increase whilst the abdomen remains open during longitudinal laparotomy. We wished to determine whether changes also occur after abdominal closure in the same animal preparation. In 10 sedated, anaesthetized paralysed, and mechanically-ventilated rats (309 +/- 33 (SD) g), resistances and elastances of the respiratory system, lung, and chest wall were measured both before longitudinal laparotomy and directly after abdominal closure. Furthermore, the resistances were also split into their initial and difference components, the former reflecting the Newtonian resistances and the latter representing the viscoelastic/inhomogeneous pressure dissipations in the system. For this purpose, the end inflation occlusion during constant inspiratory flow method was used. After laparotomy, no statistically significant changes were found in elastances and resistances of the respiratory system, lungs and chest wall (paired Student's t-test, significance level = 5%). It can be concluded that after midline xiphipubic laparotomy accompanied by bilateral ventro-dorsal infracostal incision, respiratory resistances and elastances were not different from those found in the control condition.
