Abstract

The potential advantage of each linkage on the sequence-specific DNA binding was examined via comparison of the DNA binding of peptide dimers with covalent and noncovalent dimerization domains. The results suggested that the increased sensitivity of equilibrium binding to the change in protein concentrations and the enhancement of the selectivity of specific DNA binding may be attributed to cooperativity in DNA binding. It was also suggested that cooperative DNA binding may be caused by modest dimerization only upon the specific DNA site.

Abstract

Cooperative DNA binding and reduction of the stability of the non-specific DNA binding complex accomplished the highly selective binding of G2AdCd, the peptide from the synthesis of a series of short peptides derived from the basic region of the basic leucine zipper protein GCN4. G2AdCd was found to possess both host and guest molecules in the same peptide chain. A comparison of DNA binding of the G2AdCd peptides to the single-, double-, and triple-direct repeat sequences of the CRE half-site revealed no binding at the single CRE half-site. Cooperative binding of the double-direct repeat sequence as a dimer and cooperative formation of a 3:1 G2AdCd-DNA complex for a triple-direct repeat sequence were observed.

40. A Longitudinal Study of the Impact of COVID-19 on Optimism Prediction

Abstract

The purpose of this longitudinal study was to investigate how optimistic predictions, hopelessness, and depressive symptoms changed as a result of the COVID-19 pandemic, and the causal relationships between these variables. To achieve this purpose, we used data from online surveys conducted in 2019 and 2021 among men and women aged 20–79. Based on item response theory, we developed a future prediction task for the assessment of optimistic predictions. Our comparison of online survey responses found a decline in optimistic predictions before and after the pandemic. More specifically, there were no change in predictions of negative future events, but there was the decrease in predictions of positive future events. Furthermore, we found that those who were more stressed by COVID-19 were less likely to have an optimistic view of the near future. We also found a relationship between optimistic predictions and hopelessness and depressive symptoms with lower optimism predicting more hopelessness and more depression predicting lower optimism. To prevent feelings of hopelessness, it is important to help people develop positive expectations about the future.

Abstract

Transposons are major drivers of mammalian genome evolution. To obtain new insights into the contribution of transposons to the regulation of protein translation, we here examined how transposons affected the genesis and function of upstream open reading frames (uORFs), which serve as cis‐acting elements to regulate translation from annotated ORFs (anORFs) located downstream of the uORFs in eukaryotic mRNAs. Among 39,786 human uORFs, 3,992 had ATG trinucleotides of a transposon origin, termed “transposon‐derived upstream ATGs” or TuATGs. Luciferase reporter assays suggested that many TuATGs modulate translation from anORFs. Comparisons with transposon consensus sequences revealed that most TuATGs were generated by nucleotide substitutions in non‐ATG trinucleotides of integrated transposons. Among these non‐ATG trinucleotides, GTG and ACG were converted into TuATGs more frequently, indicating a CpG methylation‐mediated process of TuATG formation. Interestingly, it is likely that this process accelerated human‐specific upstream ATG formation within transposon sequences in 5′ untranslated regions after divergence between human and nonhuman primates. Methylation‐mediated TuATG formation seems to be ongoing in the modern human population and could alter the expression of disease‐related proteins.
