12. A composite cement of high magnesium sulphate resistance

Abstract

This study investigates the magnesium sulphate resistance of chemically activated phosphorus slag-based composite cement (CAPSCC). Enough mortar specimens were prepared from phosphorus slag (80 wt.%), type II Portland cement (14 wt.%), and compound chemical activator (6 wt.%) and were exposed to 5% magnesium sulphate solution after being cured. Mortar specimens of both type II and V Portland cements (PC2 and PC5) were also prepared and used for comparison purpose. According to the test results, after 12 months of exposure, PC2, PC5 and CAPSCC exhibited 43.5, 35.2 and 25.2% reduction in compressive strength, 0.136, 0.110, and 0.026% expansion in length, and 0.91, 2.2, and 1.78% change in weight, respectively. Complementary studies by X-ray diffractometry and scanning electron microscopy revealed that CAPSCC has a very low potential for the formation of sulphate attack products, especially ettringite. The results confirm a high magnesium sulphate resistance for CAPSCC compared to PC2 and PC5., Este trabajo aborda el estudio de la resistencia al sulfato de magnesio de un cemento compuesto con base de escoria de fósforo activada químicamente (CAPSCC). Se prepararon muestras de mortero a partir de escoria de fósforo (80% en peso), cemento Portland tipo II (14% en peso) y activador químico (6% en peso) y tras el curado, se expusieron a una solución de sulfato de magnesio al 5%. También se prepararon morteros de cementos Portland de tipo II y V (PC2 y PC5) que se usaron con fines comparativos. De acuerdo a los resultados obtenidos, después de 12 meses de exposición, PC2, PC5 y CAPSCC mostraron un 43.5, 35.2 y 25.2% de reducción en la resistencia a la compresión, 0.136, 0.110, y 0.026% de expansión en longitud, y 0.91, 2.2 y 1.78% de cambio en peso, respectivamente. Estudios complementarios por difracción de rayos X y microscopía electrónica de barrido revelaron que los cementos CAPSCC tienen un potencial muy bajo para la formación de productos de ataque de sulfato, especialmente etringita. 13. Short term reactogenicity of cellular DTP vaccine in 4-6 year old children in Tehran, Iran

Abstract

Objective(s): To detcnninc the short-term reacrogenicity of diphtheria-tetanus-whole cell pertussis (DTwP) vaccine administered to preschool children in a number of health centers of Tehran in 2006. Methods: In this prospective cohort study, 337 children aged 4-6 years were injected with DTwP vaccine manufactured by Razi Institute of Iran. Reactogenicity was assessed by the parents for 7 days post vaccination using diary cards. Local (pain, redness and swelling) and systemic (fever, loss of appetite, gasterointestinal symptoms, vomiting and eczema) side effects were recorded daily. Results: Out of 337 children, 312 (92.6%) reported local reactions and 220 (65.3%) reported systemic reactions. No serious adverse events related to vaccination were reported. Among local reacuons, pain was the most frequent (86.9%), but 11 was mostly mild or moderate. Redness (52.8%) and swelling (41.2%) were the most frequently observed signs in the second day. The systemic reactions observed in children included fever (48.4%), loss of appetite (24%), gastrointestinal symptoms (5 6%), vomiting (8%) and eczema (2.7%). Only 3.6% of children had auxiliary fever above 39 chr('39')c. All signs were observed to have reduced or completely disappeared during a week. Conclusion: Compared with previous reports in Iran, reactogenicity of DTwP of Razi Institute seems to be reduced, but it was still more frequent than the internationally approved cellular vaccine counterparts. Reactogcnicity of the cellular triple vaccine may be related to the vaccine formulation or the bacterial cell fragments used in vaccine production.

Abstract

Background and Objectives: Diagnosis of mental disorders and resilience of individuals during the epidemic of diseases is necessary for planning. The purpose of this study was to determine the resilience, stress, depression and anxiety of households in Anar city at the time of the outbreak of COVID-19 disease. Materials and Methods: The present descriptive study was conducted among 453 households in Anar city (from Rafsanjan County). Data were gathered using questionnaires and in a web-based approach. Data were analyzed using independent t-tests, Tukey's post hoc test and Spearman's correlation coefficient. Results: The results showed that 251 (57.7%), 233 (51.4%) and 260 (57.3%) participants had levels of stress, anxiety and depression, respectively. The mean and standard deviation of resilience score was 37.55±19.18. There was a significant relationship between anxiety and level of education, job and age and also depression and job levels. Conclusion: Due to the high prevalence of mental disorders and low resilience during the epidemic of the disease and its consequences, psychological interventions are recommended. Abstract

This study was done to evaluate the effect of sperm source on chromatin integrity and ICSI outcomes. One hundred and thirteen samples containing epididymal aspirates of 57 obstructive azoospermic men and 56-ejaculated semen of normozoospermic men were included in this study. Sperm chromatin status was evaluated by Chromomycin A3 (CMA3), Aniline Blue (AB) and Toluidine Blue (TB). Fertilization rate and embryo quality were recorded. In epididymal group the percentage of sperms stained with AB, CMA3 and TB were significantly higher compared to ejaculate group while fertilization rate (60.6% vs. 74.04%) was significantly lower. However, embryo quality was not significantly different between two groups. In addition, abnormal sperm chromatin condensation and DNA fragmentation were not correlated with fertilization rate and embryo quality. Our results highlight the role of epididymis in sperm maturation and confirm that ICSI using ejaculated sperm is the gold standard for treatment of infertile men.

20. Fishing trend and fisheries potential analyses for the Persian Gulf and Sea of Oman, 1973-2003

Abstract

Catch data from Persian Gulf and Sea of Oman during 1973 to 2001 were used to assess the state of the fishery development in these ecosystems. Also, fishery trends were analyzed to determine catch fluctuations during the period. Catch trend for each species from 1997 to 2003 was analyzed and fishing potential of the species for the following year was predicted. Based on the assessments, we can summarize fishing development in the southern waters of Iran into three periods: undeveloped, rapid development stage and developed stage. The undeveloped stage continued till early 1981, continued by a rapid stage until 1997, reaching to maximum level of at this year and continuing ever since. Catch trend analyses for each species during 1997 to 2003 showed different patterns for each species depending on its economic value. However, the mean catch of all species showed a sustainable trend in the two marine ecosystems. It is concluded that the catch quantity for most of the species have already reached a maximum level. Therefore, in order to keep fisheries sustainable in the region, excess fishing should be avoided.

Abstract

This study was designed to find out Ruta graveolens L. functional components, which have immobilisation effect on human spermatozoa for contraceptive use. A five-step fractionation method was used to derive different components from rue aqueous extract by using hexane, chloroform, ethanol, acetone and ultrapure water. Gas chromatography-mass spectrophotometery ( GC- MS) of all fractions and the aqueous extract were performed to determine the chemical components. The immobilisation assay and membrane integrity test were also performed with four different coumarins, which were found in GC- MS in a concentration of 10 μ m. Hexane, chloroform, acetone and ethanol fractions could significantly decrease motility of sperms within the first and the second hours. Hexane fraction had also significant immediate effect. The aqueous fraction had no effect on sperm motility. Meanwhile, GC- MS revealed that aqueous extract and effective fractions had similar coumarin compounds. We performed the immobilisation assay on four different coumarins, which were found in GC- MS in a concentration of 10 μ m. Reduction of sperm motility was only significant for xanthotoxin. In the sperm viability and membrane integrity tests, hexane and ethanolic fractions could impair sperm vitality significantly, in contrast to coumarins. These results indicated that a part of immobilising effect of rue could be due to its coumarins. The possible mechanism could be blocking of spermatozoa potassium channels. [ABSTRACT FROM AUTHOR]

Abstract

Background: Pi-GST and Mu-GST are subclasses of glutathione S-transferase that present on human sperm surface and play an important role against oxidative stress. Therefore, any defects in the enzyme activity may be associated with male infertility.In this study the polymorphisms of GSTM1 and GSTP1 in association with enzyme activity and sperm parameters were studied. Methods: This case-control study involved 95 men with oligoastenoteratozoospermia and 26 controls with normozoospermia. Semen analyses were carried out according to WHO guidelines. Blood DNA was extracted using salting out procedures. GSTM1 and GSTP1 polymorphisms gene were determined through PCR-RFLP and multiplex PCR, respectively. Finally, Glutathione S-transferase activity was measured. Results: Frequencies of GSTM1 null genotype in oligoastenoteratospermic and normospermic groups were 52.1% and 53.8% respectively. There were no statistically significant differences in sperm parameters and enzyme activity between GSTM1 null and positive genotypes in two groups. There were no statistically significant differences in glutathione S-transferase activity between oligoastenoteratospermia and normospermic groups (p>0.05). All the 121 men in this study had Ile/Ile genotypes at 105 codon of GSTP1. Frequency of normal homozygote (114Ala/Ala), heterozygote (114Ala/Val) and mutant homozygote (114Val/Val) genotypes in oligoastenoteratospermic group were 81.1%, 17.9% and 1.1% respectively but in the control group they were 88.5%, 11.5% and null. Conclusions: Total glutathione S-transferase activity and sperm parameters were not affected by deficient Glutathione S-transferase activity in GSTM1 null genotype. Compensate activity of other sperm surface glutathione S-transferase isozymes, like GSTP1, may justify the cause. [ABSTRACT FROM AUTHOR]

Abstract

Ubiquitin, a 8.5 kDa peptide that marks other proteins for proteasomal degradation, tags defective spermatozoa during epididymal passage and is proposed as a biomarker for sperm quality. The present study was designed to evaluate the relationships between sperm ubiquitination, sperm chromatin integrity and semen parameters. Semen samples from 63 couples were collected and analysed according to World Health Organization criteria. Each sample was evaluated for sperm ubiquitination by the direct immunofluorescence method, using anti-ubiquitin antibodies. Chromatin integrity of the same samples was analysed using acridine orange (AO) and toluidine blue (TB) tests. A positive correlation was found between ubiquitinated spermatozoa and the percentage of spermatozoa with abnormal chromatin t AO: r = 0.58. P < 0.001 and TB: r = 0.48, P < 0.001 ). Negative correlations were obtained between sperm ubiquitination and: sperm count (r = -0.2. P = 0.048), sperm morphology (r = -0.36. P = 0.003), rapidly progressive motility (r = -0.25, P = 0.044) and slow progressive motility (r = -0.28. P = 0.022). Sperm ubiquitination was positively correlated with the percentage of immotile spermatozoa. These results show that among semen parameters, chromatin abnormality is more closely associated with sperm ubiquitination and further validate sperm ubiquitination as a suitable marker for sperm quality. Abstract

Background:Several lines of evidence show the implication of oxidative stress in the etiology of male infertility. Recently, the role of coenzyme Q10(CoQ10) in the prevention and treatment of disease has been intensively probed. However, definitive efficacy studies in oligoasthenoteratozoospermia (OAT) have not been completed yet. Aim:To evaluate the effect of CoQ10supplementation on semen parameters in idiopathic OAT (iOAT). Material/subjects and methods:A double-blind placebo controlled clinical trial was carried out. A total of 47 infertile men with iOAT were randomly assigned to receive 200 mg CoQ10daily or placebo during a 12-week period. Semen parameters were determined using microscopic evaluation according to World Health Organization guidelines. Lipid peroxidation was assessed by measuring the concentration of plasma malondialdehyde. We evaluated the total antioxidant capacity of seminal plasma. To compare variables between and within the 2 groups we used independent t-test and Paired t-test. Results:The trial showed non-significant changes in semen parameters of CoQ10group. However, concentrations of thiobarbituric acid-reactive substances were significantly (p<0.05) reduced in serum of treated groups compared with the control. Furthermore, total antioxidant capacity of seminal plasma significantly increased in the CoQ10group (p<0.05). Conclusion:Our results provide further evidence suggesting that CoQ10supplementation is associated with alleviating oxidative stress, although it does not show any significant effects on sperm concentration, motility and morphology. It may be suggested that CoQ10could be taken as an adjunct therapy in cases of OAT. Further studies are needed to draw a final conclusion.

Abstract

Background In cancer patients, chemo and radiotherapy can cause infertility by damaging spermatogenesis process. This process is based on self-renewal and differentiation of a rare population of the testicular cells called Spermatogonial Stem Cells (SSCs). Scientists have tried to isolate, enrich and culture Human spermatogonial stem cells, hoping to resolve infertility problems in cancer recovered patients in the future. Methods Spermatogonial stem cells were isolated and purified from human testicular biopsies sample consisting of at least 500,000 and at most 2,000,000 cells. Two enzymatic digestion steps were performed. Enriching methods, differential plating, and specific culture in serum-free medium with added growth factors: human GDNF, bFGF, EGF and LIF was performed on coated dishes. Results Human spermatogonial stem cell clusters were observed after 7 to 10 days in specific culture, then after several passages and successful expanding duration of 52 days, the cells were evaluated by three layer immunocytochemistry test (LSAB) to stain GPR125 protein as a surface marker in human spermatogonial stem cells. Conclusion In current study human spermatogonial stem cell were isolated and expanded with the least manipulations in comparison with the other usual isolation methods like florescent or magnetic activated cell sorting. In contrast to the other SSCs isolation and culture methods, this system is based on the testicular biopsies against large samples, thus suggested method in this study is closer to clinical usage in the future.

Abstract

R-Phycoerythrin (R-PE), a fluorescent protein from phycobiliprotein family, is isolated from red algae. Conjugation of antibodies to R-PE facilitates multiple fluorescent staining methods. In the present study polyclonal antibodies and polyclonal F(ab')2 fragment antibodies were conjugated to R-PE by two different methods. The efficiency of the methods was evaluated using Immunocytochemistry (ICC) and Sodium Dodecyl Sulfate Polyacrylamide Gel Electrophoresis (SDS-PAGE). In the first conjugation method, PE was attached to SMCC linker followed by conjugation of antibody to PE-SMCC. In the second method, SH groups were added onto R-PE molecule, while the antibody was attached to SPDP linker. Then, the antibody-SPDP molecule was conjugated to R-PE. Our results showed that the two conjugation methods did not have any abrogative effects on the antibody binding activity.

Abstract

Objective(s)Rapid tests for detection of Streptococcus agalactiae or Group B Streptococci (GBS) at the onset of labor are needed to permit early intrapartum antibiotic prophylaxis. This study aimed to evaluate the PCR assays targeting the 16S ribosomal RNA gene (16S rDNA) for detection of the GBS in comparison with a specific culture method. Materials and MethodsTwo swabs were used to obtain vaginal specimens from the 330 pregnant women attended delivery room at Hedayat hospital, Tehran, Iran. One swab was analyzed by direct plating onto selective GBS agar medium (ISLAM) and the other swab was used for a PCR assay, which amplified the 16S rDNA of S. agalactiae. Comparative study between the selective culture and the PCR assay was done among the 330 tested women.ResultsThe GBS colonization rate based on the culture results was 20.6% (68/330). Both culture and PCR methods were positive for 56 and negative for 253 women. The culture method was positive and PCR was negative in 12 women. The culture was negative and the PCR positive for 9 women. Sensitivity of the PCR assay was 82.3% and specificity was 96.5%. The positive predictive value was 86.15% and negative predictive value was 95.4%. ConclusionISLAM diagnostic procedure and PCR are rapid and reliable analyzing methods, which might be useful for accurate diagnosis of GBS colonization in pregnant women at the time of delivery.

Abstract

Universal vaccination of neonates and children against diphtheria, tetanus and pertussis has had a tremendous impact on the control of these infectious diseases worldwide. Immunization by the triple diphtheria, tetanus and whole cell pertussis vaccine (DTwP) has been applied in Iran for almost 50 years. Periodic assessment of immunogenicity of this vaccine is an important aspect of successful mass vaccination programs. The present study was performed to assess the antibody response against tetanus, diphtheria and pertussis in a group of Iranian infants vaccinated with a local DTwP vaccine. In this prospective study, 330 infants received primary vaccination at 2, 4 and 6 months of age with DTwP vaccine manufactured by Razi Institute of Iran. Blood samples were taken 2-4 weeks after the third dose to assess seroprotection and geometric mean titers (GMT) of specific antibodies. Among the 283 infants who completed the vaccination course, 98.2% and 100% developed antibodies against diphtheria and tetanus, respectively. The GMT of antibodies to tetanus, diphtheria and pertussis, were 2.09 IU/ml, 2.08 IU/ml and 8.73 EU/ml, respectively. Comparison of the results obtained from this study with those from previous studies performed in other countries revealed a similar GMT and protection rates for diphtheria and tetanus components. In the absence of well-established serological criteria, judgment about protection rate against pertussis has not been possible. A prospective vaccination study using the local DTwP vaccine in parallel to a WHO approved standard vaccine, could enable assessment of immunogenicity of the pertussis component.

Abstract

Spermatogonia are the male germ line stem cells whose life long expansion is needed for permanent production of spermatozoa. The present study was designed to examine the effect of hCG treatment on germ cell proliferation following stem cell transplantation in mice. Spermatogonial stem cells were isolated from neonatal mice testes and characterized by alkaline phosphatase, immunoreactivity and morphological analysis. hCG was injected into normal and cell transplanted mice. We then evaluated the testosterone levels and cell number in normal mice. After that, cyclin B1 gene expression was investigated in transplanted mice. Different doses of busulfan were injected to investigate the effects of chemotherapy on morphological criteria and preparation of recipient mice for transplantation. In this report we show proliferative potential of spermatogonial stem cells after cytotoxic treatment, transplantation efficiency by semi-quantitative RT-PCR, and hCG effect on stem cell regeneration in normal mice and following cell transplantation. The results indicate that spermatogonial stem cells can proliferate after transplantation, and the efficiency of their transplantation depends on hormonal treatment. Therefore, hormonal treatment after stem cell transplantation will be a powerful avenue for increasing the efficiency of transplantation and fertility restoration.
