Context Stress responses in chickens are commonly assessed from measurements of corticosterone in blood, but there is an increasing research effort to develop non-blood means of assessing the activity of the hypothalamo–pituitary (HPA) axis. It is common to measure corticosterone in the eggs and faeces. Aims We extended previous work by undertaking a study of caged laying hens comparing basal concentrations of corticosterone in plasma, faeces, egg albumen and egg yolk on a between-cage basis. We tested the hypothesis that there are positive relationships between corticosterone in plasma and corticosterone in each of the other matrices. Methods Blood samples were collected from each bird at a single point in time. In Experiment 1, these comparisons (between plasma concentrations of corticosterone on Day 1 and egg albumen, egg yolk and faecal concentrations of corticosterone on Days 1, 3 and 4 of the study) were made for hens of two ages under basal conditions, whereas, in Experiment 2, the comparisons (between plasma concentrations of corticosterone on Day 3 and egg albumen, egg yolk and faecal concentrations of corticosterone on Days 1 and 2 of the study) were made for hens housed at different space allowances with and without access to a nest box. The birds without a nest box had not had experience with a nest box prior to sampling. Key results There was a statistically significant (P = 0.012), but limited, positive relationship between plasma and egg albumen concentrations of corticosterone under basal conditions in Experiment 2. There were no other statistically significant (P > 0.05) relationships in either experiment. These results suggest that measures of corticosterone in the albumen, yolk and faeces of laying hens are unlikely to be robust predictors of basal concentrations of corticosterone in the blood. Conclusions Although there was some indication that concentrations of corticosterone in albumen may be related to concentrations in blood under basal conditions, based on all the results, this suggestion is made cautiously. Implications More comprehensive research is required to establish if measures of corticosterone in egg components and faeces are related to chronic basal activity of the hypothalamo–pituitary axis in laying hens. There is also a need to understand the impact of corticosterone on production, reproduction and welfare in hens from measures in both blood and non-blood matrices.

Current research on factors affecting the welfare of dairy calves is predominantly based on indoor, year-round calving systems. Calf rearing in these systems differs from that in more seasonal, pasture-based dairy production, meaning that risks to the welfare of dairy calves may not always be comparable between the two systems. The aim of this review was to consolidate the scientific literature relating to calf welfare in pasture-based dairy systems from birth until weaning, allowing for (1) the identification of current and emerging risks to calf welfare and (2) the formation of recommendations to mitigate these risks. Many of the risks to calf welfare discussed in this review are not exclusive to pasture-based dairies. This includes a global trend for increasing perinatal mortalities, a significant number of calves failing to achieve effective passive transfer of immunity, the low uptake of best practice pain relief when calves are disbudded, and the feeding of restricted milk volumes. In addition to these persisting welfare risks, two factors discussed in this review pose an immediate threat to the social license of dairy farming; the separation of cow and calf soon after birth and the management of surplus calves (i.e. calves not needed by the dairy industry). Several recommendations are made to improve the uptake of best-practice calf rearing and progress the development of alternative pasture-based rearing systems that accommodate changing community expectations. These include communication strategies that strengthen farmer beliefs regarding the welfare and productivity benefits achieved by best practice calf rearing and challenge beliefs regarding the associated costs. Farmers should also be encouraged to benchmark their rearing practices through improved record keeping of key rearing inputs and outcomes. Biological research is needed to advise the development of new calf rearing recommendations and the evolution of existing recommendations. Research priorities identified by this review include the effects of dystocia on the neonate and strategies to mitigate these effects, relationships between features of pen design and calf health and welfare, feasibility of dam rearing in large pasture-based dairy systems, and strategies that increase the value of the surplus calf.

Context Manipulation of milk composition in lactating ewes represents an important mechanism by which sheep breeders can improve profits through increased milk prices for cheese making and higher lamb growth rates. To this end, melatonin implantation may affect milk composition by alleviating reproductive seasonality and augmenting insulin like growth factor-1 (IGF-1). Aims This research aimed to evaluate the effects of melatonin administration on hormone levels and milk composition, and to determine its potential to manipulate milk composition of lactating Barki ewes. Methods Fifty lactating Barki ewes were divided into two groups of 25. One group received 18 mg melatonin in a single implant at 40 days after delivery, and the other group served as an untreated control. Over the following 8 weeks, milk components (fat, protein, lactose and total solids percentages), milk energy value, and blood serum hormones including IGF-1 were measured. Context In extensive grazing sheep systems, pregnant ewes undergo periods of undernutrition because gestation coincides with winter when natural pasture is of lowest quantity and poorest quality. The lamb’s weight and thermoregulatory capacity, and the ewe–lamb bond at birth, may be compromised. Maternal melatonin treatment during gestation may reverse these effects. Aim The aim was to determine the effects of melatonin treatment of single-lambing, undernourished ewes during the last third of gestation on lamb birthweights and body temperatures, and on ewe–lamb interactive behaviour after birth. Methods At Day 100 of gestation, 39 single-bearing ewes received a subcutaneous melatonin implant, and 54 ewes served as controls with no implant. Throughout gestation, the ewes remained under extensive conditions grazing on natural pasture. Measurements were made of lamb birthweight, body temperatures (surface temperature by infrared thermography and rectal temperature), and ewe–lamb behaviours during a handling test at 6–17 h after lambing. Key results There was no effect of melatonin treatment on lamb birthweight or rectal temperature, or on ewe–lamb interaction behaviours. Hip minimum surface temperature was greater in lambs from melatonin-treated ewes than lambs from control ewes (21.2°C ± 0.9°C vs 18.8°C ± 0.8°C; P = 0.05), and there was a similar trend for hip mean surface temperature (24.6°C ± 0.9°C vs 22.3°C ± 0.7°C; P = 0.06). Rump surface temperatures were greater in male than female lambs: maximum (27.9°C ± 1.2°C vs 22.9°C ± 1.2°C; P = 0.01), minimum (22.2°C ± 1.5°C vs 16.7°C ± 1.5°C; P = 0.02) and mean (25.4°C ± 1.3°C vs 20.5°C ± 1.3°C; P = 0.02). Conclusion Melatonin treatment during the last third of ewe pregnancy slightly enhanced the surface temperature of lambs at birth but did not influence ewe–lamb interaction behaviour after birth (i.e. after establishment of the ewe–lamb bond). Implications Further study in more depth is warranted into the possible effects of maternal supplementation with commercial melatonin implants on lamb development, thermoregulatory capacity, behaviour and survival rates in extensive grazing systems, including the effect on ewe–lamb behaviours immediately after birth for both singletons and twins.

Context The use of buserelin, a gonadotropin-releasing hormone (GnRH) agonist, can improve semen quality. Nevertheless, the appropriate dosage, and frequency and time of application, remain to be established owing to the subsequent negative feedback on endogenous production of testosterone described so far. Aims This study aimed to propose a new buserelin application protocol for sperm improvement in Santa Inês rams. Methods Fifteen Santa Inês rams were divided into two experimental groups: the first group (n = 8) received 2.5 μg buserelin acetate; and the second group (n = 7) received saline solution. Treatments were applied for 21 days at intervals of 6 h, totalling four hormonal or saline applications per day. In all animals, macroscopic and microscopic seminal characteristics and freezability, plasma testosterone, pixels of testicular parenchyma, and biometric analysis of testicular and accessory sex glands by ultrasound were evaluated throughout the period of hormonal application and for a further 7 days. Key results Rams receiving buserelin showed a peak of testosterone after the first week (Day 7) of application, with a drop in values at Day 14, followed by negative feedback at Day 21 and return to basal concentrations at Day 28. They also showed improvement of kinetics parameters such as straight line velocity (μm/s: +buserelin 51.5 ± 1.5 > control 46.6 ± 1.6) and linearity coefficient (%: +buserelin 53.5 ± 1.8 > control 47.5 ± 1.9). Ultrasound parameters, pixels of testicular parenchyma, and freezability were not affected by treatment. Conclusions The administration of buserelin was able to improve some sperm kinetics parameters of Santa Inês rams. Implications Low doses of this GnRH analogue can be used; however, further adjustments to the hormonal protocol are needed to obtain an effective improvement in sperm quality.

It is essential to understand responses to stress and the impact of stress on physiological and behavioural functioning of hens, so as to assess their welfare. The current understanding of stress in laying hens is comprehensively reviewed here. Most research on stress in hens has focussed on the activity of the adrenal glands, with the most common approach being to measure corticosterone, which is the predominant glucocorticoid produced by birds in response to stress. While these measures are useful, there is a need to understand how the brain regulates stress responses in hens. A greater understanding of the sympathoadrenal system and its interaction with the hypothalamo–pituitary–adrenal axis is required. There is also a lack of knowledge about the many other peptides and regulatory systems involved in stress responses in hens. The usefulness of understanding stress in hens in terms of assessing welfare depends on appreciating that different stressors elicit different responses and that there are often differences in responses to, and impacts of, acute and chronic stress. It is also important to establish the actions and fate of stress hormones within target tissues. It is the consequences of these actions that are important to welfare. A range of other measures has been used to assess stress in hens, including a ratio of heterophils to lymphocytes and haematocrit : packed cell-volume ratio and measures of corticosterone or its metabolites in eggs, excreta, feathers and the secretions of the uropygial gland. Measures in eggs have proffered varying results while measures in feathers may be useful to assess chronic stress. There are various studies in laying hens to indicate impacts of stress on the immune system, health, metabolism, appetite, and the quality of egg production, but, generally, these are limited, variable and are influenced by the management system, environment, genetic selection, type of stressor and whether or not the birds are subjected to acute or chronic stress. Further research to understand the regulation of stress responses and the impact of stress on normal functioning of hens will provide important advances in the assessment of stress and, in turn, the assessment of welfare of laying hens.

A practice that is used to extend the period of lay of hens is induced moulting. It involves restriction of nutrients, and sometimes manipulation of lighting, with the consequent loss of bodyweight and feathers, regression of the reproductive tract and cessation of egg lay. When the period of moulting is completed, the reproductive tract will regenerate and egg production will resume, thereby extending the overall period of lay. Towards the end of the laying cycle of hens, the production and quality of eggs decline. At this point, some egg farmers will induce a moult in the flocks to reduce bird-replacement costs and improve profitability. In the Australian egg industry, less than 10% of layers are moulted. There are implications for the welfare of hens that are induced to moult, particularly due to loss of bodyweight and feathers. Histopathological studies have shown the extent to which loss of feathers may affect welfare from a physical perspective, and it terms of pain. It is apparent that induced moulting can lead to an increase in aggression and injurious pecking, and birds with poor feather cover are vulnerable to damage from this pecking. Induced moulting can increase stress, although the implications of this for the welfare of hens is not well understood. The endocrine events associated with induced moulting are reasonably well known and it is possible to manipulate the endocrine system to extend the lay of hens without the need to moult. Nevertheless, these approaches are unlikely to be adopted into practice. More importantly, hens should always have feed and water available and are in good condition before undergoing a moult. This is essential from the perspective of ensuring adequate standards of welfare of birds.

This experiment studied the effects of lactation housing systems and human interaction on piglets’ responses to routine stressors. Forty litters of piglets were reared in either a standard farrowing crate (FC) or a loose farrowing and lactation pen (LP, PigSAFE pen) and received either routine contact with humans (C) or regular opportunities for positive human contact (+HC, 3 min of patting, stroking and scratching 5 times/week). Behavioural and physiological responses to routine husbandry procedures, weaning, novelty and humans were studied in addition to effects on piglet growth, injuries and survival. Compared to C piglets, +HC piglets vocalised for shorter durations (p = 0.018) during husbandry procedures and showed a lower intensity of escape behaviour during iron injection (p = 0.042) and oral vaccination (p = 0.026) at 3 d of age, capture at 2 wk of age (p &lt, 0.001), and intramuscular vaccination (p = 0.005) at 3 wk of age. +HC piglets at 2 wk of age were faster than C piglets to approach (p = 0.048) and interact (p = 0.042) with a stationary unfamiliar human. Compared to LP piglets, FC piglets showed a lower intensity of escape behaviour during capture and iron administration by a stockperson at 3 d of age (p = 0.043). FC piglets at 2 wk of age were faster than LP piglets to approach (p = 0.005) and interact (p = 0.027) with a novel object and approach (p = 0.009) and interact (p = 0.008) with an unfamiliar human. FC piglets had fewer injuries than LP piglets at 2 wk of age (p = 0.004). +HC pigs had fewer injuries than C pigs after weaning (p = 0.003). After weaning there were more pigs from LP than FC observed to be upright (both stationary, p = 0.002 and walking, p = 0.024), vocalizing (p = 0.004), nosing another pig (p = 0.035) and nosing the pen floor (p = 0.038). There were no significant effects on neutrophil:lymphocyte ratios or plasma cortisol concentrations 1.5 h after weaning. However, 25 h after weaning +HC pigs had higher haptoglobin concentrations than C pigs (p = 0.002), and C/LP pigs had higher cortisol concentrations than +HC/LP and C/FC pigs (p = 0.012). There were no significant effects on piglet growth, the number of piglets born alive or the number stillborn, however there were more piglets weaned from FC than LP (p = 0.035). The results from this experiment raise questions that require further research on the ability of pigs reared in loose pens to cope with stressors such as exposure to humans, novelty, husbandry procedures and weaning. This experiment also provides evidence that regular positive human interaction reduces pigs’ fear of humans and husbandry procedures imposed by stockpeople. More research is required to determine if any of these effects are sustained long-term.

Little is known about the effect of low-protein (LP) diets on intestinal barrier function and permeability. In the first part of this study, starting on day 9 of age, the growth performance of the birds fed 3 experimental diets in each phase of feeding (G/F: grower/finisher) was investigated. Three experimental diets were as follows: LP (170/150 g/kg CP) fortified with essential amino acids (EAA), standard protein (SP), (202/190 g/kg), and high protein (HP) (220/210 g/kg). LP and SP diets contained a similar level of EAA concentration, while the HP diet contained 10% above the Ross 308 specifications. Each diet was replicated 6 times (10 male birds per replicate). The second part investigated intestinal permeability (IP) and function on additional 72 birds. On days 14, 16, and 20, a total of 36 birds (12 birds per diet) were injected with dexamethasone (DEX) to induce leaky gut. Birds fed LP diets had lower body weight gain (BWG) and higher feed conversion ratio (FCR) compared with SP and HP diets in both grower and finisher phases of feeding. For the challenge part, DEX increased the FCR independent of diets. Diet and DEX interacted for BWG, whereby the effect of diets was only evident in sham-injected birds. Birds fed LP had a higher fluorescein isothiocyanate dextran (FITC-d) concentration indicating a more permeable intestine compared to HP, but similar to SP. DEX increased FITC-d concentration in all dietary treatments. Birds fed LP had less ileal zonula occludens-2 (ZO-2) expression in comparison with SP, but not HP. DEX increased the expression of Claudin3 and ZO-2 and reduced Claudin1 (P < 0.05) and junctional adhesion molecule 2 in the ileum. Expression of ileal Na+-dependent glucose transporter 1 (SGLT1) was upregulated in LP fed group. It is concluded that compared with SP, IP can be maintained in LP by supplementing EAA. However, when compared with HP, feeding birds with LP may lead to a higher IP. DEX had a profound independent effect on intestinal barrier function.

Confinement housing appears to be at the forefront of concern about laying hen welfare. This experiment examined the effects of floor space during rearing (315 or 945 cm2/bird) and adulthood (542 or 1648 cm2/bird) and access to a nest box on the welfare of caged laying hens. Measurements of the normality of biological functioning, such as plasma, egg albumen and yolk and fecal corticosterone concentrations, and heterophil to lymphocyte ratios, behavioral time budgets, mortality and efficiency of productivity, and measurement of hen preferences, such as choice behavior in Y maze tests, were used to assess hen welfare. There were no effects of treatment on physiological measurements. Hens given less space during adulthood spent less time mobile, inedible pecking, drinking, and preening and spent more time resting and feed pecking and sitting (P < 0.05). Hens with access to a nest box spent more time resting (P = 0.046) and less time sham dust bathing (P = 0.044) than hens without access to a nest box. There were no effects of space allowance on choice behavior for space or a nest box over food; however, hens with access to a nest box chose the nest box over food more than hens without access to a nest box (P = 0.0053). The present experiment provides no convincing evidence that either reducing space allowance in adulthood from 1648 to 542 cm2/bird or eliminating access to a nest box results in disruption of biological function. Less space and no access to a nest box did not increase the choice for more space or a nest box, respectively, over food in the preference tests. However, reduced floor space reduced behavioral freedom and denying access to a nest box eliminated the opportunity for the motivated behavior of laying their eggs in a discrete enclosed nest box, both of which presumably provide hens with the opportunity for positive affective experiences.

It costs money to improve the welfare of farm animals. For people with animals under their care, there are many factors to consider regarding changes in practice to improve welfare, and the optimal course of action is not always obvious. Decision support systems for animal welfare, such as economic cost–benefit analyses, are lacking. This review attempts to provide clarity around the costs and benefits of improving farm animal welfare, thereby enabling the people with animals under their care to make informed decisions. Many of the costs are obvious. For example, training of stockpeople, reconfiguration of pens, and administration of pain relief can improve welfare, and all incur costs. Other costs are less obvious. For instance, there may be substantial risks to market protection, consumer acceptance, and social licence to farm associated with not ensuring good animal welfare. The benefits of improving farm animal welfare are also difficult to evaluate from a purely economic perspective. Although it is widely recognised that animals with poor welfare are unlikely to produce at optimal levels, there may be benefits of improving animal welfare that extend beyond production gains. These include benefits to the animal, positive effects on the workforce, competitive advantage for businesses, mitigation of risk, and positive social consequences. We summarise these considerations into a decision tool that can assist people with farm animals under their care, and we highlight the need for further empirical evidence to improve decision-making in animal welfare.

Short day length is associated with reduced milk production in dairy ruminants. Dairy ruminants have been kept in lit sheds during winter to extend the day length and stimulate milk production. However, there studies are few on the effect of an extended photoperiod on the ensuing reproductive performance of dairy goats. The aim of this study was to examine the effect of long day photoperiod (LDPP) and exposure to bucks on milk production and plasma progesterone and prolactin in dairy goats. The study was conducted in 122 non-pregnant lactating dairy goats over 18 weeks from April to August (late autumn and winter in the Southern Hemisphere). The goats were kept in open sided sheds in which the control treatment received ambient lighting while the LDPP treatment received 16 h of light, including artificial lighting. In June, July and August synchronised does were randomly assigned each month to the presence or absence of a buck and ovulatory activity determined from plasma progesterone. Plasma progesterone concentrations were reduced (0.73 vs. 0.46 pmol, p &lt, 0.001) while prolactin concentrations were increased (0.095 vs. 1.33 ng/mL, p &lt, 0.001) in LDPP goats. The former response was most marked in late winter (0.58 vs. 0.004 pmol, p &lt, 0.001) indicating a lack of functional corpora lutea. While there was no overall effect of buck exposure on plasma progesterone concentrations there was a three-way interaction such that plasma progesterone concentrations were increased (p &lt, 0.05) by exposure to bucks in LDPP goats in August (late winter) but not at other times. Milk production was increased in LDPP goats over the latter stages of the study (1. 55 vs. 1.82 L/d, p &lt, 0.05). Also, persistency of lactation was greater in LDPP goats with fewer goats drying off (13 vs. 0%, p &lt, 0.05). These findings suggest that LDPP can increase milk production and persistence while decreasing ovulatory activity in dairy goats.

The physiology and behavior of gilts that develop in a male-biased litter can differ from gilts that develop in a female-biased litter. We hypothesized that gilts from male-biased litters will have a delayed and attenuated luteinizing hormone (LH) surge, and reduced LH pulse frequency and amplitude compared to gilts from female-biased litters. Gilts were selected at birth from male-biased (>60% males n = 10) or female-biased (>60% females n = 9) litters. From 18 wk of age, detection of puberty using daily boar contact began and their subsequent estrous periods were synchronized with oral progestogen (altrenogest). On day 3 after altrenogest withdrawal, blood samples were obtained from 6 gilts per sex bias group at 10 min intervals from 0900 to 2100 h to determine LH pulse amplitude and frequency. From 0900 on day 4, all 19 gilts were sampled every 4 h until the end of estrus to characterize LH surge dynamics. There were no differences between groups in LH pulse characteristics. Compared to gilts from female-biased litters, the LH surge in gilts from male-biased litters was delayed [56.00 ± 3.32 h vs. 43.11 ± 3.76 h (mean ± standard error of the mean (SEM)), P < 0.05], the duration was decreased [29.78 ± 2.12 h vs. 37.71 ± 1.19 h (mean ± SEM), P < 0.05] and the total secretion as measured by area under the curve was decreased (91.42 ± 9.52 ng/mL vs. 120.28 ± 9.48 ng/mL, P < 0.05). Our results indicate that a male-biased uterine environment has different effects on the tonic secretion of LH than the LH surge, with only some elements of the LH surge being affected.

There are sex differences in behaviour with males tending to be more aggressive and less fearful than females. When females are treated with androgens they display more aggressive, male like, behaviours and this suggests that androgen exposure has a role in the development of aggression. Female pigs born into litters that are more than 60% male (male biased) are likely exposed to greater androgen concentrations in utero than female pigs born into litters that are greater than 60% female (female biased). We hypothesise that the proportion of males in the litter in utero affects female pig behaviour. We predict that gilts from male biased litters will be more aggressive and less anxious than gilts from female biased litters. The behaviour of females from male biased (n = 24) and female biased (n = 30) litters was assessed at day 11 and day 21 of life with an arena test, at weaning (28 days of life) by assessing fight number and duration as well as injury scores and at 16 weeks of age with a resident intruder test. In the day 21 arena test, the pigs from female biased litters took longer to emerge into the arena than pigs from male biased litters (1.13 ± 0.05 (42.2 s) vs. 1.32 ± 0.09 (15.5 s) respectively, P = 0.016). At weaning there was a higher scratch score for piglets from male biased litter (0.84 ± 0.2 vs 0.69 ± 0.15, P = 0.015) and this continued as a trend on the day following weaning (1.76 ± 0.10 vs 1.57 ± 0.13, P = 0.080). At 16 weeks of age in the resident intruder test 48% of gilts from male biased litters initiated a fight compared to 24% of gilts from female biased litters (P = 0.08). Our data suggest that the sex ratio of the birth litter can affect behaviour of gilts, potentially having long term implications for sow aggression.

This study was conducted to test the hypothesis that between year variability in reproductive response to melatonin treatment of young Border Leicester (BL) rams in spring was related to sexual maturity at the time of treatment. Two variables of sexual maturity (mating behaviour and semen quality) were examined. In Experiment 1, mating activity of melatonin-treated and untreated rams was examined at two sites before a flock mating at one site. In Experiment 2, testosterone concentrations, semen quality and variables of in vitro fertilising capacity were examined. In Experiment 1, melatonin treatment did not alter sexual behaviour (latency to nose, total nosings, courtings including Flehmen expression, attempted mounts, mounts, ejaculations) with the exception of a lesser (P < 0.05) Flehmen expression at Site 1 whereas values for most variables were greater (P < 0.05) at Site 2. Treatment resulted in a greater pregnancy rate (89 % and 82 %, respectively; P < 0.05) and advanced distribution of pregnancies (P < 0.001) at Site 1. Testicular size and testosterone concentrations were also greater (P < 0.05) as a result of melatonin treatment in spring whilst the reverse occurred in autumn. In Experiment 2, sperm motility and in vitro fertilising capacity were greater (P < 0.05) and acrosome damage less (P < 0.05) as a result of melatonin treatment. In conclusion, variability in age at sexual maturity of young BL rams at spring mating can be reduced with melatonin treatment and is associated with differences in sexual behaviour, testicular growth, testosterone concentrations, and sperm quality.

Floor space is an important determinant of aggression and stress in group-housed sows, and the aim of the present experiment was to comprehensively examine the effects of floor space in the range of 1.45 to 2.90 m/sow from mixing until 27 d after insemination on aggression, stress, and reproduction of group-housed sows. A previous experiment on the effects of floor space indicated spatial variability across and along the research facility in both sow aggression and stress. To minimize this spatial variability within the research facility, similar-sized pens but with varying groups sizes (10-20) in 4 separate blocks of 3 contiguous pens within each of 9 time replicates (180 sows/replicate) were used to examine 6 space allowances (1.45-2.9 m/sow). Space treatments were appropriately randomized to pens. Although it may be argued that space allowance is confounded with group size in this design, there was no evidence in our previous experiment of group size effects, for pens of 10 to 80 sows, or appreciable interactions between space and group size on aggression, stress, and reproduction. In the present experiment, sows were introduced to treatments within 4 d of insemination and were floor fed 4 times per day (2.5 kg/sow per d). On both Days 2 and 26 after mixing, aggressive behavior (bites and knocks) at feeding and plasma cortisol concentrations were measured. Restricted maximum likelihood mixed model analyses were used to examine the treatment effect after accounting for replicate and random spatial location effects within replicate. There was a consistent linear effect of floor space allowance on aggression at feeding at Day 2 ( < 0.0001) and plasma cortisol concentrations at Day 2 ( = 0.0003), with aggression and stress declining with increasing space. However, there were no effects of space allowance on aggression and stress at Day 26 ( = 0.14 and = 0.79, respectively). These results show that increased floor space in the immediate post-mixing period reduces aggression and stress and that sows may adapt to reduced floor space over time. A strategy of staged-gestation penning, with more space immediately after mixing and less space later in gestation, may address both animal welfare and economic considerations, but this clearly requires further examination.

Two key hypotheses emerge in the literature regarding the impact of stress on reproduction in females of any species. First, prolonged stress impairs reproduction in females. Secondly, acute stress impairs reproduction, if it occurs at a critical time during the precisely timed series of endocrine events that induce oestrus and ovulation. We reviewed studies conducted in female pigs to find support or opposition for these hypotheses in female pigs. We also considered the role of cortisol. We found confirmation that prolonged stress or the prolonged elevation of cortisol can impair reproductive processes in female pigs, but also found that there appear to be some female pigs in which reproduction is resistant to such treatments. Reproduction in female pigs appears to be resistant to acute or repeated acute stress or elevation of cortisol, even if these occur during the series of precisely timed endocrine events that induce oestrus and ovulation. Thus, we propose modified versions of the above hypotheses that are specific to female pigs. Furthermore, while cortisol may mediate the effects of prolonged stress on reproduction in female pigs, there is evidence that, in female pigs, ACTH may require the presence of the adrenal glands to impair reproduction rather than having direct effects.

In this review, we discuss animal welfare as a complex and contested issue facing society and outline why collaborative, multi-stakeholder approaches are critical for effective policy development. Using the lens of “wicked problems” and drawing upon governance literature on policy networks, we identify important factors for working with the inherent complexity of animal welfare through the inclusion of various stakeholder perspectives. We present two case studies that illustrate policy network approaches to animal welfare and highlight the value of fostering collaboration among various stakeholder groups from the industry, community, research, and government sectors. We suggest that the influence of stakeholder networks will likely increase in coming years as newer forms of participatory governance become common. By understanding how collaborative stakeholder networks establish participatory governance, productive communication, and collective priorities, leaders in the field of animal welfare can more productively engage with stakeholders and achieve long-lasting improvements in animal welfare.

The roles of testicular hormones in the negative feedback regulation of the secretion and actions of GnRH in male domestic ruminants are reviewed, concentrating mainly on research conducted with rams. Testicular steroids have major feedback actions directly at the hypothalamus to inhibit the secretion of GnRH, although it is apparent that, under certain circumstances, the steroids also have actions directly at the pituitary gland. Further research is necessary to delineate these actions and to determine the contribution of testosterone and its primary metabolites, dihydrotestosterone and oestradiol, to negative feedback on the hypothalamo-pituitary unit. Since GnRH neurones do not possess receptors for steroids, testicular steroids must evoke other neuronal pathways to influence GnRH producing neurones. While the opioids may be important in this regard, it is necessary to determine which other neuronal pathways may also be involved to understand fully the mechanism of action of testicular steroids. It appears that the feedback regulation of the secretion of LH can be accounted for by testicular steroids, whereas the secretion of FSH is influenced by inhibin and steroids, and possibly the recently isolated proteins follistatin and activin. The actions of inhibin to suppress the secretion of FSH occur at the pituitary gland and not on the synthesis or secretion of GnRH. There is a complex interaction between testosterone and inhibin in the control of FSH secretion that results in synergistic effects during the non-breeding season but not during the breeding season. Activin has been shown to have FSH-stimulating properties and follistatin has been shown to have FSH-inhibiting properties, but it is unknown if these proteins play a physiological role in the feedback regulation of FSH in male domestic ruminants.

Understanding the mechanisms by which the body suppresses stress responses, as commonly occurs during lactation in a range of species, would provide the means to develop treatments and strategies to reduce stress in animals, especially farm animals. This would result in improvements in welfare and productivity by eliminating the detrimental impacts of stress. There has been substantial research to demonstrate that frontline stress responses are reduced during lactation, to highlight the importance of the salience of the stressor in these responses, and to identify many of the fundamental mechanisms and regions of the brain involved. Nevertheless, the extension of this knowledge to develop the means to reduce stress in the field are lacking and are required. Translation of this knowledge will lead to important benefits in terms of improved welfare and productivity in farm animals.

Simple Summary Rapidly expanding global human population has led to increased supply chain demands on animal-based farming systems and the desire for environmentally friendly products. This has also resulted because of socio-political pressure and increased public concerns over the impacts of conventional agriculture on the environment. In order to be sustainable, animal production systems must also advance animal welfare, avoiding physically and psychologically stressful situations for the animals and apply innovative methods of reducing contribution of farming practices to global climate change while also functioning at optimum productivity. Consequently, to achieve a practical and effective improvement towards environmental sustainability, animal-based agriculture should consider animal welfare assessment, objective measures of physiological stress, climate change evaluation and animal productivity in a multi-dimensional and holistic approach. Abstract In this retrospective study, we conducted a desktop-based analysis of published literature using the ScienceDirect™ search engine to determine the proportion of livestock research within the last 7 years (2015–2021) that have applied animal welfare assessment combining objective measures of physiological stress and evaluation of climate change factors in order to provide an account of livestock productivity. From the search results, 563 published articles were reviewed. We found that the majority of the literature had discussed animal production outcomes (n = 491) and animal welfare (n = 453) either individually or in conjunction with another topic. The most popular occurrence was the combination of animal welfare assessment, objective measures of stress physiology and production outcomes discussed collectively (n = 218). We found that only 125 articles had discussed the impact of climate change (22.20%) on livestock production and/or vice versa. Furthermore, only 9.4% (n = 53) of articles had discussed all four factors and published research was skewed towards the dairy sector. Overall, this retrospective paper highlights that although research into animal welfare assessment, objective measures of stress and climate change has been applied across livestock production systems (monogastrics and ruminants), there remains a shortfall of investigation on how these key factors interact to influence livestock production. Furthermore, emerging technologies that can boost the quantitative evaluation of animal welfare are needed for both intensive and extensive production systems.

There is increasing interest in using nonblood measures of glucocorticoids to assess the physiological response to chronic stress conditions. In sheep, cortisol has been measured in various matrices including saliva, feces, and wool, but comprehensive studies of the relationship between plasma concentrations of cortisol and concentrations in these nonblood matrices are lacking. Therefore, we tested the hypothesis that administration of cortisol to sheep would result in elevated concentrations of cortisol in blood, saliva, feces, and wool. Merino ewes were administered with saline or 2 mg/kg BW/d hydrocortisone acetate (HCA) by intramuscular (i.m.) injection for 28 d. This treatment was imposed to mimic circulating cortisol concentrations experienced during periods of chronic stress. Cortisol and cortisone were directly measured in plasma, saliva, and wool before, during, and after treatment with saline or HCA. A 14-d pre-treatment and a 14-d post-treatment period were used to measure time taken for glucocorticoid concentrations in each of the matrices to return to baseline levels. Cortisol was also measured in feces before, during, and after treatment. Wool growth was also measured. Before treatment, there was no difference in the concentration of cortisol or cortisone in plasma, saliva, feces, or wool in animals treated with saline or HCA. In contrast, treatment with HCA increased (P < 0.05) concentrations of both cortisol and cortisone in plasma, saliva, and wool and of cortisol in feces. In plasma, cortisol concentrations were higher than cortisone (P < 0.05), whereas saliva cortisol and cortisone concentrations did not differ significantly. In wool, the concentration of cortisone was about 19-fold higher than that of cortisol during treatment and post-treatment periods. Treatment with HCA inhibited wool growth. These results demonstrate that an increase in glucocorticoids in the blood of sheep is reflected in increases in saliva (after 7 d of treatment), feces (21 d), and wool (14 d). Therefore, measures of glucocorticoids in these matrices may provide a measure of activation of the adrenal glands over time in sheep, thereby providing a retrospective indicator of chronic stress. With respect to wool, it appears that cortisol is predominantly metabolized to cortisone in the skin or wool follicle and is stored as cortisone. Therefore wool cortisone may also provide an important measure in quantifiying chronic stress in sheep.

The aim of this study was to investigate the effect of sow confinement and nonconfinement during parturition and early lactation on the performance of sows and piglets prior to weaning. Sows and their litters were housed in either a conventional farrowing crate (control; 1.7 × 2.4 m) or a swing-sided pen (2.8 × 2.15 m). Sows housed in the swing-sided pen had the crate open (open) or closed (closed) during farrowing and opened on d 3 (LAC3) or 7 (LAC7) of lactation. Data are means ± SEM. The numbers of total and live-born piglets were not different between treatments and averaged 13.1 ± 0.4 and 12.3 ± 0.2 piglets/litter, respectively. Sows housed in a farrowing crate had a greater ( = 0.03) number of stillborn piglets compared with open sows from pens (1.1 ± 0.2 vs. 0.6 ± 0.1 piglets/litter, respectively). Live-born piglet mortality before litter equalization on d 1 was greater ( 0.05). Total live-born piglet mortality from birth until weaning for the control, closedLAC7, closedLAC3, openLAC7, and openLAC3 was 1.86 ± 0.19, 1.95 ± 0.23, 2.10 ± 0.39, 2.45 ± 0.27, and 3.74 ± 0.48, piglets/litter, respectively. Total live-born piglet mortality from birth until weaning was greater in the openLAC3 and openLAC7 treatments compared to control ( < 0.01), and openLAC3 was also greater than openLAC7, closedLAC3, and closedLAC7 ( < 0.01). The current data indicated that reducing sow confinement during parturition decreased stillborn number but increased live-born piglet mortality compared to a farrowing crate, and removing confinement on d 3 of lactation further increased total piglet mortality compared to removing sow confinement on d 7 of lactation in sows that farrowed open. Sows that were confined during farrowing and until d 3 or 7 of lactation in a swing-sided pen had a similar piglet mortality compared to those in a farrowing crate.

Activation of the hypothalamo-pituitary-adrenal (HPA) axis by psychosocial stress is attenuated during lactation. We tested the hypothesis that lactating ewes will have attenuated HPA axis responses to isolation and restraint but will have greater responses to predator stress in the form of barking dogs. We imposed two 4 h stressors: psychosocial stress (isolation and restraint of ewes) and predator stress (barking dogs). Blood was collected intravenous every 10 min from nonlactating ewes (n = 6), lactating ewes with lambs present but not able to be suckled (n = 6), and lactating ewes with lambs present and able to be suckled (n = 6). Plasma cortisol and oxytocin were measured. For nonlactating ewes, cortisol increased (P < 0.01) in response to both stressors, and these increases were greater (P < 0.01) than that in the lactating animals. For lactating ewes with lambs present but unable to be suckled, cortisol increased (P < 0.05) in response to both stressors with a greater response to barking dogs (P < 0.05). For lactating ewes with lambs present and able to be suckled, cortisol increased (P < 0.01) in response to barking dogs only. Plasma oxytocin was greater (P < 0.01) in lactating ewes than in nonlactating ewes and did not change in response to the stressors. In conclusion, lactating ewes are likely to have a greater HPA axis response to a stressor that may be perceived to threaten the welfare of themselves and/or their offspring. The role of oxytocin in attenuation of the HPA axis to stress in sheep is unclear from the current research and requires further investigation., Highlights • The response of the hypothalamo-pituitary-adrenal axis to stress is attenuated during lactation in sheep. • The extent of this attenuation is influenced by the opportunity to suckle lambs and the type of stressor. • Stressors that pose an immediate threat to the offspring evoke greater stress responses.

Anogenital distance (AGD) has been used to reflect masculinization in litter-bearing species. As masculinization affects behavior and reproduction, AGD could be measured to assist in selecting gilts with a temperament more suited to commercial production and greater reproductive potential. We hypothesized that gilts from a male-biased litter would have a longer AGD and poorer reproductive performance. In Exp. 1, AGD and weight were measured at day 1, day 21, and week 16 of age for gilts from male-biased litters (≥60% males; n = 51) and female-biased litters (≥60% females; n = 51). Sow AGD was measured 3 d after farrowing. In Exp. 2, AGD was measured at gilt selection at approximately 24 wk of age and gilts followed to second parity. Litter sex ratio affected AGD at 16 wk of age, with gilts from female-biased litters having longer AGD (mean ± SEM, 9.1 ± 0.7 mm vs. 11.0 ± 0.6 mm, P = 0.013). Anogenital distance was not different on day 1 or day 21. There was no effect of sex ratio on weight at any time, and sow AGD was not associated with the sex ratio of her litter. Gilts with an AGD longer than the mean of 11.55 mm were heavier (mean ± SEM, 118.8 ± 0.4 kg vs. 117.7 ± 0.4 kg, P = 0.023), were achieved puberty earlier (179.6 ± 0.6d vs. 182.2 ± 0.6 d, P = 0.001), were mated younger (200.6 ± 0.6 d vs. 203.2 ± 0.6 d, P = 0.001), and were more likely to be mated (91% vs. 83%, P = 0.005) than gilts with an AGD shorter than the mean. Gilts with an AGD greater than 11.55 mm had a greater born alive litter size (11.79 ± 0.20 vs. 11.20 ± 0.19, P = 0.018) compared with gilts with an AGD shorter than 11.55 mm. At 16 wk, AGD was associated with sex bias and could be used as a selection tool to predict reproductive success of the first parity, with a longer AGD being associated with gilts that had been born into a female-biased litter and that had better reproductive performance.

The physiology and behavior of gilts that develop in a male-biased litter can differ from gilts that develop in a female-biased litter. We hypothesized that gilts from male-biased litters will have a delayed and attenuated luteinizing hormone (LH) surge, and reduced LH pulse frequency and amplitude compared to gilts from female-biased litters. Gilts were selected at birth from male-biased (>60% males n = 10) or female-biased (>60% females n = 9) litters. From 18 wk of age, detection of puberty using daily boar contact began and their subsequent estrous periods were synchronized with oral progestogen (altrenogest). On day 3 after altrenogest withdrawal, blood samples were obtained from 6 gilts per sex bias group at 10 min intervals from 0900 to 2100 h to determine LH pulse amplitude and frequency. From 0900 on day 4, all 19 gilts were sampled every 4 h until the end of estrus to characterize LH surge dynamics. There were no differences between groups in LH pulse characteristics. Compared to gilts from female-biased litters, the LH surge in gilts from male-biased litters was delayed [56.00 ± 3.32 h vs. 43.11 ± 3.76 h (mean ± standard error of the mean (SEM)), P < 0.05], the duration was decreased [29.78 ± 2.12 h vs. 37.71 ± 1.19 h (mean ± SEM), P < 0.05] and the total secretion as measured by area under the curve was decreased (91.42 ± 9.52 ng/mL vs. 120.28 ± 9.48 ng/mL, P < 0.05). Our results indicate that a male-biased uterine environment has different effects on the tonic secretion of LH than the LH surge, with only some elements of the LH surge being affected.

This study directly compared the dynamics of change in plasma corticosterone concentration with the dynamics of change in tissue corticosterone concentration in laying hens. In concert, we measured the rate of gluconeogenesis, glycogenesis, and glycolysis in the liver, kidney, skeletal muscle, and heart. We evaluated these changes acutely, over 3 h in response to an adrenocorticotropic hormone (ACTH) injection, and chronically, over 24 h in response to food and water deprivation. In response to ACTH injection, there was a significant (P < 0.05) increase in plasma corticosterone concentration and a parallel significant increase in corticosterone concentration in the skeletal muscle, kidney, and heart. However, the change in corticosterone concentration in the liver did not parallel the plasma, at times it was greater than the plasma, and there was a second significant increase (P < 0.05) in corticosterone concentration in the liver after 180 min. Under these conditions, the rate of gluconeogenesis in the liver decreased and the rate of glycogenesis increased. In contrast, after 12 h and 24 h of food and water deprivation plasma corticosterone concentration was increased, and this was paralleled by increased corticosterone concentration in the liver, an increase in the rate of gluconeogenesis and a decrease in the rate of glycogenesis. After ACTH injection, glucose concentration in the liver was not significantly depleted but after 12 h or 24 h of food and water deprivation it was significantly depleted (P < 0.05). Plasma corticosterone concentration provided different insight into the effect of the stressor on hen physiology under acute and chronic conditions. Our data suggest that extending our evaluation of stress to the site of corticosterone action, that is, the target tissue, may enhance our ability to evaluate stress and the welfare of laying hens.

We tested the hypothesis that overweight/obese men aged 50–70 years will have a greater salivary cortisol, salivary alpha amylase and heart rate (HR) responses to psychological stress compared with age matched lean men. Lean (BMI=20–25 kg/m2; n=19) and overweight/obese (BMI=27–35 kg/m2; n=17) men (50–70 years) were subjected to a well-characterised psychological stress (Trier Social Stress Test, TSST) at 1500 h. Concentrations of cortisol and alpha amylase were measured in saliva samples collected every 7–15 min from 1400 to 1700 h. HR was recorded using electrocardiogram. Body weight, BMI, percentage body fat, resting systolic and diastolic blood pressure and mean arterial pressure were significantly higher (PP=0.187, P=0.288, P=0.550, respectively). There were no significant differences between the groups for pretreatment values, peak height, difference between pretreatment values and peak height (reactivity) or area under the curve for salivary cortisol, salivary alpha amylase or HR (P>0.05 for all). The results showed that, for men with a moderate level of overweight/obesity who were otherwise healthy, the response of salivary cortisol, salivary alpha amylase and HR to acute psychological stress was not impaired.

In several species, females masculinised by abnormal androgen exposure in utero have poor reproductive performance and gilts born into litters with a male bias are likely exposed to greater androgen concentrations prenatally than gilts born into female-biased litters. At 24 h of age, piglet plasma testosterone concentrations in gilts from male-biased litters (>60% male; n = 22) or female-biased litters (>60% female; n = 27) were not different. At 18 wks of age, all gilts received an injection of 400 IU equine chorionic gonadotrophin plus 200 IU human chorionic gonadotrophin to stimulate oestrus. Two weeks after the injection gilts were slaughtered and ovaries collected for determination of numbers of corpora lutea (CL). Compared to gilts from female-biased litters, gilts from male-biased litters were more likely to ovulate (86.0% vs 59.5%, P = 0.047) and had more CL (13.1 ± 1.5 vs 7.2 ± 1.7, P = 0.015). The present data indicate an effect of birth litter sex-bias on pre-pubertal physiological development, possibly involving organisational effects at the ovarian cellular level impacting on future ovarian function. Potential impacts on subsequent fertility remain to be determined.

A total of 3,120 sows, in 4 time replicates, were used to determine the effects of group size and floor space on sow welfare using behavioral, physiological, health, and fitness variables. Within 1 to 7 d postinsemination, sows were assigned randomly to treatments of a 3 by 6 factorial arrangement, with 3 group sizes (10, 30, or 80 sows/pen) and 6 floor space allowances (1.4, 1.8, 2.0, 2.2, 2.4, or 3.0 m(2)/sow). Sows were housed on partially slatted concrete floors, and overhead feeders delivered 4 times/day to provide a total of 2.5 kg of feed/sow. As pen space increased from 1.4 to 3.0 m(2)/sow, aggression at feeding decreased from about 9 to 7 bouts/sow (linear, P = 0.029) and plasma cortisol concentrations decreased from about 28 to 21 ng/mL (linear, P = 0.0089) at 2 d. Although the results are in accord with a linear decline from 1.4 to 3 m(2)/sow, the results are also in accord with a decline in these measurements from 1.4 to 1.8 m(2)/sow and no further decline greater than 1.8 m(2)/sow. Farrowing rate (percentage of inseminated sows that farrowed) also increased from about 60 to 75% as space increased from 1.4 to 3.0 m(2)/sow (linear, P = 0.012). Group size was related to skin injuries on d 9 (P = 0.0017), 23 (P = 0.0046), and 51 (P = 0.0006), with groups of 10 consistently having the lowest number of total injuries over this period. Based on the aggression and cortisol results, it is credible to judge that, within the range of floor space allowances studied, sow welfare improves with increased space. However, from a sow welfare perspective, the experiment had insufficient precision to determine what is an adequate space allowance for sows. Thus, although the results definitely support a space allowance of 1.4 m(2)/sow being too small, it is not possible to give guidance on an actual space allowance at mixing that is adequate.

Short day length (i.e. winter) is associated with reduced milk yield from dairy animals. Dairy cows and sheep have been successfully housed under lights throughout winter to extend the day length and increase milk yield, very little data exists on the effect of extended day length on the milk yield of dairy goats. The current study was conducted over an eight week period from June to August, 2009. A total of 542 Saanan and Saanan British Alpine cross goats were used, all producing >= 1.5 L of milk per day, 253 in early lactation (5-20 days in milk) and 289 in late lactation (190-210 days in milk). Does were randomly allocated to either the control or the long day photoperiod (LDPP) groups, both were housed in open sided sheds with the LDPP group exposed to 16h of light, while the other received a natural lighting regime. Milking was conducted by machine and yield was automatically recorded daily. Doe weights were measured at the beginning of the study and subsequently at four-week intervals. Milk and blood samples were collected from a subset (n=30 and n=18 respectively) of animals in each of the four groups, at day 0 and then every four weeks. Milk was analysed for protein, lactose, bulk milk cell count (SCC), fat and solids-not-fat. Blood plasma was assayed for glucose, non-esterified fatty acids (NEFA) and prolactin. Treatment with LDPP significantly increased (P < 0.001) milk yield, with the greatest response occurring in late lactation animals, who were producing 20% more milk than their control counterparts (+0.28 kg/d). An increase in milk yield was evident by week 1 in late lactation and week 4 in early lactation. Overall, goats exposed to LDPP showed a decrease in milk fat % (P=0.003), milk protein % (P < 0.001), milk solids not fat (P < 0.001) and somatic cell count (P=0.012), but a slightly higher milk lactose % (P=0.010), when compared to control animals. LDPP increased milk protein (P=0.010), lactose (P=0.032) and SNF yield (P=0.034) in late lactation animals with the response being greater after 8 weeks than after 4 weeks of treatment. Plasma prolactin (P=0.007) and glucose increased with exposure to LDPP while the same animals showed a decrease in plasma NEFA. Liveweight increased (P < 0.001) with LDPP exposure, was greater (P < 0.001) in late lactation than in early lactation and increased (P < 0.001) with week of treatment. Exposure to LDPP significantly increased milk yield and lactation persistence in Australian dairy goats, most substantially in late lactation. (C) 2013 Published by Elsevier B.V.

Glucocorticoids (corticosterone in birds and rodents and cortisol in all other mammals) are glucoregulatory hormones that are synthesized in response to a range of stimuli including stress and are regularly measured in the assessment of animal welfare. Glucocorticoids have many normal or non-stress-related functions, and glucocorticoid synthesis can increase in response to pleasure, excitement, and arousal as well as fear, anxiety, and pain. Often, when assessing animal welfare, little consideration is given to normal non-stress-related glucocorticoid functions or the complex mechanisms that regulate the effects of glucocorticoids on physiology. In addition, it is rarely acknowledged that increased glucocorticoid synthesis can indicate positive welfare states or that a stress response can increase fitness and improve the welfare of an animal. In this paper, we review how and when glucocorticoid synthesis increases, the actions mediated through type I and type II glucocorticoid receptors, the importance of corticosteroid-binding globulin, the role of 11 β-hydroxysteroid dehydrogenase, and the key aspects of neurophysiology relevant to activating the hypothalamo-pituitary-adrenal axis. This is discussed in the context of animal welfare assessment, particularly under the biological functioning and affective states frameworks. We contend that extending the assessment of animal welfare to key brain regions afferent to the hypothalamus and incorporating the aspects of glucocorticoid physiology that affect change in target tissue will advance animal welfare science and inspire more comprehensive assessment of the welfare of animals.

Neuroadrenergic function in type 2 diabetic (T2D) patients without neuropathy is poorly characterized. We therefore compared sympathetic nervous system activity at rest and during an oral glucose tolerance test in obese metabolic syndrome (MetS) subjects classified as glucose intolerant (impaired glucose tolerance [IGT]; n = 17) or treatment-naive T2D (n = 17). Untreated subjects, matched for age (mean 59 ± 1 year), sex, BMI (32.4 ± 0.6 kg/m2), and family history of diabetes were studied. We measured resting muscle sympathetic nerve activity (MSNA) by microneurography, whole-body norepinephrine kinetics by isotope dilution, insulin sensitivity by euglycemic-hyperinsulinemic clamp (steady-state glucose utilization adjusted for fat-free mass and steady-state insulin concentration [M/I]), and MetS components. T2D subjects had higher resting MSNA burst incidence (67 ± 4 versus 55 ± 3 bursts per 100 heartbeats; P = 0.05) and arterial norepinephrine levels (264 ± 33 versus 167 ± 16 pg/mL; P = 0.02), lower plasma norepinephrine clearance (by 17%; P = 0.03), and reduced neuronal reuptake compared with IGT subjects (by 46%; P = 0.04). Moreover, norepinephrine spillover responses to glucose ingestion were blunted in T2D subjects. The M/I value independently predicted whole-body norepinephrine spillover (r = −0.47; P = 0.008), whereas fasting insulin level related to neuronal norepinephrine reuptake (r = −0.35, P = 0.047). These findings demonstrate that progression to T2D is associated with increased central sympathetic drive, blunted sympathetic responsiveness, and altered norepinephrine disposition.

This review focuses on the importance of cortisol in mediating the inhibitory effects of psychosocial stress on reproduction in females. In particular, we have summarized our research in sheep where we have systematically established whether cortisol is both sufficient and necessary to suppress reproductive hormone secretion and inhibit sexual behaviour. Our findings are put into context with previous work and are used to develop important concepts as well as to identify productive further lines of investigation. It is clear that cortisol is necessary to inhibit some, but not all, aspects of reproduction in female sheep. These actions vary with reproductive state, and there are important interactions with gonadal steroids. The impact of cortisol on the tonic secretion of gonadotrophin-releasing hormone and luteinizing hormone has been investigated extensively, but less is known about the surge secretion of these hormones and their effects on sexual behaviour. Furthermore, there are separate effects of cortisol in the brain (hypothalamus) and at the anterior pituitary, illustrating that there are different mechanisms of action. Thus, although cortisol is important in mediating some of the effects of stress on reproduction, we need to look beyond cortisol and investigate some of the other mechanisms and mediators that relay the effects of stress on reproduction. In this regard, we propose that a group of neurons in the hypothalamus that co-synthesize kisspeptin, neurokinin B and dynorphin, termed KNDy cells, play important roles in mediating the effects of cortisol on reproduction. This hypothesis needs to be rigorously tested.

Aim: Insulin resistance and visceral adiposity are predisposing factors for fatty liver disease. The main objectives of this study were (i) to compare the effects of caloric restriction (CR) alone or together with moderate-intensity aerobic exercise training (CR+EX) on liver enzymes, a surrogate marker of liver injury, in obese metabolic syndrome (MetS) subjects and (ii) to identify anthropometric, metabolic, cardiovascular and dietary predictors of changes in liver enzymes. Methods: Sedentary men and women (n = 63), aged 55 ± 6 (s.d.) years with body mass index 32.7 ± 4.1 kg/m and confirmed MetS, were randomized to 12-week CR, CR+EX or no treatment (Control). Results: Weight loss averaged 7.6% in the CR and 9.1% in the CR+EX group (time effect, p < 0.001; group effect, p = 0.11); insulin sensitivity improved by 49 and 45%, respectively (both p < 0.001). Fitness (maximal oxygen consumption) increased by 19% in the CR+EX group only (p < 0.001). Alanine aminotransferase (ALT) levels decreased by 20% in the CR and 24% in the CR+EX group (time effect, both p < 0.001; group effect, p = 0.68); corresponding values for γ-glutamyltransferase (GGT) were -28 and -33%, respectively (time effect, both p < 0.001; group effect, p = 0.28). Reduction in abdominal fat mass (measured by DXA from L1 to L4) independently predicted ΔALT (r = 0.42, p = 0.005) and ΔGGT (r = 0.55, p < 0.001), whereas change in dietary saturated fat intake was independently associated with ΔALT (r = 0.35, p = 0.03). Conclusions: Reductions in central adiposity and saturated fat intake are key drivers of improvement in liver enzymes during lifestyle interventions. Exercise training did not confer significant incremental benefits in this study.

It is well known that stress inhibits normal reproductive function, including gonadotrophin secretion; however, the mechanisms and mediators involved are largely unknown. Stress impairs the secretion of luteinising hormone (LH), and it has been suggested that the RF-amide gonadotrophin-inhibitory hormone (GnIH), known as RF-amide related peptide-3 (RFRP-3) in mammalian species, may mediate this inhibitory effect of stress. If this is the case, the GnIH/RFRP system would likely be up-regulated during stress. We tested this hypothesis in ovariectomised ewes using a psychosocial stressor: isolation/restraint. Ewes were randomly allocated to control or stress (n=5 per group). Isolation/restraint stress was imposed for 90min after control sampling for 4h, whereas control ewes were sampled continuously for 5.5h. All ewes were then euthanased and brains were collected. Obesity is fast becoming the scourge of our time. It is one of the biggest causes of death and disease in the industrialized world, and affects as many as 32% of adults and 17% of children in the USA, considered one of the world's fattest nations. It can also cost countries billions of dollars per annum in direct and indirect care, latest estimates putting the USA bill for obesity-related costs at $147 billion in 2008. It is becoming clear that the pathophysiology of obesity is vastly more complicated than the simple equation of energy in minus energy out. A combination of genetics, sex, perinatal environment and life-style factors can influence diet and energy metabolism. In this regard, psychological stress can have significant long-term impact upon the propensity to gain and maintain weight. In this review, we will discuss the ability of psychological stress and ultimately glucocorticoids (GCs) to alter appetite regulation and metabolism. We will specifically focus on (i) GC regulation of appetite and adiposity, (ii) the apparent sexual dimorphism in stress effects on obesity and (iii) the ability of early life stress to programme obesity in the long term.

Five experiments were conducted to test the hypothesis that psychosocial stress interferes with the estrous cycle of sheep. In experiment 1, ewes were repeatedly isolated during the follicular phase. Timing, amplitude, and duration of the preovulatory luteinizing hormone (LH) surge were not affected. In experiment 2, follicular-phase ewes were subjected twice to a “layered stress” paradigm consisting of sequential, hourly application of isolation, restraint, blindfold, and predator cues. This reduced the LH pulse amplitude but did not affect the LH surge. In experiment 3, different acute stressors were given sequentially within the follicular phase: food denial plus unfamiliar noises and forced exercise, layered stress, exercise around midnight, and transportation. This, too, did not affect the LH surge. In experiment 4, variable acute psychosocial stress was given every 1–2 days for two entire estrous cycles; this did not disrupt any parameter of the cycle monitored. Lastly, experiment 5 examined whether the psychosocial stress paradigms of experiment 4 would disrupt the cycle and estrous behavior if sheep were metabolically stressed by chronic food restriction. Thirty percent of the food-restricted ewes exhibited deterioration of estrous cycle parameters followed by cessation of cycles and failure to express estrous behavior. However, disruption was not more evident in ewes that also encountered psychosocial stress. Collectively, these findings indicate the estrous cycle of sheep is remarkably resistant to disruption by acute bouts of psychosocial stress applied intermittently during either a single follicular phase or repeatedly over two estrous cycles.

Lifetime reproductive performance and behaviour are established in utero driven by genetics and the steroidal milieu of the uterine environment. Developing males synthesise androgens that can be absorbed by females in the litter. Consequently, the proportion of males in a litter has the potential to affect both lifetime reproductive performance and behaviour of female littermates. Although reproductive potential is understandably important for gilt selection, behaviour is becoming progressively more important as group housing of sows increases. Aggression can be a cause for removal from the breeding herd, and can also negatively affect reproductive performance and herd longevity. This review covers existing evidence of masculinisation in rodents and sheep, the mechanism causing masculinisation, and the knowledge gaps surrounding masculinisation in pigs. Premature culling of gilts is an economic and animal welfare issue in the pork industry resulting in low retention of gilts and reduced productivity. The selection of gilts needs to be improved to incorporate both lifetime reproductive potential and behaviour. We determined that there is cause to investigate the effect of the birth litter sex ratio of a gilt on her suitability for selection into the breeding herd.
