Intracranial pressure (ICP) is a major neurological parameter in animals and humans. ICP is a function of the relationship between the contents of the cranium (brain parenchyma, cerebrospinal fluid, and blood) and the volume of the skull. Increased ICP can cause serious physiological effects or even death in patients who do not quickly receive proper care, which includes ICP monitoring. Epilepsies are a set of central nervous system disorders resulting from abnormal and excessive neuronal discharges, usually associated with hypersynchronism and/or hyperexcitability. Temporal lobe epilepsy (TLE) is one of the most common forms of epilepsy and is also refractory to medication. ICP characteristics of subjects with epilepsy have not been elucidated because there are few studies associating these two important neurological factors. In this work, an invasive (ICPi) and the new minimally invasive (ICPmi) methods were used to evaluate ICP features in rats with chronic epilepsy, induced by the experimental model of pilocarpine, capable of generating the main features of human TLE in these animals.

Mossy fiber sprouting (MFS) is a frequent finding following status epilepticus (SE). The present study aimed to test the feasibility of using manganese-enhanced magnetic resonance imaging (MEMRI) to detect MFS in the chronic phase of the well-established pilocarpine (Pilo) rat model of temporal lobe epilepsy (TLE).To modulate MFS, cycloheximide (CHX), a protein synthesis inhibitor, was coadministered with Pilo in a subgroup of animals. In vivo MEMRI was performed 3 months after induction of SE and compared to the neo-Timm histologic labeling of zinc mossy fiber terminals in the dentate gyrus (DG).Chronically epileptic rats displaying MFS as detected by neo-Timm histology had a hyperintense MEMRI signal in the DG, whereas chronically epileptic animals that did not display MFS had minimal MEMRI signal enhancement compared to nonepileptic control animals. A strong correlation (r = 0.81, p0.001) was found between MEMRI signal enhancement and MFS.This study shows that MEMRI is an attractive noninvasive method for detection of mossy fiber sprouting in vivo and can be used as an evaluation tool in testing therapeutic approaches to manage chronic epilepsy.

Adiabatic pulses are sometimes considered to be mysterious and exotic entities which are difficult to understand, complex to generate and impractical to implement. This work is an attempt to bring familiarity and to fulfill the preliminary needs of anyone interested in learning more about this subject. The response of magnetization to stimuli produced by adiabatic pulses is analyzed using vector representations in a frequency modulated rotating frame. The first section deals with basic principles of amplitude and frequency modulated pulses and a vector representation in a second rotating frame is used to explain how the adiabatic condition can be satisfied. The subsequent section explains the principles of offset independent adiabaticity. These principles are then used to design optimal functions for the amplitude, frequency, and magnetic field gradient modulations for adiabatic inversion pulses. The last section considers some practical aspects for those who want to develop methodologies involving adiabatic pulses.

O câncer é uma das maiores causas de mortalidade no Brasil e no mundo, com potencial de crescimento nas próximas décadas. Um tipo de tratamento promissor é a hipertermia magnética, procedimento no qual as células tumorais morrem pelo efeito do calor gerado por partículas magnéticas após a aplicação de campo magnético alternado em frequências adequadas. Tais partículas também são capazes de atuar como agentes de contraste para imageamento por ressonância magnética, um poderoso método de diagnóstico para identificação de células neoplásicas, formando a combinação conhecida como theranostics (terapia e diagnóstico). Neste trabalho foram sintetizadas nanopartículas de óxido de ferro por método de coprecipitação com posterior encapsulação por técnica de nano spray drying, visando sua aplicação no tratamento de câncer por hipertermia e como agente de contraste para imageamento por ressonância magnética. Para a encapsulação foram utilizadas matrizes poliméricas de Maltodextrina com Polissorbato 80, Pluronic F68, Eudragit® S100 e PCL com Pluronic F68, escolhidos com o intuito de formar partículas que dispersem bem em meio aquoso e que consigam atingir alvo tumoral após administração no corpo do paciente. Parâmetros de secagem pelo equipamento Nano Spray Dryer, como temperatura, solvente e concentração de reagentes, foram avaliados. As partículas formadas foram caracterizadas por Microscopia Eletrônica de Varredura, Difração de Raios-X, Análise Termogravimétrica, Espalhamento de Luz Dinâmico, Espectroscopia de Infravermelho, magnetismo quanto a magnetização de saturação e temperatura, citotoxicidade e potencial de aquecimento. Tais procedimentos indicaram que o método de coprecipitação produziu nanopartículas de magnetita de tamanho em torno 20 nm, superparamagnéticas a temperatura ambiente, sem potencial citotóxico. A técnica de nano spray drying foi eficiente para a formação de partículas com tamanho em torno de 1 &#956m, também superparamagnéticas, biocompatíveis e com propriedades magnéticas adequadas e para aplicações pretendidas. Destaca-se a amostra com Pluronic, OF-10/15-1P, que apresentou magnetização de saturação de 68,7 emu/g e interação específica com células tumorais. Cancer is one of the greatest causes of mortality in Brazil and in the world, with growing potential for the next decades. A promising treatment alternative is magnetic hyperthermia, in which tumor cells die by the heat generated by magnetic nanoparticles after application of an alternate magnetic field in adequate frequencies. Such particles are also capable of acting as contrast agents for magnetic resonance imaging, a powerful method of diagnosis for the identification of neoplasic cells, which characterizes the combination of properties known as theranostics (therapy and diagnosis). In this work, iron oxide nanoparticles were synthesized by coprecipitation method with subsequent encapsulation by nano spray drying technique, aiming their application on cancer treatment by hyperthermia and on magnetic resonance imaging as a contrast agent. Polymeric matrices of Maltodextrin with Polysorbate 80, Pluronic F68, Eudragit® S100 and PCL with Pluronic F68 were employed for encapsulation, chosen carefully to create particles that disperse well in aqueous media and that are able to address the tumoral target after administration into the patient\'s body. Drying parameters of the Nano Spray Dryer equipment, such as temperature, dispersing medium and reagent concentrations, were evaluated. The generated particles were characterized by Scanning Electron Microscopy, X-Ray Diffraction, Thermogravimetric Analysis, Dynamic Light Scattering, Infrared Spectroscopy, by magnetism in matters of applied magnetic field and temperature, cytotoxic potential and heating potential. Such methods indicated that the coprecipitation method was able to produce magnetite nanoparticles with size of approximately 20 nm, superparamagnetic at room temperature and with no cytotoxic potential. The nano spray drying technique was efficient to produce particles with size of around 1 &#956m, biocompatible, superparamagnetic and with adequate magnetic properties for the intended applications. The sample OF-10/15-1P stands out with a saturation magnetization of 68.7 emu/g and presenting specific interactions with the tumour cells.

A Pressão intracraniana (PIC) é um dos principais parâmetros fisiológicos em animais e humanos e sua morfologia é extremamente importante. Entretanto, todos os métodos de monitoramento existentes no mercado são invasivos, existindo uma ampla demanda por sistemas não invasivos, expandindo assim o campo de pesquisas acerca desse importante parâmetro neurológico, que só não é melhor estudado devido a forma invasiva de ser monitorado. A motivação é fazer com que o monitoramento da PIC seja tão comum e tão essencial quanto é hoje o monitoramento da pressão arterial, facilitando o diagnóstico e até prognóstico de diversas doenças. Este trabalho analisa e implementa melhorias de um sistema não invasivo de monitoramento da pressão intracraniana, baseado em extensometria. Um dos objetivos, no que tange o desenvolvimento do produto, é analisar o equipamento como um todo - Sensor, Hardware, Firmware e Software - e propor melhorias a partir dos testes realizados. Os testes realizados In vivo mostraram uma boa correlação do sinal com um sistema Gold Stardard, evidenciando o potencial promissor do método. The intracranial pressure (ICP) is one of the main physiological parameters in animals and humans and its morphology is extremely important. However, all monitoring methods available in the market are invasive and there is a large demand for non-invasive systems, thus expanding the scope of research on this important neurological parameter, that just is not further studied because of the invasive method of monitoring. The motivation is to make monitoring the ICP as common and as essential, as monitoring the blood pressure is nowadays, facilitating diagnosis and even prognosis of various diseases. This work analyzes and implements improvements in a non-invasive intracranial pressure monitoring system based on extensometer. One of the goals, regarding product development, is to analyze the unit as a whole- Sensor, Hardware, Firmware and Software- and propose improvements from the tests. The in vivo tests showed a good correlation with a Gold Stardard system signal showing the promising potential of the method.

As técnicas de ressonância magnética nuclear (RM), por não serem invasivas ou destrutivas, apresentam um papel importante em diversas áreas do conhecimento como, por exemplo, medicina, química, física, setor rural, setor petrolífero, etc. Entretanto os custos de aquisição, manutenção e atualização dos equipamentos comerciais são muito altos e países em desenvolvimento, como é o caso do Brasil, sofrem com os impactos em função deste fator, principalmente nos setores ligados à pesquisa, em especial, nas áreas de concepção de novos métodos de RM. Além disso, o cenário nacional de desenvolvimento tecnológico desta área é escasso e os equipamentos comerciais fornecidos pelas empresas nem sempre oferecem a flexibilidade desejada. Baseado no uso de tecnologias inovadoras, como é o caso dos hardwares reconfiguráveis e uso de linguagem, compilador, bibliotecas e editores próprios para desenvolvimento de métodos de sequência de pulsos de RM, o Centro de Imagens e Espectroscopia in vivo por Ressonância Magnética (CIERMag) propõe uma solução alternativa através do desenvolvimento de um espectrômetro completamente digital de RM, flexível o suficiente para operar como um tomógrafo. O objetivo deste trabalho é idealizar e desenvolver a camada de software responsável pela aquisição, organização, visualização e armazenamento dos dados provenientes do espectrômetro através do subsistema ToRM Console, levando em consideração a flexibilidade proposta. Além disso, um arcabouço de ferramentas para automatização de pós-processamento, o ToRM Processing API, foi desenvolvido. Ambos os sistemas foram implementados em linguagem Python, com auxílio das principais bibliotecas de propósito científico (Numpy, Scipy, Matplotlib, etc). A realização de um experimento para estudo do comportamento do tempo de relaxação transversal de soluções de sulfato de cobre em função da variação da concentração é mostrada através do uso do método Carr-Purcell-Meiboom-Gill (CPMG). Outras aplicações do espectrômetro são mostradas através da obtenção de dados gerados por meio de sintetizadores de sinais acoplados ao espectrômetro. Os resultados mostram que o espectrômetro atualmente pode operar como um relaxômetro de RM de fácil operação. Os sistemas desenvolvidos neste trabalho mostram-se suficientemente flexíveis e preparados para operação em experimentos de Imagens e Espectroscopia de Alta Resolução, que são futuras etapas de desenvolvimento no CIERMag, tendo em vista as ferramentas desenvolvidas e testadas através do uso de dados sintetizados. The Nuclear Magnetic Resonance (NMR) techniques, which are neither invasive nor destructive, play an important role on several areas of knowledge, such as Medicine, Chemistry, Physics, Agriculture, Oil industry, among others. However, the costs associated with acquisition, maintenance and upgrade are very high, which have impacted on developing countries, as Brazil. Furthermore, the national scenario of technological development in this area is scarce and the commercial equipment provided by the companies does not always offer the desired flexibility. Based on the use of innovative technologies, which is the case of reconfigurable hardware and the use of proprietary language, compiler, libraries and editors for the development of NMR sequence pulses methods, the Magnetic Resonance Imaging and in vivo Spectroscopy Center (CIERMag) have proposed an alternative solution through the development of a completely digital NMR spectrometer, flexible enough to operate as an MRI Scanner. This dissertation reports the design and development of a software layer responsible for the acquisition, organization, visualization and storage of the data provided by the spectrometer through the ToRM Console subsystem, taking into account the proposed flexibility. A framework for post-processing automation, the ToRM Processing API, is also described. Both systems were developed in Python programming language, with the help of the main scientific purpose libraries (Numpy, Scipy, Matplotlib, etc.). Carr-Purcell-Meiboom-Gill (CPMG) method was used for the evaluation of the transverse relaxation time dependency on variable concentrations of copper sulfate solution. Other spectrometer applications are shown through the acquisition of synthesized data from simulation devices coupled to the spectrometer. The results show that the spectrometer can currently operate as an easy-to-use NMR Relaxometer. The systems described in the present work proved to be sufficiently flexible and prepared for the operation as MRI Scanners or Analytical Spectrometers, which are the future stages of development at CIERMag, considering the tools developed and tested through the use of synthesized data.

Neste trabalho, é apresentado o desenvolvimento de um IP de rede Ethernet com interface para o barramento Avalon para utilização em conjunto com o processador Nios II da Altera. O IPC foi adaptado do projeto Ethernet_tri_mode, e é capaz de transferir dados a velocidades de 1000, 100 e 10 Mbps. O desenvolvimento envolveu a adaptação do código para atingir os requisitos do projeto, feito segundo as diretrizes do CIERMag de manter todo o código em VHDL. Além disso, foi implementada uma interface de comunicação com o processador Nios II para tornar possível a configuração do sistema, bem como a transferência de dados através de um software sendo executado no processador. O IPC Ethernet foi projetado para ser aplicado no espectrômetro digital em desenvolvimento pelo CIERMag e teve como compromissos a baixa utilização de recursos lógicos do FPGA e, ao mesmo tempo, a disponibilização de uma alta taxa de transferência de dados para o espectrômetro. Como ferramenta de desenvolvimento, foi utilizada a plataforma Quartus II cujo fornecedor é a Altera. Já os testes em placa foram realizados em um kit de desenvolvimento DE3-150 da Terasic, o qual utiliza uma FPGA Stratix III, também da Altera. Com o intuito de testar e validar o sistema, foi desenvolvido um software para o processador Nios II capaz de receber e enviar dados através do IPC e com inteligência para responder pedidos do tipo ARP e PING. O subsistema de Gigabit Ethernet desenvolvido aqui já incorpora a versão corrente do Espectrômetro Digital de RM do CIERMag. In this work we expose the implementation of an Ethernet network core which interfaces to Avalon bus used along with the Nios II Altera processor. This core was adapted from the Ethernet_tri_mode project. It can transfer data at rates of 1000, 100 and 10 Mbps. The development involved the adaptation of the code to fullfil the project requirements, under the policy of the CIERMag to keep the whole coding in VHDL. Furthermore was implemented an interface to communicate with the Nios II processor to enable system configuration and data transfer through a software running on the processor. The core was projected to be applied with focus on the utilization of low FPGA logical resources with the availability of a high data transfer rate. It will be used in a digital spectrometer under development at the CIERMag. The Quartus II platform, supplied by Altera was used as the development tool. The tests on board where carried out on a DE3-150 development kit from Terasic, which has an FPGA Stratix III also from Altera. In order to test and validate the system, a software for the Nios II processor was developed, able to send and receive data via IPC and with intelligence to answer ARP and PING types requests. The developed Gigabit Ethernet subsystem is now part of the running version of the CIERMag Digital MR Spectrometer.

Considerando que a batata é o terceiro produto alimentar mais importante no mundo é necessária sua preservação e adaptação para as diferentes comunidades e condições ambientais do planeta, pois a consequência da redução da produção desse alimento pode ser a fome em todo o mundo. Um potecial causador desse problema é o aquecimento global, que é cada vez mais aceito pela ciência e as mudanças climáticas que vêm acontecendo no planeta. Alguns estudos sobre o efeito do aquecimento global e também mais especificamente do estresse hídrico sobre a cultura da batata estão sendo realizados, e são analisados os efeitos sobre cada parte da planta, como folhas, brotos, raízes, etc, porém o estudo sobre a raiz da batata é realizado de forma invasiva, ou seja, é necessário remover a planta do substrato. Nesse trabalho, apresentamos uma técnica para o estudo dos efeitos do estresse hídrico sobre a raiz da batata de forma não invasiva, portanto, os estudos serão realizados sem a remoção da planta do substrato. Dessa forma, poderão ser acompanhados os efeitos do estresse hídrico na planta em todos os estádios do desenvolvimento. Além disso, pode-se acompanhar o crescimento natural da raiz, diferentemente de outros modelos de estudo. A análise foi realizada com amostras da cultivar Ágata, plantadas em substrato sem substâncias paramagnéticas, e para isso foi utilizada a cultura hidropônica. Foram realizadas Imagens de Ressonância Magnética e em seguida um processamento e quantificação das imagens através da ferramenta matemática Multifractal. A técnica mostrou-se promissora para esse estudo, pois parâmetros multifractais mostraram-se sensíveis ao estresse hídrico, portanto, poderão ser realizados futuros estudos em outras cultivares de batata sob efeito do estresse hídrico. Considering that potato is the third most important food product in the world, it is necessary its preservation and adaptation to different communities and environmental conditions on the planet, because the consequence of reducing the production of that crop may be the hunger of people around the world. With global warming, which is increasingly more accepted by science, and the climate changes that are happening on the planet it is necessary to generate new genotypes more resistant to these changes, as well as the generation of varieties suitable for tropical climates. Some studies on the effect of global warming and also more specifically on water stress on potato crops are being conducted. In these studies, one analyze the effects on every part of the plant, such as leaves, shoots, roots, etc., but the study on potato root is conducted invasively, i.e., to observe the result is necessary to remove the plant from the substrate. In this work, we present a technique for studying the effects of water stress on the potato root in a noninvasively way, therefore, various studies may be performed without requiring the removal of the plant from the substrate. Thus, the studies may accompany the effects of water stress on the plant at all stages of development. In addition, this tool allows the observation and monitoring of the natural growth of the root, unlike the other study designs. The analysis was performed with samples of cultivar Agata, planted in cups with substrate containing no paramagnetic substances, and to make this possible hydroponic culture was used. Magnetic Resonance Imaging studies were performed followed by processing and quantification of the images obtained through the mathematical tool Multifractal . The technique has shown promising for this study because multifractal parameters were sensitive to water stress, so future studies can be conducted in other potato cultivars under effect of water stress.
