Abstract

Natural products (NPs) have long been used as a rich source of bioactive compounds for drug development. Recent technological advancements have revitalised natural products research as evidenced by increased publications in this field. In this editorial review, we highlight key points from the 2020 British Journal of Pharmacology (BJP) practical guide, which outlines standards for natural products research reports, and provide papers published in BJP between years 2020 to 2023 that demonstrate adherence to these guidelines. Looking ahead, we discuss the potential of chemical proteomics approaches to elucidate natural products mechanisms of action and identify therapeutic targets for future research. Abstract

In 2023, seventy novel drugs received market authorization for the first time in either Europe (by the EMA and the MHRA) or in the United States (by the FDA). Confirming a steady recent trend, more than half of these drugs target rare diseases or intractable forms of cancer. Thirty drugs are categorized as "first‐in‐class" (FIC), illustrating the quality of research and innovation that drives new chemical entity discovery and development. We succinctly describe the mechanism of action of most of these FIC drugs and discuss the therapeutic areas covered, as well as the chemical category to which these drugs belong. The 2023 novel drug list also demonstrates an unabated emphasis on polypeptides (recombinant proteins and antibodies), Advanced Therapy Medicinal Products (gene and cell therapies) and RNA therapeutics, including the first‐ever approval of a CRISPR‐Cas9‐based gene‐editing cell therapy. Abstract

This article features St. Lawrence University's Adirondack Semester, a small program, with enrollment limited to twelve students. The participants of the program are composed of a small group of students with shared interests and serious purpose, living harmoniously and mostly free from the temptations and distractions of the larger world, learning from senior scholars at the same time that they tutor one another. Though the courses reinforce one another, experience itself unites the elements of the program. The most important part of that experience is living in a small, close-knit community with a materially simple lifestyle that is in close contact with nature. The authors describe the effects of the program and how it brings students close to nature.

Abstract

In this review, we identify opportunities for drug discovery in the treatment of COVID-19 and, in so doing, provide a rational roadmap whereby pharmacology and pharmacologists can mitigate against the global pandemic. We assess the scope for targeting key host and viral targets in the mid-term, by first screening these targets against drugs already licensed, an agenda for drug repurposing, which should allow rapid translation to clinical trials. A simultaneous, multi-pronged approach using conventional drug discovery methods aimed at discovering novel chemical and biological means of targeting a short list of host and viral entities which should extend the arsenal of anti-SARS-CoV-2 agents. This longer term strategy would provide a deeper pool of drug choices for future-proofing against acquired drug resistance. Second, there will be further viral threats, which will inevitably evade existing vaccines. This will require a coherent therapeutic strategy which pharmacology and pharmacologists are best placed to provide. Abstract

Scientists who plan to publish in the British Journal of Pharmacology (BJP) should read this article before undertaking studies utilising anaesthetics in mammalian animals. This editorial identifies certain gaps in the reporting of details on the use of anaesthetics in animal research studies published in the BJP. The editorial also provides guidance, based upon current best practices, for performing in vivo experiments that require anaesthesia. In addition, mechanisms of action and physiological impact of specific anaesthetic agents are discussed. Our goal is to identify best practices and to provide guidance on the information required for manuscripts submitted to the BJP that involve the use of anaesthetic agents in studies with experimental animals. Abstract

Accurate detection of hypertension is crucial for clinical management of pediatric chronic kidney disease (CKD). The 2017 American Academy of Pediatrics (AAP) clinical practice guideline for childhood hypertension included new normative blood pressure (BP) values and revised definitions of BP categories. In this study, we examined the effect of applying the AAP guideline’s normative data and definitions to the Chronic Kidney Disease in Children (CKiD) cohort compared with use of normative data and definitions from the 2004 Fourth Report on the Diagnosis, Evaluation, and Treatment of High Blood Pressure in Children and Adolescents.

Abstract

The increasing scale and interconnection of many environmental challenges – from climate change to land use – has resulted in the need to collaborate across borders and boundaries of all types. Traditional centralized, top-down and sectoral approaches to governance of single-issue areas or species within social-ecological systems often have limited potential to alleviate issues that go beyond their jurisdiction. As a result, collaborative governance approaches have come to the forefront. A great deal of past research has examined the conditions under which collaborative efforts are likely to achieve desired outcomes. However, few studies have analyzed how the means to achieve successful collaborative outcomes differ based on context when examined across multiple studies. In this research, we begin to chart a means for doing this. Building onto a Context-Mechanism-Outcome (CMO) Framework, we provide a coding manual to analyse how contextual variables mediate the effects of mechanism variables on outcomes of the collaborative governance of social-ecological systems. Through the examination of four cases, we provide a proof-of-concept assessment and show the utility of the CMO framework and coding manual to draw comparisons across cases for understanding how collaborative outcomes are contingent on the social-ecological context in which they occur.
