In this article we discuss the digital image as a form of representation of likeness in the digital environment. The English word likeness entails the meaning of similarity that in the theory of rhetoric constitute persua-siveness. Likeness is an implicit and often taken for granted quality of the communicative performance of digital media. While the term image is a ty-pological classification, semiotic relations of the transfer of meaning can be described with the terms icon and simulacrum. We show their presence in the digital environment tracing their tradition of their function regarding the establishing of likeness to philosophical ideas. We exemplify with the case of the digital images as derivations from the portrait Mona Lisa that the appearance as an image of all what is displayed on the screen consti-tutes the specific likeness of digitality. The persuasiveness of digital images is in line with the theory of rhetoric in an exaggerated presence of the im-age as source of aesthetic perception with the sense of sight of the viewer.

Drug-inducible suicide systems may help to minimize risks of human induced pluripotent stem cell (hiPSC) therapies. Recent research challenged the usefulness of such systems since rare drug-resistant subclones were observed. We have introduced a drug-inducible Caspase 9 suicide system (iCASP9) into the AAVS1 safe-harbor locus of hiPSCs. In these cells, apoptosis could be efficiently induced

PathogenicACTBandACTG1gene variants, encoding the actin isoformsβCYA andγCYA, respectively, are frequently associated with theBaraitser-Winter-CerebroFrontoFacial syndrome (BWCFF-S) that includes malformations of cortical development. Here we explore whether cerebral organoids grown from BWCFF-S patient-derived induced pluripotent stem cells can provide insight into the pathogenesis underlying the cortical malformations of these patients. Cerebral organoids expressing either anACTBor anACTG1gene variant, each with a point mutation resulting in a single amino acid substitution, are reduced in size, showing smaller ventricle-like structures with a thinner ventricular zone (VZ). This decrease in VZ- progenitors is associated with a striking change in the orientation of their cleavage plane from predominantly vertical (control) to predominantly horizontal (BWCFF-S), which is incompatible with increasing VZ-progenitor abundance. The underlying cause appears to be an altered subcellular tubulin localization due to the actin mutations that affects mitotic spindle positioning of VZ-progenitors in BWCFF-S.

BACKGROUND AND AIMS Anti-neutrophil-cytoplasmatic-antibody-associated vasculitis (AAV) is a life-threatening autoimmune disease. It is a necrotizing vasculitis of small- and medium-sized vessels. T-cells play a pivotal role in pathogenesis and mediating damage in the vessels. Exogen ATP (adenosine triphosphate) is normally in a low nanomolar range. Under inflammatory conditions, various cells can release ATP to the extracellular department. In this case, ATP is a DAMP (damage-associated molecular pattern), but its effect on disease mechanisms in AAV is not well understood. It is the aim of the study to determine the effect of exogenous ATP on T-cells in healthy controls and AAV-patients. METHOD A total of 22 AAV-patients in remission and 13 healthy controls were recruited. Peripheral blood mononuclear cells (PBMC) were isolated and then labeled with a tracking dye. The PBMC were cultured for 3 days with different concentrations of ATP in presence or absence of anti-CD3/CD28 stimulation. After 3 days, the cells were restimulated with PMA/Ionomycin and Brefeldin A for 4 hours. Cytokine production and cell proliferation were assessed by flow cytometry. Baseline was defined as T-cell proliferation in absence of exogenous ATP. The fold change was calculated by: $\frac{{{\rm{T}} - {\rm{cell\ proliferation\ with\ ATP}}}}{{{\rm{T}} - {\rm{cell\ proliferation\ without\ ATP}}}}$ . The P-value for paired values was calculated by the Wilcoxon matched-pairs signed rank test. The P-value for unpaired groups was calculated by the Mann Whitney U-test. RESULTS The mean T-cell proliferation of CD4+-T-cells for healthy controls was significantly different comparing 0 µM ATP and 500 µM ATP (mean proliferated fraction, 0 µM ATP versus 500 µM ATP: 40.48% versu 22.58%, P = .0002). In presence of ATP, a suppression of T-cell proliferation was found in patients and healthy controls (patients versus healthy controls, fold change T-cell proliferation from baseline with 50 µM ATP: 0.81 ± 0.03 versus 0.79 ± 0.03, P = .60; 125 µM ATP: 0.75 ± 0.03 versus 0.70 ± 0.04, P = .38; 250 µM ATP: 0.68 ± 0.03 versus 0.59 ± 0.05, P = .17; 500 µM ATP: 0.53 ± 0.03 versus 0.54 ± 0.05, P = .77); the higher the concentration of ATP, the stronger the suppression of proliferation. IFNγ+-CD4+-T-cells of healthy controls were also suppressed by exogenous ATP (0 µM ATP versus 500 µM ATP, CD4+ T-cells: %IFNγ+: 19.94% versus 10.59%, P = .0002). INFγ production by CD4+-T-cells was reduced after addition of ATP in both patients and healthy controls (patients versus healthy controls, fold change CD4+-IFNγ+ -T-cells from baseline with 50 µM ATP: 0.69 ± 0.04 versus 0.64 ± 0.04, P = 0.45; 125 µM ATP: 0.62 ± 0.04 versus 0.58 ± 0.04, P = .63; 250 µM ATP: 0.62 ± 0.04 versus 0.49 ± 0.04, P = .11; 500 µM ATP: 0.59 ± 0.05 versus 0.51 ± 0.05, P = .56). CONCLUSION Exogenous ATP induced suppression of T-cells in healthy controls and might be important for the maintenance of immunotolerance. DAMP-mediated inhibitory mechanisms were functional in AAV-patients in remission.

To harness the full potential of human pluripotent stem cells (hPSCs) we combined instrumented stirred tank bioreactor (STBR) technology with the power of in silico process modeling to overcome substantial, hPSC‐specific hurdles toward their mass production. Perfused suspension culture (3D) of matrix‐free hPSC aggregates in STBRs was applied to identify and control process‐limiting parameters including pH, dissolved oxygen, glucose and lactate levels, and the obviation of osmolality peaks provoked by high density culture. Media supplements promoted single cell‐based process inoculation and hydrodynamic aggregate size control. Wet lab‐derived process characteristics enabled predictive in silico modeling as a new rational for hPSC cultivation. Consequently, hPSC line‐independent maintenance of exponential cell proliferation was achieved. The strategy yielded 70‐fold cell expansion in 7 days achieving an unmatched density of 35 × 106 cells/mL equivalent to 5.25 billion hPSC in 150 mL scale while pluripotency, differentiation potential, and karyotype stability was maintained. In parallel, media requirements were reduced by 75% demonstrating the outstanding increase in efficiency. Minimal input to our in silico model accurately predicts all main process parameters; combined with calculation‐controlled hPSC aggregation kinetics, linear process upscaling is also enabled and demonstrated for up to 500 mL scale in an independent bioreactor system. Thus, by merging applied stem cell research with recent knowhow from industrial cell fermentation, a new level of hPSC bioprocessing is revealed fueling their automated production for industrial and therapeutic applications., Wet lab‐derived process characteristics enabled predictive in silico modeling as a rational for suspension‐based human pluripotent stem cells (hPSC) cultivation in stirred bioreactors. Consequently, this hPSC line‐independent strategy yielded 70‐fold cell expansion in 7 days achieving a density of 35 × 106 cells/mL equivalent to 5.25 billion hPSC in 150 mL scale while maintaining a pluripotent phenotype and simultaneously reducing media requirements by 75%.

The Deipnosophists is a literary work that presents an insight into ancient rhetoric and speech from various perspectives, while its author, Athenaeus of Naucratis, has a mediating function in the process of communicating this knowledge besides other topics of knowledge about ancient cultures to the readers. Being composed of fictive speeches that join the texts of ancient writings in paraphrases and citations in the conversations of the participants during a banquet, the work reveals in different layers information about rhetoric. Rhetoric is present in the composition of the work itself. In the conversation’s speech is present as the essential form of this piece of literature and one component of this composite text. But also, the writings of other authors reflect knowledge about rhetoric as the scholarly discipline for speech. We argue that Athenaeus invents and composes here a memory that arranges topics related to the culture of banquets in speeches. The speeches range from the factuality of historical accounts about rhetoricians to the fictional story of the event and its speeches. The Deipnosophists blends speeches into each other as conversations of the framing narrative of a meeting of Athenaeus who tells the story of the event to a friend and the speeches of the deipnosophists in the actual event of the banquet. The presentation of the texts of the cited and paraphrased books on rhetoric and anecdotal information about rhetoricians and sophists in the description of the banquet rely on the intensive collection of information recorded in the medium book that the author used for the creation of his own work.

CRISPR prime-editors are emergent tools for genome editing and offer a versatile alternative approach to HDR-based genome engineering or DNA base-editors. However, sufficient prime-editor expression levels and availability of optimized transfection protocols may affect editing efficiencies, especially in hard-to-transfect cells like hiPSC. Here, we show that piggyBac prime-editing (PB-PE) allows for sustained expression of prime-editors. We demonstrate proof-of-concept for PB-PE in a newly designed lentiviral traffic light reporter, which allows for estimation of gene correction and defective editing resulting in indels, based on expression of two different fluorophores. PB-PE can prime-edit more than 50% of hiPSC cells after antibiotic selection. We also show that improper design of pegRNA cannot simply be overcome by extended expression, but PB-PE allows for estimation of effectiveness of selected pegRNAs after few days of cultivation time. Finally, we implemented PB-PE for efficient editing of an amyotrophic lateral sclerosis-associated mutation in the SOD1-gene of patient-derived hiPSC. Progress of genome editing can be monitored by Sanger-sequencing, whereas PB-PE vectors can be removed after editing and excised cells can be enriched by fialuridine selection. Together, we present an efficient prime-editing toolbox, which can be robustly used in a variety of cell lines even when non-optimized transfection-protocols are applied.

Recent research challenged the usefulness of such systems since rare drug-resistant subclones were observed that showed elimination or silencing of the transgene.We have introduced a drug-inducible Caspase9 suicide system (iCASP9) into the AAVS1 safe harbor locus of hiPSCs. In these cells, apoptosis could be efficiently induced in vitro. In mice, drug treatment generally led to rapid elimination of teratomas, but individual animals subsequently formed tumor tissue from monoallelic iCASP9 hiPSCs. Very rare drug-resistant subclones of monoallelic iCASP9 hiPSCs appeared in vitro with frequencies of ~ 3×10-8. Transgene elimination, presumably via Loss of Heterozygosity (LoH), or methylation of the CAG promoter but not methylation of the ppp1r12c locus were identified as underlying mechanisms. In contrast, we never observed any escapees from biallelic iCASP9 hiPSCs, even after treatment of up to 0.8 billion hiPSCs.In conclusion, biallelic integration of an iCASP9 system in the AAVS1 locus may substantially contribute to the safety level of iPSC-based therapies, which should be calculated by relating clonal escapee frequencies to the cell number in tumors of a size that is readily detectable during routine screening procedures.

Loss-of-function mutations in the bone morphogenetic protein receptor 2 (BMPR2) gene are common in heritable or idiopathic pulmonary arterial hypertension (PAH), and can result in functional impairment of both endothelial and vascular smooth muscle cells. Here, we report 3 PAH patient-specific induced pluripotent stem cells (iPSC) lines from 3 unrelated patients harbouring different mutations in the BMPR2 gene: a heterozygous missense mutation in exon 12, a heterozygous frame shift deletion in exon 3, and a heterozygous missense mutation in exon 11. These cell lines will serve as a valuable resource to model PAH in vitro.

This article presents a model and a case study of the narrative structures that are present in the interactive media design of multimedia applications in the mass media. As basic categories for the history and structure of media, we employ the model of the modes of the physical, analog, and digital presentation/representation. In this case study of the online edition of the New York Times, we have the case of a newspaper that in the digital edition employs multi-media applications. Contrasting the traditional concept of “narrativity” with the current status quo of the digital media outlets, we will examine the specific conditions of the multimedia applications in the mass media with this sample case of the New York Times. As narrative structure of interactive design in multimedia for mass communication, we analyze the types of narrative media and general narrativity in the New York Times with the background of its processes of interactivity and media convergence.

Transgenic hiPSC lines carrying reporter genes represent valuable tools for functional characterization of iPSC derivatives, disease modelling and clinical evaluation of cell therapies. Here, the hiPSC line ‘Phoenix’ (Haase et al., 2017) was genetically engineered using TALEN-based integration of the calcium sensor GCaMP6f and RedStarnuc reporter into the AAVS1 site. Characterization of undifferentiated cells and functional investigation of hiPSC-derived cardiomyocytes-containing BCTs showed a strong intracellular calcium transient-dependent GCaMP6f and eminent RedStarnuc signal. Therefore, our dual reporter line provides an excellent tool to facilitate monitoring of engraftment, calcium fluctuations and coupling of iPSC derivatives such as cardiomyocytes in vitro and in vivo in animal models.

Cellular therapies based on induced pluripotent stem cells (iPSCs) come out of age and an increasing number of clinical trials applying iPSC-based transplants are ongoing or in preparation. Recent studies, however, demonstrated a high number of small-scale mutations in iPSCs. Although the mutational load in iPSCs seems to be largely derived from their parental cells, it is still unknown whether reprogramming may enrich for individual mutations that could lead to loss of functionality and tumor formation from iPSC derivatives. 30 hiPSC lines were analyzed by whole exome sequencing. High accuracy amplicon sequencing showed that all analyzed small-scale variants pre-existed in their parental cells and that individual mutations present in small subpopulations of parental cells become enriched among hiPSC clones during reprogramming. Among those, putatively actionable driver mutations affect genes related to cell-cycle control, cell death, and pluripotency and may confer a selective advantage during reprogramming. Finally, a short hairpin RNA (shRNA)-based experimental approach was applied to provide additional evidence for the individual impact of such genes on the reprogramming efficiency. In conclusion, we show that enriched mutations in curated onco- and tumor suppressor genes may account for an increased tumor risk and impact the clinical value of patient-derived hiPSCs.

Primary ciliary dyskinesia (PCD) is a genetic disorder characterized by defects in motile cilia and is known to occur in about 1 in 20,000 live births (Horani and Ferkol, 2018). Among the many genes associated with PCD, NME5, a gene encoding a protein involved in ciliary function, was recently reported to be involved in PCD (Anderegg et al., 2019; Cho et al., 2020). We have established two human induced pluripotent stem cell clones from a PCD patient carrying a deletion in the NME5 gene (c.415delA (p.Ile139Tyrfs*8)).

Cardiac levels of the signal transducer and activator of transcription factor-3 (STAT3) decline with age, and male but not female mice with a cardiomyocyte-specific STAT3 deficiency (CKO) display premature age-related heart failure associated with reduced cardiac capillary density. In the present study isolated male and female CKO-cardiomyocytes exhibit increased prostaglandin (PG)-generating cyclooxygenase-2 (COX-2) expression. The PG-degrading hydroxyprostaglandin-dehydrogenase-15 (HPGD) expression is only reduced in male cardiomyocytes, which is associated with increased PGD2 secretion from isolated male but not female CKO-cardiomyocytes. Reduced HPGD expression in male cardiomyocytes derive from impaired androgen-receptor-(AR)-signaling due to loss of its co-factor STAT3. Elevated PGD2 secretion in males is associated with increased white adipocyte accumulation in aged male but not female hearts. Adipocyte differentiation is enhanced in isolated SCA-1+-cardiac-progenitor-cells (CPC) from young male CKO-mice compared to the adipocyte differentiation of male wildtype (WT)-CPC and CPC isolated from female mice. Epigenetic analysis in freshly isolated male CKO-CPC display hypermethylation in pro-angiogenic genes (Fgfr2, Epas1) and hypomethylation in the white adipocyte differentiation gene Zfp423 associated with upregulated ZFP423 expression and a shift from endothelial to white adipocyte differentiation compared to WT-CPC. The expression of the histone-methyltransferase EZH2 is reduced in male CKO-CPC compared to male WT-CPC whereas no differences in the EZH2 expression in female CPC were observed. Clonally expanded CPC can differentiate into endothelial cells or into adipocytes depending on the differentiation conditions. ZFP423 overexpression is sufficient to induce white adipocyte differentiation of clonal CPC. In isolated WT-CPC, PGD2 stimulation reduces the expression of EZH2 thereby upregulating ZFP423 expression and promoting white adipocyte differentiation.Thus, cardiomyocyte STAT3-deficiency leads to age-related and sex-specific cardiac remodeling and failure in part due to sex-specific alterations in PGD2 secretion and subsequent epigenetic impairment of the differentiation potential of CPC. Causally involved is the impaired AR signaling in absence of STAT3, which reduces the expression of the PG degrading enzyme HPGD.

Dynein axonemal heavy chain 5 (DNAH5) is part of a microtubule-associated protein complex found within the cilia of the lung. Mutations in the DNAH5 gene lead to impaired ciliary function and are linked to primary ciliary dyskinesia (PCD), a rare autosomal recessive disorder. We established two human induced pluripotent stem cell (hiPSC) lines generated from a patient with PCD and homozygous mutation in the corresponding DNAH5 gene. These cell lines represent an excellent tool for modeling the ciliary dysfunction in PCD.

Media Rhetoric. The History, Theory and Applications of Rhetoric as Mediated Communication explores the tradition and continuity of the concepts of the discipline rhetoric in media for symbolic communication processes beyond speech from the perspectives of media studies, the theory of rhetoric, and applied rhetoric as an integral empirical element of communication processes with media. Based on an introduction of the history of media, media theory and media typology, the contributions of the theory of the discipline rhetoric for the understanding of media of symbolic communication and the practical relevance of rhetoric for the use of media of communication as field of applied rhetoric in contemporary area of the public discourse are described and analysed in relation to their since ancient time existing connection to rhetoric and a classification of text-types for the coding process of mass media of contemporary media is presented.

Caudal-type homeobox 2 (CDX2) transcription factor is an important marker for early trophoblast lineages and intestinal epithelium. Due to its nuclear expression the immunostaining and sorting of viable CDX2pos cells is not possible. In this paper we report the generation and describe key characteristics of a CDX2Venus knock-in reporter hiPSC-cell line (MHHi007-A-1) which can serve as an in vitro tool to study human trophoblast and intestinal differentiation.

Objective Pannexins are channel proteins important for the release of calcium and adenosine triphosphate, which are among other functions involved in early development. Here, the expression of pannexins was investigated in induced pluripotent stem cells derived from human cord blood endothelial cells (hCBiPS2), in hematopoietic stem cell-derived induced pluripotent stem cells (HSC_F1285_T-iPS2) and in human embryonic stem cells (HES-3). The expression of pannexin (Panx) 1–3 mRNAs was analyzed in all three undifferentiated stem cell lines. Stem cells then underwent undirected differentiation into embryoid bodies and were analyzed regarding expression of germ layer-specific genes. Results Panx1, Panx2, and Panx3 mRNAs were expressed in all undifferentiated stem cell lines investigated. In comparison, Panx1 showed the highest expression among all pannexins. The undirected differentiation resulted in a mixed germ layer genotype in all three stem cell lines. Whereas the expression of Panx1 was not affected by differentiation, the expression of Panx2 was slightly increased in differentiated hCBiPS2 cells, HSC_F1285_T-iPS2 as well as HES3 cells as compared to their undifferentiated counterparts. A slight increase of Panx3 expression was observed in differentiated hCBiPS2 cells only. In conclusion, pluripotent stem cells express all three pannexin genes. This article is interested in the phenomenon ʻtext‘ both as a cognitive and linguistic concept and as a construct in the history of scholarship with a historical review of definitions and functions of the text as well as studies of concrete texts. In the tradition of art as ʻmimesis‘ (imitation), of textual criticism of Hellenism and Christian theology, of modern linguistic definitions and functions of the text, and in contemporary types of texts employed by scholars we trace the concept of ʻtext‘ until the contemporary state of research. Culminating in the question of the authority determining ʻWhat is a text and what not?‘ we face finally the need of an interdisciplinary text model for the text as a complex construct of textual sequences. This model presented here will takeʻ into account the historical, interdisciplinary, and structural aspects of the analysis of texts. We will demonstrate that a proper understanding of ʻtext‘ and its studies as a method is only possible, when texts are analyzed in a process of ʻreading‘ with the background of various academic disciplines according to the here proposed model.

Among adolescents cannabis is one of the most widely used illicit drugs. In adolescence brain development continues, characterized by neuronal maturation and synaptic plasticity. The endocannabinoid system plays an important role during brain development by modulating neuronal function and neurogenesis. Changes in endocannabinoid signaling by Δ9 -tetrahydrocannabinol (THC), the psychoactive component of cannabis, might therefore lead to neurobiological changes influencing brain function and behavior. We investigated the functional maturation and dopaminergic specification of human cord blood-derived induced pluripotent stem cell (hCBiPSC)-derived small molecule neural precursor cells (smNPCs) after cultivation with the endogenous cannabinoid anandamide (AEA) and the exogenous THC, both potent agonists at the cannabinoid 1 receptor (CB1 R). Higher dosages of 10-μM AEA or THC significantly decreased functionality of neurons, indicated by reduced ion currents and synaptic activity. A lower concentration of 1-μM THC had no marked effect on neuronal and dopaminergic maturation, while 1-μM AEA significantly enhanced the frequency of synaptic activity. As there were no significant effects on DNA methylation in promotor regions of genes important for neuronal function, these cannabinoid actions seem to be mediated by another than this epigenetic mechanism. Our data suggest that there are concentration-dependent actions of cannabinoids on neuronal function in vitro indicating neurotoxic, dysfunctional effects of 10-μM AEA and THC during human neurogenesis.

Globalization has developed into one of the most interdisciplinary topics in the fields of culture, politics, and economics since the late 20th century. This article is interested in its terminology and reflections in the arts in the case of the lyrics of contemporary U.S. American and British English popular music. Even though the term ‘globalization’ was coined in the late 20th century and is associated with the idea of a universal economic, social, and cultural process in the world, conceptual elements of ‘globalization’ can be found in human history before the term was coined. The arts conceptualized the ‘world’ as an idea long before the emergence of the concept of ‘globalization’. The elements of ‘globalization’ and its history of associated ideas are found ironized and critically revised in the contemporary arts. At this point, social and cultural criticism arises and describes the contrast between ethical values and reality. Even an individualized form of poetry like the lyric poems of 20th and 21st century U.S. American popular music reflects critically as a contribution to contemporary political discourse on the concept of ‘globalization’.

Cystic Fibrosis (CF) is a genetic disease caused by mutations in the Cystic Fibrosis Transmembrane Conductance Regulator (CFTR) gene which encodes for a chloride ion channel regulating the balance of salt and water across secretory epithelia. Here we generated an iPSC line from a CF patient homozygous for the p.Asn1303Lys mutation, a Class II folding defect mutation. This iPSC line provides a useful resource for disease modeling and to investigate the pharmacological response to CFTR modulators in iPSC derived epithelia.

Loeys-Dietz syndrome (LDS) is a rare connective tissue disorder characterized by a genetic predisposition for thoracic aortic aneurysm and dissection. Despite heterozygous loss-of-function mutations in genes for ligand, receptor, or downstream mediators of the transforming growth factor β (TGFβ) pathway, LDS is associated with a signature of high TGFβ signaling. We generated induced pluripotent stem cell (iPSC) lines from three adult LDS-patients (two male, one female) of a family with a heterozygous point mutation in exon 4 of the TGFβ-receptor1 (TGFBR1) gene (p.M253I; c.759G>A). The lines offer a valuable resource for modeling the pathophysiology of genetically mediated aortic disease.
