Among the oral route, mouth dissolving tablets (MDTs) offer a benefit for drugs with slow dissolution and having low oral bioavailability. Epalrestat is one of the best effective diabetic neuropathy medication used for treating nerve pain. The problem associated with this drug is high first pass metabolism and low solubility in acidic media as well in basic media leads to short half-life, delayed dissolution and side effects. Therefore, the goal of the current work is to developed an epalrestat MDTs tablet that will provide quick drug dissolution and a quick onset of action for the treatment of nerve pain. MDTs of epalrestat were formulated by direct compression using natural superdisintegrants obtained from the various sources such as fenugreek, gum karaya and banana powder. All of the pre- and postcompression parameter results were shown to be in accordance with established specifications. In comparison to other formulations of MDTs, formulation F3 with 15 mg (7.5%) of banana powder displayed a higher rate of dissolution. It was determined that epalrestat MDTs containing natural superdisintegrant were successfully formulated with acceptable physical and chemical properties, quick oral cavity disintegration, a quick onset of action, and improved patient compliance. Ficus religiosa L., a member of the Moraceae family, is a medicinal plant having a number of pharmacological properties. The anti-inflammatory and analgesic actions of an ethanolic extract of F. religiosa bark FRE (at 100 and 200mg/kg dosages) and the biomarker component quercetin QC (at 5 and 10mg/kg doses) were investigated. The estimate of quercetin was carried by using an HPTLC analysis of FRE. Additionally, qualitative and quantitative screening for key important phytocomponents was done using dried, ground plant stem barks. By using molecular docking, the molecular interaction profile with several anti-inflammatory drug targets was examined. Both the FRE as well as QC showed a substantial decline in paw volume when compared with the relevant control groups (p<0.01 & p<0.001). Following the administration of acetic acid to mice, the FRE and QC both demonstrate a substantial lengthening of the paw licking or leaping towards Eddy's hot plate as well as a decrease in the number of writhes (p<0.01 & p<0.001). The safe and effective option for the therapeutic management of psoriasis was attempted by preparing transferosomes of Betamethasone valerate (BV) and incorporating into formulation of gel to get more skin permeation, more effectiveness in drug entrapment and sustained release. Batches of experimental design of transferosome containing 100 mg of BV were produced by using Lipova E 80 and Span 20 as surfactant in varying quantities and incorporated in 1.5% HPMC gel base. Formulation was optimized to attain maximum drug entrapment, % drug release, ex-vivo permeation, pH, viscosity, spreadability and extrudability. The size and stability was ascertained by using Zeta sizer and and TEM was utilized to assess the surface morphology. The optimized formulation entrapped 59.87 % of the drug, pH 6.57, the average size of the particle was found to be 108.93 nm and viscosity were 436.19 centipoise.The residual analyses by gas chomatography confirms the elimination of formulation's organic solvent and stability studies were conducted in accordance of the ICH guidelines. The developed formulation showed diffusion of BV through the rat's skin at sustained rate for 10 hrs while marketed preparation sustained for 7 hrs only. It promises of localized action at the affected psoriatic sites for prolonged period with better release characteristics as compared to marketed formulation. Objective: The study aimed at assessing the KAP on Adverse Drug reaction reporting and risk management among Healthcare Professionals in Saudi Arabia. Methods: A cross-sectional survey study was conducted where Participants were recruited from an online survey via a self-reported questionnaire. The data were captured in Microsoft Excel 2021 and were analyzed using Statistical Package for Social Sciences. To examine the potential differences in the mean KAP score among Dependent Variables (Knowledge, Attitude, Practice on ADR reporting and risk management) and independent variables (Occupation) repeated measures ANOVA was conducted. A p-value below 0.05 was deemed statistically significant. Results: KAP scores of Pharmacists were (1.9032, 3.1935, 3.3226) is higher when compared with Dentists (1.6667, 2.7778, 3.6667) and General Practitioners (1.1111,1.5, 3). The factor interaction of Participants of the different healthcare fields and KAP scores were not significant (F 2.477, p value 0.097, 2 = 6.871). Conclusion: Pharmacists exhibited superior understanding and disposition towards ADR reporting. Hence, it is imperative to enhance awareness and advocate for ADR reporting. [ABSTRACT FROM AUTHOR]

Background: The purpose of this research is to develop an analytical method and validate it according to ICH guidelines for the estimation of Toremifene by RP-HPLC-PDA with molecular docking and ADMET analysis. From molecular docking, it came to know the receptor affinity specifically to estrogen receptors (ERα and ERβ), which are responsible for cancer therapy. ADMET analyses secure its therapeutic potential as well safety of the drug., Methods: An isocratic method has developed by RP-HPLC-PDA (AGILENT 1100) with symmetry of 100 mm x 4.6 mm x 5 μm particle size C18 column and optimise mobile phase is methanol: 0.1% OPA (orthophosphoric acid) water ratio of 43:57% v/v. Under different conditions like acidic, alkaline, oxidative, and neutral environments, toremifene was tested for degradation., Results: The developed method is validated in accordance with ICH guidelines. A calibration curve with an r2 value of 0.9987 has been prepared across the range of 10 to 50 μg/ml with five standard dilutions. The retention time of the drug is 5.575 minutes. The validation results are system suitability (%RSD-0.76), inter-day precision (%RSD 0.14-0.29), intraday precision (%RSD 0.08-0.34), accuracy (%RSD 0.16-0.96), and robustness (%RSD 0.16-0.35). In different intended conditions, four peaks are in 1 N HCl, two peaks in 1 N NaOH, three peaks in 10% H2O2 (1hr), and one peak in neutral., Conclusion: Toremifene, a Selective Estrogen Receptor Modulator (SERM), Drug pharmacokinetic properties and receptor binding affinity results are helpful in designing the analytical method. Developing the RP-HPLC-PDA method is found to be novel, simple and precise. Coronavirus disease-2019 is a serious health threat around the globe. Across the world, approximately 142 million people were infected, and three million deaths happened. The fast propagation is also associated with constant anxiety, mental stress, and discomfort in public and health-care professionals. Lack of approved drugs regimen to combat the pandemic challenge concretely is a challenging project for all who are committed to developing remedial assistance. However, the successful development of three vaccines gives a solid roadmap to combat this disease.
