One of the major reasons for poor cancer outcomes is the existence of cancer stem cells (CSCs). CSCs are a small subpopulation of tumor cells that can self-renew, differentiate into the majority of tumor cells, and maintain tumorigenicity. As CSCs are resistant to traditional chemotherapy and radiation, they contribute to metastasis and relapse. Thus, new approaches are needed to target and eliminate CSCs. Here, we sought to target and reduce the frequency of CSCs in melanoma by therapeutic vaccination against CSC-associated transcription factors, such as Sox2 and Nanog, and aldehyde dehydrogenase (ALDH). Toward this goal, we have identified novel immunogenic peptide epitopes derived from CSC-associated Sox2 and Nanog and synthesized synthetic high-density lipoprotein (sHDL) nanodisc vaccine formulated with Sox2, Nanog, and ALDH antigen peptides together with CpG, a Toll-like receptor 9 agonist. Vaccination with nanodiscs containing six CSC antigen peptides elicited robust T cell responses against CSC-associated antigens and promoted intratumoral infiltration of CD8+ T cells, while reducing the frequency of CSCs and CD4+ regulatory T cells within melanoma tumors. Nanodisc vaccination effectively reduced tumor growth and significantly extended animal survival without toxicity toward normal stem cells. Overall, our therapeutic strategy against CSCs represents a cost-effective, safe, and versatile approach that may be applied to melanoma and other cancer types, as well as serve as a critical component in combined therapies to target and eliminate CSCs.

Purpose:Gliomas are brain tumors with dismal prognoses. The standard-of-care treatments for gliomas include surgical resection, radiation, and temozolomide administration; however, they have been ineffective in providing significant increases in median survival. Antigen-specific cancer vaccines and immune checkpoint blockade may provide promising immunotherapeutic approaches for gliomas.Experimental Design:We have developed immunotherapy delivery vehicles based on synthetic high-density lipoprotein (sHDL) loaded with CpG, a Toll-like receptor-9 agonist, and tumor-specific neoantigens to target gliomas and elicit immune-mediated tumor regression.Results:We demonstrate that vaccination with neoantigen peptide-sHDL/CpG cocktail in combination with anti–PD-L1 immune checkpoint blocker elicits robust neoantigen-specific T-cell responses against GL261 cells and eliminated established orthotopic GL261 glioma in 33% of mice. Mice remained tumor free upon tumor cell rechallenge in the contralateral hemisphere, indicating the development of immunologic memory. Moreover, in a genetically engineered murine model of orthotopic mutant IDH1 (mIDH1) glioma, sHDL vaccination with mIDH1 neoantigen eliminated glioma in 30% of animals and significantly extended the animal survival, demonstrating the versatility of our approach in multiple glioma models.Conclusions:Overall, our strategy provides a general roadmap for combination immunotherapy against gliomas and other cancer types.

To treat and manage chronic diseases, it is necessary to continuously monitor relevant biomarkers and modify treatment as the disease state changes. Compared to other bodily fluids, interstitial skin fluid (ISF) is a good choice for identifying biomarkers because it has a molecular composition most similar to blood plasma. Herein, a microneedle array (MNA) is presented to extract ISF painlessly and bloodlessly. The MNA is made of crosslinked poly(ethylene glycol) diacrylate (PEGDA), and an optimal balance of mechanical properties and absorption capability is suggested. Besides, the effect of needles’ cross-section shape on skin penetration is studied. The MNA is integrated with a multiplexed sensor that provides a color change in a biomarker concentration-dependent manner based on the relevant reactions for colorimetric detection of pH and glucose biomarkers. The developed device enables diagnosis by visual inspection or quantitative red, green, and blue (RGB) analysis. The outcomes of this study show that MNA can successfully identify biomarkers in interstitial skin fluid in a matter of minutes. The home-based long-term monitoring and management of metabolic diseases will benefit from such practical and self-administrable biomarker detection.

Shear-thinning biomaterials (STBs) based on gelatin-silicate nanoplatelets (SNs) are emerging as an alternative to conventional coiling and clipping techniques in the treatment of vascular anomalies. Improvements in the cohesion of STB hydrogels pave the way toward their translational application in minimally invasive therapies such as endovascular embolization repair. In the present study, sodium phytate (Phyt) additives are used to tune the electrostatic network of SNs-gelatin STBs, thereby promoting their mechanical integrity and facilitating injectability through standard catheters. We show that an optimized amount of Phyt enhances storage modulus by approximately one order of magnitude and reduces injection force by ≈58% without compromising biocompatibility and hydrogel wet stability. The Phyt additives are found to decrease the immune responses induced by SNs. In vitro embolization experiments suggest a significantly lower rate of failure in Phyt-incorporated STBs than in control groups. Furthermore, the addition of Phyt leads to accelerated blood coagulation (reduces clotting time by ≈45% compared to controls) due to the contributions of negatively charged phosphate groups, which aid in the prolonged durability of STB in coagulopathic patients. Therefore, the proposed approach is an effective method for the design of robust and injectable STBs for minimally invasive treatment of vascular malformations.

Cancer stem cells (CSCs) proliferate extensively and drive tumor metastasis and recurrence. CSCs have been identified in over 20 cancer types to date, but it remains unknown how to target and eliminate CSCs in vivo. Aldehyde dehydrogenase (ALDH) is a marker that has been used extensively for isolating CSCs. Here we present a novel approach to target and reduce the frequency of ALDH(high) CSCs by vaccination against ALDH. We have identified ALDH1-A1 and ALDH1-A3 epitopes from CSCs and developed synthetic high-density lipoprotein nanodiscs for vaccination against ALDH(high) CSCs. Nanodiscs increased antigen trafficking to lymph nodes and generated robust ALDH-specific T cell responses. Nanodisc vaccination against ALDH(high) CSCs combined with anti-PD-L1 therapy exerted potent antitumor efficacy and prolonged animal survival in multiple murine models. Overall, this is the first demonstration of a simple nanovaccine strategy against CSCs and may lead to new avenues for cancer immunotherapy against CSCs.

Bone defects, with second highest demand for surgeries around the globe, may lead to serious health issues and negatively influence patient lives. The advances in biomedical engineering and sciences have led to the development of several creative solutions for bone defect treatment. This review provides a brief summary of bone graft materials, an organized overview of top-down and bottom-up (bio)manufacturing approaches, plus a critical comparison between advantages and limitations of each method. We specifically discuss additive manufacturing techniques and their operation mechanisms in detail. Next, we review the hybrid methods and promising future directions for bone grafting, while giving a comprehensive US-FDA regulatory science perspective, biocompatibility concepts and assessments, and clinical considerations to translate a technology from a research laboratory to the market. The topics covered in this review could potentially fuel future research efforts in bone tissue engineering, and perhaps could also provide novel insights for other tissue engineering applications.

Epitope density has a profound impact on B cell responses to particulate Ags, the molecular mechanisms of which remain to be explored. To dissect the role of epitope density in this process, we have synthesized a series of liposomal particles, similar to the size of viruses, that display a model self-antigen peptide at defined surface densities. Immunization of C57BL/6J mice using these particles elicited both IgM and class-switched IgG1, IgG2b, and IgG3 autoreactive Abs that depended on the epitope density. In C57BL/6 gene knockout mice lacking either functional TCRs or MHC class II molecules on B cells, the liposomal particles also elicited IgM, IgG1, IgG2b, and IgG3 responses that were comparable in magnitudes to wild-type mice, suggesting that this B cell response was independent of cognate T cell help. Notably, the titer of the IgG in wild-type animals could be increased by more than 200-fold upon replacement of liposomes with bacteriophage Qβ virus-like particles that displayed the same self-antigen peptide at comparable epitope densities. This enhancement was lost almost completely in gene knockout mice lacking either TCRs or MHC class II molecules on B cells. In conclusion, epitope density above a threshold on particulate Ags can serve as a stand-alone signal to trigger secretion of autoreactive and class-switched IgG in vivo in the absence of cognate T cell help or any adjuvants. The extraordinary immunogenicity of Qβ viral-like particles relies, in large part, on their ability to effectively recruit T cell help after B cell activation.

Conventional cancer vaccines based on soluble vaccines and traditional adjuvants have produced suboptimal therapeutic efficacy in clinical trials. Thus, there is an urgent need for vaccine technologies that can generate potent T cell responses with strong anti-tumor efficacy. We have previously reported the development of synthetic high-density protein (sHDL) nanodiscs for efficient lymph node (LN)-targeted co-delivery of antigen peptides and CpG oligonucleotides (a Toll-like receptor-9 agonist). Here, we performed a comparative study in mice and non-human primates (NHPs) to identify an ideal vaccine platform for induction of CD8+ T cell responses. In particular, we compare the efficacy of CpG class B, CpG class C, and polyICLC (a synthetic double-stranded RNA analog, a TLR-3 agonist), each formulated with antigen-carrying sHDL nanodiscs. Here, we report that sHDL-Ag admixed with polyICLC elicited robust Ag-specific CD8+ T cell responses in mice, and when used in combination with α-PD-1 immune checkpoint inhibitor, sHDL-Ag + polyICLC completely eliminated large established (~100 mm(3)) MC-38 tumors in mice. Moreover, sHDL-Gag + polyICLC induced robust Simian immunodeficiency virus Gag-specific, polyfunctional CD8+ T cell responses in rhesus macaques and could further amplify the efficacy of recombinant adenovirus-based vaccine. Notably, while both sHDL-Ag-CpG-B and sHDL-Ag-CpG-C generated strong Ag-specific CD8+ T cell responses in mice, their results were mixed in NHPs. Overall, sHDL combined with polyICLC offers a strong platform to induce CD8+ T cells for vaccine applications.

Recent studies have shown that certain combinations of Toll-like receptor (TLR) agonists can induce synergistic immune activation. However, it remains challenging to achieve such robust responses in vivo in a manner that is effective, facile, and amenable for clinical translation. Here, we show that MPLA, a TLR4 agonist, and CpG, a TLR9 agonist, can be efficiently co-loaded into synthetic high-density lipoprotein nanodiscs, forming a potent adjuvant system (ND-MPLA/CpG) that can be readily combined with a variety of subunit antigens, including proteins and peptides. ND-MPLA/CpG significantly enhanced activation of dendritic cells, compared with free dual adjuvants or nanodiscs delivering a single TLR agonist. Importantly, mice immunized with physical mixtures of protein antigens ND-MPLA/CpG generated strong humoral responses, including induction of IgG responses against protein convertase subtilisin/kexin 9 (PCSK9), leading to 17–30% reduction of the total plasma cholesterol levels. Moreover, ND-MPLA/CpG exerted strong anti-tumor efficacy in multiple murine tumor models. Compared with free adjuvants, ND-MPLA/CpG admixed with ovalbumin markedly improved antigen-specific CD8+ T cell responses by 8-fold and promoted regression of B16F10-OVA melanoma (P < 0.0001). Furthermore, ND-MPLA/CpG admixed with E7 peptide antigen elicited ~20% E7-specific CD8+ T cell responses and achieved complete regression of established TC-1 tumors in all treated animals. Taken together, our work highlights the simplicity, versatility, and potency of dual TLR agonist nanodiscs for applications in vaccines and cancer immunotherapy.

Herein, we report a simple process for the synthesis of pressure-sensitive nanogels that indicate to be appropriate in the delivery of 5-Fu. The hydrogels (Al-CD) prepare by crosslinking of alginate (Al) with modified beta Cyclodextrin (β-CD) as Crosslinker. Next, nanoparticles obtaine by an emulsification method. 5-Fu as model drug loades into the Al-CD nanogels easily by mixing it in aqueous solution with the nanoparticles. The results revealed that the Al-CD nanogels are cytocompatible. They have also a noticeable drug encapsulation (82.1 ±5.7%) while they can release (in vitro controlled) 5-Fu in conditions that imitate the intravascular pressure conditions. These nanogels can rapidly be taken up by HT-29 cells (a colon cell line). In addition, a higher 5-Fu intracellular accumulation and a significant cell death extension by apoptosis mechanism is notice when compare with free 5-Fu. Accordingly, the developed nanogels can be employe as an excellent candidate to overcome the inefficiency of 5-Fu in anticancer treatments and possibly can employe for further evaluation as a chemotherapical agent in applications beyond cancer. © 2017 Wiley Periodicals, Inc. Neutrophil elastase (NE) is a serine protease stored in the azurophilic granules of neutrophils and released into the extracellular milieu during inflammatory response or formation of neutrophil extracellular traps (NETs). Neutrophils release NETs to entrap pathogens by externalizing their cellular contents in a DNA framework decorated with anti-microbials and proteases, including NE. Importantly, excess NETs in tissues are implicated in numerous pathologies, including sepsis, rheumatoid arthritis, vasculitis, and cancer. However, it remains unknown how to effectively prevent NET formation. Here, we show that NE plays a major role during NET formation and that inhibition of NE is a promising approach for decreasing NET-mediated tissue injury. NE promoted NET formation by human neutrophils, and sivelestat, a small molecule inhibitor of NE, inhibited the formation of NETs in vitro. However, in a murine model of lipopolysaccharide-induced endotoxic shock, administration of free sivelestat did not exhibit any efficacy. To improve the efficacy of sivelestat in vivo, we have developed a nanoparticle system for delivering sivelestat. We demonstrate that nanoparticle-mediated delivery of sivelestat effectively inhibited NET formation, decreased the clinical signs of lung injury, reduced NE and other proinflammatory cytokines in serum, and rescued animals against endotoxic shock. Collectively, our data demonstrates that NE signaling can initiate NET formation and that nanoparticle-mediated inhibition of NE improves drug efficacy in prevention of NETs formation.

Here we aimed in developing a new wound dressing using chitosan in combination with polyethylene glycol (PEG) for mechanical and structural properties enhancement the percentages of polymers and method of mixing were major parameters to obtain a proper porous structure with appropriate properties and economical advantage. Hence we used wide ranges of chitosan percentage and molecular weight of PEG, and two methods of fabrication through casting and freeze drying to obtain a novel wound dressing with enhanced properties. After synthesis, different material properties were examined including swelling, biodegradability. Furthermore, samples were mechanically characterized by elastic moduli parameters. Samples with 70% and 50% of chitosan and PEG molecular weight of 2000, synthesized by casting method were selected after meeting the adequate requirements as proper candidates for the wound dressing. Their porous structure with small size pores provided an appropriate environment to enable absorbance drainage and transmission of vapor. They showed superb antibacterial property with high reduction rates of bacterial colonies of E.coli and S.aureus. Moreover, they indicated non-toxicity for L929 fibroblast cells with over 98% viability in MTT assay. Considering appropriate physical, structural and biological properties, the selected samples were introduced as wound dressing candidates.

Hybrid hydrogel fibers were fabricated using hydrogel templates. On page 2146, D. Juncker, A. Khademhosseini, and co-workers show how these fibers containing different fluorescent microbeads have been assembled using textile processes, such as braiding and weaving, to create complex 3D patterns for bioengineering applications. The fabricated constructs with clinically relevant dimensions are mechanically stable.

In this study, a highly selective voltammetric sensor is introduced for the monitoring of meloxicam by using a new carbon paste electrode (CPE). A meloxicam molecularly imprinted polymer (MIP) and a non-imprinted polymer (NIP), polymerizing on the surface of functionalized multiwall carbon nanotubes (MWCNTs), were synthesized and then used for the preparation of carbon paste electrodes (CPEs) to improve the selectivity and conductivity of the prepared sensor. The sensor was applied for the determination of meloxicam using the cathodic stripping voltammetric method. The MIP@MWCNT–CP electrode showed higher recognition ability in comparison to the NIP@MWCNT–CP. Some parameters affecting the sensor response were optimized and then the calibration curve was plotted. Under optimized conditions, two linear dynamic ranges from 4.33 × 10−5 to 2.85 × 10−3 mg L−1 and 2.85 × 10−3 to 7.53 × 10−2 mg L−1 were obtained. The detection limit and quantitation limit of the sensor were calculated to be 3.22 × 10−5 and 1.1 × 10−4 mg L−1, respectively, which indicated the sensitivity of the proposed procedure. The proposed sensor is applied to determine meloxicam in plasma samples and the recovery results were found to be between 98.0 and 102.5% using the voltammetric method with acceptable precision of less than 3.1%.

Biodegradable nanofibrous polymeric substrates are used to fabricate suturable, elastic, and flexible electronics and sensors. The fibrous microstructure of the substrate makes it permeable to gas and liquid and facilitates the patterning process. As a proof-of-principle, temperature and strain sensors are fabricated on this elastic substrate and tested in vitro. The proposed system can be implemented in the field of bioresorbable electronics and the emerging area of smart wound dressings.

We are presenting magnetic molecularly imprinted polymer nanoparticles (m-MIPs) for solid-phase extraction and sample clean-up of paracetamol. The m-MIPs were prepared from magnetite (Fe3O4) as the magnetic component, paracetamol as the template, methacrylic acid as a functional monomer, and 2-(methacrylamido) ethyl methacrylate as a cross-linker. The m-MIPs were then characterized by transmission electron microscopy, FT-IR spectroscopy, X-ray diffraction and vibrating sample magnetometry. The m-MIPs were applied to the extraction of paracetamol from human blood plasma samples. Following its elution from the column loaded with the m-MIPs with an acetonitrile-buffer (9:1) mixture, it was submitted to HPLC analysis. Paracetamol can be quantified by this method in the 1 μg L−1 to 300 μg L−1 concentration range. The limit of detection and limit of quantification in plasma samples are 0.17 and 0.4 μg L−1. The preconcentration factor of the m-MIPs is 40. The HPLC method shows good precision (4.5 % at 50 μg L−1 levels) and recoveries (between 83 and 91 %) from spiked plasma samples.

The optical phase conjugation (OPC) through photonic nanostructures in coherent optics involves the utilization of a nonlinear optical mechanism through real-time processing of electromagnetic fields. Their applications include spectroscopy, optical tomography, wavefront sensing, and imaging. The development of functional and personalized holographic devices in the visible and near-infrared spectrum can be improved by introducing cost-effective, rapid, and high-throughput fabrication techniques and low-cost recording media. Here, we develop flat and thin phase-conjugate nanostructures on low-cost ink coated glass substrates through a facile and flexible single pulsed nanosecond laser based reflection holography and a cornercube retroreflector (CCR). Fabricated one/two-dimensional (1D/2D) nanostructures exhibited far-field phase-conjugated patterns through wavefront reconstruction by means of diffraction. The optical phase conjugation property had correlation with the laser light (energy) and structural parameters (width, height and exposure angle) variation. The phase conjugated diffraction property from the recorded nanostructures was verified through spectral measurements, far-field diffraction experiments, and thermal imaging. Furthermore, a comparison between the conventional and phase-conjugated nanostructures showed two-fold increase in diffracted light intensity under monochromatic light illumination. It is anticipated that low-cost ink based holographic phase-conjugate nanostructures may have applications in flexible and printable displays, polarization-selective flat waveplates, and adaptive diffraction optics.

International audience; Hydrogel templates are formed to entrap various pre-polymers prior to their crosslinking process. Upon the completion of the crosslinking process, an independent polymer network with the same fiber geometry is formed. The hydrogel template can be removed if necessary. As the proof-of-principle, fibers from various polymers are fabricated. The fabricated hybrid polymeric fibers are bioactive and can be bioprinted or assembled using textile processes. The approach can be used for creating complex 3D constructs for various applications.

Commercial lipid emulsion of propofol (CLE) has several drawbacks including pain on injection and emulsion instability. In this paper, a novel nanocarrier system is introduced to improve stability and solubility of the poorly soluble anesthetic drug, propofol, for intravenous administration. In this paper, alginate is modified using a facile method in which the carboxylic group of alginate is grafted to octanol. The octanol–grafted alginate (Alg–C8) is then employed to prepare nanoparticles which are subsequently used for encapsulation of propofol. The nanoparticles are analyzed for their pH, osmolarity, particle size, stability, morphology and sleep recovery and the results are compared with CLE as control. It is revealed that nanoparticles have the average particle size of 180 nm ± 1.2 and spherical morphology which is less than CLE while their pH, osmolarity and profile of release of formulated nanoparticles are similar to those of CLE. In addition, the results show good chemical and physical storage stability for the nanoparticles at room temperature for at least 6 months compared to CLE as control. The animal sleep recovery test on rats shows no significant difference in time of unconsciousness and recovery of the righting reflex between nanoparticles and CLE. It is concluded that encapsulated nanoparticles introduced here could be a promising clinical intravenous system for delivery of poorly soluble anesthetic propofol. In addition, this study provides an efficient and facile method for preparing a carrier system for water insoluble drugs.

Herein, we report a magnetic molecularly imprinted polymers (m-MIPs) using Fe3O4 as a magnetic component, carvedilol as a template molecule for the solid-phase extraction (MISPE) as the sample clean-up technique combined with high-performance liquid chromatography (HPLC) and for the controlled release of carvedilol at different pH values of 1.0 (simulated gastric fluid), 6.8 (simulated intestinal fluid), and 7.4 (simulated biological fluid). The adsorption kinetics was modeled with the pseudo-first-order and pseudo-second-order kinetics, and the adsorption isotherms were fitted with Langmuir and Freundlich models. The performance of the m-MIPs for the controlled release of carvedilol was assessed and results indicated that the magnetic MIPs also have potential applications in controlled drug release. Furthermore, the m-MIPs were applied to the extraction of carvedilol from human blood plasma samples. Carvedilol can be quantified by this method in the 2–350 μg L−1 concentration range. The limit of detection and limit of quantification in plasma samples are 0.13 and 0.45 μg L−1. The results from HPLC showed good precision (3.5% for 50.0 μg L−1) and recoveries (between 85 and 93) using m-MIP from human plasma samples. © 2014 Wiley Periodicals, Inc. J. Appl. Polym. The fabrication of cell-laden structures with anisotropic mechanical properties while having a precise control over the distribution of different cell types within the constructs is important for many tissue engineering applications. Automated textile technologies for making fabrics allow simultaneous control over the color pattern and directional mechanical properties. The use of textile techniques in tissue engineering, however, demands the presence of cell-laden fibers that can withstand the mechanical stresses during the assembly process. Here, the concept of composite living fibers (CLFs) in which a core of load bearing synthetic polymer is coated by a hydrogel layer containing cells or microparticles is introduced. The core thread is drawn sequentially through reservoirs containing a cell-laden prepolymer and a crosslinking reagent. The thickness of the hydrogel layer increases linearly with to the drawing speed and the prepolymer viscosity. CLFs are fabricated and assembled using regular textile processes including weaving, knitting, braiding, winding, and embroidering, to form cell-laden structures. Cellular viability and metabolic activity are preserved during CLF fabrication and assembly, demonstrating the feasibility of using these processes for engineering functional 3D tissue constructs.

A new method to conjugate methoxy polyethylene glycol (mPEG) to C6 position of chitosan under the mild condition is introduced that improves the biocompatibility and water solubility of chitosan. Harsh deprotecting step and several purification cycles are two major disadvantages of the current methods for preparing PEGylated chitosan. In this study, the amine groups at C2 position of chitosan are protected using SDS followed by grafting the PEG. The protecting group of chitosan is simply removed by dialyzing against Tris solution. The chemical structure of the prepared polymer is characterized by FTIR and 1H NMR spectroscopy. Fourier transformed-infrared (FT-IR) and 1H NMR spectra confirmed that the mPEG is successfully grafted to C6 position of chitosan. Prepared methoxy polyethylene glycol (mPEG) is then employed to prepare the nanocapsules for the encapsulation of poor water-soluble drug, propofol. The TEM, AFM, and DLS techniques are used to characterize the prepared nanocapsules size and morphology. The results show a size of about 80 nm with spherical shape for nanocapsules. In vitro drug release is carried out to evaluate the potential of nanocarriers for the intravenous delivery of drugs. The profile of release from formulated nanocapsules is similar to those of commercial lipid emulsion (CLE). In vivo animal sleep-recovery test on rats shows a close similarity between the time of unconsciousness and recovery of righting reflex between nanoparticles and CLE. This study provides an efficient, novel, and easy method for preparing a carrier system that requires less intensive reaction conditions, fewer reaction steps, and less purification steps. In addition, the nanocapsules introduced here could be a promising nano carrier for the delivery of poor water-soluble drugs.

Current methods for preparation of PEGylated chitosan have limitations such as harsh de protecting step and several purification cycles. In the present study, a facile new method for conjugating methoxy polyethylene glycol (mPEG) to chitosan under mild condition is introduced to improve water solubility of chitosan and control the release of poor water soluble drugs. The method consists of chitosan modification by grafting the C6 position of chitosan to mPEG which is confirmed by Fourier transformed-infrared (FT-IR) and proton nuclear magnetic resonance ((1)HNMR) analyses. The amine groups at the C2 position of chitosan are protected using sodium dodecylsulfate (SDS) which is removed by dialyzing the precipitation against Tris solution. The chemical structure of the prepared polymer is characterized by FTIR and (1)HNMR. The synthesized polymer is then employed to prepare nanoparticles which are characterized by transmission electron microscopy (TEM), atomic force microscopy (AFM), scanning electron microscopy (SEM), and dynamic light scattering (DLS) for their size and morphology. The nanoparticles are used for encapsulation of ibuprofen followed by in vitro release investigation in gastrointestinal and simulated biological fluids. The chitosan nanoparticles are used as control. The PEGylated nanoparticles show a particle size of 80 nm with spherical morphology. The results clearly show that drug release from PEGylated chitosan nanoparticles is remarkably slower than chitosan. In addition, drug encapsulation and encapsulation efficiency in PEGylated nanoparticles are dependent on the amount of drug added to the formulation being significantly higher than chitosan nanoparticles. This study provides an efficient, novel, and facile method for preparing a nano carrier system for delivery of water insoluble drugs.

A polyacrylate tetracycline (TC) selective microporous molecularly imprinted polymer was prepared in three different porogenic solvents (chloroform, acetonitrile, and methanol) via precipitation polymerization, using methacrylic acid monomer, ethylene glycol dimethacrylate crosslinker, and TC as template. In all three solvents this method produced microporous particles in the scale range (200–400 nm), simply, quickly, cleanly, and in good yield. The effect of polarity of porogenic solvents on binding capacity was investigated. The imprinted polymer prepared in chloroform gave much higher binding capacity (KD = 198.6) for TC than the polymers prepared in acetonitrile (KD = 133.2) or methanol (KD = 104.7). The selectivity of imprinted polymers was evaluated by rebinding other structurally similar compounds. The results clearly indicated that the imprinted acrylate polymer exhibits an excellent selectivity toward TC, and has better ability to control the release of TC than the non-imprinted polymer.© 2012 Wiley Periodicals, Inc. Alginate is a family of natural polysaccharides, widely used in industry and medicine for many applications, with its non toxic nature, gentle sol/gel transition procedure and low cost, alginate inferior biomechanical properties have limited its utility especially in tissue engineering. Additionally, ionically cross-linked alginate hydrogels generally lose most of their initial mechanical and swelling properties within a few hours in physiological solution. In order to overcome these limitations, the referenced alginate was treated by covalent fixation of octadecyl chains onto the polysaccharide backbone by esterification. In semi dilute solution, intermolecular hydrophobic interactions of long alkyl chains result in the formation of physical hydrogels, which can then be reinforced by the addition of calcium chloride. FTIR studies clearly showed the presence of ester bonds at 1612 and 1730 cm(-1) indicating that the alkyl groups are incorporated in the backbone of resulting polymer. The endothermic peak and exothermic peak present in the DSC thermogram of Alg-C18 had shifted to lower temperatures comparing to native alginate (from 106 to 83°C and from 250 to 245°C, respectively) due to the esterification reaction that leads to high hydrophobic nature of the modified sample. From rheological experiments, it can be inferred that the combination of both calcium bridges and intermolecular hydrophobic interaction in the treated alginate leads to enhanced gel strength accompanied by more stable structure in physiological solution comparing to native sodium alginate hydrogel. Finally, the modified alginate tended to have no toxic effects on mesenchymal stem cell culture, rather it supported MSC chondrogenic differentiation.
