20. Emamectin Benzoate-Induced Hepatotoxicity in Rats with Special Reference to Protective Potential of Nigella sativa Oil

Abstract

This study was designed to explore the hepatotoxicity of emamectin in male rats and the possible effect of Nigella sativa oil (NSO) in ameliorating this. Twenty-eight male rats were used in this study. They were divided into four groups, Control group: rats orally administered distilled water; NSO group: rats administered NSO orally; EMB group: rats administered emamectin benzoate orally; and EMB+NSO group: rats orally co-administered NSO with EMB, with the administrations being performed every other day for 6 weeks. Body weight was measured, liver alanine aminotransferase (ALT), aspartate aminotransferase (AST), and alkaline phosphatase (ALP) activities were determined, and total protein and albumin levels were recorded. Histopathological examination of the liver was also performed, along with caspase-3 and TNF-α immunostaining of liver tissue. EMB treatment resulted in decreased body weight, while the co-administration of NSO modulated the EMB-induced alterations in body weight. There were also increases in the activities of serum ALT, AST, and ALP and decreases in total protein and albumin levels in the EMB group. Co-treatment with NSO significantly reduced serum ALT, AST, and ALP and improved total protein and albumin levels. Histopathological examination of the liver in the EMB group revealed the presence of different histopathological alterations that were improved by the co-administration of NSO. Immunostaining of caspase-3 and TNF-α in the liver revealed strong expression in the EMB-treated group. Meanwhile, the EMB+NSO group showed weak positivity for immunoreactive cells.

Abstract

Backgorund and Objective:Acute upper GIT bleeding is a life threatening emergency. Many scoring systems have been developed to evaluate patients with acute UGIB. The purpose of this study was to confirm value of Modified Blatchford scoring system, Full rockall score , AIM65 score &lactate level as a predictor of outcome as regard patient’s mortality, their need for ICU, duration of hospitalization and need for endoscopic intervention in the emergency department. Patients an Methods:This study was performed on 100 patients who presented to the emergency department with acute UGIB in Alexandria main university hospital. All the study scores (GBS, AIMS65, PRS, and FRS) were able to predict the clinical outcomes in the UGIB patients. Results:GBS was the best performing risk score among the four scores for predicting all the clinical outcomes except the prediction of endoscopic intervention. Lactate was a sensitive predictor for ICU admission, hospital stay, rebleeding and mortality during first 48 hrs of admission in patients presenting with UGIB.

Abstract

The Pseudomonas putida strain was primarily identified and tested in vitro against antibiotic sensitivity for several antibiotics using the disc diffusion method. This isolate was also tested against sensitivity to carvacrol oil (c) and formic acid (f). The genotyping of Pseudomonas spp. and virulotyping for P. putida isolate was carried out and verified by 16S rDNA-PCR amplification. Furthermore, we assessed the efficacy of carvacrol oil and formic acid in vivo for treatment of P. Putida infection. For the in vivo challenge, 180 fish (Nile tilapia, Oreochromis niloticus) were divided into six groups: (G1: control (unchallenged), G2: carvacrol prophylaxis (3 g/kg), G3: formic acid prophylaxis (5 mL/kg), G4: control positive (challenged), G5: carvacrol treatment (3 g/kg), and G6: formic acid treatment (5 mL/kg); 30 fish per group) with three replicates. Following the challenge, nitric oxide and lysozyme activity were measured as essential indicators for fish immunity. The antioxidant parameters including SOD and catalase were computed to reflect the antioxidant status. Furthermore, relative percent survival (RPS) and mortality percent were evaluated to indicate functional immunity. The findings of the antibiotic sensitivity test showed that ciprofloxacin exhibited the largest inhibition zone. Additionally, formic acid (f) displayed the greatest inhibition zone compared to carvacrol oil (c) and was more effective in stimulating the immune-antioxidant response compared to carvacrol oil. The tested exotoxin A (tox A), exoenzyme S (exo S), and the nan1 associated-virulence genes were identified in the P. putida isolate. Overall, the current study verified the virulence of P. putida and highlighted the promising role of dietary addition of formic acid for enhancing the immune-antioxidant indicators and for mitigating P. putida infection. 40. IMPACT OF THE ENVIRONMENTAL TEMPERATURE ON THE CHRONOLOGICAL SEQUENCES OF POSTMORTEM CHANGES IN HEPATIC AND RENAL TISSUES OF ALBINO MICE (Mus Musculus).

Abstract

The current study was carried out to detect the effect of the environmental temperature on the postmortem histological and molecular changes and their synchrony with the time since death (TSD). A total of 30 male, albino mice were sacrificed by decapitation. Carcasses have been divided into two groups, 15 in each group. The first group left at room temperature (25±3 °C) and the second one kept in the refrigerator (4 °C). Tissue specimens from livers and kidneys have been taken at a definite time interval (0, 6, 12, 24, and 36 hours after death). Tissue specimens were processed into paraffine-embedded sections stained with Hematoxylin and Eosin to investigate the histological changes. Liver and kidney taken directly after death from both animal groups showed the normal structures. However, time-dependent degenerative changes have been seen in both organs throughout the postmortem time periods (6, 12, 24, and 36 hours after death). These changes influenced both the tissue architectural configuration and the cellular integrity. The cellular changes involved cytoplasmic and nuclear elements. The cytoplasmic changes ranged from partial to complete vacuolation. The nuclear changes included pyknosis, karyorrhexis, and karyolysis. Preserving animal carcasses at a lower temperature delayed the progression of these changes. Also, the molecular findings showed that the integrity of DNA from liver and kidney either kept at room temperature or in refrigerator (4 °C) was lost in a time-dependent manner. Yet, DNA degradation occurred at a slower rate at low temperatures. In conclusion, it was evident that holding the carcasses of mice at lower temperatures may delay degenerative changes and DNA degradation. Moreover, the association of both histological and molecular testing may support each other in the determination of the TSD, especially in the earlier postmortem periods. [ABSTRACT FROM AUTHOR]

Abstract

Psammomys obesus is a terrestrial rodent's species of gerbil subfamily belong to Muridae. Zoonotic diseases associated with rodents; pose a significant threat to human health and animals. Mouse whipworm Trichuris muris has been used as a tractable model of human trichuriasis. Zoonotic cestode Raillietina sp. causes public health problem when moving from wild habitats to those inhabited by humans. Endoparasitic agents, which can be very serious in human and livestock health, are mostly homed by wild rodents. The current study investigates intestinal helminth infestation of Psammomys obesus. Thirtyseven adult rats were captured from saline marsh areas of North Coast, Egypt. Rats were examined for intestinal parasite with morphological and pathological features. Thirty-three rats were found infected with: Trichuris muris, 16 (48.48, %); Raillietina trapezoides, 4 (12.12, %); mixed infection of Trichuris muris and Raillietina trapezoides, 5(15.15%). Gross examination of both small and large intestines revealed intestinal blockage and diarrhea. Histopathological changes of the intestine showed degeneration of the intestinal mucosa; and atrophy of the villi and intestinal glands. Different parts of worms observed in the lumen of colon mucosa and at the tip of villi results in discontinuity of the muscularis mucosa and necrotic lesions on the mucosa. Also, necrotic colitis and enteritis were observed. Two types of anemia were confirmed by hematological analysis. Moreover, two types of eggs were noticed. The helminths infecting Psammomyes obesus were morphologically identified with their pathological effects. Further studies are needed to assess transmitted zoonotic parasitic diseases impact public health in Egypt. [ABSTRACT FROM AUTHOR]
