Abstract

B cells are frequently found in the margins of solid tumours as organized follicles in ectopic lymphoid organs called tertiary lymphoid structures (TLS)1,2. Although TLS have been found to correlate with improved patient survival and response to immune checkpoint blockade (ICB), the underlying mechanisms of this association remain elusive1,2. Here we investigate lung-resident B cell responses in patients from the TRACERx 421 (Tracking Non-Small-Cell Lung Cancer Evolution Through Therapy) and other lung cancer cohorts, and in a recently established immunogenic mouse model for lung adenocarcinoma3. We find that both human and mouse lung adenocarcinomas elicit local germinal centre responses and tumour-binding antibodies, and further identify endogenous retrovirus (ERV) envelope glycoproteins as a dominant anti-tumour antibody target. ERV-targeting B cell responses are amplified by ICB in both humans and mice, and by targeted inhibition of KRAS(G12C) in the mouse model. ERV-reactive antibodies exert anti-tumour activity that extends survival in the mouse model, and ERV expression predicts the outcome of ICB in human lung adenocarcinoma. Finally, we find that effective immunotherapy in the mouse model requires CXCL13-dependent TLS formation. Conversely, therapeutic CXCL13 treatment potentiates anti-tumour immunity and synergizes with ICB. Our findings provide a possible mechanistic basis for the association of TLS with immunotherapy response.

Abstract

The steel industry is the most relevant sector in emerging economies due to its application in numerous fields. However, steel manufacturing involves large energy investment and produces significant greenhouse gas emissions. The current world economic and environmental scenario therefore necessitates that improvements in the footprint of the steel industry be made without affecting its viability. Considering the present challenge, we report two possible processes for Carbon Capture and Utilization (CCU). The first process is the competitive capture of CO2–SO2, followed by CO2 valorisation to methane. However, the CO2 capture capacity and lifetime for the adsorbent after multiple cycles could be improved through preliminary desulphurization of the gas current. The improved system demonstrates net profitability in a typical stainless steel plant. Therefore, it can be implemented in an industrial setting without profitability loss to steelmaking operations, fulfilling bot the goal of reducing CO2 emissions while protecting the mainstay of the plant.

[EN] Introduction: An addiction can be defined as a pathological state that leads to dependence, blocks the control of different aspects of people¿s daily lives, influences conscience and diminishes interests in learning and communicating. Information and communication technologies are a series of electronic devices that are increasingly being used by the general population that have helped create a new lifestyle, especially for youths. Their suitable use entails many advantages and benefits, but their improper use can mean serious problems, of which inappropriate sleeping and eating habits stand out, which have several repercussions on the lives of young people. Surfing the Internet, using a mobile telephone, playing video games or watching TV have changed people¿s lifestyle. This new lifestyle is strongly influencing the young people who were born in this era of advanced technologies. Therefore, we can state that the new technologies field is one of the aspects in which young people possess considerable knowledge and skills. Objective: To know the impact of addiction to new technologies and their excessive use on young people¿s sleeping and eating habits. Method: the literature search was done in various databases via PubMed, Scielo, ProQuest and Wiley Online Library. Twelve articles that met the set criteria, conducted on excessive use or addiction of ICT and about sleeping and eating habits, were analysed. Results: The studied population included both children and adolescents (8-26 years) because the use of ICT is widespread in both these life stages. The most widely used technology was the Internet, which is a tool found in almost any electronic device. All the studies stated that prolonged ICT use makes young people more prone to sleepiness in the morning. They also confirmed that the likelihood of daytime sleepiness in Internet addicts is 5.9-fold higher than for non-addicts, and that the more time they spend using electronic means, the later they go to bed (

Abstract

We describe the discovery and characterization of antibodies with potential broad SARS-CoV-2 neutralization profiles. The antibodies were obtained from a phage display library built with the VH repertoire of a convalescent COVID-19 patient who was infected with SARS-CoV-2 B.1.617.2 (Delta). The patient received a single dose of Ad5-nCoV vaccine (ConvideciaTM, CanSino Biologics Inc.) one month before developing COVID-19 symptoms. Four synthetic VL libraries were used as counterparts of the immune VH repertoire. After three rounds of panning with SARS-CoV-2 receptor-binding domain wildtype (RBD-WT) 34 unique scFvs, were identified, with 27 cross-reactive for the RBD-WT and RBD Delta (RBD-DT), and seven specifics for the RBD-WT. The cross-reactive scFvs were more diverse than the RBD-WT specific ones, being encoded by several IGHV genes from the IGHV1 and IGHV3 families combined with short HCDR3s. Three cross-reactive scFvs and one RBD-WT specific scFv were converted to human IgG1 (hIgG1). The four antibodies blocked the RBD-WT binding to angiotensin converting enzyme 2 (ACE2). Importantly, one of the antibodies also recognized the RBD from the B.1.1.529 (Omicron) isolate, implying that the VH repertoire of the convalescent patient would protect against SARS-CoV-2 Wildtype, Delta, and Omicron. From a practical viewpoint, the triple cross-reactive antibody provides the substrate for developing therapeutic antibodies with a broad SARS-CoV-2 neutralization profile. [ABSTRACT FROM AUTHOR]

Abstract

This article presents the results of a post-mortem study of various MgO-C and MgO-C-Al pieces that are used in the electric furnaces at the Acerinox factory in Cádiz. From the chemical, thermal, mineralogical and microstructural characterization of the materials prior to use and post-mortem it has been established that, in both types of refractories, the corrosion has lead to the formation of reaction layers with different chemical and mineralogical composition, as a function of temperature and oxygen partial pressure, ranging from the hot to the cold face and strongly depending on the zone of the electric furnace where the piece is situated. Changes in graphite content have been observed in both types of materials by X-ray diffraction. These changes have been quantified by Differential Thermal Analysis. In MgO-C-Al refractories it has been observed the formation of small amounts of C3Al4 inside the pieces and MgAl2O4 in the refractories’ hot face. Keeping in mind the obtained results and the phase equilibrium diagram of Mg-C-Al-O it has been established a corrosion mechanism for both types of materials. It cannot be established by the present results that additions of Al (≈ 2 % in weight) improve the performance of MgO-C materials.Se describen los resultados del estudio post-mortem de diversas piezas de MgO-C y MgO-C-Al utilizadas en los hornos eléctricos de la factoría de Acerinox en Cádiz. De la caracterización química, térmica, mineralógica y microestructural de los materiales sin usar y post-mortem se ha establecido que la corrosión ha dado lugar, en ambos tipos de refractarios, a la formación de capas de reacción con distinta composición química y mineralógica, en función de la temperatura y presión parcial de oxígeno en la pieza, que varían desde la cara caliente a la cara fría y dependen fuertemente de la zona del horno eléctrico en que está situada la pieza. En ambos tipos de materiales se han observado por difracción de rayos X cambios en el contenido de grafito con la distancia a la cara fría. Estos cambios se han cuantificado mediante análisis térmico diferencial. En los refractarios de MgO-C-Al se observa la formación de pequeñas cantidades de C3Al4 en el interior de las piezas y de MgAl2O4 en la cara caliente del refractario. Teniendo en cuenta los resultados obtenidos y los diagramas de equilibrio de fases Mg-C-Al-O se ha establecido el mecanismo de corrosión en ambos tipos de materiales. De los resultados obtenidos no se puede establecer que adiciones de Al (≈ 2% en peso) mejoren el comportamiento en servicio de los materiales de MgO-C.

Abstract

The work has been done with ten AISI 303 stainless steel rods which have similar chemical compositions, except for sulphur which varies between 0.25 and 0.36%. These steels are studied by Scanning Electron Microscopy (SEM) and Energy Dispersive X-Ray Spectrometry (EDX). The sulphides are chemically microanalysed and the occupied volume determined by automatic image analysis. It can be inferred that the chemical composition of sulphides in AISI 303 steels is variable to some extent even though the steel chemistry is almost invariable. This could mean that solidification conditions and thermomechanical transformations are able to affect resulting in variations of sulphide compositions. Quantitative relations between sulphur and sulphide contents are established and the influence of sulphide chemical compositions on those relations is studied. The experimental results are compared with those obtained from theoretical calculations. A good approach is obtained when the difference between chemical compositions of real sulphides and the theoretical one, MnS, is taken into account. Se trabaja con una serie de diez redondos de aceros inoxidables tipo AISI 303 de composiciones químicas prácticamente iguales a excepción del azufre que varía entre 0,25 y 0,36%. Se examinan los aceros mediante Microscopía Electrónica de Barrido (MEB) y Espectrometría de Dispersión de Energías de Rayos X (EDX) acoplada al MEB, analizándose químicamente los sulfuros y determinando, mediante análisis automático de imágenes, los volúmenes ocupados por los sulfuros. Parece deducible que la composición química de los sulfuros en el AISI 303 puede ser discretamente variable aún siendo prácticamente invariable la composición química del acero. Ello podría significar que las condiciones de solidificación y las transformaciones termomecánicas pueden influir, dando como resultado variaciones en la composición química de los sulfuros. Se establecen relaciones cuantitativas entre los contenidos de azufre y de sulfuros y se estudia la influencia que en dichas relaciones tienen las composiciones químicas de los sulfuros. Se comparan los resultados experimentales con los obtenidos a partir de cálculos teóricos, obteniéndose un buen grado de aproximación cuando se tiene en cuenta que los sulfuros reales tienen composiciones químicas distintas de la del sulfuro teórico, MnS.

The oxidation kinetics in air up to 50 h at 1,373 and 1,473 K of three refractory stainless steels (25Cr20Ni, 21Cr11Ni(1), 21Cr11Ni(2)) are studied. At 1,373 K, the best results are those of 25Cr20Ni followed by those of 21Cr11Ni(1), with 0,036 % lanthanides, and the worst behaviour is that of 21Cr11Ni(2), with 0,010 % lanthanides. A 2 h oxidation treatment at 1,323 K in air produces oxidation layers from 1 to 3 μm in thickness for the three materials. Scanning electron microscopy and energy dispersive microanalysis (SEM-EDX) and X-ray diffraction (XRD) characterisation of the oxidation layers give the same characteristics for the three steels: Mn Cr2O4 spinel type in the outer part, Cr2O3 in the inner one, with a SiO2 barrier in the oxide-metal interface and a large amount of internal and intergranular precipitates (also Si02) in 21Cr11Ni materials. After ageing tests, at 1,073 K for 400 h, about 10 vol. % of the σ-phase is present in the 25Cr20Ni (AISI 310 S) material, while no a-phase is present in the 21Cr11Ni materials perhaps due to the high nitrogen content. The application of field emission SEM with EDX allows the analysis of σ-phase and carbides, nitrides and carbonitrides present. The steel 25Cr20Ni only forms, in addition to the σ-phase, M23C6 type carbides, while the steel 21Cr11Ni(1) forms these carbides and M15N nitrides (M8N in the surface) and the steel 21Cr11Ni(2) forms mainly carbonitrides M6(CN). The relatively higher nitrogen level in the 21Cr11Ni(1) alloy could explain the different behaviour of both 21Cr11Ni steels. Se parte de tres aceros inoxidables austeníticos refractarios (25Cr20Ni, 21Cr11Ni(1), 21Cr11Ni(2)) y se estudian las cinéticas de oxidación al aire, hasta 50 h a 1.373 y 1.473 K. El mejor comportamiento a 1.373 K corresponde al acero 25Cr20Ni seguido de cerca del 21Cr11Ni(1) con 0,036 % de lantánidos, mientras que el peor comportamiento corresponde al 21Cr11Ni(2) con 0,010 % de lantánidos. A 1.473 K también se comporta mejor el 25Cr20Ni, a distancia de los otros inoxidables. Un tratamiento de oxidación al aire de 2 h a 1.323 K da lugar a capas de oxidación de 1 a 3 μm de espesor en los tres materiales y, en el examen de las capas de óxidos mediante microscopía electrónica de barrido (MEB), microanálisis por dispersión de energías de rayos X (EDX) y difracción de rayos X (DRX), se observan las mismas características en los tres aceros: espinelas tipo MnCr2O4 en el exterior, Cr2O3 en el interior, con barreras de SiO2 en la interfase óxido-metal y precipitados internos e intergranulares (también de SiO2) muy abundantes en los aceros 21Cr11Ni. Los ensayos de envejecimiento de 400 h a 1.073 K producen en el acero 25Cr20Ni (AISI 310 S) alrededor de un 10 % vol. de fase σ mientras que los aceros 21Cr11Ni o no la forman o lo hacen en muy pequeña cantidad, pudiendo influir en ello los elevados niveles de nitrógeno de estos últimos. La aplicación de la MEB con filamento de emisión de campo (EC) con EDX permite el análisis de la fase a así como de los carburos, nitruros y carbonitruros presentes. El acero 25Cr20Ni sólo forma, junto con a, carburos tipo M23C6, mientras que el acero 21Cr11Ni(1) forma estos carburos y nitruros M15N (M8N en la superficie) y el acero 21Cr11Ni(2)preferentemente forma M6(CN). Esta diferencia de comportamiento de los aceros 21Cr11Ni también puede ser imputable a los niveles de nitrógeno relativamente mayores de la aleación 21Cr11Ni(1).

Abstract

Three stainless steels: 25Cr20Ni, 21Cr11Ni(1) and 21Cr11Ni(2) have been oxidised in air at 1,323 K for 2 h. The three materials form oxide layers 1 to 3 μm thick with MnCr2O4 external crystals and Cr2O3 inner ones. There are a SiO2 barrier below Cr2O3, in the oxide-metal interface. SiO2 also forms intra and intergranular precipitates in the metal, which are specially abundant in 21Cr11Ni materials. The theoretical predictions about σ-phase formation tendencies are confirmed after a treatment at 1,073 K for 400 h: 25Cr20Ni forms 10 % vol. σ-phase, while both 21Cr11Ni form few σ-phase or they do not form it at all. Se oxidan al aire durante 2 h a 1.323 K tres aceros inoxidables: 25Cr20Ni, 21Cr11Ni(1) y 21Cr11Ni(2). Los tres materiales forman capas de óxido de 1 a 3 μm de espesor, con cristales externos de MnCr2O4 e internos de Cr2O3. Debajo del Cr2O3, en la intercara óxido-metal, se forma SiO2, en forma de precipitados intra e intergranulares en el metal, especialmente abundantes en los 21Cr11Ni. Las predicciones teóricas sobre las tendencias a la formación de fase σ se ven confirmadas tras un tratamiento de 400 h a 1.073 K; el 25Cr20Ni forma un 10 % en volumen de σ, mientras que los aceros 21Cr11Ni o no forman σ, o lo hacen escasamente.

34. Evolution of the structural repertoire of the human V(H) and Vkappa germline genes.

Abstract

Variable genes of human Ig are classified in families and clans which reflect the early events of gene duplication in the evolution of the locus. This organization in multiple copies of variable genes plus the somatic processes of recombination and hypermutation allows the immune system to generate an antibody repertoire of great diversity. At present the role that somatic processes play in the generation of that diversity is understood with some detail. It is a matter of hard controversy, however, which selective pressures have shaped the evolution of the germline genes of Ig and, consequently, what the role of this germline component in the generation of the antibody diversity actually is. Previous studies of our group have showed that the structural repertoire of Ig-determined by the canonical structures-is an important factor to determine the recognition properties of the antibodies. Complete knowledge of the sequences of the human V(H) and Vkappa loci is available to analyze the evolution of the structural repertoire of these loci. Two phylogenetic gene trees were built from the functional germline genes and the evolution of the structural repertoire was studied. We report that for both loci the canonical structures are not randomly distributed within the tree. Conversely, it is shown that the evolution of the structural repertoire follows a gradual process of diversification. This indicates a correlation between the evolution of genes and the structural repertoire, although important differences are found in the patterns of evolution of the structural repertoire between V(H) and Vkappa. Based on those results we propose a primordial structural repertoire for V(H) and Vkappa. The general properties and an outline of the three-dimensional structure of this primordial repertoire are given.

Foe is probably not J.M. Coetzee's best known novel, although it is a text of great importance because of the way in which its political, literary and theoretical values are interrelated. The novel addresses a foundational myth of Western societies in the figure of Robinson Crusoe, and draws attention to its textual quality. This concern with the process of representation and the narrative quality of our beliefs is also manifested throughout the novel in other issues. Thus, in the text there is a whole panoply of reflections about the central issues affecting the very mechanics of constructing a text, such as, for example, the proper way a story should be written, the relationship between representation and its referent in the real, the problem of realism, or the question of authorship.

Abstract

18-8 stainless steel is one of the most versatile materials. Some of its uses are related to structural applications; in this case, the high mechanical properties required are obtained by controlled lamination after re-crystallisation annealing. In this work, detailed analyses by FEG-SEM and EBSD have been done to correlate the mechanical properties, the amount of martensite and the structural misorientation generated. A linear relation between yield strength and local misorientation has been obtained.

Abstract

James Joyce's Ulysses has been traditionally read as the great work of Modernism, characterized by its encyclopaedic, totalizing quality, while developing techniques to help create that effect. In spite of initial forebodings, the reader becomes accustomed to a certain narrative stability in the text. However, from "Sirens" onwards, the reader begins to feel that s/he is reading a different novel, different in fact with each new episode. The reflexion upon the quality of the episodes of the second half of the novel should force the reader to reconsider as well the previous episodes. In this way s/he will find evidence (such as the breakdown of the epistemological component or the equation between language and reality) which enables the reader to re-assess the novel also from the premises of postmodernism.

Abstract

In this study, four AISI 430 (EN 1.4016) steel samples of different initial composition and annealing treatment were submitted to uni-axial tension deformation. The as-supplied and deformed materials were characterized by electron channelling contrast imaging, electron backscatter diffraction and electron probe microanalysis. The results show a clear correlation between chemical composition, annealing treatment and strain level obtained until fracture.

47. An inter-laboratory comparison of EPMA analysis of alloy steel at low voltage

Abstract

We present the results of an inter-laboratory comparison of EPMA analysis at low voltage (5-6 kV) of three monophasic alloy steel samples. The aim of the work was to obtain an estimate of the present situation of low-voltage analysis of steel and identify needs for improvement. EPMA analyses of the samples were conducted by seven participant groups using electron microprobes and scanning electron microscopes of different kind, equipped with wavelength- and/or energy-dispersive X-ray spectrometers, and employing their own methodology of analysis. The results using WDS showed essentially an underestimation of the Cr contents, with relative deviations from the reference values ranging from -0.7 % to -17 %, and an overestimation of Fe and Ni, with relative deviations from the reference composition ranging from -4 % to +30 %, and from +14 % to +42 %, respectively. The relative deviations obtained by using EDS showed larger scatter, ranging from -16 % to +54 % for Cr, -0.4 % to +66 % for Fe and from +13 % to +90 % for Ni. Reasons for the differences observed and the scatter of results are discussed.

Austenitic-ferritic stainless steels are formed by ferrite and austenite in a variable proportion between 30 % and 70 %. Their chemical composition conditions the ageing processes that can happen during heat treatments, where phases that hardens and brittles the material can be generated. Evolution of a new duplex stainless steel with lower nickel and higher nitrogen and manganese content maintained at 1123 K (critical precipitation temperature) up to 8 hours compared to classical ones (2205 and 2507) is presented. The study, done by magnetic measurements, XRD, optical and scanning electron microscopy and microanalysis, revealed a roughly similar behaviour for the three steels. The new one presents a higher initial stability, probably due to the high nitrogen and manganese contents; however, it also produces the higher final transformation due to the higher initial ferrite. A fine comparative analysis was done to correctly identify any phase; an accurate microanalysis in every generated phase was performed. All the steels tested are initially made on ferrite and austenite. After the ageing treatment, 8 hours at 1123 K, in the 2205 duplex steel a strong precipitation of sigma phase is observed, austenite and some of the original ferrite are also present; ferrite completely transforms in the 2507 duplex and austenite, sigma, secondary austenite and chromium nitride can be found; regarding the new duplex steel DBNi, austenite, ferrite, sigma, chi and chromium nitrides are detected.Los aceros dúplex austeno-ferríticos están constituidos por ferrita y austenita en proporciones variables del 30 % al 70 %. Su composición química condiciona los procesos de envejecimiento que sufren durante los tratamientos térmicos, cuando se generan fases que endurecen y fragilizan el material. Este trabajo estudia la evolución de un nuevo inoxidable dúplex con bajo contenido en níquel y alto en nitrógeno y manganeso a 1123 K (temperatura crítica de precipitación) y tiempos de hasta 8 horas, comparándola con las de dos dúplex clásicos (2205 y 2507). El estudio, mediante medidas magnéticas, DRX y microscopías óptica y electrónica de barrido con microanálisis, reveló un comportamiento general similar en los tres aceros. El nuevo material presenta mayor estabilidad inicial, probablemente debido al elevado contenido en nitrógeno y manganeso, aunque produce la mayor cantidad de transformación final debido a la mayor cantidad inicial de ferrita. Para la identificación correcta de las fases se realizó un análisis comparativo determinando las composiciones químicas de las fases generadas. Todos los aceros ensayados están inicialmente constituidos por ferrita y austenita. Tras el envejecimiento, 8 horas a 1123 K, en el acero dúplex 2205 se observa una abundante formación de sigma, siguen presentes la austenita y parte de la ferrita original; en el dúplex 2507 la ferrita transforma totalmente y se identifican austenita, sigma, austenita secundaria y nitruros de cromo; respecto al nuevo dúplex DBNi, tras el tratamiento, se detecta austenita, ferrita, fases sigma y chi y nitruros de cromo.
