Abstract

Overnutrition or undernutrition during all or part of the reproductive cycle predisposes sows to metabolic consequences and poor reproductive health which contributes to a decrease in sow longevity and an increase in perinatal mortality. This represents not only an economic problem for the pig industry but also results in poor animal welfare. To maximise profitability and increase sustainability in pig production, it is pivotal to provide researchers and practitioners with synthesised information about the repercussions of maternal obesity or malnutrition on reproductive health and perinatal outcomes, and to pinpoint currently available nutritional managements to keep sows' body condition in an optimal range. Abstract

AbstractNumerous demands on the Doctor of Veterinary Medicine training program have the potential to reduce the amount of time allocated to food animal species in general including swine medicine, despite it being a key component of veterinary education. The objective of this study was to describe swine medicine training opportunities at North American and Caribbean veterinary education institutions. A 21-question survey was developed and distributed to veterinary colleges across North America and the Caribbean. The survey was available from October 2021 to March 2022, and one response was accepted per institution. Seventy-four percent of contacted institutions completed the survey, representing 29 veterinary colleges located in the United States of America, Canada, or the Caribbean. Responses were aggregated, analyzed, and grouped by topic: institution opportunities, curriculum opportunities, clinical opportunities, and faculty involvement in the swine medicine curricula. There was substantial variation among institutions in the delivery and resources allocated to swine medicine specific curricula. Swine veterinarians help ensure the health and well-being of animals and food safety. More research is required to evaluate the outcomes of the currently available opportunities. Concurrently, veterinary education institutions should prevent the attrition of swine educational program by investing in the support and development of swine opportunities for students.

ABSTRACTHaemophilus ducreyi, the etiologic agent of chancroid, has an obligate requirement for heme. Heme is acquired by H. ducreyifrom its human host via TonB-dependent transporters expressed at its bacterial surface. Of 3 TonB-dependent transporters encoded in the genome of H. ducreyi, only the hemoglobin receptor, HgbA, is required to establish infection during the early stages of the experimental human model of chancroid. Active immunization with a native preparation of HgbA (nHgbA) confers complete protection in the experimental swine model of chancroid, using either Freund's or monophosphoryl lipid A as adjuvants. To determine if transfer of anti-nHgbA serum is sufficient to confer protection, a passive immunization experiment using pooled nHgbA antiserum was conducted in the experimental swine model of chancroid. Pigs receiving this pooled nHgbA antiserum were protected from a homologous, but not a heterologous, challenge. Passively transferred polyclonal antibodies elicited to nHgbA bound the surface of H. ducreyiand partially blocked hemoglobin binding by nHgbA, but were not bactericidal. Taken together, these data suggest that the humoral immune response to the HgbA vaccine is protective against an H. ducreyiinfection, possibly by preventing acquisition of the essential nutrient heme.

Abstract

Porcine reproductive and respiratory syndrome virus (PRRSV) continues to cause severe reproductive and respiratory pathologies resulting in immense monetary and welfare costs for the swine industry. The vaccines against PRRSV are available; but they struggle with providing protection against the plethora of heterologous PRRSV strains. To improve PRRSV vaccine development, the aim of this study was to provide an in-depth analysis of the crucial heterologous T-cell response to type-2 PRRSV. Following PRRSV modified live virus (MLV) vaccination or infection using one high- or one low-pathogenic PRRSV-strain, this nine-week study evaluated the T-cell response to different PRRSV strains. Our results demonstrate an important role for T cells in this homo- and heterologous response. Specifically, the T-helper cells were the main responders during viremia. Their peak response at 28 dpi correlated with a reduction in viremia, and their homing receptor expression indicated the additional importance for the anti-PRRSV response in the lymphatic and lung tissue. The cytotoxic T lymphocyte (CTL) response was the strongest at the site of infection—the lung and bronchoalveolar lavage. The TCR-γδ T cells were the main responders post viremia and PRRSV induced their expression of the lymph node homing the chemokine receptor, CCR7: This indicates a crucial role for TCR-γδ T cells in the anti-PRRSV response in the lymphatic system. Abstract

Angiogenesis is an essential process during follicular development and corpora lutea (CL) formation. Recent studies have shown that vascular endothelial growth factor (VEGF) is an essential regulator of ovarian angiogenesis. Several lines of evidence have indicated that the production of VEGF is regulated by hypoxia inducible factor-1alpha (HIF-1alpha), especially under hypoxic conditions, but the expression of HIF-1alpha has not been well characterized in the porcine ovary. The present study examined the expression of HIF-1alpha mRNA and its localization in porcine ovaries at different stages of the estrous cycle. Northern blot analyses of total CL RNA indicated hybridization of the porcine HIF-1alpha probe to transcripts of approximately 3.8 kb. The mRNA expression of HIF-1alpha was highest in CL during the early luteal phase, followed by a decrease during the mid- and late-luteal phases. Using in situ hybridization, abundant HIF-1alpha mRNA was evident in follicles and CL. Within non-atretic follicles, HIF-1alpha mRNA was highly expressed in the granulosa cell layer, while weaker labeling was evident in the theca interna. These results suggest that HIF-1alpha may play a role in the regulation of cellular metabolism and blood supply during follicular growth and CL formation.

Abstract

This study evaluated the influence of concomitant infections with porcine reproductive and respiratory syndrome virus (PRRSV) and Mycoplasma hyopneumoniae on growth performance, serum metabolite concentrations, and serum insulin-like growth factor-I (IGF-I) in growing pigs. Twenty-two barrows (10 weeks of age) were treated with either an intranasal administration of PRRSV and an intratracheal infusion of M. hyopneumoniae (treatment; n = 8) or a sham inoculation with medium (sham; n = 8), or were not treated (control; n = 6). The sham pigs were matched by body weight and pair-wise fed with treatment pigs. Pigs were weighed on the day of inoculation (day 0) and at 4 weeks postinoculation (day 28). Blood samples were collected prior to inoculation and at weekly intervals for 4 weeks. Pigs in the treatment group exhibited clinical signs consistent with PRRSV infection and M. hyopneumoniae pneumonia. Diagnostic procedures confirmed that treatment pigs were inoculated with PRRSV and M. hyopneumoniae and that sham and control pigs remained free of both pathogens. Average daily gain and feed conversion did not differ among the 3 groups. The IGF-I levels differed (P < 0.05) between control and treatment pigs, even after feed intake returned to similar levels among groups. At day 7, IGF-I concentrations were greater in sham pigs compared with treatment pigs, despite similar feed intake. Sham inoculation and decreased feed intake in sham pigs did not alter serum IGF-I concentrations. Evidently, IGF-I status of pigs affected with disease is influenced by nutritional and nonnutritional factors during the disease process.
