Abstract

The onset of sub-critical crack growth during slow strain rate tensile testing (SSRT) is assessed through a combined experimental and modeling approach. A systematic comparison of the extent of intergranular fracture and expected hydrogen ingress suggests that hydrogen diffusion alone is insufficient to explain the intergranular fracture depths observed after SSRT experiments in a Ni-Cu superalloy. Simulations of these experiments using a new phase field formulation indicate that crack initiation occurs as low as 40% of the time to failure. The implications of such sub-critical crack growth on the validity and interpretation of SSRT metrics are then explored.

6. Gradient-enhanced statistical analysis of cleavage fracture

Abstract

We present a probabilistic framework for brittle fracture that builds upon Weibull statistics and strain gradient plasticity. The constitutive response is given by the mechanism-based strain gradient plasticity theory, aiming to accurately characterize crack tip stresses by accounting for the role of plastic strain gradients in elevating local strengthening ahead of cracks. It is shown that gradients of plastic strain elevate the Weibull stress and the probability of failure for a given choice of the threshold stress and the Weibull parameters. The statistical framework presented is used to estimate failure probabilities across temperatures in ferritic steels. The framework has the capability to estimate the three statistical parameters present in the Weibull-type model without any prior assumptions. The calibration against experimental data shows important differences in the values obtained for strain gradient plasticity and conventional J2 plasticity. Moreover, local probability maps show that potential damage initiation sites are much closer to the crack tip in the case of gradient-enhanced plasticity. Finally, the fracture response across the ductile-to-brittle regime is investigated by computing the cleavage resistance curves with increasing temperature. Gradient plasticity predictions appear to show a better agreement with the experiments.

10. Functional Olive Oil Production via Emulsions: Evaluation of Phenolic Encapsulation Efficiency, Storage Stability, and Bioavailability.

Abstract

Background/Objectives: Olive oil is valued for its health benefits, largely due to its bioactive compounds, including hydroxytyrosol (HTyr) and oleuropein (OLE), which have antioxidant, anti-inflammatory, and cardioprotective properties. However, many of these compounds are lost during the production process. This study developed a functional olive oil-derived product using water-in-oil emulsions (W/O) to incorporate commercial extracts rich in HTyr and OLE. Methods: HTyr and OLE were encapsulated in a W/O emulsion to preserve their bioactivity. The encapsulation efficiency (EE) was evaluated, and the performance of the emulsion was tested using an in vitro gastrointestinal digestion model. Bioaccessibility was measured by calculating the recovery percentage of HTyr and OLE during the digestion stages. Results: The results showed that OLE exhibited higher EE (88%) than HTyr (65%). During digestion, HTyr exhibited a gradual and controlled release, with bioaccessibility exceeding 80% in the gastric phase and a maintained stability throughout the intestinal phase. In contrast, OLE displayed high bioaccessibility in the gastric phase but experienced a notable decrease during the intestinal phase. Overall, the W/O emulsion provided superior protection and stability for both compounds, particularly for the secoiridoids, compared to the non-emulsified oil. Conclusions: The W/O emulsion improved the encapsulation and bioaccessibility of HTyr and OLE, constituting a promising method for enriching olive oil with bioactive phenolic compounds. Therefore, this method could enhance olive oil's health benefits by increasing the availability of these bioactive compounds during digestion, offering the potential for the development of fortified foods. Abstract

Collagen is a cornerstone protein for tissue engineering and 3D bioprinting due to its outstanding biocompatibility, low immunogenicity, and natural abundance in human tissues. Nonetheless, it still poses some important challenges, such as complicated and limited extraction processes, usually accompanied by batchto-batch reproducibility and influence of factors, such as temperature, pH, and ionic strength. In this work, we evaluated the suitability and performance of new, fibrillar type I collagen as standardized and reproducible collagen source for 3D printing and bioprinting. The acidic, native fibrous collagen formulation (5% w/w) performed remarkably during 3D printing, which was possible to print constructs of up to 27 layers without collapsing. On the other hand, the fibrous collagen mass has been modified to provide a fast, reliable, and easily neutralizable process. The neutralization with TRIS-HCl enabled the inclusion of cells without hindering printability. The cell-laden constructs were printed under mild conditions (50–80 kPa, pneumatic 3D printing), providing remarkable cellular viability (>90%) as well as a stable platform for cell growth and proliferation in vitro. Therefore, the native, type I collagen masses characterized in this work offer a reproducible and reliable source of collagen for 3D printing and bioprinting purposes.

Abstract

Drug adherence is a significant medical issue, often responsible for sub-optimal outcomes during the treatment of chronic diseases such as rheumatoid or psoriatic arthritis. Monoclonal antibodies (which are exclusively given parenterally) have been proven to be an effective treatment in these cases. The use of auto-injectors is an effective strategy to improve drug adherence in parenteral treatments since these pen-like devices offer less discomfort and increased user-friendliness over conventional syringe-based delivery. This study aims to investigate the feasibility of including a monoclonal antibody as a solid formulation inside an auto-injector pen. Specifically, the objective was to evaluate the drug stability after a concentration (to reduce the amount of solvent and space needed) and freeze-drying procedure. A preliminary screening of excipients to improve stability was also performed. The nano-DSC results showed that mannitol improved the stability of the concentrated, freeze-dried antibody in comparison to its counterpart without it. However, a small instability of the CH2 domain was still found for mannitol samples, which will warrant further investigation. The present results serve as a stepping stone towards advancing future drug delivery systems that will ultimately improve the patient experience and associated drug adherence.

Abstract

Vascular stents (VS) have revolutionized the treatment of cardiovascular diseases, as evidenced by the fact that the implantation of VS in coronary artery disease (CAD) patients has become a routine, easily approachable surgical intervention for the treatment of stenosed blood vessels. Despite the evolution of VS throughout the years, more efficient approaches are still required to address the medical and scientific challenges, especially when it comes to peripheral artery disease (PAD). In this regard, three-dimensional (3D) printing is envisaged as a promising alternative to upgrade VS by optimizing the shape, dimensions and stent backbone (crucial for optimal mechanical properties), making them customizable for each patient and each stenosed lesion. Moreover, the combination of 3D printing with other methods could also upgrade the final device. This review focuses on the most recent studies using 3D printing techniques to produce VS, both by itself and in combination with other techniques. The final aim is to provide an overview of the possibilities and limitations of 3D printing in the manufacturing of VS. Furthermore, the current situation of CAD and PAD pathologies is also addressed, thus highlighting the main weaknesses of the already existing VS and identifying research gaps, possible market niches and future directions.

32. Circulating metabolic markers after surgery identify patients at risk for severe postoperative complications: a prospective cohort study in colorectal cancer.

Abstract

Background: Early detection of postoperative complications after colorectal cancer (CRC) surgery is associated with improved outcomes. The aim was to investigate early metabolomics signatures capable to detect patients at risk for severe postoperative complications after CRC surgery. Materials and methods: Prospective cohort study of patients undergoing CRC surgery from 2015 to 2018. Plasma samples were collected before and after surgery, and analyzed by mass spectrometry obtaining 188 metabolites and 21 ratios. Postoperative complications were registered with Clavien--Dindo Classification and Comprehensive Complication Index. Results: One hundred forty-six patients were included. Surgery substantially modified metabolome and metabolic changes after surgery were quantitatively associated with the severity of postoperative complications. The strongest positive relationship with both Clavien--Dindo and Comprehensive Complication Index (β=4.09 and 63.05, P<0.001) corresponded to kynurenine/tryptophan, against an inverse relationship with lysophosphatidylcholines (LPCs) and phosphatidylcholines (PCs). Patients with LPC18:2/PCa36:2 below the cut-off 0.084 µM/µM resulted in a sevenfold higher risk of major complications (OR=7.38, 95% CI: 2.82-21.25, P<0.001), while kynurenine/tryptophan above 0.067 µM/µM a ninefold (OR=9.35, 95% CI: 3.03-32.66, P<0.001). Hexadecanoylcarnitine below 0.093 µM displayed a 12-fold higher risk of anastomotic leakage-related complications (OR=11.99, 95% CI: 2.62-80.79, P=0.004). Conclusion: Surgery-induced phospholipids and amino acid dysregulation is associated with the severity of postoperative complications after CRC surgery, including anastomotic leakage-related outcomes. The authors provide quantitative insight onmetabolic markers, measuring vulnerability to postoperative morbidity that might help guide early decision-making and improve surgical outcomes. Abstract

En Uruguay y mundialmente los jóvenes expresan bajo interés por la ciencia y las tecnologías, y se trata entonces, de renovar la educación científica para revertirlo y promover el desarrollo de la cultura científica ciudadana para el desarrollo sostenible. El principal objetivo de este proyecto, desarrollado por un equipo interinstitucional, multidisciplinario e internacional, fue promover el desarrollo de competencias de pensamiento crítico en estudiantes de las modalidades presencial y semipresencial de profesorado del CFE, potenciar la apropiación de tecnologías de bajo costo y código abierto como recursos educativos en el aula, y promover la creatividad e innovación. Se utilizó un diseño cuasi-experimental pre-test/post-test con grupo control. En el primer año se trabajó en un espacio de laboratorio físico de CFE que se equipó con tecnologías de bajo costo (MIT Little Devices Lab) y posteriormente cortadora laser, una impresora 3D, equipos de código abierto y de bajo costo de microscopía (Foldscope Instruments) y electrofisiología (Backyard Brains), herramientas e insumos de electrónica y robótica, laptop, impresora, cañón de proyección y microscopio de campo claro equipado con cámara digital. En el segundo año se desarrolló y trabajó en un laboratorio virtual interactivo de ciencias vivas que posibilitó la continuidad de la actividad práctica durante la pandemia por SARS-CoV-2. Está formado por dos estaciones experimentales descargables - Fisiología y Biología Celular- que simulan la generación de datos, que pueden ser almacenados, procesados o exportados, y una guía de recursos didácticos, disponibles en repositorio de CFE (http://repositorio.cfe.edu.uy/handle/123456789/1632). No se encontraron cambios significativos en destrezas de pensamiento crítico, probablemente debido al bajo n logrado y los trabajos-reportes elaborados por los estudiantes revelaron tanto fortalezas como debilidades.
