Little is known about early plastic biofilm assemblage dynamics and successional changes over time. By incubating virgin microplastics along oceanic transects and comparing adhered microbial communities with those of naturally occurring plastic litter at the same locations, we constructed gene catalogues to contrast the metabolic differences between early and mature biofilm communities. Early colonization incubations were reproducibly dominated by Alteromonadaceae and harboured significantly higher proportions of genes associated with adhesion, biofilm formation, chemotaxis, hydrocarbon degradation and motility. Comparative genomic analyses among the Alteromonadaceae metagenome assembled genomes (MAGs) highlighted the importance of the mannose-sensitive hemagglutinin (MSHA) operon, recognized as a key factor for intestinal colonization, for early colonization of hydrophobic plastic surfaces. Synteny alignments of MSHA also demonstrated positive selection for mshA alleles across all MAGs, suggesting that mshA provides a competitive advantage for surface colonization and nutrient acquisition. Large-scale genomic characteristics of early colonizers varied little, despite environmental variability. Mature plastic biofilms were composed of predominantly Rhodobacteraceae and displayed significantly higher proportions of carbohydrate hydrolysis enzymes and genes for photosynthesis and secondary metabolism. Our metagenomic analyses provide insight into early biofilm formation on plastics in the ocean and how early colonizers self-assemble, compared to mature, phylogenetically and metabolically diverse biofilms.

We examined membrane-spanning archaeal lipids using ultra high pressure liquid chromatography-high resolution mass spectrometry (UHPLC-HRMS) in a suite of sediment samples from both cored sequences (Messel oil shale and Lake Chala) and surface sediments (Azorean lakes) encompassing ancient and modern (Eocene to Present) lacustrine environments. Additionally we compared the lacustrine data to those of marine (Mediterranean cored sequences, Arabian Sea surface sediments and Monterey outcrop sediments) and hypersaline sediments (Vena del Gesso marls) as well as marine suspended particulate matter (SPM) from the Black Sea. Regular isoprenoidal glycerol dialkyl glycerol tetraethers (GDGTs) and glycerol dialkyl diethers (GDDs) were the most abundant membrane-spanning lipids in all investigated settings (>90 % and 84 % respectively). Interestingly, GDGTs with a cyclohexyl ring (S-GDGTs) were also present in almost all investigated lake sediments, in relative abundances of ca. 2–7 % and, for the first time, also their S-GDD counterparts were detected (2–10 %). The producers of S-GDGTs are still unknown, however our results show that it is likely that bottom water anoxia (both seasonally induced or permanent) is the driving factor for the production of these lipids, whereas previous studies suggested euxinia was required for production. Unsaturated GDGTs (uns-GDGTs, ca. 2 %) were only detected in Lake Chala sediments and surface sediments from Azorean lakes, but without accompanying uns-GDDs. GMGTs, glycerol monoalkyl glycerol tetraethers, were present in Messel oil shale and marine samples, while GMDs were only found in Messel oil shale.

Ocean currents are a key driver of plankton dispersal across the oceanic basins. However, species specific temperature constraints may limit the plankton dispersal. We propose a methodology to estimate the connectivity pathways and timescales for plankton species with given constraints on temperature tolerances, by combining Lagrangian modeling with networktheory. We demonstrate application of two types of temperature constraints: thermal niche and adaptation potential and compare it to the surface water connectivity between sample stations in the Atlantic Ocean. We find that non-constrained passive particles representative of a plankton species can connect all the stations within three years at the surface with pathways mostly along the major ocean currents. However, under thermal constraints, only a subset of stations can establish connectivity. Connectivity time increases marginally under these constraints, suggesting that plankton can keep within their favorable thermal conditions by advecting via slightly longer paths. Effect of advection depth on connectivity is observed to be sensitive to the width of the thermal constraints, along with decreasing flow speeds with depth and possible changes in pathways.

The holopelagic brown macroalgae Sargassum natans and Sargassum fluitans form essential habitats for attached and mobile fauna which contributes to a unique biodiversity in the Atlantic Ocean.However, holopelagic Sargassum natans (genotype I & VIII) and Sargassum fluitans (genotype III) have begun forming large accumulations with subsequent strandings on the western coast of Africa, the Caribbean and northern Brazil, threatening local biodiversity of coastal ecosystems and triggering economic losses. Moreover, stranded masses of holopelagic Sargassum may introduce or facilitate growthof bacteria that are not normally abundant in coastal regions where Sargassum is washing ashore. Hitherto, it is not clear how the holopelagic Sargassum microbiome varies across its growing biogeographic range and what factors drive the microbial composition. We determined the microbiome associated with holopelagic Sargassum from the Great Atlantic Sargassum Belt to coastal stranding sites in Mexico and Florida. We characterized the Sargassum microbiome via amplicon sequencing of the 16S V4 region hypervariable region of the rRNA gene. The microbial community of holopelagic Sargassum was mainly composed of photo(hetero)trophs, organic matter degraders andpotentially pathogenic bacteria from thePseudomonadaceae, Rhodobacteraceae andVibrionaceae. Sargassum genotypes S. natans I, S. natans VIII and S. fluitans III contained similar microbial families, but relative abundances and diversity varied. LEfSEanalyses further indicated biomarker genera that were indicative of Sargassum S. natans I/VIII and S. fluitans III. Bacteriohopanepolyols (BHPs) are a diverse class of lipids produced by bacteria across a wide range of environments. In this study, we aim to further identify BHPs related to ecological niches and/or specific bacteria by characterizing the distribution of BHPs in suspended particulate matter (SPM) of the water column and in sediments in a range of lakes and coastal lagoons from the Azores Archipelago, as well as in a co-culture enriched for methanotrophs. Sediment samples from Azorean lakes with low-oxygen conditions during the summer months (i.e., Azul, Verde, Funda, and Negra) contain relatively high abundances of BHPs that are typically associated with methane-oxidizing (methanotrophic) bacteria (i.e., aminotetrol, aminopentol, and methylcarbamate-aminopentol), as well as the ethenolamine-BHPs (i.e., ethenolamine-BHpentol and ethenolamine-BHhexol) and the N-formylated aminoBHPs. Both ethenolamine-BHPs and N-formylated aminoBHPs were also detected in a methanotroph–methylotroph co-culture that was enriched from a lake. In the SPM of all water columns, bacteriohopanetetrol (BHT), BHT cyclitol ether, and aminotriol are the dominant BHPs. In SPM from Lake Funda, nucleoside BHPs (i.e., Me-adenosylhopaneHG-diMen> (where HG refers to head group), N1-methylinosylhopane, 2Me-N1-inosylhopane, and Me-N1-inosylhopane) are present in low abundance or absent under oxic conditions but increase in concentration near the chemocline, suggesting potential in situ production of these nucleoside BHPs rather than an allochthonous origin. In contrast, sediments from shallow, well-mixed lakes (i.e., Empadadas, São Jorge, and Lomba) contain higher abundances of adenosylhopane and N1-methylinosylhopane, which likely originate from bacteria living in nearby soils. Since 2011, the Caribbean Islands have witnessed unprecedented massive stranding of a pelagic brown algal Sargassum spp. inducing damagesfor coastal ecosystems and economy. By accumulating heavy metals, Sargassum can play a role in contaminant transportation fromoffshore to the coast. In 2019, three genotypes of Sargassum ( S. fluitans III, S. natans I, and VIII) were sampled in seven stations along a 3400 km transect in the Atlantic Ocean. Concentrations of 15 heavy metal(loid)s elements were analyzed by Inductively Coupled Plasma Optical Emission Spectrometer (ICP-OES). Mean metal concentrations were ranked following descending order: As >Fe > Al > Mn > Cd > Zn > Ni > V > Cu > Cr > Hg. The metalloid As was the most abundant contaminant with a maximum value of 115 ppm, previously observed in the Caribbean area (80–150 ppm). At Atlantic Ocean Basin-scale, metallic element concentrations do not present spatial longitudinal gradients. Genotypes S. fluitans III and S. natans (I and VIII), present differents metal(loid)s contamination distinct patterns.

The influence of biofouling on zooplankton ingestion rates of plastics in freshwater environments has received limited attention. We investigated how biofouling of microplastics in wastewater effluent and in fresh surface water influences Daphnia magna's microplastic consumption. The differences in ingestion of the biofouled as compared with the virgin microplastics were higher for the surface water by a factor of seven compared with a factor of two for the effluent. The intake of biofouled microplastics by D. magna was higher compared with virgin plastics, but the reason for this preference should be further investigated. The fate of (micro)plastic particles in the open ocean is controlled by biological and physical processes. Here, we model the effects of biofouling on the subsurface vertical distribution of spherical, virtual plastic particles with radii of 0.01–1 mm. The biological specifications include the attachment, growth and loss of algae on particles. The physical specifications include four vertical velocity terms: advection, wind-driven mixing, tidally induced mixing and the sinking velocity of the biofouled particle. We track 10 000 particles for 1 year in three different regions with distinct biological and physical properties: the low-productivity region of the North Pacific Subtropical Gyre, the high-productivity region of the equatorial Pacific and the high mixing region of the Southern Ocean. The growth of biofilm mass in the euphotic zone and loss of mass below the euphotic zone result in the oscillatory behaviour of particles, where the larger (0.1–1.0 mm) particles have much shorter average oscillation lengths (<10 d; 90th percentile) than the smaller (0.01–0.1 mm) particles (up to 130 d; 90th percentile). A subsurface maximum particle concentration occurs just below the mixed-layer depth (around 30 m) in the equatorial Pacific, which is most pronounced for larger particles (0.1–1.0 mm). This occurs because particles become neutrally buoyant when the processes affecting the settling velocity of a particle and the seawater's vertical movement are in equilibrium. Seasonal effects in the subtropical gyre result in particles sinking below the mixed-layer depth only during spring blooms but otherwise remaining within the mixed layer. The strong winds and deepest average mixed-layer depth in the Southern Ocean (400 m) result in the deepest redistribution of particles (>5000 m). Lagoons are fragile marine ecosystems that are considerably affected by anthropogenic pollutants. We performed a spatiotemporal characterization of the microbiome of two Moroccan lagoons, Marchica and Oualidia, both classified as Ramsar sites, the former on the Mediterranean coast and the latter on the Atlantic coast. We investigated their microbial diversity and abundance using 16S rRNA amplicon- and shotgun-based metagenomics approaches during the summers of 2014 and 2015. The bacterial microbiome was composed primarily of Proteobacteria (25–53%, 29–29%), Cyanobacteria (34–12%, 11–0.53%), Bacteroidetes (24–16%, 23–43%), Actinobacteria (7–11%, 13–7%), and Verrucomicrobia (4–1%, 15–14%) in Marchica and Oualidia in 2014 and 2015, respectively. Interestingly, 48 strains were newly reported in lagoon ecosystems, while eight unknown viruses were detected in Mediterranean Marchica only. Statistical analysis showed higher microbial diversity in the Atlantic lagoon than in the Mediterranean lagoon and a robust relationship between alpha diversity and geographic sampling locations. This first-ever metagenomics study on Moroccan aquatic ecosystems enriched the national catalog of marine microorganisms. They will be investigated as candidates for bioindication properties, biomonitoring potential, biotechnology valorization, biodiversity protection, and lagoon health assessment.

Humans have made such dramatic and permanent changes to Earth's landscapes that much of it is now substantially and irreversibly altered from its preanthropogenic state. Remote islands, until recently isolated from humans, offer insights into how these landscapes evolved in response to human-induced perturbations. However, little is known about when and how remote systems were colonized because archaeological data and historical records are scarce and incomplete. Here, we use a multiproxy approach to reconstruct the initial colonization and subsequent environmental impacts on the Azores Archipelago. Our reconstructions provide unambiguous evidencefor widespread human disturbance of this archipelago starting between 700 -60+50 and 850-60+60 Common Era (CE), ca. 700 y earlier than historical records suggest the onset of Portuguese settlement of the islands. Settlement proceeded in three phases, during which human pressure on the terrestrial and aquatic ecosystems grew steadily (i.e., through livestock introductions, logging, and fire), resulting in irreversible changes. Our climate models suggest that the initial colonization at the end of the early Middle Ages (500 to 900 CE) occurred in conjunction with anomalous northeasterly winds and warmer Northern Hemisphere temperatures. These climate conditions likely inhibited exploration from southern Europe and facilitated human settlers from the northeast Atlantic. These results are consistent with recent archaeological and genetic data suggesting that the Norse were most likely the earliest settlers on the islands.

The Tropical Atlantic Ocean has recently been the source of enormous amounts of floating Sargassum macroalgae that have started to inundate shorelines in the Caribbean, the western coast of Africa and northern Brazil. It is still unclear, however, how the surface currents carry the Sargassum, largely restricted to the upper meter of the ocean, and whether observed surface drifter trajectories and hydrodynamical ocean models can be used to simulate its pathways. Here, we analyze a dataset of two types of surface drifters (38 in total), purposely deployed in the Tropical Atlantic Ocean in July, 2019. Twenty of the surface drifters were undrogued and reached only ∼8 cm into the water, while the other 18 were standard Surface Velocity Program (SVP) drifters that all had a drogue centered around 15 m depth. We show that the undrogued drifters separate more slowly than the drogued SVP drifters, likely because of the suppressed turbulence due to convergence in wind rows, which was stronger right at the surface than at 15 m depth. Undrogued drifters were also more likely to enter the Caribbean Sea. We also show that the novel Surface and Merged Ocean Currents (SMOC) product from the Copernicus Marine Environmental Service (CMEMS) does not clearly simulate one type of drifter better than the other, highlighting the need for further improvements in assimilated hydrodynamic models in the region, for a better understanding and forecasting of Sargassum drift in the Tropical Atlantic.

Alkenones are polyunsaturated long-chain methyl or ethyl ketones produced by species in the Isochrysidales, an order of haptophyte algae. Based on phylogenetic data, members of the Isochrysidales have been classified into three groups with each group showing significant differences in alkenone profiles and preferred growth environments. Common carbon chain lengths of alkenones range from 37 to 40. Extended C41 methyl (C41Me) and C42 ethyl (C42Et) alkenones have been reported in hypersaline lakes in China (Lake Alahake and Lake Balikun), Canada (Lake Snakehole) and marine sediments (e.g., ∼95 Ma in Blake-Bahama Basin). It is unclear, however, if these extended alkenones are produced by one or more groups of Isochrysidales. Here, we systematically examined alkenones from cultures of Group II (Isochrysis nuda, Isochrysis litoralis, Ruttnera lamellosa, Isochrysis galbana and Tisochrysis lutea) and Group III (Emiliania huxleyi and Gephyrocapsa oceanica) Isochrysidales and environmental samples of Group I Isochysidales. C41Me and C42Et alkenones were found in all Group II species with Isochrysis nuda producing the highest percentages, but not in alkenones produced by Group I nor Group III Isochrysidales. Our results indicate that extended C41Me and C42Et alkenones are specific biomarkers for Group II Isochrysidales. We also report the first temperature calibrations of alkenones for Isochrysis nuda and Isochrysis litoralis using culture experiments, and find temperatures inferred from extended alkenones in Balikun and Alahake surface sediments match warm-season temperatures based on Isochrysis nuda calibrations, which is further corroborated by genomic data indicating the dominance of Isochrysis nuda Isochrysidales..

Alkenone-derived paleotemperature reconstruction holds great promise in lake environments. However, theoccurrence of multiple species of alkenone-producing haptophyte algae in a single lake can complicate thetranslation of alkenone unsaturation to temperature if each species requires an individual temperature calibra-tion. Here, we present thefirst systematic monitoring of two alkenone-producing haptophytes throughout thecourse of a seasonal cycle in Lake George, North Dakota, using a combined approach of DNA sequencing andalkenone lipid characterization. Field sampling revealed a nonoverlapping haptophyte succession, with both anearly and late season haptophyte bloom event. Culturing experiments demonstrated that the two haptophytespecies responsible for these blooms had statistically similar alkenone-temperature responses, although theculture-based calibrations were distinct from the in situ calibration. Bloom timing of each haptophyte speciescorresponded to surface-water temperatures that differed by more than 10?C, revealing that changes in bloomintensities for each species will skew the sediment-inferred temperatures to a different stage of the growth sea-son. These results highlight the importance of accounting for bloom timing when interpreting alkenone-derivedtemperatures in sediment cores, especially in lakes that experience large seasonalfluctuations in water columntemperature and salinity.

Three-quarters of all marine debris (MD) consists of plastic, a reflection of their worldwide use, production and waste mismanagement. Data on MD distributions can improve our ability to effectively reduce debris that escapes onto shorelines and the ocean. In this study, the Matrix Scoring Technique (Marine Strategy Framework Directive Technical Group on Marine Litter) was applied as an approach to calculate the likelihood of single debris items originating from a series of potential sources. Factors considered were: identity and function of debris, beach location, influential activities, “mix” of debris found, presence of indicator items, and quantity of MD. The standing-stock (abundance and composition) of MD was investigated in two sandy beaches (Conceição and Porto Pim) of the Azores Archipelago (NE Atlantic) for the period 2012–2018. The results of this study show promise towards the implementation of a new classification method to determine beach debris sources in remote open-ocean areas.

Members of the order Isochrysidales are unique among haptophyte lineages in being the exclusive producers of alkenones, long‐chain ketones that are commonly used for paleotemperature reconstructions. Alkenone‐producing haptophytes are divided into three major groups based largely on molecular ecological data: Group I is found in freshwater lakes, Group II commonly occurs in brackish and coastal marine environments, and Group III consists of open ocean species. Each group has distinct alkenone distributions; however, only Groups II and III Isochrysidales currently have cultured representatives. The uncultured Group I Isochrysidales are distinguished geochemically by the presence of tri‐unsaturated alkenone isomers (C37:3b Me, C38:3b Et, C38:3b Me, C39:3b Et) present in water column and sediment samples, yet their genetic diversity, morphology, and environmental controls are largely unknown. Using small‐subunit (SSU) ribosomal RNA (rRNA) marker gene amplicon high‐throughput sequencing of environmental water column and sediment samples, we show that Group I is monophyletic with high phylogenetic diversity and contains a well‐supported clade separating the previously described “EV” clade from the “Greenland” clade. We infer the first partial large‐subunit (LSU) rRNA gene Group I sequence phylogeny, which uncovered additional well‐supported clades embedded within Group I. Relative to Group II, Group I revealed higher levels of genetic diversity despite conservation of alkenone signatures and a closer evolutionary relationship with Group III. In Group I, the presence of the tri‐unsaturated alkenone isomers appears to be conserved, which is not the case for Group II. This suggests differing environmental influences on Group I and II and perhaps uncovers evolutionary constraints on alkenone biosynthesis.

Isochrysidales species of the phylum Haptophyta are the exclusive producers of C37 to C42 long chain alkyl ketones, also called long chain alkenones (LCAs). While LCA distributions are known to vary with temperature and salinity, it is difficult to tease apart the direct effects of environmental parameters vs changes in the LCA-producing organisms. The Baltic Sea surface salinity gradient, which ranges from oligohaline (0.5–5 g/kg) to polyhaline (18–30 g/kg), represents a unique opportunity to study the relationships between salinity changes, species distribution and LCA biomarkers in a single ecosystem. LCA biomarkers revealed the presence of the three known Isochrysidales groups (Groups I, II and III) in Baltic Sea surface sediments, and the presence of Groups I and II were further confirmed with DNA sequencing. Group III Isochrysidales were present in the mixoeuhaline Skagerrak based on LCA signature alone. Groups I and II Isochrysidales were found for the first time in the Baltic Sea using a combination of LCAs and DNA biomarkers, solving an eighteen-year long mystery of Baltic Sea LCA-producing haptophyte identity. Group II Isochrysidales, which have a large salinity tolerance range, were spread over the Skagerrak and the complete Baltic Sea, but were characteristic for the central Baltic Sea. Oligohaline Group I Isochrysidales were representative for the northern Baltic Sea. However, evidence of Group I Isochrysidales in the central and southern Baltic Sea suggests a possible transport by surface currents since this group is typically confined to oligohaline conditions. Testing the recently developed ratio of isomeric C37 ketones (RIK37) against the Baltic Sea surface salinity gradient revealed a significant positive correlation. There is broad interest in disease spread through the pet trade, but empirical research on hosts and pathogens in transit along actual trade routes is notably absent. Using next-generation DNA sequencing, and partnering with the ornamental fish industry, we tracked shifts in microbial community and potential pathogen structure associated with Sailfin Tang (Zebrasoma desjardinii) along the United States (U.S.) leg of an international supply chain. We observed striking changes in microbial diversity and composition of potential pathogens, including increased dominance of vibrios of fishes in transit. Our pilot findings suggest that high investment in fishes early in the supply chain may not matter to their long-term health depending on end destination conditions.

Prophylactic antibiotics in the aquaculture and ornamental fish industry are intended to prevent the negative impacts of disease outbreaks. Research in mice and humans suggests that antibiotics may disturb microbiome communities and decrease microbiome-mediated disease resistance, also known as “colonization resistance.” If antibiotics impact fish as they do mice and humans, prophylactic administrations on aquaculture farms may increase downstream disease susceptibility in target hosts, despite short-term pathogen control benefits. We tested the effects of antibiotics on mortality after a pathogen challenge in the Poecilia sphenops black molly and subsequently tested if probiotic inoculations could reverse any antibiotic-induced losses of disease resistance. We found that antibiotic treatment significantly increased fish mortality. We further found that our two candidate probiotic bacterial species, Phaeobacter inhibens S4Sm and Bacillus pumilus RI06-95Sm, were able to colonize black molly microbiomes and reverse the negative impacts of antibiotics. Despite the positive impact on survival, probiotic treatment did not influence overall microbiome community structure or diversity. Our results suggest that subtle manipulations of microbiome composition can have dramatic impacts on host phenotype. The results of this study have implications for how antibiotic-treated microbiomes can be restored and suggest that small-scale additions may be as effective as wholesale transplants.

Ocean Sampling Day was initiated by the EU-funded Micro B3 (Marine Microbial Biodiversity, Bioinformatics, Biotechnology) project to obtain a snapshot of the marine microbial biodiversity and function of the world's oceans. It is a simultaneous global mega-sequencing campaign aiming to generate the largest standardized microbial data set in a single day. This will be achievable only through the coordinated efforts of an Ocean Sampling Day Consortium, supportive partnerships and networks between sites. This commentary outlines the establishment, function and aims of the Consortium and describes our vision for a sustainable study of marine microbial communities and their embedded functional traits.
