Lipid nanoparticles own a remarkable potential in nanomedicine, only partially disclosed. While the clinical use of liposomes and cationic lipid-nucleic acid complexes is well-established, liquid lipid nanoparticles (nanoemulsions), solid lipid nanoparticles, and nanostructured lipid carriers have even greater potential. However, they face obstacles in being used in clinics due to a lack of understanding about the molecular mechanisms controlling their drug loading and release, interactions with the biological environment (such as the protein corona), and shelf-life stability. To create effective drug delivery carriers and successfully translate bench research to clinical settings, it is crucial to have a thorough understanding of the internal structure of lipid nanoparticles. Through synchrotron small-angle X-ray scattering experiments, we determined the spatial distribution and internal structure of the nanoparticles' lipid, surfactant, and the water in them. The nanoparticles themselves have a barrel-like shape that consists of coplanar lipid platelets (specifically cetyl palmitate) that are partially covered by polysorbate 80 surfactant and retain a small amount of hydration water. Although the platelet structure was expected, the presence of surfactant molecules forming sticky patches between adjacent platelets challenges the classical core-shell model used to describe solid lipid nanoparticles. Additionally, the surfactant partially covers the water-nanoparticle interface, allowing certain lipid regions to come into direct contact with surrounding water. Understanding and control of thermal transport in solids at nanoscale is crucial to engineer and to enhance properties of a new generation of optoelectronic, thermoelectric, and photonic devices. In this regard, semiconductor superlattice structures provide a unique platform to study phenomena associated with phonon propagations in solids such as heat conduction. Transient X-ray diffraction can directly probe atomic motions and therefore is among the rare techniques sensitive to phonon dynamics in a condensed matter. Here, we study optically induced transient heat conduction in GaAs/AlAs superlattice structures using EIGER2 detector. Benchmark experiments have been performed at the Austrian SAXS beamline at Elettra Sincrotrone Trieste that operated in the hybrid filling mode. We demonstrate that drifts of experimental conditions, such as synchrotron beam fluctuations, become less essential when utilizing EIGER2 double gating mode that results in a faster acquisition of high quality data and facilitates data analysis and data interpretation., Comment: 15 pages, 9 figures, 1 table

Photoresponsive materials feature properties that can be adjusted near- instantaneously, reversibly, and with high spatiotemporal precision, by light. There is considerable interest in maximising the degree of switching, and in measuring this degree during illumination in complex environments. We study the switching of photoresponsive lipid membranes that allow precise and reversible manipulation of membrane shape, permeability, and fluidity. Though these macroscopic photoresponses are clear, it is unclear how large the changes of trans/cis ratio are, and whether they can be improved. Here, we used small-angle X-ray scattering to measure the thickness of photoswitchable lipid membranes, and correlate thickness to trans/cis ratios. This revealed an unexpected dependency of photoswitching upon aqueous phase composition, highlighting smaller-than-expected photoswitching with deionized water, and showed thickness variations twice as large as previously observed. Soft X-rays can quantitatively isomerise photolipid membranes to the all- trans state: both enabling more powerful X-ray-based membrane control and underlining the role of high energy X-rays for observation-only soft matter experiments.

In-situ small- and wide-angle scattering experiments at synchrotrons often result in massive amounts of data within seconds only. Especially during such beamtimes, processing of the acquired data online, so without mentionable delay, is key to obtain feedback on failure or success of the experiment. We thus developed SAXSDOG, a python based environment for real-time azimuthal integration of large-area scattering-images. The software is primarily designed for dedicated data-pipelines: once a scattering image is transferred from the detector onto the storage-unit, it is automatically integrated and pre-evaluated using integral parameters within milliseconds. The control and configuration of the underlying server-based processes is done via a graphical user interface SAXSLEASH, which visualizes the resulting 1D data together with integral classifiers in real time. Microfluidic mixing is an important means for in-situ sample preparation and handling while Small Angle X-Ray Scattering (SAXS) is a proven tool for characterising (macro-)molecular structures. In combination those two techniques enable investigations of fast reactions with high time resolution (<1 ms). The goal of combining a micro mixer with SAXS, however, puts constraints on the materials and production methods used in the device fabrication. The measurement channel of the mixer needs good x-ray transparency and a low scattering background. While both depend on the material used, the requirement for low scattering especially limits the techniques suitable for producing the mixer, as the fabrication process can induce molecular orientations and stresses that can adversely influence the scattering signal. Not only is it important to find a production method that results in a device with low background scattering, but it also has to be versatile enough to produce appropriate mixer designs. Here we discuss two methods - laser ablation of polycarbonate and injection moulding of Topas - which were found suitable for our needs, provided care is taken in aligning the mixing/reaction channel, where the actual measurements will be carried out. We adapted the highly successful scattering density profile (SDP) model, which is used to jointly analyze small angle x-ray and neutron scattering data from unilamellar vesicles, for use with data from fully hydrated, liquid crystalline multilamellar vesicles (MLVs). By using a genetic algorithm, this new method is capable of providing high resolution structural information, as well as determining bilayer elastic bending fluctuations from standalone x-ray data. Important structural parameters, such as bilayer thickness and area per lipid were determined for a series of saturated and unsaturated lipids, as well as binary mixtures with cholesterol. Results are in good agreement with previously reported SDP data, which used both neutron and x-ray data. The addition of deuterated and non-deuterated MLV neutron data to the analysis improved lipid backbone information, but did not improve, within experimental uncertainties, the structural information regarding bilayer thickness and area per lipid.

Silver nanocrystals have been prepared by reacting silver nitrate with ascorbic acid in aqueous solution containing a low concentration of a commercial polynaphtalene sulphonate polymer (Daxad 19). Various crystalline morphologies have been obtained simply by tuning the reaction temperature. We have investigated the nanoparticle formation mechanism at three different temperatures by in situ and time resolved Small Angle X ray Scattering measurements. By modeling the scattering intensity with interacting spherical particles in a fractal-like polymer-Ag matrix, we found signatures of nucleation, growth and assembly of primary particles of about 15-20 nm. We observed how the time evolution of both spatial correlations between primary particles and the dynamic fractal geometry of the polymer-Ag matrix could influence and determine both the aggregation mechanism and the morphology of forming nanostructures in solution.

In this work a combination of an ionization chamber with one-dimensional spatial resolution and a MicroCAT structure will be presented. The combination between gas gain operations and integrating front-end electronics yields a dynamic range as high as eight to nine orders of magnitude. Therefore this device is well suitable for medical imaging or applications such as small angle x-ray scattering, where the requirements on the dynamic of the detector are exceptional high. Basically the described detector is an ionization chamber adapted to fan beam geometry with an active area of 192 cm and a pitch of the anode strips of 150 micrometer. In the vertical direction beams as high as 10 mm can be accepted. Every read-out strip is connected to an analogue integrating electronics channel realized in a custom made VLSI chip. A MicroCAT structure utilized as a shielding grid enables frame rates as high as 10kHz. The high dynamic range observed stems from the fact that the MicroCAT enables active electron amplification in the gas. Thus a single photon resolution can be obtained for low photon fluxes even with the integrating electronics.
