Abstract

International audience; Spermatozoa have a remarkable epigenome in line with their degree of specialization, their unique nature and different requirements for successful fertilization. While bull semen is widely used in artificial insemination, the literature describing DNA methylation in bovine spermatozoa is still scarce. In the context of genomic selection, more information on the epigenetic features transferred to the embryo alongside the paternal genetic heritage is necessary, in order to improve semen quality control procedures and bull fertility. We characterized the bull sperm methylome relative to bovine fibroblasts and monocytes using reduced restriction bisulfite sequencing (RRBS). A wide majority of differentially methylated cytosines (DMCs) were less methylated in sperm than in somatic cells. Consistent with previous studies in other species, these hypomethylated DMCs were enriched for genes relevant to the germline differentiation program and sperm functions. The most remarkable observation was a dramatic enrichment for repeats and particularly for satellites, which may partly account for the lower global methylation we observed in bull sperm relative to sperm from other species (LUminometric Methylation Assay). We next investigated whether fertility biomarkers could be identified in the sperm methylome. Based on field fertility indicators, we compared fertile ejaculates, subfertile ejaculates and ejaculates from bulls with a disappointing career in contradiction with genomic predictions (Montbéliarde breed, n=10/group). The median age at production was consistent among groups and the phenotypic characterization revealed no significant differences except for the proportion of motile sperm, which was reduced in disappointing bulls. We identified 4823 DMCs targeting genes involved in signaling pathways, angiogenesis and the response to growth factors. Since DNA methylation is affected by the CpG content of the genome and its alteration by DNA polymorphism, an integrative approach was conducted to identify CpGs and SNPs in correlation, which revealed both direct and indirect interactions between the genome and the epigenome.

Abstract

The objective is to study the effects of nutrient restrictions, which induce a metabolic imbalance on the inflammatory response of the mammary gland in early lactation cows. The aim is to decipher the molecular mechanisms involved, by comparing a control, with a restriction group, a transcriptome and proteome, after an intra-mammary lipopolysaccharide challenge. Multi-parous cows were either allowed ad libitum intake of a lactation diet (n = 8), or a ration containing low nutrient density (n = 8; 48% barley straw and dry matter basis) for four days starting at 24 ± 3 days in milk. Three days after the initiation of their treatments, one healthy rear mammary quarter of 12 lactating cows was challenged with 50 µg of lipopolysaccharide (LPS). Transcriptomic and proteomic analyses were performed on mammary biopsies obtained 24 h after the LPS challenge, using bovine 44K microarrays, and nano-LC-MS/MS, respectively. Restriction-induced deficits in energy, led to a marked negative energy balance (41 versus 97 ± 15% of Net Energy for Lactation (NEL) requirements) and metabolic imbalance. A microarray analyses identified 25 differentially expressed genes in response to restriction, suggesting that restriction had modified mammary metabolism, specifically β-oxidation process. Proteomic analyses identified 53 differentially expressed proteins, which suggests that the modification of protein synthesis from mRNA splicing to folding. Under-nutrition influenced mammary gland expression of the genes involved in metabolism, thereby increasing β-oxidation and altering protein synthesis, which may affect the response to inflammation.

Abstract

International audience; The equine sector is currently growing and evolving worldwide. In Europe, it is estimated that there could be at least 6 million horses in the 27 country members, grazing 6 million hectares of permanent grassland. 400 000 full time jobs equivalent would be provided by the sector and the numbers of horses and riders are growing in the approximate range of 7% a year. Today, little is known about the horse industry but there is a growing consensus that it has changed from a primarily agricultural and industrial sector activity to one firmly rooted in sports, leisure and consumption. It is impossible to understand these transformations without understanding the society within which they are embedded. Horse welfare, population size, behaviours and potentials depend upon those of the societies in which their owners and riders live. Further, contemporary society is changing as it never has before. There is no single ‘society’. Different peoples and different places all constitute unique economies, unique social values and mores, and unique formations of the horse industry. To understand the future of the horse sector we must understand these varied social and economic formations. In this context, and given the importance of and the challenges faced by the horse industry, the number of socio-economic studies devoted to this sector has recently multiplied all over the world. The social sciences undertake research, analysis and the development of new understandings of changes in the economy, in cultural values, and in social organisation of contemporary society. Working together with traditional equine sciences, we are creating new interdisciplinary knowledge which help us understand how we got to where we are now, and where the equine sector might go in the future. That’s why a working group in socio-economy has been created within the EAAP Horse Commission. Today, it includes 67 members from 20 countries. The goal of this group is to share ideas, research and experiences, but also to think about new topics of interest for research and development and to build common projects. This presentation addresses the social and economic issues faced by equine and social scientists who are exploring the contemporary shape of the equine sector, and whose research and analysis can help begin a discussion which enables us to understand what it might become in the future.
