Acellular pertussis vaccines (aPVs) have been developed as an alternative to whole-cell pertussis vaccines (wPVs) because of their similar efficacy but reduced reactogenicity. The aPV contains 3 or more immunogenic components of Bordetella pertussis. We aimed to evaluate the immunogenicity and protective potency of an aPV vaccine produced in our laboratory consisting of pertussis toxin (PT), filamentous hemagglutinin (FHA), and pertactin (PRN) in mice. The aPV components were produced and purified from the supernatant and pellet of the bacterial culture. Two doses of the formulated vaccine, in parallel with two commercial vaccines, were administered intraperitoneally to mice at 3-week intervals. Antibody titers against aPV antigens were measured by enzyme-linked immunosorbent assay (ELISA) after primary and booster vaccinations. To assess the protective efficacy, an intranasal challenge with a live pathogenic B pertussis strain was conducted 2 weeks after the booster vaccination, and bacterial counts (colony-forming units [CFUs]) in the lungs were determined 2 hours and 10 days after the challenge. The results demonstrated a significant increase in antibody titers against all pertussis antigens in the serum of the vaccinated groups compared with the negative control group, following both the primary and booster doses. No significant differences were observed between our formulation and the commercial vaccines. Furthermore, the CFU results after the challenge showed complete eradication of infection 10 days after the challenge in all immunized groups, in contrast to the control group. Our aPV formulation, the first aPV candidate developed in Iran, exhibits immunogenicity and protective efficacy comparable to those of commercial vaccines. The integration of gas and electricity networks has emerged as a promising approach to enhance the overall flexibility of energy systems. As the transition toward sustainable and decarbonized energy sources accelerates, the seamless coordination between electricity and gas infrastructure becomes increasingly crucial. This paper presents a comprehensive review of the state-of-the-art research and developments concerning the flexibility in the operation of low-carbon integrated gas and electricity networks (IGENs) as part of the whole system approach. Methods and solutions to provide and improve flexibility in the mentioned systems are studied and categorized. Flexibility is the system's ability to deal with changes and uncertainties in the network while maintaining an acceptable level of reliability. The presented review underscores the significance of this convergence in facilitating demand-side management, renewable energy integration, and overall system resilience. By highlighting the technical, economic, and regulatory aspects of such integration, this paper aims to guide researchers, policymakers, and industry stakeholders toward effective decision-making and the formulation of comprehensive strategies that align with the decarbonization of energy systems. Hepatitis B virus (HBV) infection is a major health problem worldwide and causes almost one million deaths annually. The HBV core gene codes for two related antigens, known as core antigen (HBcAg) and e-antigen (HBeAg), sharing 149 residues but having different amino- and carboxy-terminals. HBeAg is a soluble variant of HBcAg and a clinical marker for determining the disease severity and patients' screening. Currently available HBeAg assays have a shortcoming of showing cross-reactivity with HBcAg. In this study, for the first time, we evaluated whether HBcAg-adsorbed anti-HBe polyclonal antibodies could specifically recognize HBeAg or still show cross-reactivity with HBcAg. Recombinant HBeAg was cloned in pCold1 vector and successfully expressed in Escherichia coli and after purification by Ni-NTA resin was used to generate polyclonal anti-HBe antibodies in rabbit. Purified HBeAg was further characterized by assessing its reactivity with anti-HBe in the sera of chronically infected patients and HBeAg-immunized rabbit. Sera from patients with chronic HBV infection, containing anti-HBe, specifically reacted with recombinant HBeAg, implying antigenic similarity between the prokaryotic and native HBeAg in the serum of HBV-infected patients. In addition, the designed enzyme-linked immunosorbent assay (ELISA) with rabbit anti-HBe polyclonal antibodies could detect recombinant HBeAg with high sensitivity, while high cross-reactivity with HBcAg was observed. It is noteworthy that HBcAg-adsorbed anti-HBe polyclonal antibodies still showed high cross-reactivity with HBcAg, implying that due to the presence of highly similar epitopes in both antigens, HBcAg-adsorbed polyclonal antibodies cannot differentiate between the two antigens. Background: Probiotic Lactobacillus spp. modulate immune response via interactions of their binding proteins with epithelial cells. We studied the presence of attachment proteinencoding genes (mub1, mub2, and mapA) in Lactobacillus strains with probiotic features isolated from inflammatory bowel disease (IBD) patients and their attachment strength relative to healthy individuals. Methods: Bacterial strains have been isolated from stool samples of 35 healthy and 23 IBD volunteers. Lactobacillus spp. were identified using PCR. Strains with probiotic features were determined by testing resistance against acid and bile. Isolates were assigned as nonadhesive, adhesive, and strongly adhesive strains based on the number of attached bacteria to epithelial cells. Finally, PCR was used to detect the presence of mub1, mub2, and mapA genes. Results: Probiotic lactobacilli were isolated from 35/35 and 9/23 of healthy and IBD individuals and yielded a total of 87 and 28 strains, respectively. The Mub1 gene was detected in 95.4% and 100% (p>0.05), mub2 in 95.4% and 89.3% (p>0.05), and mapA in 94.3% and 78.6% (p<0.05) of healthy and IBD isolates, respectively. The numbers of bacteria attached to epithelial cells in healthy and IBD isolates were respectively 33.68±6.00 and 12.23±3.87 in non-adhesive, 71.3±10.83 and 42.17±1.33 in adhesive, 124.40±8.59 and 104.67±5.50 in the strongly adhesive group (p< 0.05). Conclusion: Less Lactobacillus spp. with weaker attachments to epithelial cells colonize the gut in IBD than healthy individuals.
