Cowpea [Vigna unguiculata (L.) Walp.] is one of the most tolerant legume crops to drought and salt stresses. WRKY transcription factor (TF) family members stand out among plant transcriptional regulators related to abiotic stress tolerance. However, little information is currently available on the expression of the cowpea WRKY gene family (VuWRKY) in response to water deficit. Thus, we analyzed genomic and transcriptomic data from cowpea to identify VuWRKY members and characterize their structure and transcriptional response under root dehydration stress. Ninety-two complete VuWRKY genes were found in the cowpea genome based on their domain characteristics. They were clustered into three groups: I (15 members), II (58), and III (16), while three genes were unclassified. Domain analysis of the encoded proteins identified four major variants of the conserved heptapeptide motif WRKYGQK. In silico analysis of VuWRKY gene promoters identified eight candidate binding motifs of cis-regulatory elements, regulated mainly by six TF families associated with abiotic stress responses. Ninety-seven VuWRKY modulated splicing variants associated with 55 VuWRKY genes were identified via RNA-Seq analysis available at the Cowpea Genomics Consortium (CpGC) database. qPCR analyses showed that 22 genes are induced under root dehydration, with VuWRKY18, 21, and 75 exhibiting the most significant induction levels. Given their central role in activating signal transduction cascades in abiotic stress response, the data provide a foundation for the targeted modification of specific VuWRKY family members to improve drought tolerance in this important climate-resilient legume in the developing world and beyond., (Copyright © 2022 Elsevier B.V. Background: Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) promotes challenging immune and inflammatory phenomena. Though various therapeutic possibilities have been tested against coronavirus disease 2019 (COVID-19), the most adequate treatment has not yet been established. Propolis is a natural product with considerable evidence of immunoregulatory and anti-inflammatory activities, and experimental data point to potential against viral targets. We hypothesized that propolis can reduce the negative effects of COVID-19., Methods: In a randomized, controlled, open-label, single-center trial, hospitalized adult COVID-19 patients were treated with a standardized green propolis extract (EPP-AF®️) as an adjunct therapy. Patients were allocated to receive standard care plus an oral dose of 400 mg or 800 mg/day of green propolis for seven days, or standard care alone. Standard care included all necessary interventions, as determined by the attending physician. The primary end point was the time to clinical improvement, defined as the length of hospital stay or oxygen therapy dependency duration. Secondary outcomes included acute kidney injury and need for intensive care or vasoactive drugs. Patients were followed for 28 days after admission., Results: We enrolled 124 patients; 40 were assigned to EPP-AF®️ 400 mg/day, 42 to EPP-AF®️ 800 mg/day, and 42 to the control group. The length of hospital stay post-intervention was shorter in both propolis groups than in the control group; lower dose, median 7 days versus 12 days (95% confidence interval [CI] -6.23 to -0.07; p = 0.049) and higher dose, median 6 days versus 12 days (95% CI -7.00 to -1.09; p = 0.009). Propolis did not significantly affect the need for oxygen supplementation. In the high dose propolis group, there was a lower rate of acute kidney injury than in the controls (4.8 vs 23.8%), (odds ratio [OR] 0.18; 95% CI 0.03-0.84; p = 0.048). No patient had propolis treatment discontinued due to adverse events., Conclusions: Addition of propolis to the standard care procedures resulted in clinical benefits for the hospitalized COVID-19 patients, especially evidenced by a reduction in the length of hospital stay. Background: Perioperative bleeding and transfusion are important causes of morbidity and mortality in patients undergoing liver transplantation. The aim of this study is to assess whether viscoelastic tests-guided therapy with the use of synthetic factor concentrates impact transfusion rates of hemocomponents in adult patients undergoing liver transplantation., Methods: This is an interventional before-after comparative study. Patients undergoing liver transplantation before the implementation of a protocol using thromboelastometry and synthetic factor concentrates were compared to patients after the implementation. Primary outcome was transfusion of any hemocomponents. Secondary outcomes included: transfusion of red blood cells (RBC), fresh frozen plasma (FFP), cryoprecipitate or platelets, clinical complications, length of stay and in-hospital mortality., Results: A total of 183 patients were included in the control and 54 in the intervention phase. After propensity score matching, the proportion of patients receiving any transfusion of hemocomponents was lower in the intervention phase (37.0 vs 58.4%; OR, 0.42; 95% CI, 0.20-0.87; p = 0.019). Patients in the intervention phase received less RBC (30.2 vs 52.5%; OR, 0.21; 95% CI, 0.08-0.56; p = 0.002) and FFP (5.7 vs 27.3%; OR, 0.11; 95% CI, 0.03-0.43; p = 0.002). There was no difference regarding transfusion of cryoprecipitate and platelets, complications related to the procedure, hospital length of stay and mortality., Conclusions: Use of a viscoelastic test-guided transfusion algorithm with the use of synthetic factor concentrates reduces the transfusion rates of allogenic blood in patients submitted to liver transplantation., Trial Registration: This trial was registered retrospectively on November 15th, 2018 - clinicaltrials.gov - Identifier: NCT03756948.

It is well-known that positive end-expiratory pressure (PEEP) can prevent ventilator-induced lung injury (VILI) and improve pulmonary physiology in animals with injured lungs. It's uncertain whether PEEP has similar effects in animals with uninjured lungs. A systematic review of randomized controlled trials (RCTs) comparing different PEEP levels in animals with uninjured lungs was performed. Trials in animals with injured lungs were excluded, as were trials that compared ventilation strategies that also differed with respect to other ventilation settings, e.g., tidal volume size. The search identified ten eligible trials in 284 animals, including rodents and small as well as large mammals. Duration of ventilation was highly variable, from 1 to 6 hours and tidal volume size varied from 7 to 60 mL/kg. PEEP ranged from 3 to 20 cmH 2 O, and from 0 to 5 cmH 2 O, in the 'high PEEP' or 'PEEP' arms, and in the 'low PEEP' or 'no PEEP' arms, respectively. Definitions used for lung injury were quite diverse, as were other outcome measures. The effects of PEEP, at any level, on lung injury was not straightforward, with some trials showing less injury with 'high PEEP' or 'PEEP' and other trials showing no benefit. In most trials, 'high PEEP' or 'PEEP' was associated with improved respiratory system compliance, and better oxygen parameters. However, 'high PEEP' or 'PEEP' was also associated with occurrence of hypotension, a reduction in cardiac output, or development of hyperlactatemia. There were no differences in mortality. The number of trials comparing 'high PEEP' or 'PEEP' with 'low PEEP' or 'no PEEP' in animals with uninjured lungs is limited, and results are difficult to compare. Background: Spinal cord injury (SCI) is characterized by a total or partial deficit of sensory and motor pathways. Impairments of this injury compromise muscle recruitment and motor planning, thus reducing functional capacity. SCI patients commonly present psychological, intestinal, urinary, osteomioarticular, tegumentary, cardiorespiratory and neural alterations that aggravate in chronic phase. One of the neurorehabilitation goals is the restoration of these abilities by favoring improvement in the quality of life and functional independence. Current literature highlights several benefits of robotic gait therapies in SCI individuals., Objectives: The purpose of this study was to compare the robotic gait devices, and systematize the scientific evidences of these devices as a tool for rehabilitation of SCI individuals., Methods: A systematic review was carried out in which relevant articles were identified by searching the following databases: Cochrane Library, PubMed, PEDro and Capes Periodic. Two authors selected the articles which used a robotic device for rehabilitation of spinal cord injury., Results: Databases search found 2941 articles, 39 articles were included due to meet the inclusion criteria. The robotic devices presented distinct features, with increasing application in the last years. Studies have shown promising results regarding the reduction of pain perception and spasticity level; alteration of the proprioceptive capacity, sensitivity to temperature, vibration, pressure, reflex behavior, electrical activity at muscular and cortical level, classification of the injury level; increase in walking speed, step length and distance traveled; improvements in sitting posture, intestinal, cardiorespiratory, metabolic, tegmental and psychological functions., Conclusions: This systematic review shows a significant progress encompassing robotic devices as an innovative and effective therapy for the rehabilitation of individuals with SCI.

Perioperative monitoring of coagulation is vital to assess bleeding risks, diagnose deficiencies associated with hemorrhage, and guide hemostatic therapy in major surgical procedures, such as liver transplantation. Routine static tests demand long turnaround time and do not assess platelet function; they are determined on plasma at a standard temperature of 37°C; hence these tests are ill-suited for intraoperative use. In contrast, methods which evaluate the viscoelastic properties of whole blood, such as thromboelastogram and rotational thromboelastometry, provide rapid qualitative coagulation assessment and appropriate guidance for transfusion therapy. These are promising tools for the assessment and treatment of hyper- and hypocoagulable states associated with bleeding in liver transplantation. When combined with traditional tests and objective assessment of the surgical field, this information provides ideal guidance for transfusion strategies, with potential improvement of patient outcomes. RESUMO A monitorização perioperatória da coagulação é fundamental para estimar o risco de sangramento, diagnosticar deficiências causadoras de hemorragia e guiar terapias hemostáticas durante procedimentos cirúrgicos de grande porte, como o transplante hepático. Os testes estáticos, comumente usados na prática clínica, são insatisfatórios no intraoperatório, pois demandam tempo e não avaliam a função plaquetária; são determinados no plasma e realizados em temperatura padrão de 37°C. Os métodos que avaliam as propriedades viscoelásticas do sangue total, como o tromboelastograma e a tromboelastometria rotacional, podem suprir as deficiências dos testes estáticos tradicionais, uma vez que permitem avaliar a coagulação de forma rápida e qualitativa, guiando a terapia transfusional de forma adequada. A tromboelastometria rotacional mostrou-se promissora na avaliação e no tratamento de estados de hipercoagulação e hipocoagulação, associados a sangramento no transplante hepático. Estas informações, combinadas com os testes tradicionais e uma avaliação objetiva do campo cirúrgico, promovem um cenário ótimo para guiar as estratégias transfusionais e potencialmente melhorar o desfecho destes pacientes.

The scope of this study was to evaluate body dissatisfaction among adolescents, and to explore differences according to sex, socioeconomic status and body mass index. 4325 individuals aged 14-15 years old from Pelotas, Brazil, were studied. Body dissatisfaction was evaluated comparing the desired image with the perceived image, according to the Tiggemann & Wilson-Barret silhouette scale. The nutritional status was categorized in accordance with World Health Organization criteria, whereas socioeconomic status was evaluated using classifications of the Brazilian Association of Research Institutes. It was found that 27.6% of teens were overweight. Body dissatisfaction, appraised by the silhouette scale, affected 51% of boys and 65.6% of girls. The economic and nutritional status was associated with the outcome. Most overweight adolescents wished to have a slimmer silhouette than the perceived one. The economic level modified the effect of nutritional status on body dissatisfaction. Adolescents from the wealthier economic classes who were not in the ideal weight range manifested greater body dissatisfaction than the less affluent individuals and, irrespective of economic status, girls are more dissatisfied with excess body weight and boys with slimness.

Objective: To evaluate the association between the physical and social environment and physical activity (leisure-time and transport-related) in a population-based sample of adults., Methods: Cross-sectional study including 972 adults (20-69 years) living in the urban area of Pelotas, Brazil. Physical activity was measured using the long International Physical Activity Questionnaire. Environmental variables were assessed using a modified version of the Neighborhood Environment Walkability Scale., Results: 69.8% (95% CI = 66.9-72.7) and 51.9% (95% CI = 48.8-55.1) of the subjects did not reach 150 min/wk on leisure-time and transport-related physical activity, respectively. Subjects living near green areas were more likely to be active in leisure-time, as well as those who reported to live in safe neighborhoods. Transport-related physical activity was higher among individuals living in areas with garbage accumulation, and was lower among those living in neighborhoods which are difficult to walk or cycle due to traffic. Social support was strongly associated with leisure-time physical activity., Conclusions: Safety investments, which are urgently required in Brazil, are likely to have a desirable side effect at increasing physical activity at the population level. Building enjoyable and safe public spaces for physical activity practice must be prioritized.

Purpose: To evaluate the prevalence of physical activity advice, the source of the information, and the types of recommendation in a population-based sample of adults living in South Brazil., Methods: Population-based study including 972 adults living in Pelotas, Brazil. The outcome variable was based on the following question: "Has anyone ever recommended you to practice physical activity"? If the answer was positive, we asked who was responsible for the prescription (an open question, which was categorized later) and which recommendation was done., Results: The prevalence of physical activity advice was 56.2% (95% CI 52.3-60.1). Physical activity advice was mostly done by physicians (92.5%). Walking was, by far, the most frequent recommendation. Females were more likely to receive advice for physical activity practice than males (OR 1.74; 95% CI 1.30-2.31). Age, economic level, body mass index and leisure-time physical activity were positively associated with physical activity advice, while self-reported health presented an inverse association with the outcome., Conclusions: The prevalence of physical activity advice was high in this sample, suggesting that the Brazilian health system is incorporating physical activity in its routine.

Objective: To describe the level of physical activity in adolescents born in 1993 in the city of Pelotas, Brazil, and to evaluate the effect of sociocultural variables on those levels., Methods: A combined ethno-epidemiological methodology was employed. In the epidemiological study, 4 452 adolescents born in 1993 were interviewed. Physical activity level was evaluated by means of a questionnaire. Adolescents with fewer than 300 minutes of physical activity per week were classified as sedentary. The ethnographic study included 69 adolescents randomly selected from among all cohort participants. On average, three in-depth interviews were carried out (about one every 6 months), with mothers and adolescents being interviewed separately., Results: The prevalence of a sedentary lifestyle was 48.7% (95%CI: 46.5 to 50.8) in boys and 67.5% (95%CI: 65.6 to 69.5) in girls (P < 0.001). The independent variable presenting the strongest association with physical activity level was the weekly frequency with which the adolescent met friends outside of school. The ethnographic study showed that boys have more social and family support to engage in physical activities in adolescence, but that many parents associate poor school performance with the amount of time spent outside the home., Conclusions: This study suggests that physical activity is often viewed as competing with family values in adolescence. This perspective should be explored in future studies and discussed with families so that physical activity may be adopted as a cultural norm.
