Urban vegetation in China has changed substantially in recent decades due to rapid urbanization and dramatic climate change. Nevertheless, the spatial differentiation of greenness among major cities of China and its evolution process and drivers are still poorly understood. This study examined the spatial patterns of vegetation greenness across 289 cities in China in 2000, 2005, 2010, 2015, and 2018 by using spatial autocorrelation analysis on the Normalized Difference Vegetation Index (NDVI); then, the influencing factors were analyzed by using the optimal parameters-based geographical detector (OPGD) model and 18 natural and anthropogenic indicators. The findings demonstrated a noticeable rise in the overall greenness of the selected cities during 2000–2018. The cities in northwest China and east China exhibited the rapidest and slowest greening, respectively, among the six sub-regions. A significant positive spatial correlation was detected between the greenness of the 289 cities in different periods, but the correlation strength weakened over time. The hot and very hot spots in southern and eastern China gradually shifted to the southwest. While the spatial pattern of urban greenness in China is primarily influenced by wind speed (WS) and precipitation (PRE), the interaction between PRE and gross domestic product (GDP) has the highest explanatory power. The explanatory power of most natural factors decreased and, conversely, the influence of anthropogenic factors generally increased. These findings emphasize the variations in the influence strength of multiple factors on urban greenness pattern, which should be taken into account to understand and adapt to the changing urban ecosystem.

Abstract IL-17+ γδ T cells (γδ T17) are kick-starters of inflammation due to their strict immunosurveillance of xenobiotics or cellular damages and rapid response to pro-inflammatory stimulators. IL-27 is a well-recognized pleiotropic immune regulator with potent inhibitory effects on type 17 immune responses. However, its actions on γδ T17 mediated inflammation and the underlying mechanisms are less well understood. Here we find that IL-27 inhibits the production of IL-17 from γδ T cells. Mechanistically, IL-27 promotes lipolysis while inhibits lipogenesis, thus reduces the accumulation of lipids and subsequent membrane phospholipids, which leads to mitochondrial deactivation and ensuing reduction of IL-17. More importantly, Il27ra deficient γδ T cells are more pathogenic in an imiquimod-induced murine psoriasis model, while intracutaneous injection of rmIL-27 ameliorates psoriatic inflammation. In summary, this work uncovered the metabolic basis for the immune regulatory activity of IL-27 in restraining γδ T17 mediated inflammation, which provides novel insights into IL-27/IL-27Ra signaling, γδ T17 biology and the pathogenesis of psoriasis.

Objective To investigate the physicochemical and biological properties of a new calcium sulfate-based root canal sealer for deciduous teeth containing calcium sulfate hemihydrate, barium sulfate, chlorhexidine acetate, and polyethylene glycol 400 (PEG 400). Methods This study was reviewed and approved by the Ethics Committee. The calcium sulfate hemihydrate and barium sulfate powders with different mass percentages were mixed with liquid PEG 400 at a powder-to-liquid ratio of 3∶1, and chlorhexidine acetate was added to a concentration of 0.2 mg/mL according to the volume of PEG 400. The above materials were mechanically ground at 250 r/min for 24 h to obtain a calcium sulfate-based root canal sealer for deciduous teeth. The sealer was classified into different groups according to mass percentages of components. The mass percentages of components were optimized by performing time, fluidity, and radiopacity experiments, and then the pH, mass loss in vitro, and microscopic morphology of the optimal sealer were evaluated. The antimicrobial properties of the sealer were evaluated by a bacterial-material cocultivation method. The cytocompatibility of the sealer was evaluated by a CCK-8 assay and cytomorphological staining, and its biocompatibility was evaluated by a subcutaneous tissue embedding assay. Results After optimization, mass percentage of calcium sulfate hemihydrate was 80 wt%, and the mass percentage of barium sulfate was 20 wt%. The flowability and radiopacity of the sealer were in accordance with international standards. The pH stabilized between 6-7. On the 7th and 14th days, the pH in the water group was significantly greater than that in the PBS group (PP>0.05). In vitro degradation experiments, the mass loss of the sealer was approximately 15.17% during the preimmersion period, and rate of mass loss decreased after 3 weeks, reaching only approximately 8.33%. X-ray diffraction (XRD) and scanning electron microscopy (SEM) revealed that the main component of the sealer after hydration was calcium sulfate dehydrate. In bacterial growth assays and cytological tests, the sealer showed significant inhibition of the growth of E. faecalis (PPP>0.05). Both the sealer group and the control group (Vitapex and zinc oxide eugenol) caused mild inflammatory reactions in tissue sections. Conclusion In this study, a new type of root canal sealer for deciduous teeth was designed based on calcium sulfate, which has good physicochemical properties and strong antibacterial properties and meets biocompatibility requirements. This study provides an idea for the development of a new type of root canal sealer for deciduous teeth.

【Objective】 Non-pressurized mesh filters are often installed in front of pumps to filter sediments and debris. In this paper, we study the influence of different irrigation water sources and backwash device parameters on hydraulic performance of a new network filter. 【Method】 The water flow rate in the experiment ranged from 120 to 160 m3/h, and the sediment concentration in the water varied from 0.2 to 0.8 g/L. The backwash varied from 1 to 20 m3/h, and the velocity gradient in the filter box changed from 1 to 4 r/min. A multi factor dispersion analysis method was used to analyze the experimental results. 【Result】 ① The initial sediment concentration had minimal impact on hydraulic performance of the mesh filter, contributing only 0.010 8 to the overall effect. However, the influence of water flow duration on the initial sediment concentration was significant, exceeding 159%. ② The fourth model provided the best fit for the overall regression analysis, with a residual value of 0.804 4, significantly higher than other models; its coefficient of determination R2 was 0.986 1. ③ the optimal performance was achieved when flow rate was 120.198 m3/h, velocity was 1.112 r/min, and self-cleaning flow rate was 19.934 m3/h. The associated a minimum energy loss was 16.024 kPa. 【Conclusion】 The model we proposed can accurately predict the impact of operation conditions and sediment concentration in the water on the performance of the non-pressurized mesh filters. Selecting optimal operation conditions can minimize the energy loss across the filter.

As one of the important forms of equipment for underground auxiliary transportation in coal mines, monorail cranes have the advantages of large carrying capacity and strong climbing ability, and unmanned driving is its inevitable development direction in the context of intelligent construction of coal mines. In order to ensure the safe driving of the unmanned monorail crane in the underground roadway, it is particularly important for the reliable detection of track joints and key targets. Firstly, the mine image dataset was enhanced to improve its diversity. A YOLOv5 algorithm with improved channel attention mechanism ECA_s and EIOU regression loss function was proposed, and the improved YOLOv5 algorithm was experimentally analyzed, and the results showed that the recognition accuracy was increased by 9.1% and mAP by 3.6% by using the improved YOLOv5 algorithm. Secondly, a machine vision-based single-track crane track seam detection method was established, and the track seam distance was calculated according to the calibration coefficient by using image preprocessing, histogram information statistics, morphological processing and other technologies, and the results showed that the detection error of a seam image processed by the track seam detection algorithm based on machine vision was only 0.3 mm. Finally, the unmanned target detection test of the monorail crane is carried out, and the results show that the average accuracy of the improved YOLOv5 target detection algorithm reaches 90.3%, which has a higher detection accuracy. The average detection error of the seam image processed by the monorail crane rail seam detection algorithm is 0.73 mm, and the maximum detection error is not more than 1.1 mm, which has higher detection accuracy. In order to ensure the safe passage and reliability of unmanned monorail cranes in coal mines, a pragmatic and feasible detection scheme and an accurate and reliable detection algorithm are provided.

Abstract Natural killer (NK) cells play a crucial role in immune response against viral infections and tumors. However, further investigation is needed to better understand the key molecules responsible for determining the fate and function of NK cells. In this study, we made an important discovery regarding the involvement of the Hippo kinases Mst1 and Mst2 as novel regulators in maintaining mouse NK cell homeostasis. The presence of high Mst1 and Mst2 (Mst1/2) activity in NK cells is essential for their proper development, survival and function in a canonical Hippo signaling independent mode. Mechanistically, Mst1/2 induce cellular quiescence by regulating the processes of proliferation and mitochondrial metabolism, thereby ensuring the development and survival of NK cells. Furthermore, Mst1/2 effectively sense IL-15 signaling and facilitate the activation of pSTAT3-TCF1, which contributes to NK cell homeostasis. Overall, our investigation highlights the crucial role of Mst1/2 as key regulators in metabolic reprogramming and transcriptional regulation for mouse NK cell survival and function, emphasizing the significance of cellular quiescence during NK cell development and functional maturation.

Background HBM4003 is a novel anti-CTLA-4 heavy chain-only antibody, designed to enhance Treg ablation and antibody-dependent cell-mediated cytotoxicity while ensuring a manageable safety profile. This phase I trial investigated the safety, pharmacokinetics, immunogenicity and preliminary efficacy of HBM4003 plus with anti-PD-1 antibody toripalimab in patients with advanced solid tumors, especially focusing on melanoma.Methods The multicenter, open-label phase I trial was divided into two parts: dose-escalation phase (part 1) and dose-expansion phase (part 2). In part 1, HBM4003 was administered at doses of 0.03, 0.1, 0.3 mg/kg in combination with toripalimab with fixed dosage of 240 mg every 3 weeks. The recommended phase II dose (RP2D) was used in the expansion phase. Primary endpoints were safety and RP2D in part 1 and objective response rate (ORR) in part 2. Biomarkers based on cytokines and multiplex immunofluorescence staining were explored.Results A total of 40 patients received study treatment, including 36 patients treated with RP2D of HBM4003 0.3 mg/kg plus toripalimab 240 mg every 3 week. 36 participants (90.0%) experienced at least one treatment-related adverse event (TRAE), of which 10 (25.0%) patients experienced grade ≥3 TRAEs and 5 (12.5%) experienced immune-mediated adverse events (irAEs) with maximum severity of grade 3. No grade 4 or 5 irAEs occurred. Efficacy analysis set included 32 melanoma patients treated with RP2D and with available post-baseline imaging data. The ORRs of anti-PD-1/PD-L1 treatment-naïve subgroup and anti-PD-1/PD-L1 treatment-failed subgroup were 33.3% and 5.9%, respectively. In mucosal melanoma, the ORR of the two subgroups were 40.0% and 10.0%, respectively. Baseline high Treg/CD4+ratio in the tumor serves as an independent predictive factor for the efficacy of immunotherapy.Conclusions HBM4003 0.3 mg/kg plus toripalimab 240 mg every 3 week demonstrated manageable safety in solid tumors and no new safety signal. Limited data demonstrated promising antitumor activity, especially in PD-1 treatment-naïve mucosal melanoma.Trial registration number NCT04727164.

Background: Postoperative recurrence of tongue squamous cell carcinoma (TSCC) has always been a clinical problem for patients and doctors. Surgery and radiotherapy are the main treatment methods for TSCC, but reoperation often leads to functional impairment. Side effects of radiotherapy include mucosal gland damage, dry mouth, weakened or lost taste. Improved treatment is needed. Objective: To evaluate the clinical outcome of a patient with TSCC treated with Microdrop aminolevulinic acid (ALA) photodynamic treatment (PDT) twice. Methods: ALA was dissolved in 5 % lidocaine and the concentration of ALA was 20 % by Microdrop method. Then, the tumor tissue was expanded 1 cm outward, and the injection points were evenly distributed with an upper and lower left and right interval of 2–3 mm. The 1 ml syringe was used to perform the injection in the skin of the tumor area, and there was a small cuticle at each injection point. A pathologically confirmed patient with TSCC received twice Microdrop ALA-PDT treatments, which were evaluated at 1 month and 4 months later. Results: After 3 h of Microdrop ALA injection, the wavelength of semiconductor laser was set to 630 nm, and the energy of 300 mW /cm2 was irradiated for 30 min. After two treatments, the lesions were not visible, and no recurrence occurred after 4 months of follow-up. The patient's tongue function was well preserved and the cosmetic effect was good. Conclusion: Microdrop ALA-PDT may be effective in the salvage treatment of select tongue squamous cell carcinoma.

Abstract In recent years, video‐based hand–object interaction has received widespread attention from researchers. However, due to the complexity and occlusion of hand movements, hand–object interaction recognition based on RGB videos remains a highly challenging task. Here, an end‐to‐end spatio‐temporal former (STFormer) network for understanding hand behaviour in interactions is proposed. The network consists of three modules: FlexiViT feature extraction, hand–object pose estimator, and interaction action classifier. The FlexiViT is used to extract multi‐scale features from each image frame. The hand–object pose estimator is designed to predict 3D hand pose keypoints and object labels for each frame. The interaction action classifier is used to predict the interaction action categories for the entire video. The experimental results demonstrate that our approach achieves competitive recognition accuracies of 94.96% and 88.84% on two datasets, namely first‐person hand action (FPHA) and 2 Hands and Objects (H2O).

Background: Although the use of anti-PD-1 antibodies has fundamentally changed traditional cancer treatment, most patients are resistant to anti-PD-1 treatment. Glucocorticoids (GCs) play an important role in tumorigenesis and tumor progression, but the role of endogenous GCs in resistance to anti-PD-1 antibody therapy remains unclear. Methods: Single cell-derived cell lines (SCDCLs) were generated from a colorectal cancer cell line (CT26) using limiting dilution. We analyzed tumor tissues from anti-PD-1 antibody-treated and untreated mice inoculated with SCDCLs via transcriptome sequencing and flow cytometry to detect pathway activity and immune cell composition changes in the tumor microenvironment. Results: Five SCDCLs were inoculated into wild-type BALB/c mice (all tumorigenic). Single-cell clone (SCC)-2 exhibited the slowest growth rates both in vivo and in vitro compared to other single-cell clones, and better long-term survival than SCC1 and CT26. Flow cytometry showed that SCC2 tumor-bearing mice exhibited significantly higher infiltration of T cells within the tumor tissue, and higher expression of PD-1 on these T cells than the other groups in vivo. However, the SCC2 group showed no response to anti-PD-1 therapy. Transcriptome analysis revealed that the SCC2 group exhibited increased expression of genes related to GC (Hsd11b1, Sgk3, Tgfbr2, and Il7r) compared to SCC2-anti-PD-1 treated tumors. Conclusions: GC pathway activation is related to resistance to anti-PD-1 therapy.

Abstract Traditional approaches to coastal defence often struggle to reduce the risks of accelerated climate change. Incorporating nature-based components into coastal defences may enhance adaptation to climate change with added benefits, but we need to compare their performance against conventional hard measures. We conduct a meta-analysis that compares the performances of hard, hybrid, soft and natural measures for coastal defence across different functions of risk reduction, climate change mitigation, and cost-effectiveness. Hybrid and soft measures offer higher risk reduction and climate change mitigation benefits than unvegetated natural systems, while performing on par with natural measures. Soft and hybrid measures are more cost-effective than hard measures, while hybrid measures provide the highest hazard reduction among all measures. All coastal defence measures have a positive economic return over a 20-year period. Mindful of risk context, our results provide strong an evidence-base for integrating and upscaling nature-based components into coastal defences in lower risk areas.

Abstract For the problem of space dynamic target tracking with occlusion, this paper proposes an online tracking method based on the combination between the five-frame difference and Deepsort (Simple Online and Realtime Tracking with a Deep Association Metric), which is to achieve the identification first and then tracking of the dynamic target. First of all, according to three-frame difference, the five-frame difference is improved, and through the integration with ViBe (Visual Background Extraction), the accuracy and anti-interference ability are enhanced; Secondly, the YOLOv5s (You Look Only Once) is improved using preprocessing of DWT (Discrete Wavelet Transformation) and injecting GAM (Global Attention Module), which is considered as the detector for Deepsort to solve the missing in occlusion, and the real-time and accuracy can be strengthened; Lastly, simulation results show that the proposed space dynamic target tracking can keep stable to track all dynamic targets under the background interference and occlusion, the tracking precision is improved to 93.88%. Furthermore, there is a combination with the physical depth camera D435i, experiments on target dynamics show the effectiveness and superiority of the proposed recognition and tracking algorithm in the face of strong light and occlusion.

Abstract Objective Metagenomic next-generation sequencing (mNGS), as an emerging technique for pathogen detection, has been widely used in clinic. However, reports on the application of mNGS in cancer patients with severe pneumonia remain limited. This study aims to evaluate the diagnostic performance of bronchoalveolar lavage fluid (BALF) mNGS in cancer patients complicated with severe pneumonia. Methods A total of 62 cancer patients with severe pneumonia simultaneously received culture and mNGS of BALF were enrolled in this study. We systematically analyzed the diagnostic significance of BALF mNGS. Subsequently, optimization of anti-infective therapy based on the distribution of pathogens obtained from BALF mNGS was also assessed. Abstract Aims We previously showed that the nab-paclitaxel plus S-1 (NPS) regimen had promising effects against metastatic pancreatic ducal adenocarcinoma (mPDAC), whose efficacy however could not be precisely predicted by routine biomarkers. This prospective study aimed to investigate the values of mutations in circulating tumor DNA (ctDNA) and their dynamic changes in predicting response of mPDAC to NPS chemotherapy. Methods Paired tumor tissue and blood samples were prospectively collected from patients with mPDAC receiving first-line NPS chemotherapy, and underwent next-generation sequencing with genomic profiling of 425 genes for ctDNA. High mutation allelic frequency (MAF) was defined as ≥ 30% and ≥ 5% in tumor tissue and blood, respectively. Kappa statistics were used to assess agreement between mutant genes in tumor and ctDNA. Associations of mutations in ctDNA and their dynamic changes with tumor response, overall survival (OS), and progression-free survival (PFS) were assessed using the Kaplan–Meier method, multivariable-adjusted Cox proportional hazards regression, and longitudinal data analysis. Results 147 blood samples and 43 paired tumor specimens from 43 patients with mPDAC were sequenced. The most common driver genes with high MAF were KRAS (tumor, 35%; ctDNA, 37%) and TP53 (tumor, 37%; ctDNA, 33%). Mutation rates of KRAS and TP53 in ctDNA were significantly higher in patients with liver metastasis, with baseline CA19-9 ≥ 2000 U/mL, and/or without an early CA19-9 response. κ values for the 5 most commonly mutated genes between tumor and ctDNA ranged from 0.48 to 0.76. MAFs of the genes mostly decreased sequentially during subsequent measurements, which significantly correlated with objective response, with an increase indicating cancer progression. High mutations of KRAS and ARID1A in both tumor and ctDNA, and of TP53, CDKN2A, and SMAD4 in ctDNA but not in tumor were significantly associated with shorter survival. When predicting 6-month OS, AUCs for the 5 most commonly mutated genes in ctDNA ranged from 0.59 to 0.84, larger than for genes in tumor (0.56 to 0.71) and for clinicopathologic characteristics (0.51 to 0.68). Repeated measurements of mutations in ctDNA significantly differentiated survival and tumor response. Among the 31 patients with ≥ 2 ctDNA tests, longitudinal analysis of changes in gene MAF showed that ctDNA progression was 60 and 58 days ahead of radiologic and CA19-9 progression for 48% and 42% of the patients, respectively. Conclusions High mutations of multiple driving genes in ctDNA and their dynamic changes could effectively predict response of mPDAC to NPS chemotherapy, with promising reliable predictive performance superior to routine clinicopathologic parameters. Inspiringly, longitudinal ctDNA tracking could predict disease progression about 2 months ahead of radiologic or CA19-9 evaluations, with the potential to precisely devise individualized therapeutic strategies for mPDAC.
