Insufficient therapeutic strategies for acute kidney injury (AKI) necessitate precision therapy targeting its pathogenesis. This study reveals the new mechanism of the marine-derived anti-AKI agent, piericidin glycoside S14, targeting peroxiredoxin 1 (PRDX1). By binding to Cys83 of PRDX1 and augmenting its peroxidase activity, S14 alleviates kidney injury efficiently in Prdx1-overexpression (Prdx1-OE) mice. Besides, S14 also increases PRDX1 nuclear translocation and directly activates the Nrf2/HO-1/NQO1 pathway to inhibit ROS production. Due to the limited druggability of S14 with low bioavailability (2.6%) and poor renal distribution, a pH-sensitive kidney-targeting dodecanamine-chitosan nanoparticle system is constructed to load S14 for precise treatment of AKI. l-Serine conjugation to chitosan imparts specificity to kidney injury molecule-1 (Kim-1)-overexpressed cells. The developed S14-nanodrug exhibits higher therapeutic efficiency by improving the in vivo behavior of S14 significantly. By encapsulation with micelles, the AUC0‒t, half-life time, and renal distribution of S14 increase 2.5-, 1.8-, and 3.1-fold, respectively. The main factors contributing to the improved druggability of S14 nanodrugs include the lower metabolic elimination rate and UDP-glycosyltransferase (UGT)-mediated biotransformation. In summary, this study identifies a new therapeutic target for the marine-derived anti-AKI agent while enhancing its ADME properties and druggability through nanotechnology, thereby driving advancements in marine drug development for AKI.

Abstract Patients who suffer from sepsis typically experience acute lung injury (ALI). Extracellular vesicles (EVs) contain miRNAs, which are potentially involved in ALI. However, strategies to screen more effective EV-miRNAs as therapeutic targets are yet to be elucidated. In this study, functional EV-miRNAs were identified based on multiomics analysis of single-cell RNA sequencing of targeted organs and serum EV (sEV) miRNA profiles in patients with sepsis. The proportions of neutrophils and macrophages were increased significantly in the lungs of mice receiving sEVs from patients with sepsis compared with healthy controls. Macrophages released more EVs than neutrophils. MiR-125a-5p delivery by sEVs to lung macrophages inhibited Tnfaip3, while miR-221-3p delivery to lung neutrophils inhibited Fos. Macrophage membrane nanoparticles (MM NPs) loaded with an miR-125a-5p inhibitor or miR-221-3p mimic attenuated the response to lipopolysaccharide (LPS)-induced ALI. Transcriptome profiling revealed that EVs derived from LPS-stimulated bone marrow-derived macrophages (BMDMs) induced oxidative stress in neutrophils. Blocking toll-like receptor, CXCR2, or TNFα signaling in neutrophils attenuated the oxidative stress induced by LPS-stimulated BMDM-EVs. This study presents a novel method to screen functional EV-miRNAs and highlights the pivotal role of macrophage-derived EVs in ALI. MM NPs, as delivery systems of key sEV-miRNA mimics or inhibitors, alleviated cellular responses observed in sepsis-induced ALI. This strategy can be used to reduce septic organ damage, particularly lung damage, by targeting EVs.

Background: The fingertip amputation is an amputation type of the finger beyond the proximal nail fold. There is no vein available for anastomoses on the dorsal side of the finger, and the palmar vein of the finger is small and tightly attached to the skin. Therefore, it is relatively difficult to implement surgical anastomoses, which poses challenges to the clinical treatment of fingertip amputations. Case report: A 29-year-old male was admitted to the hospital due to “the amputation of the fingertips of the right index, middle, and ring fingers caused by a heavy object compression 3 h ago”. The admission examination revealed that the right index, middle, and ring fingers were completely severed at the 1/2 plane of the nail bed, with irregular sections, severe contusion, and pollution. The X-ray examination showed comminuted fractures of the distal phalanges of the right index, middle, and ring fingers. Based on these findings, the patient was diagnosed with multiple severed fingertips of the right hand (Tamai Zone 1). The patient underwent debridement, vascular exploration, and replantation of the right index, middle, and ring fingertips under emergency general anesthesia. After surgery, anti-inflammatory, spasmolytic, and anticoagulant treatment and regular dressing changes were conducted. The patient did not receive a blood transfusion, and all three fingers survived. The appearance of these fingers was favorable 3 months after surgery, and the flexion and extension of these fingers were normal. Eventually, the patient achieved excellent Chen's hand function scores. Conclusions: To the best of our knowledge, this may be the first successful case regarding the replantation of three fingertips after amputations in Tamai Zone 1 with favorable outcomes. It can be maintained that super microsurgery can be used for the replantation of multiple fingertip amputations.

Fluorescence imaging with high spatiotemporal resolution and sensitivity is employed for in vivo visualization and detection applications. Compared with visible light and the first near-infrared window (700–900 nm), the second near-infrared window (1 000–1 700 nm) imaging has a lower auto-background fluorescence, deeper tissue penetration, and a higher signal-to-noise ratio, thus highlighting its broad prospects in the biomedical field. Currently, reported second near-infrared region probes include single-walled carbon nanotubes, rare-earth nanoparticles, quantum dots, metal materials, and organic molecular dyes. Multimodal imaging can overcome the limitations of single imaging and provide more comprehensive information on the anatomical structure, tissue composition, and cellular and molecular quantification of lesions to achieve multi-level visualization. Therefore, second near-infrared window nanoprobes must be engineered for multimodal imaging. Moreover, these nanoprobes can be actively targeted by modification with antibodies, peptides, nucleotides, or biofilms to obtain detailed and accurate information on biological systems. This review describes the active targeting capabilities of various second near-infrared window nanoprobes in multimodal imaging, diagnosis, and treatment of different diseases by carrying different ligands and the common features and future application prospects of second near-infrared window fluorescent nanoprobes with targeting ability.

Abstract Background Intrauterine adhesion (IUA) is one of the most severe causes of infertility in women of childbearing age with injured endometrium secondary to uterine performance. Stem cell therapy is effective in treating damaged endometrium. The current reports mainly focus on the therapeutic effects of stem cells through paracrine or transdifferentiation, respectively. This study investigates whether paracrine or transdifferentiation occurs preferentially in treating IUA. Methods Human amniotic mesenchymal stem cells (hAMSCs) and transformed human endometrial stromal cells (THESCs) induced by transforming growth factor beta (TGF-β1) were co-cultured in vitro. The mRNA and protein expression levels of Fibronectin (FN), Collagen I, Cytokeratin19 (CK19), E-cadherin (E-cad) and Vimentin were detected by Quantitative real-time polymerase chain reaction (qPCR), Western blotting (WB) and Immunohistochemical staining (IHC). The Sprague-Dawley (SD) rats were used to establish the IUA model. hAMSCs, hAMSCs-conditional medium (hAMSCs-CM), and GFP-labeled hAMSCs were injected into intrauterine, respectively. The fibrotic area of the endometrium was evaluated by Masson staining. The number of endometrium glands was detected by hematoxylin and eosin (H&E). GFP-labeled hAMSCs were traced by immunofluorescence (IF). hAMSCs, combined with PPCNg (hAMSCs/PPCNg), were injected into the vagina, which was compared with intrauterine injection. Results qPCR and WB revealed that FN and Collagen I levels in IUA-THESCs decreased significantly after co-culturing with hAMSCs. Moreover, CK19, E-cad, and Vimentin expressions in hAMSCs showed no significant difference after co-culture for 2 days. 6 days after co-culture, CK19, E-cad and Vimentin expressions in hAMSCs were significantly changed. Histological assays showed increased endometrial glands and a remarkable decrease in the fibrotic area in the hAMSCs and hAMSCs-CM groups. However, these changes were not statistically different between the two groups. In vivo, fluorescence imaging revealed that GFP-hAMSCs were localized in the endometrial stroma and gradually underwent apoptosis. The effect of hAMSCs by vaginal injection was comparable to that by intrauterine injection assessed by H&E staining, MASSON staining and IHC. Conclusions Our data demonstrated that hAMSCs promoted endometrial repair via paracrine, preferentially than transdifferentiation.

We presented an image space branch-and-bound algorithm for globally minimizing the sum of linear ratios problem. In the algorithm, a new linearizing technique was proposed for deriving the linear relaxation problem. An image space region reduction technique was constructed for improving the convergence speed of the algorithm. Moreover, by analyzing the computational complexity of the algorithm, the maximum iterations of the algorithm were estimated, and numerical experimental results showed the potential computing benefits of the algorithm. Finally, a practical application problem in education investment was solved to verify the usefulness of the proposed algorithm.

Abstract Fundus fluorescein angiography (FFA) is a crucial diagnostic tool for chorioretinal diseases, but its interpretation requires significant expertise and time. Prior studies have used Artificial Intelligence (AI)-based systems to assist FFA interpretation, but these systems lack user interaction and comprehensive evaluation by ophthalmologists. Here, we used large language models (LLMs) to develop an automated interpretation pipeline for both report generation and medical question-answering (QA) for FFA images. The pipeline comprises two parts: an image-text alignment module (Bootstrapping Language-Image Pre-training) for report generation and an LLM (Llama 2) for interactive QA. The model was developed using 654,343 FFA images with 9392 reports. It was evaluated both automatically, using language-based and classification-based metrics, and manually by three experienced ophthalmologists. The automatic evaluation of the generated reports demonstrated that the system can generate coherent and comprehensible free-text reports, achieving a BERTScore of 0.70 and F1 scores ranging from 0.64 to 0.82 for detecting top-5 retinal conditions. The manual evaluation revealed acceptable accuracy (68.3%, Kappa 0.746) and completeness (62.3%, Kappa 0.739) of the generated reports. The generated free-form answers were evaluated manually, with the majority meeting the ophthalmologists’ criteria (error-free: 70.7%, complete: 84.0%, harmless: 93.7%, satisfied: 65.3%, Kappa: 0.762–0.834). This study introduces an innovative framework that combines multi-modal transformers and LLMs, enhancing ophthalmic image interpretation, and facilitating interactive communications during medical consultation.

Polyurethane (PU) films have established a distinctive position among various protective materials due to their outstanding chemical and physical properties. However, their limited moisture barrier performance hinders their broader applications. To solve this difficult problem, PU film is first modified by adding reactive fillers (MgO) and physical barrier fillers (Modified graphene oxide, M-GO), and then Si-DLC coating is deposited on the surface of the film by hollow cathode plasma immersion ion implantation (HCPIII) method at near room temperature. MgO can react with the water molecules diffused into the film, while the addition of M-GO extends the diffusion path of water molecules. It is revealed that the addition of 5% MgO and 0.5% M-GO, along with the deposition of a 2.56 μm Si-doped DLC coating, leads to a remarkable 14.67-fold reduction in the water vapor transmittance rate (WVTR). The result of molecular dynamics simulations shows that the reactive sheet filler captures the water molecules and reduces the WVTR. In addition, the combination method of PU body modification and surface coating is suitable for polyurethane foam (PUF) to reduce the WVTR as a double coating. It's amazing that the WVTR of PUF significantly reduces by 15.29 times after double-coating. The combination of body modification and surface coating provides an effective route for significantly improving the moisture barrier property of hydrophilic polymer film and polymer foam.

The purpose of this research was to develop a chitosan sulfobetaine (CS-SNCC) film via the solution-casting method as a biodegradable antibacterial material for biomedical applications. Chitosan and monochloro-triazine sulfobetaine were used as the raw materials for CS-SNCC preparation, and Fourier-transform infrared (FTIR), ultraviolet–visible (UV-Vis), energy-dispersive X-ray (EDX), and X-ray photoelectron spectroscopy (XPS) spectra were used to characterize and analyze the structure of the synthesized CS-SNCC. Furthermore, the swelling property, thermal stability, biodegradability, cytocompatibility, and antibacterial properties of the CS-SNCC film were comprehensively investigated and compared with those of the chitosan film. The results for the film’s enzymatic biodegradation behavior show that the CS-SNCC film undergoes a weight loss of 45.54% after 21 days of incubation. In addition, the CS-SNCC film effectively resists bacterial adhesion, prevents the formation of bacteria biofilms, and exhibits high antibacterial activity, with inactivation rates of 93.43% for Escherichia coli and 91.00% for Staphylococcus aureus. Moreover, the CS-SNCC film shows good cellular activity and cytocompatibility according to the cytotoxicity results. Therefore, the prepared biodegradable, cytocompatible, antibacterial, and biofilm-controlling CS-SNCC film has potential for biomedical applications.

In order to explore the changes in fracture mechanical properties of pipeline steel welded joints in hydrogen environment, the notched tensile in-situ hydrogen charging method was used to study the hydrogen embrittlement susceptibility of X80 steel base metal, welds and heat-affected zones under a total pressure of 12 MPa and a hydrogen partial pressure of 0.36 MPa. A high-pressure slow strain rate tensile testing machine was employed to conduct notch tensile test, and a high-pressure fracture toughness testing machine and a high-pressure corrosion fatigue testing machine were used to carry out fracture toughness and fatigue crack growth rate tests on X80 steel, respectively. The crack propagation behavior of X80 steel was studied by combining fracture toughness tests and fatigue crack propagation rate tests. Results showed that under the same conditions, compared with the base metal and heat-affected zone, the reduction of area at the weld position was significantly reduced, showing a higher susceptibility to hydrogen embrittlement. Compared with the original data in air at normal temperature and pressure, the crack tip opening displacement(CTOD) value of the weld position in the hydrogen partial pressure environment of 0.36 MPa changed slightly, and no secondary cracks appeared on the fracture surface. Compared with the air environment, the fatigue crack growth rate of X80 steel welds in a hydrogen partial pressure environment of 0.36 MPa increased by 1 order of magnitude, signifying that hydrogen could increase the fatigue crack growth rate of the material. However, according to the actual pipeline pressure fluctuation, the hydrogen embrittlement sensitivity of X80 steel was smaller under 0.36 MPa hydrogen partial pressure.

ObjectiveTo determine the serum antibody level and risk factors in the adolescent population in a county in Zhejiang Province， following the immunization with inactivated COVID-19 vaccine， and to construct a prediction model for antibody concentration.MethodsWe conducted the study in a county in Zhejiang Province， employing a stratified cluster random sampling strategy in school children who had received the inactivated COVID-19 vaccine. Data on gender， age， type of vaccine， and time of vaccination was collected. Serum samples were also collected to test for anti-S and N IgG antibody against the SARS-CoV-2 by using chemiluminescent immunoassay （CLIA）. Risk factors were determined to construct a prediction model for antibody concentration.ResultsThe IgG antibody concentration was significantly higher in girls， those who received two doses， and those who had simply received the KX vaccine . It decreased with age and time interval between the sampling and last vaccination. The prediction model constructed by random forest regression in the study had a better model fit and predictive ability than that by the multivariable linear stepwise regression.ConclusionGender， age， vaccination dose， type of vaccine， and time of vaccination are associated with vaccination effectiveness of inactivated COVID-19 vaccines in adolescents. Prediction model could predict the antibody level in the vaccinated population， which can provide a new tool for better evaluation of vaccination effectiveness against emerging infectious diseases in future.

Abstract Sugarcane is a primary sugar crop and an important source of bioenergy. Pathogens are the major factors affecting sugarcane yield and sugar content. However, the mechanisms of sugarcane defense regulation remain largely unknown, and research on prospective genetic targets for modification is scarce. As the main class of calcium sensors, calcium-dependent protein kinases (CDPKs/CPKs) play a crucial role in the immune regulatory network. Using sugarcane genomic data, we identified 229 putative ScCDPKs in primordial specie Saccharum officinarum. Searching the putative CDPKs in sugarcane cultivars from National Center for Biotechnology Information (NCBI), 12 putative ScCDPKs in cultivars were identified. Phylogenetic analysis revealed evolutionary relationships among these CDPKs in sugarcane cultivars and those in S. officinarum, Arabidopsis thaliana, and rice. Truncation mutants of ScCDPKs were introduced into the reporter system and examined for activity in inducing Sugarcane Pathogenesis Related Protein 1 (ScPR1). The results showed that truncated ScCDPK1 and ScCDPK8 induce higher expression of ScPR1 than full-length ScCDPK1 and ScCDPK8. Additionally, transient expression of truncated ScCDPK1 and ScCDPK8 exhibit stronger activity in sugarcane protoplasts for activation of ScPR1 and sugarcane Serine Protease Inhibitor (ScSPI). These results demonstrate that ScCDPK1 and ScCDPK8 possess auto-inhibitory activity. The findings of this study provide a basis for an in-depth study of the sugarcane CDPK gene family and lay the foundation for further genetic improvement.

Abstract Background Intraventricular hemorrhage (IVH) and associated hydrocephalus are significant complications of intracerebral and subarachnoid hemorrhage. Despite proximity to IVH, the immune cell response at the choroid plexus (ChP) has been relatively understudied. This study employs CX3CR-1GFP mice, which marks multiple immune cell populations, and immunohistochemistry to outline that response. Methods This study had four parts all examining male adult CX3CR-1GFP mice. Part 1 examined naïve mice. In part 2, mice received an injection 30 µl of autologous blood into right ventricle and were euthanized at 24 h. In part 3, mice underwent intraventricular injection of saline, iron or peroxiredoxin 2 (Prx-2) and were euthanized at 24 h. In part 4, mice received intraventricular iron injection and were treated with either control or clodronate liposomes and were euthanized at 24 h. All mice underwent magnetic resonance imaging to quantify ventricular volume. The ChP immune cell response was examined by combining analysis of GFP(+) immune cells and immunofluorescence staining. Results IVH and intraventricular iron or Prx-2 injection in CX3CR-1GFP mice all induced ventriculomegaly and activation of ChP immune cells. There were very marked increases in the numbers of ChP epiplexus macrophages, T lymphocytes and neutrophils. Co-injection of clodronate liposomes with iron reduced the ventriculomegaly which was associated with fewer epiplexus and stromal macrophages but not reduced T lymphocytes and neutrophils. Conclusion There is a marked immune cell response at the ChP in IVH involving epiplexus cells, T lymphocytes and neutrophils. The blood components iron and Prx-2 may play a role in eliciting that response. Reduction of ChP macrophages with clodronate liposomes reduced iron-induced ventriculomegaly suggesting that ChP macrophages may be a promising therapeutic target for managing IVH-induced hydrocephalus.

Farnesyl pyrophosphate synthase (FPPS) catalyzes the synthesis of C15 farnesyl diphosphate (FPP) from C5 dimethylallyl diphosphate (DMAPP) and two or three C5 isopentenyl diphosphates (IPPs). FPP is an important precursor for the synthesis of isoprenoids and is involved in multiple metabolic pathways. Here, farnesyl pyrophosphate synthase from Sporobolomyces pararoseus NGR (SpFPPS) was isolated and expressed by the prokaryotic expression system. The SpFPPS full-length genomic DNA and cDNA are 1566 bp and 1053 bp, respectively. This gene encodes a 350-amino acid protein with a predicted molecular mass of 40.33 kDa and a molecular weight of 58.03 kDa (40.33 kDa + 17.7 kDa), as detected by SDS-PAGE. The function of SpFPPS was identified by induction, purification, protein concentration and in vitro enzymatic activity experiments. Structural analysis showed that Y90 was essential for chain termination and changing the substrate scope. Site-directed mutation of Y90 to the smaller side-chain amino acids alanine (A) and lysine (K) showed in vitro that wt-SpFPPS catalyzed the condensation of the substrate DMAPP or geranyl diphosphate (GPP) with IPP at apparent saturation to synthesize FPP as the sole product and that the mutant protein SpFPPS-Y90A synthesized FPP and C20 geranylgeranyl diphosphate (GGPP), while SpFPPS-Y90K hydrolyzed the substrate GGPP. Our results showed that FPPS in S. pararoseus encodes the SpFPPS protein and that the amino acid substitution at Y90 changed the distribution of SpFPPS-catalyzed products. This provides a baseline for potentially regulating SpFPPS downstream products and improving the carotenoid biosynthesis pathway.

Shaanxi province is rich in coal resources and is one of the main coal-producing provinces in China, with a coal production of 746 million tons in 2022. Due to the obvious differences in geological and hydrogeological conditions in the coal mining areas of the province, there are various types of water disasters in its coal mines. With the increase of coal mining intensity in recent years, the frequency of water disasters increases significantly, and the situation of water prevention and control is challenging. On the basis of the overall analysis of the hydrogeological conditions in the coal mining areas of Shaanxi province, combined with the types of water disasters revealed in the mining process of various mining areas in the province, the regional distribution of all kinds of water disas-ters is systematically summarized, and the formation mechanism and characteristics of typical water disasters are discussed. Also, the corresponding prevention and control technology and progress are put forward. The results show that ① the main coal producing areas in Shaanxi province are divided into northern Shaanxi, Huanglong and Weibei. The Jurassic coalfield in northern Shaanxi is mainly affected by the water disaster of roof's loose sand layer, the water disaster of thick sandstone, water and sand burst disaster, and the burnt rock flood exists in some coal mines. The Huanglong coalfield is mainly threatened by roof super-thick sandstone water, separated layer water, mud and sand burst disaster or hidden danger. The Carboniferous-Permian coalfields in northern Shaanxi and Weibei are mainly threatened by the water damage of Ordovician limestone on the coal seam floor. ② The hidden danger of roof flood in the Jurassic coalfield in northern Shaanxi is mainly formed by the disturbance of roof aquifer by mining water diversion fracture zone, which can form a continuous large flow water gushing through sandstone aquifer or loose sand layer, and the water filling intensity is large in some areas. Roof flood occurs when the amount of water exceeds the capacity of the drainage system in a short time, and it can be transformed into water and sand burst disaster when the collapse zone in the thin bedrock area directly leads to the loose sand layer. When the water-conducting fracture zone affects the water-rich area of burnt rock, it can form a flood of burnt rock with large instantaneous water volume and continuous flood. The super-thick sandstone aquifer on the roof of the Huanglong coalfield is swept by the mining water-conducting fracture zone to form a high-intensity continuous water gushing and may form roof flood. When the thickness of the lower aquifer is large, the separated layer water burst disaster may be formed. When the roof strata near the coal seam is soft and easy to collapse, it can be transformed into mud and sand burst disaster. In the Carboniferous-Permian coalfield and the Weibei coalfield in northern Shaanxi, when structures such as coal seam mining floor disturbance failure zones or faults lead to the Ordovician limestone aquifers, they will cause extremely serious floor water inrush disasters, which are characterized by strong sudden occurrence and large instantaneous water volume. ③ The roof water disaster treatment technology in each region mainly includes drilling hole drainage, grouting treatment, mining parameter control and so on, and the floor water damage is mainly treated by regional grouting reinforcement and plugging. Burnt rock water disaster is mainly treated by the combination of curtain grouting and drilling exploration.

Abstract Obstruction and/or reflux compromise during venous emptying can facilitate different pathophysiologies in chronic venous insufficiency (CVI). We present a patient with persistent lower limb CVI edema caused by post-thrombotic syndrome (PTS), who responded well to femoral vein valve therapy via axillary vein bypass after unsuccessful valvuloplasty, and led a normal life. During a 12 month observation period, bridging vessels completely restored original anatomical structures. In a literature study, no similar surgeries were reported, but we show that this operation may be feasible in selected patients.

The tensile behavior of two and a half-dimensional (2.5D) woven fabric reinforced Al matrix composites involving residual stress was investigated experimentally and numerically. The residual stress and subsequent tensile behavior of the composites were predicted by multiscale finite element modeling. And the correctness of numerical simulation is verified by residual stress, tensile curve, surface strain diagram and fracture morphology. The results show that the inhomogeneous residual stress leads to the damage of the matrix and interface in the interweaving area of yarns. During the tensile process, the surface strain exhibits an obvious periodic distribution characteristic, which is related to the arrangement of yarns. With the increase of tensile strain, local matrix and interface damage accumulate and expand leading to the weft yarn cracking, matrix failure and interface debonding successively. The abrupt fracture of the composite can be attributed to the axial fracture of warp yarn, which exhibits obvious fiber fracture and pull-out characteristic.

Abstract Hantaan virus (HTNV) is a pathogenic orthohantavirus prevalent in East Asia that is known to cause hemorrhagic fever with severe renal syndrome (HFRS), which has a high fatality rate. However, a Food and Drug Administration (FDA)-approved vaccine is not currently available against this virus. Although inactivated vaccines have been certified and used in endemic regions for decades, the neutralizing antibody (NAb) titer induced by inactivated vaccines is low and the immunization schedule is complicated, requiring at least three injections spanning approximately 6 months to 1 year. Replication-competent vesicular stomatitis virus (VSV)-based vaccines provide prolonged protection after a single injection. In this study, we successfully engineered the HTNV glycoprotein (GP) in the VSV genome by replacing the VSV-G open reading frame. The resulting recombinant (r) rVSV-HTNV-GP was rescued, and the immunogenicity of GP was similar to that of HTNV. BALB/c mice immunized with rVSV-HTNV-GP showed a high titer of NAb against HTNV after a single injection. Notably, the cross-reactive NAb response induced by rVSV-HTNV-GP against Seoul virus (an orthohantavirus) was higher than that induced by three sequential injections of inactivated vaccines. Upon challenge with HTNV, rVSV-HTNV-GP-immunized mice showed a profoundly reduced viral burden in multiple tissues, and inflammation in the lungs and liver was nearly undetectable. Moreover, a single injection of rVSV-HTNV-GP established a prolonged immunological memory status as the NAbs were sustained for over 1 year and provided long-term protection against HTNV infection. The findings of our study can support further development of an rVSV-HTNV-GP-based HTNV vaccine with a simplified immunization schedule.

Abstract Liver kinase B1 (LKB1), an evolutionarily conserved serine/threonine kinase, is a master regulator of the AMPK subfamily and controls cellular events such as polarity, proliferation, and energy homeostasis. Functions and mechanisms of the LKB1-AMPK axis at specific subcellular compartments, such as lysosome and mitochondria, have been established. AMPK is known to be activated at the Golgi; however, functions and regulatory mechanisms of the LKB1-AMPK axis at the Golgi apparatus remain elusive. Here, we show that TBC1D23, a Golgi-localized protein that is frequently mutated in the neurodevelopment disorder pontocerebellar hypoplasia (PCH), is specifically required for the LKB1 signaling at the Golgi. TBC1D23 directly interacts with LKB1 and recruits LKB1 to Golgi, promoting Golgi-specific activation of AMPK upon energy stress. Notably, Golgi-targeted expression of LKB1 rescues TBC1D23 deficiency in zebrafish models. Furthermore, the loss of LKB1 causes neurodevelopmental abnormalities in zebrafish, which partially recapitulates defects in TBC1D23-deficient zebrafish, and LKB1 sustains normal neuronal development via TBC1D23 interaction. Our study uncovers a regulatory mechanism of the LKB1 signaling, and reveals that a disrupted Golgi-LKB1 signaling underlies the pathogenesis of PCH.
